Abstract:As a signature of reproductive interference (RI), we reviewed hybrid production in eusocial bumblebees in Japan, by comparing introduced Bombus terrestris with native B. ignitus in Honshu (main island of Japan) and with native B. hypocrita sapporoensis in Hokkaido (northern island of Japan). In this review, we present additional new data showing hybrid production between introduced B. terrestris and native B. ignitus in Honshu. Interspecific mating with introduced B. terrestris disrupts the reproduction of native B. h. sapporoensis and B. ignitus, which belong to the same subgenus of Bombus, through inviable egg production. This interference appears to facilitate species replacement on Hokkaido. Simultaneously, the mating frequencies for queens of B. terrestris have increased, suggesting that polyandry might evolve in response to the extent of RI between B. terrestris and B. h. sapporoensis. To suppress the population size of B. terrestris in Hokkaido, two methods have been proposed: the mass release of B. h. sapporoensis males to induce RI between the two species and the spraying of insecticides against foraging workers so that the workers will carry the insecticides back to their colonies, killing the immature bees within the colonies. A candidate insecticide type is insect growth regulator, which may disrupt larval development without any apparent effect on foraging workers.

Extreme polyandry aids the establishment of invasive populations of a social insect

Abstract:Although monandry is believed to have facilitated the evolution of eusociality, many highly eusocial insects have since evolved extreme polyandry. The transition to extreme polyandry was likely driven by the benefits of within-colony genetic variance to task specialization and/or disease resistance, but the extent to which it confers secondary benefits, once evolved, is unclear. Here we investigate the consequences of extreme polyandry on the invasive potential of the Asian honey bee, Apis cerana. In honey bees and other Hymenoptera, small newly founded invasive populations must overcome the genetic constraint of their sex determination system that requires heterozygosity at a sex-determining locus to produce viable females. We find A. cerana queens in an invasive population mate with an average of 27 males (range 16–42) that would result in the founding queen/s carrying 75% of their source population’s sex alleles in stored sperm. This mating frequency is similar to native-range Chinese A. cerana (mean 29 males, range 19–46). Simulations reveal that extreme polyandry reduces the risk, relative to monandry or moderate polyandry, that colonies produce a high incidence of inviable brood in populations that have experienced a founder event, that is, when sex allele diversity is low and/or allele frequencies are unequal. Thus, extreme polyandry aids the invasiveness of A. cerana in two ways: (1) by increasing the sex locus allelic richness carried to new populations with each founder, thereby increasing sex locus heterozygosity; and (2) by reducing the population variance in colony fitness following a founder event.

Abstract:Abstract Polyandry (multiple mating by females) is a central challenge for understanding the evolution of eusociality. Several hypotheses have been proposed to explain its observed benefits in eusocial Hymenoptera, one of which, the parasite–pathogen hypothesis (PPH), posits that high genotypic variance among workers for disease resistance prevents catastrophic colony collapse. We tested the PPH in the polyandrous wasp Vespula shidai. We infected isolated workers with the entomopathogenic fungus Beauveria bassiana and quantified their survival in the laboratory. Additionally, we conducted a paternity analysis of the workers using nine microsatellite loci to investigate the relationship between survival and the matriline and patriline membership of the workers. As predicted by the PPH, nestmate workers of different patrilines showed differential resistance to B. bassiana. We also demonstrated variation in virulence among strains of B. bassiana. Our results are the first to directly support the PPH in eusocial wasps and suggest that similar evolutionary pressures drove the convergent origin and maintenance of polyandry in ants, bees, and wasps.

Abstract:Polyandry in social Hymenoptera is associated with reduced within-colony relatedness and increased colony-level ecologic fitness. One explanation for this sees increasing within-nest genetic diversity as a mechanism for improving group task efficiency and colony competitiveness. A queen on her mating flight captures nearly 90% of her breeding population's allele potential by her tenth effective mating ( m e ~ 10 males). Under this population allele capture (PAC) model, colony fitness gains track mating number in an asymptotic manner, leveling out after about the tenth mating. A supporting theory we call the genotype composition (GC) model sees genetic novelty at mating levels higher than the m e ~ 10 asymptote, the hyperpolyandry zone, resulting from unique genotype compositions whose number are potentially infinite. Colony fitness gains under the GC model will track mating number in a linear manner. We set up field colonies with Apis mellifera queens each instrumentally mated with 1, 2, 4, 8, 16, or 32 males, creating a polyandry gradient bracketing the qualitative divide of m e ~ 10, measured tokens of colony level fitness, and collected observation hive data. Our results lead us to conclude that (1) ancestral colony traits fundamental to eusociality (cooperative brood care) respond to mating level changes at or below m e ~ 10 in a manner consistent with the PAC model, whereas (2) more derived specialized colony phenotypes (resistance to the non-native parasite Varroa destructor ) continue improving with increasing m e in a manner consistent with the GC model. By either model, (3) the mechanism for increasing colony fitness is an increase in worker task specialisms and task efficiency.

Abstract:Abstract Background Efficient breeding programs are difficult to implement in honeybees due to their biological specificities (polyandry and haplo-diploidy) and complexity of the traits of interest, with performances being measured at the colony scale and resulting from the joint effects of tens of thousands of workers (called direct effects) and of the queen (called maternal effects). We implemented a Monte Carlo simulation program of a breeding plan designed specifically for Apis mellifera ’s populations to assess the impact of polyandry versus monoandry on colony performance, inbreeding level and genetic gain depending on the individual selection strategy considered, i.e. complete mass selection or within-family (maternal lines) selection. We simulated several scenarios with different parameter setups by varying initial genetic variances and correlations between direct and maternal effects, the selection strategy and the polyandry level. Selection was performed on colony phenotypes. Results All scenarios showed strong increases in direct breeding values of queens after 20 years of selection. Monoandry led to significantly higher direct than maternal genetic gains, especially when a negative correlation between direct and maternal effects was simulated. However, the relative increase in these genetic gains depended also on their initial genetic variability and on the selection strategy. When polyandry was simulated, the results were very similar with either 8 or 16 drones mated to each queen. Across scenarios, polyandrous mating resulted in equivalent or higher gains in performance than monoandrous mating, but with considerably lower inbreeding rates. Mass selection conferred a ~ 20% increase in performance compared to within-family selection, but was also accompanied by a strong increase in inbreeding levels (25 to 50% higher). Conclusions Our study is the first to compare the long-term effects of polyandrous versus monoandrous mating in honeybee breeding. The latter is an emergent strategy to improve specific traits, such as resistance to varroa, which can be difficult or expensive to phenotype. However, if used during several generations in a closed population, monoandrous mating increases the inbreeding level of queens much more than polyandrous mating, which is a strong limitation of this strategy.

Abstract:Lone Bombus ignitus queens are known to start nests in small underground cavities. To examine the nest‐site preference of post‐hibernating queens, choice tests were carried out by providing queens with orphan colonies and empty cavities as possible nesting sites within an experimental box. Our results showed that the queens had a strong preference for takeover of an orphan colony, suggesting that nest takeover (usurpation) could occur in nature even with the presence of possible empty cavities for nesting. We compared the colony‐growth process and final production of sexuals between non‐takeover and takeover colonies. The increase in the number of egg cups was faster in the takeover colonies, suggesting that orphan broods elicit earlier oviposition by the usurping queen. Reproductives emerged earlier (significant for new queens) and in greater numbers (males) from takeover colonies than from non‐takeover colonies. Thus, post‐hibernating B. ignitus queens would search for and take over small orphan colonies to increase their fitness. Abstract:Polyandry, when females mate with more than one male, is theorised to play an important role in successful colonisation of new habitats. In addition to possible benefits from sexual selection, even mild polyandry could facilitate colonisation by protecting against inbreeding and reducing the costs of mating with incompatible or infertile males. Here, we measure the importance of mild polyandry for population viability and reproductive fitness following experimental founder events into a higher‐temperature regime. Using colonisation experiments with the model beetle Tribolium castaneum, in which females can produce offspring for up to 140 days following a single mating, we founded more than 100 replicate populations using single females that had been given the opportunity to mate with either one or two males and then tracked their subsequent population dynamics. Following population viability and fitness across 10 generations, we found that extinction rates were significantly lower in populations founded by females given polyandrous opportunities to mate with two males (9%) compared to populations founded by monogamous females (34%). In addition, populations founded by females that had been provided with opportunities to store sperm from two different males showed double the median productivity following colonisation compared to monogamous‐founded populations. Notably, we identified short‐term and longer‐term benefits to post‐colonisation populations from double‐mating, with results suggesting that polyandry acts to both protect against mating with incompatible males through the founder event, and reduce inbreeding depression as the colonisation proceeds for 10 generations. Our results therefore show that even mild polyandry provides both reproductive and genetic benefits for colonising populations.

Novel insights into paternity skew in a polyandrous social wasp

Abstract:Females of many species are polyandrous. However, polyandry can give rise to conflict among individuals within families. We examined the level of polyandry and paternity skew in the common eastern yellowjacket wasp, Vespula maculifrons, in order to gain a greater understanding of conflict in social insects. We collected 10 colonies of V. maculifrons and genotyped workers and prereproductive queens at highly variable microsatellite markers to assign each to a patriline. Genotypic data revealed evidence of significant paternity skew among patrilines. In addition, we found that patrilines contributed differentially to caste production (worker vs. queen), suggesting an important role for reproductive conflict not previously discovered. We also investigated if patterns of paternity skew and mate number varied over time. However, we found no evidence of changes in levels of polyandry when compared to historical data dating back almost 40 years. Finally, we measured a suite of morphological traits in individuals from the most common and least common patrilines in each colony to test if males that showed highly skewed reproductive success also produced offspring that differed in phenotype. Our data revealed weak correlation between paternity skew and morphological phenotype of offspring sired by different males, suggesting no evidence of evolutionary tradeoffs at the level investigated. Overall, this study is the first to report significant paternity and caste-associated skew in V. maculifrons, and to investigate the phenotypic consequences of skew in a social wasp. Our results suggest that polyandry can have important consequences on the genetic and social structure of insect societies.

Abstract:Bet-hedging via polyandry (spreading the extinction risk of the female's lineage over multiple males) may explain the evolution of female multiple mating, which is found in a wide range of animal and plant taxa. This hypothesis posits that females can increase their fitness via polyandrous mating when "unsuitable" males (i.e., males causing reproductive failure for various reasons) are frequent in the population and females cannot discriminate such unsuitable mates. Although recent theoretical studies have shown that polyandry can operate as a bet-hedging strategy, empirical tests are scarce. In the present study, we tested the bet-hedging polyandry hypothesis by using the red flour beetle Tribolium castaneum. We compared female reproductive success between monandry and polyandry treatments when females mated with males randomly collected from an experimental population, including 20% irradiated (infertile) males. In addition, we evaluated geometric mean fitness across multiple generations as the index of adaptability of bet-hedging traits. Polyandrous females showed a significantly higher egg hatching rate and higher geometric mean fitness than monandrous females. These results strongly support the bet-hedging polyandry hypothesis.

Abstract:Evidence for a trade-off between reproduction and immunity has manifested in many animal species, including social insects. However, investigations in social insect queens present a conundrum: new gynes of many social hymenopterans, such as bumble bees and ants, must first mate, then transition from being solitary to social as they establish their nests, thus experiencing confounding shifts in environmental conditions. Worker bumble bees offer an opportunity to investigate patterns of immune protein expression associated with ovary activation while minimizing extraneous environmental factors and genetic differences. Here, we use proteomics to interrogate the patterns of immune protein expression of female bumble bees ( Bombus impatiens ) by (i) sampling queens at different stages of their life cycle, then (ii) by sampling workers with different degrees of ovary activation. Patterns of immune protein expression in the haemolymph of queens are consistent with a reproduction–immunity trade-off, but equivalent samples from workers are not. This brings into question whether queen bumble bees really experience a reproduction–immunity trade-off, or if patterns of immune protein expression may actually be due to the selective pressure of the different environmental conditions they are exposed to during their life cycle.

Abstract:The best mating strategy for males differs from that of females, because females gain from mating with several males (polyandry), but males gain from monopolizing the females. As a consequence, males have evolved a variety of methods, such as the transfer of inhibitory substances from their accessory glands, to ensure exclusive paternity of the female's offspring, generally with detrimental effects on female fitness. Inhibitory substances have been identified as peptides or other specific molecules. Unfortunately, in social insects male-mating traits are investigated only poorly, although male social insects might have the same fundamental influence on female-mating behavior as found in other species. A recently developed technique for the artificial insemination of bumblebee queens allowed us to investigate which chemical compound in the mating plug of male bumblebees, Bombus terrestris L., prevents females (queens) from further mating. Surprisingly, we found that the active substance is linoleic acid, a ubiquitous and rather unspecific fatty acid. Contrary to mating plugs in other insect species, the bumblebee mating plug is highly efficient and allows the males to determine queen-mating frequencies.
Pheromonal Predisposition to Social Parasitism in the Honeybee Apis Mellifera Capensis

Abstract:In honeybees, worker reproduction is mainly regulated by pheromones produced by the brood and the queen. The source of one of the queen pheromones influencing worker reproduction has been located in the mandibular glands. In nonlaying workers, this gland's profile is dominated by fatty acids that are incorporated into the food given to the brood and to nest mates. After queen loss and onset of reproductive activity, workers are able to synthesize different fatty acids, which are normally only produced by queens and that contribute to their reproductive success. Apis mellifera capensis workers have the ability to rapidly produce queen-like mandibular profiles that could represent an important factor in their ability to behave as facultative intraspecific social parasites. Indeed, A. m. capensis workers can take over reproduction from the host queens in colonies of other subspecies. Here, we show that in the presence of their own queen, the mandibular gland profile of A. m. capensis workers is dominated by the precursor of the major compound of the queen pheromone. This is a unique trait among honeybee workers and suggests that A. m. capensis workers are primed for reproduction and that this phenomenon represents a pheromonal predisposition to social parasitism. We identified geographical variation in the ratio of queen-to worker-specific compounds in themandibular gland profile of A. m. capensis workers, which corresponds with the introgression with the neighboring subspecies A. m. scutellata.
Positive selection in cytochrome P450 genes is associated with gonad phenotype and mating strategy in social bees

Abstract:The honey bee, Apis mellifera differs from all other social bees in its gonad phenotype and mating strategy. Honey bee queens and drones have tremendously enlarged gonads, and virgin queens mate with several males. In contrast, in all the other bees, the male and female gonads are small, and the females mate with only one or very few males, thus, suggesting an evolutionary and developmental link between gonad phenotype and mating strategy. RNA-seq comparisons of A. mellifera larval gonads revealed 870 genes as differentially expressed in queens versus workers and drones. Based on Gene Ontology enrichment we selected 45 genes for comparing the expression levels of their orthologs in the larval gonads of the bumble bee Bombus terrestris and the stingless bee, Melipona quadrifasciata, which revealed 24 genes as differentially represented. An evolutionary analysis of their orthologs in 13 solitary and social bee genomes revealed four genes with evidence of positive selection. Two of these encode cytochrome P450 proteins , and their gene trees indicated a lineage-specific evolution in the genus Apis, indicating that cytochrome P450 genes may be involved in the evolutionary association of polyandry and the exaggerated gonad phenotype in social bees.

Abstract:PREMISE: Fecundity and mating outcomes commonly differ among plant populations occupying contrasting environments. If self-pollination occurs primarily among flowers within plants, contrasting reproductive outcomes among populations must reflect environmental effects on plant-pollinator interactions. Specifically, local conditions could affect features of plant phenotypes that influence pollinator behavior, in turn modifying plant reproductive outcomes.METHODS: We compared phenotypes, pollinator abundance and behavior, and female fecundity and mating in two meadow populations and two forest populations of Aconitum kusnezoffii within 3 km of each other. Mating outcomes were assessed using microsatellites.RESULTS: Meadow plants generally produced more, shorter ramets with more, larger flowers, but less nectar per flower than forest plants. These differences likely largely represent phenotypic plasticity. Individual bumble bees visited more flowers on forest plants, likely because the more abundant bees in the meadows depleted nectar availability, as indicated by briefer visits to individual flowers. Despite similar fruit set in both habitats, forest plants set more seeds per fruit. Nevertheless, meadow plants produced more seeds overall, owing to sevenfold greater flower production. Consistent with individual bees visiting fewer flowers on meadow plants, more of their seeds were outcrossed. However, the outcrossed seeds of forest plants included more male mates.CONCLUSIONS: Reproductive outcomes can vary among populations of animal-pollinated plants as a result of differences in the availability of effective pollinators and environmental effects on plant phenotypes, and their functional consequences for pollinator behavior that governs pollen dispersal.

Substances in the mandibular glands mediate queen effects on larval development and colony organization in an annual bumble bee

Abstract:Social organization is commonly dynamic, with extreme examples in annual social insects, but little is known about the underlying signals and mechanisms. Bumble bee larvae with close contact to a queen do not differentiate into gynes, pupate at an earlier age, and are commonly smaller than siblings that do not contact a queen. We combined detailed observations, proteomics, microRNA transcriptomics, and gland removal surgery to study the regulation of brood development and division of labor in the annual social bumble bee Bombus terrestris . We found that regurgitates fed to larvae by queens and workers differ in their protein and microRNA composition. The proteome of the regurgitate overlaps significantly with that of the mandibular (MG) and hypopharyngeal glands (HPG), suggesting that these exocrine glands are sources of regurgitate proteins. The proteome of the MG and HPG, but not the salivary glands, differs between queens and workers, with caste-specificity preserved for the MG and regurgitate proteomes. Queens subjected to surgical removal of the MG showed normal behavior, brood care, and weight gain, but failed to shorten larval development. These findings suggest that substances in the queen MG are fed to larvae and influence their developmental program. We suggest that when workers emerge and contribute to larval feeding, they dilute the effects of the queen substances, until she can no longer manipulate the development of all larvae. Longer developmental duration may allow female larvae to differentiate into gynes rather than to workers, mediating the colony transition from the ergonomic to the reproductive phase.
A game theoretical analysis of the mating sign behavior in the honey bee

Abstract:Queens of the honey bee, Apis mellifera (L.), exhibit extreme polyandry, mating with up to 45 different males (drones). This increases the genetic diversity of their colonies, and consequently their fitness. After copulation, drones leave a mating sign in the genital opening of the queen which has been shown to promote additional mating of the queen. On one hand, this signing behavior is beneficial for the drone because it increases the genetic diversity of the resulting colony that is to perpetuate his genes. On the other hand, it decreases the proportion of the drone's personal offspring among colony members which is reducing drone fitness. We analyze the adaptiveness and evolutionary stability of this drone's behavior with a game-theoretical model. We find that theoretically both the strategy of leaving a mating sign and the strategy of not leaving a mating sign can be evolutionary stable, depending on natural parameters. However, the signing strategy is not favored for most scenarios, including the cases that are biologically plausible in reference to empirical data. We conclude that leaving a sign is not in the interest of the drone unless it serves biological functions other than increasing subsequent queen mating chances. Nevertheless, our analysis can also explain the prevalence of such a behavior of honey bee drones by a very low evolutionary pressure for an invasion of the nonsigning strategy.

We quantified the fecundity and level of polyandry in field populations of two stored grain beetles, Tribolium castaneum (Herbst) and Rhyzopertha dominica (F.), by comparing resident with dispersing individuals at common sites. Almost all females caught in flight had mated, with paternity analysis from 110 parent–offspring arrays revealing that most females had mated with more than one male (T. castaneum: 90.6, R. dominica: 70.2%). No difference in the extent of polyandry between beetles collected from grain in storage and beetles caught in flight was detected for either species, and the degree of polyandry did not impact overall fecundity. Levels of polyandry were similar to those in recently established (first generation) laboratory cultures of both species. Our results provide the first direct evidence of polyandry in wild populations of T. castaneum and R. dominica, and that females that have mated several times have the potential to produce offspring of multiple genotypes in the field. Females thus invade resource patches (whether previously colonised or not) with considerable genetic diversity to distribute across their offspring, an advantage in a newly colonised site because it increases the chances of her offspring establishing there. This advantageous consequence of polyandry to dispersing individuals, and the fact that most of the individual beetles sampled in the field had mated multiple times, provides justification as to why polyandry should not be viewed as an evolutionary paradox.

Abstract:Nonrecombining genomic variants underlie spectacular social polymorphisms, from bird mating systems to ant social organization. Because these "social supergenes" affect multiple phenotypic traits linked to survival and reproduction, explaining their persistence remains a substantial challenge. Here, we investigate how large nonrecombining genomic variants relate to colony social organization, mating system and dispersal in the Alpine silver ant, Formica selysi. The species has colonies headed by a single queen (monogynous) and colonies headed by multiple queens (polygynous). We confirmed that a supergene with alternate haplotypes-Sm and Sp-underlies this polymorphism in social structure: Females from mature monogynous colonies had the Sm/Sm genotype, while those from polygynous colonies were Sm/Sp and Sp/Sp. Queens heading monogynous colonies were exclusively mated with Sm males. In contrast, queens heading polygynous colonies were mated with Sp males and Sm males. Sm males, which are only produced by monogynous colonies, accounted for 22.9% of the matings with queens from mature polygynous colonies. This asymmetry between social forms in the degree of assortative mating generates unidirectional male-mediated gene flow from the monogynous to the polygynous social form. Biased gene flow was confirmed by a significantly higher number of private alleles in the polygynous social form. Moreover, heterozygous queens were three times as likely as homozygous queens to be multiply mated. This study reveals that the supergene variants jointly affect social organization and multiple components of the mating system that alter the transmission of the variants and thus influence the dynamics of the system.

Abstract:The invasive B whitefly and the indigenous ZHJ2 whitefly in China of the Bemisia tabaci complex represent two previously allopatric cryptic species. Laboratory experiments have shown that the invasive B can displace the indigenous ZHJ2. Here, we observed the mating behavior and mating interactions between B and ZHJ2 to elucidate the behavioral mechanisms underlying their competition. The 2 species are able to mate with each other, although at a significantly lower frequency than that between males and females within the same species. Heterospecifically-mated females produce only male progeny, indicating reproductive isolation. Heterospecific mating had only a marginal effect on reproduction of females in either species. Detailed observations on mating interactions between the 2 species showed that B had higher sexual activity than ZHJ2. B males were more likely to interrupt courtships initiated by rival males than ZHJ2 males. Compared with ZHJ2 males, B males were also more likely to successfully interrupt courtships initiated by rival heterospecific males than by males of the same species. In addition, B males guarded B females post-copulation, whereas ZHJ2 males did not. When the 2 species co-occurred, the behavioral differences and interactions between them resulted in an increased proportion of females produced in the offspring of B but reduced proportion of females in the progeny of ZHJ2. These asymmetric mating interactions might be an important mechanism underlying the displacement of ZHJ2 by B. Our study shows that reproductive interference might contribute to the competitive exclusion between closely related species during biological invasions.

Abstract:The study of polyandry has received increasing scientific attention with an emphasis on the fitness benefits and costs that females derive from mating with multiple males. There are still gaps in our understanding of how polyandry affects female fitness, however, as many previous studies compared the fitness outcomes of a single mating vs. two or three matings and did not separate the consequences of multiple mating from the costs of sexual harassment. We therefore conducted controlled mating trials with female fruit flies (Drosophila melanogaster) that could mate at either low (every eight days), medium (every four days), or high (every other day) rates while controlling for exposure to harassment from males. We found that female lifetime fitness was highest under the high followed by the medium mating-rate conditions. Moreover, we did not detect reductions in lifespan as a consequence of higher rates of polyandry. Our results demonstrate that even at realistically high rates, polyandry can lead to net fitness benefits for females, which can have major implications for sexual selection. Specifically, we discuss the significance of our findings as they relate to competition and the evolution of secondary sex characteristics in females, and sperm competition amongst males.
