Abstract:This pioneering research investigates the manner in which cellular morphological responses are linked to the nature of genome information using deep learning. Leukemia is highly heterogeneous, meaning that different types of leukemia require different treatments and have different prognoses. Current clinical diagnostic and typing tests are complex and time‐consuming. In particular, all of these tests rely on bone marrow aspiration, which is invasive and leads to poor patient compliance, exacerbating treatment delays. Morphological analysis of peripheral blood cells (PBC) is still primarily used to distinguish between benign and malignant hematologic disorders, but it remains a challenge to diagnose and type these diseases solely by direct observation of peripheral blood(PB) smears by human experts. In this study, we apply a segmentation‐based enhanced residual network that uses progressive multigranularity training with jigsaw patches. It is trained on a self‐built annotated dataset of 21,208 images from 237 patients, including five types of benign white blood cells(WBCs) and eight types of leukemic cells. The network is not only able to discriminate between benign and malignant cells, but also to typify leukemia using a single peripheral blood cell. The network effectively differentiated acute promyelocytic leukemia (APL) from other types of acute myeloid leukemia (non‐APL), achieving a precision rate of 89.34%, a recall rate of 97.37%, and an F1 score of 93.18% for APL. In contrast, for non‐APL cases, the model achieved a precision rate of 92.86%, but a recall rate of 74.63% and an F1 score of 82.75%. In addition, the model discriminates acute lymphoblastic leukemia(ALL) with the Ph chromosome from those without. This approach could improve patient compliance and enable faster and more accurate typing of leukemias for early diagnosis and treatment to improve survival.

Abstract:Acute lymphoblastic leukemia (ALL) is a form of blood cancer that affects the white blood cells. ALL constitutes approximately 25% of pediatric cancers. Early diagnosis and treatment of ALL are crucial for improving patient outcomes. The task of identifying immature leukemic blasts from normal cells under the microscope can prove challenging, since the images of a healthy and cancerous cell appear similar morphologically. In this study, we propose a binary image classification model to assist in the diagnostic process of ALL. Our model takes as input microscopic images of blood samples and outputs a binary prediction of whether the sample is normal or cancerous. Our dataset consists of 10661 images out of 118 subjects. Deep learning techniques on convolutional neural network architectures were used to achieve accurate classification results. Our proposed method achieved 94.3% accuracy and could be used as an assisting tool for hematologists trying to predict the likelihood of a patient developing ALL.

Abstract:Traditionally, researchers, doctors, and hematologists have faced difficulties in making a timely diagnosis of leukemia. Since, unlike other malignancies, leukemia typically does not create a (tumor) that can be seen in imaging techniques like X-rays or CT scans. As a result the procedures available at medical diagnosis centres of leukemia consumes a lot of time. Influenced by the capabilities of artificial intelligence techniques diagnosing the disease, the paper introduces advanced CNN models which includes InceptionV3, DenseNet201, Xception, ResNet152V2, and two hybrid models, i.e., InceptionResNetV2 and XceptionInceptionResNetV2 for identifying and classifying normal and leukemia cancer cells. Accuracy, root mean square error, recall, precision, F1 score, and loss are used to evaluate all applied advanced learning models. The Acute lymphoblastic leukemia dataset, which is separated into two classes: normal cells and malignant leukemia cells, is used in this study. During the pre-processing stage, every image undergo enhancement and are visually shown to fetch the color channels in the shape of an RGB histogram. Later the images are augmented to produce regions of interest by generating extreme points and employing adaptive thresholding techniques before being provided to the applied models for training. During the experimentation, it was discovered that InceptionResNetV2 had the highest validation accuracy of 98.59%. Similarly, DenseNet201 had the highest precision (97.57%), followed by InceptionV3 with recall (95.77%) and F1 score (95.56%). Moreover, the confusion matrix has also been generated to obtain the models' recall, precision, and F1 score values for different classes of the dataset.

Abstract:Experimental and numerical studies were carried out to investigate the viscous hydrodynamic forces acting on a tanker model in oblique motion. Oblique towing tests with a KVLCC2 model were conducted in the towing tank of Shanghai Jiao Tong University(SJTU) in deep water. The lateral forces and yaw moments on the model at different drift angles were measured. The viscous flow around the tanker model in oblique motion was simulated by solving Reynolds-averaged Navier-Stokes equations, and the lateral force, yaw moment, pressure distribution on the hull and the wake were calculated. The experimental and numerical results of the lateral force and yaw moment acting on the hull were compared, and the effect of drift angles on the hydrodynamic forces was analyzed.

Abstract:Acute myeloid leukemia (AML) is a heterogenous blood cancer with a dismal prognosis. It emanates from leukemic stem cells (LSCs) arising from the genetic transformation of hematopoietic stem cells (HSCs). LSCs hold prognostic value, but their molecular and immunophenotypic heterogeneity poses challenges: there is no single marker for identifying all LSCs across AML samples. We hypothesized that imaging flow cytometry (IFC) paired with artificial intelligence-driven image analysis could visually distinguish LSCs from HSCs based solely on morphology. Initially, a seven-color IFC panel was employed to immunophenotypically identify LSCs and HSCs in bone marrow samples from five AML patients and ten healthy donors, respectively. Next, we developed convolutional neural network (CNN) models for HSC-LSC discrimination using brightfield (BF), side scatter (SSC), and DNA images. Classification using only BF images achieved 86.96% accuracy, indicating significant morphological differences. Accuracy increased to 93.42% when combining BF with DNA images, highlighting differences in nuclear morphology, although DNA images alone were inadequate for accurate HSC-LSC discrimination. Model development using SSC images revealed minor granularity differences. Performance metrics varied substantially between AML patients, indicating considerable morphologic variations among LSCs. Overall, we demonstrate proof-of-concept results for accurate CNN-based HSC-LSC differentiation, instigating the development of a novel technique within AML monitoring.

Abstract:The bone marrow overproduces immature cells in the malignancy known as Acute Lymphoblastic Leukemia (ALL). In the United States, about 6500 occurrences of ALL are diagnosed each year in both children and adults, comprising nearly 25% of pediatric cancer cases. Recently, many computer-assisted diagnosis (CAD) systems have been proposed to aid hematologists in reducing workload, providing correct results, and managing enormous volumes of data. Traditional CAD systems rely on hematologists' expertise, specialized features, and subject knowledge. Utilizing early detection of ALL can aid radiologists and doctors in making medical decisions. In this study, Deep Dilated Residual Convolutional Neural Network (DDRNet) is presented for the classification of blood cell images, focusing on eosinophils, lymphocytes, monocytes, and neutrophils. To tackle challenges like vanishing gradients and enhance feature extraction, the model incorporates Deep Residual Dilated Blocks (DRDB) for faster convergence. Conventional residual blocks are strategically placed between layers to preserve original information and extract general feature maps. Global and Local Feature Enhancement Blocks (GLFEB) balance weak contributions from shallow layers for improved feature normalization. The global feature from the initial convolution layer, when combined with GLFEB-processed features, reinforces classification representations. The Tanh function introduces non-linearity. A Channel and Spatial Attention Block (CSAB) is integrated into the neural network to emphasize or minimize specific feature channels, while fully connected layers transform the data. The use of a sigmoid activation function concentrates on relevant features for multiclass lymphoblastic leukemia classification The model was analyzed with Kaggle dataset (16,249 images) categorized into four classes, with a training and testing ratio of 80:20. Experimental results showed that DRDB, GLFEB and CSAB blocks' feature discrimination ability boosted the DDRNet model F1 score to 0.96 with minimal computational complexity and optimum classification accuracy of 99.86% and 91.98% for training and testing data. The DDRNet model stands out from existing methods due to its high testing accuracy of 91.98%, F1 score of 0.96, minimal computational complexity, and enhanced feature discrimination ability. The strategic combination of these blocks (DRDB, GLFEB, and CSAB) are designed to address specific challenges in the classification process, leading to improved discrimination of features crucial for accurate multi-class blood cell image identification. Their effective integration within the model contributes to the superior performance of DDRNet.
Deep Learning Models for the Diagnosis of Acute Lymphoblastic Leukemia from Bone Marrow Images : A Comprehensive Literature Review

Abstract:Introduction: Acute lymphoblastic leukemia (ALL) is an aggressive blood cancer that begins in the bone marrow. It is the most common childhood cancer and requires early and accurate diagnosis for optimal treatment outcomes. Through automated image analysis of peripheral blood smears and bone marrow biopsies, artificial intelligence (AI), particularly Deep Learning (DL), have opened new avenues for improving ALL diagnosis. While bone marrow testing is the gold standard for ALL confirmation, AI applications in diagnosing ALL from bone marrow images have received less attention in the literature than peripheral blood smears. This review aims to assess the current state of AI in ALL diagnosis using bone marrow aspirates and biopsies, with a focus on Deep Learning (DL) models such as Convolutional Neural Networks (CNNs). Methods: A comprehensive literature search, conducted on June 11th, 2023, covered major medical databases, including PubMed/MEDLINE, Scopus, Embase, and Web of Science. Relevant keywords like “acute lymphoblastic leukemia,” “deep learning,” and “neural network” were employed, without time frame restrictions. Articles were included if they assessed the metrics of AI applications for ALL diagnosis in bone marrow aspirates. Articles were excluded if they: (1) had different outcomes, (2) were reviews, (3) were abstracts only, or (4) only reported peripheral blood smear metrics. Results: The search yielded 496 articles. After eliminating duplicates (282), title and abstract screening on the Rayyan platform excluded 204 records, leaving 214 articles eligible for full-text screening. Ten relevant articles were ultimately included in the review. Diverse approaches were presented to enhance diagnostic accuracy and efficiency. One study achieved 100% accuracy in classifying leukemic cells using a unique Convolutional Leaky RELU with CatBoost and XGBoost (CLR-CXG) design. Another study employed transfer learning with CNNs, obtaining 95.3% accuracy for AML, ALL, and CML classification with DenseNet121. Furthermore, the novel “i-Net” model demonstrated 99.18% validation accuracy for white blood cancer segmentation and classification. Adaptive Multi-objective CAT algorithms achieved 99.45% accuracy in detecting bone marrow cancer cells. An AI-based system using deep learning exhibited 97.2% accuracy in diagnosing ALL, while a robust fuzzy logic algorithm combined with a radial basis function neural network achieved 82.93% accuracy in diagnosing ALL in developing countries. Additionally, AI technologies based on computer microscopy demonstrated 95% and 97.5% accuracy in diagnosing ALL and minimal residual disease, respectively. A computer-aided system achieved 97.78% accuracy in classifying ALL subtypes. Conclusion: The potential of AI-driven approaches, particularly deep learning models like CNNs, in improving ALL diagnosis from bone marrow images is highlighted in this review. Because of their superior sensitivity and specificity over peripheral blood smears, bone marrow samples are the gold standard for ALL confirmation. The morphological and cellular properties of bone marrow samples provide important information for disease classification and monitoring. While DL and CNNs have shown promise in diagnosing ALL from peripheral blood smears, their use in bone marrow samples has yet to be investigated deeply. More research and external validation are required to fully realize AI's potential in ALL diagnosis from bone marrow specimens.

Abstract:Acute lymphoblastic leukemia (ALL) is the most common variant of paediatric cancer that creates numerous immature white blood cells affecting the bone marrow. Manual diagnosis of leukemia from microscopic evaluation of stained sample slides is an exhausting process, which is less accurate and susceptible to human errors. Additionally, identifying the leukemic blast cells under the microscope is complicated due to morphological similarity with the normal cell images. In this paper, we proposed an automated method to analyse the blood smear images using Local Binary Pattern (LBP) and classify the leukemic blast cells and normal cells. We have analysed the performance of machine learning and deep learning models such as Support Vector Machine (SVM), k-Nearest Neighbor algorithm (kNN), Artificial Neural Network (ANN), and Convolutional Neural Network (CNN). For classifying ALL and normal cell images, kNN achieved an accuracy of 94.4%, SVM, and ANN achieved an accuracy of 98.6%, and CNN achieved an accuracy of 99.6%. SVM achieved the highest sensitivity of 100%.
A Hybrid Feature Fusion Deep Learning Framework for Leukemia Cancer Detection in Microscopic Blood Sample Using Gated Recurrent Unit and Uncertainty Quantification

Abstract:Acute lymphoblastic leukemia (ALL) is the most malignant form of leukemia and the most common cancer in adults and children. Traditionally, leukemia is diagnosed by analyzing blood and bone marrow smears under a microscope, with additional cytochemical tests for confirmation. However, these methods are expensive, time consuming, and highly dependent on expert knowledge. In recent years, deep learning, particularly Convolutional Neural Networks (CNNs), has provided advanced methods for classifying microscopic smear images, aiding in the detection of leukemic cells. These approaches are quick, cost effective, and not subject to human bias. However, most methods lack the ability to quantify uncertainty, which could lead to critical misdiagnoses. In this research, hybrid deep learning models (InceptionV3-GRU, EfficientNetB3-GRU, MobileNetV2-GRU) were implemented to classify ALL. Bayesian optimization was used to fine tune the model's hyperparameters and improve its performance. Additionally, Deep Ensemble uncertainty quantification was applied to address uncertainty during leukemia image classification. The proposed models were trained on the publicly available datasets ALL-IDB1 and ALL-IDB2. Their results were then aggregated at the score level using the sum rule. The parallel architecture used in these models offers a high level of confidence in differentiating between ALL and non-ALL cases. The proposed method achieved a remarkable detection accuracy rate of 100% on the ALL-IDB1 dataset, 98.07% on the ALL-IDB2 dataset, and 98.64% on the combined dataset, demonstrating its potential for accurate and reliable leukemia diagnosis.

Abstract:A lightweight deep learning model is employed to detect lymphoblastic leukemia using a high‐quality dataset. The model adopts a progressive shrinking approach to minimize its size, achieving an average detection accuracy of 92.51%. Leukemia is a hematological malignancy that significantly impacts the human immune system. Early detection helps to effectively manage and treat cancer. Although deep learning techniques hold promise for early detection of blood disorders, their effectiveness is often limited by the physical constraints of available datasets and deployed devices. For this investigation, we collect an excellent‐quality dataset of 17,826 morphological bone marrow cell images from 85 patients with lymphoproliferative neoplasms. We employ a progressive shrinking approach, which integrates a comprehensive pruning technique across multiple dimensions, including width, depth, resolution, and kernel size, to train our lightweight model. The proposed model achieves rapid identification of acute lymphoblastic leukemia, chronic lymphocytic leukemia, and other bone marrow cell types with an accuracy of 92.51% and a throughput of 111 slides per second, while comprising only 6.4 million parameters. This model significantly contributes to leukemia diagnosis, particularly in the rapid and accurate identification of lymphatic system diseases, and provides potential opportunities to enhance the efficiency and accuracy of medical experts in the diagnosis and treatment of lymphocytic leukemia

Abstract:Blood is composed of white blood cells, red blood cells, and platelets. Segmentation of the blood smear cells and extraction of features of the cells is essential in the field of medicine. Acute lymphoblastic leukemia is a form of blood cancer caused due to the abnormal increase in the production of immature white blood cells in the bone marrow. It mostly affects the children below 5 years and adults above 50 years of age. Due to the late diagnosis and cost of the devices used for the determination, the mortality rate has increased drastically. Flow cytometry technique that performs automated counting fails to identify the abnormal cells. Manual recount performed using hemocytometer are prone to errors and are imprecise. The proposed work aims to survey different computer-aided system techniques used to segment the blood smear image. The primary objective here is to derive knowledge from the different methodologies used for extracting features from white blood cells and develop a system that would accurately segment the blood smear image by overcoming the drawbacks of the previous works. The objective mentioned above is achieved in two ways. Firstly, a novel algorithm is developed to segment the nucleus and cytoplasm of white blood cell. Secondly, a model is built to extract the features and train the model. The different supervised classifiers are compared, and the one with the highest accuracy is used for the classification. Six hundred images are used in the experimentation. InfoGainAttributeEval and the Ranker Search method are used to achieve the feature selection which in turn helps in improvising the classifier performance. The result shows the classification of the acute lymphoblastic leukemia into its three respective categories namely: ALL-L1, ALL-L2, ALL-L3. The model can differentiate between a normal peripheral blood smear and an abnormal blood smear. The extracted feature values of a cancerous cell and a normal cell are also shown. The performance of the model is evaluated using the test images stained with various stains. The proposed algorithm achieved an overall accuracy of 98.6%. The promising results show that it can be used as a diagnostic tool by the pathologists.Graphical abstract

Artificial intelligence-assisted diagnosis of hematologic diseases based on bone marrow smears using deep neural networks

Abstract:Background and objectives: The morphological examination of bone marrow (BM) cells is essential in both diagnosing and treating various hematologic diseases. However, it is still done manually with a heavy workload. An artificial intelligence-assisted diagnosis support system of BM cells is highly required to reduce the workloads of examiners and improve the reproducibility of the results. Methods: In this paper, we proposed an artificial intelligence-assisted diagnosis support system of morphological examination based on bone marrow smears including cells detection, classification and prediction of leukemia types. For cell detection, we trained the novel YOLOX-s model to locate cells precisely and obtain single cell images. For cell classification, we regarded it as a fine- grained classification task and proposed a novel architecture called MLFL-Net utilizing multi-level features. Furthermore, we predicted the leukemia types on a dataset including 40 normal people (BM transplantation donors) and 40 patients of different kinds of acute leukemia according to the World Health Organization (WHO) standard. Results: We constructed a large-scale data set of 11,788 fully-annotated micrographs from 728 smears and 131,300 expert-annotated single cell images. With the data set, the detection model achieved 0.9797 AUC and 4.33% box placement error. For cell classification, the total accuracy of our proposed MLFL-Net reached 89.53% which outperformed all the other related models in identifying cell categories. In the meantime, we took acute leukemia as an example to explore the leukemia types prediction procedure of hematological disease. It generated the same diagnostic prediction as the experts gave for 92.5 percent of the cohort. Conclusion: This Artificial Intelligence-assisted system can be implemented to aid in clinical decision making and accelerate diagnosis. The method will contribute to promote the intelligence and modernization of BM cytomorphology, which has vital significance of the development of the medical career.
An ensemble-acute lymphoblastic leukemia model for acute lymphoblastic leukemia image classification

Abstract:The timely diagnosis of acute lymphoblastic leukemia (ALL) is of paramount importance for enhancing the treatment efficacy and the survival rates of patients. In this study, we seek to introduce an ensemble-ALL model for the image classification of ALL, with the goal of enhancing early diagnostic capabilities and streamlining the diagnostic and treatment processes for medical practitioners. In this study, a publicly available dataset is partitioned into training, validation, and test sets. A diverse set of convolutional neural networks, including InceptionV3, EfficientNetB4, ResNet50, CONV_POOL-CNN, ALL-CNN, Network in Network, and AlexNet, are employed for training. The top-performing four individual models are meticulously chosen and integrated with the squeeze-and-excitation (SE) module. Furthermore, the two most effective SE-embedded models are harmoniously combined to create the proposed ensemble-ALL model. This model leverages the Bayesian optimization algorithm to enhance its performance. The proposed ensemble-ALL model attains remarkable accuracy, precision, recall, F1-score, and kappa scores, registering at 96.26, 96.26, 96.26, 96.25, and 91.36%, respectively. These results surpass the benchmarks set by state-of-the-art studies in the realm of ALL image classification. This model represents a valuable contribution to the field of medical image recognition, particularly in the diagnosis of acute lymphoblastic leukemia, and it offers the potential to enhance the efficiency and accuracy of medical professionals in the diagnostic and treatment processes.

Abstract:Acute myeloid leukemia (AML) is a deadly hematological malignancy. Cellular morphology detection of bone marrow smears based on the French-American-British (FAB) classification system remains an essential criterion in the diagnosis of hematological malignancies. However, the diagnosis and discrimination of distinct FAB subtypes of AML obtained from bone marrow smear images are tedious and time-consuming. In addition, there is considerable variation within and among pathologists, particularly in rural areas, where pathologists may not have relevant expertise. Here, we established a comprehensive database encompassing 8245 bone marrow smear images from 651 patients based on a retrospective dual-center study between 2010 and 2021 for the purpose of training and testing. Furthermore, we developed AMLnet, a deep-learning pipeline based on bone marrow smear images, that can discriminate not only between AML patients and healthy individuals but also accurately identify various AML subtypes. AMLnet achieved an AUC of 0.885 at the image level and 0.921 at the patient level in distinguishing nine AML subtypes on the test dataset. Furthermore, AMLnet outperformed junior human experts and was comparable to senior experts on the test dataset at the patient level. Finally, we provided an interactive demo website to visualize the saliency maps and the results of AMLnet for aiding pathologists' diagnosis. Collectively, AMLnet has the potential to serve as a fast prescreening and decision support tool for cytomorphological pathologists, especially in areas where pathologists are overburdened by medical demands as well as in rural areas where medical resources are scarce.
Feature Fusion Based Ensemble of Deep Networks for Acute Leukemia Diagnosis Using Microscopic Smear Images

Abstract:A type of blood malignancies known as leukemia leads to elevated quantities of abnormally formed blood cells and often originates in the bone marrow. An abrupt rise in the quantity of immature blood cells is one of the distinctive characteristics of acute leukemia, that predominantly affects both children and adults. The possibility exists to drastically lower the death rate associated with acute leukemia through early detection and diagnosis. A significant and time-consuming task for the early detection of acute leukemia is microscopic examination of blood cells. In this paper, a two-staged deep learning-based computer-aided diagnosis system regarding microscopic blood smear images is proposed to assist hematologists and improve the diagnosis accuracy of acute leukemia in which for the feature fusion-based ensemble, two deep neural network branches adopting pretrained, fine-tuned EfficientNetB7 and MobileNetV3Large architectures were employed, and feature maps generated from those branches were fused and fed to the second stage of the architecture to achieve the final result. Additional dropout layers and ReLu activation were employed in the architecture to speed up the network, and compound scaling, bottleneck, and fusion architectures enhanced the overall performance. The ALLIDB1, ALLIDB2, and ASH databases were incorporated to evaluate the performances of the proposed method. The experimental findings demonstrated that the proposed approach detected acute leukemia (ALL, AML, Healthy) with an accuracy of 99.3%, F1-score of 99.3%, and AUC score of 0.997. Whereas in detecting acute lymphocytic leukemia (ALL, Healthy) and acute myeloid leukemia (AML, Healthy), the accuracies reached 100.0% and 99.8%, respectively. Thus, we believe that clinics can adopt our proposed architecture for quick and automated diagnosis.

Abstract:Recent advancements in artificial intelligence (AI) have led to numerous medical discoveries. The field of computer vision (CV) for medical diagnosis has received particular attention. Using images of peripheral blood (PB) smears, CV has been utilized in hematology to detect acute leukemia (AL). Significant research has been undertaken in the area of AL diagnosis automation in order to deliver an accurate diagnosis. This study addresses the morphological classification of atypical white blood cells (WBCs), including immature WBCs and atypical lymphocytes, in acute myeloid leukemia (AML), as observed in peripheral blood (PB) smear images. The purpose of this work is to build a classification model for atypical AML WBCs based on their distinctive features. Using a hybrid model based on geometric transformation (GT) and a deep convolutional autoencoder (DCAE), this work provides a novel technique in the field of AI for resolving the issue of imbalanced distribution of WBCs in blood samples, nicknamed the "GT-DCAE WBC augmentation model". In addition, to extract context-free atypical WBC features, this study develops a stable learning paradigm by incorporating WBC segmentation into deep learning. In order to classify atypical WBCs into eight distinct subgroups, a hybrid multiclassification model termed the "two-stage DCAE-CNN atypical WBC classification model" (DCAE-CNN) was developed. The model achieved an average accuracy of 97%, a sensitivity of 97%, and a precision of 98%. Overall and by class, the model's discriminating abilities were exceptional, with an AUC of 99.7% and a class-wise range of 80% to 100%.

ALLNet: A Hybrid Convolutional Neural Network to Improve Diagnosis of Acute Lymphocytic Leukemia (ALL) in White Blood Cells

Abstract:Due to morphological similarity at the microscopic level, making an accurate and time-sensitive distinction between blood cells affected by Acute Lymphocytic Leukemia (ALL) and their healthy counterparts calls for the usage of machine learning architectures. However, three of the most common models, VGG, ResNet, and Inception, each come with their own set of flaws with room for improvement which demands the need for a superior model. ALLNet, the proposed hybrid convolutional neural network architecture, consists of a combination of the VGG, ResNet, and Inception models. The ALL Challenge dataset of ISBI 2019 (available here) contains 10,691 images of white blood cells which were used to train and test the models. 7,272 of the images in the dataset are of cells with ALL and 3,419 of them are of healthy cells. Of the images, 60% were used to train the model, 20% were used for the cross-validation set, and 20% were used for the test set. ALLNet outperformed the VGG, ResNet, and the Inception models across the board, achieving an accuracy of 92.6567%, a sensitivity of 95.5304%, a specificity of 85.9155%, an AUC score of 0.966347, and an F1 score of 0.94803 in the cross-validation set. In the test set, ALLNet achieved an accuracy of 92.0991%, a sensitivity of 96.5446%, a specificity of 82.8035%, an AUC score of 0.959972, and an F1 score of 0.942963. The utilization of ALLNet in the clinical workspace can better treat the thousands of people suffering from ALL across the world, many of whom are children.

Abstract:Leukemia is one of the most dangerous types of malignancies affecting the bone marrow or blood in all age groups, both in children and adults. The most dangerous and deadly type of leukemia is acute lymphoblastic leukemia (ALL). It is diagnosed by hematologists and experts in blood and bone marrow samples using a high-quality microscope with a magnifying lens. Manual diagnosis, however, is considered slow and is limited by the differing opinions of experts and other factors. Thus, this work aimed to develop diagnostic systems for two Acute Lymphoblastic Leukemia Image Databases (ALL_IDB1 and ALL_IDB2) for the early detection of leukemia. All images were optimized before being introduced to the systems by two overlapping filters: the average and Laplacian filters. This study consists of three proposed systems as follows: the first consists of the artificial neural network (ANN), feed forward neural network (FFNN), and support vector machine (SVM), all of which are based on hybrid features extracted using Local Binary Pattern (LBP), Gray Level Co-occurrence Matrix (GLCM) and Fuzzy Color Histogram (FCH) methods. Both ANN and FFNN reached an accuracy of 100%, while SVM reached an accuracy of 98.11%. The second proposed system consists of the convolutional neural network (CNN) models: AlexNet, GoogleNet, and ResNet-18, based on the transfer learning method, in which deep feature maps were extracted and classified with high accuracy. All the models obtained promising results for the early detection of leukemia in both datasets, with an accuracy of 100% for the AlexNet, GoogleNet, and ResNet-18 models. The third proposed system consists of hybrid CNN–SVM technologies, consisting of two blocks: CNN models for extracting feature maps and the SVM algorithm for classifying feature maps. All the hybrid systems achieved promising results, with AlexNet + SVM achieving 100% accuracy, Goog-LeNet + SVM achieving 98.1% accuracy, and ResNet-18 + SVM achieving 100% accuracy.

Abstract:Leukemia (blood cancer) is an unusual spread of White Blood Cells or Leukocytes (WBCs) in the bone marrow and blood. Pathologists can diagnose leukemia by looking at a person's blood sample under a microscope. They identify and categorize leukemia by counting various blood cells and morphological features. This technique is time-consuming for the prediction of leukemia. The pathologist's professional skills and experiences may be affecting this procedure, too. In computer vision, traditional machine learning and deep learning techniques are practical roadmaps that increase the accuracy and speed in diagnosing and classifying medical images such as microscopic blood cells. This paper provides a comprehensive analysis of the detection and classification of acute leukemia and WBCs in the microscopic blood cells. First, we have divided the previous works into six categories based on the output of the models. Then, we describe various steps of detection and classification of acute leukemia and WBCs, including Data Augmentation, Preprocessing, Segmentation, Feature Extraction, Feature Selection (Reduction), Classification, and focus on classification step in the methods. Finally, we divide automated detection and classification of acute leukemia and WBCs into three categories, including traditional, Deep Neural Network (DNN), and mixture (traditional and DNN) methods based on the type of classifier in the classification step and analyze them. The results of this study show that in the diagnosis and classification of acute leukemia and WBCs, the Support Vector Machine (SVM) classifier in traditional machine learning models and Convolutional Neural Network (CNN) classifier in deep learning models have widely employed. The performance metrics of the models that use these classifiers compared to the others model are higher.
