Abstract:Gibberellin (GA) 2-oxidase plays a key role in the GA catabolic pathway through 2β-hydroxylation. In the present study, we isolated a CaMV 35S-enhancer activation tagged mutant, H032. This mutant exhibited a dominant dwarf and GA-deficient phenotype, with a final stature that was less than half of its wild-type counterpart. The endogenous bioactive GAs are markedly decreased in the H032 mutant, and application of bioactive GAs (GA3 or GA4) can reverse the dwarf phenotype. The integrated T-DNA was detected 12.8 kb upstream of the OsGA2ox6 in the H032 genome by TAIL-PCR. An increased level of OsGA2ox6 mRNA was detected at a high level in the H032 mutant, which might be due to the enhancer role of the CaMV 35S promoter. RNAi and ectopic expression analysis of OsGA2ox6 indicated that the dwarf trait and the decreased levels of bioactive GAs in the H032 mutant were a result of the up-regulation of the OsGA2ox6 gene. BLASTP analysis revealed that OsGA2ox6 belongs to the class III of GA 2-oxidases, which is a novel type of GA2ox that uses C20-GAs (GA12 and/or GA53) as the substrates. Interestingly, we found that a GA biosynthesis inhibitor, paclobutrazol, positively regulated the OsGA2ox6 gene. Unlike the over-expression of OsGA2ox1, which led to a high rate of seed abortion, the H032 mutant retained normal flowering and seed production. These results indicate that OsGA2ox6 mainly affects plant stature, and the dominant dwarf trait of the H032 mutant can be used as an efficient dwarf resource in rice breeding.

Abstract:A cDNA library was constructed from the root tissues of cassava variety Huanan 124 at the root bulking stage. A total of 9,600 cDNA clones from the library were sequenced with single-pass from the 5′-terminus to establish a catalogue of expressed sequence tags (ESTs). Assembly of the resulting EST sequences resulted in 2,878 putative unigenes. Blastn analysis showed that 62.6% of the unigenes matched with known cassava ESTs and the rest had no ‘hits’ against the cassava database in the integrative PlantGDB database. Blastx analysis showed that 1,715 (59.59%) of the unigenes matched with one or more GenBank protein entries and 1,163 (40.41%) had no ‘hits’. A cDNA microarray with 2,878 unigenes was developed and used to analyze gene expression profiling of Huanan 124 at key growth stages including seedling, formation of root system, root bulking, and starch maturity. Array data analysis revealed that (1) the higher ratio of up-regulated ribosome-related genes was accompanied by a high ratio of up-regulated ubiquitin, proteasome-related and protease genes in cassava roots; (2) starch formation and degradation simultaneously occur at the early stages of root development but starch degradation is declined partially due to decrease in UDP-glucose dehydrogenase activity with root maturity; (3) starch may also be synthesized in situ in roots; (4) starch synthesis, translocation, and accumulation are also associated probably with signaling pathways that parallel Wnt, LAM, TCS and ErbB signaling pathways in animals; (5) constitutive expression of stress-responsive genes may be due to the adaptation of cassava to harsh environments during long-term evolution.

Carbohydrate Repartitioning in the Rice Starch Branching Enzyme IIb Mutant Stimulates Higher Resistant Starch Content and Lower Seed Weight Revealed by Multiomics Analysis.

Abstract:The starch branching enzyme IIb mutant (be2b) in rice significantly increases the resistant starch (RS) content and leads to reduced seed weight. However, the underlying metabolic mechanisms remain unclear. Proteomic analysis indicated that upregulation of starch synthase IIa (SSIIa) and SSIIIa and downregulation of BEI and SSI were possibly responsible for the decreased short amylopectin chains (DP 6-15) and increased longer chains (DP > 16) of be2b starch. The upregulation of granule-bound starch synthase led to increased amylose content (AC). These changes in the amylopectin structure and AC accounted for the increased RS content. α-Amylase 2A showed the strongest upregulation (up to 8.45-fold), indicating that the loss of BEIIb activity enhanced starch degradation. Upregulation of glycolysis-related proteins stimulated carbohydrate repartitioning through glycerate-3-phosphate and promoted the accumulation of tricarboxylic acid cycle intermediates, amino acids, and fatty acids. The unexpected carbohydrate partitioning and enhanced starch degradation resulted in the reduced seed weight in the be2b mutant.

Abstract:The proteins with DNA-binding preference to the consensus DNA sequence (A/T) GATA (A/G) belong to a GATA transcription factor family, with a wide array of biological processes in plants. Cassava (Manihot esculenta) is an important food crop with high production of starch in storage roots. Little was however known about cassava GATA domain-containing genes (MeGATAs). Thirty-six MeGATAs, MeGATA1 to MeGATA36, were found in this study. Some MeGATAs showed a collinear relationship with orthologous genes of Arabidopsis, poplar and potato, rice, maize and sorghum. Eight MeGATA-encoded proteins (MeGATAs) analysed were all localized in the nucleus. Some MeGATAs had potentials of binding ligands and/or enzyme activity. One pair of tandem-duplicated MeGATA17-MeGATA18 and 30 pairs of whole genome-duplicated MeGATAs were found. Fourteen MeGATAs showed low or no expression in the tissues. Nine analysed MeGATAs showed expression responses to abiotic stresses and exogenous phytohormones. Three groups of MeGATA protein interactions were found. Fifty-three miRNAs which can target 18 MeGATAs were identified. Eight MeGATAs were found to target other 292 cassava genes, which were directed to radial pattern formation and phyllome development by gene ontology enrichment, and autophagy by Kyoto Encyclopaedia of Genes and Genomes enrichment. These data suggest that MeGATAs are functional generalists in interactions between cassava growth and development, abiotic stresses and starch metabolism.
Cell Wall Invertase 3 Affects Cassava Productivity via Regulating Sugar Allocation From Source to Sink

Abstract:Storage roots are the main sink for photo-assimilate accumulation and reflect cassava yield and productivity. Regulation of sugar partitioning from leaves to storage roots has not been elucidated. Cell wall invertases are involved in the hydrolysis of sugar during phloem unloading of vascular plants to control plant development and sink strength but have rarely been studied in root crops like cassava. MeCWINV3 encodes a typical cell wall invertase in cassava and is mainly expressed in vascular bundles. The gene is highly expressed in leaves, especially mature leaves, in response to diurnal rhythm. When MeCWINV3 was overexpressed in cassava, sugar export from leaves to storage roots was largely inhibited and sucrose hydrolysis in leaves was accelerated, leading to increased transient starch accumulation by blocking starch degradation and reduced overall plant growth. The progress of leaf senescence was promoted in the MeCWINV3 over-expressed cassava plants with increased expression of senescence-related genes. Storage root development was also delayed because of dramatically reduced sugar allocation from leaves. As a result, the transcriptional expression of starch biosynthetic genes such as small subunit ADP-glucose pyrophosphorylase, granule-bound starch synthase I, and starch branching enzyme I was reduced in accordance with insufficient sugar supply in the storage roots of the transgenic plants. These results show that MeCWINV3 regulates sugar allocation from source to sink and maintains sugar balance in cassava, thus affecting yield of cassava storage roots.

Overexpression of Cassava MeSTP7 Promotes Arabidopsis Seedling Development

Abstract:The sugar transporter (STP) gene family is a key regulator of plant development, which is crucial for the efficient transport and utilization of sugars during plant growth and development. In this study, we identified the MeSTP7 gene, which is highly expressed in cassava fibrous roots, early storage roots, and under hormonal treatment, including IAA, MeJA, ABA, and GA3, and abiotic stressors, such as mannitol and NaCl. A strong response was observed with exoqenous IAA. Transfecting MeSTP7 into Arabidopsis promoted early seedling growth, particularly in lateral root development. The content of endogenous hormones (IAA and MeJA) as well as soluble sugars (sucrose, fructose, and glucose) was elevated in transgenic Arabidopsis. Hormone treatments with IAA, MeJA, GA3, and ABA on transgenic Arabidopsis revealed that transgenic Arabidopsis responded positively to added 20 μM IAA. They also exhibited co-induced regulation of lateral root formation by GA3, MeJA, and ABA. qRT-PCR analysis showed that overexpression of MeSTP7 upregulated the expression of IAA14, ARF7, and ARF19 in Arabidopsis. Under IAA treatment, the expression of these genes was similarly upregulated but downregulated under MeJA treatment. These results suggest that MeSTP7 may promote Arabidopsis seedling development by increasing the content of sucrose, glucose, and fructose in roots, which in turn influences IAA-based hormonal signaling.

Abstract:Gibberellins (GAs) are a class of plant hormones that can affect plant growth and development. GA-oxidases are rate-limiting enzymes, which play a direct role in GA accumulation in plants. However, the roles of GA-oxidase on carrot (Daucus carota L.) taproot development are still unclear. In this study, two GA-oxidase genes, DcGA20ox2 and DcGA2ox1, were identified in carrot. Transgenic carrot plants were obtained by using Agrobacterium-mediated genetic transformation method. The results showed that overexpression of DcGA20ox2 significantly promoted the accumulation of active GAs in carrot, increased plant height, generated more branches, and enhanced xylem development. Overexpression of DcGA2ox1 significantly reduced the total contents of active GAs compared with the control group, resulting in a dwarf phenotype and markedly increased lignin content of the transgenic carrot. The expression profiling showed that the genes of GA metabolic pathway responded to the negative feedback regulation mechanism. At the same time, the expression of most genes in lignin biosynthesis and polymerization process was up-regulated, corresponding to the massive accumulation of lignin. These findings indicated that DcGA20ox2 and DcGA2ox1 affected carrot growth and development by regulating the levels of endogenous GAs. The results from current work might shed light on further studies aimed to regulate lignification in carrot and other crops.

Abstract:The GROWTH-REGULATING FACTOR (GRF) and its INTERACTING FACTOR (GIF) have been shown to stimulate regeneration of transgenic plants with studies reporting increased transformation efficiency in multiple species, including wheat, beet, and citrus. The present work evaluated the effects of overexpressing GRF4-GIF1 and GRF5 on the regeneration of transgenic plants in cassava ( Manihot esculenta Crantz). Effects of GRF4-GIF1 and GRF5 sequences derived from Vitis vinifera and Arabidopsis thaliana were assessed by cloning expression cassettes under control of strong constitutive promoters. Friable embryogenic callus from cassava varieties 60444 and NASE 13 were transformed with Agrobacterium tumefaciens strains LBA4404 and LBA4404 THY-, and multiple independent transgenic plant lines recovered. Expression of the morphogenic genes did not enhance transformation efficiency, nor efficiency or timing of somatic embryo regeneration or whole plant recovery above the green fluorescent protein (GFP) control. Organogenesis experiments were carried out to observe effects of transgenic expression of these genes on morphogenesis from petiole, leaf-petiole, and stem explants. Results differed between the two genotypes evaluated. Expression of Vitis vinifera GRF4-GIF1 was effective for stimulation of rapid organogenesis and shoot regeneration at 30 to 37% from leaf-petiole explants of cultivar 60444. In contrast, Arabidopsis thaliana GRF5 was superior in stimulating shoot regeneration in cultivar NASE 13 with 40 to 50% of leaf-petiole explants regenerating shoots. In both cultivars, caulogenesis occurred rapidly within 3 to 4 wk culture on medium containing the cytokinin meta-topolin. Effects of overexpression of these morphogenic genes at the whole plant level were accessed by establishing plants in the greenhouse. GRF4-GIF1 overexpression resulted in significantly shorter plants with increased leaf size and total leaf area while AtGRF5 stimulated above average storage root weight.

Abstract:Gibberellins (GAs), a class of phytohormones, act as an essential natural regulator of plant growth and development. Many studies have shown that GA is related to rhizobial infection and nodule organogenesis in legume species. However, thus far, GA metabolism and signaling components are largely unknown in the model legume Medicago truncatula. In this study, a genome-wide analysis of GA metabolism and signaling genes was carried out. In total 29 components, including 8 MtGA20ox genes, 2 MtGA3ox genes, 13 MtGA2ox genes, 3 MtGID1 genes, and 3 MtDELLA genes were identified in M. truncatula genome. Expression profiles revealed that most members of MtGAox, MtGID1, and MtDELLA showed tissue-specific expression patterns. In addition, the GA biosynthesis and deactivation genes displayed a feedback regulation on GA treatment, respectively. Yeast two-hybrid assays showed that all the three MtGID1s interacted with MtDELLA1 and MtDELLA2, suggesting that the MtGID1s are functional GA receptors. More importantly, M. truncatula exhibited increased plant height and biomass by ectopic expression of the MtGA20ox1, suggesting that enhanced GA response has the potential for forage improvement.

Discussion: Outcome of Different Timings of Radiotherapy in Implant-Based Breast Reconstructions

Abstract:T irradiated breast is increasingly seen for reconstruction because of recurrence after lumpectomy and the use of postmastectomy irradiation for large tumors or for patients with more than two to four positive nodes. Many patients do not want big flap operations with secondary operative sites and do not want to have delayed reconstruction, so the plastic surgeon is faced with working with implants and irradiated tissue. This excellent study by Nava et al. helps us to evaluate the irradiated bed, to find out what happens and to ask “why?” The authors followed 257 consecutive patients for 50 months. Group 1 consisted of patients who were not known preoperatively to need irradiation but, because of positive nodes, were given chemotherapy and irradiation postoperatively. Their expanders were changed for permanent gel implants after chemotherapy but before irradiation. Group 2 patients, in contrast, had locally advanced disease and therefore underwent neoadjuvant chemotherapy first. They then had mastectomies and tissue expanders and underwent irradiation while their tissue expanders were in. Permanent implants were placed after irradiation. Group 3 consisted of patients who did not need irradiation, underwent mastectomy with expander and chemotherapy, and were then changed to a gel implant. The authors found that the irradiated expanders had a poor outcome as compared with irradiated implants. The implant loss rate was 40 percent in the irradiated expander group and 6.4 percent in the permanent implant group. The authors conclude that chest wall irradiation is best delivered on permanent implants. They advocate that expansion be performed during chemotherapy and that, 3 weeks after finishing chemotherapy, the permanent implant should be placed. Then, irradiation can begin 3 weeks later. The outcomes of the study are summarized in Table 1. If we look at the study more closely, group 1 lost one expander to infection, one to exposure, and five to capsular contracture. Group 2 lost seven expanders to exposure, eight to infection, and five to capsular contracture. Indeed, the Baker classification of groups 1 and 2 were not significantly different. It seems that the outcome is so poor in the irradiated expanders because of dehiscence and infection. Dehiscence is caused by a wound healing problem, excess tension, or poor perfusion. Infection is caused by bacterial contamination, generally in the first 2 weeks after surgery. Why are these so much higher in the irradiated expanders? Perhaps there are some factors that explain these findings. Group 2 had chemotherapy first— does chemotherapy cause some patients to heal more slowly or to be more prone to infection when the expander is place? This was studied by Warren Peled et al.,1 Azzawi et al.,2 and Berry et al.3; all found that chemotherapy did not change the rate of infection as compared with controls. The poorly performing group also had locally advanced breast cancer. Was more skin taken in this group so that dehiscence was more common? Gross et al.4 found a failure rate of 22.7 percent in 141 irradiated expanders/implants, and failure correlated with larger tumors, smoking, and positive nodes in the axilla. Were these obese or largebreasted patients who needed higher volumes in their expanders? Berry et al.3 and the Michigan Outcome Study5 found that a body mass index greater than 30 was correlated with major complications. We see also that group 2 underwent irradiation 3 weeks after a mastectomy/axillary operation (a longer operation with more lymphatic drainage), whereas group 1 underwent irradiation 3 weeks after an implant exchange (a smaller operation, often without the need for a drain). Abstract:As a starchy and edible tropical plant, cassava (Manihot esculenta Crantz) has been widely used as an industrial raw material and a dietary source. However, the metabolomic and genetic differences in specific germplasms of cassava storage root were unclear. In this study, two specific germplasms, M. esculenta Crantz cv. sugar cassava GPMS0991L and M. esculenta Crantz cv. pink cassava BRA117315, were used as research materials. Results showed that sugar cassava GPMS0991L was rich in glucose and fructose, whereas pink cassava BRA117315 was rich in starch and sucrose. Metabolomic and transcriptomic analysis indicated that sucrose and starch metabolism had significantly changing metabolites enrichment and the highest degree of differential expression genes, respectively. Sugar transport in storage roots may contribute to the activities of sugar, which will eventually be exported to transporters (SWEETs), such as (MeSWEET1a, MeSWEET2b, MeSWEET4, MeSWEET5, MeSWEET10b, and MeSWEET17c), which transport hexose to plant cells. The expression level of genes involved in starch biosynthesis and metabolism were altered, which may result in starch accumulation. These results provide a theoretical basis for sugar transport and starch accumulation and may be useful in improving the quality of tuberous crops and increasing yield.

Abstract:The high content of cyanogenic glycosides (CG) in cassava storage tubers seriously affects human food safety. CG play crucial roles in plant growth and development and can protect cassava leaves from being masticated by herbivorous predators. Nevertheless, the regulatory mechanism of CG biosynthesis, which results in a low CG content in storage tubers and high CG content in leaves, remains poorly understood. Here, yeast one-hybrid assay was performed using a mixed cDNA library of cassava storage roots and leaves as prey and the promoter of as bait. MeCYP79D2, a cytochrome P450 protein, is the rate-limiting enzyme for CG synthesis in cassava. From this information, a candidate regulator of , that is, transcription factor MePHD1.2, was selected. MePHD1.2, which is located in the nucleus and exhibits a transcription inhibitory activity, can directly bind to PD2 segment in the promoter of , which results in its repressed expression. In cassava, the transcriptional activity of was considerably enhanced in lines, which caused an increase in the contents of linamarin and lotaustralin. Our findings unveil a novel regulatory module governing CG biosynthesis, wherein mutation of attenuates its transcription inhibition on and boosts CGs biosynthesis in cassava.

Functional Characterization of the MeSSIII-1 Gene and Its Promoter from Cassava

Abstract:Soluble starch synthases (SSs) play important roles in the synthesis of cassava starch. However, the expression characteristics of the cassava SSs genes have not been elucidated. In this study, the MeSSIII-1 gene and its promoter, from SC8 cassava cultivars, were respectively isolated by PCR amplification. MeSSIII-1 protein was localized to the chloroplasts. qRT-PCR analysis revealed that the MeSSIII-1 gene was expressed in almost all tissues tested, and the expression in mature leaves was 18.9 times more than that in tuber roots. MeSSIII-1 expression was induced by methyljasmonate (MeJA), abscisic acid (ABA), and ethylene (ET) hormones in cassava. MeSSIII-1 expression patterns were further confirmed in proMeSSIII-1 transgenic cassava. The promoter deletion analysis showed that the −264 bp to −1 bp MeSSIII-1 promoter has basal activity. The range from −1228 bp to −987 bp and −488 bp to −264 bp significantly enhance promoter activity. The regions from −987 bp to −747 bp and −747 bp to −488 bp have repressive activity. These findings will provide an important reference for research on the potential function and transcriptional regulation mechanisms of the MeSSIII-1 gene and for further in-depth exploration of the regulatory network of its internal functional elements.

Abstract:The basic helix-loop-helix (bHLH) proteins are a large superfamily of transcription factors, and play a central role in a wide range of metabolic, physiological, and developmental processes in higher organisms. However, systematic investigation of bHLH gene family in cassava ( Manihot esculenta Crantz) has not been reported. In the present study, we performed a genome-wide survey and identified 148 MebHLH s genes were unevenly harbored in 18 chromosomes. Through phylogenetic analyses along with Arabidopsis counterparts, these MebHLHs genes were divided into 19 groups, and each gene contains a similar structure and conserved motifs. Moreover, many cis -acting regulatory elements related to various defense and stress responses showed in MebHLH genes. Interestingly, transcriptome data analyses unveiled 117 MebHLH genes during postharvest physiological deterioration (PPD) process of cassava tuberous roots, while 65 MebHLH genes showed significantly change. Meanwhile, the relative quantitative analysis of 15 MebHLH genes demonstrated that they were sensitive to PPD, suggesting they may involve in PPD process regulation. Cyanogenic glucosides (CGs) biosynthesis during PPD process was increased, silencing of MebHLH72 and MebHLH114 showed that linamarin content was significantly decreased in the leaves. To summarize, the genome-wide identification and expression profiling of MebHLH candidates pave a new avenue for uderstanding their function in PPD and CGs biosynthesis, which will accelerate the improvement of PPD tolerance and decrease CGs content in cassava tuberous roots.

Abstract:Abstract Background We previously identified six drought-inducible CC-type glutaredoxins in cassava cultivars, however, less is known about their potential role in the molecular mechanism by which cassava adapted to abiotic stress. Results Herein, we investigate one of cassava drought-responsive CC-type glutaredoxins, namely MeGRXC3 , that involved in regulation of mannitol-induced inhibition on seed germination and seedling growth in transgenic Arabidopsis . MeGRXC3 overexpression up-regulates several stress-related transcription factor genes, such as PDF1.2 , ERF6 , ORA59, DREB2A , WRKY40 , and WRKY53 in Arabidopsis . Protein interaction assays show that MeGRXC3 interacts with Arabidopsis TGA2 and TGA5 in the nucleus. Eliminated nuclear localization of MeGRXC3 failed to result mannitol-induced inhibition of seed germination and seedling growth in transgenic Arabidopsis. Mutation analysis of MeGRXC3 indicates the importance of conserved motifs for its transactivation activity in yeast. Additionally, these motifs are also indispensable for its functionality in regulating mannitol-induced inhibition of seed germination and enhancement of the stress-related transcription factors in transgenic Arabidopsis . Conclusions MeGRXC3 overexpression confers mannitol sensitivity in transgenic Arabidopsis possibly through interaction with TGA2/5 in the nucleus, and nuclear activity of MeGRXC3 is required for its function.

Abstract:Cellulose is the major renewable feedstock used in a variety of industrial processes. However, its native state, organizing itself in microfibrils and interacting with other components of the cell wall complex, forms a highly recalcitrant structure. Expansin proteins may facilitate the enzymatic saccharification stage by affecting non-covalent bonds between the wall polymers. In the current study, the expression profile of five selected Expansin genes from sugarcane, an important crop in the supply of biomass lignocellulose for the ethanol industry, was analyzed in leaves, roots, and young culms. The SacEXP32 and SacEXP39 genes, potentially associated with the cell wall architecture modification in sugarcane culm, were selected for functional characterization studies in Nicotiana benthamiana using the agroinfiltration assay. Heterologous expression of SacEXP32 and SacEXP39 revealed that N. benthamiana cellulose synthase gene was positively regulated at the transcript level in transformed tissue. However, only SacEXP32 expression increased cell area and enzymatic saccharification efficiency. SacEXP32 overexpression may have influenced the network of all cell wall polymers, indicating that this gene is a potential target for biotechnological approaches to improve plant biomass industrial processing.

Abstract:Proper development of the maize kernel is of great significance for high and stable maize yield to ensure national food security. Gibberellin (GA), one of the hormones regulating plant growth, is involved in modulating the development of maize kernels. Cellulose, one of the main components of plant cells, is also regulated by gibberellin. The mechanism of hormone regulation during maize grain development is highly complicated, and reports on GA-mediated modulation of cellulose synthesis during maize grain development are rare. Our study revealed that during grain growth and development, the grain length and bulk density of GA-treated corn kernels improved significantly, and the cellulose content of grains increased, while seed coat thickness decreased. The transcription factor basic region/leucine zipper motif 53 ( bZIP53) , which is strongly correlated with cellulose synthase gene 1 ( CesA1) expression, was screened by transcriptome sequencing and the expression of the cellulose synthase gene in maize grain development after GA treatment was determined. It was found that bZIP53 expression significantly promoted the expression of CesA1 . Further, analysis of the transcription factor bZIP53 determined that the gene-encoded protein was localized in the cell and nuclear membranes, but the transcription factor bZIP53 itself showed no transcriptional activation. Further studies are required to explore the interaction of bZIP53 with CesA1 .
