Abstract:Zearalenone (ZEN) is harmful to animals and human beings, so it is very important to develop a rapid and sensitive method for the detection of ZEN. In this paper, we proposed a novel ZEN-monitoring method using two aptamers as recognition elements and EnGen LbaCas12a and Nt.AlwI nicking endonuclease as signal amplifiers. When ZEN was present, it bound to the aptamer Z0 and, Z1 was released into solution. The solution was then separated and the Nt.AlwI enzyme was added in order to form a nicking-enzyme cycle, thereby producing large amounts of the ssDNA Z3 for 30 min. The Z3 formed a CRISPR-Cas12a-Z3 complex with CRISPR-Cas12a, activated the trans-cleavage ability of Cas12a, cleaved the Quenched Reporter for 20 min, and underwent fluorescence recovery. The aptasensor was able to sensitively detect ZEN in the linear range of 1–1000 pg/mL, with a detection limit as low as 0.213 pg/mL. The detection time lasted for 2 h. Additionally, this detection technology can also be used to monitor other hazards.

Abstract:Traditional methods for detecting antibiotic and mycotoxin residues rely on large-scale instruments, which are expensive and require complex sample pretreatment processes and professional operators. Although aptamer-based electrochemical sensors have the advantages of simplicity, speed, low cost, and high sensitivity, most aptamer-based sensors lack a signal amplification strategy due to their direct use of aptamers as probes, resulting in insufficient sensitivity. To solve the sensitivity problem in the electrochemical detection process, a novel electrochemical sensing strategy was established for ultrasensitive zearalenone (ZEN) detection on the basis of exonuclease I (Exo I) and branched hybridization chain reaction (bHCR) to amplify the signal. The amplification strategy showed excellent analytical performance towards ZEN with a low detection limit at 3.1×10 −12 mol/L and a wide linear range from 10 −11 to 10 −6 mol/L. Importantly, the assay was utilized in the corn powder samples with satisfactory results, holding promising applications in food safety detection and environmental monitoring.

Abstract:Zearalenone (ZEN) poses a serious threat to human and animal health. The development of sensitive determination methods for ZEN is of great significance for ensuring the quality and safety of food. Herein, based on a stimuli-responsive aptamer-functionalized metal-organic framework (MOF) nano-container and trivalent DNA peroxidase mimicking enzyme (DNAzyme), an efficient aptasensor was constructed initially for the colorimetric determination of ZEN. The proposed aptasensor only required simple operations but exhibited outstanding specificity, reproducibility, storage stability and reusability simultaneously. Under the optimal conditions, there was a good linear relationship between the changed absorbance and logarithm concentration of ZEN within 0.01-100 ng mL(-1), and the limit of detection (LOD) could reach 0.36 pg mL(-1). Moreover, the proposed aptasensor was reliable in quantifying ZEN in spiked food samples. The current bioassay provides a promising scheme for constructing stable, specific and rapid colorimetric platforms with potential applications in the fields of food safety.

Abstract:An electrochemical sensor was developed for detecting zearalenone (ZEN) based on the mimic peptide, which was screened from the library and validated by molecular simulation and electrochemical methods. The library of the mimic peptide was constructed according to the structural analysis, molecular docking, molecular dynamics and amino acid mutation. Then, the enhanced electrical signal was attributed to gold nanoparticles (AuNPs) and reduced carboxylated graphene oxide (rGO-COOH). Under the optimal conditions, the detection limit was 0.91 pg·mL-1 (S/N = 3) with a wide linear range from 0.01 ng·mL-1 to 10 ng·mL-1. In grain samples, a good recovery rate of 84% to 105.3% was achieved, while the rate ranged from 82% to 108.8% in the commercial ELISA kits. Additionally, the electrochemical sensor exhibited the remarkable specificity, excellent stability and better reproducibility (RSD = 1.94%). This sensor has great potential for rapidly detecting ZEN in food.

Abstract:Zearalenone (ZEN) is a widespread nonsteroidal mycotoxin with estrogen-like activity. Sensitive and reliable quantification of ZEN in food is critical to ensure food safety and safeguard agricultural production. Herein, by combining the metal-organic frameworks-based nanozyme (MOFzyme) and hyaluronic acid (HA)-DNA hydrogel, a well-designed colorimetric aptasensor was developed. The HA-DNA hydrogel was deposited on the surface of the bimetallic MOFzyme via strand-induced hybridization chain reaction (HCR). Relying on the ZEN-specific aptamer integrated in hydrogel structure, the disintegration of the hydrogel network and the concomitant exposure of the encapsulated MOFzyme could be specifically triggered by the introduction of ZEN molecules. Moreover, the magnitude of hydrogel disintegration was positively correlated with the amount of ZEN, by which the quantification of ZEN can be effortlessly achieved. Benefiting from the delicate design, the satisfactory catalytic performance and stability of bimetallic MOFzyme and the appealing stimuli-responsiveness of DNA hydrogel, the developed aptasensor demonstrated superior analytical performance and ease of use. Under optimal conditions, the linear range of the aptasensor fell between 0.001 and 200 ng mL-1 with a limit of detection (LOD) of 0.8 pg mL-1. Furthermore, the aptasensor was successfully applied for the quantitative detection of ZEN in corn and soybean samples with recoveries ranging from 94.0% to 109.0%. The developed aptasensor is expected to be a prospective universal platform for accurately quantifying food or environmental hazards.
A "Turn-On" FRET Aptasensor Based on the Metal-Organic Framework-Derived Porous Carbon and Silver Nanoclusters for Zearalenone Determination

Abstract:Zearalenone (ZEN) is a fungal secondary metabolite with an estrogen-like structure. Its widespread existence and potential hazards to human and animal health call for alternative rapid determination methods to monitor ZEN in food and the environment. Herein, a robust and enzyme-free "turn-on" aptasensor based on fluorescence resonance energy transfer (FRET) was developed for the sensitive identification of ZEN. Aptamer-modified silver nanoclusters (AgNCs) and porous Fe3O4/carbon octahedra derived from the metal-organic framework (MOF) acted as the FRET energy donor-acceptor pair in fluorescence quenching. DNA-templated AgNCs can be adsorbed onto the surface of porous Fe3O4/carbon octahedra through pi-pi stacking, which will lead to the FL quenching of AgNCs. However, in the presence of the ZEN, the formation of the ZEN/aptamer complex promoted the release of AgNCs and thereby brought about the recovery of fluorescence, which can be used to indicate the concentration of ZEN. The prepared aptasensor was characterized by various physicochemical techniques such as scanning electron microscopy (SEM), transmission electron microscopy (TEM), powder X-ray diffraction (PXRD), energy-dispersive X-ray spectrometry (EDX), X-ray photoelectron spectroscopy (XPS) and circular dichroism spectroscopy (CD). By combining the excellent quenching properties and easy separation of magnetic porous Fe3O4/carbon octahedra, as well as the appealing photostability and water solubility of AgNCs, the proposed FRET platform demonstrated superior analytical performance with a linear range of 0.01-250 ng mL(-1) and a LOD as low as 2 x 10(-3) ng mL(-1). Most importantly, the practicability and reliability of the FRET-based strategy were confirmed by conducting the determination of ZEN in real samples.

Abstract:Developing rapid and efficient analytical methods is of great importance for food safety Herein, we present a novel homogeneous electrochemical aptasensor for ultrasensitive quantitative determination of zearalenone (ZEN) based on a nanocomposite probe and silica nanochannel film. X-ray photoelectron spectroscopy, Fourier transform infrared spectroscopy and UV–Vis characterization techniques confirm that graphene oxide (GO) bears an aromatic conjugated structure, along with hydroxyl and carboxyl groups, facilitating the subsequent adsorption of cationic redox hexa-ammine-ruthenium (III) (Ru(NH3)63+) and anionic ZEN aptamer, to form a Ru(NH3)63+–ZEN aptamer–GO nanocomposite probe in a homogeneous solution. Vertically-ordered mesoporous silica films (VMSF) bearing silanol groups can be simply grown on the solid indium tin oxide (ITO) electrode surface and enable the selective preconcentration of Ru(NH3)63+, eventually leading to signal amplification. Since the detachment of Ru(NH3)63+ from the GO surface by the recognized ZEN aptamer in the presence of ZEN, more free Ru(NH3)63+ is released in solution and produces enhanced redox signals at the VMSF modified ITO electrode, allowing quantitative detection of ZEN. On the basis of the above sensing strategy, the proposed homogeneity, due to the assistance of graphene, as well as of the signal amplification and anti-fouling effects of VMSF, accurate analysis of ZEN can be realized in maize and Chinese chestnut samples.

Abstract:A facile strategy was successfully developed for the detection of zearalenone (ZEN). In this assay, highly fluorescent upconversion nanoparticles were synthesized and conjugated with the complementary oligonucleotide of ZEN aptamer for use as signal probes. Magnetic nanoparticles immobilized with the ZEN aptamer were assigned as capture probes. The results exhibited that the linear correlation between the decreased luminescence intensity of the signal and the concentration of ZEN was very strong (R2=0.9957) in the range of 0.05-100μg/L. In addition, the limit of detection of the proposed method (0.126μg/kg for corn and 0.007μg/L for beer) was significantly lower than the existing methods. Furthermore, the reliability of the competitive immunoassay format was validated by comparing the results determined in real food samples to those obtained from a commercially available method. Overall, the novel aptasensor have showed great potential for rapid and accurate ZEN determination.
Ultrasensitive detection of zearalenone based on electrochemiluminescent immunoassay with Zr-MOF nanoplates and Au@MoS2 nanoflowers

Abstract:In this work, an effective competitive-type electrochemiluminescence (ECL) immunosensor was constructed for zearalenone determination by using Zr-MOF nanoplates as the ECL luminophore and Au@MoS2 nanoflowers as the substrate material. Zr-MOF have an ultra-thin sheet-like structure that accelerates the transfer of electrons, ions and co-reactant intermediates, which exhibited strong and stable anodic luminescence. The three-dimensional Au@MoS2 nanoflowers would form a thin film modification layer on the glassy carbon electrode (GCE). And its good electrical conductivity and higher specific surface area utilization further improving the sensitivity of the ECL immunosensor. Under the optimized conditions, the proposed immunosensor exhibited satisfactory stability, sensitivity and accuracy, and its ECL signal was proportional to the logarithm of ZEN concentration (0.0001-100 ng/mL) and the limit of detection (LOD) was 0.034 pg/mL. In addition, the results of recovery experiment acquired for wheat flour and pig urine samples further proved the feasibility of the immunosensor for the detection of real samples, indicating its potential for ultrasensitive detection of ZEN.

Abstract:A novel enzyme-linked immunosorbent assay based on magnetic nanoparticles and biotin/streptavidin-HRP (MNP-bsELISA) was developed for rapid and sensitive detection of zearalenone (ZEN). The detection signal was enhanced and the sensitivity of the assay was improved by combined use of antibody-conjugated magnetic nanoparticles and biotin-streptavidin system. Under the optimized conditions, the regression equation for quantification of ZEN was y = -0.4287x + 0.3132 (R² = 0.9904). The working range was 0.07-2.41 ng/mL. The detection limit was 0.04 ng/mL and IC50 was 0.37 ng/mL. The recovery rates of intra-assay and inter-assay ranged from 92.8%-111.9% and 91.7%-114.5%, respectively, in spiked corn samples. Coefficients of variation were less than 10% in both cases. Parallel analysis of cereal and feed samples showed good correlation between MNP-bsELISA and liquid chromatograph-tandem mass spectrometry (R² = 0.9283). We conclude that this method is suitable for rapid detection of zearalenone in cereal and feed samples in relevant laboratories.

Abstract:Accurate and early diagnosis of zearalenone (ZEN) is particularly significant to the food safety. Herein, we propose an ionic liquid assisted self-assembly molecular imprinting strategy for ZEN based on ionic liquid functionalized boron-doped ordered mesoporous carbon -gold nanoparticles composite (BOMC-IL-Au NPs). During the composite synthesis, increased well-dispersed and uniform Au NPs are deposited on the surface of IL modified BOMC, due to the strong electrostatic interaction between AuCl4− and positively charged IL. For molecular imprinting, the BOMC-IL-Au NPs/GCE is immersed into p-aminothiophenol (p-ATP) solution and template solution in turn. Thus, the mercapto group contained p-ATP self-assembles on the Au NPs. Subsequently, the template molecules self-assemble onto the composite to form dense template layer, because of the hydrophobic interaction, π-π and hydrogen bond between template and IL/or p-ATP. After electropolymerization, the template layer is embedded into the p-ATP polymer membrane and produces lots of imprinting sites. Hence, the obtained sensor exhibits high sensitivity and selectivity. Under the optimal conditions, zearalenone can be quantified from 5 × 10−4 to 1 ng mL−1 with the low detection limit of 1 × 10−4 ng mL−1, by using [Fe(CN)6]3-/4- probe and square wave voltammetry. This strategy can also be employed to construct sensors for the detection of other substances.

Abstract:Zearalenone (ZEN) is a type of estrogenic mycotoxin commonly occurring in cereals. The aim of this study was to design a simple, rapid, inexpensive and ultrasensitive fluorescence assay for the determination of ZEN. Here, amino-modified mesoporous silica nanoparticles (MSNs-NH2) were synthesized to be the positive charge-rich reactor. A 6-carboxy-fluorescein-labeled aptamer (aptamer-FAM) was designed as the signal probe, ZEN-capture probe and negative charge reactor. In the absence of ZEN, the negatively charged aptamer-FAM combined with the positively charged MSNs-NH2 in an electrostatic manner. In the presence of ZEN, the fluorescence intensity in the supernatant increased significantly because the aptamer-FAM could bind to ZEN instead of MSNs-NH2. Under the optimal experimental conditions, this assay exhibited excellent specificity, repeatability and a wide linearity range of 0.005–150 ng/mL, with a detection limit of 0.012 ng/mL. Additionally, it showed high recovery (83.3–101.5%) for the spiked samples. There was no statistically significant difference in the ZEN concentrations detected by the proposed assay and HPLC in naturally contaminated samples. Overall, this design provides a new strategy for the rapid, inexpensive and sensitive detection of ZEN, and it could be applied to develop fluorometric assays for different targets by the selection of appropriate aptamers.

Abstract:As an estrogenic mycotoxin found in a wide range of agricultural crops, the toxicity of Zearalenone (ZEN) poses a serious risk to human health. Accordingly, to achieve rapid detection of zearalenone in complex samples, an aptamer fluorescence sensor based on magnetic graphene oxide was developed. Compared with traditional methods, this technique has the virtues of simple operation, low cost, and reliable performance. Magnetic graphene oxide (MGO) was synthesized as a fluorescent bursting agent, using a chemical precipitation approach by depositing Fe3O4 on the surface of graphene oxide. As a fluorescent probe, an aptamer coupling with CdTe quantum dots and zearalenone was used. Following the specific binding of zearalenone and the aptamer, the affinity interaction between the fluorescent probe and MGO was weakened, resulting in the recovery of fluorescence and making the qualitative and quantitative analysis of zearalenone available via fluorescence intensity determination. The results indicated that the method's linear range was 5-120 μg·L-1 and its detection limit was 2.9 μg·L-1. In addition, the recoveries varied from 76.4 to 118.8% for crop samples, with relative standard deviations (RSDs) between 0.8 and 9.5%, which suggesting an effective method for the separation and detection of mycotoxins in actual environmental samples.

Abstract:A refined electrochemical aptamer sensing technique using PEI@Ti-MOF@Ti3C2Tx-MXene was developed for the sensitive detection of ZEN in food samples. A titanium-based metal-organic skeleton (NH2-MIL-125) was synthesized in situ using 2-aminoterephthalic acid as the organic ligand and tetrabutyl titanate as the metal center, followed by the simultaneous hybridization of Ti3C2Tx-MXene to synthesize a Ti-MOF@Ti3C2Tx-MXene composite material. These composites were subsequently functionalized with PEI and covalently linked to form a sensing platform on gold electrodes. Integrating a metal-organic framework (MOF) with MXene materials not only improved the electrochemical properties compared to those of individual elements but also decreased the stacking effect and increased the number of binding sites for the aptamer. The limit of detection (LOD) of this sensor was 1.64 fg mL-1. Additionally, the sensor could efficaciously detect ZEN in cornmeal and beer samples, exhibiting outstanding stability, reproducibility, and selectivity. This highlighted its effectiveness in applications in quality supervision and food safety.

Abstract:In this study, a facile electrochemical sensing platform based on thin-layer MoS2 and thionin (MoS2-Thi) composites was constructed for the sensitive and rapid detection of zearalenone (ZEA) in human biofluids. MoS2-Thi composites with a thin-layer MoS2 structure and large specific surface area were synthesized via a simple ultrasound exfoliation method. To further enhance the performance of the platform but without increasing the complexity of the fabrication process, ZEA antibodies were modified with Pt nanoparticles (Pt NPs) using an environmentally friendly method, where they had a much wider linear range and four times higher sensitivity compared with the original ZEA monoclonal antibody (ZEA-MAb). Under the optimal conditions, the electrochemical responses of the MoS2-Thi composite-based immunosensor were linear (R-2 = 0.9915) when the ZEA concentrations ranged from 0.01 to 50 ng mL(-1). The proposed immunosensor could detect ZEA at concentrations as low as 0.005 ng mL(-1). with excellent selectivity. The immunosensor exhibited acceptable stability with high reproducibility and accuracy, and it was applied successfully for determining ZEA in human plasma and urine samples. Our findings indicated that the proposed MoS2-Thi composite-based electrochemical sensing platform provides a new approach for the rapid and sensitive bioanalysis of ZEA, and it may also be extended to other mycotoxins for multiplex detection.

Abstract:An ultralow-potential electrochemiluminescence (ECL) aptasensor has been designed for zearalenone (ZEN) assay based on a resonance energy transfer (RET) system with SnS(2) QDs/g-C(3)N(4) as a novel luminophore and CuO/NH(2)-UiO-66 as a dual-quencher. SnS(2) QDs were loaded onto g-C(3)N(4) nanosheets and enhanced the ECL luminescence via strong synergistic effects under an ultralow potential. Abstract:Zearalenone (ZEN) is a kind of mycotoxin with estrogen-like effect. The presence of ZEN in cereals is potentially harmful to the health of humans. Herein, for the first time, the binding mechanism of ZEN aptamer with 19 bases (Z19) were systematically studied by molecular dynamics, circular dichroism and isothermal titration calorim-etry. The binding driving forces, key bases of Z19 to ZEN, the conformational changes and thermodynamic in-formation were originally found. On this basis, by employing the inherent rapid nature of fluorescence polarization and target recycling induced by Exo III, a novel fluorescence polarization aptasensor was con-structed. The realized "add and measure" detection mode can be completed within one hour. Under the optimal conditions, the detection platform showed excellent analytical performance with a linear range of 0.01-100 ng/ mL and a limit of detection as low as 0.004 ng/mL. Moreover, the practicability and reliability of the constructed strategy were confirmed with standard HPLC-MS method in corn and wheat samples. The established aptasensor with enhanced sensitivity broaden the application scope of fluorescence polarization technique, especially in detection targets of low molecular weight.

Abstract:In this work, a potential-controlled electrochromic visual biosensor was developed for detecting zearalenone (ZEN) using a distance readout strategy. The sensor chip includes a square detection area and a folded signal output area created with laser etching technology. The detection area is modified with graphene oxide and ZEN aptamer, while Prussian blue (PB) is electrodeposited onto the signal output channel. When an appropriate voltage is applied, PB in the signal output area is reduced to colorless Prussian white (PW). The target ZEN molecules have the capability to release aptamers from graphene oxide (GO) surface in the detection area, resulting in a subsequent change in the potential of the visual signal output channel. This change determines the length of the channel that changes from blue to colorless, with the color change distance being proportional to the ZEN concentration. Using this distance readout strategy, ZEN detection within the range of 1 ng/mL to 300 ng/mL was achieved, with a detection limit of 0.29 ng/mL.
