Abstract:Chromium (Cr) is a toxic heavy element that interferes with plant metabolite biosynthesis and modifies the plant rhizosphere microenvironment, affecting plant growth. However, the interactions and response mechanisms between plants and rhizosphere bacteria under Cr stress still need to be fully understood. In this study, we used Iris tectorum as a research target and combined physiology, metabolomics, and microbiology to reveal the stress response mechanism of I. tectorum under heavy metal chromium stress. The results showed that Cr stress-induced oxidative stress inhibited plant growth and development and increased malondialdehyde and oxygen free radicals content. Also, it increased ascorbate peroxidase, peroxidase activity, and superoxide dismutase activity, as well as glutathione and soluble sugar content. Microbiome analysis showed that Cr stress changed the rhizosphere bacterial community diversity index by 33.56%. Proteobacteria, Actinobacteriota, and Chloroflexi together accounting for 71.21% of the total sequences. Meanwhile, the abundance of rhizosphere dominant and plant-promoting bacteria increased significantly with increasing time of Cr stress. The improvement of the soil microenvironment and the recruitment of bacteria by I. tectorum root secretions were significantly enhanced. By metabolomic analysis, five vital metabolic pathways were identified, involving 89 differentially expressed metabolites, divided into 15 major categories. In summary, a multi-omics approach was used in this study to reveal the interaction and stress response mechanisms between I. tectorum and rhizosphere bacterial communities under Cr stress, which provided theoretical basis for plant-microbial bioremediation of Cr-contaminated soils in constructed wetlands. This may provide more valuable information for wetland remediation of heavy metal pollution.

Abstract:Soil chromium (Cr) contamination poses a significant threat to the environment and public health. A phosphate solubilizing bacterial strain Bacillus megaterium PMW-03 was used to enhance the phytoremediation of Cr(VI) contamination by Pteris vittata in phosphate mining wasteland in this study. The results showed that the strain PMW-03 could promote the solubilization of phosphate rock (PR) and the removal of Cr(VI) by Pteris vittata in the phosphate mining wasteland. Compared with the groups not inoculated with PMW-03, the available phosphate content in the rhizosphere soil of the inoculated groups increased by 17.6% and 24.9%, respectively (in the group with Cr(VI) added and the group with Cr(VI) and PR added), and the available Cr(VI) content decreased by 28.2%, 25.2% and 33.4%, respectively. In addition, the strain PMW-03 also promoted the growth of Pteris vittata by secreting indole acetic acid (IAA) and siderophore, promoting Pteris vittata to produce chlorophyll, increasing the concentration of soluble phosphate and reducing the concentration of Cr(VI) in root and shoot. The results of metagenomic analysis showed that changes in biological community structure, enrichment of related functional abundances, and activation of genes (FTH1, FTL, HSP90A, glgP, htpG, glnA, and GLUD1_2) located in microbes contributed to the microbial-phytoremediation of Cr(VI) contamination. These results provide a reference for future research on the microbial-phytoremediation of Cr(VI) contamination in phosphate mining wasteland.

Abstract:The understanding of bacterial resistance to hexavalent chromium [Cr(VI)] are crucial for the enhancement of Cr(VI)-polluted soil bioremediation. However, the mechanisms related to plant-associated bacteria remain largely unclear. In this study, we investigate the resistance mechanisms and remediation potential of Cr(VI) in a plant-associated strain, AN-B15. The results manifested that AN-B15 efficiently reduced Cr(VI) to soluble organo-Cr(III). Specifically, 84.3 % and 56.5 % of Cr(VI) was removed after 48 h in strain-inoculated solutions supplemented with 10 and 20 mg/L Cr(VI) concentrations, respectively. Transcriptome analyses revealed that multiple metabolic systems are responsible for Cr(VI) resistance at the transcriptional level. In response to Cr(VI) exposure, strain AN-B15 up-regulated the genes involved in central metabolism, providing the reducing power by which enzymes (ChrR and azoR) transformed Cr(VI) to Cr(III) in the cytoplasm. Genes involved in the alleviation of oxidative stress and DNA repair were significantly up-regulated to neutralize Cr(VI)-induced toxicity. Additionally, genes involved in organosulfur metabolism and certain ion transporters were up-regulated to counteract the starvation of sulfur, molybdate, iron, and manganese induced by Cr(VI) stress. Furthermore, a hydroponic culture experiment showed that toxicity and uptake of Cr(VI) by plants under Cr(VI) stress were reduced by strain AN-B15. Specifically, strain AN-B15 inoculation increased the fresh weights of the wheat root and shoot by 55.5 % and 18.8 %, respectively, under Cr(VI) stress (5 mg/L). The elucidation of bacterial resistance to Cr(VI) has an important implication for exploiting microorganism for the effective remediation of Cr(VI)-polluted soils.

Abstract:As chromium (Cr) in ecosystems affects human health through food chain exposure, phytoremediation is an environmentally friendly and efficient way to reduce chromium pollution in the environment. Here, we review the mechanism of absorption, translocation, storage, detoxification, and regulation of Cr in plants. The Cr(VI) form is more soluble, mobile, and toxic than Cr(III), reflecting how various valence states of Cr affect environmental risk characteristics, physicochemical properties, toxicity, and plant uptake. Plant root's response to Cr exposure leads to reactive oxygen species (ROS) generation and apoptosis. Cell wall immobilization, vacuole compartmentation, interaction of defense proteins and organic ligand with Cr, and removal of reactive oxygen species by antioxidants continue plant life. In addition, the combined application of microorganisms, genetic engineering, and the addition of organic acids, nanoparticles, fertilization, soil amendments, and other metals could accelerate the phytoremediation process. This review provides efficient methods to investigate and understand the complex changes of Cr metabolism in plants. Preferably, fast-growing, abundantly available biomass species should be modified to mitigate Cr pollution in the environment as these green and efficient remediation technologies are necessary for the protection of soil and water ecology.
The Shifts of Sediment Microbial Community Phylogenetic and Functional Structures During Chromium (VI) Reduction

Abstract:The Lanzhou reach of the Yellow River, located at the upstream of Lanzhou, has been contaminated by heavy metals and polycyclic aromatic hydrocarbons over a long-time. We hypothesized that indigenous microbial communities would remediate those contaminants and some unique populations could play an important role in this process. In this study, we investigated the sediment microbial community structure and function from the Lanzhou reach. Sediment samples were collected from two nearby sites (site A and site B) in the Lanzhou reach along the Yellow River. Sediment geochemical property data showed that site A sediment samples contained significantly (p < 0.05) higher heavy metals than site B, such as chromium (Cr), manganese (Mn), and copper (Cu). Both site A and B samples were incubated with or without hexavalent chromium (Cr (VI)) for 30 days in the laboratory, and Cr (VI) reduction was only observed in site A sediment samples. After incubation, MiSeq sequencing of 16S rRNA gene amplicons revealed that the phylogenetic composition and structure of microbial communities changed in both samples, and especially Proteobacteria, as the most abundant phylum increased from 45.1 % to 68.2 % in site A, and 50.1 % to 71.3 % in site B, respectively. Some unique OTUs and populations affiliated with Geobacter, Clostridium, Desulfosporosinus and Desulfosporosinus might be involved in Cr (VI) reduction in site A. Furthermore, GeoChip 4.0 (a comprehensive functional gene array) data showed that genes involved in carbon and nitrogen cycling and metal resistance significantly (p < 0.05) increased in site A sediment samples. All the results indicated that indigenous sediment microbial communities might be able to remediate contaminants like Cr (VI), and this information provides possible strategies for future bioremediation of the Lanzhou reach.
Chromium (vi) Uptake and Tolerance Potential in Cotton Cultivars: Effect on Their Root Physiology, Ultramorphology, and Oxidative Metabolism

Abstract:Chromium (Cr) is present in our environment as a toxic pollutant, which needs to be removed using phytoremediation technology. In present study, two transgenic cotton cultivars (J208, Z905) and their hybrid line (ZD14) were used to explore their Cr uptake and tolerance potential using multiple biomarkers approach. Four different levels of Cr (CK, 10, 50, and 100 μM) were applied. Cr caused a significant reduction in root/shoot length, number of secondary roots, and root fresh and dry biomasses at 100 μM. Cr accumulated more in roots and was found higher in hybrid line (ZD14) as compared with its parent lines (J208, Z905) at all Cr stress levels (10, 50, and 100 μM). Cr translocation was less than 1 in all cultivars. Ultrastructural studies at 100 μM Cr showed an increase in number of nuclei and vacuoles and presence of Cr dense granules in dead parts of the cell (vacuoles/cell wall). Malondialdehyde (MDA), hydrogen peroxide (H2O2), total soluble proteins, superoxide dismutase (SOD), peroxidase (POD), ascorbate peroxidase (APX), catalase (CAT), and glutathione reductase (GR) as a whole were upregulated with elevated levels of Cr. Higher Cr uptake by roots, accelerated metabolism, and Cr sequestration in dead parts of the cell indicate that these cotton cultivars can be useful for Cr accumulation and tolerance.
Chromium-Resistant Bacteria Promote the Reduction of Hexavalent Chromium in Soils

Abstract:Knowledge of the chromium (Cr) redox process in soil is important in addressing Cr bioavailability and risk assessment of contaminated soils. In this study, seven representative agricultural soils with different physicochemical properties were used to investigate the importance of microbially mediated Cr(VI) reduction and the response of soil bacterial community to Cr contamination. Chromium application increased soil bacterial diversity in Periudic Argosols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols, with an exception of Udic Ferrisols. The soil bacterial community responded to Cr contamination through changes in bacterial community structure, with Cr-resistant bacteria becoming the dominant species, and the percentage of Cr-resistant bacteria of total cultivable bacteria was 89.9, 75.2, 92.8, 65.3, 72.8, 77.3, and 65.4%, respectively, for Periudic Argosols, Udic Ferrisols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols. , , , , , , , , , and were identified as the Cr-resistant bacteria. Moreover, our results demonstrated that microbial reduction was an important Cr(VI) reduction pathway, and the relative contribution of microorganisms to Cr(VI) reduction was 14.4, 44.0, 20.6, 34.9, 21.9, 21.7, and 22.0%, respectively for Periudic Argosols, Udic Ferrisols, Calcaric Regosols, Stagnic Anthrosols, Mollisols, Typic Haplustalfs, and Ustic Cambosols. Soil properties, especially Fe(II) and soil particle distribution, affected the microbially mediated Cr(VI) reduction. These results provide useful information for the bioremediation of Cr-contaminated soils under a wide range of environmental conditions.
Impact of plant-associated bacteria biosensors on plant growth in the presence of hexavalent chromium

Abstract:Cr(VI) is a highly toxic metal produced by anthropogenic activity which may impact the environment, affecting plants and animals. In plants, chromium both as Cr(III) or Cr(VI) can be absorbed by roots, is poorly translocated and affects negatively plant growth. Plants used in phytoremediation need to cope with chromium toxicity. This work aimed to evaluate strains of Ochrobactrum tritici and Nitrospirillum amazonense, resistant and modified in order to become chromate whole-cell biosensors, as plant-protectors enabling plants to withstand contaminated soils. In vitro tests were performed in three rice varieties and one maize variety. Initial evaluations of Cr(VI) toxicity to plants showed that plants had different sensitivities and BRS 6 CHUÍ rice variety was the most resistant. The metal affected plant growth and development, essentially in roots which were totally inhibited in rice varieties at 500 μM. This effect was plant-dependent. Modified N. amazonense proved to protect maize plants independently of the inoculation dose but O. tritici showed plant specificity and some toxicity when inoculated at high numbers, inhibiting rice development but not maize. Inoculants were directly responsible for growth improvements of specific plant varieties at 1.25 ppm Cr(VI), a concentration which corresponds to a weak soil contamination. Improvements were observed relatively to the Cr(VI)-treated controls, but also relative to the untreated controls, i.e., the benefits went beyond a simple neutralization of inhibition brought by Cr(VI) toxicity.
Transcriptome and metabolome analysis reveal key genes and metabolic pathway responses in Leersia hexandra Swartz under Cr and Ni co-stress

Abstract:Phytoremediation, an eco-friendly approach for mitigating heavy metal contamination, is reliant on hyperaccumulators. This study focused on Leersia hexandra Swart, a known chromium (Cr) hyperaccumulator with demonstrated tolerance to multiple heavy metals. Our objective was to investigate its response to simultaneous Cr and nickel (Ni) stress over 12 days. Results from physiological experiments demonstrated a significant increase in the activities of antioxidant enzymes (APX, SOD, CAT) and glutathione (GSH) content under Cr and Ni stress, indicating enhanced antioxidant mechanisms. Transcriptome analysis revealed that stress resulted in the differential expression of 27 genes associated with antioxidant activity and metal binding, including APX, SOD, CAT, GSH, metallothionein (MT), and nicotinamide (NA). Among them, twenty differentially expressed genes (DEGs) related to GSH metabolic cycle were identified. Notably, GSTU6, GND1, and PGD were the top three related genes, showing upregulation with fold changes of 4.57, 6.07, and 3.76, respectively, indicating their crucial role in metal tolerance. The expression of selected DEGs was validated by quantitative real-time PCR, confirming the reliability of RNA-Seq data. Metabolomic analysis revealed changes in 1121 metabolites, with amino acids, flavonoids, and carbohydrates being the most affected. Furthermore, glucosinolate biosynthesis and amino acid biosynthesis pathways were represented in the KEGG pathway of differentially expressed metabolites (DEMs). This study provides insights into the tolerance mechanisms of L. hexandra under the co-stress of Cr and Ni, offering a new perspective for enhancing its remediation performance.
Evaluation of the phytoremediation potential of dominant plant species growing in a chromium salt-producing factory wasteland, China

Abstract:The metal contents of the soil and plant tissues in a large chromium salt-producing factory wasteland were determined to assess the properties of soil contamination and to identify plant species accumulating a range of heavy metals. Total metal contents in the factory soils presented a high heterogeneity, and the principal contaminants were Cd and Cr. All plant species examined were metal-tolerant, but to different extents. Especially, the maximum accumulation of Cd (15.61 mg kg-1) and Cr (925.07 mg kg-1) was found in Melia azedarach L. Subsequently, the Cd and Cr bioaccumulation and diverse physiological properties of M. azedarach seedlings exposed to different concentrations of Cd(II), Cr(VI), or Cd(II) + Cr(VI) in nutrient solutions were further investigated. All treated seedlings were able to survive under heavy metal stress, and the accumulation of both metals in plant tissues increased with elevation of metal exposure strength. M. azedarach showed a BCF greater than 147.56 for Cd and 36.76 for Cr. Meanwhile, the TF was lower than 0.25 for Cd and 0.32 for Cr. The highest bioaccumulation in root tissues was 2708.03 mg kg-1 Cd and 824.65 mg kg-1 Cr for seedlings cultured with 20 mg L-1 Cd(II) or 20 mg L-1 Cr(VI). Cd and Cr increased each other's uptake in seedlings although a reduced accumulation in roots occurred when exposed to the highest concentration of Cd(II) + Cr(VI) treatment (20 mg L-1). At either level of concentration, the degree of plant growth inhibition and oxidative damage caused by heavy metals was Cd(II) + Cr(VI) > Cr(VI) > Cd(II). Superoxide dismutase and peroxidase exhibited positive and effective responses to low-Cd(II) or Cr(VI) concentration stress, but their activities decreased with increasing metal exposure strength. The behavior of the non-enzymatic antioxidants (GSH, soluble protein, and proline) in plant involved in the detoxification of ROS induced by metal exposure was correlated well with higher Cd and Cr accumulations. Here, the potentiality of M. azedarach with the capacity to accumulate and stabilize Cd/Cr in metal-contaminated soil by phytoremediation process has been explored.

Abstract:Arbuscular mycorrhizal fungi (AMF) and plant growth-promoting bacteria enhance plant tolerance to abiotic stress and promote plant growth in contaminated soil. However, the interaction mechanism between rhizosphere microbial communities under chromium (Cr) stress remains unclear. This study conducted a greenhouse pot experiment and metagenomics analysis to reveal the comprehensive effects of the interaction between AMF (Rhizophagus intraradices) and nitrogen-N metabolizing plant growth promoters on the growth of Iris tectorum. The results showed that AMF significantly increased the biomass and nutrient levels of I. tectorum in contaminated soil and decreased the content of Cr in the soil. Metagenomics analysis revealed that the structure and composition of the rhizosphere microbial community involved in nitrogen metabolism changed significantly after inoculation with AMF under Cr stress. Functional genes related to soil nitrogen mineralization (gltB, gltD, gdhA, ureC, and glnA), nitrate reduction to ammonium (nirB, nrfA, and nasA), and soil nitrogen assimilation (NRT, nrtA, and nrtC) were up-regulated in the N-metabolizing microbial community. In contrast, the abundance of functional genes involved in denitrification (nirK and narI) was down-regulated. In addition, the inoculation of AMF regulates the synergies between the N-metabolic rhizosphere microbial communities and enhances the complexity and stability of the rhizosphere ecological network. This study provides a basis for improving plant tolerance to heavy metal stress by regulating the functional abundance of N-metabolizing plant growth-promoting bacteria through AMF inoculation. It helps to understand the potential mechanism of wetland plant remediation of Cr-contaminated soil.

Abstract:Agricultural soils contaminated with heavy metals poison crops and disturb the normal functioning of rhizosphere microbial communities. Different crops and rhizosphere microbial communities exhibit different heavy metal resistance mechanisms. Here, indoor pot studies were used to assess the mechanisms of grain and soil rhizosphere microbial communities on chromium (Cr) stress. Millet grain variety 'Jingu 21' (Setaria italica) and soil samples were collected prior to control (CK), 6 hours after (Cr_6h), and 6 days following (Cr_6d) Cr stress. Transcriptomic analysis, high-throughput sequencing and quantitative polymerase chain reaction (qPCR) were used for sample determination and data analysis. Cr stress inhibited the expression of genes related to cell division, and photosynthesis in grain plants while stimulating the expression of genes related to DNA replication and repair, in addition to plant defense systems resist Cr stress. In response to chromium stress, rhizosphere soil bacterial and fungal community compositions and diversity changed significantly (p < 0.05). Both bacterial and fungal co-occurrence networks primarily comprised positively correlated edges that would serve to increase community stability. However, bacterial community networks were larger than fungal community networks and were more tightly connected and less modular than fungal networks. The abundances of C/N functional genes exhibited increasing trends with increased Cr exposure. Overall, these results suggest that Cr stress primarily prevented cereal seedlings from completing photosynthesis, cell division, and proliferation while simultaneously triggering plant defense mechanisms to resist the toxic effects of Cr. Soil bacterial and fungal populations exhibited diverse response traits, community-assembly mechanisms, and increased expression of functional genes related to carbon and nitrogen cycling, all of which are likely related to microbial survival during Cr stress. This study provides new insights into resistance mechanisms, microbial community structures, and mechanisms of C/N functional genes responses in cereal plants to heavy metal contaminated agricultural soils. Abstract:Chromium (Cr) contamination in soil poses a serious security risk for the development of medicine and food with ginseng as the raw material. Microbiome are critical players in the functioning and service of soil ecosystems, but their feedback to Cr-contaminated ginseng growth is still poorly understood. To study this hypothesis, we evaluated the effects of microbiome and different Cr exposure on the soil microbial community using Illumina HiSeq high-throughput sequencing. Our results indicated that 2467 OTUs and 1785 OTUs were obtained in 16S and ITS1 based on 97% sequence similarity, respectively. Bacterial and fungal diversity were affected significantly in Cr-contaminated soil. Besides, Cr contamination significantly changed the composition of the soil bacterial and fungal communities, and some biomarkers were identified in the different classification level of the different Cr-contaminated treatments using LEfSe. Finally, a heatmap of Spearman’s rank correlation coefficients and canonical discriminant analysis (CDA) indicated that Chloroflexi, Gemmatimonadetes, Acidobacteria, Verrucomicobia, and Parcubacteria in phylum level and Acidimicrobiia, Gemmatimonadetes, and Deltaproteobacteria in class level were positively correlated with AK, AP, and NO3−-N (p < 0.05 or p < 0.01), but negatively correlated with total Cr and available Cr (p < 0.05 or p < 0.01). Similarly, in the fungal community, Tubaria, Mortierellaceae, and Rhizophagus in the phylum level and Glomeromycetes, Agaricomycetes, and Exobasidiomycetes in the class level were positively correlated with AK, AP, and NO3−-N (p < 0.05 or p < 0.01), but negatively correlated with total Cr and available Cr (p < 0.05 or p < 0.01). Our findings provide new insight into the effects of Cr contamination on the microbial communities in ginseng-growing soil.

Comparative Assessment of Toxic Metals Bioaccumulation and the Mechanisms of Chromium (Cr) Tolerance and Uptake in Calotropis procera

Abstract:Progressive pollution due to toxic metals significantly undermines global environmental sustainability efforts. Chromium (Cr) is one of the most dangerous to human health. The use of plants to rid the environment of such pollutants "phytoremediation" proves to be a promising alternative to the current remediation methods. In the present study, inductively coupled plasma optical emission spectroscopy (ICP-OES) determined Cadmium (Cd), Chromium (Cr), Copper (Cu), Nickel (Ni), and Lead (Pb) concentrations in the soil, and plants (Atriplex leucoclada, Calotropis procera, Salsola imbricata, Typha augustifolia, and Phragmites australis) root and shoots. Results showed that compared to other studied metals, Cr concentration was the highest in the soil at 111.8 mg/kg, whereas Cd records the least concentration of 0.04 mg/kg. Cr also accumulated in higher concentration in C. procera than in the soil and other plants, with up to 188.2 and 68.2 mg/kg concentration in the root and shoot, respectively. In order to understand the mechanism of Cr tolerance and uptake in C. procera, germinated seeds were irrigated with 20 mg/kg Cr and control treatment (no Cr applied) for six (6) weeks under greenhouse conditions. Fourier transformed infrared spectroscopy (FTIR) results showed high Cr complexation and binding to C. procera tissues via hydroxyl and carboxylic groups. Enzymatic assay reveals increased activities of superoxide dismutase (SOD), catalase (CAT), and glutathione reductase (GR) in Cr treated C. procera than in the control. SOD activity increased by up to six (6) folds. Therefore, we conclude that C. procera is suitable for the phytoremediation of Cr polluted arid soil. Additionally, regulation of cellular homeostasis via redox signaling is essential to the Cr tolerance and detoxification mechanism.

Abstract:ABSTRACT Soil microorganisms drive critical functions in plant-soil systems. As such, various microbial properties have been proposed as indicators of soil functioning, making them potentially useful in evaluating the recovery of polluted soils via phytoremediation strategies. To evaluate microbial responses to metal phytoextraction using hyperaccumulators, a microcosm experiment was carried out to study the impacts of Zn and/or Cd pollution and Thlaspi caerulescens growth on key soil microbial properties: basal respiration; substrate-induced respiration (SIR); bacterial community structure as assessed by PCR-denaturing gradient gel electrophoresis (DGGE); community sizes of total bacteria, ammonia-oxidizing bacteria, and chitin-degrading bacteria as assessed by quantitative PCR (Q-PCR); and functional gene distributions as determined by functional gene arrays (GeoChip). T. caerulescens proved to be suitable for Zn and Cd phytoextraction: shoots accumulated up to 8,211 and 1,763 mg kg−1 (dry weight [DW]) of Zn and Cd, respectively. In general, Zn pollution led to decreased levels of basal respiration and ammonia-oxidizing bacteria, while T. caerulescens growth increased the values of substrate-induced respiration (SIR) and total bacteria. In soils polluted with 1,000 mg Zn kg−1 and 250 mg Cd kg−1 (DW), soil bacterial community profiles and the distribution of microbial functional genes were most affected by the presence of metals. Metal-polluted and planted soils had the highest percentage of unique genes detected via the GeoChip (35%). It was possible to track microbial responses to planting with T. caerulescens and to gain insight into the effects of metal pollution on soilborne microbial communities.

Abstract:Extensive industrial activities have led to an increase of the content of chromium in the environment, which causes serious pollution to the surrounding water, soil and atmosphere. The enrichment of chromium in the environment through the food chain ultimately affects human health. Therefore, the remediation of chromium pollution is crucial to development of human society. A lot of scholars have paid attention to bioremediation technology owing to its environmentally friendly and low-cost. Previous reviews mostly involved pure culture of microorganisms and rarely discussed the optimization of bioreduction conditions. To make up for these shortcomings, we not only introduced in detail the conditions that affect microbial reduction but also innovatively introduced consortium which may be the cornerstone for future treatment of complex field environments. The aim of this study is to summary chromium toxicity, factors affecting microbial remediation, and methods for enhancing bioremediation. However, the actual application of bioremediation technology is still facing a major challenge. This study also put forward the current research problems and proposed future research directions, providing theoretical guidance and scientific basis for the application of bioremediation technology.

Abstract:In recent decades, environmental pollution with chromium (Cr) has gained significant attention. Although chromium (Cr) can exist in a variety of different oxidation states and is a polyvalent element, only trivalent chromium [Cr(III)] and hexavalent chromium [Cr(VI)] are found frequently in the natural environment. In the current review, we summarize the biogeochemical procedures that regulate Cr(VI) mobilization, accumulation, bioavailability, toxicity in soils, and probable risks to ecosystem are also highlighted. Plants growing in Cr(VI)-contaminated soils show reduced growth and development with lower agricultural production and quality. Furthermore, Cr(VI) exposure causes oxidative stress due to the production of free radicals which modifies plant morpho-physiological and biochemical processes at tissue and cellular levels. However, plants may develop extensive cellular and physiological defensive mechanisms in response to Cr(VI) toxicity to ensure their survival. To cope with Cr(VI) toxicity, plants either avoid absorbing Cr(VI) from the soil or turn on the detoxifying mechanism, which involves producing antioxidants (both enzymatic and non-enzymatic) for scavenging of reactive oxygen species (ROS). Moreover, this review also highlights recent knowledge of remediation approaches i.e., bioremediation/phytoremediation, or remediation by using microbes exogenous use of organic amendments (biochar, manure, and compost), and nano-remediation supplements, which significantly remediate Cr(VI)-contaminated soil/water and lessen possible health and environmental challenges. Future research needs and knowledge gaps are also covered. The review's observations should aid in the development of creative and useful methods for limiting Cr(VI) bioavailability, toxicity and sustainably managing Cr(VI)-polluted soils/water, by clear understanding of mechanistic basis of Cr(VI) toxicity, signaling pathways, and tolerance mechanisms; hence reducing its hazards to the environment.

Abstract:Experiments were conducted to determine the differential absorption of Cr species by tumbleweed (Salsola kali) as well as the effect of this heavy metal on plant growth and nutrient uptake. Tumbleweed seeds were grown in an agar-based media containing different concentrations of either Cr(III) or Cr(VI). The results demonstrated that the uptake of Cr was influenced by the Cr concentration in the growth medium and the speciation of this heavy metal. When supplied in the hexavalent form, the concentration of Cr in the different plant parts (2900, 790, and 600 mg kg(-1) for roots, stems, and leaves, respectively) was between 10 and 20 times higher than the amounts found when Cr was supplied in the trivalent form. In addition, it was found that in most of the experiments, Cr(III) exhibited more toxic effects on tumbleweed plants than Cr(VI). The size of roots of plants grown in 20 mg L(-1) Cr(III) were significantly smaller (p < 0.05) than those grown in 20 mg L(-1) Cr(VI). Plants exposed to 20 mg L(-1) Cr(III) produced shoots significantly shorter (p < 0.05) compared with the size of control plants and with those grown in 20 mg L(-1) Cr(VI). In addition, the absorption of macronutrients and microelements was in general lower when the plants were grown in the medium containing Cr(III). The amounts of Cr concentrated in the aerial plant parts under experimental conditions may indicate tumbleweed as a new option for the phytoremediation of Cr-contaminated soil.

Abstract:Time-resolved fluorescence decay of a single-stranded DNA decamer d(CTGAAT5CAG), where d5 is the fluorescent base 1-(beta-D-2'-deoxyribosyl)-5-methyl-2-pyrimidinone, was measured and analyzed at several temperatures. The d5 base in the decamer is resolved into three states according to their fluorescence decay lifetime characteristics and temperature dependence of their associated amplitudes: fully extended and completely unstacked state, loosely associated state, and fully stacked state. These states are in slow exchange compared to their fluorescence decay rates. The population of the fully extended and completely unstacked state is small and decreases further with increasing temperature. The loosely associated state, whose fluorescence can still be efficiently quenched by other DNA bases, occupies a large portion of the conventionally defined unstacked state. Stacking enthalpy and entropy for the d5 base with thymine or cytosine bases in the DNA decamer are calculated to be -6.6 kcal/mol and -22 cal/mol.K, respectively. This work shows that fluorescent bases in DNA can be useful to the study of local conformations of bases.
