Abstract:Residue of sulfamethazine (SMZ), a typical short-acting drug to prevent bacterial infections, in food is a threat to human health. A ternary heterogeneous metal-organic framework hybrid (Zn/Fe-MOF@PDANSs) of Zn-TCPP-MOF, MIL-101 (Fe) and polydopamine nanoparticles (PDANSs) was proposed to establish an aptasensor for the sensitive and selective detection of SMZ. In this sensor, Zn-TCPP-MOF and FAM emitted fluorescence at 609 nm and 523 nm, respectively, and the fluorescence of FAM-ssDNA could be quenched when it was adsorbed on the surface of MOF hybrid. In the presence of SMZ, the fluorescence of FAM-ssDNA recovered due to the dropping from MOF hybrid, while the fluorescence of MOF hybrid remained. With this strategy, a wide concentration range and high sensitivity of SMZ were detection. And the ternary Zn/Fe-MOF@PDANSs sensor exhibited more excellent performance than binary Zn/Fe-MOF aptasensor. In addition, the sensor showed pleasurable selectivity, and was utilized for SMZ determination in authentic chicken and pork samples, implying the fascinating potential in practical application.
Selection and truncation of aptamers for ultrasensitive detection of sulfamethazine using a fluorescent biosensor based on graphene oxide

Abstract:We developed a fluorescent aptamer/graphene oxide (GO)–based biosensor to detect sulfamethazine (SMZ) residues in animal-derived foods. The SMZ-bound aptamers were identified and screened with an improved GO-SELEX technique using non-immobilizing ssDNA library. After seven rounds of selection, six SMZ aptamers were sequenced and analyzed for secondary structure, and their affinity and specificity were assessed by binding assays. The truncated aptamer (SMZ1S: 5′-CGTTAGACG-3′) with a unique stem-loop structure showed the highest affinity (Kd = 24.6 nM) to SMZ and was used to develop a GO-based fluorescent aptasensor. The binding mechanism between SMZ1S and SMZ was further analyzed by molecular docking. Under optimal conditions, the fluorescent aptasensor showed low detection limits (0.35 ng/mL) and a wide dynamic linear range (from 2 to 100 ng/mL). The aptasensor was also validated against real samples spiked with SMZ, which showed a fluorescence recovery from 93.9 to 108.8% and a coefficient of variation of < 12.7%. Taken together, these results suggest that this novel aptasensor can be used to sensitively, selectively, and accurately detect SMZ residues in foods.

Novel CoOOH-based fluorescent aptasensor for rapid and sensitive detection of sulfamethazine in environmental samples

Abstract:Sulfamethazine (SMZ) has been widely used in animal husbandry and exposed to water and soil environments, posing potential threat to human health and ecological environment. Hence, we designed a CoOOH-based aptasensor, the fluorescence resonance energy transfer between FAM-labeled aptamer and CoOOH was used to sensitively and selectively detect SMZ in water and soil environments. Molecular docking and molecular dynamics simulations were used to predict binding mechanisms of SMZ and aptamer. Under optimized conditions, the aptasensor exhibited high sensitivity and selectivity with a linear range of 5–40 ng/mL and a limit of detection of 2.43 ng/mL. The recoveries of the aptasensor were 84.6–115.8% in water and soil samples with relative standard deviations below 9%, and the detection results were highly consistent with high-performance liquid chromatography. Therefore, this developed aptasensor was a reliable tool and could be applied to monitoring of SMZ in environmental samples.

Abstract:Antibiotics residues are still an important and noticeable issue, and realizing the ultra-sensitive, sample, and rapid detection of residual antibiotics in the environment is the basis for evaluating and monitoring the pollution status. Herein, a new bimetallic PtNi nanozyme-driven dual-amplified photoelectrochemical (PEC) aptasensor was developed for sulfamethazine (SMZ) assay based on the superior photosensitive material of Z-scheme heterostructured Co9S8@ In doped CdS nanotubes (Co9S8@In-CdS NTs). In this protocol, bimetallic alloyed nanotube PtNi was introduced as the mimic enzyme to instead of bio-enzyme for constructing of signal amplification strategy. On the basis of the outstanding photosensitivity of Co9S8@In-CdS NTs and highly catalytic property of PtNi nanozyme, the proposed PEC aptasensor exhibits a wide linear range of 0.05 pg mL–1 ~ 1.0 ng mL–1, and a low detection limit of 37.2 fg mL–1. This work put forwards a novel approach for the accurate analysis of antibiotics in the environment.

Abstract:Sulfamethazine (SMZ) is one of the most used broad-spectrum antibiotics owing to its low cost and high efficacy towards bacterial diseases. This workreports a novel label-free SMZ sensor based on para-sulfonatocalix[4]arene (pSC4) capped gold nanoparticles (pSC4-AuNPs) for colorimetric detection through the host–guest interaction. The existence of SMZ resulted in the aggregation of pSC4-AuNPs and can be observed through colorimetric assay. A good linear relationship in the range 2.5 ~ 20 nM was obtained with a correlation coefficient of 0.9908. The limit of detection for SMZ was 1.39 nM. High recoveries (90.18–107.06%) were obtained, and RSD ranged from 1.21 to 2.05%. The color changes can be observed from red to gray within 10 min. Combining the supermolecule’s recognition and AuNP’s optical performance, the method paves a new, easy, and rapid way for small target sensing.

Abstract:Herein, we developed a dual fluorescent aptasensor based on mesoporous silica to simultaneously detect sulfadimethoxine (SDM) and oxytetracycline (OTC) in animal-derived foods. We immobilized two types of aptamers modified with FAM and CY5 on the silica surface by base complementary pairing reaction with the cDNA modified with a carboxyl group and finally formed the aptasensor detection platform. Under optimal conditions, the detection range of the aptasensor for SDM and OTC was 3-150 ng/mL (R 2 = 0.9831) and 5-220 ng/mL (R 2 = 0.9884), respectively. The limits of detection for SDM and OTC were 2.2 and 1.23 ng/mL, respectively. The limits of quantification for SDM and OTC were 7.3 and 4.1 ng/mL, respectively. Additionally, the aptasensor was used to analyze spiked samples. The average recovery rates ranged from 91.75 to 114.65% for SDM and 89.66 to 108.94% for OTC, and all coefficients of variation were below 15%. Finally, the performance and practicability of our aptasensor were confirmed by HPLC, demonstrating good consistency. In summary, this study was the first to use the mesoporous silica-mediated fluorescence aptasensor for simultaneous detection of SDM and OTC, offering a new possibility to analyze other antibiotics, biotoxins, and biomolecules.

Abstract:In the quest to address the mounting concerns over sulfonamide antibiotic residues in food, which pose significant threats to public health and food safety, this study introduces a cutting‐edge detection method. Surface molecular imprinting on silicon nanoparticles is harnessed to fabricate a novel fluorescent sensor that exploits the luminescent properties of cadmium telluride (CdTe) quantum dots. This innovative approach aims to detect residual sulfonamide antibiotics with high specificity and sensitivity. At the heart of this research is the development of a core‐shell nanostructure, where silicon dioxide serves as the core, and a molecularly imprinted polymers (MIPs) layer, tailored to recognize sulfamethazine (SM2), forms the shell. The pivotal advancement in this sensor design is the integration of highly fluorescent CdTe quantum dots within the MIPs layer, which significantly enhances the signal response, enabling the detection of SM2 with remarkable precision. The synthesis of this sensor employs a novel strategy, utilizing 3‐aminopropyltriethoxysilane as the functional monomer, while tetraethyl orthosilicate and ammonium hydroxide act as catalysts to facilitate the polymerization reaction. This meticulous process yields a stable core‐shell structure with active fluorescent properties. Experimental results reveal that under optimal conditions, the sensor exhibits a robust linear response to SM2 concentrations ranging from 10 to 60 μmol L−1, with a detection limit as low as 0.78 μmol L−1. Furthermore, when applied to real food samples, such as honey, the sensor not only demonstrates high recovery rates of 92.3%–98.1%, but also maintains a low relative standard deviation of less than 2.5%. The implications of this study are far‐reaching, offering a promising avenue for the rapid and reliable monitoring of antibiotic residues in the food supply chain, thereby safeguarding consumer health and upholding food safety standards.

Abstract:The misuse of antibiotics may contaminate the environment and cause harm to human health. Therefore, rapid and accurate detection of antibiotics is essential. In this study, a novel electrochemiluminescence resonance energy transfer (ECL-RET) pair was designed using a new ECL emitter (CPM, Ce-TBAPy) as the donor and Co-MOF@AuPt as the acceptor. Moreover, a highly sensitive and specific "on-off-on" ECL aptasensor was constructed for detecting sulfadiazine (SDZ). The aptasensor exhibited a broad linear range (from 10.0 fg mL-1 to 100 ng mL-1) for the SDZ concentration, with limit of detection and limit of quantification values of 1.14 fg mL-1and 3.75 fg mL-1, respectively. The aptasensor achieved good results in spiking experiments with milk and egg samples, and successfully quantified SDZ in fish meal quality control sample. The prepared aptasensor presents great potential for food and environmental safety by detecting antibiotics.

Abstract:Abstract In this work, a dual‐emission ratiometric fluorescence sensor was prepared and is used for the detection of sulfadiazine drug. In this paper, the hydrothermal method was used to synthesize CQD (fluorescence response signal) and sulfhydryl modified cadmium telluride quantum dots (fluorescence reference signal), and the molecularly imprinted polymer is synthesized following. After eluting the template molecule SDZ, the blot recognition site remains on the surface of the imprinted polymer. During the process of detection, due to electron transfer between SDZ and CQDs, holes of stimulated electrons are filled. It leads fluorescence quenching around 430 nm, which greatly improves sensor selection performance. Compared with traditional single‐emission fluorescence sensors, ratiometric fluorescence sensors have a wider linear range and higher detection accuracy. Under the optimal conditions, the sensor exhibited excellent performance in terms of low detection limit (4.2 nmol L −1 ) and its satisfactory linear range (2–20 μmol L −1 ). This intelligent response change mode also provide prospect for other sulfonamide antibiotics detection and offer a reference to signal transform for other molecular imprinting technology. Abstract:An electrochemical biosensor was proposed utilizing an improved amplification strategy for the rapid detection of sulfamethazine (as a model target) in aquatic environments. In this competitive immunoassay, cetyltrimethylammonium bromide-capped gold nanoparticles (CTAB-AuNPs) were used as a signal amplifier and electrode matrix and coated with an antigen-antibody (Cag-Ab(1)) specific binding system as a recognition unit for the target compound. In addition, silver nanoparticle labels were functionalized with dendritic fibrous nanosilica (DFNS@AgNPs) and decorated onto chitosan/single walled carbon nanohorn (CS/SWCNH)-modified glass carbon electrodes (GCEs), which improved the electron transfer rate and increased the surface area, enabling more coating antigens to be captured. Under acidic conditions, massive amounts of the Ag+ bound to the surface of the AuNPs dissolved, and consequently, formed Ag+@CTAB-AuNP complexes, which resulted in a distinctly improved peroxidase-like activity and enhanced current response. Furthermore, the destroyed Ab(1)-Ab(2)-DFNS conjugation greatly decreased the impedance, bringing about the amplification of the electrochemical signals. After optimization of the parameters, the proposed approach exhibited excellent performance, including good sensitivity (LOD, 0.0655 ng/mL) and satisfactory accuracy (recoveries, 79.02%-118.39%; CV, 3.18%-9.82%), which indicates the great potential of this strategy for the rapid detection of trace pollutants in the environments.

Abstract:Fluorescence-based aptasensors possess high sensitivity but are complicated and usually require multistep labeling and modification in method design, which severely limit the practical applications. Here, a label-free fluorescence-based aptasensor, consisting of aptamer, gold nanoparticles (AuNPs), and cadmium telluride (CdTe) quantum dots (QDs), was developed for the detection of sulfadimethoxine (SDM) in water and fish based on the specific recognition of SDM-aptamer and the inner filter effect of QDs and AuNPs. In the absence of a target, AuNPs dispersed in salt solution because of the aptamer protection, which could effectively quench the fluorescence emission of QDs, while in the presence of SDM, AuNPs aggregated due to the specific recognition of SDM-aptamer to SDM, which resulted in fluorescence recovery. A linear response of SDM concentrations in the range of 10–250 ng mL −1 ( R 2 = 0.99) was obtained, and the detection limit was 1.54 ng mL −1 (3σ, n = 9), far below the maximum residue limit (100 ng mL −1 ) of SDM in edible animal tissues regulated by China and the European Commission. The fluorescence-based aptasensor was applied to the detection of SDM in aquaculture water and fish samples with high accuracy, excellent precision, and ideal selectivity. The results indicated that the developed aptasensor was simple in design, easy to operate, and could be used to detect rapidly and accurately SDM in water and fish samples.

Abstract:Sulfadiazine is an environmental pollutant derived from abuse of antibiotics. Its content in environmental water is closely related to human health. Thus, a novel dual-emission surface molecularly imprinted nanosensor is designed for the specific adsorption and detection of sulfadiazine. In the system, blue emissive carbon quantum dots wrapped with silica served as the internal reference signal for eliminating background interference, while red emissive thioglycolic acid modified CdTe quantum dots (CdTe QDs), which are low dimensional semiconductor materials by the combination of cadmium and tellurium with excellent optical properties, were encapsulated in the imprinted layer to offer recognition signal. The fluorescence of CdTe quantum dots was quenched and the fluorescence quenching degree of carbon quantum dotswas inconspicuous with the increase of concentration of sulfadiazine, thereby reflecting the color change. The detection of sulfadiazine was successfully achieved in a concentration range of 0.25-20 mu mol/L with detection limit of 0.042 mu mol/L and nanosensors had specific recognition for sulfadiazine over its analogues. Compared to single-emission fluorescence sensors, ratiometric fluorescence nanosensors had wider linear range and higher detection accuracy. Furthermore, the nanosensors were also successfully applied for the determination of sulfadiazine in real water and milk samples with acceptable recoveries. The study provides a feasible method for the detection of sulfadiazine and a reference for the detection of sulfonamides.
Ultrasensitive and selective detection of sulfamethazine in milk via a Janus-labeled Au nanoparticle-based surface-enhanced Raman scattering-immunochromatographic assay

Abstract:Sulfamethazine (SM2) is an antibacterial drug,which has been extensively used in human and veterinary medicine, long-term consumption of which may lead to the accumulation of sulfonamides in the body. Detection of sulfonamides often uses microbiological approaches, mass spectrometry and chromatography, which are expensive and time-consuming. Surface-enhanced Raman scattering-based immunochromatographic assay (SERS-ICA) has been recently applied in the detection. Herein, a Janus-labeled Au nanoparticle with subnanosized SiO 2 -monoclonal antibody and SERS reporter (DTNB) modified simultaneously ( mAb AuNp DTNB ) has been developed in a SERS-based lateral flow immunosensor, which can be used for rapid, quantitative and ultrasensitive detection of sulfamethazine residue in milk. The mAb AuNp DTNB exhibits a specific array on a paper stripe, which not only identifies sulfamethazine but also straightforwardly exposes the Raman reporter between the AuNps via self-assembly. The detection sensitivity of SERS-ICA for sulfamethazine reached 0.1 pg/mL, which was far below the previously published value by ELISA and the maximum residue limit set by the European Union. The entire SERS-ICA detection for sulfamethazine was completed within 15 min. Furthermore, high accuracy for this assay was exhibited in the spiking experiment with a recovery percentage of 88.1%–112.7%. The results demonstrated that this SERS-ICA can potentially be applied in point-of-care testing as an ultrasensitive and quantitative to semi-quantitative analytical method.

Abstract:Abstract Sulfamethoxazole (SMX) is a sulfonamide antibiotic primarily used to treat urinary tract infections and used in veterinary and industrialized husbandry to treat diseases and food additives. Like other antibiotics, SMX is considered as a pollutant in water and food that threaten local life. This study developed a surface plasmon resonance (SPR) sensor chip that is fast, highly selective, and reusable, and requires no pretreatment for detecting SMX. As a receptor, SMX imprinted methacrylic acid-2-hydroxyethyl methacrylate-ethylene glycol dimethacrylate polymer [poly(MAA-HEMA-EGDMA)] was used. The surface of the gold SPR chips was coated with a drop-casting method. The nanofilm coated chips were characterized by scanning electron microscopy (SEM), atomic force microscopy (AFM), ellipsometer, contact angle measurement, and Fourier-transform infrared spectrometry (FTIR). Imprinting factor (IF) was calculated as: Δ R [MIP(molecularly imprinted polymers)]/Δ R [NIP(non-imprinted)]=12/3.5=3.4. Limit of detection (LOD) and limit of quantification (LOQ) values were calculated with 3 s/m and 10 s/m methods, and the results were found to be 0.001 1 μg/L for LOD 0.003 4 μg/L for LOQ. Adsorption studies on both standard SMX solution and commercial milk samples were applied. Also, we investigated the developed chip’s reusability, storability, and selectivity with amoxicillin and cefalexin.

Simple preparation of fluorescence probe based on CdTe quantum dots combined with MIP for selective detection of sulfadimidine

Abstract:A simple fluorescence sensor for the detection of sulfadimidine (SM2) was fabricated by molecular imprinting technology. Cadmium telluride quantum dots (CdTe QDs) as the fluorescent material, 3-aminopropyltriethoxysilane (APTES) as functional monomer, sulfadimidine as template molecule and tetraethoxysilane (TEOS) as cross-linker, the CdTe QDs were embedded in silica nano-particles by reverse microemulsion method. Under optimal detection condition, response is linear in the 2–30 μmol L−1 SM2 concentration condition with the detection limit was 0.4 μmol L−1. In addition, the correlation coefficient was 0.9857 and the imprinting factor was 4.82. The fluorescent probe was successfully applied to the determination of SM2 in tap water and milk sample with the spiked recoveries ranging from 99.8 to 103.2%. It gets a good specific recognition ability and potential practical application value.

Abstract:Identification of sulfonamides (SAs) residues in food is vital for human health. A set of 4-channel sensor array was constructed by carbon dots (CDs) embedded in photonic crystal molecularly imprinted (PCMIP@CDs) film which included 3 PCMIP@CDs units and 1 PCNIP@CDs unit to determine typical SAs: sulfadimethoxine, sulfathiazole, sulfaguanidine, sulfamethazine, sulfadiazine. Under the optimal conditions, the response time of the sensor array was only 200 s. Moreover, 300 fluorescence response signals (4 sensor units × 5 sulfonamides × 3 concentrations × 5 repeats) were processed by pattern recognition technique to analyze the ability of the sensor array to recognize 5 kinds of SAs. Subsequently, the linear discrimination analysis (LDA) method was used to identify the five SAs simultaneously with 100 % classification accuracy and the limit of detection was 0.01-0.26 nmol/L. Moreover, the proposed method can effectively identify-five SAs in water and fish samples.

Abstract:The widespread use of sulfonamide (SA) antibiotics in animal husbandry has led to residues of SAs in the environment, causing adverse effects to the ecosystem and a risk of bacterial resistance, which is a potential threat to public health. Therefore, it is highly desirable to develop simple, high-throughput methods that can detect multiple SAs simultaneously. In this study, we isolated aptamers with different specificities based on a multi-SA systematic evolution of ligands by the exponential enrichment (SELEX) strategy using a mixture of sulfadimethoxine (SDM), sulfaquinoxaline (SQX), and sulfamethoxazole (SMZ). Three aptamers were obtained, and one of them showed a similar binding to all tested SAs, with dissociation constant (Kd) ranging from 0.22 to 0.63 μM. For the other two aptamers, one is specific for SQX, and the other is specific for SDM and sulfaclozine. A label-free detection method based on the broad-specificity aptamer was developed for the simultaneous detection of six SAs, with detection of limits ranging from 0.14 to 0.71 μM in a lake water sample. The aptasensor has no binding for other broad-spectrum antibiotics such as β-lactam antibiotics, quinolones, tetracyclines, and chloramphenicol. This work provides a promising biosensor for rapid, multiresidue, and high-throughput detection of SAs, as well as a shortcut for the preparation of different specific recognition elements required for the detection of broad-spectrum antibiotics.

Abstract:A novel, molecularly imprinted, upconversion fluorescence probe (UCNP@MIFP) for sulfonamide sensing was fabricated by Pickering emulsion polymerization using UCNP@SiO2 particles as the stabilizer and sulfamethazine/sulfamerazine as the co-templates. The synthesis conditions of the UCNP@MIFP were optimized, and the synthesized probe was characterized by scanning electron microscopy, Fourier transform infrared spectrometer, thermogravimetric analyzer, and fluorescence spectrometer. The UCNP@MIFPs showed a good adsorption capacity and a fast kinetic feature for the template. The selectivity experiment revealed that the UCNP@MIFP has a broad-spectrum molecular recognition capability. Good linear relationships were obtained over the concentration range of 1-10 ng/mL for sulfamerazine, sulfamethazine, sulfathiazole, and sulfafurazole, with low limits of detection in the range of 1.37-2.35 ng/mL. The prepared UCNP@MIFP has the potential to detect four sulfonamide residues in food and environmental water.
A highly sensitive luminescent aptasensor utilizing MOF-74-co encapsulation of luminol in a 'turn-on' mode for streptomycin detection

Abstract:This study focused on detecting streptomycin (STR) residues using a luminescent aptasensor encapsulated with aptamer. Utilizing MOF-74-Co with peroxidase-like activity, luminol was enclosed in its pores. The specific STR aptamer acted as a gatekeeper, ensuring excellent performance. Upon exposure to STR, the aptamers detached, releasing luminol and amplifying the luminescent signal through MOF-74-Co catalytic activity. A linear relationship between fluorescence intensity and STR concentration (50 nM ∼ 5 × 106 nM) was established, with a limit of detection of 0.065 nM. The sensor exhibited high selectivity for STR even in the presence of other aminoglycoside antibiotics. Applied to tea, egg, and honey samples, the sensor showed recovery rates of 91.38-100.2%, meeting safety standards. This MOF-based aptasensor shows promise for detecting harmful residues.

Abstract:An electrochemical sensor based on single-atom cobalt-anchored nitrogen-doped carbon catalysts (Co/N‒C SACs), synthesized by pyrolyzing zeolitic imidazolate frameworks (Co-ZIF-8), was constructed to quantify sulfamethoxazole (SMX), a member of the sulfonamide family of antibiotics, in environmental water samples. The quantification depends on the electrocatalytic oxidation of the aromatic ‒NH2 group in the SMX molecule to the hydroxylamine group (–NHOH). The sensor was fabricated by depositing the Co/N‒C SAC suspension (in a mixture of water, ethanol, and Nafion) on a glassy carbon (GC) electrode surface. The response of the sensor showed a linear relationship with SMX in a wide range of concentration (0.08–66.5 μM) with a limit of detection (LOD) of ~15 nM at a signal-to-noise (S/N) ratio of 3. The sensor also had high repeatability, reproducibility, and stability. Moreover, it exhibited high selectivity and resistance to interference in the presence of other antibiotics (trimethoprim, ciprofloxacin, and norfloxacin selected randomly), endocrine disruptors (17β-estradiol), pesticides (carbaryl), common aqueous ions (Na+, K+, Fe3+, Cu2+, Ca2+, Cl–, Br–, CO32–, SO42–, and NO3–), and common surfactants (sodium dodecyl sulfate and Triton X-100). Furthermore, the sensor was used to quantify the SMX in environmental water samples (lake water, groundwater, and secondary wastewater) with high accuracy. This electrochemical method can be used for quantifying SMX in environmental water samples for various applications, including biomedical analysis, environmental pollutant detection, and water safety evaluation.
