Abstract:Nickel (Ni) stress adversely affects plant growth and biomass accumulation, posturing severe menace to crop production and food security. The current study aimed to determine the putative role of sodium nitroprusside (SNP) in mitigating Ni-induced phytotoxicity and identify the underlying defense mechanisms in maize, which are poorly understood. Our findings showed that SNP significantly augmented plant growth, biomass, and photosynthesis-related attributes (Fv/Fm, Fm, qP ETR, and fPSII) through diminishing Ni uptake and translocation in root and shoot tissues of maize under Ni stress conditions. In parallel, exogenous SNP substantially relieved maize seedlings from Ni-induced stress by enhancing enzymatic (SOD, CAT, and GPX) and non-enzymatic (phenol and flavonoids) antioxidant defenses and reducing oxidative stress indicators (MDA and H2O2). The results revealed that SNP treatment increased the content of organic osmolyte glycine betaine and the activity of GST, concomitantly with ATP and ionic exchange capacity (including Ca2+-ATPase and Mg2+-ATPase), advocating its sufficiency to promote plant growth and avert Ni-induced stress in maize plants. The only exception was the production of organic acids (citric, oxalic, malic, and formic acids), which was reduced as SNP treatment relieved maize seedlings from Ni-induced oxidative damage. The application of SNP also displayed higher expression of defense- and detoxifying-related genes than in control treatments. Together, our data highlighted the mechanism involved in the amelioration of Ni toxicity by SNP; thus, suggesting a potential role of SNP in mitigating the adverse effects of Ni-contaminated soils to boost growth and yield of crop plants, that is, maize.
Exogenous Glycinebetaine Regulates the Contrasting Responses in Leaf Physiochemical Attributes and Growth of Maize under Drought and Flooding Stresses

Abstract:Flooding and drought are the two most devastating natural hazards limiting maize production. Exogenous glycinebetaine (GB), an osmotic adjustment agent, has been extensively used but there is limited research on its role in mitigating the negative effects of different abiotic stresses. This study aims to identify the different roles of GB in regulating the diverse defense regulation of maize against drought and flooding. Hybrids of Yindieyu 9 and Heyu 397 grown in pots in a ventilated greenhouse were subjected to flooding (2–3 cm standing layer) and drought (40–45% field capacity) at the three-leaf stage for 8 d. The effects of different concentrations of foliar GB (0, 0.5, 1.0, 5.0, and 10.0 mM) on the physiochemical attributes and growth of maize were tested. Greater drought than flooding tolerance in both varieties to combat oxidative stress was associated with higher antioxidant activities and proline content. While flooding decreased superoxide dismutase and guaiacol peroxidase (POD) activities and proline content compared to normal water, they all declined with stress duration, leading to a larger reactive oxygen species compared to drought. It was POD under drought stress and ascorbate peroxidase under flooding stress that played crucial roles in tolerating water stress. Foliar GB further enhanced antioxidant ability and contributed more effects to POD to eliminate more hydrogen peroxide than the superoxide anion, promoting growth, especially for leaves under water stress. Furthermore, exogenous GB made a greater increment in Heyu 397 than Yindieyu 9, as well as flooding compared to drought. Overall, a GB concentration of 5.0 mM, with a non-toxic effect on well-watered maize, was determined to be optimal for the effective mitigation of water-stress damage to the physiochemical characteristics and growth of maize.

Abstract:Abstract Background Compared with other abiotic stresses, drought stress causes serious crop yield reductions. Poly-γ-glutamic acid (γ-PGA), as an environmentally friendly biomacromolecule, plays an important role in plant growth and regulation. Results In this project, the effect of exogenous application of γ-PGA on drought tolerance of maize ( Zea mays . L) and its mechanism were studied. Drought dramatically inhibited the growth and development of maize, but the exogenous application of γ-PGA significantly increased the dry weight of maize, the contents of ABA, soluble sugar, proline, and chlorophyll, and the photosynthetic rate under severe drought stress. RNA-seq data showed that γ-PGA may enhance drought resistance in maize by affecting the expression of ABA biosynthesis, signal transduction, and photosynthesis-related genes and other stress-responsive genes, which was also confirmed by RT–PCR and promoter motif analysis. In addition, diversity and structure analysis of the rhizosphere soil bacterial community demonstrated that γ-PGA enriched plant growth promoting bacteria such as Actinobacteria , Chloroflexi , Firmicutes , Alphaproteobacteria and Deltaproteobacteria . Moreover, γ-PGA significantly improved root development, urease activity and the ABA contents of maize rhizospheric soil under drought stress. This study emphasized the possibility of using γ-PGA to improve crop drought resistance and the soil environment under drought conditions and revealed its preliminary mechanism. Conclusions Exogenous application of poly-γ-glutamic acid could significantly enhance the drought resistance of maize by improving photosynthesis, and root development and affecting the rhizosphere microbial community.

Abstract:Salt stress is a common abiotic stress that limits the growth, development and yield of maize (Zea mays L.). To better understand the response of maize to salt stress and the mechanism by which exogenous glycine betaine (GB) alleviates the damaging effects of salt stress, the morphology, physiological and biochemical indexes, and root transcriptome expression profiles of seedlings of salt-sensitive inbred line P138 and salt-tolerant inbred line 8723 were compared under salt stress and GB-alleviated salt stress conditions. The results showed that under salt stress the growth of P138 was significantly inhibited and the vivo ion balance was disrupted, whereas 8723 could prevent salt injury by maintaining a high ratio of K+ to Na+. The addition of a suitable concentration of GB could effectively alleviate the damage caused by salt stress, and the mitigating effect on salt-sensitive inbred line P138 was more obvious than that on 8723. Transcriptome analysis revealed that 219 differentially expressed genes (DEGs) were up-regulated and 153 DEGs were down-regulated in both P138 and 8723 under NaCl treatment, and that 487 DEGs were up-regulated and 942 DEGs were down-regulated in both P138 and 8723 under salt plus exogenous GB treatment. In 8723 the response to salt stress is mainly achieved through stabilizing ion homeostasis, strong signal transduction activation, increasing reactive oxygen scavenging. GB alleviates salt stress in maize mainly by inducing gene expression changes to enhance the ion balance, secondary metabolic level, reactive oxygen scavenging mechanism, signal transduction activation. In addition, the transcription factors involved in the regulation of salt stress response and exogenous GB mitigation mainly belong to the MYB, MYB-related, AP2-EREBP, bHLH, and NAC families. We verified 10 selected up-regulated DEGs by quantitative real-time polymerase chain reaction (qRT-PCR), and the expression results were basically consistent with the transcriptome expression profiles. Our results from this study may provide the theoretical basis for determining maize salt tolerance mechanisms and the mechanism by which GB regulates salt tolerance.

Abstract:Plant growth and development are adversely affected by various environmental stresses. In this study, we investigated the possible function of glycinebetaine (GB) in improving abiotic stress tolerance in maize plants by exogenous application. Results indicated that GB application could effectively improve maize germination percentages, biomass weights and grain yield per plant under abiotic stresses. Moreover, GB-treated plants showed higher reactive oxygen species (ROS)-scavenging capacities, therefore less oxidative destruction than control plants under stresses. In addition, abscisic acid (ABA) levels were increased in GB-treated plants compared to control plants. The comparison of transcriptome profile was performed to analyze the possible mechanism underlying the enhanced tolerance mediated by GB. Many differentially expressed genes (DEGs) between GB-treated and control plants were found to be involved in responses to oxidative stress, abiotic stress and ABA. Taken together, these results indicated that GB might enhance abiotic stress tolerance in maize by regulating ROS-scavenging capacities and ABA-mediated stress-responsive pathways.

New Genes Identified as Modulating Salt Tolerance in Maize Seedlings Using the Combination of Transcriptome Analysis and BSA

Abstract:(1) Background: Salt stress is an abiotic factor that limits maize yield and quality. A highly salt-tolerance inbred AS5 and a salt-sensitive inbred NX420 collected from Ningxia Province, China, were used to identify new genes for modulating salt resistance in maize. (2) Methods: To understand the different molecular bases of salt tolerance in AS5 and NX420, we performed BSA-seq using an F2 population for two extreme bulks derived from the cross between AS5 and NX420. Transcriptomic analysis was also conducted for AS5 and NX420 at the seedling stage after treatment with 150 mM of NaCl for 14 days. (3) Results: AS5 had a higher biomass and lower Na+ content than NX420 in the seedling stage after treatment with 150 mM NaCl for 14 days. One hundred and six candidate regions for salt tolerance were mapped on all of the chromosomes through BSA-seq using F2 in an extreme population. Based on the polymorphisms identified between both parents, we detected 77 genes. A large number of differentially expressed genes (DEGs) at the seedling stage under salt stress between these two inbred lines were detected using transcriptome sequencing. GO analysis indicated that 925 and 686 genes were significantly enriched in the integral component of the membrane of AS5 and NX420, respectively. Among these results, two and four DEGs were identified as overlapping in these two inbred lines using BSA-seq and transcriptomic analysis, respectively. Two genes (Zm00001d053925 and Zm00001d037181) were detected in both AS5 and NX420; the transcription level of Zm00001d053925 was induced to be significantly higher in AS5 than in NX420 (41.99 times versus 6.06 times after 150 mM of NaCl treatment for 48 h), while the expression of Zm00001d037181 showed no significant difference upon salt treatment in both lines. The functional annotation of the new candidate genes showed that it was an unknown function protein. (4) Conclusions: Zm00001d053925 is a new functional gene responding to salt stress in the seedling stage, which provides an important genetic resource for salt-tolerant maize breeding.

Abstract:In maize (Zea mays L.) production, grain-filling stage is a pivotal phase that drought occurring would irreversibly cause leaf senescence and yield loss. Meanwhile, drought induces the disequilibrium of carbohydrates that inevitably produces the toxic substance methylglyoxal (MG) that plays dual roles including cytotoxic metabolite or signaling molecule in plants. However, how exogenous MG influences maize yield formation in response to drought during grain filling remains unknown. In this study, maize plants were exposed to moderate and severe drought conditions from 15 to 28 days after pollination with foliar spraying of MG (0, 15, 25, and 35 mM). Notably, MG application significantly increased kernel number and retrieved yield loss by 14–48% under drought, demonstrating an increased resistance of the plants. Interestingly, high (25–35 mM) and low concentrations (15–25 mM) of MG application under moderate and severe drought performed the highest yield output, respectively. To investigate the mechanisms by which MG enhanced drought resistance, we confirmed that MG application postponed leaf senescence under both drought conditions and significantly improved photosynthesis under severe drought during filling stage. Specifically, MG application escalated the levels of soluble sugar and sucrose while suppressing endogenous MG accumulation by activating glyoxalase system in leaf during the early phase of drought stress and well-watered condition. Collectively, these results demonstrate that exogenous MG application enhances drought tolerance during maize grain filling, possibly through regulation on the homeostasis of endogenous MG and sugars. These findings provide a new approach to secure yield against drought stress in maize production.

Abstract:Improving crop salt tolerance is an adaptive measure to climate change for meeting future food demands. Previous studies have reported that glycine betaine (GB) plays critical roles as an osmolyte in enhancing plant salt resistance. However, the mechanism underlying the GB regulating plant Na + homeostasis during response to salinity is poorly understood. In this study, hydroponically cultured maize with 125 mM NaCl for inducing salinity stress was treated with 100 μM GB. We found that treatment with GB improved the growth of maize plants under non-stressed (NS) and salinity-stressed (SS) conditions. Treatment with GB significantly maintained the properties of chlorophyll fluorescence, including Fv/Fm, ΦPSII, and ΦNPQ, and increased the activity of the antioxidant enzymes for mitigating salt-induced growth inhibition. Moreover, GB decreased the Na + /K + ratio primarily by reducing the accumulation of Na + in plants. The results of NMT tests further confirmed that GB increased Na + efflux from roots under SS condition, and fluorescence imaging of cellular Na + suggested that GB reduced the cellular allocation of Na + . GB additionally increased Na + efflux in leaf protoplasts under SS condition, and treatment with sodium orthovanadate, a plasma membrane (PM) H + -ATPase inhibitor, significantly alleviated the positive effects of GB on Na + efflux under salt stress. GB significantly improved the vacuolar activity of NHX but had no significant effects on the activity of V type H + -ATPases. In addition, GB significantly upregulated the expression of the PM H + -ATPase genes, ZmMHA2 and ZmMHA4 , and the Na + /H + antiporter gene, ZmNHX1. While, the V type H + -ATPases gene, ZmVP1 , was not significantly regulated by GB. Altogether these results indicate that GB regulates cellular Na + homeostasis by enhancing PM H+-ATPases gene transcription and protein activities to improve maize salt tolerance. This study provided an extended understanding of the functions of GB in plant responses to salinity, which can help the development of supportive measures using GB for obtaining high maize yield in saline conditions.

Abstract:Maize (Zea mays L.) is one of the major staple crops providing human food, animal feed, and raw material support for biofuel production. For its growth and development, maize requires essential macronutrients. In particular, nitrogen (N) plays an important role in determining the final yield and quality of a maize crop. However, the excessive application of N fertilizer is causing serious pollution of land area and water bodies. Therefore, cultivating high-yield and low-N-tolerant maize varieties is crucial for minimizing the nitrate pollution of land and water bodies. Here, based on the analysis of the maize leaf transcriptome and proteome at the grain filling stage, we identified 3957 differentially expressed genes (DEGs) and 329 differentially abundant proteins (DAPs) from the two maize hybrids contrasting in N stress tolerance (low-N-tolerant XY335 and low-N-sensitive HN138) and screened four sets of low-N-responsive genes and proteins through Venn diagram analysis. We identified 761 DEGs (253 up- and 508 down-regulated) specific to XY335, whereas 259 DEGs (198 up- and 61 down-regulated) were specific to HN138, and 59 DEGs (41 up- and 18 down-regulated) were shared between the two cultivars under low-N-stress conditions. Meanwhile, among the low-N-responsive DAPs, thirty were unique to XY335, thirty were specific to HN138, and three DAPs were shared between the two cultivars under low-N treatment. Key among those genes/proteins were leucine-rich repeat protein, DEAD-box ATP-dependent RNA helicase family proteins, copper transport protein, and photosynthesis-related proteins. These genes/proteins were involved in the MAPK signaling pathway, regulating membrane lipid peroxidation, and photosynthesis. Our results may suggest that XY335 better tolerates low-N stress than HN138, possibly through robust low-N-stress sensing and signaling, amplified protein phosphorylation and stress response, and increased photosynthesis efficiency, as well as the down-regulation of 'lavish' or redundant proteins to minimize N demand. Additionally, we screened glutathione transferase 42 (ZmGST42) and performed physiological and biochemical characterizations of the wild-type (B73) and gst42 mutant at the seedling stage. Resultantly, the wild-type exhibited stronger tolerance to low N than the mutant line. Our findings provide a better understanding of the molecular mechanisms underlying low-N tolerance during the maize grain filling stage and reveal key candidate genes for low-N-tolerance breeding in maize.

Abstract:Abstract Drought is one of the major environmental stresses that negatively affect the maize ( Zea mays L.) growth and production throughout the world. Foliar applications of plant growth regulators, micronutrients or osmoprotectants for stimulating drought-tolerance in plants have been intensively reported. A controlled pot experiment was conducted to study the relative efficacy of salicylic acid (SA), zinc (Zn), and glycine betaine (GB) foliar applications on morphology, chlorophyll contents, relative water content (RWC), gas-exchange attributes, activities of antioxidant enzymes, accumulations of reactive oxygen species (ROS) and osmolytes, and yield attributes of maize plants exposed to two soil water conditions (85% field capacity: well-watered, 50% field capacity: drought stress) during critical growth stages. Drought stress significantly reduced the morphological parameters, yield and its components, RWC, chlorophyll contents, and gas-exchange parameters except for intercellular CO 2 concentration, compared with well water conditions. However, the foliar applications considerably enhanced all the above parameters under drought. Drought stress significantly (p < 0.05) increased the hydrogen peroxide and superoxide anion contents, and enhanced the lipid peroxidation rate measured in terms of malonaldehyde (MDA) content. However, ROS and MDA contents were substantially decreased by foliar applications under drought stress. Antioxidant enzymes activity, proline content, and the soluble sugar were increased by foliar treatments under both well-watered and drought-stressed conditions. Overall, the application of GB was the most effective among all compounds to enhance the drought tolerance in maize through reduced levels of ROS, increased activities of antioxidant enzymes and higher accumulation of osmolytes contents.

Exogenous Sorbitol Application Confers Drought Tolerance to Maize Seedlings through Up-Regulating Antioxidant System and Endogenous Sorbitol Biosynthesis

Abstract:This study aims to explore the impacts of exogenous sorbitol on maize seedlings under polyethylene glycol (PEG)-simulated drought stress. Six treatments were set: normal condition (CK), PEG (P), 10 mM sorbitol (10S), PEG plus 10 mM sorbitol (10SP), 100 mM sorbitol (100S) and PEG plus 100 mM sorbitol (100SP). Maize seedlings' growth under PEG-simulated drought stress was significantly inhibited and exogenous sorbitol largely alleviated this growth inhibition. The seedlings under 10SP treatment grew much better than those under P, 100S and 100SP treatments and no significant difference in growth parameters was observed between the control and 10S treatment. The seedlings treated with 10SP had higher contents of soluble sugar, soluble protein, proline, ascorbic acid (AsA), reduced glutathione (GSH), sorbitol and relative water content, higher activities of antioxidant enzymes and aldose reductase, but lower contents of malondialdehyde (MDA), H2O2 and relative electrical conductivity than those treated with P, 100S and 100SP. qRT-PCR analysis showed that the transcript levels of genes encoding putative aldose reductase (AR) under P treatment were significantly up-regulated in sorbitol-applied treatments. Taken together, the results demonstrated that exogenous sorbitol application conferred drought tolerance to maize seedlings by up-regulating the expression levels of AR-related genes to enhance the accumulation of intracellular osmotic substances such as sorbitol and improve antioxidant systems to tone down the damage caused by drought stress.

Abstract:Drought stress seriously affects the growth, development, yield, and quality of maize. This study aimed to investigate the effects of exogenous 5-ALA on root morphology and physiological changes in maize seedlings and to detect its regulatory network. The results showed that adding 25 mg/L 5-ALA accelerated root morphogenesis (root average diameter, main root length, total root length, and root surface area) and promoted dry matter accumulation and free radical removal. Transcriptome analysis showed that after applying exogenous 5-ALA, differently expressed genes (DEGs) were mainly involved in histidine metabolism, amino acid biosynthesis, plasma membrane components, secondary active sulfate transmembrane transporter activity, and anion reverse transporter activity. Two inbred lines specifically responded to organelle and structural molecular activity, and 5-ALA may regulate maize roots to achieve drought tolerance through these two pathways. In addition, candidate genes that may regulate maize root growth were screened by weighted gene co-expression network analysis (WGCNA). These genes may play important roles in alleviating drought stress through lignin synthesis, heat shock proteins, iron storage and transport, calcium binding proteins, and plasma membrane regulation of exogenous regulator 5-ALA. Our results may provide a theoretical basis for clarifying the response of maize seedling roots to drought and the mechanism of exogenous hormones in alleviating drought.

Regulation Mechanisms of Nitrogen-Doped Carbon Dots in Enhanced Maize Photosynthesis under Drought Stress

Abstract:Herein, greenhouse experiments were designed to reveal the role of nitrogen-doped carbon dots (N-CDs) in enhancing maize drought tolerance. Two humidity conditions were created: adequate watering (soil moisture, 75%) and drought stress (soil moisture, 35%). Corn seedlings were harvested after spraying the N-CD solution (5 mg·L–1) on maize leaves for 5 days. The results indicated that foliar application of N-CDs increased the net photosynthesis rate (28.6%) of maize, and the fresh and dry weights of roots and shoots increased by 224.5, 360.0, 230.8, and 63.3% under drought stress, respectively. N-CDs showed high reactive oxygen species (ROS)-scavenging activity, resulting in enhanced superoxide dismutase activity (26.7%) and reduced malondialdehyde enzyme activity (18.9%). Besides, N-CDs could be used as light-harvesting materials to improve the light utilization efficiency, upregulate psbA gene expression (81.7-fold), and promote fast synthesis of the D1 protein, which could repair photosystem II under drought stress. Therefore, foliar-sprayed N-CDs could improve photosynthesis through multiple pathways under drought stress: light harvesting, photoprotection, and light repairing. Then, N-CD exposure reduced the corn yield loss under drought by nearly 30% compared with those of the control groups in a full life cycle study. Therefore, this study found for the first time that N-CD-enabled nanoagriculture could ensure crop growth and yield under drought stress, which would be important for global crop cultivation and a promising alternative to deal with the global climate change.

Abstract:Water deficit is a key limiting factor for limiting yield in maize (Zea mays L.). It is crucial to elucidate the molecular regulatory networks of stress tolerance for genetic enhancement of drought tolerance. The mechanism of drought tolerance of maize was explored by comparing physiological and transcriptomic data under normal conditions and drought treatment at polyethylene glycol- (PEG-) induced drought stress (5%, 10%, 15%, and 20%) in the root during the seedling stage. The content of saccharide, SOD, CAT, and MDA showed an upward trend, proteins showed a downward trend, and the levels of POD first showed an upward trend and then decreased. Compared with the control group, a total of 597, 2748, 6588, and 5410 differentially expressed genes were found at 5%, 10%, 15%, and 20% PEG, respectively, and 354 common DEGs were identified in these comparisons. Some differentially expressed genes were remarkably enriched in the MAPK signaling pathway and plant hormone signal transduction. The 50 transcription factors (TFs) divided into 15 categories were screened from the 354 common DEGs during drought stress. Auxin response factor 10 (ARF10), auxin-responsive protein IAA9 (IAA9), auxin response factor 14 (ARF14), auxin-responsive protein IAA1 (IAA1), auxin-responsive protein IAA27 (IAA27), and 1 ethylene response sensor 2 (ERS2) were upregulated. The two TFs, including bHLH 35 and bHLH 96, involved in the MAPK signal pathway and plant hormones pathway, are significantly upregulated in 5%, 10%, 15%, and 20% PEG stress groups. The present study provides greater insight into the fundamental transcriptome reprogramming of grain crops under drought.

Exogenous Melatonin Reinforces Photosynthesis, Antioxidant Defense and Gene Expression to Ameliorate Na2CO3 Stress in Maize

Abstract:Salt stress can seriously affect the growth and development of maize (Zea mays L.), resulting in a great yield loss. Melatonin (MT), an indole hormone, is a potential enhancer of plant tolerance against salt stress. However, the complex mechanisms of MT application in enhancing maize salt tolerance are still unclear. Herein, three-leaf seedlings of salt-susceptible P138 and its salt-resistant ethyl methane sulfonate (EMS)-104 mutant were cultured with or without 150 μM MT application under 0 and 100 mM Na2CO3 treatments for seven days, to systematically explore the response mechanisms of exogenous MT in improving the salt tolerance of maize. The results showed that salt stress triggered an escalation in reactive oxygen species production, enhanced multiple antioxidant enzymes' activities, impaired cellular membrane permeability, inhibited photosynthetic pigment accumulation, and ultimately undermined the vigor and photosynthetic prowess of the seedlings. While suitable MT application counteracted the detrimental impacts of Na2CO3 on seedlings' growth and photosynthetic capacity, the seedling length and net photosynthetic rate of P138 and EMS-104 were increased by 5.5% and 18.7%, and 12.7% and 54.5%, respectively. Quantitative real-time PCR (qRT-PCR) analysis further showed that MT application activated the expression levels of antioxidant enzyme-related genes (Zm00001d025106, Zm00001d031908, Zm00001d027511, and Zm00001d040364) and pigment biosynthesis-related genes (Zm00001d011819 and Zm00001d017766) in both maize seedlings under Na2CO3 stress; they then formed a complex interaction network of gene expression, multiple physiological metabolisms, and phenotype changes to influence the salt tolerance of maize seedlings under MT or Na2CO3 stress. To sum up, these observations underscore that 150 μM MT can alleviate salt injury of maize seedlings, which may provide new insights for further investigating MT regulation mechanisms to enhance maize seedlings' salt resistance.

Integrative Effects of Zinc Nanoparticle and PGRs to Mitigate Salt Stress in Maize

Abstract:Salinity is one of the most critical problems for agricultural development and threatens future food safety. Therefore, we aimed to investigate root application of zinc oxide nanoparticles (ZnO-NPs; 0, 50, 100 mg/L), 24-epibrassinolide (EBL; 0, 0.02, 0.04 μM), and their combinations on the growth and performance of maize (Zea mays L.) as a model plant grown under salt stress (i.e., 0, 5 and 10 dS m−1) in a hydroponic system. The results showed that the highest salt stress negatively affected growth, physiological, and biochemical traits of maize. However, the application of EBL, ZnO-NPs, and their combinations significantly mitigated salt stress and improved the growth and performance of the physiological system in maize plants. In particular, the combination treatment of 100 mg/L ZnO-NPs + 0.02 μM EBL surpassed all other root treatments and resulted in the highest root and shoot growth, leaf area, relative leaf water content, net photosynthesis, total chlorophyll content, and uptake of zinc (Zn) and potassium (K). Furthermore, it minimized salt stress by reducing Na uptake, Na/K ratio, and proline in stressed maize plants. For example, the combination treatment of 100 mg/L ZnO-NPs + 0.02 μM EBL improved root length by +175%, shoot length by +39%, leaf area by +181%, RWC by +12%, net photosynthesis by +275, total chlorophyll content by +33%, and total phenolic content by +38%, in comparison to those obtained from the control, respectively. Furthermore, it enhanced the roots and leaves uptake of Zn under high salt stress treatment (i.e., 10 dS m−1) by +125% and +94%, and K+ by +39% and +51%, as compared to those grown without any of NPs or EBL treatments, respectively. Thus, the root application of 100 mg/L ZnO-NPs + 0.02 μM EBL can be a potential option to mitigate salt stress and improve the physiological, biochemical, and performance of strategy crops such maize.

Abstract:A technique is described for measurement of oxidation and energy production rate in vitro in small unrestrained animals. The method consists of a combined system of radiorespirometry and indirect calorimetry in an open air circuit. This makes possible the study of the following parameters: whole body energy production rate, net oxidation of fat, carbohydrates and protein, RQ, specific energy production from exogenous labelled substrates, specific activity of (14CO2) in expired air and cumulative oxidation of labelled substrates. The accuracy of this system has been evaluated by various methods such as calibration with high precision gases, the burning of methanol and longitudinal studies in growing and weight-stable rodents. The overall accuracy was found to be approximately +/- 2% for parameters related to whole body energy metabolism and better than +/- 1% for collection of labelled expired CO2. This includes methodological and interindividual variations among inbred animals. The system also allows the use of intravenous and intragastric infusion lines during long-term periods without any significant restriction of the rats.
