Abstract:Chlorpyrifos is a widely used organophosphate insecticide and ubiquitously detected in the environment. However, little attention has been paid to its endocrine disrupting effect to non-target organisms. In the present study, zebrafish was exposed to 13 and 65 μg/L of chlorpyrifos for 7 and 10 days to determine the induced neurotoxicity and the alteration of thyroid metabolism. The 120 h LC50 and LC10 of chlorpyrifos was estimated as 1.35 mg/L and 0.62 mg/L based on the acute embryo toxicity assay, respectively. The acetylcholinesterase (AChE) inhibitory was detected by 13 μg/L chlorpyrifos and could be reversed by the co-exposure of 100 and 1000 μg/L anticholinergic agent atropine. For thyroid hormone level, 13 and 65 μg/L of chlorpyrifos induced increased free T3 levels in 10 dpf (days post-fertilization). The expression of thyroid related genes in 7 and 10 dpf exposed zebrafish were measured by the quantitative Real-Time PCR (qRT-PCR) assay. The mRNA expression of tshba, thrb, crhb, ttr, tpo, ugt1ab and slc5a5 had significant change. However, the alterations of thyroid hormone and mRNA expression could be partly rescued by the addition of atropine. The molecular docking of chlorpyrifos and T3 to the thyroid receptor β in zebrafish using homology modelling and CDOCKER procedures shown weaker binding ability of chlorpyrifos compared to T3. Therefore, we concluded that the disturbance of thyroid signaling in zebrafish might arise from the developmental neurotoxicity induced by chlorpyrifos.
Changes of Embryonic Development, Locomotor Activity, and Metabolomics in Zebrafish Co‐exposed to Chlorpyrifos and Deltamethrin

Abstract:Organophosphates (OPs) and pyrethroids (PYRs) are extensively used pesticides and often occur in the form of mixture, whereas little was known about their joint toxicities. We aim to investigate the individual and joint effects of OPs and PYRs exposure on zebrafish embryo by employing chlorpyrifos (CPF) and deltamethrin (DM) as representatives. Zebrafish embryos at 2 hours post fertilization (hpf) were exposed to CPF (4.80, 39.06, and 78.13 μg/L), DM exposure (0.06, 1.60, and 3.19 μg/L), and CPF + DM (4.80 + 0.06, 39.06 + 1.60, and 78.13 + 3.19 μg/L) until 144 hpf. Embryonic development, locomotor activity, and metabolomic changes were recorded and examined. Results displayed that individual exposure to CPF and DM significantly increased the mortality and malformation rate of zebrafish embryos, but decreased hatching rate was only found in CPF + DM co‐exposure groups (p < .05). Meanwhile, individual CPF exposure had no detrimental effect on locomotor activity, high dose of individual CPF exposure decreased the swimming speed but had adaptability to the conversion from dark to light, whereas high dose of CPF + DM co‐exposure exhibited not only significant decline in swimming speed but also no adaptability to the repeated stimulations, suggesting deficit in learning and memory function. In metabolomic analysis, individual CPF exposure mainly influenced the metabolism of glycerophospholipids and amino acids, individual DM exposure mainly influenced glycerophospholipids, and CPF + DM co‐exposure mainly influenced glycerophospholipids and amino acids. Taken together, our findings suggested the embryonic toxicities and neurobehavioral changes caused by CPF and/or DM exposure. The disorder metabolomics of glycerophospholipids and amino acids might be involved in the underlying mechanism of those toxicities.

Abstract:Background: Organophosphate insecticides (OPs) are used worldwide, yet despite nearly ubiquitous exposure in the general population, few have been studied outside the laboratory. Fetal brains undergo rapid growth and development, leaving them susceptible to long-term effects of neurotoxic OPs. The objective here was to investigate the extent to which prenatal exposure to OPs affects infant motor development.Methods: 30 OPs were measured in umbilical cord blood using gas chromatography tandem mass spectrometry in a cohort of Chinese infants. Motor function was assessed at 6-weeks and 9-months using Peabody Developmental Motor Scales 2nd edition (PDMS-2) (n = 199). Outcomes included subtest scores: reflexes, stationary, locomotion, grasping, visual-motor integration (V-M), composite scores: gross (GM), fine (FM), total motor (TM), and standardized motor quotients: gross (GMQ), fine (FMQ), total motor (TMQ).Results: Naled, methamidophos, trichlorfon, chlorpyrifos, and phorate were detected in = 10% of samples. Prenatal naled and chlorpyrifos were associated with decreased 9-month motor function. Scores were 0.55, 0.85, and 0.90 points lower per 1 ng/mL increase in log-naled, for V-M (p = 0.04), FM (p = 0.04), and FMQ (p = 0.08), respectively. For chlorpyrifos, scores were 0.50, 1.98, 0.80, 1.91, 3.49, 2.71, 6.29, 2.56, 2.04, and 2.59 points lower for exposed versus unexposed infants, for reflexes (p = 0.04), locomotion (p = 0.02), grasping (p = 0.05), V-M (p < 0.001), GM (p = 0.007), FM (p = 0.002), TM (p < 0.001), GMQ (p = 0.01), FMQ (p = 0.07), and TMQ (p = 0.008), respectively. Girls appeared to be more sensitive to the negative effects of OPs on 9-month motor function than boys.Conclusions: We found deficits in 9-month motor function in infants with prenatal exposure to naled and chlorpyrifos. Naled is being aerially sprayed to combat mosquitoes carrying Zika virus, yet this is the first nonoccupational human study of its health effects. Delays in early-motor skill acquisition may be detrimental for downstream development and cognition.

Abstract:Pyrethroid and organophosphate pesticides are two of the most commonly used classes of insecticide worldwide. At sublethal concentrations, permethrin (a pyrethroid) and chlorpyrifos (an organophosphate) impact behavior in model fish species. We investigated behavioral effects of environmentally relevant concentrations of permethrin or chlorpyrifos on early larval delta smelt Hypomesus transpacificus , a Critically Endangered teleost species endemic to the San Francisco Bay Delta, California, USA. Using a photomotor behavioral assay of oscillating light and dark periods, we measured distance moved, turn angle, meander, angular velocity, rotations, thigmotaxis (time spent in the border versus center), and swim speed duration and frequency. The lowest concentrations of permethrin used in the tests (0.05 and 0.5 μg l -1 ) caused significant increases in distance moved at 72 and 96 h, respectively. At 48, 72, and 96 h of exposure, 5 μg l -1 of permethrin caused a hyperactive state in which the larvae significantly decreased thigmotaxis, quickly turning in short bouts of activity, characterized by significant increases in rotations and freezing events. Larvae exposed to 0.05 μg l -1 chlorpyrifos significantly increased thigmotaxis at 72 and 96 h. In response to 5 μg l -1 chlorpyrifos, larvae significantly increased velocity at 72 h exposure, and significantly increased freezing events at 96 h. Behavioral data on larval delta smelt exposed to contaminants present in their limited habitat have the potential to aid evaluations of the suitability of spawning and rearing habitats for this endangered species, thus improving conservation management strategies focused on this sensitive life stage.

Early Environmental Exposure to Oxytetracycline in Danio Rerio May Contribute to Neurobehavioral Abnormalities in Adult Zebrafish.

Abstract:The common antibiotic oxytetracycline (OTC) is nowadays commonly found in natural aquatic environments. However, the underlying mechanisms of low-dose OTC exposure and its neurotoxic effects on aquatic animals remain unknown. In this study, we exposed zebrafish larvae to environmental concentrations of OTC in early life and performed neurobehavioral, 16S rRNA gene sequencing, and transcriptomic analyses. OTC exposure resulted in hyperactivity of larvae and a significant reduction in the number of neurons in the midbrain. The expression levels of 15 genes related to neural function changed. Additionally, the composition of 65 genera of the gut microbiota of larvae was altered, which may be one of the reasons for the abnormal neural development. We further studied the long-term outcomes among adult fish long after cessation of OTC exposure. OTC treatment caused adult fish to be depressive and impulsive, symbolizing bipolar disorder. Adult fish exposed to OTC had significantly fewer neurons and their gut bacteria composition did not recover 104 days after terminating OTC exposure. Finally, we analyzed the correlation between the gut microbiota of larvae, genes related to neural function, and metabolites of adult fish brain tissue. The results showed that the abundance of several members of the biome in larvae was related to the transcription levels of genes related to neural function, which were related to the metabolic levels in the adult brain. In conclusion, our study showed that early-life exposure to environmental concentrations of OTC can lead to persistent neurobehavioral abnormalities until adulthood through dysbiosis in the gut microbiota.

Abstract:Chlorpyrifos is a widely used organophosphorus insecticide because of its high efficiency and overall effectiveness, and it is commonly detected in aquatic ecosystems. However, at present, the impact of chlorpyrifos on the aquatic micro-ecological environment is still poorly understood. Here, we established aquatic microcosm systems treated with 0.2 and 2.0 µg/L chlorpyrifos, and employed omics biotechnology, including metagenomics and 16S rRNA gene sequencing, to investigate the effect of chlorpyrifos on the composition and functional potential of the aquatic and zebrafish intestinal microbiomes after 7 d and 14 d chlorpyrifos treatment. After 14 d chlorpyrifos treatment, the aquatic microbial community was adversely affected in terms of its composition, structure, and stability, while its diversity showed only a slight impact. Most functions, especially capacities for environmental information processing and metabolism, were destroyed by chlorpyrifos treatment for 14 d. We observed that chlorpyrifos increased the number of risky antibiotic resistance genes and aggravated the growth of human pathogens. Although no clear effects on the structure of the zebrafish intestinal microbial community were observed, chlorpyrifos treatment did alter the metabolic capacity of the zebrafish. Our study highlights the ecological risk of chlorpyrifos to the aquatic environment and provides a theoretical basis for the rational use of pesticides in agricultural production.

Abstract:While triphenyl phosphate (TPhP) has been frequently detected in surface water and wildlife, its adverse effects on the gonadal development and reproductive behaviors of fish remain unclear. In this study, Japanese medaka (Oryzias latipes) were exposed to TPhP at concentrations of 134.1, 299.1, and 1429.5 ng/L from hatching [0 days posthatching (dph)] to sexual maturity (100 dph). TPhP induced gonadal intersex in male medaka in all exposure groups, and a significant increase was observed in the 1429.5 ng/L exposure group, with an incidence of 26.2% (11 of 42; p < 0.01). TPhP exposure also caused abnormal chasing behavior, with a lowest observable effective concentration (LOEC) of 299.1 ng/L, and reduced the desire of males for females in the 1429.5 ng/L group, demonstrating toxicity for fish reproductive behaviors. The anti-androgenic activity of TPhP via both androgen suppression and AR blocking was proposed to be the major mechanism of the observed effects. On the basis of dose-dependent inhibition of successful mating, fertilization, and hatching of eggs, environmentally relevant concentrations of Tphp pose a risk to fish reproduction.

Abstract:Daphnia magna is a test organism used for ecological risk assessments of pesticides, but little is known about the expression levels of cytochrome P450s (CYP)s and their changes after pesticide exposure in the less than 24‐h‐olds used for ecotoxicity tests. In this study, D. magna juveniles were exposed to 0.2 μg/L of chlorpyrifos under the conditions for acute immobilization test as specified by the OECD test guideline for 24 h, and then the gene expression was compared between the control and chlorpyrifos‐exposure groups by RNA‐sequencing analysis, with a focus on CYP genes. Among 38 CYP genes expressed in the control group, seven were significantly up‐regulated while two were significantly down‐regulated in the chlorpyrifos‐exposure group. Abstract:Chlorpyrifos (CPF) has been extensively used in the world and frequently found in natural environments, might cause a range of environmental issues and pose a health risk to aquatic species. However, investigation of its toxic effects on offspring after parental exposure has been neglected, especially for aquatic organisms such as fish. In the current study, the effects of chronic CPF exposure (3 and 60 μg/L) on adult zebrafish (F0) was investigated to determine its influence on adult reproductive capacity and offspring (F1 and F2). The results showed the existence of CPF both in F0 ovaries and F1 embryos and larvae, indicating that CPF could be transferred directly from the F0 adult fish to F1 offspring. After 90 d exposure, we observed that F0 female fish showed increased proportion of perinucleolar oocyte in the ovaries, decreased proportion of mature oocyte, and decreased egg production, but not in F1 adult. The transcriptomic analysis revealed that the disruption of metabolism during oocyte maturation in the CPF treatment zebrafish might interfere with F0 oocytes development and quality and ultimately influence offspring survival. For the larvae, the parental CPF exposure distinctly inhibited heart rate at 72 and 120 hpf and increased the mortality of F1 but not F2 larvae. The changes of biochemical indicators confirmed a disturbance in the oxidative balance, induced inflammatory reaction and apoptosis in F1 larvae. Furthermore, the changing profiles of mRNA revealed by RNA-seq confirmed an increased susceptibility in F1 larvae and figured out potential disruptions of ROS metabolism, immune system, apoptosis, and metabolism pathways. Taken together, these results show that chronic CPF treatment can induce reproductive toxicity, and parental transfer of CPF occurs in fish, resulting in transgenerational alters in F1 generation survival and transcription that raising concerns on the ecological risk of CPF in the natural environment.

Abstract:Perfluorooctane sulfonic acid (PFOS) has been widely detected in environment and organisms. PFOS has been identified as the driving agent for the behavioral changes of zebrafish larvae, while the underlying molecular mechanism remains unclear. In this study, zebrafish embryos/larvae were exposed to 0, 0.04, 0.1, 0.4 and 1 μM PFOS for 166 h. The locomotor behaviors and the mRNA transcription of genes in neuromuscular system were detected. Exposure to PFOS did not affect the hatching/death rates and body length, but increased the heart beat rates and frequency of spontaneous tail coiling. Locomotor behavior in zebrafish larvae of 0.4 and 1 μM PFOS groups were increased in the light condition. Additionally, the levels of acetylcholine (Ach) in 0.4 μM PFOS group and dopamine (DA) in 0.1, 0.4 and 1 μM PFOS groups were found to be significantly increased. The expression of genes related to the synthesis and decomposition of ACh,the synthesis and receptor of DA, and fosab was increased in the different PFOS treatment groups, while the expression of all the other genes of the neuromuscular system were significantly reduced. The findings of this investigation demonstrated that PFOS exposure may alter the locomotor behavior of zebrafish through disrupting the expressions of genes in neuromuscular system. The disturbed process of neurotransmitter transmission and muscle contraction caused by PFOS may be the dominant mechanism of hyperactivity in zebrafish.

Interaction between chronically HIV‐infected promonocytic cells and human umbilical vein endothelial cells: role of proinflammatory cytokines and chemokines in viral expression modulation

Abstract:HIV type 1 expression was significantly up‐regulated in chronically infected promonocytic cell line (U1) co‐cultured with human umbilical vein endothelial cells (HUVEC). Virus replication, evaluated as supernatant p24 release, was higher when U1 were co‐cultured with IL‐1β‐activated HUVEC than with unstimulated HUVEC. When non‐adherent U1 were removed from co‐cultures, the remaining U1 cells adherent to the endothelial monolayer still showed enhanced HIV replication in comparison with an equal number of U1 cultured alone. While addition of adhesion molecule blocking antibodies (anti‐intercellular adhesion molecule‐1 (ICAM‐1), ‐vascular cell adhesion molecule‐1 (VCAM‐1), ‐CD18 and ‐very late antigen‐4 (VLA‐4)) strongly inhibited adherence of U1 cells to endothelial monolayers, such treatment resulted in only a partial reduction in p24 release. Furthermore, HIV replication in U1 cells was enhanced on culture in HUVEC‐conditioned media. Such data suggest that soluble mediators secreted by endothelial monolayers may modulate HIV‐1 expression. Indeed, addition of cytokine and chemokine antagonists to both U1/HUVEC co‐cultures and to U1 cultured in HUVEC‐conditioned media clearly down‐regulated p24 release. Anti‐IL‐6, anti‐tumour necrosis factor‐alpha (TNF‐α) and, particularly, anti‐MCP‐1 MoAbs reduced p24 release, while anti‐IL‐8 polyclonal antiserum and IL‐1 receptor antagonist (IL‐1Ra) had no significant effect. Thus, the interaction between HUVEC and infected monocytic cells up‐regulates HIV‐1 replication predominantly through production of endothelium‐derived soluble factors including MCP‐1, TNF‐α and IL‐6. This phenomenon may influence the passage of HIV‐1 from latency to productive replication and enhance virus spreading during physiological and/or pathological contact of monocytes with endothelium.
Oral Exposure to Tributyltin Induced Behavioral Abnormality and Oxidative Stress in the Eyes and Brains of Juvenile Japanese Medaka (Oryzias latipes)

Abstract:The widely used compound tributyltin (TBT), which can be continuously detected in aquatic species and seafood, may induce diverse adverse effects on aquatic organisms. However, little is known regarding the mechanistic links between behavioral abnormality and oxidative stress in different fish tissues in response to oral TBT exposure. Herein, juvenile Japanese medaka (Oryzias latipes) were orally exposed to TBT at 1 and 10 ng/g-bw/d for four weeks. After exposure, the locomotor activity and social interaction of juvenile medaka were found to be significantly reduced in the 10 ng/g-bw/d TBT-exposed group. Furthermore, the antioxidant biomarkers in different tissues of juvenile medaka showed different levels of sensitivity to TBT exposure. The eye superoxide dismutase (SOD) activities markedly increased in both groups exposed to 1 and 10 ng/g-bw/d TBT, while the eye and brain malondialdehyde (MDA) levels increased in the higher dose group. Furthermore, the eye and brain ATPase activities markedly declined in the 1 ng/g-bw/d TBT-exposed group. A correlation analysis revealed that the decreased locomotor activity and social interaction in medaka were associated with the eye antioxidant enzyme (i.e., SOD and catalase (CAT)) activity and brain oxidative damage level. Thus, our findings suggested that there might be some mechanistic links between the behavioral abnormality induced by TBT exposure and oxidative stress in the eyes and brains of medaka. Thus, our findings indicate that the impacts of oral exposure to TBT should be considered to better assess its risk to the aquatic ecosystem and human health.

Abstract:Metabolic signals such as insulin, leptin and glucose are known to alter hypothalamic function. Although insulin and leptin are known to directly alter hypothalamic areas that regulate reproduction, the mechanisms by which glucose alters reproductive function are not as clear. Catecholaminergic neurons in the A1/C1 region in the hindbrain are glucose-responsive and project to the arcuate nucleus. To determine if this pathway is involved in the regulation of sex behavior and luteinizing hormone (LH) secretion, this catecholaminergic pathway was lesioned with injections of saporin conjugated with anti-dopamine-β-hydroxylase (DSAP) or unconjugated saporin (SAP) in adult male rats. Rats were given glucoprivic challenges and feeding and sex behavior was observed. As was expected, the DSAP-treated rats showed decreased feeding during glucoprivation (250 mg/kg 2-deoxy-D-glucose, 2DG) compared to SAP controls. Glucoprivation caused a significant reduction in sex behavior in both SAP and DSAP animals equally, compared to saline treatments (p < 0.05). At the end of the experiment, animals were given a final challenge with 2DG or saline, euthanized by decapitation and trunk blood was assayed for plasma LH levels. In situ hybridization analysis revealed that 2DG treatment caused a significant reduction in GALP mRNA in SAP controls compared to saline treatment. This reduction in GALP mRNA was prevented with DSAP treatment. In SAP animals, 2DG elicited a significant decrease in plasma LH levels (p < 0.05); this reduction in plasma LH was absent in the DSAP-treated male rats. These data indicate that the A1/C1 efferents to the ventromedial hypothalamus are involved in the glucostatic regulation of GALP mRNA, feeding behavior and LH secretion, but not sex behavior in the adult male rat.
Ecological risk assessment for typical organophosphorus pesticides in surface water of China based on a species sensitivity distribution model

Abstract:The ecological risks posed by widespread organophosphorus pesticide (OPs) pollution in the surface waters of China remain unclear. In this study, species sensitivity distribution (SSD) parametric statistical approaches were coupled with fully acute and chronic toxicity data to fit the sensitivity distributions of different aquatic species to five typical OPs: dimethoate, malathion, parathion-methyl, trichlorfon, and dichlorvos. Crustaceans exhibit the highest sensitivity to OPs, whereas algae are the least sensitive. The acute hazardous concentrations that affected 5 % of the species (HC 5 ) were 0.112, 0.001, 0.001, 0.001, and 0.001 mg/L for dimethoate, malathion, parathion-methyl, trichlorfon, and dichlorvos, respectively, whereas their chronic HC 5 values were 0.004, 0.004, 0.053, 0.001, and 0.0005 mg/L, respectively. Hence, dichlorvos is highly toxic and poses greater risk to non-target aquatic species. The evaluation data revealed varying geographical distribution characteristics of the ecological risks from OPs in 15 freshwater aquatic systems across different regions of China. Dichlorvos posed the highest risk in the basins of Zhejiang and Guangdong Provinces, with the highest chronic Risk Quotient (RQ) and Hazard Index (HI) at 9.34 and 9.92, respectively. This is much higher than what was collected and evaluated for foreign rivers (the highest chronic RQ and HI in foreign rivers were 1.65 and 2.24, respectively). Thus, dichlorvos in the surface waters of China poses a substantial ecological risk to aquatic organisms, and may endanger human health.

Abstract:Metolachlor (MT) is one of the most important pesticides applied to corn and other crops for controlling broadleaf and grass weeds. However, the effects of MT on the thyroid system in fish remain to be elucidated. In the present experiment, transcription of genes related to the thyroid system, including thyrotropin releasing hormone (Trh), deiodinase 2 (Dio2), thyroid hormone receptor α (Thrα), and thyroid hormone receptor β (Thrβ), were induced by MT in a sex-, developmental stage-, and tissue- specific manner when medaka were exposed to various concentrations of MT for 14days. The transcriptional levels of the genes were only significantly altered in both juvenile and adult female medaka in response to MT exposure. And the lowest concentrations able to significantly induce transcription of the selected genes were 10 and 100μg/L in juvenile and adult female medaka, respectively. In adult female medaka, a significant up-regulation of these genes was detected only in the brain, with little or no effect in the liver. Furthermore, MT-induced (100μg/L) transcription of thyroid system-related genes was enhanced significantly in male juvenile medaka in the presence of estrogen (E2) (50 and 100ng/L). Moreover, the mRNA levels of Thrα and Thrβ in males increase with the combined treatments of 100μg/L MT and 100ng/L E2. Dio2 increased when exposed to 100μg/L MT and 50 or 100ng/L E2. The information obtained in the present study suggests that MT has the potential to influence several steps of the hypothalamus–pituitary–thyroid (HPT) axis homeostasis and to disrupt the thyroid system in medaka.
Effects of Chlorpyrifos on Cholinesterase and Serine Lipase Activities and Lipid Metabolism in Brains of Rainbow Trout (Oncorhynchus mykiss)

Abstract:Chlorpyrifos is an organophosphorus insecticide that elicits acute toxicity through inhibition of acetylcholinesterase (AChE), leading to acetylcholine accumulation and prolonged stimulation of cholinergic receptors throughout the central and peripheral nervous systems. Previous studies have indicated that neurodevelopment may also be impaired through alternative pathways, including reduction of cyclic adenosine monophosphate (cAMP)-catalyzed downstream events. The upstream initiating events that underlie noncholinergic neurological actions of chlorpyrifos and other organophosphorus compounds remain unclear. To investigate the potential role of fatty acid signaling disruption as a mechanism of toxicity, lipid metabolism and fatty acid profiles were examined to identify alterations that may play a critical role in upstream signaling in the central nervous system (CNS). Juvenile rainbow trout were treated for 7 days with nominal chlorpyrifos concentrations previously reported to diminish olfactory responses (10, 20, and 40 μg/l). Although lethality was noted higher in doses, measured chlorpyrifos concentrations of 1.38 μg/l (nominal concentration 10 μg/l) significantly reduced the activity of AChE and two serine lipases, monoacylglycerol lipase, and fatty acid amide hydrolase in the brain. Reductions in lysophosphatidylethanolamines (16:0, 18:0, 18:1, and 22:6) derived from the phosphatidylethanolamines and free fatty acids (palmitic acid 16:0, linolenic acid 18:3, eicosadienoic acid 20:2, arachidonic acid 20:4, and docosahexaenoic acid 22:6) were also noted, suggesting that chlorpyrifos inhibited the metabolism of select phospholipid signaling precursors at sublethal concentrations. These results indicate that in addition to AChE inhibition, environmentally relevant chlorpyrifos exposure alters serine lipase activity and lipid metabolites in the trout brain, which may compromise neuronal signaling and impact neurobehavioral responses in aquatic animals.
Sublethal neurotoxicity of organophosphate insecticides to juvenile coho salmon

Abstract:For decades, organophosphate (OP) insecticides have been used as chemical control agents in watersheds that support at-risk populations of Pacific salmon throughout western North America. Spray drift, runoff, and other processes transport OPs to critical surface water habitats for migratory salmonids. While most OPs share a common mechanism of action (i.e., inhibition of neuronal acetylcholinesterase, or AChE), they typically vary in toxic potency. Moreover, dose-response relationships for exposure and sublethal neurotoxicity (e.g., brain AChE inhibition) in salmonids have not been defined for many OPs. Here we exposed juvenile coho salmon (Oncorhynchus kisutch) to five common anticholinesterase insecticides (dimethoate, ethoprop, naled, phorate and phosmet) that are widely used on agricultural, commercial, residential, and public lands. Each of the five pesticides produced a concentration-dependent inhibition of AChE enzyme activity. The effective concentration for 50 % AChE inhibition (96-hr EC50) indicated the highest toxicity for phorate (EC50 = 0.57 μg/L) followed by phosmet (3.3 μg/L), naled (7.8 μg/L), ethoprop (90.6 μg/L) and dimethoate (273 μg/L). These findings can inform 1) relative hazard analyses for OP use near sensitive aquatic habitats, 2) predictions of sublethal OP mixture toxicity, and 3) ecological risk assessments for threatened or endangered species of Pacific salmon.

Abstract:A simple and useful characterization of many predictive models is in terms of model structure and model parameters. Accordingly, uncertainties in model predictions arise from uncertainties in the values assumed by the model parameters (parameter uncertainty) and the uncertainties and errors associated with the structure of the model (model uncertainty). When assessing uncertainty one is interested in identifying, at some level of confidence, the range of possible and then probable values of the unknown of interest. All sources of uncertainty and variability need to be considered. Although parameter uncertainty assessment has been extensively discussed in the literature, model uncertainty is a relatively new topic of discussion by the scientific community, despite being often the major contributor to the overall uncertainty. This article describes a Bayesian methodology for the assessment of model uncertainties, where models are treated as sources of information on the unknown of interest. The general framework is then specialized for the case where models provide point estimates about a single‐valued unknown, and where information about models are available in form of homogeneous and nonhomogeneous performance data (pairs of experimental observations and model predictions). Several example applications for physical models used in fire risk analysis are also provided.
