Abstract:Bacillus thuringiensis (Bt) Cry toxins have been widely used in the development of genetically modified organisms (GMOs) for pest control. This work aimed to establish more cost effective and broader detection methods for commonly used Cry toxins. Using ligand blot and bio-layer interferometry, we confirmed that a recombinant toxin-binding fragments derived from Helicoverpa armigera cadherin-like protein (HaCad-TBR) could broadly bind Cry1Ab, Cry1Ac, Cry2Aa, and Cry2Ab with the affinity of 0.149, 0.402, 120, and 4.12 nM, respectively. Based on the affinity results, a novel receptor-antibody sandwich assay broadly detecting Cry1A and Cry2 toxins was developed by using HaCad-TBR as capture molecules, and anti-Cry1A/Cry2A polyclonal antibodies (pAbs) as the detection antibodies. The detection limit (LOD) for Cry1Ab, Cry1Ab, Cry2Aa, and Cry2Ab were 5.30, 5.75, 30.83 and 13.70 ng/mL. To distinguish Cry1A and Cry2A toxins in a singular test, anti-Cry1A pAbs and anti-Cry2A pAbs were labelled with different quantum dots (QDs). The LOD for the four toxins by receptor-QDs-pAbs sandwich assay were calculated to be 1.36, 4.71, 17.48, and 7.54 ng/mL, respectively. The two developed methods were validated by spiked rice and corn samples, suggesting they may potentially be used in monitoring and quantifying Cry toxins in food and environment.

Abstract:Bacillus thuringiensis (Bt) Cry toxins have been widely used in the development of genetically modified organisms (GMOs) for pest control. This work aimed to establish more cost effective methods for used Cry2Aa toxins. Three immunoassay methods (IC-ELISA, DAS-ELISA, and CLEIA) were successfully developed in this work. The mAb was used as the detecting antibody, for the IC-ELISA, the range of IC20 to IC80 was 1.11 μg/mL - 60.70 μg/mL, and an IC50 of 10.65 μg/mL. For the DAS-ELISA, the limit of detection (LOD) and limit of quantitation (LOQ) were 10.76 ng/mL and 20.70 ng/mL, respectively. For the CLEIA, the LOD and LOQ were 6.17 ng/mL and 7.40 ng/mL, respectively. The scFv-based detections were the most sensitive for detecting Cry2Aa. The LOD and LOQ for the DAS-ELISA were 118.75 ng/mL and 633.48 ng/mL, respectively. The LOD and LOQ for the CLEIA, read as 37.47 ng/mL and 70.23 ng/mL, respectively. The fact that Cry2Aa toxin was recovered in spiked grain samples further demonstrated that the approaches might be used to identify field samples. These methods provided good sensitivity, stability, and applicability for detecting Cry2Aa toxin, promising ultrasensitive monitoring and references for Cry toxins risk assessment.

Abstract:Crystal proteins (Cry) from Bacillus thuringiensis (Bt) are well-known pesticidal proteins in genetically modified (GM) crops and often used as the target proteins in detection of GM food. Instead of the classic mouse hybridoma, new rabbit monoclonal antibodies (McAb) were first produced in detection of GM food. Rabbit McAb and polyclonal antibodies (PcAb) were prepared using Bt-Cry1Ac protein as the immune antigen (Ag). Sandwich ELISA with either PcAb as the coating antibody and peroxidase horseradish (HRP) labeled McAb as the detecting antibody (PcAb-Ag-McAb), or McAb as the coating antibody and HRP labeled PcAb as the detecting antibody (McAb-Ag-PcAb), was developed for detecting Bt-Cry1Ac protein. The analytical sensitivity of McAb-Ag-PcAb was higher with the linear detection range of 0.97-62.50 ng mL-1 and the limit of detection (LOD) of 0.24 ng mL-1. There was no cross-reaction with Cry1Ab and Cry1c proteins and the mean recovery was 103.35% in corn leaves. The LOD and the linear detection range of the proposed ELISA are more satisfactory than other ELISA systems using an anti-Cry1Ac PcAb or two Cry1Ac-specific McAb for detection of the target protein and the research provides an effective instrument for detecting Cry1Ac protein in GM crops and their derivatives.
A Fast And Sensitive Quantitative Lateral Flow Immunoassay For Cry1ab Based On A Novel Signal Amplification Conjugate

Abstract:A novel lateral flow immunoassay (LFIA) signal amplification strategy for the detection of Cry1Ab based on amplification via a polylysine (PL) chain and biotin-streptavidin system (BSAS) is described. In this system, multiple fluorescence dyes (FL) were directly coated on the surface of PL and conjugated with antibody via the BSAS for construction of novel signal amplification (FLPL-BSAS-mAb1) conjugates, in which FL, PL and BSAS were employed to improve the sensitivity of LFIA. Compared with conventional LFIA, the sensitivity of FLPL-BSAS-mAb1-based LFIA was increased by approximately 100-fold. Quantified linearity was achieved in the value range of 0-1,000 pg/mL. The limit of detection (LOD) was reached 10 pg/mL after optimization of reaction conditions. To our knowledge, this represents one of the most sensitive LFIA for Cry1Ab yet reported. Furthermore, the detection time for this method was about 10 min. Therefore, it should be an attractive alternative compared to conventional immunoassays in routine control for Cry1Ab.

Abstract:Using the hybridoma cells 2D3 which secrete anti-Cry1 monoclonal antibody as DNA source, the variable region genes of heavy chain (VH) and light chain (VL) were amplified and assembled as single-chain variable fragment (scFv). The scFv-2D3 genes were cloned into pET-26b vectors for expression and scFv-2D3 expression was identified by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and western blot (WB). The activity of scFv-2D3 was determined by indirect enzyme-linked immune sorbent assay (ELISA). As a result, scFv-2D3 was successfully constructed and expressed, with specific recognition activities to the Cry1 toxins. Homologous modeling and molecular docking results showed that VL was the main combination domain of scFv-2D3 with Cry1 toxins. Therefore, VH and VL were separately expressed and then assayed by indirect ELISA, indicating that both VH and VL could recognize the Cry1 toxin, and the activity of VL was higher than that of VH, which confirming the molecular docking result. Afterward, double light chain antibodies (VL-VL) were designed and produced, and the binding activities of VL-VL against Cry1Aa, Cry1Ab, and Cry1Ac were significantly enhanced with OD450 values improved 1.18, 1.44, and 1.22 times, respectively, compared with scFv-2D3. For VL-VL-based double antibody sandwiched ELISA (DAS-ELISA), the limits of detection (LOD) and limits of quantification (LOQ) were 10.5–16.0 and 12.8–21.5 ng mL−1 for seven Cry1 toxins, respectively, which were lower than scFv-2D3 (14.8–29.2 and 39.0–50.3 ng mL−1). The results were beneficial to developing high-throughput and high-sensitive immune-detecting technology for Cry1 toxins.

Abstract:A method of extracting and purifying Cry1Ab protein(Bt toxin) from Cry1Ab transgenic rice was established. Most of the Bt toxin present in the tissue of Cry1Ab transgenic rice was extracted effectively with a solution of 50 mmol/LNa2CO3 and NaHCO3. The crude protein containing Bt toxin was obtained after the pretreatment of Cry1Ab transgenic rice with ultra-filtration, ammonium sulfate precipitation and centrifugation. The dialysed crude protein was futher separated on DEAE Sephadex A-50 columns and Sephadex G-150 columns. The protein bound on DEAE Sephadex A-50 gel was eluted with buffer solution B(10 mmol/L tris-HCl buffer+1.0 mmol/L EDTA, pH=8.0) mixed with 0.1, 0.3, 0.5 and 0.8 mol/l. NaCl in a discontinuous gradient elution mode. The peak of the Bt toxin eluted from the columns was identified by ELISA and bioassayed with larvae of tobacco hornworms and silkworms. The purity and the bioactivity of the Bt toxin were determined by means of SDS-PAGE and larvicidal assay, respectively. The purity of the Bt toxin obtained by this method is high, and its insecticidal activity is retained after the toxin is purified.

Abstract:Traditional ELISA methods of using animal immunity yield antibodies for detection Cry toxin. Not only is this incredibly harmful to the animals, but is also time-intensive. Here we developed a simple method to yield the recognition element. Using a critical selection strategy and immunoassay we confirmed a clone from the Ph.D-C7C phage library, which has displayed the most interesting Cry1Ab-binding characteristics examined in this study (Fig. 1). The current study indicates that isolating peptide is an alternative method for the preparation of a recognition element, and that the developed assay is a potentially useful tool for detecting Cry1Ab.

Abstract:Enzyme-linked immunoadsorbent assay (ELISA) was utilized to probe the conformation change of CryIAb protein expressed by transgenic CryIAb rice based on the change of its binding capacity with antibody resulting from conformation transition. Stability of CryIAb protein in different temperature, solvent composition, pH and different organic solvents were measured. Results showed that the antibody on the micro-well plate of ELISA kits could sensitively recognized conformation transition of CryIAb protein without need of highly purified protein. The conformation change induced by temperature was depended on the solvent composition. Relatively high stability could be achieved in sodium sulfate ( Na2SO4) solution. In contrast, sodium dodecyl sulfate ( SDS) accelerated its temperature-induced conformation transition. At room temperature, 25% ( V/V) organic solvent such as methanol, ethanol, 2-propanol and acetonitrile could significantly cause CryIAb protein conformation change. Among all the solvents tested, the acetonitrile worked strongest. The extent of the transition in alcohols was highly correlation with the hydrophobicity of the solvents. pH also had an effect on the conformation transition of CryIAb protein. The native structure of CryIAb protein was stable in pH 8 similar to 10. Too acidic or alkaline media destabilized the protein as. demonstrated by the decrease of ELISA results. Moreover, humic acids from soil could remarkably stabilize the conformational of CryIAb protein even in acidic condition.

Abstract:In order to practice the concept of science education cooperation， in the experimental teaching of pathogenic biology and immunology， a comprehensive experimental project is designed for the optimization and application of antibody microarray in the multiple mycotoxins determination. Based on the principle of competitive reaction mode， the antibody microarray layout and reaction hardware devices are designed， as well as the horseradish peroxidase labeled mycotoxin complete antigen is prepared. After optimizing the relevant reaction conditions， the antibody microarray that can realize multiple detection of mycotoxins is established. The results show that this immunological detection method can realize the simultaneous quantitative detection of four mycotoxins with high sensitivity and stability. This comprehensive experiment combines the prevention and control needs of mycotoxins with research hotspots of immune detection technology， which can deepen students’ understanding and mastery of classroom knowledge and enhance the comprehensiveness and systematisms of learning. Compared with confirmatory experiments， this comprehensive experiments integrated with scientific research achievements can consolidate students’ theoretical knowledge and experimental operation skills， cultivate and stimulate their innovative thinking and research enthusiasm， improve their comprehensive quality.
Photoelectrochemical and visual dual-mode sensor for efficient detection of Cry1Ab protein based on the proximity hybridization driven specific desorption of multifunctional probe

Abstract:Currently, the development of sensitive and visual strategy for Cry1Ab detection, particularly using a switchable dual-mode detection system based on a single component, remains a great challenge. Here, a photoelectrochemical (PEC) and visual dual-mode sensor was designed for Cry1Ab detection based on a proximity hybridization driven multifunctional probe. In the presence of Cry1Ab, specific desorption of the antibody-DNA conjugate was achieved via sufficient proximity hybridization, leading to the selective release of the multifunctional signal probe, i.e., antibody-labeled single-stranded DNA-gold nanoparticles (Ab1-S1-AuNPs). The released Ab1-S1-AuNPs reduced the photocurrent signal and produced a colored response, thereby achieving PEC and visual dual-mode detection based on a single component. Owing to the different signal generation mechanisms, two independent signals were obtained simultaneously, which provided self-verification to improve reliability and accuracy. Taking advantage of the PEC sensitive detection and visual prediction, the dual-mode sensor achieved efficient detection of the Cry1Ab protein. The developed sensor was successfully used to determine Cry1Ab in corn, wheat, and soil samples with satisfactory results. This method offers a promising biosensing platform for the on-site detection of Cry1Ab protein.

Abstract:The azygos venous system of one Weddell seal (Leptonychotes weddelli) has been described. Both left and right azygos trunks were present with segmental anastomoses between them. The right azygos trunk was the larger and opened into the anterior vena cava; the left azygos trunk terminated in the left costocervical vein. At each segmental level the azygos trunks communicated with collecting sinuses into which drained blood from intercostal spaces and the extradural intravertebral vein. These collecting sinuses were themselves connected to each other by longitudinal anastomoses. The relationship of this system to blood flow during diving is briefly discussed.

Abstract:A novel method for the immunoassay of genetically modified organisms (GMOs) protein Cry1Ab was proposed. First, nano magnetic beads with the diameters around 400 nm were prepared by the hydrothermal method. Then, the magnetic beads prepared were modified with -NH2 group using the sol-gel approach. The antibody was conjugated on the surface of magnetic beads through bi-functional agent glutaraldehyde. The immunoassay was carried out in the commercial nucleic acid extractor. The detection limit of this method for Cry1Ab is as low as 1 ppb, which is comparable to that of the common ELISA methods. This automatic way is promising because of its rapidity and simplicity.

Sequence analysis revealed that one gene encoded a polypeptide of 673 amino acid residues with a molecular mass of 76.3 kDa, 38% identical to Cry10Aa, and the other encoded a polypeptide of 687 amino acid residues with a molecular mass of 77.1 kDa, 74% identical to Cry30Aa. These two novel crystal protein genes were designated as and by Bt Insecticidal Crystal Proteins Nomenclature Committee, respectively. The Cry54Aa1 and Cry30Fa1 proteins retained five conserved regions commonly found in the existing Cry proteins. Cry54Aa1 protein exhibited insecticidal activities against (Lepidoptera), (Lepidoptera), and (Diptera) when its encoding gene was expressed in an host strain.
Purification of Cry1Ab protein by multi-cycles ion-exchange chromatography and its peptide mass mapping

Abstract:The coarse purified Cry1Ab toxin was further purified on Q-Sepharose FF ion-exchange column by multi-cycles separation method.Ion strength of the mobile phase increased stepwise in the elution process,in which the concentration of NaCl increased from 0.1 mol·L-1 to 1.0 mol·L-1 dissolved in Tris-HCl buffer.Results showed that Cry1Ab protein could be eluted when the concentration of NaCl reached 0.4 mol·L-1.and the chromatographic mechanism might be in accordance with the molecular replacing mechanism.The highly purified Cry1Ab protein could be obtained after the protein sample was separated by three cycles of the chromatographic process.The purity and apparent molecular weight of the protein were determined by SDS-PAGE.The protein band on the gel was in-situ digested by trypsin,and the peptide mass mapping was analyzed by MALDI-TOF.The resulting peptides mass was searched on Mascot database,and 80% coverage with known Cry1Ab protein was found.

Abstract:Impedimetric analysis method is an important tool for food safety detection. In this work, a novel impedimetric microfluidic analysis system consisted of a printed gold electrode chip and a microfluidic flow cell was developed for sensitive and selective detection of transgenic protein Cry1Ab. AntiCry1Ab aptamer coated magnetic beads were used to recognize transgenic protein Cry1Ab and form Cry1Ab-aptamer modified magnetic beads. After separation, the obtained Cry1Ab-aptamer modified magnetic beads were dissolved in 0.01 M mannitol and followed by injection into the microfluidic flow cell for impedimetric measurement. At the frequency of 358.3 Hz, the impedance signal shows a good linearity with the concentrations of Cry1Ab protein at a range from 0 to 0.2 nM, and the detection limit is 0.015 nM. The results demonstrate that the impedimetric microfluidic analysis system provides an alternative way to enable sensitive, rapid and specific detection of transgenic protein Cry1Ab.
Phage-displayed nanobody-based fluorescence-linked immunosorbent assay for the detection of Cry3Bb toxin in corn

Abstract:Cry toxins have been widely used as biopesticides and applied in transgenic crops for the control of pests, raising public concerns for unforeseen environmental and health risks. In this study, five phage-displayed nanobodies against Cry3Bb toxin were selected from a naive nanobody library by screening. Using purified phage-displayed nanobody as capture antibody, a novel double-antibody sandwich fluorescence-linked immunosorbent assay (FLISA) based on quantum dot beads (QBs) was established for the determination of Cry3Bb toxin. The developed method exhibited a linear working range of 31.25–500 ng/mL and a low detection limit of 8.45 ng/mL. The recoveries of Cry3Bb toxin from spiked corn samples were ranging from 93.1% to 116.8% with a coefficient of variation lower than 9.4%. Molecular modeling results showed that key amino acids in the CDR3 region of nanobody P27 combine with Cry3Bb toxin through hydrogen bonds and hydrophobic interactions. These results indicated that the phage-displayed nanobody-based QBs-FLISA may provide a possible tool for detecting Cry3Bb toxin in corn.

Abstract:The diamondback moth, Plutella xylostella (L.), is a worldwide pest of cruciferous crops and can rapidly develop resistance to many chemical insecticides. Although insecticidal crystal proteins (i.e., Cry and Cyt toxins) derived from Bacillus thuringiensis (Bt) have been useful alternatives to chemical insecticides for the control of P. xylostella, resistance to Bt in field populations of P. xylostella has already been reported. A better understanding of the resistance mechanisms to Bt should be valuable in delaying resistance development. In this study, the mechanisms underlying P. xylostella resistance to Bt Cry1Ac toxin were investigated using two-dimensional differential in-gel electrophoresis (2D-DIGE) and ligand blotting for the first time. Comparative analyses of the constitutive expression of midgut proteins in Cry1Ac-susceptible and -resistant P. xylostella larvae revealed 31 differentially expressed proteins, 21 of which were identified by mass spectrometry. Of these identified proteins, the following fell into diverse eukaryotic orthologous group (KOG) subcategories may be involved in Cry1Ac resistance in P. xylostella: ATP-binding cassette (ABC) transporter subfamily G member 4 (ABCG4), trypsin, heat shock protein 70 (HSP70), vacuolar H(+)-ATPase, actin, glycosylphosphatidylinositol anchor attachment 1 protein (GAA1) and solute carrier family 30 member 1 (SLC30A1). Additionally, ligand blotting identified the following midgut proteins as Cry1Ac-binding proteins in Cry1Ac-susceptible P. xylostella larvae: ABC transporter subfamily C member 1 (ABCC1), solute carrier family 36 member 1 (SLC36A1), NADH dehydrogenase iron-sulfur protein 3 (NDUFS3), prohibitin and Rap1 GTPase-activating protein 1. Collectively, these proteomic results increase our understanding of the molecular resistance mechanisms to Bt Cry1Ac toxin in P. xylostella and also demonstrate that resistance to Bt Cry1Ac toxin is complex and multifaceted.
