Abstract:The physiological profiling on community level and analysis of 16S rRNA V6 tag based on high throughput sequencing were used to characterize and to compare microbial community structure, diversity, and bacterial phylogeny from sediments before and after remediation with the help of nitrate. The experiment was conducted in situ within a heavily polluted river located in the south of China. Total 2900 m(2) area of sediment were selected on river bed for in situ test. The results showed that about 86.5% of nitrate on average can be consumed within 14 days since injection into contaminated sediment in river and about 86.5% on average of the odor matters of AVS (acidic volatile sulfide) can be removed. It was found that the microbial community richness and diversity decreased during remediation process after nitrate injection due to overgrowth of functional bacteria and then revived on the 14th day nearly the end of remediation process. The most abundant phyla were Proteobacteria (44.10%), Firmicutes (7.22%) and Cloroflexi (6.70%) while 24.90% of the sequences were from bacterial domain. Proteobacteria were found significantly abundant in treated sediments on the 7th day, which were resulted from significant abundance of beta-proteobacteria, epsilon-proteobacteria and gamma-proteobacteria. On the contrary, Firmicutes, Cloroflexi, Actinobacteria and Deltaproteobacteria were significantly abundant in untreated sediments and in sediments after 14 days treatment. Microbial community in sediment before remediation was closely adapted to the sediment conditions with serious sulfide pollution. Injection of nitrate into sediment played multi-roles as stimulation of nitrate reducing bacteria (NRB), inhibition of sulfate reducing bacteria (SRB) as well as increase of oxidation-reduction potential (ORP). Electrical Stimulation Accelerated Phenanthrene Biodegradation Coupling with Nitrate Reduction in Groundwater

Abstract:Phenanthrene (PHE) and nitrate co-contamination widely exists in the groundwater. Anaerobic bioremediation of the two suffered from the slow removal rate and limited electron transfer within microbes. Herein electrical stimulation (ES) was proved to stimulate the simultaneous removal of PHE and nitrate anaerobically. High removal of PHE and nitrate (90.94 % and 100 %, respectively) was obtained within 24 h in the ES-bioreactor, two times higher than the control anaerobic bioreactor (32.16 %). ES-bioreactor exhibited a good PHE removal performance within initial PHE ranging from 5 to 25 mg/L. In ES-bioreactor, extracellular polymeric substances increased PHE bioavailability and acted as protective capsules. Nicotinamide adenine dinucleotide (NADH) and cytochrome C (cyt-C) facilitated the extracellular and intracellular electron exchange during PHE biotransformation. ES significantly selected Acetobacterium, PHE-degraders (f_Bacteroidetesvadin HA17, Limno-bacter and Stenotrophomonas) and electrogens (Clostrium_sensu_stricto_13) to participate in PHE decomposition. Acetobacterium decomposed complex organics to acetate, proving accessible substrates for electrogens to main-tain an efficient electron flow. Enzymes regulating bacterial chemotaxis, flagellar assembly and carbohydrate metabolism were up-regulated with ES, suggesting the indispensable role of such enzymatic processes in ES-bioreactor. Overall, this work demonstrates ES can promote anaerobic bioremediation of PHE and nitrate in groundwater and, thus, opens up a new bioelectrochemistry application scenario.
Simultaneous degradation of n-hexane and production of biosurfactants by Pseudomonas sp. strain NEE2 isolated from oil-contaminated soils.

Abstract:The presence of surfactants in biofilters could enhance hydrophobic VOC removal. In this study, blood agar plate, methylene blue agar plate and a culture with n-hexane as the only carbon source were used to screen strains that could biodegrade n-hexane and produce biosurfactants simultaneously. The effects of n-hexane concentration on n-hexane removal and biosurfactant production were also investigated. Results showed that such a strain identified to be Pseudomonas sp. Strain NEE2 was successfully isolated from oil-polluted soils. The biosurfactants production by this strain were dependent on the initial concentration of n-hexane (132-2640 mg/L). At the concentration of 2640 mg/L of n-hexane, the biosurfactants promoted n-hexane removal. At 132 mg/L of n-hexane, n-hexane removal efficiency on day 2 exceeded 60%. The synergistic mechanisms of n-hexane removal and biosurfactant production by Pseudomonas sp. Strain NEE2 were discussed including the enhanced mass transfer from gas to liquid phase, within the biofilm phase and biodegradation at the presence of biosurfactants as well as the consequently enhanced production of the biosurfactants. These results in this study proved that it is possible for microorganisms utilizing the synergistic effect of hydrophobic VOC degradation and biosurfactant production for cost-effective hydrophobic VOC removal in biofilters. Abstract:The topoisomerase II (TOP2)-associated DNA cleavage activity and the DNA sequence preference of 20 antitumor drugs, including 15 TOP2-interacting compounds, have been defined. Four major classes of drugs have been identified: (i) those which enhanced the stabilization of cleavable complexes at a single major site (e.g. amsacrine, doxorubicin), or (ii) at many sites (e.g. etoposide, azatoxin), with chemically related compounds having very similar, although not identical, cleavage patterns (e.g. etoposide, GL331 and Top-53); (iii) those which inhibited DNA breakage (e.g. aclarubicin, actinomycin D); and (iv) those which did not visibly interfere with TOP2-mediated cleavable complexes (e.g. ICRF-187, camptothecin). All drugs tested induced similar overall patterns of sites of preferred DNA cleavage, in the presence either of the two known isoforms, TOP2alpha or TOP2beta, although relative intensities of signals at each position varied. It has been further shown that etoposide and its derivatives blocked the religation step downstream of the DNA cleavage step, whereas amsacrine, ellipticine, azatoxin and genistein acted upstream through enhancement of DNA cleavage. The information provided by this mechanistically based comparison can now be exploited in designing or synthesizing novel TOP2-interacting agents.
Simultaneous Removal of Nitrate, Nitrobenzene and Aniline from Groundwater in a Vertical Baffled Biofilm Reactor

Abstract:The challenge of simultaneous removal of nitrobenzene (NB), aniline (AN) and nitrate from groundwater in a single bioreactor is mainly attributed to the persistence of AN to degradation with anoxic denitrification con-ditions. In this work, simultaneous removal of NB (100 mu M), AN (100 mu M) and nitrate (1 mM) was achieved within 8 h with a COD/N ratio of 8 in a vertical baffled biofilm reactor (VBBR). By setting DO concentration at 0.4-0.5 mg L-1 to create a micro-aerobic condition, NB removal rate was accelerated without accumulation of AN, and AN could serve as electron donors for denitrification after ring cleavage. High-throughput sequencing showed that biofilm was predominated by denitrifiers (Luteimonas, Planctomyces, Thiobacillus, Thauera and so on) and NB-degrading bacteria (Pseudomonas), and biodiversity varied vertically along the height of the reactor. A dominantly anaerobic pathway for reducing NB to AN was identified by PICRUSt analysis, as the predicted genes involved in aerobic transformation of NB were several magnitudes lower than those in the anaerobic pathway. This study provided a new insight to the role of oxygen in robust bioremediation groundwater contaminated with NB, AN and nitrate.

Abstract:Biological denitrification from simulated contaminated groundwater using ethanol as an external carbon source was investigated with an entrapped mixed microbial cell (EMMC) system. Ethanol was used as an external carbon source to serve both as the energy source for the nitrate removal reaction and substrate for microorganisms in the system. The hydraulic retention time and chemical oxygen demand (COD)/nitrogen ratio significantly affect nitrate removal performance and, therefore, must be monitored carefully to achieve high rates of nitrate and COD removal. The effect of packing ratios of carriers in the reactor on nitrate removal performance was also investigated. The removal of nitrate and COD in reactors with various carrier packing ratios (12, 22, and 35%) is not significantly different during steady‐state operation. However, the start‐up periods in these three reactors are significantly different. The highest packing ratio can result in a fast removal of nitrate during the start‐up periods. Results of solids retention time (SRT) analysis indicate that a long SRT in the EMMC system contributes to the stable removal of nitrate and to fast start up of the system. The increase of SRT is related to the increase in days of operation, SRT is maintained at a level of more than 200 days.

Abstract:The aim of this study was to investigate the effect of adsorption by a composite active medium (nitrogen-degrading bacteria immobilized on scoria) on the removal of ammonium nitrogen (NH4+-N), nitrite nitrogen (NO2–N) and nitrate nitrogen (NO3–N). Transport of these three forms of nitrogen was investigated using columns with different flow rates, initial concentrations and depths. Immobilization of the bacteria on the scoria significantly enhanced the ability of the composite active medium to remove nitrogen during the remediation process. As the flow rate increased, the shear force increased and solute diffusion decreased, thereby improving the tolerance of the bacteria for the three forms of nitrogen and reducing the penetration time. Increasing the initial nitrogen concentration resulted in a decrease in the adsorption rate constant (KTh). The nitrogen-degrading bacteria immobilized on scoria rapidly reached saturation with regards to their nitrogen adsorption capacity. The initial nitrogen concentration of each media layer was different, and the composite active medium had a certain blocking effect, which resulted in a slow increase in the concentration of nitrogen in the deeper media layer. The transport experiments showed that the process accords with the thomas model and the Bohart-Adams model. The contents of the three forms of nitrogen were within acceptable standards for drinking water after 2 months of a column containing the composite active medium. The composite active medium can be used for in situ and ex situ remediation of groundwater containing excessive nitrogen. High-throughput sequencing analysis was used to monitor the composition of the bacterial community present within the composite active medium. During the remediation process, there were only slight changes in the structure and composition of the nitrogen-degrading bacterial community, although there were clear differences in abundance. Pseudomonas, Stenotrophomonas and Serratia were the three bacterial genera that were effective removal of all three forms of nitrogen.

Abstract:Emulsified nano-zero-valent iron (EZVI) is a modified form of bare nanoiron with improved transportability and targetability for the remediation of organic-solvents polluted soil and groundwater. In this work, EZVI (50–150 nm) was prepared by coating an emulsified vegetable oil membrane on the surface of Fe nanoparticles. EZVI was well-dispersed and less aggregation was observed. Batch experiments were conducted in anaerobic conditions to investigate the kinetics of nitrobenzene reduction by EZVI and the influences of oil concentration, initial iron content, and initial pH. Results indicated that the kinetics of nitrobenzene reduction by EZVI followed a pseudo-first-order kinetics. The observed rate constant of nitrobenzene is 0.0942 min−1. The oil concentration of 1 and 2 % tended to be preferred concentrations. The rate of nitrobenzene degradation and aniline formation increased with increasing iron content. The low pH is favorable to the nitrobenzene reduction by EZVI.

[Development and Evaluation of a Sustainable Long-release Carbon Material Applied for In-Situ Remediation of Groundwater Nitrogen Pollution].

Abstract:Due to slow flow rates and inter-substance reactions in groundwater, remediation requires the addition of materials with sustained release properties. This research uses agricultural waste and zero-valent iron (Fe0), coupling biology and chemistry, to research and develop a sustainable long-release carbon material with a synergistic physico-habitat, and to evaluate its performance, also taking into account the occurrence of nitrogen in groundwater. The material developed has a double-layer structure with an inner core and an outer shell. The core, consisting of agricultural waste, Fe0, and other raw materials, constitutes the repair layer. Agricultural waste provides a carbon source for microorganisms, and Fe0 can quickly remove nitrate via chemical reactions and reduce DO to develop an anaerobic environment in water. The shell provides a solute permeation layer and consists of primary minerals and other components. This can slow the release of TOC from the core and adsorb secondary contaminants. Physical properties testing showed that the materials core was uniformly cross-linked, and its shell exhibited a clear uniform pore structure (SEM). Favorable mechanical compression was recorded for particle strengths of up to 80-105 N. With a density of 1.1 g·cm-3, the material did not float in water. Experiments showed that the material had excellent sustained release. The amount[Max:21-25 mg·(g·L)-1] and rate[Max:0.185 mg·(g·L·d)-1] of TOC release exhibited a steady state trend, but fluctuated greatly in the case of agricultural wastes[(Max amount:53-75 mg·(g·L)-1, Max rate:0.455 mg·(g·L·d)-1]. In terms of further functional gene abundance, materials leachate was found to be conducive to denitrifying bacteria. In early denitrification and oxygen trapping experiments, the Fe0 chemical reaction was dominant for reduction of nitrogen and DO, facilitating denitrification. However, biological denitrification gradually dominated. Differences in denitrification rates between iron-free and iron-containing materials were smaller, as was the correlation between denitrification rate and iron content. These results indicate the formation of physico-habitat synergistic denitrification in the materials system.

Rapid enrichment of AnAOB with a novel vermiculite/tourmaline modification technology for enhanced DEAMOX process

Abstract:The DEnitrifying AMmonium OXidation (DEAMOX) has been proven to be a promising process treating contaminated surface water containing ammonia and nitrate, while the enrichment of the slow-growing anammox bacteria (AnAOB) remains a challenge. In this study, a novel polyurethane-adhesion vermiculite/tourmaline (VTP) modified carrier was developed to achieve effective enrichment of AnAOB. The results demonstrated that the VTP-1 (vermiculite: tourmaline = 1:1) system exhibited the greatest performance with the total nitrogen removal efficiency reaching 87.6% and anammox contributing 63% to nitrogen removal. Scanning electron microscope analysis revealed the superior biofilm structure of the VTP-1 carrier, providing attachment for AnAOB. The addition of VTP-1 promoted the secretion of EPS (extracellular polymeric substances) by microorganisms, which increased to 85.34 mg/g VSS, contributing to the aggregation of anammox cells. The favorable substrate microenvironment created by NH4+ adsorption and NO2- supply via partial denitrification process facilitated the growth of AnAOB. The relative abundance of Candidatus Brocadia and Thauera increased from 0.04% to 0.3%-1.03% and 2.06% in the VTP-1 system, respectively. This study sheds new light on the anammox biofilm formation and provides a valid approach to initiate the DEAMOX process for low nitrogen polluted water treatment.
Surfactant-assisted biodegradation of nitrobenzene in groundwater by sulfided nano-zero valent iron activated persulfate: Synergistic effect, mechanism, and application

Abstract:Groundwater contamination by dense non-aqueous phase liquids (DNAPLs), particularly nitrobenzene, represents a significant environmental challenge due to their chemical stability, persistence, and low solubility. This study aims to develop a synergistic approach for the biodegradation of nitrobenzene in groundwater, leveraging a combined system of Tween 80-assisted solubilization, sulfidized nano-zero valent iron (S-nZVI), and persulfate (PS) activation. The reduction process is facilitated by S-nZVI, while PS activation generates strong oxidizing radicals, and Tween 80 enhances nitrobenzene solubilization, thus improving the overall treatment efficacy. Laboratory experiments, including both batch and column studies, were conducted to evaluate the performance of this approach. Furthermore, a persulfate slow-release gel system was introduced to provide sustained activation, leading to improved long-term degradation efficiency. The results demonstrated that the synergistic combination of S-nZVI, PS, and Tween 80 led to significantly enhanced degradation of nitrobenzene and its byproduct aniline, achieving degradation rates of up to 96.74% for nitrobenzene and 100% for aniline after 6 h under optimal conditions. Additionally, the oxidation rate of aniline reached 91.53% within 5 min when the optimal dosage of 1.2 mM PS was used. The Tween-80/S-nZVI/PS slow-release gel system further achieved cumulative degradation rates of 87.49% for nitrobenzene over 14 pore volumes in a column study, demonstrating its potential for practical applications in groundwater remediation. These findings provide a promising new direction for the remediation of contaminated groundwater.

Abstract:Large amounts of nitrogen fertilized to food production are lost via denitrification and leaching. Dissimilatory nitrate reduction to ammonium (DNRA) is a bioprocess competing with denitrification, and it conserves bioavailable nitrogen in soil ecosystems. The carbon to nitrogen (C/N) ratio has long been believed essential to balance the competition between DNRA and denitrification, but increasing evidence indicates that DNRA may occur unrelated to C/N ratio via a largely unknown mechanism. Taking Geobacter as representative bacteria, we found that the kinetic of nitrite bio-reduction to ammonium was the key to enhancing DNRA and demonstrated it in pure and mixed cultures and soil ecosystems. Compared to other two strains of DNRA bacteria, Geobacter sulfurreducens was confirmed for the first time to conduct a 3-fold more rapid DNRA (NO 2 − →NH 4 + ) when it grew with the kinetically matched nitrite accumulator Delftia tsuruhatensis (NO 3 − →NO 2 − ). They assisted and restricted each other to bring their population ratio close to the theoretical yield ratio of 2.7:1 while performing cross-feeding with nitrite and ammonium. This rapid DNRA was further demonstrated to conserve 2–8 times more reactive nitrogen in organic deficient soils, and up to 40% of 15 NO 3 − was reduced to ammonium. Our results found a sustainable way of nitrogen retention in soils, and have broader implications for understanding nitrogen turnover in ecosystems.

Sulfide-rich Sediments Remediation by Slow-Release Calcium Nitrate with Low Risk: Experiments in a Continuous Flow Reactor

Abstract:Nitrate-driven sulfide oxidation has been proven to be a cost-effective approach for controlling sediment odor. However, injecting liquid nitrate (LN) directly into sediment carries ecological risks of nitrate escape and release of pollutants into overlying water. In this study, slow-release nitrate granules (GN) were prepared and applied in a continuous flow reactor to assess the differences between GN and LN in sediment remediation and control of this ecological risk. The results showed that the GN slowly released calcium nitrate, resulting in a decrease in acid volatile sulfide (AVS) from an initial 1328 ± 42.7mg/kg to 490 ± 341 and 212 ± 32.8mg/kg in the upper (U) and deeper (D) layers of the sediment, respectively. No significant decrease in AVS concentrations was observed in the LN treatments. Compared with LN-D and -U, the NH4+-N production and PO43--P release doses were reduced by 29.5% and 52.0% for GN-D and 41.2% and 37.8% for GN-U, respectively. Taxonomic analysis showed that sulfide-driven denitrifiers such as Thiobacillus, Rhodanobacter, Sulfurimonas, and Pseudomonas became dominant in the GN treatments, which were 2.8-6.7, 27.0-201.8, 1.5-3.0, and 3.0-496 times that of the LN treatments, respectively. The abundance of sulfur oxidation genes (e.g., sat and aprB) and denitrifying genes (e.g., nirS and nosZ) were also upregulated to higher levels by GN. Moreover, the genes (e.g., nrfA) responsible for the isomerization of nitrate into ammonium were down-regulated by GN. The results showed that the slow-released calcium nitrate should be a promising way for in-situ remediation of sulfide-rich sediment.
Comparison of different ecological remediation methods for removing nitrate and ammonium in Qinshui River, Gonghu Bay, Taihu Lake

Abstract:Ecological remediation is one of the most practical methods for removing nutrients from river ecosystems. In this study, transformation and fate of nitrate and ammonium among four different ecological restoration treatments were investigated by stable 15 N isotope pairing technique combined with quantitative polymerase chain reaction and high-throughput sequencing technology. The results of 15 N mass-balance model showed that there were three ways to the fate of nitrogen: precipitated in the sediment, absorbed by Elodea nuttallii ( E. nuttallii ), and consumed by microbial processes (denitrification and anaerobic ammonium oxidation (anammox)). The results shown that the storage of 15 NH 4 + in sediments was about 1.5 times as much as that of 15 NO 3 − . And much more 15 NH 4 + was assimilated by E. nuttallii , about 2 times as much as 15 NO 3 − . Contrarily, the rate of microbial consuming 15 NO 3 − was higher than converting 15 NH 4 + . As for the group with 15 NO 3 − added, 29.61, 45.26, 30.66, and 51.95 % were accounted for 15 N-labeled gas emission. The proportions of 15 NH 4 + loss as 15 N-labeled gas were 16.06, 28.86, 16.93, and 33.09 % in four different treatments, respectively. Denitrification and anammox were the bacterial primary processes in N 2 and N 2 O production. The abundances of denitrifying and anammox functional genes were relatively higher in the treatment with E. nuttallii -immobilized nitrogen cycling bacteria (E-INCB) assemblage technology applied. Besides, microbial diversity increased in the treatment with E. nuttallii and INCB added. The 15 NO 3 − removal rates were 35.27, 49.42, 50.02, and 65.46 % in four different treatments. And the removal rates of 15 NH 4 + were 24, 34.38, 48.84, and 57.74 % in treatments A, B, C, and D, respectively. The results indicated that E-INCB assemblage technology could significantly promote the nitrogen cycling and improve nitrogen removal efficiency.
Dynamic Succession of Groundwater Functional Microbial Communities in Response to Emulsified Vegetable Oil Amendment during Sustained In Situ U(VI) Reduction.

Abstract:A pilot-scale field experiment demonstrated that a one-time amendment of emulsified vegetable oil (EVO) reduced groundwater U(VI) concentrations for 1 year in a fast-flowing aquifer. However, little is known about how EVO amendment stimulates the functional gene composition, structure, and dynamics of groundwater microbial communities toward prolonged U(VI) reduction. In this study, we hypothesized that EVO amendment would shift the functional gene composition and structure of groundwater microbial communities and stimulate key functional genes/groups involved in EVO biodegradation and reduction of electron acceptors in the aquifer. To test these hypotheses, groundwater microbial communities after EVO amendment were analyzed using a comprehensive functional gene microarray. Our results showed that EVO amendment stimulated sequential shifts in the functional composition and structure of groundwater microbial communities. Particularly, the relative abundance of key functional genes/groups involved in EVO biodegradation and the reduction of NO3-, Mn(IV), Fe(III), U(VI), and SO42- significantly increased, especially during the active U(VI) reduction period. The relative abundance for some of these key functional genes/groups remained elevated over 9 months. Montel tests suggested that the dynamics in the abundance, composition, and structure of these key functional genes/groups were significantly correlated with groundwater concentrations of acetate, NO3-, Mn(II), Fe(II), U(VI), and SO42-. Our results suggest that EVO amendment stimulated dynamic succession of key functional microbial communities. This study improves our understanding of the composition, structure, and function changes needed for groundwater microbial communities to sustain a long-term U(VI) reduction.

Abstract:Excessive nitrate is a critical concern for a long-term greenhouse production system and potentially pollutes groundwater through leaching. Bacillus megaterium NCT-2 can be used to remove excessive nitrate. Enrichments were made from salinized soil, with and without strain NCT-2, to examine the influence of Bacillus megaterium NCT-2 on the nitrate metabolic potential and microbial composition under various carbon sources using detection of nitrogen content, high-throughput sequencing and quantitative real-time PCR. The results demonstrated that strain NCT-2 enhanced the ability of enriched microorganisms to convert nitrate to ammonium with some but not all carbon sources. The microbial composition and their functional potential had a significant relationship with the strain NCT-2 and carbon sources in the concentrates. Specifically, the enrichments with strain NCT-2 significantly increased the relative abundance of Bacillus and Pseudomonas genera, while decreased the relative abundance of Halomonas genus compared to enrichments without NCT-2 strain. Furthermore, in terms of carbon sources, D-cellobiose promoted the enrichment of Enterobacter genus, methyl-glucoside favored the enrichments of Photobacterium or Serratia genera, and D-mannitol favored the enrichments of Photobacterium, Serratia, or Halomonas genera. In , the application of Bacillus megaterium NCT-2 will increase the potential of microorganisms to remove nitrate from the environment. Moreover, the current study demonstrates that different carbon sources can also increase the potential for excess nitrate removal by changing the microbial community composition.

Abstract:Excessive nitrate in waterbody, which usually leads to eutrophication, can be removed by microbial denitrification processes. However, denitrification rate is greatly constrained by the spatial separation of nitrate in the upper oxic water layer from the electron donors close to the anoxic sediment. Herein, we constructed a genetically engineered bacterium (GEB), by co-expressing MO2.1 from Moringa oleifera and FLO5 from Saccharomyces cerevisiae, to simultaneously adsorb and self-flocculate nitrate from water. The GEB could migrate 45.5% of the nitrate to the bottom layer in actual water. Expressed FLO5 is electrically neutral and could trigger bacterial flocculation under the mediation of extracellular polysaccharides. Certain cationic arginine sites in MO2.1 can form strong electrostatic adsorption with NO3-, achieving bacterial self-flocculation, sedimentation and nitrate removal. The construction of multifunctional GEB could realize waterbody nitrate migration and is important to studies on new functional bacteria, water purification technology as well as geochemical nitrogen cycle.

Abstract:The extensive usage of ammonium sulfate as the leaching agent to extract rare earth elements led to widespread ammonia nitrogen (NH4+-N) pollution in the tailing soils of ion-adsorbed rare earth deposits in southern China. However, the cost-effective technologies to tackle with the long-termretention of NH4+-N in the rare earth mining soil have been largely unresolved. In this study, we developed a cost-effective approach to activate soil nitrification by the co-application of alkaline materials and organic fertilizer. The co-application of 0.3 % of organic fertilizer and 0.1 % similar to 0.2 % of CaO or MgO or Mg(OH)(2) stimulated a soil NH4+-N decrease rate of 2.01-7.58 mg kg(-1) d(-1) and a soil NO3--N accumulation rate of 1.56-7.09 mg kg(-1) d(-1). Noting that only if the soil pH was elevated to 7.81-9.00, the NH4+-N decrease rate and NO3--N accumulation rate were dependent on the proton consumption capacity of the alkaline materials. The application of CaCO3 could not stimulate soil nitrification possibly due to the soil pH was uncapable to be elevated to above 7.68. The qPCR, amplicon sequencing, and nitrification inhibitor batch incubation results demonstrated that organic fertilizer supplied active ammonia-oxidizing bacteria Nitrosomonas europaea. The proliferation of Nitrosomonas europaea in the alkaline materials and organic fertilizer co-applied soil was responsible for the soil nitrification. Furthermore, the application of commercial denitrifying bacteria inoculum promoted the removal of accumulated NO3--N. The findings of this study provide a lost-cost technology to remove NH4+-N from the rare earth mining soil.

Abstract:Nanotechnology and biotechnology offer promising avenues for bolstering food security through the facilitation of soil nitrogen (N) sequestration and the reduction of nitrate leaching. Nonetheless, a comprehensive and mechanistic evaluation of their effectiveness and safety remains unclear. In this study, a soil remediation strategy employing nano-Fe 3 O 4 and straw in N-contaminated soil was developed to elucidate N retention mechanisms via diverse metagenomics techniques. The findings revealed that subsoil amended with straw, particularly in conjunction with nano-Fe 3 O 4 , significantly increased subsoil N content (53.2%) and decreased nitrate concentration (74.6%) in leachate. Furthermore, the enrichment of functional genes associated with N-cycling, sulfate, nitrate, and iron uptake, along with chemotaxis, and responses to environmental stimuli or microbial collaboration, effectively mitigates nitrate leaching while enhancing soil N sequestration. This study introduces a pioneering approach utilizing nanomaterials in soil remediation, thereby offering the potential for the cultivation of safe vegetables in high N input greenhouse agriculture.

Abstract:Azo dyes wastewater contains refractory pollutant and nitrogen, which threatens human health and ecological environment when discharged into environment directly. Electron shuttle (ES) is able to participate in the extracellular electron transfer, and thus enhances the removal efficiency of refractory pollutant. However, the continuous dosing of soluble ES would rise operation cost and cause contamination inevitably. In this study, a type of insoluble ES (carbonylated graphene oxide (C-GO)) was developed and melt blended into polyethylene (PE) to prepare novel C-GO-modified suspended carriers. Compared to those of conventional carrier (31.60%), the surface active sites of novel C-GO-modified carrier increased to 52.95%. An integrated hydrolysis/acidification (HA, filled with C-GO-modified carrier) - anoxic/aerobic (AO, filled with clinoptilolite-modified carrier) process was applied to remove azo dye acid red B (ARB) and nitrogen simultaneously. ARB removal efficiency was significantly improved in the reactor filled with C-GO-modified carriers (HA2) compared to the reactor filled with conventional PE carriers (HA1) or activated sludge (HA0). Total nitrogen (TN) removal efficiency of the proposed process increased by 25.95-32.64% compared to the reactor filled with activated sludge. Moreover, the intermediates of ARB were identified by liquid chromatograph-mass spectrometer (LC-MS), and the degradation pathway of ARB through ES was proposed. C-GO-modified carriers induced ARB-removal-related bacterial enrichment (such as Chloroflexi, Lactivibrio, Longilinea, Bacteroidales and Anaerolineaceae). Besides, the relative abundance of denitrifiers and nitrifiers in the AO reactor filled with clinoptilolite-modified carrier was increased by 11.60% compared with activated sludge. Copy numbers of genes related to membrane transport, carbon/energy metabolism and nitrogen metabolism increased significantly on the surface-modified carriers. This study proposed an efficient approach for simultaneous azo dyes and nitrogen removal, showing potential in actual application.
