Abstract:Fish oils are a rich source of polyunsaturated fatty acids, including eicosapentaenoic acid and docosahexaenoic acid, which are reported to exhibit therapeutic effects in a variety of human diseases. However, these oils are highly susceptible to degradation due to oxidation, leading to rancidity and the formation of potentially toxic reaction products. The aim of this study was to synthesize a novel emulsifier (HA-PG10-C18) by esterifying hyaluronic acid with poly(glyceryl)10-stearate (PG10-C18). This emulsifier was then used to formulate nanoemulsion-based delivery systems to co-deliver fish oil and coenzyme Q10 (Q10). Q10-loaded fish oil-in-water nanoemulsions were fabricated, and then their physicochemical properties, digestibility, and bioaccessibility were measured. The results indicated that the environmental stability and antioxidant activity of oil droplets coated with HA-PG10-C18 surpassed those coated with PG10-C18 due to the formation of a denser interfacial layer that blocked metal ions, oxygen, and lipase. Meanwhile, the lipid digestibility and Q10 bioaccessibility of nanoemulsions formulated with HA-PG10-C18 (94.9 and 69.2%) were higher than those formulated with PG10-C18 (86.2 and 57.8%), respectively. These results demonstrated that the novel emulsifier synthesized in this study could be used to protect chemically labile fat-soluble substances from oxidative damage, while still retaining their nutritional value.
Novel Multi-Phase Nano-Emulsion Preparation for Co-Loading Hydrophilic Arbutin and Hydrophobic Coumaric Acid Using Hydrocolloids

Abstract:This study was designed to co-load hydrophilic arbutin and hydrophobic coumaric acid in nano-sized multi-phase emulsion (W/O/W) using hydrocolloids and examined for its various stability and bioaccessibility parameters. The creaming stability and sensory evaluation of emulsion in Lactobacillus beverage and its release kinetics in simulated oral and gastrointestinal conditions were also studied. Results revealed that multi-phase emulsion exhibited small particle size and low polydispersity index with high zeta potential. Higher encapsulation efficiency was also observed. Moreover, the multi-phase emulsion showed good storage stability under different conditions. The multi-phase emulsion also showed controlled release of arbutin and coumaric acid in simulated conditions with increasing bioaccessibility. Stability of these compounds under unfavorable conditions was also found to be increased. Moreover, high creaming stability and sensory evaluation score of the multi-phase emulsion suggested that the formulation was capable of retaining arbutin and coumaric acid in Lactobacillus beverage without affecting its sensory properties. These results indicated that the multi-phase nano-emulsion (W/O/W) could effectively co-deliver hydrophilic arbutin and hydrophobic coumaric acid and thus could be a novel delivery method for increasing bioaccessibility of these nutraceutically important phenolic compounds.
Synthesis and Characterization of Fucoidan/chitosan-Coated Nanoliposomes for Enhanced Stability and Oral Bioavailability of Hydrophilic Catechin and Hydrophobic Juglone

Abstract:This study aimed to improve the thermodynamic performance of nanoliposomes (NLs) using fucoidan (F) as the second-layer coating biopolymer along with chitosan (CS), to control the delivery and bioavailability of catechin (C) and juglone (J). The stabilized liposomal carrier of F/CS-conjugated JC-NL (F-CS-JC-NL) was developed with optimum concentrations of CS (0.09 wt%) and F (0.10 wt%), with the highest encapsulation efficiency of juglone (95.47%) and catechin (90.88%). Physicochemical characterization revealed that F-CS-JC-NL disclosed improved stability under different pH and ionic strengths, with the maximum juglone/catechin retention under thermal, oxidative and storage conditions. In vitro digestion revealed that NL double-coating (F-CS-JC-NL) significantly reduced compound leakage in the gastrointestinal tract, providing a controlled release and better bioavailability of juglone/catechin compared to CS-JC-NL and JC-NL. Conclusively, this study provides a novel NL-based de-livery carrier with enhanced physicochemical stability and controlled release that might have promising use in delivering functional ingredients.

Abstract:O/W emulsion and its spray-dried microcapsule contain the oil phase and the protein matrix, providing the potential to co-encapsulate different antioxidants. However, antioxidants were generally encapsulated in the oil phase of microcapsule, its protein matrix is rarely used. It is first to prove the possibility to encapsulate resveratrol in the emulsified oil droplets at high wall/core ratios. The optimal microcapsule with 1.75% surface oil was fabricated with 15% transglutaminase-crosslinked WPI (TGase-WPI) and 5% gum Arabic (GA). Resveratrol mainly located in the protein matrix of initial emulsion and reconstituted microcapsule. The effects of TGase-WPI/GA microcapsule and resveratrol co-encapsulation on DHA/EPA and lipid hydroperoxides/TBARS were different. The interfacial protein, the partition of resveratrol in the emulsified oil droplets and its storage stability and inhibitory effect on size change of reconstituted microcapsules increased as the polyphenol increased. These results expand the potential use of spray-dried microcapsules as co-encapsulation carriers.

Abstract:Resveratrol (Res), a non-flavonoid polyphenolic compound extracted from natural plants, has many pharmacological effects such as anti-tumor, organ protection, improvement of cardiovascular disease, and anti-inflammation. However, due to its poor water solubility and light-degradable property, its bioavailability is restricted. In this study, the hyaluronic acid (Ha)-coated poloxamer 188 (F68) and poloxamer 407 (F127) and Ha-coated Res nanomicelles (Ha-F68/F127-Res-Mc) were prepared by film dispersion method. The nanomicelles were evaluated by indicators of encapsulation efficiency ( EE ), drug loading ( DL ), particle size, polydispersity index and ζ-potential. Besides, using the Box–Behnken design (BBD) based response surface methodology (RSM), obtained the optimum prescription process to obtain the Ha-F68/F127-Res-Mc. Compared with the Res active pharmaceutical ingredient (API), Ha-F68/F127-Res-Mc had higher water solubility and photostability, and could achieve sustained release. Compared with the Res active pharmaceutical ingredient (API), Ha-F68/F127-Res-Mc had higher water solubility and photostability, and could achieve sustained release. In conclusion, the prepared nanomicelles have stronger cytotoxic effect on MCF-7 than doxorubicin (DOX). This will provide a basis for the development of Res antitumor injection, and provide a reference for the research on nano-delivery systems of other BCS II drugs in the future.

Abstract:Resveratrol (RES) is an important functional substance with multiple active properties. However, RES is susceptible to natural environmental conditions that reduce its bioactivity. To improve the bioavailability of RES, in this study, Catechin and Fe3+/Ca2+ were selected to form supramolecules, which were then coated on the surface of hydrophobic RES nanoparticles (RES NPs) to create composite RES NPs. The obtained composite RES NPs demonstrated higher antioxidant capacity and better photo-thermal stability than RES NPs. Additionally, a pectin (PE) dietary gel was designed as a delivery carrier for RES. The results showed that the incorporation of composite RES NPs not only endowed the gels with significant dietary activity but also enhanced the texture, water retention capacity and hydrophobicity. After 28 days of storage, the retention rate of RES could be maintained above 90 % in the dietary gels. Meanwhile, the controlled release of RES was achieved in in vitro simulated digestion.
Preparation and Characterization of Fish Oil Pickering Emulsions Stabilized by Resveratrol-Loaded Gliadin/Chitin Nanocrystal Composite Nanoparticles

Abstract:Unsaturated fatty acids present in fish oil offer various physiological benefits to the human body. However, their susceptibility to oxidation severely limits their potential applications. The purpose of this study was to develop Pickering emulsions stabilized from a composite of resveratrol-loaded gliadin nanoparticles and oxidized chitin nanocrystals (GR/OC) to protect fish oil from oxidation. The effects of the GR/OC composite on the characterizations of fish oil Pickering emulsions were investigated, including the microstructure, physicochemical properties (stability and rheological behavior), and digestion properties in vitro. The results revealed that an increased concentration of the GR/OC composite significantly reduced the droplet size and improved the ambient stability of the emulsions (in terms of pH, ionic strength, temperature, and storage time). Confocal laser scanning microscopy images depicted that the GR/OC nanoparticles were uniformly dispersed at the interface between water and fish oil (W-O interface). This distribution formed a protective envelope around the droplets. Remarkably, the addition of 2% GR/OC nanoparticles stabilized the Pickering emulsions and showed the most positive effect on the antioxidant capacity compared to that of the control group. These stabilized emulsions maintained lower peroxide values and acid values, which were 1.5 times less than those of the blank control during the 14 day accelerated oxidation experiment. Furthermore, the Pickering emulsions stabilized by GR/OC nanoparticles exhibited the ability to protect fish oil from contamination by gastric juices and facilitate the intestinal absorption of omega-3 polyunsaturated fatty acids. The findings suggest that these GR/OC-stabilized Pickering emulsions offer a promising alternative for delivering fish oils in various industries, including the food industry.

Abstract:A patient who developed primary brain lymphoma 6 years following whole brain irradiation due to a low-grade glioma is described. The patient had no evidence of congenital or acquired immunodeficiency state and achieved a good and prompt response to aggressive chemotherapy, including high-dose methotrexate. The previous radiation therapy is implicated in the etiology of the lymphoma because of the geometric coincidence, the relatively long latency period and the different histology. A brief review of current literature is reported.

Abstract:Liposome-based drug delivery systems have been widely used in drug and gene delivery. However, issues such as instability, immune clearance, and poor targeting have significantly limited their clinical utility. Consequently, there is an urgent need for innovative strategies to improve liposome performance. In this study, we explore the interaction mechanisms of hyaluronic acid (HA), a linear anionic polysaccharide composed of repeating disaccharide units of d-glucuronic acid and N-acetyl-d-glucosamine connected by alternating β-1,3 and β-1,4 glycosidic linkages, and its octanoylated derivates (OHA) with liposomes using extensive coarse-grained molecular dynamics simulations. The octyl moieties of OHA spontaneously inserted into the phospholipid bilayer of liposomes, leading to their effective coating onto the surface of liposome and enhancing their structural stability. Furthermore, encapsulating liposome with OHA neutralized their surface potential, interfering with the formation of a protein corona known to contribute to liposomal immune clearance. Importantly, the encapsulated OHA maintained its selectivity and therefore targeting ability for CD44, which is often overexpressed in tumor cells. These molecular-scale findings shed light on the interaction mechanisms between HA and liposomes and will be useful for the development of next-generation liposome-based drug delivery systems.
Improvement of NaCas/DGMO complex emulsion on resveratrol stability, in vitro bioaccessibility, in vivo bioavailability and gut microbiota

Abstract:Evaluation for biological impact of food emulsions is fundamental for their application. In present study, we utilized a NaCas-DGMO (sodium caseinate-decylglycerol monooleate) stabilized emulsion to improve resveratrol's (Res) stability, and bioavailability. The in vivo interaction between complex emulsion and gut microbiota was further explored. Results indicated NaCas-DGMO emulsion achieved a loading rate of 92 % for Res and significantly enhanced storage and photo stability of Res. In vitro gastrointestinal digestion highlighted a significant improvement in Res's bioaccessibility. In vivo pharmacokinetic tests showed a notable 3.1-fold increase in oral bioavailability, with a prolonged Tmax of 6 h post-administration. Gut microbiota analysis revealed that the emulsion promoted beneficial bacteria, like Blautia, which produce short-chain fatty acids. Consequently, the findings proved potential of NaCas-DGMO stabled emulsion as carriers for bioactive substances in the food industry. The innovative methodology employed in assessing biological effects provides valuable insights for future research in related field.

Abstract:The aim of this study was to optimize the formation of sodium caseinate (CS) and gum arabic (GA) complexes through the Maillard reaction and to evaluate their effectiveness in improving the emulsification properties and stability of docosahexaenoic acid (DHA) nanoemulsions. First, the best target polysaccharides were selected, and the best modification conditions were determined using orthogonal experiments. Secondly, the response surface experiments were used to optimize the preparation process of the emulsion. The stability, in vitro digestion characteristics, and rheological characteristics of the emulsion prepared by means of CS-GA were compared with the emulsion prepared using a whey protein isolate (WPI). After the orthogonal test, the optimal modification conditions were determined to be a reaction time of 96 h, a CS-GA mass ratio of 1:2, a reaction temperature of 60 °C, and a degree of grafting of 44.91%. Changes in the infrared (IR), Raman, ultraviolet (UV), and endogenous fluorescence spectra also indicated that the complex structure was modified. The response surface test identified the optimal preparation process as follows: an emulsifier concentration of 5 g/L, an oil-phase concentration of 5 g/L, and a homogenization frequency of five, and the emulsion showed good stability. Therefore, the use of a nanoemulsion as a nanoscale DHA algal oil delivery system is very promising for extending the shelf life and improving the stability of food.

Abstract:In recent years, active substances have been extensively applied in the fields of food, cosmetics, and pharmaceuticals. However, their preservation and transportation have posed challenges due to issues such as oxidation and photodegradation. This study proposes a method for synthesizing Zein-Hyaluronic Acid (Zein-HA) conjugate particles via the Schiff base reaction, utilizing these conjugate particles to encapsulate and protect active substances within a stable emulsion system. Compared to zein, the modified conjugate particles exhibit significantly improved dispersibility, amphiphilicity, interfacial affinity, and emulsifying properties. Consequently, these particles are capable of stabilizing high internal phase Pickering emulsions with an oil phase volume fraction of up to 80 ( v /v)%, thereby enabling the carriage of a higher load of active components. Furthermore, the prepared emulsions demonstrate excellent storage stability, resistance to ionic strength (250–2000 mM NaCl), and outstanding antioxidative characteristics. Moreover, after 8 h of UV light exposure, the retention rates of the active substances (curcumin, astaxanthin, and resveratrol) exceed 60 %. Therefore, these emulsions hold substantial potential to be applied as a carrier system in the food, cosmetics, and pharmaceutical industries.

Abstract:Novel nanoparticles ( Z -R/H) were successfully fabricated by a resveratrol-grafted zein covalent conjugate (Z-R) combined with quaternary ammonium chitosan (HTCC), which were used as stabilizers to prepare peppermint oil (PO) Pickering emulsions with antioxidant activity. HTCC effectively adjusted wettability of Z -R conjugate, and three-phase contact angle of Z-R/H 3:1 was moderate (95.01°). The influencing factors of Pickering emulsion formation, including volume fraction of PO, concentration of Z -R/H, and mass ratio of Z-R to HTCC, were evaluated by droplet size, ζ-potential, microscopic observation, and stability index analysis. Pickering emulsions stabilized by Z -R/H 3:1 showed excellent physical stability under heat treatment. Z-R/H nanoparticles adsorbed on the oil-water interface yielded a dense filling layer as a physical barrier to improve the emulsion stability, which was validated by confocal laser-scanning microscopy. After 4 weeks of storage, retention rate of PO in Pickering emulsion stabilized by Z -R/H 3:1 remained high (72.1 %). Electronic nose analysis showed that Z -R/H 3:1 -stabilized emulsion effectively prevented volatilization of PO aroma components. Additionally, PO and Z-R/H nanoparticles provided an additive antioxidant effect of Pickering emulsions against DPPH and ABTS free radicals. In summary, these novel Z -R/H nanoparticle offer promising applications as a stabilizer with great potential in preparing functional Pickering emulsions to improve essential oil delivery.

Abstract:High internal phase Pickering emulsions (HIPPEs) stabilized with octenyl succinic anhydride starch/chitosan complexes were examined as a topical delivery vehicle for resveratrol. All resveratrol-loaded HIPPEs showed stable gel-like network structures, with the droplet size and microrheological properties largely dependent on the complex concentrations. HIPPEs exhibited strong stability when subjected to light, high temperature, UV radiation and freeze-thaw treatment, and resveratrol retention was greatly improved with the increasing addition of complexes and resveratrol. High amounts of resveratrol facilitated the antioxidant activity of HIPPEs, whereas sustained release of resveratrol was mainly related to the existence of complex interfacial layers. Moreover, HIPPEs overcome the stratum corneum barrier, with an approximately 3–5-fold increase in resveratrol deposition in deep skin compared to bulk oil. In conclusion, the emulsion composition (especially at the particle level) was vital for the effectiveness of HIPPEs as a carrier, which may provide new opportunities to design topical delivery systems.

Abstract:The application of many hydrophilic and hydrophobic nutraceuticals is limited by their poor solubility, chemical stability, and/or bioaccessibility. In this study, a novel Pickering high internal phase double emulsion co-stabilized by modified pea protein isolate (PPI) and sodium alginate (SA) was developed for the co-encapsulation of model hydrophilic (riboflavin) and hydrophobic (β-carotene) nutraceuticals. Initially, the effect of emulsifier type in the external water phase on emulsion formation and stability was examined, including commercial PPI (C-PPI), C-PPI-SA complex, homogenized and ultrasonicated PPI (HU-PPI), and HU-PPI-SA complex. The encapsulation and protective effects of these double emulsions on hydrophilic riboflavin and hydrophobic β-carotene were then evaluated. The results demonstrated that the thermal and storage stabilities of the double emulsion formulated from HU-PPI-SA were high, which was attributed to the formation of a thick biopolymer coating around the oil droplets, as well as thickening of the aqueous phase. Encapsulation significantly improved the photostability of the two nutraceuticals. The double emulsion formulated from HU-PPI-SA significantly improved the in vitro bioaccessibility of β-carotene, which was mainly attributed to inhibition of its chemical degradation under simulated acidic gastric conditions. The novel delivery system may therefore be used for the development of functional foods containing multiple nutraceuticals.

Abstract:Self‐assembly and application performance of ferulic acid modified sodium hyaluronate Hyaluronic acid (HA) is a mucopolysaccharide alternately linked by glucuronic acid and n‐acetylglucosamine, which is usually used as a moisturizer in cosmetics. In this study, HA derivative (FA‐HA) was prepared by modifying sodium hyaluronate with ferulic acid (FA). The chemical and morphological structure of FA‐HA were characterized by UV, FTIR, 1H‐NMR, DLS and TEM. The results showed that FA was grafted onto HA chains via ester bonds and self‐assembled into nanoparticles in aqueous solution. The introduction of FA endowed the nanoparticles with antioxidant function. The FRAP value of 10 mg/mL FA‐HA was 1.985 mM, and the clearance rate of O2−, DPPH+ and ABTS+ could reach 83.13%, 88.86% and 99.48% respectively. Moreover, FA‐HA can be used as Pickering emulsifiers to stabilize the emulsion. The results showed that the size of emulsion droplets decreased as the concentration of FA‐HA increased. When the mass ratio of oil to water was 7/3, the emulsion droplets were the smallest. The addition of sodium chloride will increase the size of emulsion droplets, destroying the stability of the emulsion. In addition, FA‐HA was also universal and was capable of emulsifying alkanes, esters, and natural oils, which was expected to be applied in the cosmetics and food fields.

Abstract:Emulsion gels synergistically stabilized by different polysaccharides and modified shea butter were reported in this work. The effects of different oil phase contents and polysaccharide types on particle size and rheological properties of emulsion gels were investigated. Modified shea butter molecules are closely arranged on the surface of oil droplets, polysaccharide molecules can be combined and twined with modified shea butter molecules by hydrogen bonding, and gel matrix formed by dissolving in water can suspend, support and immobilize droplets to enhance emulsifying ability. Emulsion gels exhibited good environmental stability against prolonged storage, heating and freezing. These emulsion gels were used to encapsulate and deliver α-tocopherol acetate. The encapsulation efficiency was higher over 30 days compared with the unencapsulated olive oil control group. This research would be of importance not only for extending the knowledge about emulsion gels by modified natural vegetable oil and polysaccharides, but also for the development of novel, eco-friendly and sustainable emulsion gels for food, cosmetic and medicine formulations.

Hybrid Shear-thinning Hydrogel Integrating Hyaluronic Acid with ROS-Responsive Nanoparticles

Abstract:Nanoparticle (NP) supra-assembly offers unique opportunities to tune macroscopic hydrogels' mechanical strength, material degradation, and drug delivery properties. Here, synthetic, reactive oxygen species (ROS)-responsive NPs are physically crosslinked with hyaluronic acid (HA) through guest-host chemistry to create shear-thinning NP/HA hydrogels. A library of triblock copolymers composed of poly(propylene sulfide)-bl-poly(N,N-dimethylacrylamide)-bl-poly(N,N-dimethylacrylamide-co-N-(1-adamantyl)acrylamide) are synthesized with varied triblock architectures and adamantane grafting densities and then self-assembled into NPs displaying adamantane on their corona. Self-assembled NPs are mixed with β-cyclodextrin grafted HA to yield eighteen NP/HA hydrogel formulations. The NP/HA hydrogel platform demonstrates superior mechanical strength to HA-only hydrogels, susceptibility to oxidative/enzymatic degradation, and inherent cell-protective, antioxidant function. The performance of NP/HA hydrogels is shown to be affected by triblock architecture, guest/host grafting densities, and HA composition. In particular, the length of the hydrophilic second block and adamantane grafting density of self-assembled NPs significantly impacts hydrogel mechanical properties and shear-thinning behavior, while ROS-reactivity of poly(propylene sulfide) protects cells from cytotoxic ROS and reduces oxidative degradation of HA compared to HA-only hydrogels. This work provides insight into polymer structure-function considerations for designing hybrid NP/HA hydrogels and identifies antioxidant, shear-thinning hydrogels as promising injectable delivery platforms for small molecule drugs and therapeutic cells.
