Abstract:It is essential to reduce nitrogen losses and to improve nitrogen conversion during organic waste composting because of environmental protection and sustainable development. To reveal newly domesticated ammonifying microorganisms (AM) cultures on the ammonification and nitrogen conversion during the composting, the screened microbial agents were inoculated at 5 % concentration (in weight basis) into cow manure compost under five different treatments: sterilized distilled water (Control), Amm-1 (mesophilic fungus-F1), Amm-2 (mesophilic bacterium-Z1), Amm-3 (thermotolerant bacterium-Z2), and Amm-4 (consortium: F1, Z1, and Z2), and composted for 42 days. Compared to control, AM inoculation prolonged the thermophilic phases to 9–19 days, increased the content of NH 4 + -N to 1.60–1.96 g/kg in the thermophilic phase, reduced N 2 O and NH 3 emissions by 22.85–61.13 % and 8.45–23.29 %, increased total Kjeldahl nitrogen, and improved cell count and viability by 12.09–71.33 % and 66.71–72.91 %. AM was significantly associated with different nitrogen and microbial compositions. The structural equation model (SEM) reveals NH 4 + -N is the preferable nitrogen for the majority of bacterial and fungal growth and that AM is closely associated with the conversion between NH 3 and NH 4 + -N. Among the treatments, inoculation with Amm-4 was more effective, as it significantly enhanced the driving effect of the critical microbial composition on nitrogen conversion and accelerated nitrogen ammonification and sequestration. This study provided new concepts for the dynamics of microbial in the ammonification process of new AM bacterial agents in cow manure compost, and an understanding of the ecological mechanism underlying the ammonification process and its contribution to nitrogen (N) cycling from the perspective of microbial communities.

Performance of microbial inoculation and tricalcium phosphate on nitrogen retention and conversion: Core microorganisms and enzyme activity during kitchen waste composting

Abstract:The substantial release of NH3 during composting leads to nitrogen (N) losses and poses environmental hazards. Additives can mitigate nitrogen loss by adsorbing NH3/NH4, adjusting pH, and enhancing nitrification, thereby improving compost quality. Herein, we assessed the effects of combining bacterial inoculants (BI) (1.5%) with tricalcium phosphate (CA) (2.5%) on N retention, organic N conversion, bacterial biomass, functional genes, network patterns, and enzyme activity during kitchen waste (KW) composting. Results revealed that adding of 1.5%/2.5% (BI + CA) significantly (p < 0.05) improved ecological parameters, including pH (7.82), electrical conductivity (3.49 mS/cm), and N retention during composting. The bacterial network properties of CA (265 node) and BI + CA (341 node) exhibited a substantial niche overlap compared to CK (210 node). Additionally, treatments increased organic N and total N (TN) content while reducing NH4+-N by 65.42% (CA) and 77.56% (BI + CA) compared to the control (33%). The treatments, particularly BI + CA, significantly (p < 0.05) increased amino acid N, hydrolyzable unknown N (HUN), and amide N, while amino sugar N decreased due to bacterial consumption. Network analysis revealed that the combination expanded the core bacterial nodes and edges involved in organic N transformation. Key genes facilitating nitrogen mediation included nitrate reductase (nasC and nirA), nitrogenase (nifK and nifD), and hydroxylamine oxidase (hao). The structural equation model suggested that combined application (CA) and microbial inoculants enhance enzyme activity and bacterial interactions during composting, thereby improving nitrogen conversion and increasing the nutrient content of compost products.

Abstract:To advance in the assessment of therapeutic interventions for diseases that progress slowly but that lead ultimately to permanent disability, such as multiple sclerosis, rheumatoid arthritis or Crohn’s disease [CD], it is essential to evaluate the disease-modifying effects of available treatments. It is also necessary to separate these long-term effects from the short-term effects on symptoms that these treatments may afford. This separation of improvement in symptoms from change in primary disease progression can be difficult. In clinical trials, the measure of the effects of a particular therapeutic intervention is commonly done using indices. The majority of indices that have been developed for assessment of CD are measures of the severity of inflammatory activity, including evaluations based on symptoms and biomarkers [Crohn’s Disease Activity Index [CDAI]],1 endoscopy [Crohn’s Endoscopic Index of Severity [CDEIS]],2 or radiology [Magnetic Resonance Index of Activity [MaRIA]].3 Nevertheless, these are not indices reflecting purely inflammatory activity; each of these indices may be influenced by irreversible damage in the absence of active inflammation; for example, CDAI may increase due to bile salt–induced diarrhoea after ileal resection, CDEIS may increase due to presence of scars or fixed stenotic lesions, and MaRIA may increase due to persistent fibrotic thickening of the bowel wall. Abstract:This study investigates the application of specific microbial inoculants in the composting process of pig and chicken manure, resulting in a 9% increase in total nitrogen content. The introduction of these inoculants instigated a beneficial shift in the microbial community, augmenting nitrogen retention and fixation, while concurrently diminishing the prevalence of pathogens and antibiotic resistance genes. This research highlights the potential of microbial interventions to enhance the quality of agricultural compost and mitigate environmental risks. This study examined the effects of three Bacillus strains and one Saccharomyces cerevisiae strain on nitrogen transformation and microbial communities in pig and chicken manure compost. The findings revealed that the use of compound microbial inoculants increased the compost temperature, accelerated moisture reduction, enhanced cellulase activity, and stimulated the accumulation of NH4+‐N, NO3−‐N, and total nitrogen (TN), resulting in a 9% increase in TN content. The abundance of Firmicutes decreased by 3.95% at the maturation phase, while Actinobacteria and Bacteroidetes increased by 1.64% and 1.85%, respectively. Inoculation led to an increase in amoA, nxrA and nifH gene copy numbers, while simultaneously reducing the abundance of nirK, nosZ and nirS genes. It also resulted in an increase in functional enzyme levels, specifically nif and amo, with a corresponding decrease in nor. Clostridium, Phascolarctobacterium, Eubacterium and Faecalibacterium from the class Clostridium, which have a significant correlation with nifH and nxrA genes, suggest their likely crucial role in nitrogen retention and fixation. Inoculation aided in the removal of pathogenic bacteria and antibiotic resistance genes (ARGs) like fluoroquinolones, nucleosides and nitroimidazole. This study provides effective theoretical support for the mechanism of nitrogen retention and fixation, and for improving the quality of compost.

Abstract:Microbial inoculation plays a crucial role in enhancing the efficiency of biowaste conversion. In this study, we investigated the effects of inoculating a complex microbial agent called the BP consortium, comprising Bacillus licheniformis, Bacillus subtilis, and Penicillium chrysogenum , at different stages of swine manure (SM) aerobic composting, specifically the initial stage and cooling stage. Our objective was to examine the impact of BP inoculation on nitrogen conversion, enzyme activity, and microbial community dynamics through a 35-day aerobic composting experiment. Compared to the group without the inoculant, initial-stage inoculation of BP led to a remarkable increase of 23.12% and 140.91% in the germination index and NO 3 − -N content of the final product, respectively. Furthermore, it effectively reduced NH 3 and N 2 O releases by 65.73% and 18.76% during the SM composting process. The activities of urease also exhibited significant increase at the conclusion of the experiment. Additionally, we observed a proportional increase in the number of Proteobacteria and Actinobacteria during the thermophilic stage of composting. However, BP inoculation during the cooling phase did not demonstrate a positive effect on nitrogen retention. Nevertheless, the findings from this investigation provide valuable guidance for the implementation of BP consortium inoculation to mitigate nitrogen loss during SM aerobic composting.

Abstract:Biofertilizers are considered as alternatives to chemical fertilizers for enhancing plant growth. However, only a few microorganisms responsible for nitrogen mineralization have been exploited. Furthermore, mineralization after biofertilizer application has not been well studied. In this paper, mineralization was assessed in soil samples inoculated or not with the ammonifier Bacillus amyloliquefaciens DT over a five-week incubation period. The results showed that ammonifying bacteria amendment increased CO2 and N2O emission, as well as inorganic nitrogen and total soluble nitrogen concentrations. Importantly, after inoculation, the net nitrogen mineralization rate increased by 23.23% and 32.16% in the unfertilized control and pig manure with chemical fertilizer soil samples, respectively. High-throughput sequencing of internal transcribed spacer sequences and 16S rRNA showed that ammonifier amendment changed the composition of the microbial community. Notably, members of the bacterial phyla Firmicutes and Actinobacteria and genera Lysobacter and Sphingopyxis were enriched, while those belonging to the fungal phylum Ascomycota were depleted after B. amyloliquefaciens DT inoculation, verifying that this ammonifier DT contributed to disease suppression in terms of community structure. Thus, it is concluded that B. amyloliquefaciens DT can serve as a potential biofertilizer for promoting plant growth owing to its abilities of mineralization and disease suppression.

Abstract:Humus is the main product of organic waste composting in agriculture. The conversion of carbon and nitrogen in composting is controlled by ammonifying microorganisms (AM), but their role and mechanisms in humus formation are unclear. This study used multiple spectroscopic and microbiological techniques to explore how new ammonifying microorganism cultures (NAMC) molecularly affect humus formation in cow manure and straw compost. NAMC increased humus by 3.45 % – 9.46 %, resulting in enhanced humic acid and reduced fulvic acid. NAMC (especially Amm-4) also altered the chemical properties of humus, producing highly reactive amides and creating binding sites for low molecular weight compounds. NAMC enhanced the formation of O-H and C = O functional groups, which contributed to the formation of the aromatic structure and facilitated the three-dimensional polymerization of humus. Furthermore, NAMC promoted the Maillard and lignin pathways of humus formation, as revealed by structural equation models. This study provides new insights into the molecular mechanisms of NAMC-mediated humus formation during composting.

Unraveling impacts of inoculating novel microbial agents on nitrogen conversion during cattle manure composting: Core microorganisms and functional genes

Abstract:The impacts of microbial agents on nitrogen conversion during composting is still not entirely clear. In this research, a novel microbial agent containing two thermotolerant nitrifying bacteria was identified and its impacts on nitrogen conversion, bacterial structure and functional genes during cattle manure composting were investigated. The results revealed that the inoculation enhancing the maturation of compost, increased the total nitrogen by 13.6-26.8%, reduced NH3 emission and the N2O emission by 24.8-36.1% and 22.7-32.1%, respectively. Particularly, the microbial agents mixed Acinetobacter radioresistens and Bacillus nitratireducens (1:1, treatment group 1) had the best nitrogen preservation effect. Furthermore, the inoculation not only produced diverse diazotroph community but could strength the co-occurrence between core microorganisms to promote nitrogen metabolism. The metagenomic analysis demonstrated that the inoculation decreased the abundance of nitrate reduction gene (nirS, norC, nap and nif), and increased the abundance of hao, thus facilitating nitrification and suppressing NH3 and N2O emission.

Abstract:To address the challenge of increasing nitrogen retention in compost, this study investigated the effects of microbial communities on denitrification and ammonia assimilation during sludge composting by inoculating microbial inoculants . The results showed that the retention rates of total Kjeldahl nitrogen (TKN) and humic acid (HA) in MIs group (with microbial inoculants) were 4.94 % and 18.52 % higher than those in the control group (CK), respectively. Metagenomic analysis showed that Actinobacteria and Proteobacteria were identified as main microorganisms contributing to denitrification and ammonia assimilation. The addition of microbial agents altered the structure of the microbial community, which in turn stimulated the expression of functional genes. During cooling period, the ammonia assimilation genes glnA , gltB and gltD in MIs were 15.98 %, 24.84 % and 32.88 % higher than those in CK, respectively. Canonical correspondence analysis revealed a positive correlation between the dominant bacterial genera from the cooling stage to the maturity stage and the levels of NO 3 − -N, NH 4 + -N, HA, and TKN contents. NH 4 + -N was positively correlated with HA, indicating NH 4 + -N might be incorporated into HA. Heat map and network analyses revealed NH 4 + -N as a key factor affecting functional genes of denitrification and ammonia assimilation, with Nitrospira identified as the core bacteria in the microbial network. Therefore, the addition of microbial agents could increase nitrogen retention and improve compost product quality.

Abstract:In this study, three thermophilic heterotrophic nitrifiers were isolated and inoculated into the semi-permeable membrane-covered composting of sewage sludge as thermophilic microbial (TM) agent. The physicochemical properties and microbial communities as well as the metabolism of microorganisms during composting were analyzed. Compared to CK, the inoculation of TM agent prolonged the thermophilic stage from 4 days to 7 days, increased the degradation of organic matters by 14.06 %, and promoted humification of compost. Meanwhile, TM agent reduced the NH 3 emission by 34.64 % while the NO 3 − -N content was 2.72 folds that of CK, resulting lower nitrogen loss. Additionally, an obvious succession of microbial communities occurred and the inoculation of TM agent enriched organic matters-degrading microorganisms ( Bacillus , Thermobispora , Planifilum and Tepidimicrobium ) and decreased the abundance of pathogens ( Aspergillus and Cephalotrichum ). Besides, many microorganisms related to nitrogen transformation were detected during the composting. The analysis of functional genes revealed that the nitrification was facilitated while denitrification process might be inhibited, establishing an efficient nitrogen retention system. This research provided an effective agent for sludge composting and indicated that the inoculation of thermophilic nitrifiers was a promising strategy to improve composting efficiency and reduce nitrogen loss.

Effect of simplified inoculum agent on performance and microbiome during cow manure-composting at industrial-scale

Abstract:A simplified inoculum agent, only comprising Bacillus subtilis and Aspergillus niger, was utilized for industrial-scale cow-manure composting to investigate its impact on composting performance and microbiome. Inoculants elevated the average and peak temperatures by up to 7 and 10 °C, respectively, during the thermophilic stage, reduced organic matter content, and raised germination index. Inoculation also extended the period of composting above 50 °C from 12 to 26 days. Sequencing unveiled significant shifts in microbial diversity, composition, and function. Aspergillus thrived during the mesophilic phase, potentially initiating composting, whereas Bacillus, Lysinibacillus, and Clostridium were enriched during the thermophilic stage. Metagenomic sequencing revealed an increased abundance of carbohydrate-active enzymes and glycometabolism-related genes responsible for lignocellulose degradation and heat generation after inoculation. These enriched microbes and functional genes contributed to organic matter degradation and temperature maintenance during thermophilic stage, expediting composting. This suggests the effectiveness of this simplified inoculum in industrial-level cow-manure composting.

Abstract:Microorganisms can play a crucial role in the efficiency for composting, which are essential for converting the organic wastes into a well-stabilized, value added product. However, the activity of most of the key functional microorganisms were inhibited due to the limited special nutrient substances or other physiochemical factors during composting, which further affected the quality of compost. The study was conducted to investigate the effects of enriched ammonium (NH4+-N) and organic nitrogen (Org-N) on indigenous microbial community and whether nitrogen (N) nutrient additions could modify the special species during composting. The results showed that the abundance and structure of bacterial community had distinctly diverse responses to different N nutritional treatments (no nutrient addition, NH4+-N addition, and Org-N addition). The addition of N sources enhanced the abundance of corresponding uncultured indigenous species negatively related to the factor of NH4+ and Org-N in redundancy analysis (RDA) during composting but the effect of NH4+ was more significant than Org-N. Nonmetric multidimensional scaling ordination (NMDS) demonstrated that both the two N additions changed bacterial community but had different duration for affecting bacterial composition. Conclusively, an optimized method for regulating the key stains with special biological capacity is proposed by controlling the single limiting-nutrient factor sharply decreasing at one of composting stages and negatively related to the key species in RDA.
Control of nitrogen and odor emissions during chicken manure composting with a carbon-based microbial inoculant and a biotrickling filter

Abstract:Although aerobic composting is usually utilized in livestock manure disposal, the emission of odorous gases from compost not only induces harm to the human body and the environment, but also causes loss of nitrogen, sulfur, and other essential elements, resulting in a decline in product quality. The impact of biotrickling filter (BTF) and insertion of carbon-based microbial agent (CBMA) on compost maturation, odor emissions, and microbial population during the chicken manure composting were assessed in the current experiment. Compared with the CK group, CBMA addition accelerated the increase in pile temperature (EG group reached maximum temperature 10 days earlier than CK group), increased compost maturation (GI showed the highest increase of 41.3% on day 14 in EG group), resulted in 36.59% and 14.60% increase in NO 3 − -N content and the total nitrogen retention preservation rate after composting. The deodorization effect of biotrickling filter was stable, and the removal rates of NH 3 , H 2 S, and TVOCs reached more than 90%, 96%, and 56%, respectively. Furthermore, microbial sequencing showed that CBMA effectively changed the microbial community in compost, protected the ammonia-oxidizing microorganisms, and strengthened the nitrification of the compost. In addition, the nitrifying and denitrifying bacteria were more active in the cooling period than they were in the thermophilic period. Moreover, the abundance of denitrification genes containing nirS , nirK , and nosZ in EG group was lower than that in CK group. Thus, a large amount of nitrogen was retained under the combined drive of BTF and CBMA during composting. This study made significant contributions to our understanding of how to compost livestock manure while reducing releases of odors and raising compost quality.

Abstract:Microbial inoculation plays a significant role in promoting the efficiency of biowaste conversion. This study investigates the function of Streptomyces-Bacillus Inoculants (SBI) on carbon (C) and nitrogen (N) conversion, and microbial dynamics, during cow manure (10% and 20% addition) and corn straw co-composting. Compared to inoculant-free controls, inoculant application accelerated the compost's thermophilic stage (8 vs 15 days), and significantly increased compost total N contents (+47%) and N-reductase activities (nitrate reductase: +60%; nitrite reductase: +219%). Both bacterial and fungal community succession were significantly affected by DOC, urease, and NH 4 + -N, while the fungal community was also significantly affected by cellulase. The contribution rate of Cupriavidus to the physicochemical factors of compost was as high as 83.40%, but by contrast there were no significantly different contributions (∼60%) among the top 20 fungal genera. Application of SBI induced significant correlations between bacteria, compost C/N ratio, and catalase enzymes, indicative of compost maturation. We recommend SBI as a promising bio-composting additive to accelerate C and N turnover and high-quality biowaste maturation. SBI boosts organic cycling by transforming biowastes into bio-fertilizers efficiently. This highlights the potential for SBI application to improve plant growth and soil quality in multiple contexts.

Abstract:This study conducted composting experiments using cow manure and wheat straw, employing five different treatment methods (CK, magnetized water treatment-M, magnetized water combined with biochar treatment-SM, magnetized water combined with ferrous sulfate treatment-FM, magnetized water combined with Bacillus megaterium treatment-GM), aiming to accelerate the composting maturation process and reduce carbon loss. The results of the experiments showed that the SM treatment entered the thermophilic phase on the second day and lasted for 9 days, resulting in the best maturation effect. The FM treatment increased the carbon content in the compost, effectively preserving carbon sources. principal component analysis analysis revealed significant differences in microbial community structures due to different treatments. Additionally, structural equation modeling observations indicated that different treatments affected the compost environment, thereby influencing microbial activity and carbon content. Overall, the SM treatment positively affected the compost maturation process, while the FM treatment effectively preserved carbon sources in the compost.

Abstract:The bioaugmented mechanical composting (BMC) of kitchen waste with rapid heating, dehydration, and humification is promising for nitrogen conservation. The combination of ammonia assimilation and rapid humification in BMC may be a distinct nitrogen conservation pathway, converting NH4 +-N to recalcitrant organic nitrogen reservoirs. However, questions remain regarding the existence of this pathway and its enhancing mechanism. Thus, nitrogen transformation during a BMC process and a conventional pile composting (CPC) process were compared in this study. Results indicated a significant increase (68.53–75.56%) in the proportion of initial total nitrogen in raw materials preserved in recalcitrant humins in BMC compared to CPC. The significantly higher activity of glutamate dehydrogenase (64.65 ± 7.10 U·mg−1) and glutamine synthase (29.92 ± 3.37 U·mg−1) associated with ammonia assimilation in BMC facilitated the conversion of NH4 + from ammonification into amino acids at the thermophilic phase. These amino acids were then rapidly polymerised with other humic precursors and gradually condensed into heterocyclic and quaternary nitrogen in humins. Further analysis of the bacterial communities and their potential functions revealed that Firmicutes (e.g., Bacillus) and Proteobacteria (e.g., Acinetobacter), which are capable of mineralisation, were enriched by rapid heating and dehydration in BMC. The resulting accumulated NADPH, α-ketoglutaric acid, and humic precursors activated thermophiles associated with ammonia assimilation (e.g., Ureibacillus, Kroppenstedtia, Virgibacillus, and Tepidimicrobium) and humification (e.g., Sacchsromonospora and Bacilli), accelerating nitrogen transformation. This study provides a theoretical foundation for the development of novel nitrogen conservation strategies of coupling ammonia assimilation with rapid humification through enhanced mineralisation during composting.

Abstract:This study proposed combining food waste (FW) and cattle manure (CM) in composting to improve the product maturity. The findings suggested that the inclusion of CM effectively extended the thermophilic stage, facilitated the decomposition of cellulose, and enhanced the production of humus-like substances by enhancing beneficial microbial cooperation. Adding 40 % CW was optimal to reduce the nitrogen loss, increase the cellulose degradation rate to 22.07 %, increase germination index (GI) to 140 %, and reduce normalized antibiotic resistance gene (ARG) abundances. Adding CW could promote the transformation of protein-like compounds, thereby enhancing the humification process of organic substances. Structural equation modeling further verified that the temperature was the key factor affecting humification production, while the main driver for ARGs was physiochemical parameters. This study shows that co-composting of FW and CM offers the potential to promote humification, reduce ARG abundance, and improve fertilizer quality for utilization of both biowastes in the field.

Abstract:This work aimed to investigate the microbial mechanisms for the improvement of composting efficiency driven by the compound microbial inoculum (MI) ( Bacillus subtilis SL-44, Enterobacter hormaechei Rs-189 and Trichoderma reesei ) during co-composting of spent mushroom substrate (SMS) and chicken manure (CM). The treatments used in the study were as follows: 1) MI (inoculation with microbial inoculum), 2) CI (inoculation with commercial microbial inoculum), and 3) CK (without inoculation). The results demonstrated that MI increased the seed germination index (GI) by 25.11%, and contents of humus, humic acid (HA) and available phosphorus (AP) were correspondingly promoted by 12.47%, 25.93% and 37.16%, respectively. The inoculation of MI increased the temperature of the thermophilic stage by 3–7 °C and achieved a cellulose degradation rate of 52.87%. 16S rRNA gene analysis indicated that Actinobacteria (11.73–61.61%), Firmicutes (9.46–65.07%), Proteobacteria (2.86–32.17%) and Chloroflexi (0.51–10.92%) were the four major phyla during the inoculation composting. Bacterial metabolic functional analysis revealed that pathways involved in amino acid and glycan biosynthesis and metabolism were boosted in the thermophilic phase. There was a positive correlation between bacterial communities and temperature, humification and phosphorus fractions. The average dry weight, fresh weight and seedling root length in the seedling substrates adding MI compost were 1.13, 1.23 and 1.06 times higher than those of the CK, respectively. This study revealed that biological inoculation could improve the composting quality and efficiency, potentially benefiting the resource utilization of agricultural waste resources.

Abstract:The discovery of complete ammonia oxidizers (comammox) has dramatically altered our perception of nitrogen (N) biogeochemistry. However, their functional importance vs. the canonical ammonia oxidizers (i.e., ammonia oxidizing-archaea (AOA) and bacteria (AOB)) in agroecosystems is still poorly understood. Accordingly, a new assay using acetylene, 3,4-dimethylpyrazole phosphate (DMPP), and 1-octyne was adopted to assess the ammonia (NH3) oxidation and nitrous oxide (N2O) production activity of these functional guilds in a subtropical Inceptisol under long-term different fertilization regimes. These regimes include CK (no fertilizer control), synthetic fertilizer only (NPK), organic manure only (M) and organic manure plus synthetic fertilizer (MNPK). AOA dominated NH3 oxidation in the M treatment, while AOB dominated both NH3 oxidation and N2O production in all treatments except M. Comammox always played a minor role in both NH3 oxidation and N2O production across all treatments. Both M and MNPK treatments significantly increased the activity and growth of comammox. Compared to NPK, comammox exhibited increases of 270 % and 326 % in the NH3 oxidation rates, and increases of 1472 % and 563 % in the N2O production rates in M and MNPK, respectively. Random forest model revealed that copper (Cu), comammox abundance, and dissolved organic nitrogen (DON) were the most important predictors for the NH3 oxidation rates of comammox. Redundancy analyses (RDA) showed that fertilizer treatments significantly altered the community composition of NH3 oxidizers, and pH was the overarching parameter underpinning the community shift of the NH3 oxidizers. Overall, this study provides evidence that comammox play a minor yet unneglectable role in the nitrification of agroecosystems, and the long-term addition of organic manure stimulates the growth and activity of comammox in a subtropical Inceptisol.
