Abstract:A novel mixed-mode expanded bed adsorbent with anion-exchange properties was explored with benzylamine as the functional ligand. The cellulose composite matrix, densified with stainless steel powder, was prepared with the method of water-in-oil suspension thermal regeneration. High activation levels of the cellulose matrix were obtained with allyl bromide because of the relative inertness of the allyl group under the conditions of the activation reaction. After the formation of the bromohydrin with N-bromosuccinimide and coupling with benzylamine, the activated matrix was derived to function as a mixed-mode adsorbent containing both hydrophobic and ionic groups. The protein adsorption capacity was investigated with bovine serum albumin as a model protein. The results indicated that the prepared adsorbent could bind bovine serum albumin with a high adsorption capacity, and it showed salt tolerance. Effective desorption was achieved by a pH adjustment across the isoelectric point of the protein. The interactions between the cell and adsorbent were studied, and the bioadhesion was shielded by the adjustment of the salt concentration above 0.1M. Stable fluidization in the expanded bed was obtained even in a 2% (dry weight) yeast suspension. The direct capture of target proteins from a biomass-containing feedstock without extra dilution steps could be expected with the mixed-mode adsorbent prepared in this work, and this would be especially appropriate for expanded bed adsorption applications. Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:In this study, the interaction forces between different cellulosic nanomaterials and a protein domain belonging to cellulose binding modules family 1 (CBM1) were investigated at the molecular scale. Cellulose binding modules are protein domains found in carbohydrate active enzymes having an affinity toward cellulosic materials. Here, the binding force of a fusion protein containing a cellulose binding module (CBM1) produced recombinantly in E. coli was quantified on different cellulose nanocrystals immobilized on surfaces. Adhesion of the CBM on cellulose with different degrees of crystallinity as well as on chitin nanocrystals was examined. This study was carried out by single molecule force spectroscopy using an atomic force microscope, which enables the detection of binding force of individual molecules. The study contains a preliminary quantification of the interactions at the molecular level that sheds light on the development of new nanocellulose-based nanocomposites with improved strength and elasticity.

Abstract:Cellulose nanocrystal (CNC) is a relatively new natural nanoparticle derived from plants. Several biomedical applications are envisioned for CNC. Although its safety upon systemic administration is still questionable, it may serve as a worthy substrate for the adsorption of blood proteins in ex vivo settings for diagnostic purposes. The interaction between CNC and Human serum albumin (HSA) may influence both structure and function of other adsorbed proteins on the CNC surface. Herein, molecular dynamics (MD) simulation and circular dichroism spectroscopy (CD) were utilized to study the conformational changes of HSA in the presence and absence of the CNC. The secondary structure study results show that the HSA structure was conserved during the interaction. Fluorescence spectroscopy indicates that the CNC has a quenching property on the intrinsic fluorescence of HSA. Furthermore, increasing CNC concentrations led to a blue shift in the HSA fluorescence emission at 344 nm. The Gibbs free energy demonstrated the HSA and CNC interaction is spontaneous; the MMPBSA outcomes also approved that electrostatic and van der Waals (vdW) interactions were the major forces in the HSA-CNC complex. The results of this study shed light on the adsorption of HSA on CNC and the nature of their interactions.

Insights into controlling bacterial cellulose nanofiber film properties through balancing thermodynamic interactions and colloidal dynamics

Abstract:In recent years, nanocellulose has emerged as a sustainable and environmentally friendly alternative to traditional petroleum-derived structural polymers. Sourced either from plants, algae, or bacteria, nanocellulose can be processed into colloid, gel, film and fiber forms. However, the required fundamental understanding of process parameters that govern the morphology and structure-property relationships of nanocellulose systems, from colloidal suspensions to bulk materials, has not been developed and generalized for all forms of cellulose. This further hinders the more widespread adoption of this biopolymer in applications. Our study investigates the dispersion of cellulose nanofibers (CNFs) produced by a bacterial-yeast co-culture, in solvents, highlighting the role of thermodynamic interactions in influencing their colloidal behavior. By adjusting Hansen solubility parameters, we controlled the thermodynamic relationship between CNFs and solvents across various concentrations, studying the dilute to semi-dilute regimes. Rheological measurements revealed that the threshold at which a concentration-based regime transition occurs is distinctly solvent-dependent. Complementing rheological analysis with small angle X-ray scattering and zeta potential measurements, our findings reveal that enhancing CNF-solvent interactions increases excluded volume in the dilute regime, emphasizing the importance of the balance between fiber-fiber and fiber-solvent interactions. Moreover, we investigated the transition from colloidal to solid state by creating films from dispersions with varying interaction parameters in semi-dilute regimes. Through mechanical testing and scanning electron microscopy imaging of the fracture surfaces, we highlight the significance of electrokinetic effects in such transitions, as dispersions with higher electrokinetic stabilization gave rise to stronger and tougher films despite having less favorable thermodynamic interaction parameters. Our work provides insights into the thermodynamic and electrokinetic interplay that governs bacterial CNF dispersion, offering a foundation for future application and a deeper understanding of nanocellulose's colloidal and structure-property relationships.

Abstract:At ultralow shear rate (~0.01 s−1), acting below the yield stress of the aqueous gel, adsorption of calcium carbonate nanoparticles (<~100 nm) onto cellulose nanofibrils is induced without pigment–pigment preflocculation. Dispersant-free and polyacrylate treated dispersed carbonate particles are compared. Initially, it is seen that the polyacrylate dispersed material does not adsorb, whereas the dispersant-free carbonate adsorbs readily under the controlled ultralow shear conditions. However, repeated cycles of ultralow shear with intermittent periods in the rest state eventually induce the effect as initially seen with the dispersant-free calcium carbonate. The fibril suspension in the bulk is slightly acidic. The addition of buffer to a controlled pH in the case of the dispersant treated particles maintained a similar delay in the onset of adsorption, but adsorption occurred eventually after repeated cycles. During this cycling process, in parallel, the pH gradually drops under repeated cycles of ultralow shear, opposite to expectation, given the buffering capacity of calcium carbonate. The conductivity, in turn, progressively increases slightly at first and then significantly. The action of surface bound water on the nanofibril is considered key to the action of adsorption, and the condition of ultralow shear suggests that the residence time of the particle in contact with the nanofibril, acting under controlled strain against diffusion in the gel, is critical. It is proposed that under these specific conditions the calcium carbonate nanoparticles act as a probe of the nanofibril surface chemistry. The hydrogen bonded water, known to reside at the nanofibril surface, is thus considered the agent in the carbonate-surface interaction, effectively expressing an acid dissociation, and the calcium carbonate nanoparticles act as the probe to reveal it. An important phenomenon associated with this acid dissociation behaviour is that the adsorbed calcium carbonate particles subsequently act to flocculate the otherwise stable cellulose material, leading to release of water held in the aqueous gel matrix structure. This latter effect has major implications for the industrial ease of use of micro and nanofibrillar cellulose at increased solids content. This novel mechanism is also proposed for use to enhance the dewatering capability in general of complex cellulose-containing gel-like water-holding suspensions.

Abstract:Cellulose II nanospheres (NPcat, 63 nm diameter) bearing a soft and highly accessible shell are produced and endow rapid and significant binding (16 mg cm−2) with protein (bovine serum albumin,BSA). In all‐water PEG/dextran emulsion systems, BSA and NPcat diffuse to droplet interfaces through electrostatic effects, forming an elastic layer structure enveloping the emulsion droplets and leading to stable W/W emulsions. Pickering water‐in‐water (W/W) emulsions stabilized by biobased colloids are pertinent to engineering biomaterials with hierarchical and confined architectures. In this study, stable W/W emulsions are developed through membranization utilizing biopolymer structures formed by the adsorption of cellulose II nanospheres and a globular protein, bovine serum albumin (BSA), at droplet surfaces. The produced cellulose II nanospheres (NPcat, 63 nm diameter) bearing a soft and highly accessible shell, endow rapid and significant binding (16 mg cm−2) with BSA. NPcat and BSA formed complexes that spontaneously stabilized liquid droplets, resulting in stable W/W emulsions. It is proposed that such a system is a versatile all‐aqueous platform for encapsulation, (bio)catalysis, delivery, and synthetic cell mimetics.

Abstract:Nanobiocatalysis is a new frontier of emerging nanosized material support in enzyme immobilization application. This paper is about a comprehensive review on cellulose nanofibers (CNF), including their structure, surface modification, chemical coupling for enzyme immobilization, and potential applications. The CNF surface consists of mainly –OH functional group that can be directly interacted weakly with enzyme, and its binding can be improved by surface modification and interaction of chemical coupling that forms a strong and stable covalent immobilization of enzyme. The knowledge of covalent interaction for enzyme immobilization is important to provide more efficient interaction between CNF support and enzyme molecule. Enzyme immobilization onto CNF is having potential for improving enzymatic performance and production yield, as well as contributing toward green technology and sustainable sources.

Universal method for protein bioconjugation with nanocellulose scaffolds for increased cell adhesion

Abstract:Bacterial nanocellulose (BNC) is an emerging biomaterial since it is biocompatible, integrates well with host tissue and can be biosynthesized in desired architecture. However, being a hydrogel, it exhibits low affinity for cell attachment, which is crucial for the cellular fate process. To increase cell attachment, the surface of BNC scaffolds was modified with two proteins, fibronectin and collagen type I, using an effective bioconjugation method applying 1-cyano-4-dimethylaminopyridinium (CDAP) tetrafluoroborate as the intermediate catalytic agent. The effect of CDAP treatment on cell adhesion to the BNC surface is shown for human umbilical vein endothelial cells and the mouse mesenchymal stem cell line C3H10T1/2. In both cases, the surface modification increased the number of cells attached to the surfaces. In addition, the morphology of the cells indicated more healthy and viable cells. CDAP activation of bacterial nanocellulose is shown to be a convenient method to conjugate extracellular proteins to the scaffold surfaces. CDAP treatment can be performed in a short period of time in an aqueous environment under heterogeneous and mild conditions preserving the nanofibrillar network of cellulose.

Abstract:Abstract This study describes the preparation and characterization of full atomistic models of amorphous cellulose and calcium carbonate (CaCO 3 ) nanocomposite to assess its mechanical properties within and beyond the elastic limit via molecular dynamics simulations. The interactions by hydrogen bond and conformation of the cellulose molecules from the assessment of torsional angles were specifically monitored during the tensile stretching simulations to get deep understanding of the possible structural changes produced in the material during the deformation. On the one hand, the results showed a favorable interaction of the cellulose matrix with the calcium carbonate nanoparticle, with the electrostatic contribution being dominant over the van der Waals component. The determined mechanical elastic constants indicated that the inclusion of the CaCO 3 nanoparticle provided an increase on the rigidity of the composite system of 15%, 18% and 19% in the Young, shear or bulk modulus, respectively. On the other hand, using extension and compression simulations, the recovery capacity of the material systems was also assessed in terms of plastic deformation. The elastoplastic behavior was observed for either the neat or the CaCO 3 nanocomposite, with an elastic limit around 2.5%. The results also showed that the presence of the CaCO 3 nanoparticle produced higher values of plastic deformation in the composite material compared to the neat cellulose system and thus decreased the flexibility of the material. A hysteresis mechanism was identified together with irreversible conformational changes on the cellulose molecules which would explain the plastic deformation observed on the cellulosic systems. It was concluded that the higher plastic deformations observed in the nanocomposite system would be a result of the disruption of the network of hydrogen bonds and the associated decrease on the number of possible interactions. Abstract:Crystalline nanocellulose is widely used for example, in the paper-making and food industries, as support matrix material or reinforcement of polymer materials, but also in drug carrier and nanomedicine applications. Interestingly, aqueous solutions of cellulose are extremely sensitive to small amounts of added salt yet mere considerations of charge screening leave open questions regarding the mechanisms, especially for unmodified cellulose in aqueous solutions. Here, we map NaCl ion distributions and the effect of added NaCl salt on the hydration of I cellulose nanocrystal (CNC) surfaces by atomistic detail molecular dynamics simulations with explicit water solvent. The simulations reveal the dependency of the hydration layers of the six surfaces of CNCs on the ions, as well as NaCl ion binding sites, and preferences in terms of binding free energy for the ions near CNC surfaces at different NaCl concentrations. We discuss the modelling results against our prior rheology characterization of cellulose solutions. Together, the results indicate that the high sensitivity of cellulose aqueous solutions to added salt rises from the ions near the surface changing locally the ordering and structure of the hydration layers of the CNC surfaces. The revealed mechanism of salt-induced viscosity changes in cellulose aqueous solutions allows advanced design of gelling CNC systems for various end uses and may also guide tuning cellulose interactions by different solvent environments.

Abstract:Bacterial nanocellulose (BNC) is a biodegradable polysaccharide with unique properties that make it an attractive material for various industrial applications. This study focuses on the strain Komagataeibacter medellinensis ID13488, a strain with the ability to produce high yields of BNC under acidic growth conditions and a promising candidate to use for industrial production of BNC. We conducted a comprehensive investigation into the effects of downstream treatments on the structural and mechanical characteristics of BNC. When compared to alkaline-treated BNC, autoclave-treated BNC exhibited around 78% superior flexibility in average, while it displayed nearly 40% lower stiffness on average. An SEM analysis revealed distinct surface characteristics, indicating differences in cellulose chain compaction. FTIR spectra demonstrated increased hydrogen bonding with prolonged interaction time with alkaline solutions. A thermal analysis showed enhanced thermal stability in alkaline-treated BNC, withstanding temperatures of nearly 300 °C before commencing degradation, compared to autoclaved BNC which starts degradation around 200 °C. These findings provide valuable insights for tailoring BNC properties for specific applications, particularly in industries requiring high purity and specific mechanical characteristics.

Abstract:Electrochemical biosensing devices are known for their simple operational procedures, low fabrication cost, and suitable real-time detection. Despite these advantages, they have shown some limitations in the immobilization of biochemicals. The development of alternative materials to overcome these drawbacks has attracted significant attention. Nanocellulose-based materials have revealed valuable features due to their capacity for the immobilization of biomolecules, structural flexibility, and biocompatibility. Bacterial nanocellulose (BNC) has gained a promising role as an alternative to antifouling surfaces. To widen its applicability as a biosensing device, BNC may form part of the supports for the immobilization of specific materials. The possibilities of modification methods and in situ and ex situ functionalization enable new BNC properties. With the new insights into nanoscale studies, we expect that many biosensors currently based on plastic, glass, or paper platforms will rely on renewable platforms, especially BNC ones. Moreover, substrates based on BNC seem to have paved the way for the development of sensing platforms with minimally invasive approaches, such as wearable devices, due to their mechanical flexibility and biocompatibility.

Abstract:Biomaterials of different nature have been and are widely studied for various biomedical applications. In many cases, biomaterial assemblies are designed to mimic biological systems. Although biomaterials have been thoroughly characterized in many aspects, not much quantitative information on the molecular level interactions between different biomaterials is available. That information is very important, on the one hand, to understand the properties of biological systems and, on the other hand, to develop new composite biomaterials for special applications. This work presents a systematic, quantitative analysis of self- and cross-interactions between films of collagen I (Col I), collagen IV (Col IV), laminin (LN-521), and cellulose nanofibrils (CNF), that is, biomaterials of different nature and structure that either exist in biological systems (e.g., extracellular matrices) or have shown potential for 3D cell culture and tissue engineering. Direct surface forces and adhesion between biomaterials-coated spherical microparticles and flat substrates were measured in phosphate-buffered saline using an atomic force microscope and the colloidal probe technique. Different methods (Langmuir-Schaefer deposition, spin-coating, or adsorption) were applied to completely coat the flat substrates and the spherical microparticles with homogeneous biomaterial films. The adhesion between biomaterials films increased with the time that the films were kept in contact. The strongest adhesion was observed between Col IV films, and between Col IV and LN-521 films after 30 s contact time. In contrast, low adhesion was measured between CNF films, as well as between CNF and LN-521 films. Nevertheless, a good adhesion between CNF and collagen films (especially Col I) was observed. These results increase our understanding of the structure of biological systems and can support the design of new matrices or scaffolds where different biomaterials are combined for diverse biological or medical applications.
Regioselective and Water-Promoted Surface Esterification of Never-Dried Cellulose Fibers Towards Nanofibers with Adjustable Surface Energy

Abstract:A new regioselective route is introduced for surface modification of biological colloids in the presence of water. Taking the case of cellulose nanofibers (CNFs), we demonstrate a site-specific (93% selective) reaction between the primary surface hydroxyl groups (C6-OH) of cellulose and acyl imidazoles. CNFs bearing C6-acetyl and C6-iso-butyl groups, with a degree of substitution up to 1 mmol·g–1 were yielded upon surface esterification, affording nanofibers of adjustable surface energy. The morphological and structural features of the nanofibers remained largely unaffected, but the regioselective surface reactions enabled tailoring of interfacial interactions, as demonstrated in oil/water Pickering emulsions. Our method precludes the need for drying or solvent exchange to organic solvents for surface esterifications, which are otherwise demanded in the fabrication of esterified colloids and polysaccharides and can be potentially implemented into reactive extrusion and compounding of high-solid content colloids. The proposed acylation is expected to open new and sustainable approaches that benefit from the presence of water and the high selectivity.

Nanocellulose as a New Degradable Suspension Additive for High-Density Calcium Brines

Abstract:Abstract Calcium-based brines, principally calcium chloride and calcium bromide, find widespread use as completion fluids and base fluids for reservoir drill-in fluids (RDF). The high ionic strength of these brines, combined with the chemical compatibility issues inherent with divalent ions, such as calcium, makes identifying additives for developing functional fluids challenging. While additives, such as hydroxyethyl cellulose, can provide viscosity to calcium brines, imparting the suspension properties needed for the transport of solid bridging agents or drill cuttings can be particularly problematic in divalent brines. Even the widely used and potent suspension agent, xanthan gum, in addition to being difficult to degrade, suffers from gelation and precipitation under certain conditions in high-calcium environments. In this study, nanocellulose, which has previously shown promise as a suspension additive in freshwater and low-salinity brines, was evaluated in high-density brine fluids. Extensive laboratory evaluation of the commercially sourced nanocellulose was conducted in sodium chloride, calcium chloride, and calcium bromide brines. Rheological determinations were made of simple systems containing only brine plus the novel additive, as well as fully formulated RDF containing the additive with sized calcium carbonate bridging solids, alkalinity, and filtration control additives. This data was compared before and after aging at various temperatures and was further compared to other biopolymers that are used today in divalent brines. Stability of the fluids and the capability of the formulated fluids to suspend solids were also examined. Fluid-loss control of full formulations was compared under API conditions. Single-phase return permeability studies were conducted. While results in sodium brines were mixed, nanocellulose was shown to outperform existing biomaterials in calcium brines. Nanocellulose was shown to be readily hydrated in the challenging high-density divalent environment and provided superior suspension to currently used fluid components at similar concentrations. Rheological performance of nanocellulose was found to be synergistic with bridging solids. In one particularly interesting example, it was also shown to complement a synthetic polymer in a high-temperature calcium bromide application. Nanocellulose provides superior rheological performance and reduced formation damage potential compared to conventional materials in difficult high-density calcium chloride and bromide brines. The improved rheological profile and enhanced thermal stability of fluids formulated with nanocellulose, combined with a greater diversity of removal options, can facilitate expansion of the utility of calcium-based brines in well construction and completion applications.

Abstract:Concentrated fabric conditioners are water-based formulations containing 10 wt. % of cationic surfactants that are deposited on textile fibers during the rinse cycle in a washing machine to make them smoother and softer to touch. In topical formulations, the concentration of cationic surfactants is reduced by half, this reduction being compensated by the addition of environment-friendly polysaccharides. Using atomic force microscopy (AFM), quartz crystal microbalance with dissipation (QCM-D) and ellipsometry, the deposition of formulations with or without polysaccharides on model cellulose substrates is studied. We found that 180 nm long cellulose nanocrystals (CNCs) deposited by spin-coating on amorphous silicon dioxide or on quartz crystal sensors provide a good model of cellulose surfaces. QCM-D results reveal strong electrostatic interactions of the surfactant vesicles and polysaccharides with the CNC layer. In the presence of polysaccharides, the adsorbed quantities are less (by 60%) than the sum of the respective amounts of each component, the structure of the interface being however more homogenous and rigid. This outcome suggests that surface techniques coupled with CNC coated substrates are promising for studying interactions of current formulations with cellulose surfaces.
