Abstract:Stochastic resonance (SR) effect has been discovered in non-dynamical threshold systems such as sensory systems. This paper presents a network simulating basic structure of a sensory system to study SR. The neuronal network consists of two layers of the Hodgkin-Huxley (HH) neurons. Compared with single HH model, subthreshold stimulating signals do not modulate output signal-noise ratio, thus a fixed level of noise from circumstance can induce SR for the various stimulating signals. Numeric experimental results also show that noises do not always deteriorate the capability of the detection of suprathreshold input signals.

Abstract:Stochastic resonance is a phenomenon, wherein the response of a nonlinear system to a weak periodic signal is enhanced at a nonzero level of noise. In order to study the ability of the network in weak signal detection, Hodgkin-Huxley neuronal model was adopted for constructing a double layer neuronal network. Stochastic resonance in a single neuron was characterized with signal-to-noise ratio for showing the positive role of noise in subthreshold signal detection. But the optimal level of noise for stochastic resonance in a single neuron must be adjusted as the nature of the external signal changes. This has been thought to impose a limitation on the use of stochastic resonance in weak signal detection. The double layer neuronal network was investigated and the characteristics of stochastic resonance was observed. Results indicate that this network has the ability of detecting the subthreshold signal varying in a range of amplitude at a fixed noise level and the noise does not degrade the ability of the network to detect suprathreshold signals. Thus, the double layer neuronal network does not suffer from the limitation of stochastic resonance in a single neuron. This suggests a positive role of noise in sensory systems.

Abstract:We systematically investigate the stochastic dynamics of a single Hodgkin–Huxley neuron driven by stochastic excitatory and inhibitory input spikes via unreliable synapses in this paper. Based on the mean-filed theory, a novel intrinsic neuronal noise regulation mechanism stemming from unreliable synapses is presented. Our simulation results show that, under certain conditions, the stochastic resonance phenomenon is able to be induced by the unreliable synaptic transmission, which can be well explained by the theoretical prediction. To a certain degree, the results presented here provide insights into the functional roles of unreliable synapses in neural information processing.

Abstract:Noise is widespread at all levels of the nervous system, affecting the generation, transmission, and response of neural signals. However, significant challenges remain in effectively separating signal and noise experimentally; at the same time, a growing number of modeling studies suggest that noise is not always harmful and exhibits many positive effects on the nervous system. This paper first reviews the main sources and properties of noise in the nervous system, as well as the physiological basis for modulating factors such as temperature, channel blocking, time delay, and electromagnetic effects that are often considered in conjunction with noise. Secondly, based on different neural models, various nonlinear effects induced by noise at different levels, such as neurons and neural networks, are explored. Nonlinear physical phenomena such as resonance, synchronization, and signal transmission promotion fully demonstrate the positive role of noise in the response and transmission of neural signals. Finally, the stochastic effects in neuronal systems are summarized, and potential applications are proposed.

Abstract:Activity-dependent plasticity plays an important role in biological neural network learning. Unlike the backpropagation-based learning in artificial neural networks that depends on supervised signals, biological neurons adjust themselves using their own historical behaviors as a clue to benefit information processing. Inspired by this biological neural mechanism, this paper proposes a novel neuron model, named Activity-Dependent Neuron (ADN). The basic idea of ADN is to add a gate in the neuron model to control its signal conduction capability, that is, the gate will facilitate transmission of important signals while suppress trivial signals. This idea can be achieved by self-tuning of activation behaviors. We also develop an iterative training algorithm, so that the ADNs can be smoothly incorporated into deep neural networks to jointly learn with the network weights. Experimental results found that the ADNs can efficiently improve the noise-resistant capability. Compared with the state-of-the-art, it is more robust to unforeseen noises. It does not need noise data for training.

Abstract:The random transitions of ion channels between conducting and non-conducting states generate a source of internal fluctuations in a neuron, known as channel noise. The standard method for modeling fluctuations in the states of ion channels uses continuous-time Markov chains nonlinearly coupled to a differential equation for voltage. Beginning with the work of Fox and Lu, there have been attempts to generate simpler models that use stochastic differential equation (SDEs) to approximate the stochastic spiking activity produced by Markov chain models. Recent numerical investigations, however, have raised doubts that SDE models can preserve the stochastic dynamics of Markov chain models. We analyze three SDE models that have been proposed as approximations to the Markov chain model: one that describes the states of the ion channels and two that describe the states of the ion channel subunits. We show that the former channel-based approach can capture the distribution of channel noise and its effect on spiking in a Hodgkin-Huxley neuron model to a degree not previously demonstrated, but the latter two subunit-based approaches cannot. Our analysis provides intuitive and mathematical explanations for why this is the case: the temporal correlation in the channel noise is determined by the combinatorics of bundling subunits into channels, and the subunit-based approaches do not correctly account for this structure. Our study therefore confirms and elucidates the findings of previous numerical investigations of subunit-based SDE models. Moreover, it presents the first evidence that Markov chain models of the nonlinear, stochastic dynamics of neural membranes can be accurately approximated by SDEs. This finding opens a door to future modeling work using SDE techniques to further illuminate the effects of ion channel fluctuations on electrically active cells.

Abstract:Neuronal membrane potentials fluctuate stochastically due to conductance changes caused by random transitions between the open and close states of ion channels. Although it has previously been shown that channel noise can nontrivially affect neuronal dynamics, it is unknown whether ion-channel noise is strong enough to act as a noise source for hypothesised noise-enhanced information processing in real neuronal systems, i.e. 'stochastic facilitation.' Here, we demonstrate that biophysical models of channel noise can give rise to two kinds of recently discovered stochastic facilitation effects in a Hodgkin-Huxley-like model of auditory brainstem neurons. The first, known as slope-based stochastic resonance (SBSR), enables phasic neurons to emit action potentials that can encode the slope of inputs that vary slowly relative to key time-constants in the model. The second, known as inverse stochastic resonance (ISR), occurs in tonically firing neurons when small levels of noise inhibit tonic firing and replace it with burst-like dynamics. Consistent with previous work, we conclude that channel noise can provide significant variability in firing dynamics, even for large numbers of channels. Moreover, our results show that possible associated computational benefits may occur due to channel noise in neurons of the auditory brainstem. This holds whether the firing dynamics in the model are phasic (SBSR can occur due to channel noise) or tonic (ISR can occur due to channel noise).

However, current studies of the effects of channel blockage and noise on neuronal response states are mainly limited to single-compartment neurons. This study introduces an analytical method to explore theintrinsic mechanism of channel blockage and noise effects on the response states of multi-compartment neurons, by using the smooth Pinsky-Rinzel two-compartment neuron model as a case study. Potassium, sodium, and calcium ion channel blockage coefficient are separately introduced to develop a smooth Pinsky-Rinzel neuron model with ion channel blockage. Methods such as single-parameter bifurcation analysis, double-parameter bifurcation analysis, coefficient of variation, and frequency characteristics analysis are utilized to examine the effects of various ion channel blockages on neuronal response states. Additionally, smooth Pinsky-Rinzel neuron Subunit noise model and conductance noise model are constructed to investigate their response characteristics by using interspike interval analysis and coefficient of variation indicators. Subthreshold stimulation is used to explore the presence of stochastic resonance phenomena. Single-parameter bifurcation analysis of the ion channel blockage model elucidates the dynamic processes of two torus bifurcations and limit point bifurcations in Pinsky-Rinzel neuron firing under potassium ion blocking. Double-parameter bifurcation analysis reveals a nearly linear increase in the Hopf bifurcation node of potassium ions with input current, whereas sodium ions exhibit a two-stage pattern of linear decline followed by exponential rise. The analysis of average firing frequency and coefficient of variation indicates that the moderate potassium channel blockage promotes firing, sodium channel blockage inhibits firing, and calcium channel blockage shows the complex characteristics but mainly promotes firing. Subthreshold stimulation of the channel noise model demonstrates the stochastic resonance phenomena in both models, accompanied by more intense chaotic firing, highlighting the positive role of noise in neural signal transmission. The interspike interval and coefficient of variation indicators show consistent variation levels for both noise models, with the conductance model displaying greater sensitivity to membrane area and stronger encoding capabilities. This study analyzes the general frequency characteristics of potassium and sodium ions in a multi-compartment neuron model through ion channel blocking model, providing special insights into the unique role of calcium ions. Further, the study explores stochastic resonance by using ion channel noise model, supporting the theory of noise-enhanced signal processing and offering new perspectives and tools for future studying complex information encoding in neural systems. By constructing an ion channel blockage model, the effects of potassium and sodium ions on the frequency characteristics of multi-compartment neurons are analyzed and the special influences of calcium ions are revealed. Using the ion channel noise model, the stochastic resonance is investigated, supporting the theory that the noise enhances signal processing. Abstract:Cortical excitability is sensitive to noise perturbations despite homeostatic regulation. However, modeling the effect of noise in a heterogeneous neuronal population is a mathematical challenge. Using a mean‐field model of a randomly connected excitatory population, we investigated how noise affects excitability at the resting state. Stable equilibrium analysis revealed that increasing noise intensity enhances excitability, indexed by synaptic conductance. Further, this enhancement disappeared when excitability is sufficiently high due to strong mean input current, but not influenced by the connection probability of the population. Finally, the mechanism of noise‐induced enhancement was explained by eigenvalues moving toward the imaginary axis. © 2024 Institute of Electrical Engineers of Japan and Wiley Periodicals LLC.

Abstract:We consider a classical space-clamped Hodgkin-Huxley model neuron stimulated by synaptic excitation and inhibition with conductances represented by Ornstein-Uhlenbeck processes. Using numerical solutions of the stochastic model system obtained by an Euler method, it is found that with excitation only there is a critical value of the steady state excitatory conductance for repetitive spiking without noise and for values of the conductance near the critical value small noise has a powerfully inhibitory effect. For a given level of inhibition there is also a critical value of the steady state excitatory conductance for repetitive firing and it is demonstrated that noise either in the excitatory or inhibitory processes or both can powerfully inhibit spiking. Furthermore, near the critical value, inverse stochastic resonance was observed when noise was present only in the inhibitory input process. The system of 27 coupled deterministic differential equations for the approximate first and second order moments of the 6-dimensional model is derived. The moment differential equations are solved using Runge-Kutta methods and the solutions are compared with the results obtained by simulation for various sets of parameters including some with conductances obtained by experiment on pyramidal cells of rat prefrontal cortex. The mean and variance obtained from simulation are in good agreement when there is spiking induced by strong stimulation and relatively small noise or when the voltage is fluctuating at subthreshold levels. In the occasional spike mode sometimes exhibited by spinal motoneurons and cortical pyramidal cells the assunptions underlying the moment equation approach are not satisfied.

Noise-induced behaviors in neural mean field dynamics

Abstract:The collective behavior of cortical neurons is strongly affected by the presence of noise at the level of individual cells. In order to study these phenomena in large-scale assemblies of neurons, we consider networks of firing-rate neurons with linear intrinsic dynamics and nonlinear coupling, belonging to a few types of cell populations and receiving noisy currents. Asymptotic equations as the number of neurons tends to infinity (mean field equations) are rigorously derived based on a probabilistic approach. These equations are implicit on the probability distribution of the solutions which generally makes their direct analysis difficult. However, in our case, the solutions are Gaussian, and their moments satisfy a closed system of nonlinear ordinary differential equations (ODEs), which are much easier to study than the original stochastic network equations, and the statistics of the empirical process uniformly converge towards the solutions of these ODEs. Based on this description, we analytically and numerically study the influence of noise on the collective behaviors, and compare these asymptotic regimes to simulations of the network. We observe that the mean field equations provide an accurate description of the solutions of the network equations for network sizes as small as a few hundreds of neurons. In particular, we observe that the level of noise in the system qualitatively modifies its collective behavior, producing for instance synchronized oscillations of the whole network, desynchronization of oscillating regimes, and stabilization or destabilization of stationary solutions. These results shed a new light on the role of noise in shaping collective dynamics of neurons, and gives us clues for understanding similar phenomena observed in biological networks.

Abstract:We continue the work of a series of previous studies of a mathematical model that describes the mean-field limit behavior of a homogeneous network of excitatory point spiking neurons. Contrary to other models, here noise is intrinsic to the neurons through a membrane-potential dependent spiking rate probability, that we assume to be given by a power law. This allows one to collapse several sources of neural noise into a single function, which aids for a more treatable mathematical and comptuational analysis. In particular, we give pseudo-analytical expressions for the invariant distributions of membrane potentials across the population and study their stability computationally. The neurons are assumed to be connected both by chemical and possibly electrical (gap junction) synapses and can also undergo a leakage of ions with the extracellular medium. The distributions are of compact support whenever leakage or gap junction rates are non-zero and an infinite discontinuity appears at the maximum potential in the population. This happens invariably for the leakage (with no gap junctions) and gap junction (without leakage) cases. However, these discontinuous distributions might only be stable for linear spiking rate functions. It was recently shown how the network can present highly synchronous states of global oscillations in its activity when leakage is not present and gap junctions are strong enough. Here we confirm how oscillations persist also when weak leakage is considered. Thus, these dendritic (rather than axonal) gap junctions could play a role in the high levels of neural synchrony necessary for the development of neural systems at young ages. The model, thus, presents a rich phenomenology and capability to reproduce several biological features of neural networks despite of its mathematical simplicity, hence presenting a powerful tool for high performance computing.

Abstract:We propose a stochastic dynamical model of noisy neural networks with complex architectures and discuss activation of neural networks by a stimulus, pacemakers and spontaneous activity. This model has a complex phase diagram with self-organized active neural states, hybrid phase transitions, and a rich array of behavior. We show that if spontaneous activity (noise) reaches a threshold level then global neural oscillations emerge. Stochastic resonance is a precursor of this dynamical phase transition. These oscillations are an intrinsic property of even small groups of 50 neurons.

Abstract:The response of neurons is highly sensitive to the stimulus. The stimulus can be associated with a direct injection in vitro experimentation (e.g., time dependent and independent inputs); or post-synaptic potentials resulting from the interaction of many neurons. A typical incoming stimulus resembles a noise which in principle can be described as a random variable. In computational neuroscience, the noise has been extensively studied for different setups. In this study, we investigate the effect of noisy inputs in a minimal network of two identical leaky integrate-and-fire (LIF) neurons interacting with finite pulses. In particular, we consider a Gaussian white noise as a standard function for stochastic modelling of neurons, while taking into account the pulse width as an elementary component for the signal transmission. By exploring the role of noise and finite pulses, the two neurons show a synchronous spiking behaviour characterized by fluctuations in the interspike intervals. Above some critical values the synchronous regime collapses onto asynchronous dynamics. The abrupt change in such dynamics is accompanied by a hysteresis, i.e., the coexistence of synchronous and asynchronous firing behaviour.

Characterization of subthreshold voltage fluctuations in neuronal membranes

Abstract:Synaptic noise due to intense network activity can have a significant impact on the electrophysiological properties of individual neurons. This is the case for the cerebral cortex, where ongoing activity leads to strong barrages of synaptic inputs, which act as the main source of synaptic noise affecting on neuronal dynamics. Here, we characterize the subthreshold behavior of neuronal models in which synaptic noise is represented by either additive or multiplicative noise, described by Ornstein-Uhlenbeck processes. We derive and solve the Fokker-Planck equation for this system, which describes the time evolution of the probability density function for the membrane potential. We obtain an analytic expression for the membrane potential distribution at steady state and compare this expression with the subthreshold activity obtained in Hodgkin-Huxley-type models with stochastic synaptic inputs. The differences between multiplicative and additive noise models suggest that multiplicative noise is adequate to describe the high-conductance states similar to in vivo conditions. Because the steady-state membrane potential distribution is easily obtained experimentally, this approach provides a possible method to estimate the mean and variance of synaptic conductances in real neurons.

Abstract:Neuronal models based on the Hodgkin-Huxley equation form a fundamental framework in the field of computational neuroscience. While the neuronal state is often modeled deterministically, experimental recordings show stochastic fluctuations, presumably driven by molecular noise from the underlying microphysical conditions. In turn, the firing of individual neurons gives rise to an electric field n extracellular space, also thought to affect the firing pattern of nearby neurons. We develop a multiscale model which combines a stochastic ion channel gating process taking place on the neuronal membrane, together with the propagation of an action potential along the neuronal structure. We also devise a numerical method relying on a split-step strategy which effectively couples these two processes and we experimentally test the feasibility of this approach. We finally also explain how the approach can be extended with Maxwell's equations to allow the potential to be propagated in extracellular space.

Abstract:Conductance-based equations for electrically active cells form one of the most widely studied mathematical frameworks in computational biology. This framework, as expressed through a set of differential equations by Hodgkin and Huxley, synthesizes the impact of ionic currents on a cell's voltage--and the highly nonlinear impact of that voltage back on the currents themselves--into the rapid push and pull of the action potential. Later studies confirmed that these cellular dynamics are orchestrated by individual ion channels, whose conformational changes regulate the conductance of each ionic current. Thus, kinetic equations familiar from physical chemistry are the natural setting for describing conductances; for small-to-moderate numbers of channels, these will predict fluctuations in conductances and stochasticity in the resulting action potentials. At first glance, the kinetic equations provide a far more complex (and higher-dimensional) description than the original Hodgkin-Huxley equations or their counterparts. This has prompted more than a decade of efforts to capture channel fluctuations with noise terms added to the equations of Hodgkin-Huxley type. Many of these approaches, while intuitively appealing, produce quantitative errors when compared to kinetic equations; others, as only very recently demonstrated, are both accurate and relatively simple. We review what works, what doesn't, and why, seeking to build a bridge to well-established results for the deterministic equations of Hodgkin-Huxley type as well as to more modern models of ion channel dynamics. As such, we hope that this review will speed emerging studies of how channel noise modulates electrophysiological dynamics and function. We supply user-friendly MATLAB simulation code of these stochastic versions of the Hodgkin-Huxley equations on the ModelDB website (accession number 138950) and http://www.amath.washington.edu/~etsb/tutorials.html.

Abstract:The effect of intrinsic channel noise is investigated for the dynamic response of a neuronal cell with a delayed feedback loop. The loop is based on the so-called autapse phenomenon in which dendrites establish not only connections to neighboring cells but as well to its own axon. The biophysical modeling is achieved in terms of a stochastic Hodgkin-Huxley model containing such a built in delayed feedback. The fluctuations stem from intrinsic channel noise, being caused by the stochastic nature of the gating dynamics of ion channels. The influence of the delayed stimulus is systematically analyzed with respect to the coupling parameter and the delay time in terms of the interspike interval histograms and the average interspike interval. The delayed feedback manifests itself in the occurrence of bursting and a rich multimodal interspike interval distribution, exhibiting a delay-induced reduction of the spontaneous spiking activity at characteristic frequencies. Moreover, a specific frequency-locking mechanism is detected for the mean interspike interval.

Abstract:Multi-electrode arrays (MEA) are increasingly used to investigate spontaneous neuronal network activity. The recorded signals comprise several distinct components: Apart from artefacts without biological significance, one can distinguish between spikes (action potentials) and subthreshold fluctuations (local fields potentials). Here we aim to develop a theoretical model that allows for a compact and robust characterization of subthreshold fluctuations in terms of a Gaussian statistical field theory in two spatial and one temporal dimension. What is usually referred to as the driving noise in the context of statistical physics is here interpreted as a representation of the neural activity. Spatial and temporal correlations of this activity give valuable information about the connectivity in the neural tissue. We apply our methods on a dataset obtained from MEA-measurements in an acute hippocampal brain slice from a rat. Our main finding is that the empirical correlation functions indeed obey the logarithmic behaviour that is a general feature of theoretical models of this kind. We also find a clear correlation between the activity and the occurence of spikes. Another important insight is the importance of correcly separating out certain artefacts from the data before proceeding with the analysis.
