Abstract:As a non-invasive cancer therapy method, photodynamic therapy (PDT) shows tremendous promise in clinical cancer treatment. Light-activated singlet oxygen production of photosensitizers (PSs) is the prerequisite for cancer PDT, and the use of organic photosensitizers is always limited by visible light-based activation, hydrophilicity, biocompatibility, selectivity and quantum yield of singlet oxygen. Currently, both zinc porphyrin- and BODIPY-based structures have been widely used in the development of PDT PSs. Here, we developed a novel naphthalimide bridged zinc porphyrin/BODIPY molecule (Por-BDP-1) with two poly(ethylene glycol) (PEG) chains, in which D-A structure was constructed between the naphthalimide group and porphyrin group. After self-assembly into nanoparticles, Por-BDP-1 NPs (Diameter: 122.4 nm) could quench fluorescence in 600-700 nm, bind with calf thymus-DNA, and produce singlet oxygen during light-irradiation (laser: 680 nm, 1.0 W/cm2). In addition, Por-BDP-1 NPs effectively killed HeLa cells with a IC50 value = 44.8 mu g/mL and showed a lower dark toxicity under the same conditions. All our results demonstrated that our naphthalimide bridged zinc porphyrin/BODIPY nano-photosensitizer is a promising nanoagent for PDT in the clinic.

Abstract:Antimicrobial photodynamic therapy has considerable promise in the fight against bacterial infections. The superior photophysical characteristics of porphyrins have made them effective photosensitizers in the field of phototherapy. Herein, the light-induced antimicrobial effects of three porphyrins with different substituents have been compared. 5,10,15,20-tetrakis(4-hydroxyphenyl) porphyrin (THPP) shows superior photosensitizing activity and antimicrobial ability under irradiation with green light. THPP can also inhibit and destroy mature Staphylococcus aureus biofilms under irradiation. This work provides a reference for the rational design of photosensitizers for application in antimicrobial photodynamic therapy.

Abstract:The threat posed by antibiotic-resistant bacteria and the challenge of biofilm formation has highlighted the inadequacies of conventional antibacterial therapies, leading to increased interest in antibacterial photodynamic therapy (aPDT) in recent years. This approach offers advantages such as minimal invasiveness, low systemic toxicity, and notable effectiveness against drug-resistant bacterial strains. Porphyrins and their derivatives, known for their high molar extinction coefficients and singlet oxygen quantum yields, have emerged as crucial photosensitizers in aPDT. However, their practical application is hindered by challenges such as poor water solubility and aggregation-induced quenching. To address these limitations, extensive research has focused on the development of porphyrin-based nanomaterials for aPDT, enhancing the efficacy of photodynamic sterilization and broadening the range of antimicrobial activity. This review provides an overview of various porphyrin-based nanomaterials utilized in aPDT and biofilm eradication in recent years, including porphyrin-loaded inorganic nanoparticles, porphyrin-based polymer assemblies, supramolecular assemblies, metal-organic frameworks (MOFs), and covalent organic frameworks (COFs). Additionally, insights into the prospects of aPDT is offered, highlighting its potential for practical implementation.
Amphiphilic Tetracationic Porphyrins Are Exceptionally Active Antimicrobial Photosensitizers: in Vitro and in Vivo Studies with the Free-Base and Pd-chelate

Abstract:Antimicrobial photodynamic inactivation (aPDI) employs the combination of nontoxic photosensitizing dyes and visible light to kill pathogenic microorganisms regardless of drug-resistance, and can be used to treat localized infections. A meso-substituted tetra-methylpyridinium porphyrin with one methyl group replaced by a C12 alkyl chain (FS111) and its Pd-derivative (FS111-Pd) were synthesized and tested as broad-spectrum antimicrobial photosensitizers when excited by blue light (5 or 10 J/cm(2)). Both compounds showed unprecedented activity, with the superior FS111-Pd giving 3 logs of killing at 1 nM, and eradication at 10 nM for Gram-positive methicillin-resistant Staphylococcus aureus. For the Gram-negative Escherichia coli, both compounds produced eradication at 100 nM, while against the fungal yeast Candida albicans, both compounds produced eradication at 500 nM. Both compounds could be categorized as generators of singlet oxygen (phi(Delta) = 0.62 for FS111 and 0.71 for FS111-Pd). An in vivo study was carried out using a mouse model of localized infection in a partial thickness skin abrasion caused by bioluminescent Gram-negative uropathogenic E. coli. Both compounds were effective in reducing bioluminescent signal in a dose-dependent manner when excited by blue light (405 nm), but aPDI with FS111-Pd was somewhat superior both during light and in preventing recurrence during the 6 days following PDT.

Abstract:Bacterial infection poses a significant threat to human health, and the emergence of antibiotic-resistant strains has exacerbated the situation. Antimicrobial photodynamic therapy (aPDT) has emerged as a promising antibiotic-free treatment option that employs reactive oxygen species (ROS) to cause oxidative damage to bacteria and surrounding biomolecules for treating microbial infections. This review summarizes the recent progress in the development of organic photosensitizers, including porphyrins, chlorophyll, phenothiazines, xanthenes and aggregation-induced emission photosensitizers, for aPDT. A detailed description of innovative therapeutic strategies that rely on the infection microenvironment or the unique structural properties of bacteria to amplify the therapeutic effects is provided. Moreover, the combination of aPDT with other therapy strategies such as antimicrobial peptide therapy, photothermal therapy (PTT) or gas therapy, is described. Finally, the current challenges and perspectives of organic photosensitizers for clinical antibacterial applications are discussed.
Metalloporphyrin–indomethacin conjugates as new photosensitizers for photodynamic therapy

Abstract:Photodynamic therapy (PDT) is a promising cancer treatment approach with the advantages of low toxicity and noninvasive characteristics. In this study, a series of metalloporphyrin–indomethacin conjugates tethered with poly(ethylene glycol) (PEG) chains were prepared and characterized. The singlet oxygen production of the conjugates was evaluated through 2′, 7′-dichlorofluorescin (DCFH) method. Because of the heavy atom effect, the metal porphyrin complexes exhibited the higher singlet oxygen ( 1 O 2 ) quantum yield than that of free base porphyrin. The order of 1 O 2 yield of the synthesized porphyrins was PtPor > PdPor > ZnPor > Por. The MTT assay using HeLa cells verified the low cytotoxicity of porphyrin–indomethacin conjugates in the dark. Upon irradiation, the platinated porphyrin (PtPor) showed the highest therapeutic activity among these conjugates, probably due to its high efficiency of 1 O 2 generation. The cellular uptake and subcellular localization of the conjugates were further evaluated through a confocal laser scanning microscope. The results showed that the conjugates were primarily localized in the lysosomes of HeLa cells.

Abstract:In recent years cancer has become a global problem and burden to the health system. Photodynamic therapy (PDT) and sonodynamic therapy (SDT) are promising alternatives for cancer treatment. It is very important to use phthalocyanines as sensitizers in such treatments. In this study, a new type of Zn-phthalocyanine compound ( 2 ) peripherally substituted with 4-Hydroxy-7-methoxyquinoline was synthesized and characterized, and its photodynamic and sono-photodynamic efficiencies were examined. Additionally, the electronic and spectroscopic properties of the compounds were calculated theoretically using Density Functional Theory (DFT) and Time-Dependent Density Functional Theory (TD-DFT) methods. The effect of the solvent on the reactivity of the compounds was examined by the Conductive-Like Polarizable Continuum Model (CPCM). The results have indicated that singlet oxygen quantum yield has increased in the sono-photochemical study (Φ Δ =0.98) compare to the photodynamic study (Φ Δ =0.70). Experimental and theoretical study results show that the reactivity of the new ligand increases with Zn metal.

Rational Design of Self-Assembled Cationic Porphyrin-Based Nanoparticles for Efficient Photodynamic Inactivation of Bacteria

Abstract:Bacterial infection has become an urgent health problem in the world. Especially, the evolving resistance of bacteria to antibiotics makes the issue more challenging, and thus new treatments to fight these infections are needed. Antibacterial photodynamic therapy (aPDT) is recognized as a novel and promising method to inactivate a wide range of bacteria with few possibilities to develop drug resistance. However, the photosensitizers (PSs) are not effective against Gram-negative bacteria in many cases. Herein, we use conjugated meso-tetra(4-carboxyphenyl)porphine (TCPP) and triaminoguanidinium chloride (TG) to construct self-assembled cationic TCPP-TG nanoparticles (NPs) for efficient bacterial inactivation under visible light illumination. The TCPP-TG NPs can rapidly adhere to both Gram-negative and Gram-positive bacteria and display promoted singlet oxygen (1O2) generation compared with TCPP under light irradiation. The high local positive charge density of TCPP-TG NPs facilitates the interaction between the NPs and bacteria. Consequently, the TCPP-TG NPs produce an elevated concentration of local 1O2 under light irradiation, resulting in an extraordinarily high antibacterial efficiency (99.9999% inactivation of the representative bacteria within 4 min). Furthermore, the TCPP-TG NPs show excellent water dispersity and stability during 4 months of storage. Abstract:Bacterial infections pose a significant threat to human health and entail substantial economic losses. Due to the broad-spectrum antibacterial effect and low susceptibility to drug resistance, photodynamic therapy (PDT), a nontraditional antibacterial approach, has garnered a lot of attention. In PDT, the selection of photosensitizer (PS) is crucial because it directly affects the efficiency and safety of the treatment. As a versatile fluorophore, the advantages of 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) used as a PS for antibacterial PDT are mainly reflected in its high quantum yield of singlet oxygen, easy modification, and exceptional photostability. Through strategic chemical modifications of the BODIPY structures, it is possible to enhance their photodynamic antibacterial activity and refine their selectivity for bacterial killing. This review focuses on the application of BODIPY-based PSs for treating bacterial infections. According to the design strategies of photodynamic antibacterial materials incorporating BODIPY, a variety of representative therapeutic agents having emerged in recent years are classified and discussed, aiming to offer insights for future research and development in this field.
Porphyrin photosensitizer molecules as effective medicine candidates for photodynamic therapy: electronic structure information aided design

Abstract:Traditional photosensitizers (PS) in photodynamic therapy (PDT) have restricted tissue penetrability of light and a lack of selectivity for tumor cells, which diminishes the efficiency of PDT. Our aim is to effectively screen porphyrin-based PS medication through computational simulations of large-scale design and screening of PDT candidates via a precise description of the state of the light-stimulated PS molecule. Perylene-diimide (PDI) shows an absorption band in the near-infrared region (NIR) and a great photostability. Meanwhile, the insertion of metal can enhance tumor targeting. Therefore, on the basis of the original porphyrin PS segments, a series of metalloporphyrin combined with PDI and additional allosteric Zn-porphyrin-PDI systems were designed and investigated. Geometrical structures, frontier molecular orbitals, ultraviolet-visible (UV-vis) absorption spectra, adiabatic electron affinities (AEA), especially the triplet excited states and spin–orbit coupling matrix elements (SOCME) of these expanded D–A porphyrin were studied in detail using the density functional theory (DFT) and time-dependent density functional theory (TDDFT) methods. PS candidates, conforming type I or II mechanism for PDT, have been researched carefully by molecular docking which targeted Factor-related apoptosis (Fas)/Fas ligand (Fasl) mediated signaling pathway. It was found that porphyrin-PDI, Fe 2 -porphyrin-PDI, Zn-porphyrin-PDI, Mg-porphyrin-PDI, Zn-porphyrin combined with PDI through single bond (compound 1 ), and two acetylenic bonds (compound 2 ) in this work would be proposed as potential PS candidates for PDT process. This study was expected to provide PS candidates for the development of novel medicines in PDT.

Abstract:Nowadays, infectious diseases persist as a global crisis by causing significant destruction to public health and the economic stability of countries worldwide. Especially bacterial infections remain a most severe concern due to the prevalence and emergence of multi-drug resistance (MDR) and limitations with existing therapeutic options. Antibacterial photodynamic therapy (APDT) is a potential therapeutic modality that involves the systematic administration of photosensitizers (PSs), light, and molecular oxygen (O2) for coping with bacterial infections. Although the existing porphyrin and non-porphyrin PSs were effective in APDT, the poor solubility, limited efficacy against Gram-negative bacteria, and non-specific distribution hinder their clinical applications. Accordingly, to promote the efficiency of conventional PSs, various polymer-driven modification and functionalization strategies have been adopted to engineer multifunctional hybrid phototherapeutics. This review assesses recent advancements and state-of-the-art research in polymer-PSs hybrid materials developed for APDT applications. Further, the key research findings of the following aspects are considered in-depth with constructive discussions: i) PSs-integrated/functionalized polymeric composites through various molecular interactions; ii) PSs-deposited coatings on different substrates and devices to eliminate healthcare-associated infections; and iii) PSs-embedded films, scaffolds, and hydrogels for regenerative medicine applications.
Photodynamic studies and photoinactivation of Escherichia coli using meso-substituted cationic porphyrin derivatives with asymmetric charge distribution

Abstract:The photodynamic activities of novel asymmetrically meso-substituted cationic porphyrins, 5,10-di(4-methylphenyl)-15,20-di(4-trimethylammoniumphenyl)porphyrin iodide 1 and 5-(4-trifluorophenyl)-10,15,20-tris(4-trimethylammoniumphenyl)porphyrin iodide 2 and its metal complex with Pd(II) 3, have been investigated in both homogeneous medium bearing photooxidizable substrates and in vitro on a typical gram-negative bacterium Escherichia coli. The amphiphilic character of porphyrin 2 was increased by the presence of a high-lipophilic trifluoromethyl group and its photophysical properties changed by forming a complex with Pd(II). Absorption and fluorescence spectroscopic studies were compared in different media. Fluorescence quantum yields (phi(F)) of 0.16 for 1 in tetrahydrofuran and 0.08 for 2 in N, N-dimethylformamide (DMF) were calculated, whereas no significant emission was detected for Pd(II) porphyrin 3. The singlet molecular oxygen, O(2)((1)Delta(g)), production was evaluated using 9,10-dimethylanthracene in DMF yielding relative values of 1, 0.55 and 0.47 for porphyrins 3, 2 and 1, respectively. A faster decomposition of l-tryptophan was obtained using Pd(II) porphyrin 3 as sensitizer with respect to the free-base porphyrins 1 and 2. In biological medium, the behavior of cationic porphyrins 1-3 were compared with that of 5-(4-carboxyphenyl)-10,15,20-tris(4-methylphenyl)porphyrin 4, which was used as a noncationic sensitizer. These porphyrins are rapidly bound to E. coli cells in 5 min and the amount of cell-bound sensitizer is not appreciably changed incubating the cultures for longer times. The recovered porphyrin 2 after one washing step reaches a value of approximately 2.9 nmol/10(6) cells and this amount remains high even after three washes, indicating that this sensitizer is tightly bound to cells. Photosensitized inactivation of E. coli was analyzed using cells without and with one washing step. In both cases, a higher photoinactivation of cells was found for tricationic porphyrin 2 and 3, causing a approximately 5.5 log (99.999%) decrease of cell survival, when treated with 10 microM of sensitizer. Under these conditions, a lower effect was found for porphyrin 1 (approximately 4 log) whereas sensitizer 4 did not produce appreciable photodamage. The results were also confirmed by growth delay experiments. These studies show that the amphiphilic tricationic porphyrin 2 and 3 bearing a trifluoromethyl group can be a promising model for phototherapeutic agents with potential applications in inactivation of bacteria by photodynamic therapy.
Structural Optimization of BODIPY Photosensitizers for Enhanced Photodynamic Antibacterial Activities

Abstract:Enhancing the interactions between photosensitizers and bacteria is key to developing effective photodynamic antibacterial agents. However, the influence of different structures on the therapeutic effects has not been systematically investigated. Herein, 4 BODIPYs with distinct functional groups, including the phenylboronic acid (PBA) group and pyridine (Py) cations, were designed to explore their photodynamic antibacterial activities. The BODIPY with the PBA group (IBDPPe-PBA) exhibits potent activity against planktonic Staphylococcus aureus (S. aureus) upon illumination, while the BODIPY with Py cations (IBDPPy-Ph) or both the PBA group and Py cations (IBDPPy-PBA) can significantly minimize the growth of both S. aureus and Escherichia coli (E. coli). In particular, IBDPPy-Ph can not only eliminate the mature S. aureus biofilm and E. coli biofilm in vitro, but also promote the healing of the infected wound. Our work provides an alternative for reasonable design of photodynamic antibacterial materials.
Supramolecular Porphyrin Photosensitizers: Controllable Disguise and Photoinduced Activation of Antibacterial Behavior.

Abstract:A series of supramolecular photosensitizers were fabricated from porphyrin derivatives (Por) containing quaternary ammonium groups with cucurbit[7]uril (CB[7]) based on host-guest interactions. The antibacterial activity of Por in the dark could be turned off upon binding with CB[7], whereas the antibacterial activity under white-light illumination could be turned on. In addition, its antibacterial efficiency could be greatly enhanced by introducing metal ions. When Pd(II) was introduced into porphyrin, its antibacterial efficiency was enhanced from 40 to 100%. It should be noted that these small molecules showed little to no cytotoxicity toward mammalian cells even at concentrations higher than those under the antibacterial condition studied. This line of research will provide a strategy for germicides consisting of quaternary ammonium groups to fight against bacterial accumulation in the long term and holds huge potential for application in the real world.

Abstract:Although the idea of combining the light of photodynamic therapy (PDT) and the sound of sonodynamic therapy (SDT) and converting into a much more effective therapeutic treatment method, sonophotodynamic therapy (SPDT), is very wise and sensitizing agents such as phthalocyanines produce higher amounts of singlet oxygen ( 1 O 2 ) with this method, the usage of SPDT for boron-dipyrromethene ( BODIPY )-based theranostics has put forward by our group for the first time and no studies have been published by other groups in the literature so far. Therefore, as our continuing motivation in this direction, in the present study, a novel near-infrared ( NIR ) BODIPY fluorophore bearing thienyl-pyridine substituent at meso -position, iodines as heavy atoms at 2-,6- positions and distyryl segments at 3-,5- positions which enlarge π-conjugation of the molecule has been synthesized successfully. The structural characterizations of the target NIR BODIPY 2 has been accomplished by the conventional spectroscopic methods ( 1 H NMR, 13 C NMR, FT-IR (ATR), UV–Vis and HRMS/MS) and elemental analysis. In order to evaluate the singlet oxygen generation capacity of NIR BODIPY 2 , photochemical and sonophotochemical methods have been employed comparatively. The singlet oxygen quantum yield (Φ Δ ) of NIR BODIPY 2 was calculated 0.37 using by photochemical method, this value increased to 0.66 by sonophotochemical study. It can be said that these results will open new horizons for researchers in terms of understanding sonophotochemical measurements of BODIPY derivatives for future in vitro or in vivo studies.

Abstract:Antimicrobial photodynamic therapy (aPDT) has become a potent contender in the fight against microbial infections, especially in the context of the rising antibiotic resistance crisis. Recently, there has been significant interest in polyphenolic natural products as potential photosensitizers (PSs) in aPDT, given their unique chemical structures and inherent antimicrobial properties. Polyphenolic natural products, abundant and readily obtainable from natural sources, are generally regarded as safe and highly compatible with the human body. This comprehensive review focuses on the latest developments and future implications of using natural polyphenols as PSs in aPDT. Paramount polyphenolic compounds, including curcumin, hypericin, quercetin, hypocrellin, celastrol, riboflavin, resveratrol, gallic acid, and aloe emodin, are elaborated upon with respect to their structural characteristics, absorption properties, and antimicrobial effects. Furthermore, the aPDT mechanism, specifically its targeted action on microbial cells and biofilms, is also discussed. Polyphenolic natural products demonstrate immense potential as PSs in aPDT, representing a promising alternate approach to counteract antibiotic-resistant bacteria and biofilm-related infections.

Abstract:Photodynamic therapy (PDT) is a treatment method consisting of common combination of oxygen, light energy and a light absorbing molecule called a photosensitizer. In this work, four new compounds consisting of BODIPY precursors and BODIPY-cyclotriphosphazene derivatives were synthesized to investigate the PDT effects. The chemical structures of the compounds were characterized and then their photophysical properties were determined by spectroscopic techniques. The precursor BODIPYs and their cyclotriphosphazene derivatives exhibited similar properties such as strong absorption intensity, high photostability and low fluorescence profile in the NIR region. Additionally, the singlet oxygen production capacities of these compounds were determined using the photobleaching technique of 1,3-diphenylisobenzofuran (DPBF) under light illumination. By introducing iodine atoms into the molecule, which are responsible for the intersystem transition (ISC) enhancement, a more efficient singlet oxygen production was achieved in both the iodinated-BODIPY and its cyclotriphosphazene derivative. Anticancer activities of the precursor BODIPYs and their cyclotriphosphazene derivatives in the absence and presence of light illumination were evaluated on cancerous cell lines (PC3 and DU145) and non-tumorigenic prostate epithelial PNT1a cell The compounds triggered the death of cancer cell PC3 the more significantly in the presence of red light compared to the healthy cells (PNT1a).

Abstract:Traditional photosensitizers show limited singlet oxygen generation in hypoxic infection lesions, which greatly suppress their performance in antibacterial therapy. Meanwhile, there still is lack of feasible design strategy for developing hypoxia-overcoming photosensitizers agents. Herein, radical generation of π-conjugated small molecules is efficiently manipulated by an individual selenium (Se) substituent. With this strategy, the first proof-of-concept study of a Se-anchored oligo (thienyl ethynylene) (OT-Se) with high-performance superoxide radical (O2 •−) and hydroxyl radical (•OH) generation capability is present, and achieves efficient antibacterial activities towards the clinically extracted multidrug-resistant bacteria methicillin-resistant S. aureus (MRSA) and carbapenem-resistant E. coli (CREC) at sub-micromolar concentration under a low white light irradiation (30 mW/cm2). The water-dispersible OT-Se shows a good bacteria-anchoring capability, biocompatibility, and complete elimination of multidrug-resistant bacteria wound infection in vivo. This work offers a strategy to boost type-I photodynamic therapy (PDT) performance for efficient antibacterial treatments, advancing the development of antibacterial agents.
