Abstract:A sensitive, reliable, and cost-effective detection for SARS-CoV-2 was urgently needed due to the rapid spread of COVID-19. Here, a "signal-on" magnetic-assisted PEC immunosensor was constructed for the quantitative detection of SARS-CoV-2 nucleocapsid (N) protein based on Z-scheme heterojunction. Fe3O4@SiO2@Au was used to connect the capture antibody to act as a capture probe (Fe3O4@SiO2@Au/Ab1). It can extract target analytes selectively in complex samples and multiple electrode rinsing and assembly steps were avoided effectively. CdTe QDs sensitized TiO2 coated on the surface of SiO2 spheres to form Z-scheme heterojunction (SiO2@TiO2@CdTe QDs), which broadened the optical absorption range and inhibited the quick recombination of photogenerated electron/hole of the composite. With fascinating photoelectric conversion performance, SiO2@TiO2@CdTe QDs were utilized as a signal label, thus further realizing signal amplification. The migration mechanism of photogenerated electrons was further deduced by active material quenching experiment and electron spin resonance (ESR) measurement. The elaborated immunosensor can detect SARS-CoV-2 N protein in the linear range of 0.005-50 ng mL-1 with a low detection limit of 1.8 pg mL-1 (S/N = 3). The immunosensor displays extraordinary sensitivity, strong anti-interference, and high reproducibility in detecting SARS-CoV-2 N protein, which envisages its potential application in the clinical diagnosis of COVID-19.

Abstract:Dependable, specific and rapid diagnostic methods for severe acute respiratory syndrome beta-coronavirus (SARS-CoV-2) detection are needed to promote public health interventions for coronavirus disease 2019 (COVID-19). Herein, we have established an entropy-driven amplified electrochemiluminescence (ECL) strategy to detect the RNA-dependent RNA polymerase (RdRp) gene of SARS-CoV-2 known as RdRp-COVID which as the target for SARS-CoV-2 plays an essential role in the diagnosis of COVID-19. For the construction of the sensors, DNA tetrahedron (DT) is modified on the surface of the electrode to furnish robust and programmable scaffolds materials, upon which target DNA-participated entropy-driven amplified reaction is efficiently conducted to link the Ru (bpy)(3)(2+) modified S3 to the linear ssDNA at the vertex of the tetrahedron and eventually present an "ECL on" state. The rigid tetrahedral structure of the DT probe enhances the ECL intensity and avoids the cross-reactivity between single-stranded DNA, thus increasing the sensitivity of the assays. The enzyme-free entropy-driven reaction prevents the use of expensive enzyme reagents and facilitates the realization of large-scale screening of SARS-CoV-2 patients. Our DT-based ECL sensor has demonstrated significant specificity and high sensitivity for SARS-CoV-2 with a limit of detection (LOD) down to 2.67 fM. Additionally, our operational method has achieved the detection of RdRp-COVID in human serum samples, which supplies a reliable and feasible sensing platform for the clinical bioanalysis.

Abstract:Direct detection of SARS-CoV-2 in biological specimens is often challenging due to the low abundance of viral components and lack of enough sensitivity. Herein, we developed a new type of chemiluminescent functionalized magnetic nanomaterial for sensitive detection of the SARS-CoV-2 antigen. First, HAuCl4 was reduced by N-(aminobutyl)-N-(ethylisoluminol) (ABEI) in the presence of amino magnetic beads (MB-NH2) to generate ABEI-AuNPs, which were directly assembled on the surface of MB-NH2. Then, Co2+ was modified onto the surface to form MB@ABEI-Au/Co2+ (MAA/Co2+). MAA/Co2+ exhibited good chemiluminescence (CL) and magnetic properties. It was also found that it was easy for the antibody to be connected with MAA/Co2+. Accordingly, MAA/Co2+ was used as a sensing interface to construct a label-free immunoassay for rapid detection of the N protein in SARS-CoV-2. The immunoassay showed a linear range from 0.1 pg/mL to 10 ng/mL and a low detection limit of 69 fg/mL, which was superior to previously reported methods for N protein detection. It also demonstrated good selectivity by virtue of magnetic separation, which effectively removed a sample matrix after immunoreactions. It was successfully applied for the detection of the N protein in spiked human serum and saliva samples. Furthermore, the immunoassay was integrated with an automatic CL analyzer with magnetic separation to detect the N protein in patient serums and rehabilitation patient serums with satisfactory results. Thus, the CL immunoassay without a complicated labeling procedure is sensitive, selective, fast, simple, and cost-effective, which may be used to combat the COVID-19 pandemic. Finally, the CL quenching mechanism of the N protein in the immunoassay was also explored.

Abstract:The repeated outbreaks of coronavirus disease 2019 (COVID-19) have seriously threatened human health and economic development. Therefore, it is still significant to explore an efficient method for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) detection. Herein, we designed a dual-mode colorimetric and fluorescent immunosensor based on Fe 2+ doped ZIF-67 derivatives-based composites, Pt nanoparticles loaded on CoFe nanoparticles embedded porous carbon-nitrogen hexahedron (CoFe/NC@Pt), for detecting SARS-CoV-2 nucleocapsid protein. The doping of Fe 2+ generated bimetallic CoFe nanoparticles and formed more defects after pyrolysis. Meanwhile, the peroxides-like activity significantly improved owning to the synergistic effect of Co and Fe nanoparticles. Furthermore, loaded Pt nanoparticles not only provided more active sites, but also could couple antibodies by modifying functional groups. CoFe/NC@Pt exhibited good peroxidase-like activity and could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to blue oxides in the presence of H 2 O 2 , generating visible color signals. While the oxTMB could quench the fluorescence of carbon dots through inner filter effect, thereby providing fluorescent signals. The limit of detection were 0.022 ng/mL (colorimetric) and 0.018 ng/mL (fluorescent). And the specificity of the immunosensor was verified by comparing with other interferents. Therefore, this dual-mode immunosensor with good performance provided a reliable strategy for diagnosis of COVID-19.

Abstract:In this work, we propose a Ti3C2Tx-based electrochemiluminescence (ECL) sensing platform, combined with a programmable entropy-driven cycling strategy and inverted DNA tetrahedral nanomachines to detect the SARS-CoV-2 RdRp gene. The use of Ti3C2Tx in the ECL sensing platform provides unique advantages that improve the assay's sensitivity. Firstly, the SARS-CoV-2 RdRp gene triggers an entropy-driven cyclic amplification reaction to obtain a large number of Output strands. Subsequently, the Output strand reacts with an inverted DNA tetrahedral swing arm strand to start the self-driven process. Finally, the ferrocene (Fc)-labeled hairpin DNA strand of the inverted DNA tetrahedron is severed to achieve the signal change. Under optimal conditions, the ECL platform integrated with entropy-driven cycle amplification reaction and inverted DNA tetrahedral nano-machines obtained a wide range of 100 aM to 10 nM and a limit of detection (LOD) of 48.3 aM for the SARS-CoV-2 RdRp gene. Moreover, the recovery rates of 97% to 103.3% and 98.34% to 103% were obtained in real environments and human pharyngeal swabs, respectively. This ECL sensing platform, which combines an entropy-driven cycle amplification reaction and inverted DNA tetrahedral nanomachines, opens up new opportunities for the detection of other substances.

Abstract:A novel porphyrin‐based heterobimetallic 2D MOF, [(ZnTCPP)Co2(MeIm)] (1) is constructed to act as an excellent electrochemiluminescence probe for rapid nonamplified detection of SARS‐CoV‐2. High electrocatalytic activity with tunable luminescence is crucial for the development of electrochemiluminescence (ECL) luminophores. In this study, a porphyrin‐based heterobimetallic 2D metal organic framework (MOF), [(ZnTCPP)Co2(MeIm)] (1), is successfully self‐assembled from the zinc(II) tetrakis(4‐carboxyphenyl)porphine (ZnTCPP) linker and cobalt(II) ions in the presence of 2‐methylimidazole (MeIm) by a facile one‐pot reaction in methanol at room temperature. On the basis of the experimental results and the theoretical calculations, the MOF 1 contains paddle–wheel [Co2(‐CO2)4] secondary building units (SBUs) axially coordinated by a MeIm ligand, which is very beneficial to the electron transfer between the Co(II) ions and oxygen. Combining the photosensitizers ZnTCPP and the electroactive [Co2(‐CO2)4] SBUs, the 2D MOF 1 possesses an excellent ECL performance, and can be used as a novel ECL probe for rapid nonamplified detection of the RdRp gene of SARS‐CoV‐2 with an extremely low limit of detection (≈30 aM).

Abstract:Early detection of the coronavirus is acknowledged as a crucial measure to mitigate the spread of the pandemic, facilitating timely isolation of infected individuals, and disrupting the transmission chain. In this study, we leveraged the properties of synthesized Ag-MOF, including high porosity and increased flow intensity. Electrochemical techniques such as cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were employed to develop an economical and portable sensor with exceptional selectivity for COVID-19 detection. The methodology involves the deposition of Ag-MOF onto the surface of a Glassy Carbon Electrode (GCE), which resulted in a progressive augmentation of electric current. Subsequently, the targeted antibodies were applied, and relevant tests were conducted. The sensor demonstrated the capacity to detect the virus within a linear range of 100 fM to 10 nM, boasting a noteworthy Limit of Detection (LOD) of 60 fM. The entire detection process could be completed in a brief duration of 20 min, exhibiting high levels of accuracy and precision, outperforming comparable techniques in terms of speed and efficacy.

Abstract:The COVID-19 pandemic caused by SARS-CoV-2 has precipitated a global public health emergency, necessitating the urgent development of rapid and accurate virus detection methods. Compared with the current single-signal detection methods commonly used, dual-mode detection has inherent self-verification and self-correction functions and is expected to provide more reliable and interference-resistant detection results. In this work, a rapid approach for SARS-CoV-2 detection was developed using an electrochemical and colorimetric immunosensor based on a NiFe layered double hydroxide (LDH) loaded with silver citrate (NiFe-LDH@AgCit). The electrochemical sensor is capable of measuring the electrical signal of the transition from Ag to Ag+ in the presence of SARS-CoV-2. At the same time, the color reaction of 3,3 ',5,5 '-tetramethylbenzidine with & sdot;OH can be used to build a colorimetric sensor. In electrochemical and colorimetric sensing systems, the linear detection ranges were 0.0005-10 and 0.001-10 ng mL- 1, respectively. The corresponding limits of detection reached as low as 0.167 and 0.333 pg mL- 1, respectively. In addition, the dual-signal sensor has been favorably used for the detection of SARS-CoV-2 in human serum, and the recovery ranges were 99.0 %-101.6 % and 100.6 %-104.5 %, respectively. In conclusion, the proposed dual-signal sensing technique offers novel insights for the prompt diagnosis of virus-related illnesses, which has wide application prospects.
A novel photoelectrochemical immunosensor based on TiO2@Bi2WO6 hollow microspheres and Ag2S for sensitive detection of SARS-COV-2 nucleocapsid protein

Abstract:Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-COV-2) is a cluster of β coronaviruses. The 2019 coronavirus disease (COVID-19) caused by SARS-COV-2 is emerging as a global pandemic. Thus, early diagnosis of SARS-COV-2 is essential to prevent severe outbreaks of the disease. In this experiment, a novel label-free photoelectrochemical (PEC) immunosensor was obtained based on silver sulfide (Ag2S) sensitized titanium dioxide@bismuth tungstate (TiO2@Bi2WO6) nanocomposite for quantitative detection of SARS-COV-2 nucleocapsid protein. The constructed TiO2@Bi2WO6 hollow microspheres had large specific surface area and could produce high photocurrent intensity under visible light illumination. Ag2S was in-situ grown on the surface of thioglycolic acid (TGA) modified TiO2@Bi2WO6. In particular, TiO2@Bi2WO6 and Ag2S formed a good energy level match, which could effectively enhance the photocurrent conversion efficiency and strength the photocurrent response. Ascorbic acid (AA) acted as an effective electron donor to effectively eliminate photogenerated holes. Under optimal experimental conditions, the constructed immunosensor presented a supersensitive response to SARS-COV-2 nucleocapsid protein, with a desirable linear relationship ranged from 0.001 to 50 ng/mL for nucleocapsid protein and a lower detection limit of 0.38 pg/mL. The fabricated sensor exhibited a wide linear range, excellent selectivity, specificity and stability, which provided a valuable referential idea for the detection of SARS-COV-2.

Abstract:The COVID-19 pandemic has become a global catastrophe, affecting the health and economy of the human community. It is required to mitigate the impact of pandemics by developing rapid molecular diagnostics for SARS-CoV-2 virus detection. In this context, developing a rapid point-of-care (POC) diagnostic test is a holistic approach to the prevention of COVID-19. In this context, this study aims at presenting a real-time, biosensor chip for improved molecular diagnostics including recombinant SARS-CoV-2 spike glycoprotein and SARS-CoV-2 pseudovirus detection based on one-step-one-pot hydrothermally derived CoFeBDCNH 2 -CoFe 2 O 4 MOF-nanohybrids. This study was tested on a PalmSens-EmStat Go POC device, showing a limit of detection (LOD) for recombinant SARS-CoV-2 spike glycoprotein of 6.68 fg/mL and 6.20 fg/mL in buffer and 10% serum-containing media, respectively. To validate virus detection in the POC platform, an electrochemical instrument (CHI6116E) was used to perform dose dependent studies under similar experimental conditions to the handheld device. The results obtained from these studies were comparable indicating the capability and high detection electrochemical performance of MOF nanocomposite derived from one-step-one-pot hydrothermal synthesis for SARS-CoV-2 detection for the first time. Further, the performance of the sensor was tested in the presence of Omicron BA.2 and wild-type D614G pseudoviruses.

Abstract:Surface-enhanced Raman scattering (SERS) technique with high sensitivity, reliable specificity, and rapid recognition ability exhibits attractive promise for the effective fast-monitoring of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Herein, a novel SERS-active immunoassay kit for SARS-CoV-2 nucleocapsid (N) protein was prepared by in-situ growing gold (Au) nanowire forests (NFs) onto Ti 3 C 2 T x , which was then modified onto polymethyl methacrylate (PMMA) matrix and encapsulated into kit. It was noted that the Au nanowires with fibrous structures which vertically anchored on Ti 3 C 2 T x served as perfect channels to promote photo-induced charge transfer. The synergistic action of electromagnetic and chemical effects resulted in an enhancement factor (EF) of 1.27 × 10 7 . Furthermore, the unreliable fluctuation of the enhanced signal was eliminated by using the intrinsic Raman signal of the flexible PMMA platform, achieving an improved correlation coefficient (R 2 ) value from 0.950 to 0.990. Moreover, the as-designed immunoassay kit with both high sensitivity and remedied quantitative ability rendered by the Ti 3 C 2 T x @Au NFs-PMMA composite exhibited a powerful performance in the practical detection of N-protein with concentration low to 5.0 × 10 −8 mg/mL.

Abstract:The rapid epidemic around the world of coronavirus disease 2019 (COVID-19), caused by the SARS-CoV-2 virus, proves the need and stimulates efforts to explore efficient diagnostic tests for the sensitive detection of the SARS-CoV-2 virus. An aggregation-induced electrochemiluminescence (AIECL) sensor was developed for the ultrasensitive detection of the SARS-CoV-2 nucleocapsid (N) protein in this work. Tetraphenylethylene doped in zeolite imidazole backbone-90 (TPE-ZIF-90) showed highly efficient aggregation-induced emission (AIE) to endow TPE-ZIF-90 with high ECL intensity. Upon the capture of the SARS-CoV-2 N protein by immune recognition, an alkaline phosphatase (ALP)-modified gold nanoparticle (AuNP)-decorated zinc oxide (ZnO) nanoflower (ALP/Au-ZnO) composite was introduced on the sensing platform, which catalyzed L-ascorbate-2-phosphate trisodium salt (AA2P) to produce PO43- and ascorbic acid (AA). Based on a multiquenching of the ECL signal strategy, including resonance energy transfer (RET) between TPE-ZIF-90 and Au-ZnO, disassembly of TPE-ZIF-90 triggered by the strong coordination between PO43- and Zn2+, and RET between TPE-ZIF-90 and AuNPs produced in situ by the AA reductive reaction, the constructed AIECL sensor achieved highly sensitive detection of the SARS-CoV-2 N protein with a low limit of detection of 0.52 fg/mL. With the merits of high specificity, good stability, and proven application ability, the present RET- and enzyme-triggered multiquenching AIECL sensor may become a powerful tool in the field of SARS-CoV-2 virus diagnosis.

Abstract:Tracking and monitoring of low concentrations of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can effectively control asymptomatic transmission of current coronavirus disease 2019 (COVID-19) in the early stages of infection. Here, we highlight an electrochemical immunosensor for sensitive detection of SARS-CoV-2 antigen marker spike protein. The surface-clean Pd-Au nanosheets as a substrate for efficient sensing and signal output have been synthesized. The morphology, chemical states and excellent stable electrochemical properties of this surface-clean heterostructures have been studied. Functionalized superparamagnetic nanoparticles (MNPs) were introduced as sample separators and signal amplifiers. This biosensor was tested in phosphate buffered saline (PBS) and nasopharyngeal samples. The results showed that the sensor has a wide linear dynamic range (0.01 ng mL-1 to 1000 ng mL-1) with a low detection limit (0.0072 ng mL-1), which achieved stable and sensitive detection of the spike protein. Therefore, this immunosensing method provides a promising electrochemical measurement tool, which can furnish ideas for early screening and the reasonable optimization of detection methods of SARS-CoV-2.

Abstract:Here we put forward an innovative "one master and two servants" strategy for enhancing the ECL performance. A novel ECL luminophore named Zr-TCPP/NH 2 -BDC ( 2 ) was synthesized by self-assembly of meso-tetra(4-carboxyphenyl)porphine (TCPP) and 4-aminobenzoic acid (NH 2 -BDC) with Zr clusters. 2 has a porous structure and a highly ordered structure, which allows the molecular motion of TCPP to be effectively confined, thereby inhibiting nonradiative energy transfer. Importantly, 2 has a higher and more stable ECL signal. To further improve the sensitivity of the sensor, we use polydopamine-coated manganese dioxide ( 2 ), which has a double quenching effect, as the quencher. The nucleocapsid (N) protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2-N) is one of the ideal markers for the early diagnosis of COVID-19, and its sensitivity detection is of great significance for the prevention and treatment of COVID-19. Thus, we constructed a quenching-type ECL sensor for the ultrasensitive detection of the SARS-CoV-2-N. Its linear range is 10 fg/mL∼1 μg/mL and the calculated detection limit is 1.4 fg/mL (S/N = 3). The spiked recoveries are 97.40–103.8%, with the relative standard deviations (RSD) under 3.0%. More importantly, the technique offers a viable way to identify and diagnose viral infections early.

Abstract:Electrochemiluminescence lateral flow immunoassay (ECL‐LFIA) strips are prepared by fabricating three electrodes on a nitrocellulose membrane, which simplifies electrode fixation and reduces the distance between the ECL luminophore and electrode surface, promoting ECL reactions. Using ruthenium(II) complex‐loaded dendritic mesoporous silica nanospheres as tags (PEI‐Ru/dSiO2), the ECL‐LFIA strip enables the rapid and quantitative detection of disease biomarkers. Lateral flow immunoassays (LFIA) are widely used for the cost‐effective and rapid detection of diverse analytes. However, traditional LFIA suffers from difficulties in providing quantitative results and has low sensitivity. Herein, LFIA is combined with electrochemiluminescence (ECL), a leading transduction technique with high sensitivity and wide dynamic range, to achieve highly sensitive and quantitative detection of severe acute respiratory syndrome coronavirus nucleocapsid protein (SARS‐CoV‐2 N protein). Ruthenium(II)‐based complexes are synthesized and loaded into dendritic mesoporous silica nanospheres (PEI‐Ru/dSiO2), which possessed central‐radial pore channels and served as tags for ECL‐LFIA. The electrodes are fabricated on a nitrocellulose (NC) membrane, which simplifies the structure of the ECL‐LFIA. PEI‐Ru/dSiO2 is captured on the electrode surface via a sandwich immunoreaction, which enhances the ECL signal by decreasing the distance between PEI‐Ru/dSiO2 and the electrode surface. Using 2,2‐bis(hydroxymethyl)‐2,2′,2′'‐nitrilotriethanol (BIS‐TRIS) as coreactant, the ECL‐LFIA is used for detecting SARS‐CoV‐2 N protein, with a linear range of 1–104 ng mL−1 and a limit of detection (LOD) of 0.52 ng mL−1. ECL‐LFIA can also be used to detect analytes in complex matrices. These results demonstrate that the prepared ECL‐LFIA has great potential as a point‐of‐care testing platform for the rapid and quantitative detection of disease biomarkers.

Abstract:Serological antibody tests are useful complements of nuclei acid detection for SARS-CoV-2 diagnosis, which can significantly improve diagnostic accuracy. However, antibody detection in serum or plasma remains challenging to do with high sensitivity. In this study, Ag nanoparticles with ultra-thin Au shells embedded with 4-mercaptobenzoic acid (MBA) (Ag MBA @Au) were manufactured and then assembled onto Fe 3 O 4 surface by electrostatic interaction to construct the Fe 3 O 4 -Ag MBA @Au nanoparticles (NPs) with magnetic-Raman-colorimetric properties. Based on the composite nanoparticles, a colorimetric and Raman dual-mode lateral flow immunoassay (LFIA) for ultrasensitive identification of SARS-CoV-2 nucleocapsid (N) protein antibody was constructed. The magnetic nanoparticles (Fe 3 O 4 NPs) were acted as the core and coated a layer of Ag MBA @Au particles on the surface by electrostatic interaction to prepare Fe 3 O 4 -Ag MBA @Au NPs, which can amplify the SERS signal due to multiple Ag MBA @Au particles concentrated on a single magnetic nanoparticle. Moreover, the Fe 3 O 4 -Ag MBA @Au NPs facilitated pre-purifying sample using magnetic separation, and complex matrix interference would be greatly decreased in the detection. The Fe 3 O 4 -Ag MBA @Au NPs modified with N protein recognized and bound with N protein antibodies, which were trapped on the T-line, forming color band for observing detection. Under optimal conditions, the N protein antibodies could be qualitatively detected in colorimetric mode with the visual limit of 10 −8 mg/mL and quantitatively detected by SERS signals between 10 −6 and 10 −10 mg /mL with 0.08 pg/mL detection limit. The coefficients variations (CV) of intra-assay was 8.0%, whereas of inter-assay was 11.7%, confirming of good reproducibility. Finally, this approach was able to discriminate between positive, negative, and weakly positive samples when detecting 107 clinical serum samples. The process enables highly sensitive quantitative assays that are valuable for evaluating disease processes and guiding treatment.

Abstract:The prompt and accurate point-of-care test (POCT) for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in infected persons or virus-containing environmental samples is of great importance. The present work reports a highly integrated electrochemiluminescence/electrochemical (ECL/EC) sensor for determination of SARS-CoV-2 pseudoviruses, in which bio-recognition element (SARS-CoV-2 IgG antibody), bifunctional probe (tris (2,2'-bipyridyl) ruthenium (Ru(bpy)32+)), and amplification material (gold nanoparticles (Au NPs)) are designed into bipolar silica nanochannel array (bp-SNA). bp-SNA consisting of homogeneous two-layer mesoporous silica films bears inner silanol groups and outer amino groups, generating a solid "electrostatic nanocage" for stable confinement of Ru(bpy)32+ and Au NPs inside the nanochannels and further providing functional sites for covalent modification of SARS-CoV-2 IgG antibody. Owing to the preconcentration capacity of bp-SNA and amplified effect of Au NPs, ECL or EC signals of Ru(bpy)32+ can be remarkably promoted and thereby increase the analytical performance, which can be diminished by immunorecognization of target SARS-CoV-2 pseudoviruses on the sensing interface. The developed integrated ECL/EC sensor based on Ru@AuNPs/bp-SNA modified solid indium tin oxide electrode enables the sensitive analysis of SARS-CoV-2 pseudoviruses by ECL mode with a linear range of 50 TU mL-1-5000 TU mL-1, as well as the EC mode with a linear range of 100 TU mL-1-5000 TU mL-1. Moreover, the designed sensor showed satisfactory results in the analyses of saliva and pond water samples. When flexible electrode substate (polyethylene terephthalate) is employed, Ru@AuNPs/bp-SNA has great potential to integrate with KN95 face masks for direct detection of SARS-CoV-2 pseudoviruses produced from breathing, talking and coughing processes, which could provide an efficient platform for POCT diagnosis.

Abstract:Carbonaceous immunosensors are ideal nanoplatforms for developing rapid, precise, and ultra-specific diagnostic kits capable of early detection of viral infectious illnesses such as COVID-19. However, developing a proper carbonic immunosensor requires stepwise protocols to find optimum operating conditions to minimize drawbacks. Herein, for the first time and through a stepwise protocol, activation, and monoclonal IgG antibody mounting capability of multi-walled carbon nanotubes (MWCNTs) at two diverse outer diameters (ODs), viz., 20-30 nm and 50-80 nm, and graphene deriv atives (graphene oxide (GO) and reduced graphene oxide (rGO)) were examined and compared with each other toward finding the prime carbonaceous nanomaterial(s) for maximized antibody loading efficiency along with an ideal detection limit (DL) and sensitivity. Next, the effect of common amplifying agents, i.e., Au nanostars (Au NSs) and Ag nanowires (Ag NWs), on the total performance of the best carbonaceous structure was carefully assessed, and the responsible detection mechanism is investigated in detail. Next, the developed carbonaceous immunosensors were assessed via voltammetric and impedance assays, and their performances toward specific detection of SARS-CoV-2 antigen through immunoreaction were examined in detail. The study's outcome showed the superior performance of conjugated rGO-based immunosensor with Au NSs toward specific and quick (1 min) detection of SARS-CoV-2 antigen in biological fluids compared with other 1D/2D carbonaceous nanomaterials.
