Abstract:Diabetes mellitus during pregnancy, which can be classified into pregestational diabetes and gestational diabetes, has become much more prevalent worldwide. Maternal diabetes fosters an intrauterine abnormal environment for fetus, which not only influences pregnancy outcomes, but also leads to fetal anomaly and development of diseases in later life, such as metabolic and cardiovascular diseases, neuropsychiatric outcomes, reproduction malformation, and immune dysfunction. The underlying mechanisms are comprehensive and ambiguous, which mainly focus on microbiota, inflammation, reactive oxygen species, cell viability, and epigenetics. This review concluded with the influence of intrauterine hyperglycemia on fetal structure development and organ function on later life and outlined potential mechanisms that underpin the development of diseases in adulthood. Maternal diabetes leaves an effect that continues generations after generations through gametes, thus more attention should be paid to the prevention and treatment of diabetes to rescue the pathological attacks of maternal diabetes from the offspring.
SIRT3 alleviates high glucose-induced chondrocyte injury through the promotion of autophagy and suppression of apoptosis in osteoarthritis progression

Abstract:A growing amount of epidemiological evidence proposes diabetes mellitus (DM) to be an independent risk factor for osteoarthritis (OA). Sirtuin 3 (SIRT3), which is mainly located in mitochondria, belongs to the family of nicotinamide adenine dinucleotide (NAD + )-dependent protein deacetylases and is involved in the physiological and pathological processes of cell regulation. The aim of this study was to investigate the effects of SIRT3 on diabetic OA and underlying mechanisms in the prevention of type 2 DM (T2DM)-induced articular cartilage damage. High-fat and high-sugar diets combined with streptozotocin (STZ) injection were used for establishing an experimental T2DM rat model. The destabilization of medial meniscus (DMM) surgery was applied to induce the rat OA model. Primary rat chondrocytes were cultivated with a concentration of gradient glucose. Treatment with intra-articular injection of SIRT3 overexpression lentivirus was achieved in vivo, and intervention with SIRT3 knockdown was performed using siRNA transfection in vitro. High glucose content was found to activate inflammatory response, facilitate apoptosis, downregulate autophagy, and exacerbate mitochondrial dysfunction in a dose-dependent manner in rat chondrocytes, which can be deteriorated by SIRT3 knockdown. In addition, articular cartilage damage was found to be more severe in T2DM-OA rats than in DMM-induced OA rats, which can be mitigated by the intra-articular injection of SIRT3 overexpression lentivirus. Targeting SIRT3 is a potential therapeutic strategy for the alleviation of diabetic OA.

Intrauterine hyperglycaemia during late gestation caused mitochondrial dysfunction in skeletal muscle of male offspring through CREB/PGC1A signaling

Abstract:Background: Maternal diabetes mellitus can influence the development of offspring. Gestational diabetes mellitus (GDM) creates a short-term intrauterine hyperglycaemic environment in offspring, leading to glucose intolerance in later life, but the long-term effects and specific mechanism involved in skeletal muscle dysfunction in offspring remain to be clarified. Methods: Pregnant mice were divided into two groups: The GDM group was intraperitoneally injected with 100 mg/kg streptozotocin on gestational days (GDs) 6.5 and 12.5, while the control (CTR) group was treated with vehicle buffer. Only pregnant mice whose random blood glucose level was higher than 16.8 mmol/L beginning on GD13.5 were regarded as the GDM group. The growth of the offspring was monitored, and the glucose tolerance test was performed at different time points. Body composition analysis and immunohistochemical methods were used to evaluate the development of lean mass at 8 weeks. The exercise capacity and grip strength of the male mouse offspring were assessed at the same period. Transmission electron microscopy was used to observe the morphology inside skeletal muscle at 8 weeks and as a foetus. The genes and proteins associated with mitochondrial biogenesis and oxidative metabolism were investigated. We also coanalyzed RNA sequencing and proteomics data to explore the underlying mechanism. Chromatin immunoprecipitation and bisulfite-converted DNA methylation detection were performed to evaluate this phenomenon. Results: Short-term intrauterine hyperglycaemia inhibited the growth and reduced the lean mass of male offspring, leading to decreased endurance exercise capacity. The myofiber composition of the tibialis anterior muscle of GDM male offspring became more glycolytic and less oxidative. The morphology and function of mitochondria in the skeletal muscle of GDM male offspring were destroyed, and coanalysis of RNA sequencing and proteomics of foetal skeletal muscle showed that mitochondrial elements and lipid oxidation were consistently impaired. In vivo and in vitro myoblast experiments also demonstrated that high glucose concentrations impeded mitochondrial organisation and function. Importantly, the transcription of genes associated with mitochondrial biogenesis and oxidative metabolism decreased at 8 weeks and during the foetal period. We predicted Ppargc1α as a key upstream regulator with the help of IPA software. The proteins and mRNA levels of Ppargc1α in the skeletal muscle of GDM male offspring were decreased as a foetus (CTR vs. GDM, 1.004 vs. 0.665, p = 0.002), at 6 weeks (1.018 vs. 0.511, p = 0.023) and 8 weeks (1.006 vs. 0.596, p = 0.018). In addition, CREB phosphorylation was inhibited in GDM group, with fewer activated pCREB proteins binding to the CRE element of Ppargc1α (1.042 vs. 0.681, p = 0.037), Pck1 (1.091 vs. 0.432, p = 0.014) and G6pc (1.118 vs. 0.472, p = 0.027), resulting in their decreased transcription. Interestingly, we found that sarcopenia and mitochondrial dysfunction could even be inherited by the next generation. Conclusions: Short-term intrauterine hyperglycaemia significantly reduced lean mass in male offspring at 8 weeks, resulting in decreased exercise endurance and metabolic disorders. Disrupted organisation and function of the mitochondria in skeletal muscle were also observed among them. Foetal exposure to hyperglycaemia decreased the ratio of phosphorylated CREB and reduced the transcription of Ppargc1α, which inhibited the transcription of downstream genes involving in mitochondrial biogenesis and oxidative metabolism. Abnormal mitochondria, which might be transmitted through aberrant gametes, were also observed in the F2 generation.
Intrauterine hyperglycemia impairs endometrial receptivity via up-regulating SGK1 in diabetes

Abstract:Diabetes is a complex metabolic disorder which can adversely affect reproductive function. SGK1 is found to be up-regulated in multiple tissues of diabetic patients. However, the effects of diabetes on endometrial SGK1 expression and endometrial receptivity remain unknown. In this study, we established a streptozotocin-induced diabetic mouse model and observed reduced implantation sites, retarded development of pinopodes, increased SGK1, and aberrant expression of LIF and MUC1 in the endometrial epithelium. We injected the uterine lumen of normal mice with high-glucose solution and cultured endometrial cells in high-glucose medium to mimic intrauterine hyperglycemia. Both studies provided compelling evidence that hyperglycemia could lead to diminished embryo implantation and dysregulated SGK1, LIF and MUC1. Additionally, through over-expression of SGK1 in vivo and in vitro , we found that enhanced SGK1 also decreased LIF expression, increased MUC1 expression, and attenuated embryo implantation rate. We further identified that hyperglycemia-activated SMAD2/3 might be responsible for the enhancement of SGK1 and verified directly the interaction between SMAD3 and corresponding SMAD binding elements within SGK1 promoter. Taken together, our study confirmed the association between diabetes-related hyperglycemia and endometrial receptivity defects. Hyperglycemia-induced SGK1 has a tremendous role in this pathological process, rendering it as an attractive therapeutic target for diabetes-related reproductive disorders.
Intrauterine Hyperglycemia Induces Hepatic Steatosis in Mouse Offspring Via Altered Lipid Gene Expressions

Abstract:We aimed to determine the effects of antenatal exposure to intrauterine hyperglycemia on offspring's lipid metabolism and the potential mechanism. We examined lipid metabolism of male offspring of gestational diabetes mellitus (GDM) and detected relevant genes expressions to explore the underlying mechanism. Pregnant ICR mice were injected with a single injection of streptozotocin to establish a GDM model. We examined birth weight, 3-week and 8-week body weight (BW), liver weight and fat weight of the offspring of control (F1-C) and GDM (F1-GDM). Further, intraperitoneal glucose tolerance testing (IPGTT) was performed at the age of 3 and 8 weeks. Hepatic content of triglyceride (TG) and total cholesterol (TC) were measured as well as serum concentrations of lipids. Frozen liver samples were stained with Oil-Red-O and hematoxylin. Quantitative real-time PCR were performed to detect genes expressions associated with lipid metabolism. For each group, n>8. Statistical analysis was performed by unpaired two-tailed Student t test (version 22.0; SPSS). P values <0.05 were considered statistically significant. We found birth weight of F1-GDM offspring was significant lower than F1-C (p<0.001). After fostered by normoglycemic mice, there were no differences in BW, liver weight and visceral fat weight at 8-week age. The IPGTT was normal at 3 weeks, whereas at 8-week age, impaired glucose tolerance (IGT) was found in F1-GDM group (p<0.05). In F1-GDM group, the hepatic TG and TC levels were significantly increased (p<0.05, p<0.05). We found that the serum TG level in the F1-GDM group was higher than that in the control group, while F1-GDM group had lower HDL levels than F1-C (p<0.01, p<0.001, respectively). There were no differences in serum TC, LDL, NEFA, ALT and AST between the two groups. An increased level of Oil-Red-O staining with larger and more numerous lipid droplets in liver tissues was detected in F1-GDM group at the age of 8 weeks. Hepatic genes involved in fatty acid synthesis and cholesterol synthesis were up-regulated, whereas fatty acid oxidation and cholesterol transportation were down-regulated. Our results suggest that intrauterine hyperglycemia exposure contributes to early onset of hepatic steatosis associated with changes in hepatic gene expression and lipid handling.
Intrauterine Hyperglycemia Exposure Results in Intergenerational Inheritance Via DNA Methylation Reprogramming on F1 PGCs

Abstract:Background The existing reports about intergenerational or transgenerational effects of intrauterine hyperglycemia have included both intrauterine and postnatal metabolic exposure factors, while the impact of intrauterine hyperglycemia per se has not been assessed alone. A number of studies suggest DNA methylation reprogramming of gametes plays a crucial role in the metabolic inheritance, but it is unclear when and how DNA methylation patterns are altered when exposed to intrauterine hyperglycemia. In this study, we selected nondiabetic F1- and F2-gestational diabetes mellitus (GDM) male mice as founders to examine metabolic changes in the next generation and performed methylome sequencing of day 13.5 primordial germ cells (PGCs) from F1-GDM to explore the underlying epigenetic mechanism. Results We found that intrauterine hyperglycemia exposure resulted in obesity, insulin resistance, and/or glucose intolerance in F2 male mice, but no metabolic changes in F3 male mice at 8 weeks. Using reduced representation bisulfite sequencing, we found DNA methylome of day 13.5 PGCs from F1-GDM fetuses revealed differently methylated genes enriched in obesity and diabetes. Methylation validation of the insulin resistance and fat accumulation gene Fyn showed a consistent hypomethylation status in F1 PGCs, F1 fetal testes, sperm from F1/C-GDM mice, and somatic cells from F2-GDM male mice. In contrast, no methylation alteration was observed in F2-GDM male germ cells and F3-GDM somatic cells. Conclusion We provide evidence that intrauterine hyperglycemia exposure per se contributes to intergenerational metabolic changes in the F2 but not F3 generation. And the aberrant DNA methylation reprogramming occurs as early as day 13.5 in PGCs of the F1 generation. Our findings suggest that intrauterine exposure alone is sufficient to cause the epigenetic inheritance in F2 offspring, and the epigenetic memory carried by DNA methylation pattern could be erased by the second wave of methylation reprogramming in F2 PGCs during fetal development.

Abstract:Brown adipose tissue (BAT) is an exclusive tissue of nonshivering thermogenesis. It is fueled by lipids and glucose and involved in energy and metabolic homeostasis. Intrauterine exposure to hyperglycemia during gestational diabetes mellitus may result in abnormal fetal development and metabolic phenotypes in adulthood. However, whether intrauterine hyperglycemia influences the development of BAT is unknown. In this study, mouse embryos were exposed to the intrauterine hyperglycemia environment by injecting streptozocin into pregnant mice at 1 d post coitum (dpc). The structure of BAT was examined by hematoxylin and eosin staining and immunohistochemical analysis. The glucose uptake in BAT was measured in vivo by [18F]-fluoro-2-deoxyglucose-micro-positron emission tomography. The gene expression in BAT was determined by real-time PCR, and the 5'-C-phosphate-G-3' site-specific methylation was quantitatively analyzed. Intrauterine hyperglycemia exposure resulted in the impaired structure of BAT and decreased glucose uptake function in BAT in adulthood. The expressions of the genes involved in thermogenesis and mitochondrial respiratory chain in BAT, such as Ucp1, Cox5b, and Elovl3, were down-regulated by intrauterine hyperglycemia exposure at 18.5 dpc and at 16 wk of age. Furthermore, higher methylation levels of Ucp1, Cox5b, and Elovl3 were found in offspring of mothers with streptozotocin-induced diabetes. Our results provide the evidence for enduring inhibitory effects of intrauterine hyperglycemia on BAT development in offspring. Abstract:AIM:Gestational diabetes mellitus (GDM) has been shown to be associated with a high risk of diabetes in offspring. In mitochondria, the inhibition of pyruvate dehydrogenase (PDH) activity by PDH phosphorylation is involved in the development of diabetes. We aimed to determine the role of PDH phosphorylation in the liver in GDM-induced offspring glucose intolerance. RESULTS:PDH phosphorylation was increased in lymphocytes from the umbilical cord blood of the GDM patients and in high glucose-treated hepatic cells. Both the male and female offspring from GDM mice had elevated liver weights and glucose intolerance. Further, PDH phosphorylation was increased in the livers of both the male and female offspring from GDM mice, and elevated acetylation may have contributed to this increased phosphorylation. MATERIALS AND METHODS:We obtained lymphocytes from umbilical cord blood collected from both normal and GDM pregnant women. In addition, we obtained the offspring of streptozotocin-induced GDM female pregnant mice. The glucose tolerance test was performed to assess glucose tolerance in the offspring. Further, Western blotting was conducted to detect changes in protein levels. CONCLUSIONS:Intrauterine hyperglycemia induced offspring glucose intolerance by inhibiting PDH activity, along with increased PDH phosphorylation in the liver, and this effect might be mediated by enhanced mitochondrial protein acetylation.

Abstract:Osteoarthritis is the most common degenerative joint disease and causes major pain and disability in adults. It has been reported that mitochondrial dysfunction in chondrocytes is associated with osteoarthritis. Sirtuins are a family of nicotinamide adenine dinucleotide-dependent histone deacetylases that have the ability to deacetylate protein targets and play an important role in the regulation of cell physiological and pathological processes. Among sirtuin family members, sirtuin 3, which is mainly located in mitochondria, can exert its deacetylation activity to regulate mitochondrial function, regeneration, and dynamics; these processes are presently recognized to maintain redox homeostasis to prevent oxidative stress in cell metabolism. In this review, we provide present opinions on the effect of mitochondrial dysfunction in osteoarthritis. Furthermore, the potential protective mechanism of SIRT3-mediated mitochondrial homeostasis in the progression of osteoarthritis is discussed.

Abstract:Intrauterine hyperglycemia of gestational diabetes mellitus (GDM) leads to long-term metabolic dysfunction in generations. Previous studies include both intrauterine exposure and postnatal adult diabetic conditions as influence factors. The aim of our study is to assess the impact of intrauterine hyperglycemia per se on F2 generation's metabolism via germ cells. Animal model study. A streptozotocin-induced GDM mouse model was established. Only male individuals were studied. Healthy F1-GDM male mice were crossed with normal female to produce F2 generation. Body weight, glucose tolerance test (GTT), insulin tolerance test (ITT), serum fasting insulin and lipids parameters were determined at 8 week in both generations, homeostasis model of insulin resistance (HOMA-IR) index was then calculated. Reduced Representation Bisulfite Sequencing (RRBS) was performed on primordial germ cells (PGCs) of E13.5 male F1-GDM and control fetus. Ingenuity Pathway Analysis (IPA) was used to identify the top related physiological pathways. Comparisons between two groups were performed using Student t-tests. One-way ANOVA was used to compare differences among multiple groups. For each group, n>10. A P<0.05 was considered statistically significant. (1) F1-GDM male mice which showed no significant difference in body weight, GTT, ITT, HOMA-IR index and serum parameters with control mice (P>0.05), were chosen as healthy founders to produce F2 generation. (2) Some of F2-GDM offspring (28/41) exhibited glucose intolerance (P<0.001). (3) All the F2-GDM offspring exhibited higher ITT curve than control (P<0.001 of normal glucose tolerance individuals, P<0.05 of glucose intolerance individuals, vs. control). All the F2-GDM offspring exhibited higher HOMA-IR index (P<0.01 of normal glucose tolerance individuals vs. control, P<0.05 of glucose intolerance individuals vs. control). (4) F2-GDM offspring had higher body weight than control mice (P<0.001 of normal glucose tolerance individuals, P<0.001 of glucose intolerance individuals, vs. control). (5) Glucose intolerance F2 mice showed higher triacylglycerol and non-esterified fatty acid than control (P<0.05). (6) RRBS revealed 1842 differentially methylated genes. These genes were mainly involved in cell cycle, HGF signaling, ERK/MAPK signaling, insulin receptor signaling and PI3K/AKT signaling pathways. Intrauterine hyperglycemia exposure per se contributes to glycolipid metabolism dysfunction in F2 generation. The aberrant methylation erasure in PGCs may provide one possible epigenetic mechanism underlies the transmission effect.

Abstract:Objective:To explore the role of AKT-mTOR signaling pathway and SIRT1 in fetal growth and development programming of gestational diabetes mellitus(GDM).Methods:Fifteen cases of GDM and 15 cases of normal maternal placental tissue were collected in the First Affiliated Hospital of Chongqing Medical University from December 2014 to June 2015.Western blot was used to detect the expression of AKT-mTOR and SIRT1 in human placenta.Human transformed primary extravillous trophoblast cell line (HTR8/ SVneo) cells were used to establish the model of GDM,which were divided into normal group,osmotic control,and high glucose group.After the treatment,the expressions of AKT-mTOR and SIRT1 were detected by Western blot;Flow cytometry was used to detect the apoptosis rate in each group.On day 18.5 of gestation,db/+ mice were anesthetized with pentobarbitone and a caesarean section was performed.The placentas were excised and frozen immediately for genotyping,and then the wild type placentas were selected for GDM group.C57 mice placentas were control group(normal).Then the expressions of AKT-mTOR were detected by Western blot.Results:The phosphorylation of AKT and mTOR in placenta of GDM group was significantly lower than that of normal group (0.347 ± 0.031 vs.1.000 ± 0.175,P=0.004;0.465 ± 0.045 vs.1.000 ± 0.098,P=0.000).Similarly,the expression of SIRT1 was obviously decreased in GDM group than in normal group (0.682 ± 0.055 vs.1.000 ± 0.127,P=0.044).In the cell model,high glucose could decrease AKT and mTOR phosphorylation (0.512 ± 0.056 vs.1.103 ± 0.111,P=0.023;0.262 ± 0.091 vs.1.153 ± 0.057,P=0.001) and SIRT1 expression(0.472 ± 0.034 vs.1.013 ± 0.098,P=0.040).The apoptosis rate in high glucose group was significantly increased than that in norm al group (14.550 ± 1.624 vs.9.547 ± 0.685,P=0.032).In the animal model,the phosphorylation of AKT and mTOR also was significantly decreased in GDM group (0.527 ± 0.080 vs.1.000 ± 0.055,P=0.003;0.418 ± 0.059 vs.1.000 ± 0.084,P=0.001).Conclusion:The intrauterine environment of high glucose may inhibit placental AKT-mTOR signaling pathways,thereby programming offspring development track.

Abstract:BACKGROUND:Excessive androgen exposure during pregnancy has been suggested to induce diabetic phenotypes in offspring in animal models. The aim of this study was to investigate whether pregestational maternal hyperandrogenism in human influenced the glucose metabolism in offspring via epigenetic memory from mother's oocyte to child's somatic cells.METHODS:Of 1782 reproductive-aged women detected pregestational serum androgen, 1406 were pregnant between 2005 and 2010. Of 1198 women who delivered, 1116 eligible mothers (147 with hyperandrogenism and 969 normal) were recruited. 1216 children (156 children born to mothers with hyperandrogenism and 1060 born to normal mother) were followed up their glycometabolism in mean age of 5years. Imprinting genes of oocyte from mothers and lymphocytes from children were examined. A pregestational hyperandrogenism rat model was also established.FINDINGS:Children born to women with hyperandrogenism showed increased serum fasting glucose and insulin levels, and were more prone to prediabetes (adjusted RR: 3.98 (95%CI 1.16-13.58)). Oocytes from women with hyperandrogenism showed increased insulin-like growth factor 2 (IGF2) expression. Lymphocytes from their children also showed increased IGF2 expression and decreased IGF2 methylation. Treatment of human oocytes with dihydrotestosterone upregulated IGF2 and downregulated DNMT3a levels. In rat, pregestational hyperandrogenism induced diabetic phenotypes and impaired insulin secretion in offspring. In consistent with the findings in human, hyperandrogenism also increased Igf2 expression and decreased DNMT3a in rat oocytes. Importantly, the same altered methylation signatures of Igf2 were identified in the offspring pancreatic islets.INTERPRETATION:Pregestational hyperandrogenism may predispose offspring to glucose metabolism disorder via epigenetic oocyte inheritance. Abstract:Gestational diabetes mellitus (GDM) is associated with an increased risk of metabolic disorders in offspring in later life. Although mounting evidence suggests that therapy for GDM could improve neonatal health, whether the therapy confers long-term metabolic benefits to offspring in their later adult lives is not known. Here, using a mouse model of diabetes in the latter half of pregnancy to mimic human GDM, we find that the efficient insulin therapy for GDM confers significant protection against glucose intolerance and obesity in offspring fed a normal chow diet. However, the therapy fails to protect offspring when challenged with a high-fat diet, especially for male offspring. Genome-wide DNA methylation profiling of pancreatic islets from male offspring identified hypermethylated regions in several genes that regulate insulin secretion, including Abcc8, Cav1.2, and Cav2.3 that encode K-ATP or Ca2+ channels, which are associated with reduced gene expression and impaired insulin secretion. This finding suggests a methylation-mediated epigenetic mechanism for GDM-induced intergenerational glucose intolerance. It highlights that even efficient insulin therapy for GDM is insufficient to fully protect adult offspring from diet-induced metabolic disorders.

Abstract:Diabetes mellitus is prevalent among women of reproductive age, and many women are left undiagnosed or untreated 1 . Gestational diabetes has profound and enduring effects on the long-term health of the offspring 2 , 3 . However, the link between pregestational diabetes and disease risk into adulthood in the next generation has not been sufficiently investigated. Here we show that pregestational hyperglycaemia renders the offspring more vulnerable to glucose intolerance. The expression of TET3 dioxygenase, responsible for 5-methylcytosine oxidation and DNA demethylation in the zygote 4 , is reduced in oocytes from a mouse model of hyperglycaemia (HG mice) and humans with diabetes. Insufficient demethylation by oocyte TET3 contributes to hypermethylation at the paternal alleles of several insulin secretion genes, including the glucokinase gene ( Gck ), that persists from zygote to adult, promoting impaired glucose homeostasis largely owing to the defect in glucose-stimulated insulin secretion. Consistent with these findings, mouse progenies derived from the oocytes of maternal heterozygous and homozygous Tet3 deletion display glucose intolerance and epigenetic abnormalities similar to those from the oocytes of HG mice. Moreover, the expression of exogenous Tet3 mRNA in oocytes from HG mice ameliorates the maternal effect in offspring. Thus, our observations suggest an environment-sensitive window in oocyte development that confers predisposition to glucose intolerance in the next generation through TET3 insufficiency rather than through a direct perturbation of the oocyte epigenome. This finding suggests a potential benefit of pre-conception interventions in mothers to protect the health of offspring.
Intergenerational Neurocognitive Effects of Intrauterine Hyperglycemia on Male Offspring

Abstract:Background Studies on human and animals suggest associations between gestational diabetes mellitus (GDM) with increased susceptibility to develop neurological disorders in offspring. However, researches have focused on the neurodevelopment consequences of the first filial (F1) offspring. We hypothesize that the intrauterine hyperglycemia exposure will alter epigenetic reprogramming in F1 sperm, and carry risks of passing on molecular defects to the second filial (F2). Results We found that intrauterine hyperglycemia exposure resulted in memory impairment in both F1 and F2 males from the F1-GDM male offspring. Transcriptome profiling of F1 and F2 hippocampi revealed that differentially expressed genes were enriched in learning, memory, cognition, neurotransmission, synaptic plasticity, and postsynaptic specialization. Again, enrichment curves computed by Gene set enrichment analysis (GSEA) of F1 hippocampi were highly consistent with F2. Interestingly, combined analysis of F1 sperm methylome and gene expression of F2 hippocampi screened out several hypermethylation-low expression genes which are associated with abnormal central nervous system development (particularly synapse development and homeostasis), such as Camk2b, Dlgap1, Wnt5a, Tubb2b and so on. Conclusions These findings implicate that the male germ line is a major player in transgenerational phenotypic transmission. Taken together, our results for the first time suggest that intrauterine hyperglycemia leads to transgenerational cognitive impairment, and, sperm methylome is a potential epigenetic mechanism for the effects of GDM.

Abstract:Studies on humans and animals suggest associations between gestational diabetes mellitus (GDM) with increased susceptibility to develop neurological disorders in offspring. However, the molecular mechanisms underpinning the intergenerational effects remain unclear. Using a mouse model of diabetes during pregnancy, we found that intrauterine hyperglycemia exposure resulted in memory impairment in both the first filial (F1) males and the second filial (F2) males from the F1 male offspring. Transcriptome profiling of F1 and F2 hippocampi revealed that differentially expressed genes (DEGs) were enriched in neurodevelopment and synaptic plasticity. The reduced representation bisulfite sequencing (RRBS) of sperm in F1 adult males showed that the intrauterine hyperglycemia exposure caused altered methylated modification of F1 sperm, which is a potential epigenetic mechanism for the intergenerational neurocognitive effects of GDM.

Abstract:OBJECTIVES:The Developmental Origins of Health and Disease Science indicate that chronic diseases in adulthood are associated with prenatal and early-life traits. Our study aimed to explore the metabolic phenotype of offspring from advanced paternal age (APA) and the inherited alterations in sperm. METHODS:3-month-old (Young father, YF-F0) and 21-month-old male (Old Father, OF-F0) C57BL/6J mice were used to study paternal aging's effect on offspring. Blood glucose testing, lipid analysis, indirect calorimetry and RNA sequencing were performed. RESULTS:The characterized metabolic changes in OF-F1 male mice offspring were glucose intolerance, hepatic lipid accumulation, increased adipocytes and impaired energy balance that lasted until they were elderly. Gene expression in both 8-week-old and 52-week-old offspring livers significantly altered in lipid metabolism- and thermogenesis-related pathways. PPAR signaling pathway was activated in both young and elderly offspring livers as indicated by significant upregulation of Cyp7a1, Cyp8b1, Cyp4a10, Cyp4a31, Fabp2, and Scd1. These targeted genes were also confirmed to be increased in offspring adipocytes. Furthermore, when examined the differentially expressed genes in F0 and F1 sperm, upregulated pathways including cholesterol metabolism, type II diabetes mellitus and endocrine resistance were strongly related to the APA offspring phenotype. Importantly, approximately 46.7% of enriched pathways in the sperm of APA offspring were consistent with those of APA fathers. CONCLUSIONS:These findings added evidence of the connection between paternal gametes and alterations in progeny genome and raised the possibility that inherited alterations in sperm contribute to the intergenerational effects of paternal aging offspring's chronic metabolic risks.

Abstract:1. (Z)-1,3-Dichloro[2-14C]propene([14C](Z)-DCP) when dosed orally to rats gave 82-84% of the radioactivity in the urine 24 h after treatment. Most of this 14C (92%) was present as N-acetyl-S-((Z)-3-chloroprop-2-enyl)cysteine ((Z)-DCP mercapturic acid).2. When (Z)-DCP was incubated with glutathione and rat liver cytosol (containing glutathione S-alkyl transferase), very rapid loss of (Z)-DCP was observed. The product of this reaction was S-[(Z)-3-chloroprop-2-enyl)]glutathione.

Abstract:The sustained global rise in the prevalence of diabetes mellitus over the last decades increasingly affects young adults. Thus, 7%–20% of pregnancies are complicated by maternal diabetes.1 ‘Diabetic mothers give birth to diabetic babies’ is one oversimplified conclusion from human and animal studies demonstrating that maternal hyperglycaemia predisposes the offspring to the development of diabetes.2 However, little is known about the molecular mechanisms underlying this phenomenon known as ‘metabolic memory’. A recent article published in Nature reveals that glucose intolerance – a sign of prediabetic status – is transmitted from a diabetic mother to the next generation via the female oocytes. Mounting evidence has shown that maternal diabetes during the gestational period can make offspring more likely to increase their risk of diabetes.3 To distinguish the effect of pre-gestation and lactation, the authors fertilized the oocytes from STZ-induced hyperglycaemia female mice in vitro with healthy male sperm and transferred the 2-cell embryo to normal glycaemia foster female mice. The approach of in vitro fertilization and embryo transfer enables the researchers only to focus on the effect of oocytes on metabolic memory transmission. The mice generated from hyperglycaemic oocytes had normal growth trajectories but exhibited impaired glucose tolerance compared with controls. The authors showed that glucose intolerance in offspring was attributed to impaired glucose-stimulated insulin secretion by pancreatic islets beta cells. This glucose intolerance was more pronounced in male than female offspring, which worsened with age and high-fat diet in both sexes, suggesting that F1 generation inherited the diabetes susceptibility from the hyperglycaemia-exposed oocytes. Interestingly, this metabolic phenotype was detected only in F1 offspring, not persisted in F2 generation (at least without metabolic phenotype till 24-week old), indicating that maternal hyperglycaemia had an intergenerational rather than a transgenerational effect on offspring health via oocytes. Given that when sperm exposed to a prediabetic status, the transgenerational metabolic inheritance was observed in F2 offspring via paternal germline, indicating that the maternal germline-dependent diabetes inheritance differs from paternal germline.4 One general mechanism by which early life exposures could be linked to acquired traits is the alteration of epigenetic marks. In mammals, efficient genome-wide DNA methylation reprogramming occurs in the pre-implantation embryo and the germline, and these two rounds of epigenetic erasure are required to remove the acquired epigenetic signatures to ensure that the embryo properly reflects the genetic blueprint characteristic of each species.5 Epigenetic reprogramming is two sides of the same coin. Reprogramming events act as natural barriers aimed precisely at maintaining the pluripotency of the zygote and ensuring proper embryo development.6 However, each reprogramming window shows active epigenetic regulators and unstable epigenetic markers,7 which processes are susceptible to any environmental factors and can be transmitted to the following generation. Thus, one possibility is that maternal hyperglycaemia might disturb the epigenome during post-fertilization, leading to acquired traits of glucose intolerance in offspring. To test the hypothesis, researchers first compared the mRNA transcriptome of oocytes from the hyperglycaemia mice and controls. Among all the differentially expressed genes, Tet3 transcripts were detected significantly downregulated in hyperglycaemia oocytes. Tet3 expression was also decreased in oocytes from another diabetic mouse model and diabetic women. The mammalian TET family includes three members, TET1, TET2, and TET3, which can catalyse the conversion of 5-position of cytosine (5mC) to 5-hydroxymethylcytosine (5hmC).8 TET3 is abundantly expressed in mature oocytes and acts as a maternal factor critical for paternal DNA demethylation.9 Investigators found that the oxidation of methylated DNA level was markedly lower in the paternal pronuclei of zygotes from hyperglycaemic mice. To evaluate the effects of reduced maternal TET3 and paternal genome 5hmC on offspring glucose metabolism, the authors carried out whole-genome bisulfite sequencing on offspring E18.5 pancreas. Numerous regions were hypermethylated in foetal pancreas generated from hyperglycaemia oocytes, and differentially hypermethylated regions (hyper-DMRs) in the promoters of several genes involved in insulin secretion, including a pancreas-specific promoter that drives pancreatic transcription of Gck. Gck encodes glucokinase, a key rate-limiting enzyme that regulates glucose-stimulated insulin secretion.10 The authors went on to show the hyper-DMRs of Gck derived from the paternal genome and persisted in a developmental stage from pre-implantation embryo to postnatal adulthood. However, it is difficult to exclude the possibility that TET3 is also involved in the maternal demethylation of metabolic genes. To explore whether reduced maternal Tet3 expression was the only contributor to Gck hypermethylation and glucose intolerance in offspring, they generated a Tet3 knockout mouse, in which Tet3 expression was insufficient due to oocytes-specific Tet3 heterozygous and homozygous knockout, mimicked the effect of maternal hyperglycaemia. Offspring from genetically engineered Tet3+/− and Tet3−/− oocytes exhibited altered DNA methylation status of Gck and glucose intolerance, which was similar to those from hyperglycaemia oocytes. Additionally, a supplemental injection of exogenous Tet3 mRNA to mature oocytes abolished the effects of hyperglycaemia or Tet3 deletion. Previous results showed that paternal germline was susceptible to metabolic challenges,4 and this study provided evidence that female germline was also vulnerable to environmental factors when they are maturing. High glucose exposure led to a significant reduction in Tet3 mRNA levels in mice and human oocytes during in vitro maturation, due to accelerating Tet3 mRNA degeneration. This finding in germline cells coincided with somatic cells exposed to hyperglycaemia,11 reflecting that high glucose possibly causes Tet3 destabilization. It will be exciting to explore the mechanisms by which molecular events control the Tet3 mRNA or protein stability. Taken together, this study revealed maternal and paternal co-contribution to oocytes origin of diabetic susceptibility. Hyperglycaemia directly compromised maternal oocytes’ Tet3 levels and further indirectly defected paternal DNA methylation reprogramming via TET3 insufficiency, finally causing the perturbations of insulin secretion gene expression and glucose intolerance in next generation (Figure 1). Future research is required to determine whether the TET3-mediated inheritance is relevant to humans and, if so, the mechanism and molecular targets by which high glucose regulates TET3 stability and expression will hopefully revolutionize preventative and therapeutic approaches for diabetes management.
