Abstract:This aim of this study was to evaluate the whitening efficacy of the water extract from bamboo shavings (WEBS) in malignant melanoma cells (B-16) of mice. The safety of WEBS was evaluated by the acute oral toxicity test, the repeated skin irritation test and the acute eye irritation test was also evaluated. WEBS showed strong inhibitory effects against the activity of tyrosinase in B-16 cells in a dose dependent manner, and was more potent than arbutin. The melanin content was significantly inhibited by WEBS and the cytotoxicity of WEBS was lower than that of arbutin and hydroquinone. WEBS inhibited mushroom tyrosinase and the maximum was 65.05% at 16 mg/ml with an IC50 of 6 mg/ml. The LD50 was larger than 5000 mg/kg body weight and WEBS was found to be non-toxic and non-irritating. Abstract:Hyperpigmentation is an increased deposition of melanin in the skin. The effects of ethanolic extract from the brown alga Sargassum serratifolium (ESS) on melanogenic protein expressions in B16F10 mouse melanoma cells were examined to elucidate its hypopigmenting properties. ESS remarkably reduced melanin synthesis in α-melanocyte stimulating hormone (α-MSH)-stimulated B16F10 cells. Western blot analysis revealed that ESS attenuated the expression of melanogenic enzymes, tyrosinase, and tyrosinase-related protein 1, by cyclic adenosine monophosphate (cAMP)-responsive element-binding protein (CREB)-mediated downregulation of microphthalmia-associated transcription factor (MITF). ESS inhibited accumulation of cellular cAMP that leads to inhibition of CREB phosphorylation. Moreover, ESS activated extracellular signal-regulated kinase (ERK), but not Akt and other mitogen-activated protein kinases, which is responsible for posttranslational downregulation of MITF. Therefore, ESS attenuated α-MSH-stimulated hyperpigmentation in B16F10 cells through modulation of CREB/ERK signaling pathways. Three antimelanogenic compounds such as sargahydroquinoic acid, sargachromenol, and sargaquinoic acid were identified in ESS depending on inhibition of melanin synthesis. These findings suggest that ESS could be a potential agent in the treatment of hyperpigmentation-related skin disorders.

Abstract:The present study investigated the photoprotective effect of the ultrasonic-assisted ethanol extract (USHE) from Sargassum horneri, a brown seaweed containing fucosterol (6.22 ± 0.06 mg/g), sulfoquinovosyl glycerolipids (C23H43O11S, C25H45O11S, C25H47O11S, C27H49O11S), and polyphenols, against oxidative damage in ultraviolet B (UVB)-exposed HaCaT keratinocytes. USHE indicated antioxidant activity in ferric-reducing antioxidant power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging. After screening experiments, 15.6, 31.3, and 62.5 μg/mL concentrations of USHE and ascorbic acid as positive control were selected to be used throughout the investigation. USHE increased cell viability by markedly reducing the production of intracellular reactive oxygen species (ROS) in UVB-exposed HaCaT keratinocytes. Additionally, USHE reduced the apoptosis and sub-G1 cell population and increased the mitochondrial membrane potential. Moreover, USHE modulated the protein expression levels of anti-apoptotic molecules (Bcl-xL, Bcl-2, and PARP) and pro-apoptotic molecules (Bax, cleaved caspase-3, p53, cleaved PARP, and cytochrome C). This modulation accorded with the upregulation of cytosolic heme oxygenase (HO)-1, NAD(P)H quinone oxidoreductase 1 (NQO 1), and nuclear factor erythroid-2-related factor 2 (Nrf2), collectively known as components of the antioxidant system. These findings suggest that USHE has a photoprotective effect on UVB-exposed HaCaT keratinocytes and can be utilized to develop cosmeceuticals for UVB protection.

Inhibitory Effects of (-)-epigallocatechin-3-gallate on Melanogenesis in Ultraviolet A-Induced B16 Murine Melanoma Cell

Abstract:Purpose: To investigate the anti-melanogenesis effect of green tea compound, (-)-epigallocatechin-3-gallate (EGCG), on B16 murine melanoma cell irradiated by ultraviolet A (UVA) in the search for natural skin-lightening alternative agents.Methods: B16 murine melanoma cells by UVA (9.0 J/cm(2)) for 0 to 32 min and then incubated in Dulbecco's Modified Eagle's Medium (DMEM) with EGCG (0-200 mu g/mL) for 2 days. Cell viability was determined by MTT method and cell protein was quantified using a PA102 Bradford protein assay kit. Activity of tyrosinase (TRY) was determined based on the oxidation rate of 3,4-dihydroxy phenylalanine (DOPA). The ultra-structure of the melanosomes was observed by transmission electron microscopy (TEM).Results: TRY activity and melanin concentration were increased to 146.70 +/- 10.28 % (p < 0.05) and 157.06 +/- 6.37 % (p < 0.05), respectively, by 9.0 J/cm(2) UVA irradiation for 8 min, compared to blank control without UV A and EGCG. EGCG inhibited the UV A induced increase in TRY activity and melanin level, and the optimum concentration of EGCG was 25 mu g/mL. TRY activity and melanin concentration were decreased to 64.71 +/- 4.41 (p < 0.05) and 86.24 +/- 5.15 % (p < 0.05), respectively, compared to blank (control) which was neither treated by UVA nor by EGCG. TEM showed that UVA induced the formation of melanosomes while EGCG inhibited UVA-induced melanosome maturation.Conclusion: EGCG inhibits UVA-induced melanogenesis via suppression of TRY activity and melanosome maturation and is thus a potential alternative to melanogenesis inhibitor.

Abstract:Introduction: Melanogenesis, the process responsible for producing melanin pigment, hinges on the activation of the amino acid L-tyrosine by the pivotal enzyme tyrosinase. In cases of hyperpigmentation induced by ultraviolet radiation, there's a surge in both tyrosinase enzyme levels and melanin production. Sargassum polycystum is currently under active investigation for its potential in this regard. This review aims to delve deeper into the properties of Sargassum polycystum and its impact on tyrosinase expression and melanin production. Literature Review: Sargassum polycystum, a brown seaweed commonly found in coastal regions with abundant coral reefs, harbors active compounds like flavonoids, fucoidan, fucoxanthin, and phloroglucinol. Fucoidan, an extract from brown seaweed, exhibits promising anti-inflammatory and antioxidant properties. Recent studies have revealed its ability to impede the melanogenesis process. Specifically, it was observed that fucoidan from brown seaweed extract could dampen α-MSH stimulation while enhancing the activation of the ERK-MAPK pathway, thereby acting as a negative regulator for melanogenesis. Fucoxanthin, another component, showcases antioxidant capabilities. Furthermore, phloroglucinol, found in Sargassum polycystum, exerts tyrosinase inhibitory effects by chelating copper within the enzyme. Flavonoids like catechin, hesperidin, kaempferol, and baicalein in Sargassum polycystum demonstrate inhibitory effects on melanogenesis activation pathways, including suppression of the Erk1/2 pathway, MiTF, and direct inhibition of tyrosinase. Consequently, this leads to reduced tyrosinase expression and melanin production, ultimately attenuating the melanogenesis process. Conclusion: Sargassum polycystum emerges as a promising candidate for combating hyperpigmentation induced by exposure to ultraviolet B radiation.

Abstract:The 2, 3, 5, 4'-tetrahydroxystilbene-2-O-beta-D-glucoside (THSG), a water-soluble active component extracted from dried tuber root of Polygonum multiflorum, has been found to induce pigmentation in B16 cells, but the details of the underlying mechanism remain unknown. The present study was conducted to investigate the mechanism of stimulatory effect of THSG on melanogenesis using B16F1 melanoma cells.Several experiments were performed in B16F1 melanoma cells. We studied melanin content, tyrosinase activity, cell viability, and performed reverse transcription polymerase chain reaction and Western blots for proteins involved in melanogenesis.THSG increased the melanin content and tyrosinase activity in a concentration-dependent manner and treatment with 10 microg/ml THSG enhanced the expression of tyrosinase time-dependently in B16 cells. We then investigated whether THSG influences the expression of microphthalmia-associated transcription factor (MITF), which is required for tyrosinase expression. THSG was found to induce sustained MITF up-regulation and cAMP response element (CRE) binding protein (CREB) activation, suggesting that THSG-mediated MITF activation may be cAMP dependent. Furthermore, Western blot analysis revealed that THSG elevated the level of phosphorylation of p38 mitogen-activated protein kinase (p38 MAPK) significantly at 1-6 h; a p38 MAPK inhibitor, SB203580, almost completely attenuated the THSG-mediated up-regulation of melanin synthesis and induction of MITF and tyrosinase expression.THSG exerts its stimulatory effect on melanogenesis by MAP kinase activation and MITF-induction of tyrosinase.

Abstract:Hyperpigmentation is a type of pigmentary disorder induced by overexpression of melanin content activated severe esthetic problems as melasma, freckle, ephelides, lentigo and other forms on human skin. Several whitening agents have restricted use because of their side effects or stability such as kojic acid, ascorbic acid and hydroquinone can act as cytotoxic substance which associated to dermatitis and skin cancer. To find for the safe substance, this study aimed to find for the ability of several components in Sucrier banana peel (SBP) extracts to inhibit melanogenesis process through p38 signaling pathway in B16F10 mouse melanoma cells. Tyrosinase activity and the cellular melanin content were dose dependent manner decreasing after SBP treatment. Furthermore, SBP decreased the expression of melanogenesis relate protein as microphthalmia-associated transcription factor (MITF) and tyrosinase protein after 24 hours incubation with α-melanocyte stimulating hormones (MSH) stimulating. The findings demonstrated that SBP contained an effective agent for hyperpigmentation inhibitor through p38 signaling pathways without any effect to ERK pathway, and subsequent down-regulate MITF expression and tyrosinase enzyme family production.

Abstract:Cutaneous pigmentation is an important phenotypic trait whose regulation, despite recent advances, has yet to be completely elucidated. Melanogenesis, a physiological process of melanin production, is imperative for organism survival as it provides protection against the environmental insults that majorly involve sunlight-induced skin photodamage. However, immoderate melanin synthesis can cause pigmentation disorders associated with a psychosocial impact. In this study, the hypopigmentation effect of (2-methylbutyryl)shikonin, a natural product present in the root extract of Lithospermum erythrorhizon, and the underlying mechanisms responsible for the inhibition of melanin synthesis in α-MSH-stimulated B16F10 cells and C57BL/6J mice was studied. Non-cytotoxic concentrations of (2-methylbutyryl)shikonin significantly repressed cellular tyrosinase activity and melanin synthesis in both in vitro and in vivo models (C57BL/6J mice). (2-Methylbutyryl)shikonin remarkably abolished the protein expression of MITF, tyrosinase, tyrosinase-related protein 1, and tyrosinase-related protein 2, thereby blocking the production of pigment melanin via modulating the phosphorylation status of MAPK proteins, viz., ERK1/2 and p38. In addition, specific inhibition of ERK1/2 attenuated the inhibitory effects of (2-methylbutyryl)shikonin on melanin synthesis, whereas selective inhibition of p38 augmented the inhibitory effect of BSHK on melanin synthesis. Moreover, topical application of (2-methylbutyryl)shikonin on C57BL/6J mouse tails remarkably induced tail depigmentation. In conclusion, with these findings, we, for the first time, report the hypopigmentation effect of (2-methylbutyryl)shikonin via inhibition of cellular tyrosinase enzyme activity, subsequently ameliorating the melanin production, thereby indicating that (2-methylbutyryl)shikonin is a potential natural therapy for hyperpigmentation disorders.

Abstract:The medical records of 84 postmenopausal women treated with oral norethisterone acetate (NTA) for advanced breast cancer were retrospectively analyzed. Treatment was devoid of significant toxicity. Twenty-one patients were not evaluable for treatment response either because of insufficient data or inadequate treatment trial. Complete plus partial response was obtained in 21 (33.3 %) of the 63 evaluable patients, with a median duration of 10 months. Disease stabilization was observed in 16 (25.4 %) patients for a median duration of 5 months, while 26 patients (41.3 %) showed progressive disease while on treatment. The best response was observed in women with dominant soft part disease and an age over 70 (CR+PR 48 %). The literature on norethisterone acetate is reviewed and compared with present results. The role of progestational agents in the treatment of advanced mammary carcinoma is discussed.
Anti-melanogenic effects of Aster spathulifolius extract in UVB-exposed C57BL/6J mice and B16F10 melanoma cells through the regulation of MAPK/ERK and AKT/GSK3β signalling

Abstract:Objectives: Pharmacological studies of Aster spathulifolius Maxim(AS) have demonstrated its anti-allergy, anti-viral and anti-obesity effects, however, its anti-melanogenic effects is still unclear. In this study, the effects of AS extract (ASE) on the inhibition of melanin synthesis were investigated in vitro and in vivo. Methods: To perform this study, the contents of melanin and tyrosinase activity were analysed in B16F10 melanoma cells. Western blotting was carried out to determine the underlyling mechanism. Additionally, we investigated the effect of this extract on hyperpigmentation in C57bL/6J mice induced by 3, 6 and 9 weeks of UVB irradiation. Key findings: AS extract led to reduced melanin synthesis through the regulation of MITF and its downstream signals. Furthermore, ASE increased the phosphorylation of MAPK/ERK and Akt/GSK3β signalling pathway components. In vivo study, hypopigmentation effects were also observed. The melanocyte activity and the distribution of melanin granules were decreased in UVB-irradiated mice treated with ASE. Conclusions: These results suggest that the ASE may be promising as an active anti-melanogenic component, and further investigations should be performed regarding its potential as a whitening agent in the field of cosmetics.
Effect of hydrosol obtained from Camellia japonica branch on α-MSH-induced melanin and tyrosinase activity in B16F10 melanoma cells

Abstract:Hydrosols that originate from various aromatic botanical sources in natural or organic settings contain a spectrum of fragrant compounds, which can be extracted from leaves, stems, peels, flowers, and roots. These compounds are known to exhibit diverse medicinal properties. However, there is limited research on hydrosols extracted from Camellia japonica branches, specifically in terms of their potential to inhibit tyrosinase. Hence, the aim of this study was to investigate the chemical composition of these hydrosols and their effects on inhibiting tyrosinase. Hexamethyl-cyclotrisiloxane (38.1%) and vanillin (25.3%) were identified as the primary constituents in the hydrosol through gas chromatography–mass spectrometry analysis. The inhibitory effects of the hydrosol, in comparison with the positive control arbutin, were evaluated against mushroom tyrosinase, revealing significant tyrosinase inhibitory properties for both the hydrosol and arbutin. Furthermore, in the presence of α-melanocyte-stimulating hormone, the hydrosol notably diminished melanogenesis, resulting in a substantial reduction in melanin production. Genetic and protein analyses were conducted to uncover the mechanisms behind the hydrosol’s inhibition of tyrosinase and reduction of melanin. The results suggest that the hydrosol may effectively shield melanocytes from detrimental factors associated with tyrosinase-related proteins. The anti-tyrosinase activity of the hydrosol indicates its potential for promoting skin lightening.

Abstract:Melanin production is an important process that prevents the host skin from harmful ultraviolet radiation; however, an overproduction of melanin results in skin diseases. In the present study, we determined the antioxidative and anti-melanogenic activities of polyphenol- and flavonoid-enriched rice seed extracts in melan-a cells. The polyphenol and flavonoid content of Hopum (HP) and Sebok (SB) rice seed extracts was measured. The antioxidant capacity was determined using the ABTS radical scavenging method. SB contained high amounts of polyphenols and flavonoids, which significantly increased antioxidative activity compared with HP. Various concentrations of these extracts were evaluated in a cytotoxicity using melan-a cells. At 100 μg/mL, there was no significant difference for all treatments compared with untreated cells. Therefore, 100 μg/mL was selected as a concentration for the further experiments. SB significantly suppressed the phosphorylation/activation of p-38 MAPK, increased the expression of phosphorylated ERK 1/2 and Akt, and downregulated the microphthalmia-associated transcription factor (MITF). This resulted in decreased levels of tyrosinase and tyrosinase-related protein-1 and -2. These results indicate the potential of polyphenol- and flavonoid-enriched rice seed as a treatment for hyperpigmentation.

Abstract:SIR, Standard therapy for discoid lupus erythematosus (DLE) consists of potent topical steroids, with the attendant risk of steroid dermopathy. We report a patient who responded to sulphasalazine on two occasions with relapse on withdrawal of this systemic therapy. A 57-year-old woman was diagnosed as having DLE 18 years previously. The facial eruption, characterized by red, scaly plaques that were associated with scarring, could only be controlled for 12 years by the daily application of a potent steroid, occasional intralesional steroids and a sun screen. There was thinning of the skin and telangectasia. A presumptive diagnosis of ulcerative colitis was made 6 years ago when she was abroad and she was treated with sulphasalazine 500 mg four times a day. Within 2 months she noted an improvement of her facial rash and was able to reduce the frequency of application of the topical steroid to twice a week. After 18 months, in the absence of proven colitis, her sulphasalazine was stopped, without a relapse of her bowel symptoms. However, there was a marked deterioration of her DLE, resulting in increased usage of topical steroid after 3 months. Following 2 years of poor control with topical therapy, the patient was recommenced on sulphasalazine 500 mg twice a day, with some improvement. Complete control was achieved after a further year when the dose was increased to 500 mg four times a day. The patient has remained on this dose of sulphasalazine for the past year, requiring only very occasional topical steroid therapy and has suffered no side-effects.

Abstract:Background The pursuit for safe and efficacious skin-whitening agents has prompted a dedicated exploration of plant-derived compounds. Notably, Tagetes erecta L. flowers has been used as a medicinal extract and possessed in vitro mushroom tyrosinase activity. However, whether polyphenol-enriched fraction extracted from T. erecta L. flowers (TE) regulates melanogenesis within cellular and animal models has not yet been investigated. Purpose This study aimed to investigate the TE as a prospective inhibitor of melanogenesis. Methods Through advanced UPLC-QTof/MS analysis, the components of TE were analyzed. Anti-melanogenic effects of TE were evaluated in α-melanocyte-stimulating hormone (α-MSH)-stimulated B16F10 melanoma cells by measuring cell viability assay, extracellular and intracellular melanin biosynthesis, cyclic adenosine monophosphate (cAMP) production, and melanogenesis-related gene and protein expression. Zebrafish larvae were employed for in vivo studies, assessing both heart rate and melanogenesis. Furthermore, molecular docking analyses were employed to predict the interaction between TE components and the melanocortin 1 receptor (MC1R). Direct binding activity of TE components to MC1R was compared with [Nle 4 , d -Phe 7 ]-MSH (NDP-MSH). Results TE was found to contain significant phenolic compounds such as patulitrin, quercetagetin, kaempferol, patuletin, and isorhamnetin. This study revealed that TE effectively inhibits melanin biosynthesis in both in vitro and in vivo models. This inhibition was attributed to interference of TE with the cAMP–cAMP response element-binding protein (CREB)–microphthalmia-associated transcription factor (MITF)–tyrosinase pathway, which plays a pivotal role in regulating melanogenesis. Importantly, TE exhibited the remarkable ability to curtail α-MSH-induced melanogenesis in zebrafish larvae without impacting heart rates. Molecular docking analyses predicted that the components of TE possibly interact with the melanocortin 1 receptor, suggesting their role as potential inhibitors of melanin biosynthesis. However, through the direct binding activity compared with NDP-MSH, any TE components did not directly bind to MC1R, suggesting that TE inhibits α-MSH-induced melanogenesis by inhibiting the cAMP-mediated intracellular signaling pathway. The assessment of anti-melanogenic activity, conducted both in vitro and in vivo, revealed that patulitrin and patuletin exhibited significant inhibitory effects on melanin formation, highlighting their potency as major contributors. Discussion This investigation demonstrated the considerable potential of TE as a natural remedy endowed with remarkable anti-melanogenic properties. The demonstrated capacity of TE to attenuate melanin production by modulating the cAMP–CREB–MITF–tyrosinase pathway underscores its central role in management of disorders associated with excessive pigmentation. Importantly, the implications of these findings extend to the cosmetics industry, where TE emerges as a prospective and valuable ingredient for the formulation of skin-whitening products. The elucidated interactions between TE components and MC1R not only provide insight into a potential mechanism of action but also elevate the significance of this study. In summary, this study not only contributes to our comprehension of pigmentation-related conditions but also firmly establishes TE as a secure and natural strategy for the regulation of melanin production. The innovative aspects of TE propel it into the forefront of potential interventions, marking a noteworthy advancement in the pursuit of effective and safe solutions for pigmentation disorders.

Mechanistic Insights into the Stimulatory Effect of Melanogenesis of 4-Methylcoumarin Derivatives in B16F10 Melanoma Cells

Abstract:Vitiligo is a skin condition characterized by the loss of pigment, resulting in white patches on various parts of the body. It occurs when melanocytes, the cells that are responsible for producing skin pigment, are destroyed or stop functioning. This study aimed to investigate the melanogenic potential of various 4-methylcoumarin (4MC) derivatives, including 6-methoxy-4-methylcoumarin (6M-4MC), 7-methoxy-4-methylcoumarin (7M-4MC), 7-amino-4-methylcoumarin (7A-4MC), 6,7-dihydroxy-4-methylcoumarin (6,7DH-4MC), 7,8-dihydroxy-4-methylcoumarin (7,8DH-4MC), and 6,7-dimethoxy-4-methylcoumarin (6,7DM-4MC), in B16F10 melanoma cells. Our findings revealed that, while 4MC, 7A-4MC, 6,7DH-4MC, and 7,8DH-4MC did not exhibit any effect on melanin production, significant stimulation of melanogenesis was observed with 6M-4MC, 7M-4MC, and 6,7DM-4MC, with 6M-4MC demonstrating the most pronounced effect. 6M-4MC significantly stimulated melanin production and tyrosinase activity in a concentration-dependent manner in B16F10 cells. A Western blot analysis revealed that 6M-4MC increased the expression levels of microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein-1 (TRP-1), and tyrosinase-related protein-2 (TRP-2). Further mechanistic studies showed that 6M-4MC inhibited extracellular signal-regulated kinase (ERK) and protein kinase B (AKT), which led to the upregulation of MITF and TRP proteins and subsequent activation of melanin synthesis. Additionally, 6M-4MC activated GSK3β phosphorylation, reduced β-catenin phosphorylation, and stimulated melanogenesis via the GSK3β/β-catenin pathway. Moreover, a primary skin irritation test was conducted on the upper backs of 32 healthy female volunteers to assess the potential irritation or sensitization from 6M-4MC when applied topically at concentrations of 50 μM and 100 μM. The test results showed no adverse effects on the skin. Collectively, these findings suggest that 6M-4MC may be a promising pigmentation stimulator for use in cosmetics and in the medical treatment of hypopigmentation disorders, particularly in the treatment of skin conditions such as vitiligo.

Tyrosinase inhibitory activity of Sargassum fusiforme and characterisation of bioactive compounds

Abstract:Introduction: Sargassum fusiforme (Harvey) Setchell, also known as Tot (in Korean) and Hijiki (in Japanese), is widely consumed in Korea, Japan, and China due to its health promoting properties. However, the bioactive component behind the biological activity is still unknown. Objectives: We aimed to optimise the extraction conditions for achieving maximum tyrosinase inhibition activity by using two sophisticated statistical tools, that is, response surface methodology (RSM) and artificial neural network (ANN). Moreover, high-resolution mass spectrometry (HRMS) was used to tentatively identify the components, which are then further studied for molecular docking study using 2Y9X protein. Methodology: RSM central composite design was used to conduct extraction using microwave equipment, which was then compared to ANN. Electrospray ionisation tandem mass spectrometry (ESI-MS/MS) was used to tentatively identify bioactive components, which were then docked to the 2Y9X protein using AutoDock Vina and MolDock software. Results: Maximum tyrosinase inhibition activity of 79.530% was achieved under optimised conditions of time: 3.27 min, temperature: 128.885°C, ethanol concentration: 42.13%, and microwave intensity: 577.84 W. Furthermore, 48 bioactive compounds were tentatively identified in optimised Sargassum fusiforme (OSF) extract, and among them, seven phenolics, five flavonoids, five lignans, six terpenes, and five sulfolipids and phospholipids were putatively reported for the first time in Sargassum fusiforme. Among 48 bioactive components, trifuhalol-A, diphlorethohydroxycarmalol, glycyrrhizin, and arctigenin exhibited higher binding energies for 2Y9X. Conclusion: Taken together, these findings suggest that OSF extract can be used as an effective skin-whitening source on a commercial level and could be used in topical formulations by replacing conventional drugs.
Inhibitory Effects of Salidroside and Paeonol on Tyrosinase Activity and Melanin Synthesis in Mouse B16F10 Melanoma Cells and Ultraviolet B-induced Pigmentation in Guinea Pig Skin

Abstract:Salidroside, the major active component of Rhodiola rosea, a herb with antioxidant, free radical scavenging and tyrosinase inhibitory effects, has been recently reported in protecting the kerationcytes from the UV radiation, suggesting the potential of this component in depigmentation. Paeonol is isolated from Moutan Cortex Radicis with anti-inflammation/microbial activities, was reported to induce the down-regulation of microphthalmia-associated transcription factor and subsequently tyrosinase. To testify the potential of these compounds as melanin formation inhibitors for hyperpigmentation therapy, the influence of salidroside and paeonol on pigmentation was investigated. With arbutin as a positive control, salidroside and paeonol were evaluated for their inhibitory effect on the cell viability, tyrosinase activity and melanin synthesis in B16F10 melanoma cells, as well as their effects in UVB-induced hyperpigmentation in brown guinea pig skins. It was demonstrated that the significant inhibition of salidroside (33.0%) and paeonol (22.2-30.9%) on the tyrosinase activity is slightly lower than that of arbutin (18.4-44.7%). However, salidroside exhibited the dose-dependent inhibition (30.6-42.0%) in melanin synthesis at a low concentration of 100 μM, paeonol and arbutin expressed inhibition rates of 27.4-37.2% and 25.8-45.6% within 500-1000 μM. The in vivo topical application of these compounds was demonstrated to obviously decrease the hyperpigmentation on UVB stimulated guinea pig skin. This study provided the original evidence for the salidroside and paeonol as therapeutic agents for pigmentation disorder and skin lightening, with further clinical investigation of these compounds in the field of depigmentation was suggested.

Abstract:Sargassum horneri (SH) is widely consumed as a healthy seaweed food in the Asia–Pacific region. However, the bioactive components contributing to its biological activity remain unknown. Herein, we optimized multifrequency ultrasonic-assisted extraction conditions to achieve higher antioxidant activity using a response surface methodology and an artificial neural network. High-resolution mass spectrometry (HRMS; negative mode) was used to tentatively identify the secondary metabolites in the optimized SH extract, which were further tested against oxidative stress in RAW264.7 cells. Additionally, the identified compounds were analyzed in silico to determine their binding energies with the Keap1 protein (4L7B). We identified 89 compounds using HRMS, among which 19 metabolites (8 polyphenolics, 2 flavonoids, 2 lignans, 2 terpenes, 2 tannins, 2 sulfolipids, and 1 phospholipid) were putatively reported for the first time in SH. The in vitro results revealed that optimized SH extract inhibited oxidative stress via the Nrf2/MAPKs/HO-1 pathway in a dose-dependent manner. This result was validated by performing in silico simulation, indicating that sargaquinoic acid and glycitein-7-O-glucuronide had the highest binding energies (−9.20 and −9.52 Kcal/mol, respectively) toward Keap1 (4L7B). This study offers a unique approach for the scientific community to identify potential bioactive compounds by optimizing the multivariant extraction processing conditions, which could be used to develop functional and nutraceutical foods.

Abstract:The present study aimed to examine the potential inhibitory activity of oleoylethanolamide (OEA) on α-melanocyte stimulating hormone (α-MSH)-stimulated melanogenesis and the molecular mechanism(s) involved in the process in B16 mouse melanoma cells. Our data demonstrated that OEA markedly inhibited melanin synthesis and tyrosinase activity in α-MSH-stimulated B16 cells. In addition, the expression of melanogenesis-related proteins, such as melanocortin-1 receptor (MC1R), microphthalmia-associated transcription factor (MITF), tyrosinase-related protein-1 (TRP-1) and tyrosinase, was suppressed in a concentration-dependent manner by OEA. In addition, OEA may suppress melanogenesis through a peroxisome proliferator-activated receptor α (PPARα)-independent pathway. Moreover, OEA activated ERK, Akt, p38 pathways and inhibits CREB pathway in α-MSH-stimulated B16 cells. The specific ERK inhibitor PD98059 partly blocked OEA-inhibited melanin synthesis and tyrosinase activity and partly abrogated the OEA-suppressed expression of melanogenic proteins. Furthermore, OEA presented remarkable inhibition on the body pigmentation in the zebrafish model system. Our findings demonstrated that OEA is an effective inhibitor of hyperpigmentation through activation of ERK, Akt and p38 pathways, inhibition of the CREB pathway, and subsequent down-regulation of MITF, TRP-1 and tyrosinase production.

Abstract:The inhibitory effects of Salix pseudo-lasiogyne water extract (SPWE) on melanogenesis were evaluated using intracellular tyrosinase activity and melanin content in B16F10 cells exposed to α-melanocyte-stimulating hormone (α-MSH). Meanwhile, the antioxidative capacity of SPWE was also investigated. SPWE suppressed intracellular tyrosinase activity and decreased the melanin content in B16F10 cells. The results showed that tyrosinase protein expression was suppressed by the SPWE-treated group. SPWE inhibited mRNA and protein expression of microphthalmia-associated transcription factor (MITF), tyrosinase-related protein (TRP)-1, and TRP-2. SPWE inhibited the intracellular reactive oxygen species level in B16F10 cells. SPWE not only demonstrated 1,1-diphenyl-2-picrylhydrazyl stable free radical (DPPH), and 2,2-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS)-free radical scavenging activity but also possessed chelation capacity and ferric ions (Fe3+) reducing power. We concluded that anti-melanogenesis and antioxidant effect of SPWE may result from the down-regulation MITF and tyrosinase expression and the inhibition ROS generation. Overall, SPWE might be a potential candidate substance for whitening agent about pigmentation.
