Abstract:The ubiquitous presence of micro- and nanoplastics (MNPs) in soil has raised concerns regarding their potential effects on terrestrial plants. The coexistence and interactions between MNPs and heavy metals altering their phytotoxicity deserves further investigation. In this study, we explored the impacts of various concentrations of polystyrene nanoplastics (PS-NPs) and cadmium (Cd) alone or in combination on the growth and development of Arabidopsis thaliana. Additionally, we examined the effects of combined stress on the uptake and translocation of Cd within Arabidopsis thaliana. Our findings revealed several key insights: PS-NPs exhibited the capability to internalize in the maturation zone of Arabidopsis roots; the presence of Cd changed the particle size and zeta potential of PS-NPs; the presence of PS-NPs heightened Cd accumulation in the underground parts of Arabidopsis seedlings, leading to a stronger oxidative stress response in these regions; the composite stress exerted a more pronounced effect on the growth and development of Arabidopsis compared to individual stresses. Interestingly, while higher PS-NPs concentrations hindered Cd migration from roots to leaves, they also acted as carriers for Cd uptake in Arabidopsis roots. These findings shed light on the combined impacts of MNPs and heavy metals on plant physiology, offering theoretical insights to guide risk assessment strategies for MNPs and heavy metals in terrestrial ecosystems.

Abstract:Micro and nanoplastics (MPs/NPs) coupled with heavy metals are prevalent in both aquatic and terrestrial ecosystems. Their ecological toxicity and combined adverse effects have obtained significant concern. Past studies primarily focused on how MPs/NPs influence the behavior of heavy metals. Yet, the possible effects of heavy metals on MP/NP transport and toxicity within co-contaminated systems are still not well-understood. In this study, we conducted split-root experiments to explore the transport and toxicity of polystyrene (PS) particles of varying sizes in parsley seedlings, both with and without the addition of cadmium (Cd). Both the PS-NPs (100 nm) and PS-MPs (300 nm) traveled from the PS-spiked roots (Roots-1) to the non-PS-spiked roots (Roots-2), with or without Cd, possibly because of phloem transport. Furthermore, the presence of Cd reduced the accumulation and movement of PS-NP/MP in the roots, likely due to the increased positive charge (Cd 2+ ) on the PS surface. PS-NPs/MPs in both Roots-1 and Roots-2 were observed using transmission electron microscopy (TEM). When Cd was added to either Roots-1 (PS + Cd|H) or Roots-2 (PS|Cd), there was a minor reduction in the chlorophyll a and carotenoids content in leaves with PS|H. The adverse impacts of MPs|H on both indicators were influenced by the MP concentration. However, chlorophyll b significantly increased in the PS|H, PS + Cd|H, and PS|Cd treatments. Consequently, the chlorophyll a /b ratio declined, indicating inhibition of photosynthesis. The dehydrogenase content showed a minor change in Roots-1 and Roots-2 without Cd stress, whereas it significantly decreased on the Cd-spiked side and subsequently inhibited root growth. In contrast, the marked rise in glutathione (GSH) levels within Cd-spiked roots suggested, based on Gaussian analysis, that GSH and Cd chelation were instrumental in mitigating Cd toxicity. When Cd was introduced to both Roots-1 and Roots-2 simultaneously (PS + Cd|Cd), the aforementioned index showed a notable decline.

Abstract:To investigate the toxicological effects of polystyrene microplastics (PS-MPs), cadmium (Cd), and their combined contamination on the growth and physiological responses of V. faba seedlings, this experiment employed a hydroponic method. The Hoagland nutrient solution served as the control, changes in root growth, physiological and biochemical indicators of V. faba seedlings under different concentrations of PS-MPs (10, 100 mg/L) alone and combined with 0.5 mg/L Cd. The results demonstrated that the root biomass, root vitality, generation rate of superoxide radicals (O 2 ·− ), malondialdehyde (MDA) content, and superoxide dismutase (SOD) activity increased with increasing concentration under the influence of PS-MPs alone, while the soluble sugar content and peroxidase (POD) activity decreased. In the combined treatment with Cd, the trends of these indicators are generally similar to the PS-MPs alone treatment group. However, root vitality and SOD activity showed an inverse relationship with the concentration of PS-MPs. Furthermore, laser confocal and electron microscopy scanning revealed that the green fluorescent polystyrene microspheres entered the root tips of the V. faba and underwent agglomeration in the treatment group with a low concentration of PS-MPs alone and a high concentration of composite PS-MPs with Cd.

Abstract:Biodegradable plastics, such as poly(butylene adipate terephthalate) (PBAT) and polylactic acid (PLA), are potential alternatives to conventional polyethylene (PE), both of which are associated with the production of microplastics (MPs). However, the toxicity of these compounds on medicinal plants and their differential effects on plant morphophysiology remain unclear. This study supplemented soils with MPs sized at 200 μm at a rate of 1% w/w and incubated them for 50 days to investigate the impact of MPs on the growth and metabolites of dandelion (Taraxacum mongolicum Hand.-Mazz.). The results demonstrated that the investigated MPs decreased the growth of dandelion seedlings, induced oxidative stress, and altered the activity of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase). Based on the comprehensive toxicity assessment results, the ecological toxicity was in the following order: PE MPs > PBAT MPs > PLA MPs. Metabolomics analyses revealed metabolic reprogramming in dandelion plants, leading to the enrichment of numerous differentially accumulated metabolites (DAMs) in the leaves. These pathways include carbohydrate metabolism, energy metabolism, and biosynthesis of secondary metabolites, suggesting that dandelions respond to MP stress by enhancing the activity of sugar, organic acid, and amino acid metabolic pathways. In addition, phenolic acids and flavonoids are critical for maintaining the balance in the antioxidant defense system. Our results provide substantial insights into the toxicity of biodegradable MPs to plants and shed light on plant defense and adaptation strategies. Further assessment of the safety of biodegradable MPs in terrestrial ecosystems is essential to provide guidance for environmentally friendly management.

Abstract:With advances in plastic resource utilization technologies, polystyrene (PS) and sulfonated polystyrene (SPS) microplastics continue to be produced and retained in environmental media, potentially posing greater environmental risks. These plastics, due to their different physicochemical properties, may have different environmental impacts when compounded with other pollutants. The objective of this study was to investigate the combined toxic effects of PS and SPS on wheat using cadmium (Cd) as a background contaminant. The results demonstrated that Cd significantly impeded the normal growth of wheat by disrupting root development. Both PS and SPS exhibited hormesis at low concentrations and promoted wheat growth. Under combined toxicity, PS reduced oxidative stress and promoted the uptake of essential metal elements in wheat. Additionally, KEGG pathway analysis revealed that PS facilitated the repair of Cd-induced blockage of the TCA cycle and glutathione metabolism. However, high concentrations of SPS in combined toxicity not only enhanced oxidative stress and interfered with the uptake of essential metal elements, but also exacerbated the blocked TCA cycle and interfered with pyrimidine metabolism. These differences are related to the different stability (Zeta potential, Hydrodynamic particle size) of the two microplastics in the aquatic environment and their ability to carry heavy metal ions, especially Cd. The results of this study provide important insights into understanding the effects of microplastics on crops in the context of Cd contamination and their environmental and food safety implications.

Abstract:Background: Activator protein 1 is a transcription factor involved in the regulation of proinflammatory mediators. Activation of phagocytes by lipopolysaccharide depends on the expression of CD14 on the cell surface. In this study, we investigated the effects of morphine and nitric oxide on CD14 expression and activator protein 1 activation in human blood monocytes and neutrophils as well as the leukocyte cell line HL‐60. Methods: Whole blood was incubated with morphine, the nitric oxide donor S‐nitroso‐N‐acetyl‐penicillamine, naloxone or nitric oxide synthase inhibitors N&ohgr;‐nitro‐l‐arginine and N&ohgr;‐nitro‐l‐arginine‐methylester and stimulated with lipopolysaccharide. Activator protein 1 nuclear content was determined by flow cytometry in human blood neutrophils and monocytes. CD14 expression on neutrophils was measured after incubation with fluorescein isothiocyanate‐labelled antibodies. Electric mobility shift assay served for evaluation of activator protein 1 nuclear binding in HL‐60 cells. Results: Incubation of whole blood with morphine and subsequent stimulation with lipopolysaccharide decreased activator protein 1 nuclear content. Exposure to naloxone before morphine treatment abolished morphine‐induced inhibition of activator protein 1 activity in human blood monocytes and neutrophils. Nitric oxide synthase inhibitors also reversed morphine's effects. CD14 expression on neutrophils was reduced after morphine treatment. These effects were antagonized by nitric oxide synthase inhibitors and naloxone. Conclusion: Morphine inhibits activator protein 1 activation by a &mgr; opioid receptor pathway coupled to nitric oxide as second messenger. The decrease in CD14 expression caused by morphine may play a role in inhibition of activator protein 1 activation following lipopolysaccharide treatment of phagocytes.

Abstract:Microplastics are widespread in freshwater environments, their biological effects and combined effects of other pollutants have attracted extensive attention. In this study, we investigated the adsorption properties of heavy metals onto polystyrene (PS) microplastics as well as the bioavailability and toxicity of microplastics and heavy metals by hydroponic wheat seedlings experiment. Results showed that PS microplastics (0.5 mu m, 100 mg/L) had no significant effect on wheat seedlings growth, photosynthesis, and reactive oxygen species (ROS) content. However, PS microplastics could adsorb copper and cadmium, with a predominantly chemisorption. The accumulation of copper and cadmium in wheat seedlings reduced in the presence of PS microplastics, which meant the toxic effect by heavy metals might be mitigated. Compared with single heavy metals treatments, the combination of PS microplastics and heavy metals increased chlorophyll content, enhanced photosynthesis and reduced the accumulation of ROS. These findings suggest that PS microplastics (0.5 mu m, 100 mg/L) have a mitigating effect on the bioavailability and toxicity of copper and cadmium.
Enhanced microalgal toxicity due to polystyrene nanoplastics and cadmium co-exposure: From the perspective of physiological and metabolomic profiles

Abstract:As important emerging contaminants, nanoplastics can act as vectors for other environmental pollutants, resulting in their migration throughout ecosystems and altering their toxicity. In this study, the fluorescent dye label aggravated the toxicity of polystyrene (PS) nanoplastics (100 nm diameter particles) to microalgae Euglena gracilis. Therefore, the toxicity of non-fluorescent labelled PS alone and in combination with divalent cadmium (Cd2+) on Euglena gracilis in the environmentally relevant concentrations was investigated. Results revealed that co-exposure to 50 μg/L (1.1 × 1010 particles/L) PS and 50 μg/L Cd2+ resulted in synergistic effects, significantly inhibiting microalgal growth by 28.76%. Superoxide dismutase, peroxidase and extracellular polymeric substances were distinctly enhanced in co-exposure treatments compared to the control, indicating that cellular antioxidant defense responses were activated. LC-MS-based metabolomic analysis suggested that PS and Cd2+ exposure alone or in combination induced significant disruption to carbohydrate and purine metabolism-related pathways, as compared to controls. As part of the PS and Cd2+ stress response, differential metabolites involved in lipid metabolism and amino acid metabolism provide antioxidants and cell membrane protective molecules. Overall, this combined physiological and metabolomic analysis approach provides a better understanding of the potential risks posed by nanoplastics and heavy metal pollution in aquatic ecosystems.

Abstract:The coexistence of microplastics (MPs) and toxic metal contaminants in soils is becoming increasingly common, thereby posing serious threat to soil–plant systems. Cadmium (Cd) is the most common metal contaminant in soil and can easily combine with MPs, thereby altering its bioavailability. However, few studies have focused on the co-pollution of MPs and Cd, particularly the complex phytotoxicity caused by their interaction and the effect of co-exposure on Cd uptake in plants. We conducted pot experiments to compare the effects of exposure to polystyrene (PS) and Cd, as well as the effects of co-exposure (PS + Cd), on the physiological characteristics of Brassica chinensis L. and explored the regulatory factors of MPs on Cd uptake in plant tissues. The results showed that plant biomass, photosynthetic parameters, and chlorophyll content significantly decreased (p < 0.05) with increasing PS doses under treatment with MPs alone. Although the negative effects of PS and Cd co-exposure on plants were higher than those of PS alone, however, the addition of MPs reduced the toxicity effects of Cd on plants and decreased the uptake and accumulation of Cd by plants compared with the Cd treatment alone. Furthermore, plants can resist the increased malondialdehyde content and oxidative stress induced by PS and Cd exposure by increasing the activities of superoxide dismutase and peroxidase. Under the PS + Cd treatment, linear models showed that soil organic carbon and sucrase activity were the key variables affecting Cd uptake by plant shoots and roots, respectively. The results of the partial least squares path modeling further showed that PS indirectly affected Cd uptake by B. chinensis by significantly affecting the physicochemical properties of soil, Cd concentration, and enzyme activity. Our results provide a new perspective and an important reference for further understanding the effects of MPs on the bioavailability and fate of heavy metals.

Abstract:Nanoplastics (NPs) arouse increasing attention recently. Many studies have shown possible negative effects on plants exposed to NPs, but more data are needed on various species of plants to elucidate the potential phytotoxicity mechanisms of NPs. In this work, a pot experiment was conducted to analyze toxicity induced by polystyrene nanoplastics (PSNPs; particle size, 75.10 nm) at diverse doses (0.05, 0.1, 0.25, 0.5, 1.0 mg/g dry soil weight) to seedlings of garlic ( Allium sativum L.). The photosynthetic pigments, antioxidant enzyme activities, common phytohormones and styrene contents, and nutritional quality of leaves of A. sativum seedlings were determined. Results showed that the toxic effects of PSNPs on plants at 42 days after exposure were stronger than those at 28 days. Chlorophyll a, chlorophyll b, as well as carotenoids levels within A. sativum leaves treated with PSNPs for 42 days decreased, with the highest decreasing rates of 40.22%, 41.29%, and 27.38%, respectively. The malondialdehyde (MDA) level within A. sativum leaves was noticeably up-regulated at 0.5 and 1.0 mg/g of PSNPs. Furthermore, the guaiacol peroxidase (POD) and catalase (CAT) activities and the abscisic acid (ABA) contents increased significantly with increasing the concentrations of PSNPs. After exposure for 42 days, allicin, soluble sugar and soluble protein levels decreased with increasing the concentration of PSNPs. The highest styrene content in leaves of A. sativum was obtained for the treatment group of the 0.5 mg/g PSNPs, showing an increase of 23.73% relative to control. Collectively, PSNPs were translocated into the leaves of A. sativum from roots, significantly affected the photosynthetic pigments contents, and significantly induced oxidative stress in A. sativum . Furthermore, high doses of PSNPs exposure impaired the A. sativum 's nutritive quality. These research results can provide basic data for accurately evaluating the ecological toxicity of NPs on crops.

Abstract:Microplastics (MPs) occur widely in terrestrial ecosystems. However, information on the interaction of MPs with metals in terrestrial ecosystems is lacking in the literature. The present study investigated the effects of two types of MPs (high-density polyethylene (HDPE) and polystyrene (PS)) with different dosages (i.e., 0, 0.1%, 1%, and 10%) on the uptake and effects of Cd in maize plants grown in an agricultural soil. Results showed that addition of Cd at a 5 mg/kg caused inhibited plant growth and resulted in high Cd accumulation in plant tissues. Polyethylene alone showed no significant phytotoxic effects, but a high-dose of HDPE (10%) amplified Cd phytotoxicity. Polystyrene negatively affected maize growth and phytoxicity further increased in the presence of Cd. Both HDPE and PS caused soil diethylenetriaminepentaacetic acid (DTPA)-extractable Cd concentrations to increase but did not significantly affect Cd uptake into plant tissues. In the soil without Cd addition, HDPE decreased soil pH, while PS did not significantly alter soil pH. However, in the soil spiked with Cd, both HDPE and PS increased pH. Overall, impacts on plant growth and Cd accumulation varied with MP type and dose, and PS induced substantial phytotoxicity. In conclusion, co-occurring MPs can change Cd bioavailability, plant performance, and soil traits. Our findings highlight the ecological impacts that could occur from the release of MPs into soil.

Abstract:Nanoplastics, as a hot topic of novel contaminants, lack extensive concern in higher plants; especially the potential impact and mechanism of nanoplastics on legume crops remains elusive. In this study, the toxicity of polystyrene nanoplastics (PS-NPs, 200 nm) with diverse doses (control, 10, 50, 100, 200, 500 mg/L) to soybean and mung bean plants grown hydroponically for 7 d was investigated at both the macroscopic and molecular levels. The results demonstrated that the root length of both plants was markedly suppressed to varying degrees. Similarly, mineral elements (Fe, Zn) were notably decreased in soybean roots, consistent with Cu alteration in mung bean. Moreover, PS-NPs considerably elevated malondialdehyde (MDA) levels only in soybean roots. Enzyme activity data indicated mung bean exhibited significant damage only at higher doses of PS-NPs stress than soybean, implying mung bean is more resilient. Transcriptome analysis showed that PS-NPs stimulated the expression of genes associated with the antioxidant system in plant roots. Furthermore, starch and sucrose metabolism might play a key role in coping with PS-NPs to enhance soybean resistance, but the MAPK pathway was enriched in mung bean. Our findings provide valuable perspectives for an in-depth understanding of the performance of plants growing in waters contaminated by nanoplastics.

Abstract:With the large-scale use of plastic products in daily life and the progress of industrial production, it has had a serious impact on the soil environment. To identify the effect of heavy metal and microplastics on the development of plants, we examined the changes of physiological and biochemical indexes of Brassica chinensis var. chinensis under different concentrations of heavy metal cadmium and polyethylene stress with a pot experiment. We set three cadmium concentration gradients—0, 10, 20 mg/kg and three polyethylene concentration gradients—0, 18, 36 g/kg in the experiment. The results showed that both cadmium concentration and polyethylene concentration have significant effects on chlorophyll content, enzyme activity, and root development (except for the effect of polyethylene concentration on root volume, superoxide anion free radical content, and peroxidase activity), and the addition of polyethylene alleviated the stress effect of cadmium on biomass, root system, and chlorophyll content. The stress of cadmium and polyethylene reduced the leaf and root biomass of B. chinensis var. chinensis and inhibited the activities of enzymes related to chlorophyll synthesis, while the content of superoxide anion free radicals in the leaves increased, causing the enhancement of catalase activity. In addition, the content of photosynthetic pigments decreased, suggesting that the capture of light energy by B. chinensis var. chinensis will be reduced. These findings provide a new insight into the toxic effects of microplastics and heavy metals on B. chinensis var. chinensis , and further apply to the ecological risk assessment of microplastics and heavy metals on higher plants.

Abstract:The effects of polystyrene nanoplastics (PSNPs) on the physiological and molecular metabolism of corn seedlings were examined by treating corn (Zea mays L.) seedlings with 100, 300, and 500 nm diameter PSNPs and examining plant photosynthetic characteristics, antioxidant enzyme systems, and molecular metabolism. After 15 days of exposure to PSNPs with different particle sizes (50 mg·L-1), the photosynthetic characteristics of the plant remained stable, and the maximum photochemical quantum yield (Fv/Fm) and non-photochemical quenching coefficient (NPQ) had no significant effects. The root microstructure was damaged and the antioxidant enzyme system was activated, and the content of malondialdehyde (MDA) was significantly increased by 2.25-4.50-fold. In addition, 100 nm and 300 nm PSNPs exposure caused root superoxide dismutase (SOD) activity to increase 1.28-fold and 1.53-fold, and glutathione-peroxidase (GSH-PX) activity increased 1.30-fold and 1.58-fold. Non-targeted metabolomics analysis identified a total of 304 metabolites. Exposure to 100, 300, and 500 nm PSNPs led to the production of 85 (upregulated: 85, downregulated: 0), 73 (upregulated: 73, downregulated: 0), and 86 (upregulated: 84, downregulated: 2) differentially expressed metabolites, respectively, in the plant roots. Co-expressed differential metabolites accounted for 38.2% of the metabolites and indicated a metabolic imbalance primarily in organic acids and derivatives in the root system. The most significant enrichment pathways were those of alanine, aspartate, and glutamate metabolism. Overall, exposure to PSNPs of different particle sizes activated the root antioxidant enzyme system and interfered with plant basic metabolism. The alanine, aspartate, and glutamate metabolic pathways appear to be closely related to plant mechanisms for tolerance/detoxification of PSNPs.

Toxicity and fate of cadmium in hydroponically cultivated lettuce (Lactuca sativa L.) influenced by microplastics

Abstract:Although more attention has been paid to microplastics (MPs) pollution in environment, research on the synthetic influence of microplastic and heavy metals remains limited. To help fill this information gap, we investigated the adsorption behavior of virgin polyvinyl chloride microplastics (PVCMPs) (≤450 µm white spherical powder) on cadmium (II). The effects on seed germination, seedling growth, photosynthetic system, oxidative stress indicators of lettuce, and changes in Cd bioavailability were evaluated under Cd2+ (25 μmol/L), PVCMPs (200 mg/L), and PVCMP-Cd combined (200 mg/L + 25 μmol/L) exposures in hydroponic system. The results demonstrated that the PVCMPs effectively adsorbed Cd ions, which validated by the pseudo-second-order kinetic and the Langmuir isotherm models, indicating the sorption of Cd2+ on the PVCMPs was primary chemisorption and approximates monomolecular layer sorption. Compared to MPs, Cd significantly inhibits plant seed germination and seedling growth and development. However, Surprising improvement in seed germination under PVCMPs-Cd exposure was observed. Moreover, Cd2+ and MPs alone or combined stress caused oxidative stress with reactive oxygen species (ROS) including H2O2, O2- and Malondialdehyde (MDA) accumulation in plants, and substantially damaged to photosynthesis. With the addition of PVCMPs, the content of Cd in the leaves significantly (P<0.01) decreased by 1.76-fold, and the translocation factor and Cd2+removal rate in the water substantially (P<0.01) decreased by 6.73-fold and 1.67-fold, respectively in contrast to Cd2+ stress alone. Therefore, it is concluded the PVCMP was capable of reducing Cd contents in leaves, alleviating Cd toxicity in lettuce. Notably, this study provides a scientific foundation and reference for comprehending the toxicological interactions between microplastics and heavy metals in the environment.
Effect of foliar application of nanoparticles on growth, physiology, and antioxidant enzyme activities of lettuce (Lactuca sativa L.) plants under cadmium toxicity

Abstract:Nanotechnology has attracted the interest of scientists due to its wide range of application specifically in agriculture. Nanoparticles (NPs) may act as a promising materials to alleviate cadmium (Cd) stress in plants. This study aims to assess the impact of multiple nanoparticles including nSiO 2 (50 mg L −1 :100 mg L −1 ), nTiO 2 (20 mg L −1 :60 mg L −1 ), nZnO (50 mg L −1 :100 mg L −1 ), nFe 3 O 4 (100 mg L −1 :200 mg L −1 ), nCuO (50 mg L −1 :100 mg L −1 ), and nCeO 2 (50 mg L −1 :100 mg L −1 ) in combination with CdCl 2 (5 μM) to mitigate Cd toxicity in lettuce through foliar application in hydroponic solution. Current findings indicate that foliar application of nSiL + Cd (50 mg L −1 ), nZnL + Cd (50 mg L −1 ), and nTiL + Cd (20 mg L −1 ) is more effective in improving growth, biomass, root architecture, and elevated photosynthetic efficiency, which might be attributed to the increasing uptake of essential micronutrient (K, Mg, Ca, Fe, Zn) under Cd stress. Similarly, treatment with nanoparticles leads to reduced accumulation of ROS and MDA in lettuce, while enhancing the SOD, POD, CAT, and APX activities. The results showed that nanoparticles have high tolerance against Cd as depicted by the inhibition in Cd accumulation by 3.2–58% and 10–72% in roots as well as edible parts of lettuce, respectively. In addition, Cd alone reduces the morphological traits, antioxidant enzyme activity, and photosynthetic activity, while increasing the ROS, MDA, and Cd accumulation in lettuce. This comprehensive study suggests the role of nanoparticles in reducing Cd toxicity in lettuce, signifying their importance as stress mitigation agents. However, long-term pot, priming, and field trials are needed to identify the optimal nanoparticle for the lettuce under variable environmental conditions.

Abstract:Engineered nanoparticles (ENPs) and nanoplastics (NPs) are typical nanoparticles in terrestrial environments. Till now, few studies have compared their toxicity and mechanism to plants. Here we investigated the effects of CuO, nZVI ENPs and polystyrene (PS) NPs on lettuce growth, metabolic functions, and microbial community structure. Results showed that low concentrations of nanoparticles decreased root biomass and promoted photosynthetic indicators, whereas increased reactive oxygen species (ROS) were detected in roots exposed to high concentrations of nanoparticles. High-dose CuO ENP exposure significantly raised the MDA content by 124.6 % compared to CK, causing the most severe membrane damage in the roots among the three types of nanoparticles. Although linoleic acid metabolism was down-regulated, the roots alleviated CuO stress by up-regulating galactose metabolism. Uptake of PS by roots similarly caused ROS production and activated the oxidative stress system by altering amino acid and vitamin metabolism. Faster microbial responses to nanoparticles were observed in the nZVI and PS networks. The root toxicity was indirectly mediated by ion release, NP uptake, or ROS generation, ultimately impacting root cell metabolism, rhizospheric microorganism and plant growth. These findings provide theoretical basis for assessing environmental impact of nanoparticles and their possible ecological risks.

Abstract:As nanoplastics and persistent organic pollutants are broadly distributed in aquatic ecosystems and pose a potential threat to ecosystem, most pertinent studies have focused on aquatic animals, while studies on freshwater plants have been rarely reported. Therefore, we analyzed the single and combined toxicological impacts of various concentrations of 80 nm polystyrene nanoplastics (PS-NPs) including 0.5, 5, 10, and 20 mg/L and polychlorinated biphenyl-52 (PCB-52, 2,2',5,5'- tetrachlorobiphenyl) at 0.1 mg/L on the aquatic plant Spirodela polyrhiza (S. polyrhiza) after a 10-day hydroponic experiment. Laser confocal scanning microscopy (LCSM) showed the accumulation of PS-NPs mainly in the root surface and the lower epidermis of leaves, and the enrichment of PS-NPs was aggravated by the presence of PCB-52. PS-NPs at 10 mg/L and 20 mg/L alone or in combination with PCB-52 notably inhibited the growth of S. polyrhiza, reduced the synthesis of chlorophylls a and b, and increased the activities of superoxide dismutase (SOD) and peroxidase (POD) as well as malondialdehyde (MDA) levels, and induced osmotic imbalance (soluble protein and soluble sugar contents) (p < 0.05). However, a single treatment with low levels of PS-NPs had positive effects on the growth (0.5 mg/L) and photosynthetic systems (0.5, 5 mg/L) of S. polyrhiza, while co-exposure exacerbated the damaging impacts of PS-NPs on the antioxidant defense system of S. polyrhiza, which was more pronounced in the roots. Furthermore, correlation analysis revealed that plant growth parameters were positively correlated with chlorophyll a and b content and negatively correlated with soluble sugars, antioxidant enzymes, lipid peroxidation, and carotenoid content (p < 0.05). These results provide data to improve the understanding of the single and combined ecotoxicological effects of nanoplastics and polychlorinated biphenyls (PCBs) in aquatic plants and their application in phytoremediation measures.
