Abstract:OBJECTIVE:To provide basis of environmental factors of genuine crude drug, Salvia miltiorrhiza root.METHOD:On-the-spot investigation and indoor chemical analysis were made to study the physicochemical properties of growing soil and content of inorganic elements of S. miltiorrhiza, and obtained data were analysed with SPSS 10.0 software.RESULT:S. miltiorrhiza root of high harvest area accumulated Cu and Zn. In soil principal component analysis, no principal component was obvious. So the drug has good adaptability in ecological environment of soil.CONCLUSION:Ecological environment of soil isn't leading factor in forming genuine crude S. miltiorrhiza.

Abstract:Mineral elements are important components of medicinal herbs, and their concentrations are affected by many factors. In this study, Ca, Mg, Na, K, Fe, Mn, Cu, and Zn concentrations in wild Saposhnikovia divaricata and its rhizosphere soil collected from seven locations at two different times in China were measured, and influences of rhizosphere soil on those minerals in plant were evaluated. The results showed that mean concentrations of eight minerals in plant samples decreased in the order: Ca > Mg > Na > K > Fe > Zn > Mn > Cu, and those in the soil samples followed the following order: Na > Fe > Ca > K > Mg > Mn > Zn > Cu. Mean concentrations of Ca, Na, Mg, and K in plants were higher than those in soils, while higher mean concentrations of the other four minerals were found in soils. It was found that there was a positive correlation of Mg, Na, and Cu concentrations in the plant with those in the soil respectively, but a negative correlation of Mn concentration in plant with that in the soil. Except Ca, K, and Mn, the other five minerals in plant were all directly affected by one or more chemical compositions of soil. The results also indicate that pH value and concentrations of total nitrogen, Mg, Mn, and Cu in soil had significant correlations with multimineral elements in plant. In a word, mineral elements uptake of S. divaricata can be changed by adjusting the soil fertility levels to meet the need of appropriate quality control of S. divaricata.

Abstract:The root of Salvia miltiorrhiza Bunge (SMB) has been widely used to treat cardiovascular diseases. However, the contents of secondary metabolites in the roots from different production areas are significantly different, and the impact of soil factors on this accumulation remains unclear. Therefore, this study aimed to elucidate the regularity of variation between the active components and soil factors through targeted metabolomics and chemical dosimetry. Soils were collected from five different cities (A, B, C, D, and E) and transplanted into the study area. The results showed that there were significant differences in the soil fertility characteristics and heavy metal pollution levels in different soils. Ten water- and twelve lipid-soluble metabolites were identified in SMBs grown in all soil types. SMBs from D cities exhibited the highest total tanshinone content (p < 0.05). The salvianolic acid B content in SMBs from E cities was the highest (p < 0.05). Interestingly, correlation analysis revealed a significant negative correlation between the accumulation of lipid-soluble and water-soluble metabolites. Double-matrix correlation analysis demonstrated that available potassium (AK) was significantly negatively correlated with salvianolic acid B (r = −0.80, p = 0.0004) and positively correlated with tanshinone IIA (r = 0.66, p = 0.008). Conversely, cadmium (Cd) and cuprum (Cu) were significantly positively and negatively correlated with salvianolic acid B (r = 0.96, p 0.52, p 0.60, p 0.31, p < 0.05) were the primary drivers of the differences in the active components of SMBs. These findings provide a theoretical framework for modulating targeted metabolites of SMB through soil factors, with significant implications for the cultivation and quality control of medicinal plants.

Abstract:The roots of Salvia miltiorrhiza Bunge. are commonly used in the treatment of cardiovascular diseases, and tanshinones and salvianolic acids are its main active ingredients. However, the composition and content of active ingredients of S. miltiorrhiza planted in different regions of the soil environment are also quite different, which adds new difficulties to the large-scale and standardization of artificial cultivation. Therefore, in this study, we measured the active ingredients in the roots of S. miltiorrhiza and the contents of rhizosphere soil elements from 25 production areas in eight provinces in China, and used the data to develop a prediction model based on BP (back propagation) neural network. The results showed that the active ingredients had different degrees of correlation with soil macronutrients and trace elements, the prediction model had the best performance (MSE = 0.0203, 0.0164; R 2 = 0.93, 0.94). The artificial neural network model was shown to be a method that can be used to screen the suitable cultivation sites and proper fertilization. It can also be used to optimize the fertilizer application at specific sites. It also suggested that soil testing formula fertilization should be carried out for medicinal plants like S. miltiorrhiza , which is grown in multiple origins, rather than promoting the use of “special fertilizer” on a large scale. Therefore, the model is helpful for efficient, rational, and scientific guidance of fertilization management in the cultivation of S. miltiorrhiza .

Abstract:Salvia miltiorrhiza is one of the famous traditional Chinese medicines for treating cardiovascular and cerebrovascular diseases. Tanshinone and phenolic acids are the main active compounds of Salvia miltiorrhiza, whereas the distribution patterns of the two kinds of components are still unclear among Salvia spp. In this work, high-performance liquid chromatography was applied to analyze the distribution patterns of major components in the roots and leaves of 58 Salvia spp. The results showed that the distribution patterns of tanshinone and phenolic acids in Salvia spp. varied significantly. Phenolic acid components such as rosmarinus acid, caffeic acid, and danshensu are widely distributed in the roots and leaves, and phenolic acids in the leaves of Salvia spp. are generally higher than that in roots. Tanshinones are mainly detected in the roots of Salvia przewalskii, Salvia trijuga, Salvia castanea, and Salvia yunnanensis. The content of major components of the different species varied significantly. The content of phenolic acids in most Salvia spp. generally followed the pattern of salvianolic acid B > rosmarinic acid > danshensu > caffeic acid both in the roots and leaves. Tanshinone IIA and cryptotanshinone were the main lipophilic components of Salvia spp. distributed in southwest China. A correlation between the distribution pattern of tanshinone and the genetic relationship of species was indicated in the work. This research systematically reveals the distribution patterns of tanshinone and phenolic acids in Salvia spp., providing a theoretical basis for the development and utilization of medicinal resources of Salvia.

Abstract:We investigated the effects of the complex cultivation of Salvia miltiorrhiza on microbial communities, secretions, yield, and active ingredients, and the mechanism of action between microbial communities, secretions, and S. miltiorrhiza growth and development. Neither maize nor soybean was suitable to grow with S. miltiorrhiza , but sesame significantly increased salvinone content, the active ingredient of S. miltiorrhiza , and Tanshinone IIA, Tanshinone I, and Cryptotanshinone increased by 27.06%, 22.76%, and 26.41%, respectively, which increased the abundance and number of microbial communities in S. miltiorrhiza roots. 16S rRNA results showed that the most abundant bacterial phyla were Proteobacteria and Acidobacteriota, and their number increased with compound planting of sesame and S. miltiorrhiza . Salvia inter-root secretions affected the microbial community and Salvia growth and development, and lipids and lipid-like molecules significantly reduced Salvia yield and active ingredients. Overall, different plant secretions can lead to differences in the natural environment and Salvia root growth and development, and the composite planting of sesame with Salvia can improve inter-root microbial communities, enhance Salvia quality, and make fuller use of land resources.

Abstract:Tanshinones are abietane diterpenoid quinone compounds with diverse biological activities and pharmacological effects found in Salvia miltiorrhiza . Leveraging the high-density genetic map established through our prior research endeavors, we conducted a quantitative trait locus (QTL) analysis pertaining to the concentrations of three major tanshinone components, cryptotanshinone, tanshinone I, and tanshinone IIA, in S. miltiorrhiza . This extensive investigation was conducted across three distinct planting environments, ultimately identifying a comprehensive repertoire of 27 discernible QTLs. These QTLs were mapped onto four distinct linkage groups (LG), namely LG1, LG5, LG6, and LG7, which explained 3.11%-37.85% phenotypic variation. Candidate genes were projected based on consistent QTLs detected for each active ingredient in three environments. Nineteen putative candidate genes involved in the regulation of tanshinone biosynthesis were identified. These genes participate in primary metabolic and multiple branching terpenoid biosynthesis pathways, forming a complex regulatory network. Our findings have the potential to offer novel insights into advancing the understanding of the regulatory mechanisms governing tanshinone biosynthesis. Furthermore, these results establish crucial groundwork for gene discovery, marker-assisted selection breeding, and map-based cloning of functional genes associated with tanshinone content in S. miltiorrhiza .

Abstract:Salvia miltiorrhiza , known as danshen, is one of most valued medicinal plants in China. Although it has been cultivated since ancient times, an optimal culture system needs to be standardized for this important species. Here, we explored the phytochemical properties of S. miltiorrhiza with the treatments of rare earth elements (REEs) to develop an optimal tissue culture system. Four-week-old in vitro-grown S. miltiorrhiza plantlets were used as explants. The experiment was conducted in a randomized block design on a Murashige and Skoog (MS) medium containing 0.2 mg·L −1 naphthaleneacetic acid (NAA) to induce rooting at four different concentrations (50, 100, 200, and 300 μM) of REEs such as cerium (Ce), lanthanum (La), or praseodymium (Pr), respectively. Compared with all REEs at different concentrations, 100 μM Pr induced greater root length than Ce or La at any concentrations. Concomitantly, 0.38 μg tanshinone IIA/mg dry weight (DW) was observed, which was 54.84% higher than in the control. Similarly, chlorophyll content, antioxidant enzyme activity, and secondary metabolite were enhanced in rooting medium supplemented with 100 μM Pr. Therefore, this study showed that 100 μM Pr is an adequate concentration in the optimal culture system for promoting plant growth as well as enhancing secondary metabolite content in S. miltiorrhiza .

Abstract:Salvia miltiorrhiza Bunge, a member of the Lamiaceae family, is valued in traditional Chinese Medicine. Its dried root (named Danshen) has been used for hundreds of years, primarily for the treatment of cardiovascular and cerebrovascular diseases. Tanshinones are the main active ingredients in S. miltiorrhiza and exhibit significant pharmacological activities, such as antioxidant activity, anti-inflammatory activity, cardiovascular effects, and antitumor activity. Danshen dripping pill of Tianshili is an effective drug widely used in the clinical treatment of cardiovascular diseases. With the increasing demand for clinical drugs, the traditional method for extracting and separating tanshinones from medicinal plants is insufficient. Therefore, in combination with synthetic biological methods and strategies, it is necessary to analyze the biosynthetic pathway of tanshinones and construct high-yield functional bacteria to obtain tanshinones. Moreover, the biosynthesis of tanshinones has been studied for more than two decades but remains to be completely elucidated. This review will systematically present the composition, extraction and separation, pharmacological activities and biosynthesis of tanshinones from S. miltiorrhiza, with the intent to provide references for studies on other terpenoid bioactive components of traditional Chinese medicines and to provide new research strategies for the sustainable development of traditional Chinese medicine resources.

Abstract:Plants of the genus Salvia produce various types of phenolic compounds and tanshinones which are effective for treatment of coronary heart disease. Salvia miltiorrhiza and S. castanea Diels f. tomentosa Stib are two important members of the genus. In this study, metabolic profiles and cDNA-AFLP analysis of four samples were employed to identify novel genes potentially involved in phenolic compounds and tanshinones biosynthesis, including the red roots from the two species and two tanshinone-free roots from S. miltiorrhiza. The results showed that the red roots of S. castanea Diels f. tomentosa Stib produced high contents of rosmarinic acid (21.77 mg/g) and tanshinone IIA (12.60 mg/g), but low content of salvianolic acid B (1.45 mg/g). The red roots of S. miltiorrhiza produced high content of salvianolic acid B (18.69 mg/g), while tanshinones accumulation in this sample was much less than that in S. castanea Diels f. tomentosa Stib. Tanshinones were not detected in the two tanshinone-free samples, which produced high contents of phenolic compounds. A cDNA-AFLP analysis with 128 primer pairs revealed that 2300 transcript derived fragments (TDFs) were differentially expressed among the four samples. About 323 TDFs were sequenced, of which 78 TDFs were annotated with known functions through BLASTX searching the Genbank database and 14 annotated TDFs were assigned into secondary metabolic pathways through searching the KEGGPATHWAY database. The quantitative real-time PCR analysis indicated that the expression of 9 TDFs was positively correlated with accumulation of phenolic compounds and tanshinones. These TDFs additionally showed coordinated transcriptional response with 6 previously-identified genes involved in biosynthesis of tanshinones and phenolic compounds in S. miltiorrhiza hairy roots treated with yeast extract. The sequence data in the present work not only provided us candidate genes involved in phenolic compounds and tanshinones biosynthesis but also gave us further insight into secondary metabolism in Salvia.
Effects of smoke-water and smoke-isolated karrikinolide on tanshinones production in Salvia miltiorrhiza hairy roots

Abstract:Smoke–water (SW) and the biologically-active compound karrikinolide (KAR1), isolated from plant-derived smoke, is known to show stimulating effects on the plant growth of agricultural and horticultural crops. However, little knowledge has been obtained on their effects on the secondary metabolism of medicinal plants. In this study, hairy root cultures of Salvia miltiorrhiza, one of the most renowned herbs in traditional Chinese medicine (commonly known as “danshen”), were treated with SW at dilutions of 1:500 (v/v), 1:1000 (v/v), 1:2000 (v/v) or KAR1 solution at 10−9 M. The effects of SW and KAR1 treatment on the content of tanshinones (tanshinone I, tanshinone IIA, cryptotanshinone and dihydrotanshinone) and the expressions pattern of genes involved in biosynthesis of tanshinones: Acetyl CoA acyltransferase (AACT), 3-hydroxy-3-methylglutaryl CoA synthase (HMGS), 3-hydroxy-3-methylglutaryl CoA redutase (HMGR), 1-deoxy-D-xylulose5-phosphate synthase (DXS), 1-deoxy-D-xylulose5-phosphate reductoisomerase (DXR), 4-(5-pyrophosphate cytidine) -2-C-methyl-Derythritol kinase (CMK), Cobaxyl pyrophosphate synthase (CPS), isopentenyl diphosphate isomerase (IPPI) and geranylgeranyl diphosphate synthase (GGPPS) in S.miltiorrhiza hairy root during elicitation in the S.miltiorrhiza hairy roots were evaluated. The results showed that both SW and KAR1 treatments markedly increased the content of tanshinones in hairy roots of S.miltiorrhiza. The content of tanshinone I was significantly improved, and was evaluated as 3.1-, 3.3-, 1.3- and 1.2-fold of that in the control on 1, 3, 5 and 9 d after treatment with SW 1:1000, respectively. The content of tanshinone IIA was slightly increased by SW and KAR1. The effects of SW treatments on cryptotanshinone content were concentration-dependent. Compared to the control, the accumulation of dihydrotanshinone increased significantly (p < 0.05) with SW and KAR1(p < 0.05) treatments in the earlier stage (day 1–3post-treatment). Enzymes of the tanshinone pathways showed different responses to the treatments of SW and KAR1 due to their multiple roles in the secondary metabolite pathways. The transcript of HMGS was more sensitive to the SW and KAR1 elicitation, whilst the responses of DXS, DXR, CPS and GGPPS to SW and KAR1 treatments were later than AACT and HMGS. Our results indicate that smoke–water and KAR1 have the potential to be used as elicitors to increase the accumulation of secondary metabolites in S.miltiorrhiza hairy roots, as well as to provide insights into the mechanisms of biosynthesis of liposoluble tanshinones in S.miltiorrhiza.
Effects of Cerous Nitrate on Growth and Tanshinone Production in Salvia Miltiorrhiza Hairy Roots

Abstract:Salvia miltiorrhiza, a popular traditional Chinese medicine, is widely used for treatments in cardiotonic disease. Tanshinones are a group of bioactive ingredients in S. miltiorrhiza. In this study, Ce3+ was used as an elicitor to enhance tanshinones production in S. miltiorrhiza hairy roots. The results showed that contents of dihydrotanshinone I (DTI) and cryptotanshinone (CT) were significantly enhanced by 50 μmol/L Ce3+, and reached to 0.875 mg/g and 0.271 mg/g, respectively. However, tanshinone II A (TIIA) and tanshinone I (TI) contents were significantly decreased to 59% and 62% of the control. Simultaneously, expressions of genes (HMGR, DXR, DXS1, DXS2 and GGPPS) involved in tanshinone biosynthesis were upregulated by Ce3+. Responses of DXS1, DXS2 and GGPPS to Ce3+ treatments were later than HMGR and DXR. We speculated that branch pathways of DTI and CT biosynthesis were probably different from TIIA and TI. This work will help us understand biosynthetic mechanism of tanshinones in plants.
Deciphering the effects of genotype and climatic factors on the performance, active ingredients and rhizosphere soil properties of Salvia miltiorrhiza

Abstract:Introduction: Salvia miltiorrhiza Bunge is an important medicinal herb, which is widely cultivated in most parts of China. It has attracted considerable attention because of its pharmacological properties and potential health benefits. Methods: We used a field experiment to determine the effects of different genotypes and climatic factors on the performance (plant biomass, morphological parameters), active ingredients, rhizosphere soil physicochemical properties and microbial composition of S. miltiorrhiza at five cultivation locations. Results: The results showed that these parameters were significantly different in the six different genotypes of S. miltiorrhiza from five producing areas. Genotype and soil physicochemical properties were the main factors affecting the growth traits of S. miltiorrhiza , while genotype, climate and soil physicochemical properties were the main factors affecting the content of active components of S. miltiorrhiza . Microbial phospholipid fatty acid analysis showed that the biomass of Gram-positive and Gram-negative bacteria was affected by the genotypes of S. miltiorrhiza plants, while the biomass of arbuscular mycorrhizal fungi, fungi, Gram-positive and Gram-negative bacteria was affected by climate factors. Discussion: Based on the main results, DS993 was the most suitable genotype for S. miltiorrhiza in the five producing areas from the perspective of comprehensive growth traits and medicinal components, while DS993 and DS2000 were suitable for planting in Shandong province from the perspective of origin. DS996 is not suitable for all of the above production areas. These results are helpful to understand the ecological adaptability of different genotypes of S. miltiorrhiza resources, and to select appropriate S. miltiorrhiza genotypes for specific planting areas, so as to maximize yield and quality.

Comparative transcriptome analyses of different Salvia miltiorrhiza varieties during the accumulation of tanshinones

Abstract:Salvia miltiorrhiza (Labiatae) is an important medicinal plant in traditional Chinese medicine. Tanshinones are one of the main active components of S. miltiorrhiza . It has been found that the intraspecific variation of S. miltiorrhiza is relatively large and the content of tanshinones in its roots of different varieties is also relatively different. To investigate the molecular mechanisms that responsible for the differences among these varieties, the tanshinones content was determined and comparative transcriptomics analysis was carried out during the tanshinones accumulation stage. A total of 52,216 unigenes were obtained from the transcriptome by RNA sequencing among which 23,369 genes were differentially expressed among different varieties, and 2,016 genes including 18 diterpenoid biosynthesis-related genes were differentially expressed during the tanshinones accumulation stage. Functional categorization of the differentially expressed genes (DEGs) among these varieties revealed that the pathway related to photosynthesis, oxidative phosphorylation, secondary metabolite biosynthesis, diterpenoid biosynthesis, terpenoid backbone biosynthesis, sesquiterpenoid and triterpenoid biosynthesis are the most differentially regulated processes in these varieties. The six tanshinone components in these varieties showed different dynamic changes in tanshinone accumulation stage. In addition, combined with the analysis of the dynamic changes, 277 DEGs (including one dehydrogenase, three CYP450 and 24 transcription factors belonging to 12 transcription factor families) related to the accumulation of tanshinones components were obtained. Furthermore, the KEGG pathway enrichment analysis of these 277 DEGs suggested that there might be an interconnection between the primary metabolic processes, signaling processes and the accumulation of tanshinones components. This study expands the vision of intraspecific variation and gene regulation mechanism of secondary metabolite biosynthesis pathways in medicinal plants from the “omics” perspective.

Abstract:Salvia miltiorrhiza B., an herb used in traditional Chinese medicine, has been widely used to prevent and treat cardiovascular and other diseases. Currently, the majority of medicinal plants used in the US are imported from foreign countries, which involves transportation, quality control, and other issues. The objective of this study was to investigate the effect of nitrogen fertilization on growth and content of tanshinone I, tanshinone IIA, cryptotanshinone, and salvianolic acid B for Salvia miltiorrhiza B. in Mississippi. Plants were fertilized with one of five nitrogen (N) rates (0, 2, 4, 6, or 8 g N/plant from NH4NO3). Plants were harvested in November 2020 and 2021. Plants treated with 8 g N had higher plant growth index, leaf SPAD value, shoot and root number, shoot and root weight, maximum root length and diameter, shoot: root ratio, N concentration in root, and content of bioactive components compared to plants treated with 0, 2, 4 g N. Plants receiving 6 g N had similar shoot number, maximum root length, maximum root diameter, root weight and content of bioactive components compared to plants receiving 8 g N. However, plants receiving 6 g N had higher photosynthetic activity compared to plants receiving the higher N rate. Higher N rates increased plant growth and content of tested bioactive compounds.

Abstract:Climate change profoundly influences the geospatial distribution of secondary metabolites and causes the geographical migration of plants. We planted seedlings of the same species in eighteen ecological regions along a latitudinal gradient in eastern and western China, in order to explore the regulation of multi-climatic factors on active ingredient accumulation in Salvia miltiorrhiza Bunge. The correlations between six active ingredient contents and ten climatic factors were investigated to clarify their relationships. We found that climatic factors not only regulated active ingredient contents but also markedly influenced their composition and led to a specific geospatial distribution of these active ingredients in China. The main climatic factors include the air temperature, precipitation, atmospheric vapour pressure and sunshine duration. Future warming in high-latitude regions could cause continued northward expansion of planting areas suitable for S. miltiorrhiza. The effect of extreme climatic conditions on active ingredients should not be overlooked. The findings of this study can help farmers scientifically choose suitable cultivation regions in the future. Furthermore, this study provides an innovative idea for the exploration of secondary metabolic responses to changing ecological factors in medicinal plants.

Abstract:With the continuous increase in saline–alkali land, sustainable development of the global environment and ecology have been seriously affected. This study compared the absorption and accumulation patterns of 11 elements in different parts (roots, stems and leaves) of different leaf Na regulation strategies of the pioneer plant Chenopodiaceae in saline–alkali land and evaluated the effects of soil nutrient status and soil salinity on the distribution of plant elements. The results showed that the changes in the content of Ca, Mg and Na in plants are affected by the salt-tolerant type and on different parts. Soil salinity had no significant effect on element concentrations in different parts of plants. The Pearson correlation coefficient showed that the correlation between plants and soil elements was different, and different parts of plants had the characteristics of selective absorption of soil elements. The salt tolerance type and soil mineral element concentrations explained most of the variation observed in element concentrations in Chenopodiaceae plants; the soil salinity property played only a minor role. It was concluded that the genetic factors are the prerequisite in the composition pattern of leaf elements in Chenopodiaceae, and soil factors are the key to determining element accumulation. These conclusions provide an effective reference for evaluating plant breeding and its response to environmental change in saline–alkali arid areas in Hulunbuir grassland and other parts of the world.

[Distribution characteristics and correlation analysis of inorganic elements in Alismatis rhizoma and rhizospheric soils in different habitats]

Abstract:Objective: To investigate the distribution characteristics and correlation of inorganic element contents in Alismatis Rhizoma and rhizospheric soils from three habitats (Jianou, Fujian; Guangchang, Jiangxi; Pengshan, Sichuan). Methods: The inorganic element contents were detected with inductively coupled plasma mass spectrometry (ICP-MS). Results: The results showed that inorganic element contents in Alismatis Rhizoma from different habitats presented regular distribution characteristics. In all samples, the contents of S, P, K, Mg and Ca were relatively high. The contents of S, P, K, Fe, Mg, Ca, Al and Zn in Alismatis Rhizoma from Fujian with low-content harmful heavy metal were more than those of other two places. Alismatis Rhizoma from different habitats showed remarkable ability to enrich P, S, Zn, Mg and Cu, especially to P and S. The contents of inorganic element in Alismatis Rhizoma and rhizosperic soil showed certain correlation. Conclusion: These results provide scientific evidence for artificial modification of effective components biosynthesis in Alisma orientalis, and it could also provide a reference for forming mechanism of genuine characteristics and quality of Alismatis Rhizoma.
Effect of rhizosphere soil microbial communities and environmental factors on growth and the active ingredients of Angelica sinensis in Gansu Province, China

Abstract:The growth and accumulation of active ingredients of Angelica sinensis were affected by rhizosphere soil microbial communities and soil environmental factors. However, the correlationship between growth and active ingredients and soil biotic and abiotic factors is still unclear. This study explored rhizosphere soil microbial community structures, soil physicochemical properties, enzyme activities, and their effects on the growth and active ingredient contents of A. sinensis in three principal cropping areas. Results indicated that the growth indices, ligustilide, ferulic acid contents, and soil environmental factors varied in cropping areas. Pearson correlation analysis revealed that the growth of A. sinensis was affected by organic matter, total nitrogen, total phosphorus, and available phosphorus; ferulic acid and ligustilide accumulation were related to soil catalase and alkaline phosphatase activities, respectively. Illumina MiSeq sequencing showed that the genera Mortierella and Conocybe were the dominant fungal communities, and Sphingomonas , Pseudomonas , Bryobacter , and Lysobacter were the main bacterial communities associated with the rhizosphere soil. Kruskal–Wallis one-way ANOVA and Spearman correlation conjoint analysis demonstrated a significant positive correlation ( p < 0.001) among the composition of the rhizosphere microbial communities at all three sampling sites. The growth and active ingredient accumulation of A. sinensis not only was significantly susceptible to the bacterial communities of Sphingomonas , Epicoccum , Marivita , Muribaculum , and Gemmatimonas but also were significantly influenced by the fungal communities of Inocybe , Septoria , Tetracladium , and Mortierella ( p < 0.05). Our findings provide a scientific basis for understanding the relationship between the growth and active ingredients in A. sinensis and their corresponding rhizosphere soil microbial communities, soil physicochemical properties, and enzyme activities.

Abstract:The feasibility of using chemometric techniques associated with multi-element analysis for the classification of three varieties of Codonopsis Radix samples was investigated, and the correlation between environmental factors and metal element content was further explored. It was found that R. Codonopsis was a good source of potassium (average content of 9420 μg/g), magnesium (average content of 1590 μg/g), calcium (average content of 388 μg/g) and iron (average content of 333 μg/g). The statistical analysis results indicated that twenty-two elements content (K, Ca, Mg, Fe, Se, Na, Zn, V, Cu, Cr, Mn, Ni, Co, Sr, Pb, Cd, As, Hg, Al, Ba, Tl and U) were significantly different among three R. Codonopsis varieties, hence it was possible to discriminate R. Codonopsis according to their varieties by LDA (correct classification rate of 98.6% and cross-validation rate of 95.9%). In addition, the correlation analysis results indicated that the relative humidity showed significantly negative correlation with essential elemental content such as K, Mg, Ca and Se (correlation coefficients ranged from -0.321 to -0.564, p < 0.05); the annual average precipitation exhibited significantly positive correlation with toxic elemental elements such as As, Cd, Pb and Hg (correlation coefficients ranged from 0.266 to 0.897, p < 0.05).
