Abstract:The cellular components of the immune system and the inflammatory milieu that it can generate is a central theme in many diseases including cancer. Immune cells can be manipulated by tumor cells to favor a pro-tumor microenvironment resulting in tumor progression. Ovarian cancer can alter its microenvironment favoring tumor growth by suppressing effector T cells as well as recruiting myeloid-derived cells, Th17 cells, γδ T cells, as well as non-immune cells such as adipose cells to aid in the generation or the propagation of the pro-inflammatory milieu. The human microbiome maintains a delicate balance between pro- and anti-inflammatory mechanisms, essential for gut homeostasis, and has critical roles in immune system development and metabolism. Alterations in the microbiome results in dysbiosis, quantitative and qualitative shifts in microbial populations, and contributes to chronic inflammation in various diseases including cancer. We highlight the role that the gut microbiota may play in cancer initiation and/or progression as well as its impact on cancer therapy. The association and interactions between the microbiome, both gut microbiota as well as infectious virus, with ovarian cancer, is reviewed here. Understanding the mechanisms by which the microbiome modulates the innate and adaptive immune response and contributes to an inflammatory milieu in cancer may offer insights into novel therapeutic targets.

Abstract:Humans and other mammals are colonized by microbial agents across the kingdom which can represent a unique microbiome pattern. Dysbiosis of the microbiome has been associated with pathology including cancer. We have identified a microbiome signature unique to ovarian cancers, one of the most lethal malignancies of the female reproductive system, primarily because of its asymptomatic nature during the early stages in development. We screened ovarian cancer samples along with matched, and non-matched control samples using our pan-pathogen array (PathoChip), combined with capture-next generation sequencing. The results show a distinct group of viral, bacterial, fungal and parasitic signatures of high significance in ovarian cases. Further analysis shows specific viral integration sites within the host genome of tumor samples, which may contribute to the carcinogenic process. The ovarian cancer microbiome signature provides insights for the development of targeted therapeutics against ovarian cancers.

Abstract:Abstract Background: The tumor microenvironment has immense potential to change a patient’s prognosis and therapeutic response. One of the emerging components of interest in the tumor microenvironment is the presence of intracellular microbes. Numerous studies have focused on colon and pancreatic cancer because of their proximity to the commensals in the gut. The female reproductive system is another site in our body where the rich vaginal microbiome has the potential to affect nearby organs and impact diseases. However, there is still little investigation done to understand the impact of microbes on Ovarian Cancer (OC). Objective: To understand the role of the microbiome in OC metastasis, to develop new therapies to target the tumor microbiome Methods: We used a co-culture study, the parameters of which were optimized to study the infection of microbes in different cell lines. Using a wide array proteome profiler, we looked for cytokines and chemokines to understand changes in signaling post-infection. Protein levels in the conditioned media using ELISA. Changes in actin, paxillin, and LC3B (autophagy marker) were monitored using immunofluorescence microscopy and western blots. Migration was studied by using transwell assay and spheroid cultures. Results: We have identified two species of bacteria preferentially enriched in human ovarian tumors based on recent findings in the literature. We have demonstrated that these microbes are better at infecting OC cells in vitro over non-specific bacteria, can reside intracellularly, and are metabolically active. Upon infection, this intracellular uptake drives the release of pro-inflammatory cytokines and chemokine signaling by cancer and normal ovarian surface cells. We have also observed increased autophagy, changes in the actin cytoskeleton and reduced Paxillin foci on cancer cells infected with OC-associated microbes. This has led to increased migration in them; however, inactivated microbes did not change their migratory behavior. The infection also caused individual tumor cells to form more numerous and larger colonies when seeded into soft agar, suggesting an effect on cancer cell migration. Furthermore, we have shown that these microbes also elicit a more robust reaction in immune cells like Dendritic cells and macrophages, causing them to secrete chemokines like CCL 2,3,4,5, which also cause the cancer cells to migrate and have the potential to recruit neutrophils to drive the tumor microenvironment towards an immunosuppressive phenotype. Conclusion: Most ovarian cancer patients are diagnosed at Stage III/IV, where cancer has metastasized, and is unresponsive to immunotherapy as the cancer is considered “cold”. Through functional studies, we were able to gain insights into the role of the microbiome, which is associated with the clinical outcomes of OC, hoping to potentiate the existing therapies by probing the tumor microbiome. Abstract:The gut microbiota has emerged as an important consideration in clinical oncology. The role of the microbiome in cancer extends beyond causation and cancer risk. It is now known that the microbiome not only acts at a local epithelial level in the gut but also modifies immune responses within intestinal and extraintestinal tumors. Microbial signaling influences the clinical course of cancer including the efficacy, bioavailability, and toxicity of chemotherapeutic and immunotherapy agents. This has focused research on microbiota profiling in different cancer states with an aim of developing prognostic biomarkers of risk. The potential value of microbiome manipulation with live biotherapeutics or microbial transplantation has also become a realistic consideration. Maintenance of microbial diversity in patients with cancer is a variable challenge given the modifying influences of the tumor itself, chemotherapy, nutritional status, and sporadic antimicrobial therapy. Here, we address current evidence for the role of the microbiome in cancer therapy.

Abstract:Breast cancer-related mortality remains high worldwide, despite tremendous advances in diagnostics and therapeutics; hence, the quest for better strategies for disease management, as well as the identification of modifiable risk factors, continues. With recent leaps in genomic technologies, microbiota have emerged as major players in most cancers, including breast cancer. Interestingly, microbial alterations have been observed with some of the established risk factors of breast cancer, such as obesity, aging and periodontal disease. Higher levels of estrogen, a risk factor for breast cancer that cross-talks with other risk factors such as alcohol intake, obesity, parity, breastfeeding, early menarche and late menopause, are also modulated by microbial dysbiosis. In this review, we discuss the association between known breast cancer risk factors and altered microbiota. An important question related to microbial dysbiosis and cancer is the underlying mechanisms by which alterations in microbiota can support cancer progression. To this end, we review the involvement of microbial metabolites as effector molecules, the modulation of the metabolism of xenobiotics, the induction of systemic immune modulation, and altered responses to therapy owing to microbial dysbiosis. Given the association of breast cancer risk factors with microbial dysbiosis and the multitude of mechanisms altered by dysbiotic microbiota, an impaired microbiome is, in itself, an important risk factor.
Impact of initiatives to reduce public dental waiting lists in Queensland, Australia.

Abstract:Information on public dental service waiting lists is available as part of the Queensland Government open data policy. Data were analysed for the period December 2012 to December 2013, to present the total number and percentage of people waiting for care and who have waited beyond the desirable period. Over the 1-year study period, the number of people on the waiting list decreased from 130 546 to 77 146, a difference of 40.9%. A decrease of 80.6% was found for those waiting beyond the desirable period for care. The largest decrease was for general care (44.9%). The initiatives to reduce the public dental waiting list appear to have been successful in significantly reducing the number of people waiting in general and especially those waiting beyond the desirable period. The initiatives to decrease waiting lists represent a downstream approach and are less likely to have any significant impact on the prevention of oral diseases. As waiting lists are reduced, more emphasis should be placed on upstream approaches such as health promotion, specific protection measures and targeting high-risk individuals for oral diseases.

As a primary instigator of inflammation, the majority of microbiomes naturally residing within our bodies engage in a mutually beneficial symbiotic relationship. Nevertheless, alterations in the microbiome's composition or breaches in the normal barrier function can disrupt the internal environment's homeostasis, potentially leading to the development and progression of various diseases, including tumors. The investigation of tumor-related microbiomes has contributed to a deeper understanding of their role in tumorigenesis. This review offers a comprehensive overview of the microbiome alterations and the associated inflammatory changes in ovarian cancer. It may aid in advancing research to elucidate the mechanisms underlying the ovarian cancer-associated microbiome, providing potential theoretical support for the future development of microbiome-targeted antitumor therapies and early screening through convenient methods.

Abstract:Human gut microbiota physiologically and actively participates as a symbiont to a wide number of fundamental biological processes, such as absorption and metabolism of nutrients, regulation of immune response and inflammation; gut microbiota plays also an antitumor role. However, dysbiosis, resulting from a number of different situations—dysmicrobism, infections, drug intake, age, diet—as well as from their multiple combinations, may lead to tumorigenesis and is associated with approximately 20% of all cancers. In a diagnostic, prognostic, therapeutic, and epidemiological perspective, it is clear that the bifaceted role of microbiota needs to be thoroughly studied and better understood. Here, we discuss the anti- and pro-tumorigenic potential of gut and other microbiota districts along with the causes that may change commensal bacteria from friend to foes.

Abstract:Bacterial dysbiosis is evolving as an advocate for carcinogenesis and has been associated with pancreatic cancer progression and survival outcomes. The gut and pancreas of cancer patients harbor a unique microbiome that differs significantly from that of healthy individuals. We believe that the pancreatic cancer microbiome regulates tumorigenesis by altering host cell function and modulating immune cells, skewing them toward an immunosuppressive phenotype. Moreover, altering this pathogenic microbiome may enhance the efficacy of current therapies in pancreatic cancer and improve survival outcomes. This review highlights the findings on microbial modulation across various pre-clinical and clinical studies and provides insight into the potential of targeting the microbiome for pancreatic cancer therapy.

Abstract:Recent evidence has shown that the human microbiome is associated with various diseases, including cancer. The salivary microbiome, fecal microbiome, and circulating microbial DNA in blood plasma have all been used experimentally as diagnostic biomarkers for many types of cancer. The microbiomes present within local tissue, other regions, and tumors themselves have been shown to promote and restrict the development and progression of cancer, most often by affecting cancer cells or the host immune system. These microbes have also been shown to impact the efficacy of various cancer therapies, including radiation, chemotherapy, and immunotherapy. Here, we review the research advances focused on how microbes impact these different facets and why they are important to the clinical care of cancer. It is only by better understanding the roles these microbes play in the diagnosis, development, progression, and treatment of cancer, that we will be able to catch and treat cancer early.

Abstract:In recent times, the microbiome has been increasingly recognized as having a hand in various disease states that include cancer as a part. Our commensal and symbiotic microbiota, in addition to pathogens with oncogenesis features, have tumor-suppressive characteristics. Our nutrition and other environmental influences can modulate some microbial species representatives within our digestive system and other systems. The microbiota has recently shown a two-way link to cancer immunotherapy for both the prognosis and the therapeutic aspects. Preclinical results indicated that microbiota modification could be transformed into a novel technique to improve cancer therapy's effectiveness. This article aimed to review recent development in our understanding of the microbiome and its relationship to cancer cells and discuss how the microbiome stimulates cancer and its clinical and therapeutic applications. Such information was selected and extracted from the PubMed, Web of Science, and Google Scholar databases for published data from 2000 to 2020 using relevant keywords containing a combination of terms, including the microbiome, cancer, immune response, immune response, and microbiota. Finally, we concluded that studying the human microbiome is necessary because it provides a thorough understanding of humans' interaction and their indigenous microbiota. The microbiome provides useful insight into future research studies to optimize these species to fight life-threatening diseases such as cancer and has rendered the microbiome a successful cancer treatment strategy.

Abstract:Cancer remains a public health concern worldwide, with its incidence increasing worldwide and expected to continue growing during the next decades. The microbiome has emerged as a central factor in human health and disease, demonstrating an intricate relationship between the microbiome and cancer. Although some microbiomes present within local tissues have been shown to restrict cancer development, mainly by interacting with cancer cells or the host immune system, some microorganisms are harmful to human health and risk factors for cancer development. This review summarizes the recent evidence concerning the microbiome and some of the most common cancer types (i.e., lung, head and neck, breast, gastric, colorectal, prostate, and cervix cancers), providing a general overview of future clinical approaches and perspectives.

Abstract:Ovarian cancer (OC) remains a significant health challenge globally, with high mortality rates despite advancements in treatment. Emerging research suggests a potential link between OC development and genital dysbiosis, implicating alterations in the microbiome composition as a contributing factor. To investigate this correlation, a meta-analysis was conducted following PRISMA and MOOSE guidelines, involving eight studies encompassing 3504 patients. Studies investigating the role of upper and inferior genital tract dysbiosis were included, with particular reference to HPV infection and/or history of pelvic inflammatory disease. The analysis revealed no significant difference in genital dysbiosis prevalence between OC patients and healthy controls. Although previous literature suggests associations between dysbiosis and gynecologic cancers, such as cervical and endometrial cancers, the findings regarding OC are inconclusive. Methodological variations and environmental factors may contribute to these discrepancies, underscoring the need for standardized methodologies and larger-scale studies. Despite the limitations, understanding the microbiome's role in OC development holds promise for informing preventive and therapeutic strategies. A holistic approach to patient care, incorporating microbiome monitoring and personalized interventions, may offer insights into mitigating OC risk and improving treatment outcomes. Further research with robust methodologies is warranted to elucidate the complex interplay between dysbiosis and OC, potentially paving the way for novel preventive and therapeutic approaches.

Abstract:Inflammation has been confirmed to exist in the tumor microenvironment, while the risk of cancer occurrence increases in cases of chronic inflammation. It is estimated that approximately 10% to 20% of cancers are associated with chronic infections and attendant inflammation. Bacteria, both pathogenic and commensal, viruses, and fungi actively participate in the development and maintenance of inflammation and tumor growth in humans. The exposome, which is a sum of human environmental exposures, such as industrial diet, consumed drugs, and toxins, affects the composition and function of the human microbiome, which could lead to dysbiosis and disorders in tissue homeostasis through different mechanisms, including the intensification of the immune response, activation and abnormal proliferation, and disruption to epithelial barrier integrity. Presently, science remains at the stage of revealing the complexity associated with the mechanisms involved in building relationships that cover the microbiome-inflammation-tumor, yet it is already known how important it is to care for microbial homeostasis of the organism.

Furthermore, we explore the unique microbial diversity within breast tissue, indicating potential imbalances brought about by cancer and highlighting specific microbes as promising therapeutic targets. Emphasizing a holistic One Health approach, this review underscores the importance of integrating insights from human, animal, and environmental health to gain a deeper understanding of the complex microbe–cancer interplay. The female reproductive system has a unique microbiome that helps keeping health maintenance and protects against infection. Contrary to other mucosal sites, the vaginal microbiota often exhibits limited diversity and contains few Lactobacilli species, which were categorized into five distinct community state types using high-throughput 16S rRNA gene sequencing. According to current research, Human Papillomavirus (HPV) acquisition, persistence, and cervical cancer development are influenced by increasing vaginal microbiome diversity and decreased Lactobacillus spp. This review explores the intricate relationship between vaginal microbiota and HPV in the context of cervical cancer development. It discusses the impact of microbiota diversity and dysbiosis on the acquisition, persistence, and clearance of HPV. The review also emphasizes the role of vaginal microbiota in maintaining a healthy female reproductive system and explores the potential of various interventions, such as probiotics and prebiotics, in restoring a healthy vaginal environment. The human microbiome participates in the modulation of human metabolism by influencing several intricate pathways. The association between specific bacteria or viruses and the efficacy of cancer treatments and the occurrence of treatment-related toxicity in cancer patients has been reported. However, the understanding of the interaction between the host microbiome and the cancer treatment response is limited, and the microbiome potentially plays a greater role in the treatment of cancer than reported to date. Here, we provide a thorough review of the potential role of the gut and locally resident bacterial microbiota in modulating responses to different cancer therapeutics to demonstrate the association between the gut or locally resident bacterial microbiota and cancer therapy. Probable mechanisms, such as metabolism, the immune response and the translocation of microbiome constituents, are discussed to promote future research into the association between the microbiome and other types of cancer. We conclude that the interaction between the host immune system and the microbiome may be the basis of the role of the microbiome in cancer therapies. Future research on the association between host immunity and the microbiome may improve the efficacy of several cancer treatments and provide insights into the cause of treatment-related side effects.
