Abstract:Ricin, a category-B agent for bioterrorism, and Shiga toxins (Stxs), which cause food poisoning bind to the ribosomal P-stalk to depurinate the sarcin/ricin loop. No effective therapy exists for ricin or Stx intoxication. Ribosome binding sites of the toxins have not been targeted by small molecules. We previously identified CC10501 , which inhibits toxin activity by binding the P-stalk pocket of ricin toxin A subunit (RTA) remote from the catalytic site. Here, we developed a fluorescence polarization assay and identified a new class of compounds, which bind P-stalk pocket of RTA with higher affinity and inhibit catalytic activity with submicromolar potency. A lead compound, RU-NT-206, bound P-stalk pocket of RTA with similar affinity as a five-fold larger P-stalk peptide and protected cells against ricin and Stx2 holotoxins for the first time. These results validate the P-stalk binding site of RTA as a critical target for allosteric inhibition of the active site.

Abstract:OBJECTIVE:To study lysosomes involvement in the degradation of ricin A chain.METHODS:A lysosome-targeted singal KFERQ was added to the C terminus of rRTA by DNA recombinant technology. A pKK223.3 expression system in E. coli was used to produce recombinant ricine A chain (rRTA) and rRTA-KFERQ. Recombinant proteins were purified by affinity chromatography using Blue-Sepharose 6B. The cytotoxicity of recombinant proteins was measured by the MTT method.RESULTS:Recombinant RTA-KFERQ was 49.87%, 54.18% and 88.68% less cytotoxic than RTA itself on the three cell lines HEPG2, Hela and A549, respectively.CONCLUSION:Lysosomes can degrade, but not completely inactivate RTA in different cells, suggesting cells may have other degradation pathways for RTA.

Abstract:Ricin enters the cells by receptor-mediated endocytosis, followed by translocation across the membranes of intracellular organelles. A trans-Golgi retention peptide signal YQRL was fused to the C-terminus of ricin A chain (RTA) by polymerase chain reaction. The recombinant RTA and RTA–YQRL were expressed in Escherichia coli using plasmid pKK223.3 under the control of a tac promoter. The recombinant proteins were purified by affinity chromatography on a Blue-Sepharose 6B column. The cytotoxicities of RTA and the fusion toxin RTA–YQRL were measured by the MTT assay in HeLa, SKOV-3, and WISH cells following fluid-phase endocytosis. The rRTA–YQRL was 2-, 10-, and 40-fold more cytotoxic than rRTA itself in the three cell lines, respectively. The results indicate that addition of a TGN retention signal YQRL to the C-terminus of RTA can markedly increase its cytotoxicity, suggesting TGN may play an important role in the intracellular routing and translocation of RTA.
Detection of functional ricin by immunoaffinity and liquid chromatography-tandem mass spectrometry

Abstract:The toxin ricin is a biological weapon that may be used for bioterrorist purposes. As a member of the group of ribosome-inactivating proteins (RIPs), ricin has an A-chain possessing N-glycosidase activity which irreversibly inhibits protein synthesis. In this paper, we demonstrate that provided appropriate sample preparation is used, this enzymatic activity can be exploited for functional ricin detection with sensitivity similar to the best ELISA and specificity allowing application to environmental samples. Ricin is first captured by a monoclonal antibody directed against the B chain and immobilized on magnetic beads. Detection is then realized by determination of the adenine released by the A chain from an RNA template using liquid chromatography coupled to tandem mass spectrometry. The immunoaffinity step combined with the enzymatic activity detection leads to a specific assay for the entire functional ricin with a lower limit of detection of 0.1 ng/mL (1.56 pM) after concentration of the toxin from a 500 microL sample size. The variability of the assay was 10%. Finally, the method was applied successfully to milk and tap or bottled water samples.

Abstract:Objective To design and express a novel peptide based on ricin toxin antibody in E. coli, and to evaluate its biological activity. Methods Based on the crystal structure of ricin toxin A chain (RTA) and the RTA-rRNA interact in the complex model, the steric conformation of RTA was theoretical modeled and its functional domain was preliminarily determined. The humanized single-domain RTA antibody was designed rationally by computer-guidod molecular design method. Its coding sequence was ob- tained by overlapping extension PCR, and cloned into the pET-32a vector. The fusion protein was then ex-pressed in E. coli BL21 (DE3), identified by Western blot, and purified with Ni-NTA agarose. The binding and neutralizing activity of this novel peptide for riein was evaluated by competitive ELlSA assay and MTT assay. Results A recombinant human single-domain antibody expressing a polypeptide against RTA in the CDR3 loop was designed. The fusion protein was successfully expressed in E. coll. The purified protein can bind to ricin, and neutralize its activity in SP2/0 viability assay. Conclusion The success of the novel pep-tide based on riein toxin antibody provides a novel method to develop new generation of ricin antagonists.

Abstract:Here we describe an affinity molecule-directed surface plasmon resonance (SPR) immunosensor for a label-free, differentiation and quantification of ricin and abrin from their structural highly like agglutinin biotoxins. By an introduction of protein G as the affinity capturing molecule, we fulfilled a complete strategy contains (i) screening monoclonal antibodies to be paired in a sandwiched format, (ii) differentiate quantification from the agglutinin, (iii) ascertain of active from inactive biotoxin, and (iv) structural identification of captured biotoxins on a single chip. By the aid of an enrichment step from immunomagnetic beads, we could accurately measure ricin or abrin with a concentration lowered to 0.6 ng/mL (10 pM) in different complex matrices such as stevia, protein powder, and human plasma, with linear ranges of two or three orders of magnitude, and satisfied recovery. We then differentially quantified the mixed crude extracts from castor beans and jequirity peas, and real samples from the fourth OPCW biotoxin exercise to prove the practical availability. We further provided a SPR-mass spectrometric evidence directly obtained from Protein G affinity chip via a noncovalent molecule surface for the first time for definitely structural identification for crude extracts.

Abstract:We have developed an aptamer-based assay for the food toxin ricin. It is based on a competitive quenching strategy along with time-resolved fluorescence detection. The assay involves the following steps: (a) The aptamer is immobilized on the europium-doped KGdF4 nanoparticles (NPs); (b) these NPs are added to the sample where ricin binds to the aptamer; (c) graphene oxide (GO) is added and competitively binds to the aptamer on the NPs that are not blocked by ricin; this causes the quenching of the fluorescence of the NPs; (d) fluorescence is detected at 593 nm in a microplate reader in the time-resolved mode at an excitation wavelength of 273 nm, a delay time of 100 μs, and a gating time of 1 s. Under optimal conditions, the calibration plot is linearly related to the concentration of ricin in the 50 pg·mL−1 to 50 ng·mL−1 range (R2 = 0.9975), and the limit of detection is 8 pg·mL−1. The method was compared to a standard ELISA, and correlation was excellent. The assay presented here provides a sensitive, dependable and convenient platform that is expected to have promising applications for the homogeneous assay of various other target analytes.

Abstract:Previous studies of inhibitors of ricin A chain (RA) mainly focused on the analogues of adenine and ribosomal RNA (rRNA) substrates. In this paper, a novel antagonist peptide (named PT) was designed rationally based on the crystal structure of the complex RA–rRNA. Theoretical results had clearly revealed the blockage of PT in the RA–rRNA interaction. The competitive inhibition experiment indicated that PT could significantly inhibit the binding activity of RA with anti-RA antibody. In order to further prove the competitive effect of PT against RA, N-glycosidase antagonizing activity of PT in cell-free system was evaluated using luciferase protein synthesis inhibition assay. Consequent data demonstrated that, at a RA level (0.022 nM) giving 50% decrease of protein synthesis in the absence of the peptide, protein synthesis could be recovered by the peptide for up to 80% at a level of 0.1 microgram/ml. This study highlights the interest of computation-aided method in the design of novel peptides with the ability to block the deleterious biological effects of RA. In addition, the method of luciferase protein synthesis inhibition assay in cell-free system which should provide rapid, sensitive, selective, and quantitative assessment may be developed to evaluate the potential antagonizing activity of RA inhibitors.

Abstract:Objective To study the inhibitory effect of pterin acid (PTA) against ricin and recombinant ricin A chain protein.Methods Luciferase protein synthesis inhibition assay in a cell-free system and in vitro cytotoxicity experiments were performed to assess the biological activity of ricin and rRTA treated with PTA.Results The result showed that PTA could significantly inhibit the activity of ricin and rRTA in a dose-dependent manner.Conclusion PTA might be used as a small molecular probe to develop an evaluating system for ricin/RTA small molecular inhibitor in vitro.The cell-free system adopted in the current study could also serve as a necessary basis for screening some novel small molecular compounds against ricin and RTA in the future.

Abstract:Ricin and abrin are highly potent plant-derived toxins, categorized as type II ribosome-inactivating proteins. High toxicity, accessibility, and the lack of effective countermeasures make them potential agents in bioterrorism and biowarfare, posing significant threats to public safety. Despite the existence of many effective analytical strategies for detecting these two lethal toxins, current methods are often hindered by limitations such as insufficient sensitivity, complex sample preparation, and most importantly, the inability to distinguish between biologically active and inactive toxin. In this study, a cytotoxicity assay was developed to detect active ricin and abrin based on their potent cell-killing capability. Among nine human cell lines derived from various organs, HeLa cells exhibited exceptional sensitivity, with limits of detection reaching 0.3 ng/mL and 0.03 ng/mL for ricin and abrin, respectively. Subsequently, toxin-specific neutralizing monoclonal antibodies MIL50 and 10D8 were used to facilitate the precise identification and differentiation of ricin and abrin. The method provides straightforward and sensitive detection in complex matrices including milk, plasma, coffee, orange juice, and tea via a simple serial-dilution procedure without any complex purification and enrichment steps. Furthermore, this assay was successfully applied in the unambiguous identification of active ricin and abrin in samples from OPCW biotoxin exercises.

Abstract:The ultrasensitive and rapid detection of ricin B toxin (RTB) is essential for food safety and environmental monitoring. Herein, a dual-mode magnetic relaxation switch (MRS) and fluorescence (FL) biosensing strategy was developed to efficiently detect RTB using fluorescent magnetic nanoparticles (MNP300@SiO2(FITC)). Meanwhile, the as-prepared composite MNP300@SiO2(FITC) exhibited superior biocompatibility and increased FL readout and was coupled with aptamer (Apt) to form a captured probe. Magnetic nanoparticles, 30 nm in diameter (MNP30), were coupled to a Blocker to form a paired probe to compete with RTB for Apt binding. The presence of the RTB triggered the dual-mode detection switch, thus, weakening the magnetic and fluorescent signals. Compared with the single-mode detection method, the Δ T2 and Δ FL intensity here exhibited an excellent linear relationship with logarithm of RTB concentrations at 0.001–500 ng/mL and 0.005–500 ng/mL, and obtained ultrahigh sensitivities of 0.8 pg/mL and 3 pg/mL, respectively. In addition, the dual-mode biosensor gained satisfactory spiked recoveries and relative standard deviations for quantitative detection of spiked RTB in edible oil and tap water samples. To our knowledge, this is the first study to describe the accurate quantification of RTB using a sensitive MRS-FL biosensor. We anticipate that this strategy will provide novel avenues for the development of dual-mode sensing assays.

Activity of Recombinant Toxin Ricin A Chain and Several Wild Type Single Chain Ribosome Inactivating Proteins

Abstract:Recombinant ricin toxin A chain(rRTA)is one of the important elements used for constructing immunotoxin,which may be useful as a treatment of many diseases.To obtain rRTA,ricin toxin A chain gene was cloned into pExSecⅠ vector and expressed in E.coli BL21(DE3),the proportion of expressed rRTA in total bacteria protein was about 20%.The expression products obtained a purity of 80% or so by a CM Sepharose column.Recombinant ricin toxin A chain,and several wild type single chain ribosome inactivating proteins,were analyzed by cleavage assay of supercoiled DNA and 3 H Leu labeled protein synthesis inhibition assay.The results suggest that rRTA had the similar activity as the others while there had no linear correlation between the two methods.
A potential therapeutic peptide-based neutralizer that potently inhibits Shiga toxin 2 in vitro and in vivo

Abstract:Shiga toxin 2 (Stx2) is a major virulence factor in infections with Stx-producing Escherichia coli (STEC), which can cause serious clinical complications in humans, such as hemolytic uremic syndrome (HUS). Recently, we screened and identified two peptide-based Stx2 neutralizers, TF-1 and WA-8, which specifically and directly bind to Stx2. Computer simulations suggested that the majority of TF-1 or WA-8 binds tightly at the receptor-binding site 3 of Stx2. The two peptides also effectively inhibited the cytotoxic activity of Stx2 by blocking the binding of Stx2 to target cells. TF-1 exhibits remarkable therapeutic potency in both mice and rat toxicity models. In mice toxicity models, TF-1 provided full protection when mice were injected with 5 LD 50 of Stx2. In rat toxicity models, TF-1 reduced fatal tissue damage and completely protected rats from the lethal challenges of Stx2. In these rats, TF-1 significantly decreased the concentration of Stx2 in blood and diminished tissue distribution levels of Stx2. Furthermore, TF-1 effectively protected rats from the pathological effects caused by Stx2, especially in the kidney, thymus, adrenal gland, and lung. Taken together, these results indicate that TF-1 is a promising therapeutic agent against the pathogenicity of Stx2.

Abstract:Objective To construct a prokaryotic expression vector in which the maximum level of rRTA-Trx fusion protein was attained with homogeneous,soluble,and full bioactive,and to prepare anti-rRTA monoclonal antibody(mAb) and establish sandwich ELISA for ricin detection.Methods A pair of RTA special primer was designed based on computer-aided design,RNA secondary structure,and also Escherichia coli partialism codon.RTA gene was cloned from PUC19/RA to pET32a(+) by PCR.Prokaryotic expression vector pET32a(+)/rRA/JM109 was eatablished and identified by double enzyme digestion and DNA sequencing.After induction with IPTG,the rRTA protein was obtained,and then purified by Ni2+-NTA resin column and evaluated by ELISA and Western blot assays.An anti-RTA mAb was obtained by using hybridoma technique and identified by Western blot assay.A sandwich ELISA for ricin detection was also established.Results a prokaryotic expression vector pET32a(+)/rRA/BL21 was constructed.Recombinant RTA proteins with high biological activity were successfully produced in the pET32a vector expression system.After purification with Ni2+-NTA resin column,the extraction showed a yield of approximately 10-20 mg/L.An anti-RTA mAb was obtained with hybridoma technique and specific ELISA screening.Western blotting showed that the mAb had specific binding ability to RTA in reduced SDS-PAGE.A sandwich ELISA for detects ricin was established with a ricin detection limit of 3.125 ng/mL.Conclusion rRTA,anti-RTA mAb,and a sandwich ELISA for ricin detection have practical use in biotherapy against carcinoma and in prophylaxis,diagnosis and therapeutics against bioterror attack of in the future.

Abstract:The retrograde transport inhibitor Retro-2 has a protective effect on cells and in mice against Shiga-like toxins and ricin. Retro-2 causes toxin accumulation in early endosomes and relocalization of the Golgi SNARE protein syntaxin-5 to the endoplasmic reticulum. The molecular mechanisms by which this is achieved remain unknown. Here, we show that Retro-2 targets the endoplasmic reticulum exit site component Sec16A, affecting anterograde transport of syntaxin-5 from the endoplasmic reticulum to the Golgi. The formation of canonical SNARE complexes involving syntaxin-5 is not affected in Retro-2-treated cells. By contrast, the interaction of syntaxin-5 with a newly discovered binding partner, the retrograde trafficking chaperone GPP130, is abolished, and we show that GPP130 must indeed bind to syntaxin-5 to drive Shiga toxin transport from the endosomes to the Golgi. We therefore identify Sec16A as a druggable target and provide evidence for a non-SNARE function for syntaxin-5 in interaction with GPP130.

Evaluation of an Electrochemiluminescence Assay for the Rapid Detection of Ricin Toxin

Abstract:In this article, we detail a comprehensive laboratory evaluation of an immunoassay for the rapid detection of ricin using the Meso Scale Diagnostics Sector PR2 Model 1800. For the assay evaluation, we used inclusivity, exclusivity, and informational panels comprised of extracts of 35 near-neighbor plant cultivar-extracts, 66 lectins, 26 white powders, 16 closely related toxins and proteins/toxoids, and a pool of 30 BioWatch filter extracts. The results show that the Meso Scale Diagnostics ricin detection assay exhibits good sensitivity and specificity with a limit of detection of 1.2 ng/mL. However, the dynamic range of the assay for the quantitation of ricin was limited. We observed a hook effect at higher ricin concentrations, which can lead to potential false negative results. A modification of the assay protocol that incorporates extra wash steps can decrease the hook effect and the potential for false negative results.

Abstract:Ricin and its A chains can be used to conjugate with monoclonal antibodies to prepare immunotoxins. Ricin A chain (RTA) and its modification RTA-KDEL (ER-retrieval signal) were expressed with the pKK223.3 system in Escherichia coli under control of a tac promoter. The recombinant proteins can be purified by one-step affinity chromatography on a column of Blue-Sepharose 6B. The toxicities of RTA and its mutant RTA-KDEL were evaluated by the MTT assay in HeLa, MCF, and ECV-304 cells following fluid-phase endocytosis. RTA-KDEL was somewhat more cytotoxic than RTA itself in the different cell lines. The results suggest that rRTA-KDEL may be useful for the synthesis of more potent immunotoxins.
The impact of a marriage enrichment program on relationship factors.

Abstract:This study represented a continuing effort to assess the effects of a marriage enrichment program previously used by Kilmann, Moreault, and Robinson. Eleven couples who responded to newspaper and radio announcements received fair-fight training followed by sexual enhancement. Six couples recruited in the same manner received no treatment but responded to the measures at the same time intervals as the experimental couples. In comparison with the control couples the experimental couples reported a significant increase in marital adjustment and significantly greater actualized relationship. From a videotaped problem-solving task the experimental couples were rated as exhibiting significantly more positive communication patterns than the control couples. The findings offer support for the marriage enrichment program, although larger samples are needed to determine which treatment components are most beneficial for defined couples.

Abstract:A simple two-step protocol for modification of atomic force microscopy (AFM) tip and substrate by using a "click reaction" has been developed. The modified tip and substrate would be applied to detect trace amounts of ricin by using atomic force microscopy. A key feature of the approach is the use of a PEG (polyethylene glycol) derivative functionalized with one thiol and one azide ending group. One end of the PEG was attached to the gold-coated AFM tip by a strong Au-thiol bond. The azide group hanging at the other end of the immobilized PEG was used for the attachment of an antiricin antibody modified with an alkyne group using a "click reaction". The latter reaction is highly efficient, compatible with the presence of many functional groups and could proceed under mild reaction conditions. In a separate step, ricin was immobilized on the gold substrate surface that was modified by active esters. For this process, a novel bifunctional reagent was employed containing an active ester and a thioctic acid moiety. By these modification processes, AFM recognition imaging was used to detect the toxin molecules and the results show fg/mL detection sensitivity, surpassing the existing detection techniques. With measurement of the unbinding force between the antiricin antibody and ricin, which was statistically determined to be 64.89 +/- 1.67 pN, the single molecular specificity of this sensing technique is realized.
