Abstract:Microplastic (MP) and nanoplastic (NP) could cause gut microbiota alterations. Although micro/nanoplastic (MNP) degradation is attracting increasing scientific interest, the evaluation of MNP reduction in gut needs to be further investigated. This study aimed to determine whether partial reduction of polystyrene MNP in gut could affect the immunity, gut microbiota and metabolome of mice. Serum eotaxin/CCL11 was at a lower level in the mice exposed to 200 μg and 500 μg NP (i.e., 2NP and 5NP groups, respectively) compared to those exposed to 500 μg MP (i.e., 5 MP group), while serum IL-2 and IL-4 were both greater in the 5NP group compared to the 5 MP group. The gut bacterial alpha diversity, fungal diversity and evenness were all similar among the MNP and control groups. However, the gut fungal richness was greater in both the 5NP and 5 MP groups compared to the control group. The gut bacterial and fungal compositions were both different between the MNP and control groups. Multiple gut bacteria and fungi showed different levels between the 2NP and 5NP groups, as well as between the 2NP and 5 MP groups. Increased Staphylococcus and decreased Glomus were determined in the 2NP group compared to both the 5NP and 5 MP groups. A Lactobacillus phylotype was found as the sole gatekeeper in the bacterial network of the 2NP group, while a Bifidobacterium phylotype contributed most to the stability of the bacterial networks of both the 5NP and 5 MP groups. Multiple differential gut metabolic pathways were found between the 2NP and 5NP/5 MP groups, and mTOR signaling pathway was largely upregulated in the 2NP group compared to both the 5NP and 5 MP groups. The relevant results could help with the evaluation of partial reduction of MNP in gut.

Abstract:Nanoplastics adsorb surrounding organic contaminants in the environment, which alters the physicochemical properties of contaminants and affects associated ecotoxicological effects on aquatic life. The current work aims to explore the individual and combined toxicological implications of polystyrene nanoplastics (80 nm) and 6:2 chlorinated polyfluorinated ether sulfonate (Cl-PFAES, trade name: F-53B) in an emerging freshwater fish model Hainan Medaka ( Oryzias curvinotus ). Therefore, O. curvinotus were exposed to 200 μg/L of PS-NPs or 500 μg/L of F-53B in the single or mixture exposure for 7 days to investigate the effects on fluorescence accumulation, tissue damage, antioxidant capacity and intestinal flora. The PS-NPs fluorescence intensity was significantly higher in the single exposure treatment than it in combined exposure treatment ( p < 0.01). Histopathological results showed that exposure to PS-NPs or F-53B inflicted varying degree of damages to the gill, liver, and intestine, and these damage were also present in the corresponding tissues of the combined treatment group, illustrating a stronger extent of destruction of these tissues by the combined treatment. Compared to the control group, combined exposure group elevated the malondialdehyde (MDA) content, superoxide dismutase (SOD) and catalase (CAT) activities except in the gill. In addition, the adverse contribution of PS-NPs and F-53B on the enteric flora in the single and combined exposure groups was mainly characterised in the form of reductions in the number of probiotic bacteria (Firmicutes) and this reduction was aggravated by the combined exposure group. Collectively, our results indicated that the toxicological effects of PS-NPs and F-53B on pathology, antioxidant capacity and microbiomics of medaka may be modulated by the interaction of two contaminants with mutually interactive effects. And our work offers fresh information on the combined toxicity of PS-NPs and F-53B to aquatic creatures along with a molecular foundation for the environmental toxicological mechanism.

Abstract:The toxic effects of organic pollutants and nanoplastics on fish have been extensively studied, but there is limited research available on their combined toxicity to bivalves. This research aimed to investigate the accumulation and ecotoxicological impacts such as antioxidant capacity, histopathology and intestinal microbiota in white hard clam Meretrix lyrata , resulting from 7 days of single and mixture exposure to 3,3′,4,4′-tetrachlorobiphenyl (PCB77, 0.1 mg/L) and polystyrene nanoplastics (PS-NPs, 80 nm, 1 mg/L). Our findings revealed that PS-NPs accumulated in various tissues such as the intestine, gill, mantle, foot, and siphon. And when compared to the PCB-PSNPs (PP) co-exposure group, the intestinal fluorescence intensity mediated by plastic particles in the PS-NPs (PS group) was significantly higher. The gill, digestive gland, and intestine were all damaged to varying extent by single exposure to PS-NPs or PCB77, according to histopathological analysis, which was aggravated by PP group. Moreover, the co-exposure induced a higher level of oxidative stress, which reflected by increase of activities of superoxide dismutase, catalase, glutamate oxaloacetate transaminase and glutamic-pyruvic transaminase and malondialdehyde content. In addition, the intestine microbial composition was dramatically altered by the combined exposure, reducing the abundance of probiotics such as Firmicutes, thereby posing a great threat to the health and metabolism of M. lyrata . In conclusion, our findings showed that PS-NPs and PCB77 co-exposure induced a higher toxicity to M. lyrata , including histopathological changes, altered antioxidant capacity and intestinal microbiota disruption. This study provides novel insights into PCB77 and PS-NPs' combined toxicity to marine organisms and its underlying molecular mechanisms of ecotoxicological effects.

Abstract:As important emerging contaminants, nanoplastics can act as vectors for other environmental pollutants, resulting in their migration throughout ecosystems and altering their toxicity. In this study, the fluorescent dye label aggravated the toxicity of polystyrene (PS) nanoplastics (100 nm diameter particles) to microalgae Euglena gracilis. Therefore, the toxicity of non-fluorescent labelled PS alone and in combination with divalent cadmium (Cd2+) on Euglena gracilis in the environmentally relevant concentrations was investigated. Results revealed that co-exposure to 50 μg/L (1.1 × 1010 particles/L) PS and 50 μg/L Cd2+ resulted in synergistic effects, significantly inhibiting microalgal growth by 28.76%. Superoxide dismutase, peroxidase and extracellular polymeric substances were distinctly enhanced in co-exposure treatments compared to the control, indicating that cellular antioxidant defense responses were activated. LC-MS-based metabolomic analysis suggested that PS and Cd2+ exposure alone or in combination induced significant disruption to carbohydrate and purine metabolism-related pathways, as compared to controls. As part of the PS and Cd2+ stress response, differential metabolites involved in lipid metabolism and amino acid metabolism provide antioxidants and cell membrane protective molecules. Overall, this combined physiological and metabolomic analysis approach provides a better understanding of the potential risks posed by nanoplastics and heavy metal pollution in aquatic ecosystems.

[Long-term development of carotid stenosis treated by endovascular means: a prospective study in a neurovascular unit].

Abstract:AIMS Even today, unlike endarterectomy, long-term development in carotid angioplasty has still not been well defined. The aim of this study is to describe both the short- and long-term development and the rate of restenosis in a consecutive series of patients with carotid stenosis who underwent endovascular treatment at the Hospital Universitario La Fe; a distinction is made between patients with and without symptoms. PATIENTS AND METHODS From 1999 to 2010, 426 patients were submitted to a carotid angioplasty (25.5% were asymptomatic patients). Clinical follow-ups were conducted in the case of 374 patients. They had annual check-ups and every year a neurosonological study was performed. The aim was to determine what complications occurred during the first 30 days, as well as the long-term complications, to establish the rate of restenosis and to evaluate possible differences between patients with and without symptoms. RESULTS The morbidity and mortality rate in the first month was 4.2% (4.4% in symptomatic patients and 3.8% in asymptomatic patients). The mean follow-up time was 55 months: 8% of patients presented strokes, 11% suffered from myocardial infarct and 24.3% died, with no significant differences between patients with or without symptoms. The rate of restenosis was 17.3% at five years, 7.5% equal to or above 50% and, of those, 1.1% were symptomatic. Five patients with significant restenosis were successfully treated with endovascular techniques. CONCLUSIONS Carotid angioplasty is an effective, safe technique in the hands of an experienced professional; our findings are good, both in the short and the long term, with a low rate of restenosis. If this latter condition occurs, vascular intervention techniques are safe and efficient.
The role of gut microbiota in mediating increased toxicity of nano-sized polystyrene compared to micro-sized polystyrene in mice

Abstract:Microplastics (MPs) are widespread environmental contaminants that have been detected in animals and humans. However, their toxic effects on terrestrial mammals and the underlying mechanisms are still not well understood. Herein, we explored the role of gut microbiota in mediating the toxicity of micro- and nano-sized polystyrene plastics (PS-MPs/PS-NPs) using an antibiotic depleted mice model. The results showed that PS-MPs and PS-NPs exposure disrupted the composition and structure of the gut microbiota. Specifically, these particles led to an increase in pathogenic Esherichia-shigella, while depleting probiotics such as Akkermansia and Lactobacillus. Comparatively, PS-NPs particles had more pronounced effect, leading to obviously shifted the colon transcriptional profiles characterized by inducing the enrichment of colon metabolism and immune-related pathways (i.e., upregulated in genes like udgh, ugt1a1, ugt1a6a, ugt1a7c and ugt2b34). Additionally, both PS-MPs and PS-NPs induced oxidative stress, gut-liver damage and systemic inflammation in mice. Mechanistically, we confirmed that PS particles disturbed gut microbiota, activating TLR2-My88-NF-κB pathway to trigger the release of inflammatory cytokine IL-1β and TNF-α. The damage and inflammation caused by both size of PS particles was alleviated when the gut microbiota was depleted. In conclusion, our findings deepen the understanding of the molecule mechanisms by which gut microbiota mediate the toxicity of PS particles, informing health implications of MPs pollution.
Polystyrene nanospheres-induced hepatotoxicity in swamp eel ( Monopterus albus ): From biochemical, pathological and transcriptomic perspectives

Abstract:As ubiquitous emerging pollutants, microplastics (MPs) in aquatic environments have aroused critical global concerns. Despite the occurrence and characteristics of MPs in freshwater agroecosystems well-described by our previous study, their ecotoxicological implications in Monopterus albus remains unfathomed. Herein, we dissected toxic effects and mechanisms of PS-NPs exposure against M. albus hepatic tissues at concentrations of 0.5 (L), 5 (M), 10 (H) mg/L for 28 days using physiochemical measurements, histopathological analysis and transcriptomic sequencing. Results showed that upon PS-NPs treatments, levels of ROS, MDA, 8-OHdG and MFO activity were significantly enhanced relative to the control (C) group, while SP content and T-AOC activity were dramatically suppressed, suggesting ROS burst, lipid peroxidation and DNA damage may occur in liver tissues. This oxidative damage further triggered impaired hepatic function and histopathology, disordered lipid metabolism and hepatocyte apoptosis, as reflected by significantly diminished activities of GPT, GOT, ACP, AKP and LDH, paralleled with augmented levels of TG, TC and HSI as well as Cytc and Caspase-3,8,9 activities. Noticeably, concentration-dependent rises of apoptotic rate, vacuolar degeneration and lipid deposition were manifest in TUNEL, H&E and ORO staining. In addition, a total of 375/475/981 up-regulated as well as 260/611/1422 down-regulated DEGs in C vs L, C vs M and C vs H categories were identified based on RNA-seq, respectively. These DEGs were significantly annotated and enriched into GO terms (membrane, cytoplasm, response to stimuli, oxidation-reduction process) as well as KEGG pathways (ether lipid metabolism, apoptosis, chemical carcinogenesis-reactive oxygen species, non-alcoholic fatty liver disease). Moreover, signaling cascades Keap1-Nrf2, p53 and PPAR were either substantially initiated or dysregulated to orchestrate PS-NPs hepatotoxicity featuring oxidative damage, hepatocyte apoptosis and lipid steatosis. Collectively, this study not only expounded on toxicological mechanisms whereby PS-MPs exerted deleterious effects on M. albus , but also pointed to ecological risks of PS-MPs-induced hepatoxicity and lipid steatosis in this commercially-important species.

Abstract:The pervasive presence of nanoplastics (NPs) in marine environments poses a threat to marine organisms. Gills, as the organ in direct contact with the environment in marine invertebrates, maybe the first to accumulate NPs. To date, the toxic effects of NPs on the gills of marine invertebrates are still largely unknown. In this study, the response of multiple biomarkers (i.e., total antioxidant capacity, the activity of acetylcholine, ion content and transport enzyme, metabolic enzymes, and lipids content) in mussels Mytilus galloprovincialis exposed to polystyrene nanoplastics (PS-NPs) for 7 days were evaluated. Significant inductions of total antioxidant capacity (T-AOC) and inhibition of acetylcholine (AChE) activity were detected after 7 days of PS-NPs exposure. PS-NPs also triggered significant alteration in ion content (Na + and K + ) and suppressed the activities of the ion transport enzyme (Na + /K + -ATPase). Moreover, we found the activity of metabolic enzymes (succinate dehydrogenase and pyruvate kinase) and lipids content (triacylglycerol and cholesterol) were significantly altered, suggesting the interference of PS-NPs on energy metabolism and lipid metabolism. This investigation provides substantial information to understand the physical responses of invertebrate gills to PS-NPs exposure. Given the crucial ecological roles of invertebrates, the presence of PS-NPs in the marine environment may have far-reaching impacts on population abundance, biodiversity, and stability of the marine ecosystem.

Abstract:Nano-plastics (NPs) have emerged as prevalent contaminants in aquatic ecosystems, gaining significant research interest. Nonetheless, limited research has addressed the toxicity mechanisms associated with PS-NPs (polystyrene nanoplastics) of varying particle sizes. In this investigation, genotoxicity, growth patterns, hepatopancreatic damage, and intestinal flora alterations in freshwater shrimp Neocaridina palmata (Shen 1948), subjected to 35 days PS-NPs exposure (two size PS-NPs: 75 nm and 200 nm were used for this experiment, and five concentrations were set: 0 mg/L, 0.5 mg/L, 2.5 mg/L, 5 mg/L, and 10 mg/L concentrations PS-NP concentrations were examined using RNA sequencing, histopathological analyses, enzyme activity assessments, and 16S rRNA sequencing. Noteworthy variations in differentially expressed genes (DEGs) were identified across groups exposed to different PS-NPs sizes. We observed that PS-NPs predominantly instigated cellular component-related processes and induced apoptosis and oxidative stress across tissues via the mitochondrial pathway. Although the 200 nm-PS-NPs are stronger than the 75 nm-PS-NPs in terms of fluorescence intensity, 75 nm-PS-NPs are more likely to promote apoptosis than 200 nm-PS-NPs. PS-NPs impeded standard energy provision in N. palmata, potentially contributing to decreased body length and weight. Moreover, PS-NPs inflicted damage on intestinal epithelial and hepatopancreatic tissues and significantly modified intestinal microbial community structures. Specifically, PS-NPs-induced intestinal damage was marked by a decline in some probiotics (notably Lactobacilli) and a surge in pathogenic bacteria. Moreover, supplementing N. palmata with Lactobacilli appeared ameliorate oxidative stress and strengthen energy metabolism. Our findings provided valuable insights into crustacean toxicity mechanisms when subjected to PS-NPs and the potential risks that different PS-NPs sizes posed to terrestrial ecosystems.
Polystyrene microplastics induce molecular toxicity in Simocephalus vetulus: A transcriptome and intestinal microorganism analysis

Abstract:The global prevalence and accumulation of plastic waste is leading to pollution levels that cause significant damage to ecosystems and ecological security. Exposure to two concentrations (1 and 5 mg/L) of 500 nm polystyrene (PS)-nanoplastics (NPs) for 14 d was evaluated in Simocephalus vetulus using transcriptome and 16 s rRNA sequencing analyses. PS-NP exposure resulted in stress-induced antioxidant defense, disturbed energy metabolism, and affected the FoxO signaling pathway, causing neurotoxicity. The expression of Cyclin D1 (CCND), glucose-6-phosphatase (G6PC) and phosphoenolpyruvate carboxykinase (PCK) genes was decreased compared to the control, whereas the expression of caspase3 (CASP3), caspase7 (CASP7), Superoxide dismutase (SOD), Heat shock protein 70 (HSP70), MPV17, and Glutathione S-transferase (GST) genes was increased, thus, suggesting that NP ingestion triggered oxidative stress and disrupted energy metabolism.. PS-NPs were present in the digestive tract of S. vetulus after 14 days of exposure. In addition, the abundance of the Proteobacteria and opportunistic pathogens was elevated after PS-NPs exposure. The diversity and homeostasis of the S. vetulus gut microbiota were disrupted and the stability of intestinal barrier function was impaired. Multiomic analyses highlighted the molecular toxicity and microbial changes in S. vetulus after exposure to NPs, providing an overview of how plastic pollution affects freshwater organisms and ecosystems.
Transcriptomic analysis reveals nanoplastics-induced apoptosis, autophagy and immune response in Litopenaeus vannamei

Abstract:Increasing attention is being paid to the toxic physiological effects of nanoplastics (NPs) on aquatic organisms. However, few studies have systematically evaluated the regulatory mechanisms of NPs on immune response in crustaceans. In this study, a 28-day chronic exposure experiment was conducted in which shrimps were exposed to various 80-nm polystyrene NPs concentrations (0, 0.1, 1, 5 and 10 mg/L). Transcriptomic analysis was used to investigate the regulatory mechanisms of NPs in immune response of Litopenaeus vannamei. With increasing NPs concentration, the total hemocyte count (THC) content decreased, while phagocytosis rate (PR) and respiratory burst (RB) showed trends of first rising and then falling. High concentration (10 mg/L) of NPs caused the destruction of hepatopancreas tissue structure, the shedding of microvilli, the increase number of hepatocyte apoptosis and autophagy structure. With increasing NPs concentration, the lysozyme (Lys), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities first increased and then decrease, while contents of lipid peroxidation and malondialdehyde increased; the expression levels of Toll, MyD88, GPx, SOD, proPO, Lys, and ALF generally increased at first and then decreased. Transcriptional sequencing analysis showed that the pathway of differentially expressed genes in KEGG enrichment mainly included lysosome (ko04142), apoptosis (ko04210) pathways, indicating that the NPs mainly affected the immune regulatory mechanism. Further analysis by Gene Set Enrichment Analysis (GSEA) showed that the up-regulation pathways of NPs activation mainly included immune response-related pathways such as mitochondrial autophagy, DNA repair, autophagosomes signaling pathway. Our results indicated that NPs exposure induced oxidative stress, apoptosis and autophagy in shrimps. This study provides a basis for further understanding of the mechanisms of antioxidant immune regulation by NPs in shrimp and may serve as a reference for healthy ecological culture of shrimp.

Abstract:Shelf-life of scientific discoveries is usually less than few decades because science itself is an ever evolving discipline. Therefore, it is truly remarkable that pyloromyotomy described by Conrad Ramstedt (Fig. 1) is celebrating centennial this year. In October 1912, Ramstedt described the cure of hypertrophic pyloric stenosis (HPS) and that serendipitous discovery has stood the test of 100 years! (Fig. 2) It is also the most satisfying of all surgical procedures that offers consistent cure and it has saved millions of infants from premature death. Athena is pleased to join the fraternity of pediatric surgery in celebrating Ramstedt’s achievement [1]. Colossal success of a curative procedure usually obviates the need for further scientific research by solving the underlying problem. Athena is curious to know the impact of Ramstedt on the research and literature pertaining to HPS. She searched Pubmed titles using the key words “pyloromyotomy”, or “hypertrophic pyloric stenosis”. Contrary to the expectations, the number of research articles has progressively increased over the last century (Fig. 3). Some of the recent research papers conclude diametrically opposite of the views held in the past.

Studying the transcriptomic and metabolic responses of mice to nanoplastic-contaminated water is critical for understanding molecular-level toxicity of nanoplastics (NPs), but there are few studies on this topic. To analyze the effects of different concentrations of polystyrene (PS) nanoplastic-contaminated water on mice at the transcriptome and metabolism of spleens to study the molecular toxicity. Here, testing of histopathology of spleen of female mice was performed after drinking water containing 0.1 μm PS-NPs (1 mg/mL and 50 mg/mL) at different concentrations for 49 days, respectively. The spleen tissue samples were subjected to metabolome and transcriptome sequencing. Four differentially expressed genes were randomly chosen for qRT-PCR to confirm the correctness of transcriptome sequencing. Common Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis showed that a large number of differential genes and differential metabolites mainly focused on immune, inflammation, neurodegenerative disease, cardiovascular disease, nervous, etc. in the organism systems module; lipid, amino acid, taurine and hypotaurine metabolisms, etc. in the metabolism module; signaling translation, signaling molecules and interaction, and neuroactive ligand-receptor interaction, etc. in the environmental information processing. The results showed that pathway analysis at transcriptome and metabolome levels confirmed that the immune system of mice was affected after drinking water contaminated with polystyrene nanoplastics.
Toxicity of polystyrene nanoplastic and copper oxide nanoparticle in Artemia salina : single and combined effects on stress responses

Abstract:Nanoparticles, such as copper oxide nanoparticle (CuO NP) and polystyrene nanoplastic (PSNP), are increasingly released into aquatic environments, and pose potential risks to aquatic animals such as brine shrimps. Understanding the toxicity of these nanoparticles, especially when combined, is very important to assess their environmental effects. Therefore, this work describes the toxicity of polystyrene nanoplastic (PSNP) and CuO nanoparticles (CuO NPs) for brine shrimp ( Artemia salina ). The body length and stress biomarkers, including the activity of SOD, CAT, GST, Acid phosphatase, AChE, level of MDA and GSH, and expression of the hsp70 gene were quantified. The 48h-EC50 values for PSNP, CuO NPs, and their combination were determined as 1.024 and 5.089, and 0.512 mg L -1 , respectively. The combined exposure groups showed the highest growth inhibition. This was associated with increased activity of SOD and GST, decreased activity of CAT, a significant decrease in the level of GSH, a significant increase in the MDA level, and expression of the hsp70 gene (P<0.05). Moreover, an increased ACP and reduced AChE activity were observed in exposure groups. This study indicated that PSNP and CuO NPs have synergistic toxicity for A. salina , underscoring the importance of further investigation into their combined effect on aquatic animals.

Abstract:Nanoplastics (NPs, <1000 nm) may adsorb organic pollutants in the aquatic environment, thereby, influencing their bioavailability to organisms. This study aims to investigate the individual and combined toxicity of perfluorooctane sulfonate (PFOS) and virgin yellow-green fluorescent polystyrene NPs (200 nm) at sub-lethal doses on the marine mussel Perna viridis. Our results demonstrated that both PFOS single and PFOS-NP co-exposure at 1000 μg/L significantly increased PFOS distribution in the gills, gonads, and visceral mass (p < 0.05), compared to the control. Further, PFOS single and PFOS-NP co-exposures at 100 and 1000 μg/L significantly increased the reactive oxygen species (ROS) levels in mussel tissues that consequently altered the responses of antioxidant entities including MDA, CAT, SOD, GR, and GST. The transcriptional profiling of oxidative stress-related genes (cyp4, hsp22, hsp60, gst-omega, and gst-pi), showed significantly downregulated expressions at the lowest level of co-exposure (PFOS 10 μg/L) in all tissues, especially in gills, compared to the control group. Overall, the enhanced integrated biomarker response (EIBR) revealed PFOS-NP co-exposure at 1000 μg/L, as the most stressful circumstance to induce mixture toxicity, at which more structural damage to the gills and gonads were observed than single PFOS/NPs exposure. In , the co-exposure significantly enhanced the PFOS bioaccumulation and ROS levels in mussel tissues, resulting in altered antioxidant and genetic responses, suggesting that NPs could affect the distribution of PFOS between P. viridis and seawater. Hence, further studies should be conducted to unveil the interactive toxic effects of NPs and PFOS on marine organisms.

Abstract:In the marine environment, plastic pollution may occur simultaneously with hypoxia. However, current ecological risk assessments of nanoplastics have rarely considered the impact of additional environmental factors, such as hypoxia. In this study, we investigated the effect of polystyrene nanospheres (PS-NPs) on the digestive performance (antioxidant system and digestive enzymes) of mussels Mytilus galloprovincialis under different patterns of hypoxia (normoxia, constant hypoxia, and fluctuating hypoxia). The result showed that PS-NPs caused oxidative damage in the digestive glands of mussels, while all patterns of hypoxia exacerbated this oxidative damage. Activities of four digestive enzymes (α-amylase, cellulase, trypsin, and lipase) were examined. Among these, the activity of the α-amylase was inhibited by PS-NPs, and the inhibition was aggravated by all the hypoxia patterns. The cellulase activity and trypsin activity was enhanced by PS-NPs, and the increase was further stimulated by hypoxia. Lipase activity was not affected by PS-NPs alone, but significant inhibition was detected after the coexposure to PS-NPs and hypoxia. Conclusively, the combined stress of hypoxia and nanoplastics can significantly affect the digestive performance of mussels and may alter the mussel nutrient uptake strategy. Our work has provided new insight into the ecological risk assessment of plastics under global climate change.

Abstract:Microplastics (MPs) pollution and salinity variation are two environmental stressors, but their combined effects on marine mollusks are rarely known. Oysters (Crassostrea gigas) were exposed to 1 × 104 particles L-1 spherical polystyrene MPs (PS-MPs) of different sizes (small polystyrene MPs (SPS-MPs): 6 μm, large polystyrene MPs (LPS-MPs): 50-60 μm) under three salinity levels (21, 26, and 31 psu) for 14 days. Results demonstrated that low salinity reduced PS-MPs uptake in oysters. Antagonistic interactions between PS-MPs and low salinity mainly occurred, and partial synergistic effects were mainly induced by SPS-MPs. SPS-MPs induced higher lipid peroxidation (LPO) levels than LPS-MPs. In digestive glands, low salinity decreased LPO levels and glycometabolism-related gene expression, which was related to salinity levels. Low salinity instead of MPs mainly affected metabolomics profiles of gills through energy metabolism and osmotic adjustment pathway. In conclusion, oysters can adapt to combined stressors through energy and antioxidative regulation.

Abstract:Microplastics (MPs) are currently one of the major environmental problems due to the large amount released in aquatic and terrestrial ecosystems as well as their potential impacts on organisms and human health. However, the molecular mechanisms underlying MP toxicity remain elusive and insufficiently understood. In this study, oysters were exposed to irregular MPs composed of polyethylene and polyethylene terephthalate at concentrations of 10 and 1000 μg L−1 for 21 days under static conditions, and physiological responses and changes in antioxidant enzymatic activity were investigated. Moreover, the protein expression profiles in the digestive glands were analyzed by using the isobaric tags for relative and absolute quantitation (iTRAQ) method. And the expression of ten potentially regulated genes was analyzed by qRT-PCR in different tissues under MP exposure. It was demonstrated that PE and PET MP exposure had no distinct adverse effects on physiological rates, while inducing oxidative stresses in the oyster. Kyoto encyclopedia of genes and genomes (KEGG) enrichment analysis of DEPs revealed that MP exposure mainly disturbed the "arachidonic acid metabolism", "linoleic acid metabolism" and "glycerophospholipid metabolism" processes in oysters. Gene Ontology enrichment analysis revealed that MPs affected the oxidation-reduction process, lipid metabolism process and pentose phosphate pathway. Moreover, a significant increase in the mRNA expression of genes related to lipid and aerobic metabolism and apoptosis pathways was observed after MP exposure. In summary, our results indicated that MPs could induce oxidative stress and alter lipid and glucose metabolism processes in oysters. Additionally, the weight of evidence (WOE) model analysis suggested that high concentrations of MPs were more hazardous than low concentrations and typically higher for PET than PE. This study provided the effects of PE and PET MP toxicity on oysters at the molecular level, which will contribute to better understanding the defense mechanisms of oysters against MP exposure.

Abstract:Nanoplastics (NPs) are one of the most hazardous marine litters, having the potential to cause far-reaching impacts on the environment and humankind. The effect of NPs on fish health has been studied, but their impact on the subcellular organelles remains unexplored. The present investigation studied the possible implications of polystyrene-nanoplastics (PS-NPs) on the hematology, tissue organization, and endoplasmic reticulum (ER) stress-related proteins in Nile tilapia (Oreochromis niloticus). Fish were exposed to ∼100 nm PS-NPs at environmentally relevant (0.1 mg/L), and sublethal (1, 10 mg/L) concentrations for 14 days through water exposure. The growth performance and hematological parameters such as erythrocytes, hemoglobin, hematocrit, and leucocytes decreased, while thrombocytes increased with PS-NPs dose-dependently. The gills, liver, kidney, and heart tissues displayed increasing degrees of pathology with increased concentrations of PS-NPs. The gills showed severe epithelial hyperplasia and lamellar fusion. The liver had an abstruse cellular framework, membrane breakage, and vacuolation. While glomerular and tubular atrophy was the most prominent pathology in the kidney tissue, the heart displayed extensive myofibrillar loss and disorderly arranged cardiac cells. The ER-stress-related genes such as bip, atf6, ire1, xbp1, pkr, and apoptotic genes such as casp3a, and bax were over-expressed, while, the anti-apoptotic bcl2 was under-expressed with increasing concentrations of PS-NPs. Immunohistochemistry and blotting results of GRP78, CHOP, EIF2S, and ATF6 in gills, liver, kidney, and heart tissues affirmed the translation to ER stress proteins. The results revealed the sub-lethal adverse effects and the activation of the ER-stress pathway in fish with sub-chronic exposure to PS-NPs.
