Abstract:The heterotrophic nitrification-aerobic denitrification (HNAD) process can remove nitrogen and organic carbon under aerobic conditions. To get the in-depth mechanism of the HAND process, a strain named Acinetobacter johnsonii ZHL01 was isolated, and enzyme activity, electron transport, energy production, and gene expression of the strain were studied with small-molecule carbon sources, including sodium citrate, sodium acetate, sodium fumarate, and sodium succinate. The HNAD pathway of ZHL01 was NH4+→NH2OH → NO, and nitrogen balance analysis shows that ZHL01 could assimilate and denitrify 58.29 ± 1.05 % and 16.58 ± 1.07 % of nitrogen, respectively. The assimilation, the nitrification/denitrification, and the respiration processes were regulated by the concentration of reduced nicotinamide adenine dinucleotide (NADH) produced from the different metabolic pathways of small-molecule carbon sources. The HNAD process occurs to reduce intracellular redox levels related to NADH concentrations. This discovery provides a theoretical basis for the practical application of HAND bacteria.

Abstract:The heterotrophic nitrification and aerobic denitrification (HNAD) process can simultaneously remove nitrogen and chemical oxygen demand under completely aerobic conditions, which has more advantages than the conventional nitrogen removal process. This review summarized the history of the HNAD process, and the distribution of the microorganisms which can accomplish the HNAD process, and then, elucidated the effects of carbon source, C/N ratio, temperature, dissolved oxygen (shaking speed), heavy metals, salinity, and antibiotic on the performance of the HNAD bacteria. Moreover, the metabolic pathways and the mechanism of the HNAD process are discussed. At last, the problems and outlook relating to the study of the HNAD process were presented. In the HNAD process, the metabolisms of organic matter, heterotrophic nitrification, and aerobic denitrification are interlinked via the electron transportation systems which makes the HNAD process more complicated. Revealing the driven mechanism of the HNAD process is crucial for the application of the HNAD process in the future.

Heterotrophic nitrification processes driven by glucose and sodium acetate: New insights into microbial communities, functional genes and nitrogen metabolism from metagenomics and metabolomics

Abstract:Heterotrophic nitrification (HN) bacteria use organic carbon sources to remove ammonia nitrogen (NH4+-N); however, the mechanisms of carbon and nitrogen metabolism are unknown. To understand this mechanism, HN functional microbial communities named MG and MA were enriched with glucose and sodium acetate, respectively. The NH4+-N removal efficiencies were 98.87 % and 98.91 %, with 88.06 % and 69.77 % nitrogen assimilation for MG and MA at 22 h and 10 h, respectively. Fungi (52.86 %) were more competitive in MG, and bacteria (99.99 %) were dominant in MA. Metagenomic and metabolomic analyses indicated that HN might be a signaling molecule (NO) in the production and detoxification processes when MG metabolizes glucose (amo, hao, and nosZ were not detected). MA metabolizes sodium acetate to produce less energy and promotes nitrogen oxidation reduction; however, genes (hao, hox, and NOS2) were not detected. These results suggest that NO and energy requirements induce microbial HN.
Genomics and nitrogen metabolic characteristics of a novel heterotrophic nitrifying-aerobic denitrifying bacterium Acinetobacter oleivorans AHP123

Abstract:A novel aerobic strain of Acinetobacter oleivorans AHP123 was isolated from activated sludge, which could conduct heterotrophic nitrification and denitrification simultaneously. This strain has excellent NH4+-N removal ability, with 97.93% removal rate at 24-hour. To identify the metabolic pathways of this novel strain, genes of gam, glnA, gdhA, gltB, nirB, nasA, nar, nor, glnK and amt were detected by genome analysis. Through RT-qPCR, it was found that the expression of key genes confirmed two possible ways of nitrogen removal in strain AHP123: nitrogen assimilation and heterotrophic nitrification aerobic denitrification (HNAD). However, the absence of some common HNAD genes (amo, nap and nos) suggested that strain AHP123 might have a different HNAD pathway from other HNAD bacteria. Nitrogen balance analysis revealed that strain AHP123 assimilated most of the external nitrogen sources into intracellular nitrogen.

Abstract:Anaerobic ammonium oxidation (anammox) is a biological process by which ammonium is oxidized to dinitrogen gas by using nitrite as the electrons acceptor. Anaerobic ammonium-oxidizing bacteria play an important role in nitrogen removal from wastewater and global N-cycle. The study of metabolism of anammox bacteria will help us understand the anammox mechanism and develop anammox biotechnology. Anammox bacteria are chemoautotrophic bacteria that use CO, or HCO3- as carbon source and obtain their energy from the conversion of ammonium and nitrite into dinitrogen gas. Hydrazine has been detected as an intermediate in the anammox pathway, while hydroxylamine and nitric oxide have not been detected yet. The genomic data indicate that anammox bacteria fix carbon dioxide through acetyl-CoA pathway. The proposed anammox pathway is consistent with the available experimental data, thermodynamical calculation and biochemical determination and as well as the Ockham's razor principal.

Abstract:Ammonium (NH4+)is not only nitrogen source that can support methanotrophic growth, but also it can inhibit methane (CH4) oxidation by competing with CH4 for the active site of methane monooxygenase. NH4+ conversion and its feedback effect on the growth and activity of methanotrophs were evaluated with Methylosinus sporium used as a model methanotroph. Nitrogen sources could affect the CH4-derived carbon distribution, which varied with incubation time and nitrogen concentrations. More CH4-derived carbon was incorporated into biomass in the media with NH4+-N, compared to nitrate-nitrogen (NO3--N), as sole nitrogen source at the nitrogen concentrations of 10-18 mmol L-1. Although ammonia (NH3) oxidation activity of methanotrophs was considerably lower, only accounting for 0.01-0.06% of CH4 oxidation activity in the experimental cultures, NH4+ conversion could lead to the pH decrease and toxic intermediates accumulation in the their habits. Compared with NH4+, nitrite (NO2-) accumulation in the NH4+ conversion of methanotroph had stronger inhibition on its activity, especially the joint inhibition of NO2- accumulation and the pH decrease during the NH4+-N conversion. These results suggested that more attention should be paid to the feedback effects of NW conversion by methanotrophs to understand effects of NH4+ on CH4 oxidation in the environments. Abstract:Although heterotrophic nitrification-aerobic denitrification (HN-AD) is promising in nitrogen removal, it remains unclear for most HN-AD strains in physiological characteristics and metabolic mechanisms. In this study, a newly isolated strain Acinetobacter sp. Z1 converted not only inorganic nitrogen, but also organic nitrogen to N 2 . Among them, urea was the preferential nitrogen substrate. Single-factor experiments showed that efficient HN-AD process occurred with acetate as carbon source, C/N ratios of 12 for NH 4 + -N and 15 for NO 3 – -N, pH 8, 30 °C, DO of ∼5.8 mg/L and salinity less than 1.5 %. Subsequently, response surface analysis was applied to predict the optimal growth conditions. Its complete genome annotation in combination with enzymatic activity assay and nitrogen balance calculation showed that at least four pathways involved in nitrogen metabolism. This work indicates that ureolytic strain Z1 could be prepared as bacterial agents with other HN-AD strains to treat urea-containing wastewater like urine from urban community.

Abstract:Combined partial-denitrification and anammox process has received growing interest for treating wastewaters with high nitrate concentrations, but the underlying molecular mechanisms are currently poorly understood due to the complexity in bacterial community composition and functionality of the coupled system. This study applied metagenomic and metatranscriptomic approaches to gain comprehensive and deep insights into the nitrogen metabolic pathways in a lab-scale partial-denitrification/anammox bioreactor showing stable nitrogen removal. Stoichiometric analysis showed that most of nitrogen was removed by the coupled system (similar to 92.1%), but dissimilatory denitrification also contributed to nitrogen gas production. The latter was further evidenced by nosZ gene transcripts in the metatranscriptome. Metagenomic and metatranscriptomic analyses identified Candidatus Jettenia caeni as the most abundant and active anammox bacteria with relative abundance and mRNA expression of over 5% and 27%, respectively, while Thauera appeared as the major denitrifying bacteria in the system (relative abundance and mRNA expression exceeded 8% and 10%, respectively). mRNA levels of hdh/hzo and hzsA/B/C genes in Candidatus Jettenia caeni and Candidatus Brocadia sinica were much higher than those of other functional genes, confirming the major roles of the anammox bacteria in nitrogen metabolism. In addition, nrfA/B/C/D genes were mainly expressed in anammox bacteria, implying their involvement in that dissimilatory nitrite reduction to ammonia (especially for species Candidatus Brocadia sinica). Nitrate reduction process in Thauera was mainly achieved by the activation of the napA/B/C genes instead of narG/H genes. This study extablished the nitrogen metabolic network based on the comprehensive analyses of the abundance and mRNA expression of functional genes, which might improve our understanding of the nitrogen metabolic mechanisms of the combined partial-denitrification and anammox process.
Aerobic carbon metabolism modulates nitrite ammonifiers for inhibiting nitrogen loss as revealed by microcosm experiment of agricultural upland soil

Abstract:Denitrification and dissimilatory nitrate reduction to ammonium (DNRA), two divergent nitrogen metabolism pathways, share nitrite as a common intermediate during anaerobic reduction. However, these pathways influence the environment and soil nutrients differently. Unlike denitrification, which involves nitrogen loss through gaseous nitrogen emission and nitrate leaching, DNRA is beneficial for the conservation of nitrogen. Nevertheless, the key mechanism underlying the joint regulation of aerobic carbon metabolism to nitrite ammonifiers in soil is still unclear, although previous studies have reported factors such as carbon and oxygen can independently affect DNRA. Here, microcosm experiments with agricultural upland soil were conducted under different aeration conditions supplemented with labile carbon to analyze the process of anaerobic nitrogen metabolism. The results indicated that denitrification exclusively dominated nitrite reduction when the soil was directly placed in an anaerobic environment. Nonetheless, a significant increase in DNRA activity and the attenuation of denitrification were detected when the soil was incubated aerobically with the addition of glucose prior to anaerobic incubation. Specifically, up to 55.8% of nitrite reduction switched to nitrogen conservation mainly via DNRA under high-carbon conditions. Quantitative assays of the nrfA gene and metagenomic data revealed a significant increase in DNRA-related genes after aerobic carbon metabolism. Furthermore, nrfA gene sequence analysis revealed a significant shift in the composition of nitrite ammonifiers community. These results indicate that nitrate/nitrite metabolic flux in the soil could be regulated to enhance DNRA by stimulating facultative anaerobic nitrite ammonifiers such as Sedimentibacter under alternating aerobic and anaerobic environments with carbon metabolism.

Metagenomic insights into nitrite accumulation in sulfur-based denitrification systems utilizing different electron donors: Functional microbial communities and metabolic mechanisms

Abstract:Sulfur-based autotrophic denitrification (SADN) offers new pathway for nitrite supply. However, sequential transformation of nitrogen and sulfur forms, and the functional microorganisms driving nitrite accumulation in SADN with different reduced inorganic sulfur compounds (RISCs), remain unclear. Desirable nitrite accumulation was achieved using elemental sulfur (S0-group), sulfide (S2--group) and thiosulfate (S2O32--group) as electron donors. Under equivalent electron supply conditions, S2O32--group exhibited a superior nitrate conversion rate (NCR) of 0.285 kg N/(m³·d) compared to S2--group (0.103 kg N/(m³·d)). Lower NCR in S2--group was attributed to sulfide strongly inhibiting energy metabolism process of TCA cycle, resulting in reduced reaction rates. Moreover, the NCR of S0-group (0.035 kg N/(m³·d)) was poor due to the chemical inertness of S0. Specific microbial communities were selectively enriched in phylum level, with Proteobacteria increasing to an astonished 96.27-98.49 %. Comprehensive analyses of functional genus, genes, and metabolic pathways revealed significant variability in the active functional genus, with even the same genus showed significant metabolic differences in response to different RISCs. In S0-group, Thiomonas (10.0 %) and Acidithiobacillus (5.1 %) were the primary contributor to nitrite accumulation. Thiobacillus was the most abundant sulfur-oxidizing bacteria in S2--group (43.84 %) and S2O32--group (18.92 %). In S2--group, it contributed to nitrite accumulation, while in S2O32--group, it acted as a complete denitrifier (NO3--N→N2). Notably, heterotrophic denitrifying bacteria, Comamonas (12.52 %), were crucial for nitrite accumulation in S2O32--group, predominating NarG while lacking NirK/S. Overall, this work advances our understanding of SADN systems with different RISCs, offering insights for optimizing nitrogen and sulfur removal.
A fully-differential CMOS low-pass notch filter for biosignal measurement devices with high interference rejection

Abstract:A fifth-order fully-differential CMOS low-pass notch filter (LPNF) with high interference rejection is presented to provide an interface to the instrumentation amplifier (IA) in biopotential and bioimpedance measurement devices. The LPNF employs six operational transconductance amplifiers with differential difference input stage, which have low transconductances in nanosiemens range to enable the use of small capacitors that are feasible for on-chip integration. The filter design was simulated using IBM 0.13-μm CMOS technology. It has a bandwidth of 47.7 Hz, covering the electroencephalography (EEG) frequencies from 1 to 40 Hz with a notch of 73.2 dBc at the 60 Hz power line frequency. The fully-differential structure provides an inherent advantage of high common-mode rejection and high power supply rejection as reflected by system simulations (IA and LPNF) resulting in a common-mode rejection ratio (CMRR) of 101.3 dB and a power supply rejection ratio (PSRR) of 64.7 dB.
Fulvic acid mediated highly efficient heterotrophic nitrification-aerobic denitrification by Paracoccus denitrificans XW11 with reduced C/N ratio

Abstract:Reducing the C/N ratio requirements for heterotrophic nitrification-aerobic denitrification (HNAD) is crucial for its practical application; however, it remains underexplored. In this study, a highly efficient HNAD bacterium, Paracoccus denitrificans XW11, was isolated. The HNAD characteristics of XW11 were studied, and the redox mediator fulvic acid (FA) was used to reduce the C/N requirements. Whole-genome sequencing revealed multiple denitrification genes in XW11; however, nitrification genes were not identified, because heterotrophic nitrification-related gene sequences were not included in the database. However, the nitrogen removal related enzyme activity test revealed complete nitrification and denitrification pathways. Reverse transcription PCR showed that the membrane-bound nitrate reductase (NarG), rather than the periplasmic nitrate reductase, was responsible for aerobic denitrification. The conventional nitrite reductase (NirS) also does not mediate nitrite denitrification. When the C/N ratio was 10, the ammonia removal efficiency of the Control was 71.71 % and the addition of FA increased it to 86.12 %. Transcriptomic analysis indicated electron flow from the carbon source to FA without proton transmembrane transport, and the presence of FA constructs another electron transfer system. The redox potential of oxidized FA/reduced FA is 0.3679 V, avoiding competition for electrons from Complex III. Thus, ammonia monooxygenase obtains electrons more easily, thereby promoting nitrification. The enzyme activity test of the nitrification process confirmed this view. In addition, NarG expression increased, and the denitrification process was enhanced. Overall, FA improved HNAD efficiency by facilitating electron transfer to the nitrogen dissimilation process, offering a novel approach to reduce the C/N requirement of HNAD.
Nitrogen Removal and N2O Accumulation during Hydrogenotrophic Denitrification: Influence of Environmental Factors and Microbial Community Characteristics.

Abstract:Hydrogenotrophic denitrification is regarded as an efficient alternative technology of removing nitrogen from nitrate-polluted water that has insufficient organics material. However, the biochemical process underlying this method has not been completely characterized, particularly with regard to the generation and reduction of nitrous oxide (N2O). In this study, the effects of key environmental factors on hydrogenotrophic denitrification and N2O accumulation were investigated in a series of batch tests. The results show that nitrogen removal was efficient with a specific denitrification rate of 0.66 kg N/(kg MLSS·d), and almost no N2O accumulation was observed when the dissolved hydrogen (DH) concentration was approximately 0.40 mg/L, the temperature was 30 °C, and the pH was 7.0. The reduction of nitrate was significantly affected by the pH, temperature, inorganic carbon (IC) content, and DH concentration. A considerable accumulation of N2O was only observed when the pH decreased to 6.0 and the temperature decreased to 15 °C, where little N2O accumulated under various IC and DH concentrations. To determine the microbial community structure, the hydrogenotrophic denitrifying enrichment culture was analyzed by Illumina high-throughput sequencing, and the dominant species were found to belong to the genera Paracoccus (26.1%), Azoarcus (24.8%), Acetoanaerobium (11.4%), Labrenzia (7.4%), and Dysgonomonas (6.0%).

Integrative chemical and omics analysis of the ammonia nitrogen removal characteristics and mechanism of a novel oligotrophic heterotrophic nitrification-aerobic denitrification bacterium

Abstract:A novel oligotrophic heterotrophic nitrification-aerobic denitrification bacterium designated as Pseudomonas sp. N31942, was isolated from a eutrophic lake. Strain N31942 exhibits high ammonia nitrogen removal ability in oligotrophic environment as ammonia nitrogen can be efficiently (86.97 %) removed within 10 h with no accumulation of nitrite. In the nitrification process, strain N31942 can convert ammonia into nitrate in the absence of hydroxylamine oxidase and nitrite oxidoreductase. As for the denitrification process, nitrate or nitrite were reduced to ammonia and further converted into glutamate by dissimilatory nitrate reduction pathway. Transcriptomic analysis detected 2080 differentially expressed genes. Among them, the expression of the related genes in dissimilatory nitrate reduction process was all up-regulated at low ammonia concentrations, which indicates that the strain has excellent nitrogen removal efficiency for further nitrogen removal. Integrative omics analyses revealed that strain N31942 may have two possible pathways for the NH4+-N removal as direct GDH/GS-GOGAT pathway (NH4+-N → Glutamate) and indirect GDH/GS-GOGAT pathway (NH4+-N → NH2OH → NO2−-N → NO3−-N → NO2−-N → NH4+-N → Glutamate). Moreover, strain N31942 also has excellent nitrogen removal ability for real sewage and 77.21 % total nitrogen could be removed within 48 h. The results presented here provide new insights into ammonia nitrogen removal characteristics and mechanism of heterotrophic nitrification-aerobic denitrification bacterium under oligotrophic conditions.

Abstract:The combined effects of carbon source (HAc, HPr, Glu, Glu + HAc) and nitrate concentration (40, 80 mg/L labeling as R 40 , R 80 ) on partial denitrification (PD) were discussed at C/N ratio of 2.5 (COD = 100, 200 mg/L). The optimal NO 2 − -N and NTR reached to 67.03 mg/L, 99.14% in HAc-R 80 system, and denitrification kinetics revealed the same conclusion, corresponding to higher COD utilization rate (CUR: 58.46 mgCOD/(gVSS·h)), nitrate reduction rate (N a RR: 29.94 mgN/(gVSS·h)) and nitrite accumulation rate (N i AR: 29.68 mgN/(gVSS·h)). The preference order was HAc > HPr > Glu + HAc > Glu in both R 40 and R 80 systems due to different metabolic pathways, however, the NO 2 − -N accumulation and kinetic parameters of R 80 group were dramatically higher than those in R 40 for the same carbon source. The R 80 group facilitated more concentrated biodiversity (607–808 OTUs) with Terrimonas and norank_f_Saprospiraceae responsible for high NO 2 − -N accumulation in HAc and HPr served systems, while norank_f_norank_o_Saccharimonadales and OLB13 dominated the Glu containing systems.

Abstract:The anaerobic/aerobic/anoxic (A/O/A) process is a promising biotechnology to intensify denitrification in low carbon/nitrogen (C/N) wastewater treatment, but the neglected typical rate-limiting step—nitrification—would hinder its wider application. Heterotrophic nitrification driven by intracellular carbon (PHAs) could enhance nitrification and achieve endogenous simultaneous nitrification and denitrification (ESND) in the A/O/A process, but its feasibility remains unexamined. Here we established four A/O/A-SBRs at different C/N ratios (3, 7.5, 12, and 16.5) to address the above-mentioned knowledge gaps. The results showed that organic supplementation promoted both nitrification and denitrification (performance and relevant enzymatic activities) until organic overdose (C/N = 16.5) exacerbated niche competitions from other non-functional heterotrophs. qPCR and batch tests indicated that high C/N ratios inhibited autotrophic nitrifiers, and heterotrophic nitrifiers (HNB) dominated in the enhanced nitrification. Given the high HNB contribution (43.7%) and low COD variation (< 10 mg L −1 ) in the SND (76.4%) of CN12, we proposed a potential SND pathway based on heterotrophic nitrification and denitrification driven by PHAs and verified it with batch tests. Microbial and functional analyses suggested that CN12 favored the intracellular carbon transformation and harbored the minimum autotrophic nitrifiers, supporting the dominance of ESND in the enhanced SND. Our findings expand the understanding of the relationships between intracellular carbon transformation and SND and provide a novel nitrogen removal pathway for the practical application of the A/O/A process.

Abstract:To address the limitations inherent in both sulfur autotrophic denitrification (SAD) and heterotrophic denitrification (HD) processes, this study introduces a novel approach. Three carbon sources (glucose, methanol, and sodium acetate) were fed into the SAD system to facilitate the transition towards mixotrophic denitrification. Batch experiments were conducted to explore the effects of influencing factors (pH, HRT) on the denitrification performance of the mixotrophic system. Carbon source dosages were varied at 12.5%, 25%, and 50% of the theoretical amounts required for HD (18, 36, and 72 mg/L, respectively). The results showed distinct optimal dosages for each of the three organic carbon sources. The mixotrophic system, initiated with sodium acetate at 25% of the theoretical value, demonstrated the highest denitrification performance, achieving NO3--N removal efficiency of 99.8% and the NRR of 6.25 mg/(L·h). In contrast, the corresponding systems utilizing glucose (at 25% of the theoretical value) and methanol (at 50% of the theoretical value) achieved lower removal efficiency of 77.0% and 88.4%, respectively. The corresponding NRRs were 4.85 mg/(L·h) and 5.65 mg/(L·h). Following the transition from SAD to a mixotrophic system, the abundance of Thiobacillus decreased from 78.5% to 34.4% at the genus level, and the mixotrophic system cultivated a variety of other denitrifying bacteria (Thauera, Aquimonas, Azoarcus, and Pseudomonas), indicating an enhanced microbial community structure diversity. The established artificial neural network (ANN) model accurately predicted the effluent quality of mixotrophic denitrification, which predicted values closely aligning with experimental results (R2 > 0.9). Furthermore, initial pH exerted greater relative importance for COD removal and sulfur conversion, while the relative importance of HRT was more pronounced for NO3--N removal.

Abstract:The heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria harbored remarkable removal capacity in eliminating single nitrogen pollutant, but their comprehensive performance in dealing with mixed nitrogen sources remained unknown. In this study, the denitrification capacity of HN-AD bacteria Paracoccus denitrificans HY-1 was explored through whole-genome sequencing and nitrogen metabolic pathway analysis, and the complex nitrogen sources with gradient concentration were used to validate its nitrogen removal efficiency under both aerobic and anaerobic conditions. The results demonstrated that strain HY-1 harbored an efficient degradation capacity on three nitrogen sources, and the removal rates of NH 4 + -N, NO 2 − -N and NO 3 − -N were 10.67 mg·L −1 ·h −1 , 27.81 mg·L −1 ·h −1 , and 22.05 mg·L −1 ·h −1 , respectively. The denitrification rate of 15.33 mg·L −1 ·h −1 was obtained under anaerobic condition with mixed nitrogen sources. Also, the simultaneous nitrification denitrification phosphorus removal (SNDPR) trial revealed that strain HY-1 could eliminate PO 4 3− -P with a rate of 0.38 mg·L −1 ·h −1 under coexistence condition of the three nitrogen sources. Combined with the nitrogen balance analysis, the results indicated that strain HY-1 possesses a great potential for application in complex nitrogen and phosphorus contaminated environments.

Abstract:The effect of different carbon sources was studied relative to the treatment effect of aerobic denitrifying bacteria during the treatment of nitrate-containing wastewater as well as the influence of co-metabolism flora. Three carbon sources, i.e., glucose, citric acid, and sodium acetate, were selected to study the changes in the pH, nitrate nitrogen, chemical oxygen demand removal, nitrite nitrogen concentration, physical and chemical properties of sludge and volatile fatty acids, and high-throughput sequencing to study any changes in flora. The results showed that the denitrification ability of the system using citric acid as the carbon source was stronger than the denitrification ability of the system using glucose or sodium acetate as the carbon source. The removal of nitrate nitrogen in the system was the result of the co-metabolism of acid producing bacteria and aerobic denitrifying bacteria. Due to the greater number of types and greater amount of dominant bacteria in the sludge domesticated and cultured with citric acid as a carbon source, the co-metabolism flora formed was more stable, so it could better remove nitrate nitrogen.

Abstract:Harnessing nitrous oxide (N2O)-reducing bacteria is a promising strategy to reduce the N2O footprint of engineered systems. Applying a preferred organic carbon source as an electron donor accelerates N2O consumption by these bacteria. However, their N2O consumption potential and activity when fed different organic carbon species remain unclear. Here, we systematically compared the effects of various organic carbon sources on the activity of N2O-reducing bacteria via investigation of their biokinetic properties and genomic potentials. Five organic carbon sources-acetate, succinate, glycerol, ethanol, and methanol-were fed to four N2O-reducing bacteria harboring either clade I or clade II nosZ gene. Respirometric analyses were performed with four N2O-reducing bacterial strains, identifying distinct shifts in DO- and N2O-consumption biokinetics in response to the different feeding schemes. Regardless of the N2O-reducing bacteria, higher N2O consumption rates, accompanied by higher biomass yields, were obtained with acetate and succinate. The biomass yield (15.45 ± 1.07 mg-biomass mmol-N2O-1) of Azospira sp. strain I13 (clade II nosZ) observed under acetate-fed condition was significantly higher than those of Paracoccus denitrificans and Pseudomonas stutzeri, exhibiting greater metabolic efficiency. However, the spectrum of the organic carbon species utilizable to Azospira sp. strain I13 was limited, as demonstrated by the highly variable N2O consumption rates observed with different substrates. The potential to metabolize the supplemented carbon sources was investigated by genomic analysis, the results of which corroborated the N2O consumption biokinetics results. Moreover, electron donor selection had a substantial impact on how N2O consumption activities were recovered after oxygen exposure. Collectively, our findings highlight the importance of choosing appropriate electron donor additives for increasing the N2O sink capability of biological nitrogen removal systems.
