Abstract:Ohmic heating may improve bioactive compounds and processing, ensuring food safety of beverages, liquid and pasty food, or liquid with solid pieces. Due to those traits, this study conducted a comparison between ohmic heating technology and conventional heating (CH), with a focus on assessing the impact of both methods on functional compounds (such as angiotensin-converting enzyme inhibition, α-amylase and α-glucosidase inhibition, and antioxidant activity) in both fresh and thawed raw sheep milk, which had been frozen for up to 3 mo. Different ohmic heating conditions were applied and compared to CH (3.33-8.33 V/cm vs. CH [73°C/15 s]). A total of 18 peptides with some functional activities were identified by MALDI-TOF mass spectrometry analysis. Ohmic heating samples presented the highest activities related to health, followed by CH and raw milk samples; antioxidant activity range was from 0.11% to 0.71%, antihypertensive activity ranged from 0.20% to 0.72%, and antidiabetic activity ranged from 0.21% to 0.79%. Of 51 volatile compounds detected, some were degraded by freezing, storing, and heating the sheep milk. This study showed for the first time that ohmic heating processing improved sheep milk bioactive peptides and preserved volatile compounds.

Abstract:Galactooligosaccharides (GOS) are functional oligosaccharides obtained enzymatically from lactose. The interest in GOS has increased due to health benefits, such as the strength of the immune system and the support of a healthy gut microbiome. This review contributes to the state of the art of the domain of GOS, building an integrated overview highlighting 4 key elements that the food industry should consider when developing healthier and cleaner foods: food technological applications of GOS, prebiotic health benefits, microbiome modulation according to GOS structure, and fundamentals on process elaboration of GOS. GOS are a good strategy for fat and sugar replacement, as well as for calorie reduction, nutritional enhancement, processing aid, and texture improvement. Moreover, GOS supplementation is a potential approach to the improvement of bowel and immune health, cancer, renal injury, obesity, depression and anxiety treatment, and the enhancement of mineral absorption, mainly via microbiome modulation. Nevertheless, this modulation depends on GOS concentration, chain length, type, and orientation of linkage, and those aspects are shaped during the processing of GOS. Thus, the product development process of foods should have a holistic consideration of previous factors when selecting GOS as ingredients to address the desired functionalities efficiently. Current literature has discrepancies considering the structural heterogenicity of available GOS sources. Abstract:Prebiotics are non-digestible ingredients that exert significant health-promoting effects on hosts. Galactooligosaccharides (GOS) have remarkable prebiotic effects and structural similarity to human milk oligosaccharides. They generally comprise two to eight sugar units, including galactose and glucose, which are synthesized from substrate lactose by microbial β-galactosidase. Enzyme sources from probiotics have received particular interest because of their safety and potential to synthesize specific structures that are particularly metabolized by intestinal probiotics. Owing to advancements in modern analytical techniques, many GOS structures have been identified, which vary in degree of polymerization, glycosidic linkage, and branch location. After intake, GOS adjust gut microbiota which produce short chain fatty acids, and exhibit excellent biological activities. They selectively stimulate the proliferation of probiotics, inhibit the growth and adhesion of pathogenic bacteria, alleviate gastrointestinal, neurological, metabolic and allergic diseases, modulate metabolites production, and adjust ion storage and absorption. Additionally, GOS are safe and stable, with high solubility and clean taste, and thus are widely used as food additives. GOS can improve the appearance, flavor, taste, texture, viscosity, rheological properties, shelf life, and health benefits of food products. This review systemically covers GOS synthesis, structure identifications, metabolism mechanisms, prebiotic bioactivities and wide applications, focusing on recent advances.

Free comment as a valuable approach to characterize and identify the drivers of liking of high-protein flavored milk drink submitted to ohmic heating

Abstract:Flavored milk drink is a popular dairy product traditionally processed by pasteurization, which is a safe and robust process. Still, it can imply a greater energy expenditure and a more significant sensorial alteration. Ohmic heating (OH) has been proposed as an alternative to dairy processing, including flavored milk drink. However, its impact on sensory characteristics needs to be evidenced. This study used Free Comment, an underexplored methodology in sensory studies, to characterize five samples of high-protein vanilla-flavored milk drink: PAST (conventional pasteurization 72 °C/15 s); OH6 (ohmic heating at 5.22 V/cm); OH8 (ohmic heating at 6.96 V/cm); OH10 (ohmic heating at 8.70 V/cm), and OH12 (ohmic heating at 10.43 V/cm). Free Comment raised similar descriptors to those found in studies that used more consolidated descriptive methods. The employed statistical approach allowed observation that pasteurization and OH treatment have different effects on the sensory profile of products, and the electrical field strength of OH also has a significant impact. PAST was slightly to moderately negatively associated with "acid taste," "fresh milk taste," "smoothness," "sweet taste," "vanilla flavor," "vanilla aroma," "viscous," and "white color." On the other hand, OH processing with more intense electric fields (OH10 and OH12) produced flavored milk drinks strongly associated with the "in natura" milk descriptors ("fresh milk aroma" and "fresh milk taste"). Furthermore, the products were characterized by the descriptors "homogeneous," "sweet aroma," "sweet taste," "vanilla aroma," "white color," "vanilla taste," and "smoothness." In parallel, less intense electric fields (OH6 and OH8) produced samples more associated with a bitter taste, viscosity, and lumps presence. Sweet taste and fresh milk taste were the drivers of liking. In conclusion, OH with more intense electric fields (OH10 and OH12) was promising in flavored milk drink processing. Furthermore, the free comment was a valuable approach to characterize and identify the drivers of liking of high-protein flavored milk drink submitted to OH.

Abstract:As a prebiotics supplemented in infant formula (IFs), galactooligosaccharide (GOS) also has many other biological activities, but the Maillard reaction characteristics are still unclear. This study investigated the Maillard reactivity of GOS and effects on advanced glycation end products (AGEs) formation during IFs processing. The results showed that both AGEs and HMF formation were temperature-dependent and reached the maximum at pH 9.0 in Maillard reaction system of GOS and Nα-acetyl-L-lysine. Acidic conditions accelerated HMF formmation, but protein cross-linking was more likely to occur in alkaline conditions. Degrees of polymerization (DP) of GOS had no significant effect on AGEs formation (except pyrraline), but the greater the DP, the higher the concentration of HMF and pyrraline. Besides, compared with arginine and casein, lysine and whey protein were more prone to Maillard reaction with GOS. GOS promoted AGEs formation in a dose-dependent manner during IFs processing. These results provide a reliable theoretical basis for application of GOS in IFs.

Abstract:Lactobacilli should resist stress environments in industry process and gastrointestinal tract before exerting their beneficial effects. To explore the possible stabilizers in probiotic products, prebiotic oligosaccharides were investigated. We investigated the effect of four selected oligosaccharides on the survival of probiotic Lactobacillus plantarum and L. acidophilus to simulated stress conditions. It was found that the tolerance of lactobacilli to simulated artificial gastrointestinal juice, heat treatment and phenol solution was obviously enhanced in fructo-oligosaccharides (FOS) and xylo-oligosaccharides (XOS) group. In addition, chito-oligosaccharides (COS), manno-oligosaccharides (MOS) and glucose also had positive effect compared with control group (without sugar). Prebiotic oligosaccharides, especially XOS and FOS added in medium have protection function to lactobacilli in stress environments. The protection function of oligosaccharides may correlate with the bacteria growth, which was stimulated by these oligosaccharides. Prebiotic oligosaccharides may be used as stabilizers in probiotic products.

Abstract:The present investigation was undertaken to develop highland barley milk fortified with functional endogenous β-glucan extracted from the bran. Apparent viscosity, storage stability, ΔBS, TSI, centrifugal sedimentation rate, particle size analysis, zeta potential, and confocal laser scanning microscopy were used to characterize the stability of highland barley milk. The results showed that the addition of β-glucan effectively improved the stability of highland barley milk by increasing the viscosity. The SI of the sample with β-glucan increased after 14 days of storage and the absolute value of zeta potential increased compared with the control one. The contact angle increased significantly from 23.6° to 34.5°. ΔBS showed that the supernatant separation was significantly alleviated. The span of particle size became narrow and the distribution was more uniform. CLSM showed that β-glucan changed the distribution of protein and starch, thus increasing the repulsion between particles to enhance stability. Moreover, the HB milk tended to be stable with an increasing additional amount. Combined with the sensory evaluation, the final concentration of 1.2% (w/v) was selected.

Abstract:We used the Halomonas sp. S62 β-galactosidase to synthesize galactooligosaccharides from lactose, to explore galactooligosaccharides synthesis efficiency. To improve the yield of galactooligosaccharides, the reaction conditions were optimized, and the orthogonal test was carried out by the reaction temperature, p H, enzyme dosage and substrate concentration. The optimal reaction conditions were as follows: reaction temperature 40 ℃, p H 7. 0, enzyme dosage 50 U/m L, substrate concentration 300 g/L.The maximal yield of galactooligosaccharides was (41. 91±0. 27) ％ at 6 h and the lactose consumption rate was (82. 47±0. 38) ％.Within 4-8 h, the yield of galactooligosaccharides was above 40％ and the lactose consumption rate was over80％.The yield of galactooligosaccharides was maintained above 40％ within 4-8 h of the reaction. The consumption rate of lactose was all above 80％. The utilization rate of lactose was increased while the high production of galactooligosaccharides was achieved, which was beneficial to the reduction of production cost, laying the foundation for the industrial application of low-temperature S62 β-galactosidase.
Use of whey permeate containing in situ synthesised galacto-oligosaccharides for the growth and preservation of Lactobacillus plantarum

Abstract:Galacto-oligosaccharides (GOS) are prebiotics that have a beneficial effect on human health by promoting the growth of probiotic bacteria in the gut. GOS are commonly produced from lactose in an enzymatic reaction catalysed by β-galactosidase, named transglycosylation. Lactose is the main constituent of whey permeate (WP), normally wasted output from the cheese industry. Therefore, the main goal of this work was to optimise the synthesis of GOS in WP using β-galatosidase from Aspergillus oryzaea. WP and whey permeate enzymatically treated (WP-GOS) were used as culture media of Lactobacillus plantarum 299v. Lb. plantarum 299v attained the stationary phase in approximately 16 h, reaching 3·6 and 4·1×108 CFU/ml in WP and WP-GOS, respectively. The in situ synthesised GOS were not consumed during growth. No significant differences were observed in the growth kinetics of microorganisms in both media. After fermentation, microorganisms were dehydrated by freeze-drying and spray-drying and stored. The recovery of microorganisms after fermentation, dehydration and storage at 4 °C for at least 120 d was above 108 CFU/g. These studies demonstrated that WP is an appropriate substrate for the synthesis of GOS and the obtained product is also adequate as culture medium of Lb. plantarum 299v. The coexistence of GOS and dehydrated viable probiotic microorganisms, prepared using an effluent as raw material, represents the main achievement of this work, with potential impact in the development of functional foods.
Optimization for Galactooligosaccharides Synthesis: A Potential Alternative for Gut Health and Immunity

Abstract:AIMS:Present study was conducted to optimize the processing parameters for galactooligosaccharides (GOS) synthesis from whey powder followed by exploring its prebiotic efficiency.MAIN METHODS:All factors (initial lactose concentration, pH, reaction time, temperature and enzyme to substrate ratio; E/S) were analyzed by single factor analysis and optimization for GOS yield was done following the orthogonal experimental design. For in vivo analysis, 60 mice were equally divided into four groups (normal control, NC; low, medium, and high dose of GOS, LG, MG and HG) and fed varying levels (0, 0.25, 0.5 and 1.0 g/kg bw per day) of GOS, for 30 days and sampling was done at the end of experiment regarding gut health, immunity, cecal microbiota and metabolites.KEY FINDINGS:Optimum yield of GOS (25.1%) was obtained at reaction time 25 min, temperature 50 °C, pH 4.5 and the enzyme to substrate ratio (E/S) of 20 U/g. In vivo experiment, shallower crypt and greater villus to crypt ratio (V/C) were found in the duodenum of LG treatment compared to NC mice (P < .05). The GOS promotes thymus development and improve immunity. Intervention with GOS increased the population of bifidobacterium and lactobacillus in MG and bifidobacterium in LG mice (P < .05), and was accompanied by decreased proliferation of desulfovibrio. Correlation analysis also supported the above findings.SIGNIFICANCE:This study optimized the processing parameters for GOS preparation and provided data encouraging to suggest that GOS can be a potential option to improve the gut health and immunity.

Abstract:The impact of prebiotics on the structural characteristics of yogurt is an important aspect of evaluating its functional properties. This study aimed to evaluate and compare the effects of several commonly used prebiotics, including fructooligosaccharide (FOS), galactooligosaccharide (GOS), inulin (INU), polydextrose (PDX), and xylooligosaccharide (XOS), on the gel properties of milk protein and the structural features, with respect to the texture and rheology, of stirred yogurt during and after fermentation. The results revealed that the supplementation of INU, PDX, and XOS was involved in the construction of protein networks during fermentation, promoting a viscous and more elastic gel structure, due to the enhanced protein–water interactions. This resulted in a significant increase (p < 0.05) in structural stability (higher critical strain (γc) and greater thixotropy), firmness, cohesiveness, and rheology (G′ and G′′) and a significant decrease (p < 0.05) in the loss of yogurt during centrifugation. Conversely, the supplementation of GOS and FOS did not appear to be involved in the construction of the protein network and barely affected the rheological properties of the gel during fermentation. However, a significant increase (p 0.05) in loss during centrifugation were still observed in the yogurt. These findings could be useful for a comprehensive assessment of the application potential of these prebiotics in yogurt, when combined with their respective prebiotic properties.

Lifetime of solder joint and delamination in flip chip assemblies

Abstract:The effects of underfill on thermomechanical behaviors of two types (B and D) of flip chip packages, with different bumping size and stand-off height were investigated under thermal cycling by both experiments and finite element simulation. The results show that the use of underfill encapsulant increases tremendously (/spl sim/20 times) the thermal fatigue lifetime of SnPb solder joint, and weakens the effects of stand-off height on the reliability, and changes the deformation mode of the package. It was found that the thermal fatigue crack occurs in the region with a maximum plastic strain range, and the Coffin-Manson type equation could then be used for both packages with and without underfill. The effects of material models of underfill, i.e. constant elasticity (EC) and temperature dependent elasticity (ET) as well as the viscoelasticity (VE), on the thermomechanical behaviors of flip chip package were also studied in the simulation. The VE model gives comparatively large plastic strain range, and big displacements in shear direction, as well as sequentially low lifetime of solder joints. The ET model gives the close results to VE model and could be used instead of VE in simulations for the purpose of simplicity. Underfill delamination analysis of flip chip on low-cost board is also presented. The delamination propagation rates at the interface between chip and underfill have been measured by using C-SAM inspection of flip chip assemblies under thermal cycle loading. The experimental measurement was done in four cases, that were type B with fine solder joint and fractured solder joint, and type D with fine and fractured solder joint. In the finite element simulations the strain energy release rates G and the phase angles /spl phi/ near the delamination crack tip were calculated for four measurement cases by employing the fracture mechanical method. The Paris half-empirical equations were determined from the delamination propagation rates measured and the energy release rates simulated. Meanwhile, the energy release rates G with a different delamination crack length were also simulated. The G /spl sim/ a curve representing a convex shape when the crack propagates and indicates that the delamination crack may be stable. After propagating for a certain length the crack will be arrested in the flip chip assembly.
A novel process for the production of high-purity galactooligosaccharides (GOS) using consortium of microbes

Abstract:Galactooligosaccharides (GOS) are nondigestible dietary fibers which have a beneficial effect on human health by promoting the growth of probiotic bacteria in the gut. In addition, other health benefits have been reported from oligosaccharides consumption such as stimulation of intestinal mobility, colon cancer prevention, mineral absorption as well as protection against certain pathogenic bacterial infections. The goal of this research was to develop an efficient biotransformation system using a consortium of microbes for the production of ≥85% pure GOS and reusing the cell biomass in repeated cycles of biotransformation. Production of GOS by lactose transgalactosylation using whole cells of Sporobolomyces singularis MTCC 5491 as a source of β-galactosidase and monosaccharides utilization by yeast isolate (NUTIDY007) were studied. For increasing the purity of GOS, growth and bioconversion parameters on the transgalactosylation by the whole cells were investigated. Further, continuous production of GOS was studied in a reactor with microfiltration membrane system. A maximum GOS purity of 42% was achieved using single culture of S. singularis. Under optimized conditions, single culture of S. singularis produced a maximum of 56% pure GOS. Addition of second culture to the reaction mixture for utilization of glucose significantly increased the GOS purity from 56% to ≥85%. The product consisted of tri- to penta-galactooligosaccharides. Trisaccharides were the main component of the reaction mixture. A maximum productivity of 10.9 g/L/hr was obtained under the optimum conditions.
Bioconversion of spent coffee grounds to prebiotic mannooligosaccharides – an example of biocatalysis in biorefinery

Abstract:Spent coffee ground (SCG), an agro-industrial waste, have a high content of polysaccharides such as mannan, making it ideal for utilisation for the production of nutraceutical oligosaccharides. Recently, there has been growing interest in the production of mannooligosaccharides (MOS) for health promotion in humans and animals. MOS are reported to exhibit various bioactive properties, including prebiotic and antioxidant activity. In this study, SCG was Vivinal pretreated using NaOH, characterized and hydrolysed using a Bacillus sp. derived endo -β-1,4-mannanase, Man26A, for MOS production. Structural analyses using Fourier-transform infrared spectroscopy (FT-IR) and thermogravimetric analysis (TGA) were conducted to assess the efficacy of the pretreatment. Lignin removal by the pretreatment from SCG was clearly shown by TGA. FT-IR, on the other hand, showed the presence of α-linked D -galactopyranoside (812 cm −1 ) and β-linked D -mannopyranoside residues (817 cm −1 ) in both SCG samples, signifying the presence of mannan. Hydrolysis of pretreated SCG by Man26A produced mannobiose and mannotriose as the main MOS products. The effect of simulated gastric conditions on the MOS was investigated and showed this product to be suitable for oral administration. Finally, the prebiotic effect of the MOS on the growth of selected beneficial bacteria was investigated in vitro ; showing that it enhanced Lactobacillus bulgaricus , Bacillus subtilis and Streptococcus thermophilus growth. These findings suggest that SCG is a viable source for the production of MOS which can be orally administered as prebiotics for effecting luxuriant growth of probiotic bacteria in the host's digestive tract, leading to a good health status.

This study investigated the fortification of sugar-free white chocolate with Lacticaseibacillus rhamnosus GG microcapsule co-encapsulated with beet residue extract. The chocolates were evaluated for moisture, water activity, texture, color properties, melting, physicochemical, and probiotic stability during storage. Furthermore, the survival of L. rhamnosus GG and the bioaccessibility of phenolic compounds were investigated under in vitro simulated gastrointestinal conditions. Regarding the characterization of probiotic microcapsules, the encapsulation efficiency of L. rhamnosus GG was > 89 % while the encapsulation efficiency of phenolic compounds was > 62 %. Chocolates containing probiotic microcapsules were less hard and resistant to breakage. All chocolates had a similar melting behavior (endothermic peaks between 32.80 and 34.40 °C). After 120 days of storage at 4 °C, probiotic populations > 6.77 log CFU/g were detected in chocolate samples. This result demonstrates the potential of this matrix to carry L. rhamnosus GG cells. Regarding the resistance of probiotic strains during gastric simulation, the co-encapsulation of L. rhamnosus GG with beet extract contributed to high counts during gastrointestinal transit, reaching the colon (48 h) with viable cell counts equal to 11.80 log CFU/g. Finally, one of our main findings was that probiotics used phenolic compounds as a substrate source, which may be an observed prebiotic effect.

Abstract:β-glucans come from cereals that have been located within compounds with prebiotic activity. They have presented several bioactivities that have determined their high functional value. The aim of this study was to identify the influence of oat β-glucan on the survival and proteolytic activity of Lactobacillus rhamnosus GG in a milk fermentation through an experimental design to optimize the process. For β-glucan extraction after dry milling of oats, two methods were applied: with and without enzymatic inactivation of the semolina. The highest extraction yield (45.25 g/L) was obtained with enzymatic inactivation. For the optimization of survival and proteolytic activity, a central design composed of axial points with two factors on three levels was used. Control factors were β-glucan and inoculum concentrations. According to response surface, the best survival growth rate of probiotic was observed with 4.38% of inoculum and 22.46 g/L of β-glucan, and the highest production of free amino groups was observed with 4.18% of inoculum and 22.71 g/L of β-glucan. Thus, β-glucan promotes the proteolytic activity of Lb. rhamnosus GG in milk fermentation.

Abstract:Galacto-oligosaccharides (GOS) are the prebiotic constituents with various functional qualities that promote the optimal proliferation of gut microbiota and are consequently beneficial to human health. GOS are not absorbed by humans and animals, but they play numerous health-related positive roles in the human body. Currently, their production has occupied much attention due to the increasing demand for functional foods. It could be produced by enzymatic and chemical approaches. Many microbial enzymes have been utilized for GOS production but the main source of these enzymes is lactose. Moreover, characterization of GOS structures is also a necessary and critical effort for understanding their mechanism of action. Hence, this review is written to understand the extensive but related features for the synthesis of GOS which includes: the enzymatic production of GOS through various enzymes and the types of enzymes such as free, immobilized, recombinant, crude, and purified enzymes that are involved in their synthesis, the techniques used in the purification of GOS, the analysis and quantification of GOS through various practices, and the functional properties of GOS are also discussed in this review. It also aims to combine new findings with current information in order to improve understanding of GOS formation and other factors related to its functionality.
Sugar-Free Milk Chocolate as a Carrier of Omega-3 Polyunsaturated Fatty Acids and Probiotics: A Potential Functional Food for the Diabetic Population

Abstract:Chocolate is an adequate matrix to deliver bioactive ingredients. However, it contains high sugar levels, one of the leading causes of chronic degenerative diseases. This work aimed to evaluate the effects of milk chocolate reformulation with alternative sugar sweeteners (Sw; isomalt + stevia), probiotics (Prob), and ω-3 polyunsaturated fatty acids (PUFAs) on its physicochemical properties and consumers’ acceptability. Lactobacillus plantarum 299v (L. p299v) and Lactobacillus acidophilus La3 (DSMZ 17742) were added as Prob strains, and fish oil (FO) was added as the source of ω-3 PUFAs. Prob addition resulted in chocolates with >2 × 107 colony forming unit (CFU) per serving size (12 g). Except for Prob, aw values of all treatments were <0.46. Sw and Sw + Prob presented the nearest values to the control in hardness, whereas Sw without FO increased fracturability. FO, Sw + FO, and Sw + Prob + FO contained 107.4 ± 12.84, 142.9 ± 17.9, and 133.78 ± 8.76 mg of ω-3 PUFAs per chocolate, respectively. Prob + FO increased the resistance of chocolate to shear stress, while Sw + FO showed a similar flow behavior to the control. The consumers’ acceptability of Sw + Prob chocolate was adequate, while Sw + Prob + FO had higher acceptability than Prob + FO. Health benefits of reformulated milk chocolates requires further assessment by in vitro, in vivo and clinical studies.

Abstract:Background: Different types of reduced-lactose yogurt, obtained by lactose hydrolysis using β-galactosidase enzyme, are commercially available. The breakdown of lactose modifies the carbohydrate profile, including the production of prebiotic galacto-oligosaccharides (GOS), which could affect the survival and activity of starter and probiotic cultures and the parameters of yogurt quality. The extension of these changes is dependent on the yogurt matrix composition. This study aimed to evaluate the influence of lactose hydrolysis on GOS, lactose, volatile profile and physicochemical parameters of different yogurt varieties during storage. Results: The presence of β-galactosidase enzyme did not affect either the global composition or the survival of cultures. Overall, the hydrolyzed products had lower acidity than traditional ones. GOS were found at similar levels in fresh hydrolyzed yogurts, whereas in traditional yogurts they were not detected. The proportion of ketones, acids and aldehydes seems to be more dependent on yogurt variety than on addition of the enzyme. Likewise, the storage period affected the volatile fraction to different degree; the increase in acid compounds was more pronounced in hydrolyzed than in traditional yogurts. Conclusion: This work shows that it is possible to obtain different varieties of reduced-lactose yogurt, some of them with additional benefits to health such as reduced fat, reduced calories, added with probiotic/inulin and enriched in GOS, with similar characteristics to traditional products. © 2016 Society of Chemical Industry.
