Abstract:Panax ginseng C.A.Meyer is a perennial herb of the Araliaceae family.As the use of traditional Chinese medicine as a new resource food supply becomes more and more popular and sales of P.ginseng are increasing worldwide.The therapeutic potential of P.ginseng has been studied extensively,and ginsenosides,the principal active ingredients of P.ginseng,are shown to be involved in diverse biological and pharmacological activities.Currently, there are several commercial extract preparations such as G115 (marketed as Ginsana),which is the standardized extracts of the roots of P.ginseng and contains 4% ginsenosides.The modern action mechanism of P.ginseng had not been known until ginsenosides were isolated in 1963.Much effort has since been focused on evaluating the function and elucidating the molecular mechanism of each ginsenoside.A variety of publications on P.ginseng and ginsenosides has been growing exponentially since 1975.This article reviews the important biological and pharmacological activities of triterpenoids isolated from P.ginseng over the last few years.These include safety assessment,and central nervous and cardio-cerebral vascular regulation,immunomodulating, endocrine system and anti-stress, anti-inflammation, cytotoxicity against tumor cells and anticancer, antidiabetic and antiobesity,lowering blood lipid,hepatoprotection,anti-osteoporosis,anti-fatigue,as well as anti-aging activities and so on, which might greatly contribute to provide the scientific basis for Evidence-Based Medicine in clarifying the effective substance basis of P.ginseng.

Abstract:The panax genus is a widely used medicinal plant with good biological activity. As one of the main active components of the Panax genus, polysaccharides have various pharmacological effects. This review summarizes the latest research reports on ginseng, American ginseng, and Panax notoginseng polysaccharides and compares the differences in extraction, isolation and purification, structural characteristics, and biological activities. The current research mainly focuses on ginseng polysaccharides, and the process of extraction, isolation, and structure analysis of each polysaccharide is roughly the same. Modern pharmacological studies have shown that these polysaccharides have antioxidants, antitumor, immunomodulatory, antidiabetic, intestinal protection, skin repair, and other biological activities. This review provides new insights into the differences between the three kinds of ginseng polysaccharides which will help to further study the medicinal value of ginseng in traditional Chinese medicine.

P. ginseng, a special herb used in medicine and food, is one of the main research objects for qualitative and quantitative analysis of metabolomics and has gradually become the focus of researchers globally. Here, we conducted a comprehensive summary and analysis of numerous studies published in ginseng metabolomics. This review aims to provide more novel ideas for the quality evaluation, development, and clinical application of ginseng in the future and offer more useful technical references for the modernization and internationalization of herbal medicine based on metabolomics.

Abstract:Panax notoginseng is a famous perennial herb widely used as material for medicine and health-care food. Due to its various therapeutic effects, research work on P. notoginseng has rapidly increased in recent years, urging a comprehensive review of research progress on this important medicinal plant. Here, we summarize the latest studies on the representative bioactive constituents of P. notoginseng and their multiple pharmacological effects, like cardiovascular protection, anti-tumor, and immunomodulatory activities. More importantly, we emphasize the biosynthesis and regulation of ginsenosides, which are the main bioactive ingredients of P. notoginseng. Key enzymes and transcription factors (TFs) involved in the biosynthesis of ginsenosides are reviewed, including diverse CYP450s, UGTs, bHLH, and ERF TFs. We also construct a transcriptional regulatory network based on multi-omics data and predicted candidate TFs mediating the biosynthesis of ginsenosides. Finally, the current three major biotechnological approaches for ginsenoside production are highlighted. This review covers advances in the past decades, providing insights into quality evaluation and perspectives for the rational utilization and development of P. notoginseng resources. Modern omics technologies facilitate the exploration of the molecular mechanisms of ginsenoside biosynthesis, which is crucial to the breeding of novel P. notoginseng varieties. The identification of functional enzymes for biosynthesizing ginsenosides will lead to the formulation of potential strategies for the efficient and large-scale production of specific ginsenosides.

Abstract:Ginseng (Panax ginseng C. A. Meyer) is known as a valuable traditional Chinese herb for tens of centuries. Previous pharmacological and phytochemical investigations indicate that polysaccharides are a main biologically active ginseng component that possesses different bioactivities such as antitumor, immunoregulation, antioxidation, hepatoprotection, and anti-inflammation. Work is needed to summarize the full aspects of these polysaccharides systematically to boost their further investigation and application. In this review, studies on the extraction, purification, characterization, and bioactivity are introduced in detail. Moreover, the structure–bioactivity relationship of ginseng polysaccharides is explored and discussed. The uronic acids richer in ginseng polysaccharides tend to have higher biological effect, and the α-(1→4)-GalpA and α-(1→4)-Galp in the main chain are essential for the anti-inflammatory and antiviral effects. We believe our work proposes a helpful bibliography for further innovation and promotes these natural products in functional food.

Integrated metabolomic and transcriptomic analyses revealed the distribution of saponins in Panax notoginseng

Abstract:Panax notoginseng is famous for its important therapeutic effects. Saponins are bioactive compounds found in different parts and developmental stages of P. notoginseng plants. Thus, it is urgently to study saponins distribution in different parts and growth ages of P. notoginseng plants. In this study, potential biomarkers were found, and their chemical characteristic differences were revealed through metabolomic analysis. High-performance liquid chromatography data indicated the higher content of saponins (i.e., Rg1, Re, Rd, and Rb1) in the underground parts than that in the aerial parts. 20(S)-Protopanaxadiol saponins were mainly distributed in the aerial parts. Additionally, the total saponin content in the 3-year-old P. notoginseng plant (188.0 mg/g) was 1.4-fold higher than that in 2-year-old plant (130.5 mg/g). The transcriptomic analysis indicated the tissue-specific transcription expression of genes, namely, PnFPS, PnSS, PnSE1, PnSE2, and PnDS, which encoded critical synthases in saponin biosyntheses. These genes showed similar expression patterns among the parts of P. notoginseng plants. The expression levels of these genes in the flowers and leaves were 5.2fold higher than that in the roots and fibrils. These results suggested that saponins might be actively synthesized in the aerial parts and transformed to the underground parts. This study provides insights into the chemical and genetic characteristics of P. notoginseng to facilitate the synthesis of its secondary metabolites and a scientific basis for appropriate collection and rational use of this plant.

Abstract:Ginseng, an ancient medicinal herb, is used in oriental medicine for the treatment of various diseases. Saponins are the main bioactive components of ginseng, but the multiple glucosyl side chains on its molecules prevent ginsenosides from entering the blood through the intestinal membrane, thus reducing the efficacy. The preparation of rare ginsenosides, which are easy to be absorbed by human body and have higher drug activity, has been widely practiced by removing the sugar group of natural ginsenosides in vitro . Rare ginsenosides Rg3 and Rh2 have been approved as drugs or health supplements to improve immune function. This review summarizes the preparation methods of ginsenoside Rg3 and Rh2 in recent years. Ginsenoside Rg3 and Rh2 were prepared by biotransformation of protopanaxadiol type ginsenoside, with the highest conversion rate of 98.19% and 95.89% in the laboratory, respectively. At present, improving the conversion rate and reducing the production cost are still the bottleneck of industrial scale production of Rg3 and Rh2 through the deglycosylation directly from Rb1, Rb2, Rb3, Rc and Rd in the crude extract of ginseng. In addition, ginsenosides Rg3 and Rh2 play anti-inflammatory, anticancer, cardiovascular protective, immunomodulatory, neuroprotective, anti-diabetic, anti-fatigue, anti-allergic, anti-aging, antioxidant and other pharmacological effects by activating AMPK, JNK, NF-κB, MAPKs, P13K/AKT/mTOR and other signaling pathways. As potential drugs for prevention and treatment of various diseases, ginsenoside Rg3 and Rh2 need to further clarify other underlying mechanisms of action through in vitro and in vivo experiments.

Abstract:As a kind of commonly used traditional Chinese medicine, ginseng has a high reputation at home and abroad. The research of ginseng has been expanded to medicine, pharmacy, biology, food science and other fields, with great achievements in recent years. Ginseng contains ginsenosides, volatile oil, carbohydrates, amino acids, polypeptides, inorganic elements and othser chemical constituents. Each component has extensive physiological activity, and is the base of ginseng's effect. After processing, the complicated changes are taken place in the constituents of ginseng, and some new substances produced. This paper aims to review the studies on chemical constituents and their mechanisms during ginseng processing, and the ideas, methods and the direction of the development of traditional Chinese medicine processing in the future.

Abstract:The Chinese people discovered ginseng and used it as a revitalizing agent since time immemorial. They are still the world's major consumers of this plant drug. The commercial product of ginseng comes from two species of the genus Panax in the family Araliaceae. These species are P. ginseng C. A. Meyer which is the source plant of the Chinese, Korean and Japanese brands of ginseng, and P. quinquefolius L., which is the source of American ginseng. Phytogeographically, ginseng demonstrates the classical bicentric pattern of plant distribution, with closely related species in eastern Asia and in eastern North America. Ecologically, ginseng is an undergrowth of hardwood mixed deciduous forest. It prefers the northern or the northeastern slope of a hill. Species of the genus Tilia are good indicators of the proper environmental condition for the growth of ginseng. Morphologically, ginseng is a perennial herb with fleshy root, a single annual stem bearing a whorl of palmately compound leaves, and a terminal simple umbel of small 5-merous flowers. The flowers are soon followed by pea-sized fruits developed from inferior ovaries. The fruits are red when ripe. Ginseng is propagated by seed. The commercial products of ginseng consist primarily of roots 2-20 years old. Within this age range, the older the root the higher the market value, provided they are grown in proper conditions. The methods of curing the roots change the color and shape of the products. Chinese ginseng is prepared from roots bleached, boiled, steamed, or sugared in curing. The cultural background for the uses of ginseng by the Chinese people is explained. Ginseng may be used alone in the form of tea, powder, or as a masticatory. It is also used in combination with other drugs of animal, mineral, or plant origin. Forty-two recipes are selected from Pen-ts'oa kang-mu and translated into English for the first time to show the various ways by which ginseng is used in traditional Chinese medicine. A systematic summary of the companion plant drugs of ginseng is presented in the form of a table, showing the distribution of the species in the plant kingdom. The scientific names of the species are given in full.

Abstract:ETHNOPHARMACOLOGICAL RELEVANCE: Panax notoginseng (Burk.) F. H. Chen, also called Sanqi, is a widely used traditional Chinese medicine, which has long history used as herbal medicines. It is currently an important medicinal material in China, holding the first place in the sale volume of the whole patent medicines market in China, and the market size of the single species has exceeded 10 billion yuan. In addition, P. notoginseng is an important constituent part of many famous Chinese patent medicines, such as Compound Danshen Dripping Pills and Yunnan Baiyao. P. notoginseng saponins (PNSs), which are the major active components of P. notoginseng, are a kind of chemical mixture containing different dammarane-type saponins. Many studies show that PNSs have been extensively used in medical research or applications, such as atherosclerosis, diabetes, acute lung injury, cancer, and cardiovascular diseases. In addition, various PNS preparations, such as injections and capsules, have been made commercially available and are widely applied in clinical practice.AIM OF THE REVIEW: Since the safety and efficacy of compounds are related to their qualitative and quantitative analyses, this review briefly summarizes the analytic approaches for PNSs and their biological effects developed in the last decade.METHODOLOGY: This review conducted a systematic search in electronic databases, such as Pubmed, Google Scholar, SciFinder, ISI Web of Science, and CNKI, since 2009. The information provided in this review is based on peer-reviewed papers and patents in either English or Chinese.RESULTS: At present, the chromatographic technique remains the most extensively used approach for the identification or quantitation of PNSs, coupled with different detectors, among which the difference mainly lies in their sensitivity and specificity for analyzing various compounds. It is well-known that PNSs have traditionally strong activity on cardiovascular diseases, such as atherosclerosis, intracerebral hemorrhage, or brain injury. The recent studies showed that PNSs also responded to osteoporosis, cancers, diabetes, and drug toxicity. However, some other studies also showed that some PNSs injections and special PNS components might lead to some biological toxicity under certain dosages.CONCLUSION: This review may be used as a basis for further research in the field of quantitative and qualitative analyses, and is expected to provide updated and valuable insights into the potential medicinal applications of PNSs.

Medical Mistrust and Less Satisfaction With Health Care Among Native Americans Presenting for Cancer Treatment

Abstract:Purpose. To assess barriers to cancer care among Native Americans, whose health outcomes compare unfavorably with those of the general U.S. population. Methods and patients. We undertook a comparative community-based participatory research project in which newly-diagnosed cancer patients were prospectively surveyed using novel scales for medical mistrust and satisfaction with health care. Socio-demographic information was obtained. Mean scale scores for mistrust and satisfaction were analyzed by race. Multivariable models were used to adjust for income, education level, and distance lived from cancer care institute. Results. Participation refusal rate was 38%. Of 165 eligible patients, 52 were Native American and 113 where non-Hispanic White. Native Americans expressed significantly higher levels of mistrust (p =.0001) and lower levels of satisfaction (p = .0001) with health care than Whites. In multivariable analyses, race was the only factor found to be significantly predictive of higher mistrust and lower satisfaction scores. Conclusion. Native Americans exhibit higher medical mistrust and lower satisfaction with health care.

Abstract:The steamed Panax ginseng C. A. Mey. (Renshen in Chinese), Panax notoginseng (Burk.) F. H. Chen (Sanqi in Chinese), and Panax quinquefolium L. (Xiyangshen in Chinese) not only facilitate storage, but also increase the content of rare saponins and enhance their clinical application value. The traditional steaming process has high energy consumption, low efficiency, and lacks standardized operating procedures and unified standards. This paper retrieves the research literature on the steaming process parameters, rare saponin increments, and efficacy enhancement results of ginseng plants. By summarizing the effects of different steaming processes on rare saponins and pharmacodynamics in P. ginseng, P. notoginseng and P. quinquefolium, we explored new steaming methods and established a new quality evaluation system to provide guarantee for the effectiveness and safety of clinical medication and provide scientific basis for its rational use.

Abstract:Ginseng is members of the genus Panax in the Araliaceae family,which is well-known as the king of herbs,and has been an important medicinal resource all over the world.Many studies have been conducted to elucidate magical healing activities of ginsengs.Since most studies have been focused on the roots and rhizomes of ginseng as traditional medicinal parts,scientists are less attracted by ginseng stems-leaves.The prices of the roots and rhizomes of ginseng are expensive because it takes 4-6 years for growing from the seed,whereas ginseng stems-leaves which the prices are low and can be harvested every year.Therefore,if the ingredients from the stems-leaves have a similar or new biological activity as those from the roots and rhizomes,their utility values will be improved.Ginsenosides,the active components of the roots and rhizomes of ginseng,are shown to be involved in modulating multiple physiological activities.As a continuation of our summary on ginseng, this article will review the triterpenoids isolated from the roots and rhizomes,stems-leaves,fruits and seeds of ginseng as well as red ginseng to provide the scientific basis for research and development of modern Chinese medicine derived from ginseng.Until now,201 triterpenoid compounds have been isolated and identified from ginseng.Among them,189 compounds are classified into dammarane-type and their derivatives,10 ones are classified into oleanolic acid-type and 2 ones are classified into lupane-type,which dammarane-type triterpenoids and their derivatives are predominantly dominant type not only in amount but also in structural varieties.

Abstract:Ginseng,the roots and rhizomes of Panax ginseng C.A.Mey.(Araliaceae),is one of the most commonly used traditional Chinese medicines.Of the three main kinds of ginseng,fresh ginseng is crude ginseng harvested from the land of planting,white ginseng is sun-dried ginseng,and red ginseng is produced by steaming raw ginseng.In this article,the studies on ginsenosides in the fresh ginseng,sun-dried ginseng(Ginseng Radix et Rhizoma)and red ginseng(Ginseng Radix et Rhizoma Rubra)were summarized,and the characteristics of chemical structure-types of the aglycone were investigated,to provide scientific foundation for their bioactive material base establishment.The chemical structure-types of the aglycones in the fresh and sun-dried ginsengs were oversimplified,which were divided into the three types of protopanaxadiol,protopanaxatriol and oleanolic acid.The chemical diversity of aglycone types in red ginseng were revealed,which might greatly contribute to the bioactive diversity of red ginseng.

Abstract:Ginseng has been used as a traditional herb in Asian countries for thousands of years. It contains a large number of active ingredients including steroidal saponins, protopanaxadiols, and protopanaxatriols, collectively known as ginsenosides. In the last few decades, the anti-oxidative and anti-cancer effects of ginseng, in addition to its effects on improving immunity, energy and sexuality, and combating cardiovascular diseases, diabetes mellitus, and neurological diseases have been studied in both basic and clinical research. Ginseng could be a valuable resource for future drug development; however further higher quality evidence is required. Moreover, ginseng may have drug interactions although the available evidence suggests it is a relatively safe product. An Integrated Biochemical, Proteomics, and Metabolomics Approach for Supporting Medicinal Value of Panax ginseng Fruits

Abstract:Panax ginseng roots are well known for their medicinal properties and have been used in Korean and Chinese traditional medicines for 1000s of years. However, the medicinal value of P. ginseng fruits remain poorly characterized. In this study, we used an integrated biochemical, proteomics, and metabolomics approach to look into the medicinal properties of ginseng fruits. DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS [2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid)] assays showed higher antioxidant activities in ginseng fruits than leaves or roots. Two-dimensional gel electrophoresis (2-DE) profiling of ginseng fruit proteins (cv. Cheongsun) showed more than 400 spots wherein a total of 81 protein spots were identified by mass spectrometry using NCBInr, UniRef, and an in-house developed RNAseq (59,251 protein sequences)-based databases. Gene ontology analysis showed that most of the identified proteins were related to the hydrolase (18%), oxidoreductase (16%), and ATP binding (15%) activities. Further, a comparative proteome analysis of four cultivars of ginseng fruits (cvs. Yunpoong, Gumpoong, Chunpoong, and Cheongsun) led to the identification of 22 differentially modulated protein spots. Using gas chromatography-time of flight mass spectrometry (GC-TOF MS), 66 metabolites including amino acids, sugars, organic acids, phenolic acids, phytosterols, tocopherols, and policosanols were identified and quantified. Some of these are well known medicinal compounds and were not previously identified in ginseng. Interestingly, the concentration of almost all metabolites was higher in the Chunpoong and Gumpoong cultivars. Parallel comparison of the four cultivars also revealed higher amounts of the medicinal metabolites in Chunpoong and Gumpoong cultivars. Taken together, our results demonstrate that ginseng fruits are a rich source of medicinal compounds with potential beneficial health effects.
