Abstract:The purpose of this study was to determine the capacity of indigenous microbes in tailings to degrade bitumen aerobically, and if acetate biostimulation further improved degradation. Fluid fine tailings, from Base Mine Lake (BML), were used as microbial inocula, and bitumen in the tailings served as a potential carbon source during the experiment. The tailings were capped with 0.22 mu m-filtered BML surface water with or without BML bitumen and acetate addition and incubated for 100 days at 20 degrees C. CO2 production and petroleum hydrocarbon reductions (50-70% for the biostimulation treatment) in the tailings were observed. DNA was extracted directly from the tailings, and increased bacterial density was observed by qPCR targeting the rpoB gene in the biostimulated group. 16 S rRNA sequencing was used to determine microbial composition profiles in each treatment group. The microbial communities indigenous to the tailings shifted after the bitumen was added. Acidovorax, Rhodoferax, Pseudomonas and Pseudox-anthomonas spp. significantly increased compared to the original microbial community and demonstrated tolerance to bitumen-based toxicity. The first three genera showed more potential for biostimulation treatment with acetate and may be important bitumen/hydrocarbon-degraders in an oil sands end pit lake environment. Naphthenic acid anaerobic biodegrading consortia enriched from pristine sediments underlying oil sands tailings ponds

Abstract:Seepage from oil sands tailings ponds (OSTP), which contain toxic naphthenic acids (NAs), can infiltrate into groundwater. Clay sediment layer beneath is a critical barrier for reducing the infiltration of NAs into the sand sediment layer, where groundwater channels reside. Biodegradation has great potential as a strategy for NAs removal, but little is known about NAs biodegradability and potential functional microbes in these pristine sediments. This study investigated the potential for anaerobic biodegradation of NAs by microbial consortia enriched from clay and sand sediments underlying OSTP, amended with either acid extracted organics or Merichem NAs, under nitrate- and sulfate-reducing conditions. Degradation of NAs only be detected after DOC concentration reached to steady state after 163 days. Microbial community analysis shows that different electron acceptors, sediment types, and NAs sources associated with specific microbial taxa and can explain 14.8, 13.9 % and 5% of variation of microbial community structures, respectively. The DOC and methane were the most important geochemical properties for microbial community variations. This study approved the potential capability of indigenous microbial communities from the pristine sediments in NA degradation, demonstrating the barrier function of pristine clay sediments underlying OSTP in prohibiting organic contaminants from entering into groundwater.

Abstract:The aerobic and anaerobic biodegradation of polyethylene glycols (PEGs) in a model wastewater was investigated employing sludge microbes from Xiamen Terylene plant. The effect of molecular weight (MW) on the aerobic/anaerobic biodegradation of PEGs was assessed by performing shake flasks/sealed flasks experiments, respectively, using organic model wastewater containing PEG 600, 6000 and 20,000. In aerobic biodegradation, although there were differences between the degradation processes, the three kinds of PEG were all degraded by about 80% in 5 days, regardless of the wide diversity of MW. In anaerobic biodegradation, about 50% degradation of PEG 600 in 9 days, 40% degradation of PEG 6000 in 10 days and 80% degradation of PEG 20,000 in 6 days were obtained. The effect of nutrition on anaerobic degradation was also investigated. The biodegradation rate of PEG 6000 increased sequentially in the organic, inorganic and enriched organic media. The latter two were similar to each other. In the enriched organic media PEG 6000 was degraded by 50% in 10 days and 70% in 14 days, while in the organic media the degradation rate was 23% in 10 days and 50% in 14 days. It was notable that the anaerobic microbes could use PEG 6000 as sole carbon source, and PEG MW up to 20,000 was efficiently biodegraded in this study, either in aerobic or anaerobic way.
Anaerobic biodegradation of partially hydrolyzed polyacrylamide in long-term methanogenic enrichment cultures from production water of oil reservoirs

Abstract:The increasing usage of partially hydrolyzed polyacrylamide (HPAM) in oilfields as a flooding agent to enhance oil recovery at so large quantities is an ecological hazard to the subsurface ecosystem due to persistence and inertness. Biodegradation of HPAM is a potentially promising strategy for dealing with this problem among many other methods available. To understand the responsible microorganisms and mechanism of HPAM biodegradation under anaerobic conditions, an enrichment culture from production waters of oil reservoirs were established with HPAM as the sole source of carbon and nitrogen incubated for over 328 days, and analyzed using both molecular microbiology and chemical characterization methods. Gel permeation chromatography, High-pressure liquid chromatography and Fourier-transformed infrared spectroscopy results indicated that, after 328 days of anaerobic incubation, some of the amide groups on HPAM were removed and released as ammonia/ammonium and carboxylic groups, while the carbon backbone of HPAM was converted to smaller polymeric fragments, including oligomers and various fatty acids. Based on these results, the biochemical process of anaerobic biodegradation of HPAM was proposed. The phylogenetic analysis of 16S rRNA gene sequences retrieved from the enrichments showed that Proteobacteria and Planctomycetes were the dominant bacteria in the culture with HPAM as the source of carbon and nitrogen, respectively. For archaea, Methanofollis was more abundant in the anaerobic enrichment. These results are helpful for understanding the process of HPAM biodegradation and provide significant insights to the fate of HPAM in subsurface environment and for possible bioremediation.

Abstract:High molecular weight (106–3 × 107 Da) polyacrylamide (PAM) is commonly used as a flocculant in water and wastewater treatment, as a soil conditioner, and as a viscosity modifier and friction reducer in both enhanced oil recovery and high volume hydraulic fracturing. These applications of PAM can result in significant environmental challenges, both in water management and in contamination of local water supplies after accidental spills. This paper provides a short review of current applications of high molecular weight PAM, including the potential for PAM degradation by chemical, mechanical, thermal, photolytic, and biological processes. Methods for treating wastewater containing partially degraded PAM are then discussed along with issues related to the potential toxicity and mobility of PAM in the environment after disposal or accidental release.

Abstract:Polyacrylamide (PAM) is generally employed in wastewater treatment processes such as sludge dewatering and therefore exists in the sludge. Furthermore, it degrades slowly and can deteriorate methane yield during anaerobic digestion (AD). The impact or fate of PAM in AD under thermophilic conditions is still unclear. This study mainly focuses on PAM degradation and enhanced methane production from PAM-added sludge during 15 days of thermophilic (55°C) AD compared to mesophilic (35°C) AD. Sludge and PAM dose from 10 to 50 g/kg TSS were used. The results showed that PAM degraded by 76% to 78% with acrylamide (AM) content of 0.2 to 3.3 mg/L in thermophilic AD. However, it degraded only 27% to 30% with AM content of 0.5 to 7.2 mg/L in mesophilic AD. The methane yield was almost 230 to 238.4 mL g VSS on the 8th day in thermophilic AD but was 115.2 to 128.6 mL/g VSS in mesophilic AD. Mechanism investigation revealed that thermophilic AD with continuous stirring not only enhanced PAM degradation but also boosted the organics release from the sludge with added PAM and gave higher methane yield than mesophilic AD.

Abstract:To deal with polyacrylamide (PAM) wastewater, granular sludge formed in glucose-fed sequencing batch reactors (SBR) was employed to cultivate PAM-degrading granules. Three replicated SBRs were operated with increasing PAM concentration in the influent from 67 to 670 mg L-1, and the hydraulic retention time was increased at the same time from 1 d to 6 d during the six-phase of the 43 d acclimation period. The well-acclimated PAM-degrading granules were different from the seeding granules in colour, mean diameter, biomass density and settle ability, and could use PAM as the sole carbon and nitrogen source. In the batch experiments, PAM degradation rate by granules was determined as 2.23 mg PAM g(-1) MLSS h(-1). According to the analysis of the intermediates of PAM biodegradation, PAM was degraded initially through hydrolysis of the amide group, and no acrylamide monomer was detected. With the help of LC/MS, the main intermediate was identified as polyacrylic acid with a low molecular weight. Therefore, the PAM-degrading granular sludge may be employed for removing PAM in the wastewater produced from tertiary oil recovery that uses polymeric flooding technology.

Abstract:Polyacrylamide (PAM) possesses unique characteristics, including high water solubility, elevated viscosity and effective flocculation capabilities. These properties make it valuable in various sectors like agriculture, wastewater treatment, enhanced oil recovery, and mineral processing industries, contributing to a continually expanding market. Despite its widespread use globally, understanding its environmental fate at the soil-water interface remains limited. This article aims to provide an overview of the occurrence, degradation pathways, toxicity, and risks associated with PAM in the bioenvironment. The findings indicate that various degradation pathways of PAM may occur in the bioenvironment through mechanical, thermal, chemical, photocatalytic degradation, and/or biodegradation. Through a series of degradation processes, PAM initially transforms into oligomers and acrylamide (AM). Subsequently, AM may undergo biodegradation, converting into acrylic acid (AA) and other compounds such as ammonia. Notably, among these degradation intermediates, AM demonstrates high biodegradability, and the bioaccumulations of both AM and AA are not considered significant. Ensuring the sustainable use of PAM necessitates a comprehensive understanding among policymakers, scholars, and industry professionals regarding PAM, encompassing its properties, applications, degradation pathways, toxic effect on humans and the environment, and relevant regulations. Additionally, this study offers insights into future priority research directions, such as establishing of a reliable source-to-destination supply chain system, determining the maximum allowable amount for PAM in farmlands, and conducting long-term trials for the PAM-containing demolition residues.

Application of an indigenous microorganisms-based fixed-bed GAC-biofilm reactor for passive and sustainable treatment of oil sands process water through combined adsorption and biodegradation processes

Abstract:In this study, a fixed-bed biofilm reactor (biofilter) was developed and applied for oil sands process water (OSPW) remediation by using granular activated carbon (GAC) as packing media. Using quantitative polymerase chain reaction (qPCR) detection, the total bacterial copy number (16S) in the GAC biofiltration system was found to reach a relatively stable level (1.3 ± 0.2 × 109 copies/g GAC) after 62 days of operation, and the thickness of biofilm on GAC surface was 26.7 ± 4.3 μm based on the scan of confocal laser scanning microscopy (CLSM). The established GAC-biofilter showed 95.4% naphthenic acids (NAs) removal from raw OSPW after 2 months of operation. The GAC-biofilter also showed 88.3% NAs removal after a long operation time (2 years), indicating its sustainable bioremediation capacity for OSPW. 16S and 18S rRNA gene-targeted metagenomic sequencing showed that the microbial community in the GAC biofilter had higher diversity and richness than that found in the sand biofilter which was used for OSPW treatment previously. Comamonadaceae and Saccharomycotina were found to be the dominant bacterial and fungal families in the GAC biofilter, respectively. Xenobiotic metabolism function of the microbial community may contribute significantly to the biodegradation of NAs. The GAC biofiltration process is a promising passive OSPW treatment approach that can be used in-situ.

Microbially-accelerated consolidation of oil sands tailings. Pathway I: changes in porewater chemistry

Abstract:Dispersed clay particles in mine tailings and soft sediments remain suspended for decades, hindering consolidation and challenging effective management of these aqueous slurries. Current geotechnical engineering models of self-weight consolidation of tailings do not consider microbial contribution to sediment behavior, however, here we show that microorganisms indigenous to oil sands tailings change the porewater chemistry and accelerate consolidation of oil sands tailings. A companion paper describes the role of microbes in alteration of clay chemistry in tailings. Microbial metabolism in mature fine tailings (MFT) amended with an organic substrate (hydrolyzed canola meal) produced methane (CH4) and carbon dioxide (CO2). Dissolution of biogenic CO2 lowered the pH of amended MFT to pH 6.4 vs. unamended MFT (pH 7.7). About 12% more porewater was recovered from amended than unamended MFT during 2 months of active microbial metabolism, concomitant with consolidation of tailings. The lower pH in amended MFT dissolved carbonate minerals, thereby releasing divalent cations including calcium (Ca(2+)) and magnesium (Mg(2+)) and increasing bicarbonate (HCO(-) 3) in porewater. The higher concentrations increased the ionic strength of the porewater, in turn reducing the thickness of the diffuse double layer (DDL) of clay particles by reducing the surface charge potential (repulsive forces) of the clay particles. The combination of these processes accelerated consolidation of oil sands tailings. In addition, ebullition of biogenic gases created transient physical channels for release of porewater. In contrast, saturating the MFT with non-biogenic CO2 had little effect on consolidation. These results have significant implications for management and reclamation of oil sands tailings ponds and broad importance in anaerobic environments such as contaminated harbors and estuaries containing soft sediments rich in clays and organics.
Mutagenicity of enantiomers of penbutolol.

Abstract:With a view to examine the effect of chirality and the cause of batch-to-batch variation in the mutagenicity of penbutolol, penbutolol enantiomers - isopenbutolol [R(+)-enantiomer] and penbutolol [S(-)-enantiomer] - and two batches of Betapressin were tested employing the Ames Salmonella tester strain TA98. The mutagenic activity of R(+)-enantiomer was found to be similar to that of a batch of penbutolol with a high content of this optical isomer. The pharmaceutical form of penbutolol, Betapressin, exhibited either less or equal mutagenic effectiveness to the S(-)-enantiomer. In the presence of the S9 mix, the mutagenicity of R(+)-enantiomer was only slightly affected in the low dose range of 40 to 160 micrograms/plate. A metabolite of penbutolol, (RS) l"-dehydropenbutolol, did not cause an increase in the number of revertants/plate.
Efficient Degradation of Polyacrylamide Using a 3-Dimensional Ultra-Thin SnO2-Sb Coated Electrode

Abstract:Polyacrylamide (PAM) is widely used in polymer flooding processes to increase oil recovery while the byproduct of PAM-containing wastewater is a serious environmental issue. In this study, electrochemical oxidation process (EAOP) was applied for treating PAM wastewater using a new type of 3-dimensional ultra-thin SnO2-Sb electrode. Nano-sized catalysts were evenly dispersed both on the surface and inside of a porous Ti filter forming nano-thickness catalytic layer that enhances the utilization and bonding of catalysts. This porous Ti electrode showed 20% improved OH center dot production and 16.3 times increased accelerated service life than the planar Ti electrode. Using this electrode to treat 100 mg L-1 PAM, the TOC removal efficiency reached over 99% within 3 h under current density of 20 mA cm(-2). The EAOP could fastly break the long-chain PAM molecules into small molecular intermediates. With the porous electrode treating 5 g L-1 PAM under current density of 30 mA cm(-2), EAOP reduced 94.2% of average molecular weight in 1 h and 92.0% of solution viscosity in 0.5 h. Moreover, the biodegradability of PAM solution was significantly improved as the solution BOD5/COD ratio raised from 0.05 to 0.41 after 4 h treatment. The degradation pathway of PAM was also investigated.

Abstract:The water-soluble polyacrylamide (PAM) can accumulate in ecosystems and cause serious environmental pollution. Biological approach achieves poor PAM degradation efficiency, due to the extreme resistance of PAM to the microbial metabolism. In the present work, the potential of bioelectrochemical system (BES) as an effective tool to degrade the PAM is adequately evaluated. The closed-circuit operation of BES obtains COD removal efficiencies of 29.2 and 33.6 % for the PAM and polyacrylic acid (PAA), respectively. In comparison, 4.3 and 2.7 % of COD are removed after the PAM and PAA are treated in the open-circuit BES, and 7.3 and 6.6 % are removed in the aerobic BES. These results suggest the bioelectricity generation is crucial to trigger the activity of bioanode for the effective degradation of PAM. Bioelectricity generation not only favors the decomposition of carbon backbone but also facilitates the hydrolysis of amide group in the side-chain of PAM. Microbial attack on the carbon backbone of PAM is proposed to initiate at the head-to-head linkage, resulting in the formation of ether bond within the shortened carbon chain. The Ignavibacterium sp. and phenotypically uncharacterized bacteria are classified as the dominant species on the anode of PAM-fed BES.
Uncovering Anaerobic Hydrocarbon Biodegradation Pathways in Oil Sands Tailings from Two Different Tailings Ponds via Metabolite and Functional Gene Analyses

Abstract:Oil sands tailings, a slurry of alkaline water, silt, clay, unrecovered bitumen, and residual hydrocarbons generated during bitumen extraction, are contained in ponds. Indigenous microbes metabolize hydrocarbons and emit greenhouse gases from the tailings. Metabolism of hydrocarbons in tailings ponds of two operators, namely, Canadian Natural Upgrading Limited (CNUL) and Canadian Natural Resources Limited (CNRL), has not been comprehensively investigated. Previous reports have revealed sequential and preferential hydrocarbon degradation of alkanes in primary cultures established from CNUL and CNRL tailings amended separately with mixtures of hydrocarbons (n-alkanes, iso-alkanes, paraffinic solvent, or naphtha). In this study, activation pathway of hydrocarbon biodegradation in these primary cultures was investigated. The functional gene analysis revealed that fumarate addition was potentially the primary activation pathway of alkanes in all cultures. However, the metabolite analysis only detected transient succinylated 2-methylpentane and 2-methylbutane metabolites during initial methanogenic biodegradation of iso-alkanes and paraffinic solvent in all CNUL and CNRL cultures amended with iso-alkanes and paraffinic solvent. Under sulfidogenic conditions (prepared only with CNUL tailings amended with iso-alkanes), succinylated 2-methylpentane persisted throughout incubation period of ~ 1100 days, implying dead-end nature of the metabolite. Though no metabolite was detected in n-alkanes- and naphtha-amended cultures during incubation, assA/masD genes related to Peptococcaceae were amplified in all CNUL and CNRL primary cultures. The findings of this present study suggest that microbial communities in different tailings ponds can biodegrade hydrocarbons through fumarate addition as activation pathway under methanogenic and sulfidogenic conditions.

Abstract:Mass polyacrylamide-containing wastewater from tertiary oil recovery needs to be treated in time. So the treatment technology focuses on biodegradation of PAM in this paper. The course that 7 strains of PAM biodegradation bacteria were separated from oilfield wastewater was introduced. The nutrient system of the bacteria was studied with Molecular biological identification method. The result showed that PAM could be the only source of N and C for the 7 strains, but the rate of growth was slow. The biodegradation rate of PAM accelerated when Liquid Paraffin and yeast extract were added in the experiment. Four of the 7 strains belong to Actinobacteria, two belong to Alphaproteobacteria, and one belongs to Bacillus. The homology of the 7 strains and the known bacteria in the data base was over 98%.

Abstract:Cationic polyacrylamide (cPAM) is a widely used flocculant to dewater sewage sludge (SS) for high-solids anaerobic digestion (AD), and its degradation is crucial since it would release toxic acrylamide (AM) once entering environment. Hydrothermal treatment (HTT) is an efficient method to enhance the AD efficiency of SS. However, the effects of cPAM on AD of SS and the degradation of cPAM during HTT-AD process have not be studied. The study showed cPAM at 20 mg/g TS increased methane yield of SS from 127.0 to 138.9 ml CH4/g TS in HTT-AD process, and the biodegradability of cPAM was 76.3%, which was much higher than that (7.4%) without HTT. In HTT-AD process, the enrichment of certain microbes (e.g. Gelria sp.) was observed, which might be related with cPAM degradation. HTT decreased the molecular weight (MW) of cPAM, and resulted in the production of 2-hydroxy-ethyl-trimethylammonium, ammonia, trimethylamine, and ethanol. Methane potential tests of the main HTT products also showed they were easily to be degraded. Overall, HTT-AD integrated process was an efficient method to reduce environmental risk of cPAM as well as increase energy output (biogas), and the study also provided insights into the degradation mechanism of cPAM during HTT.
Microbial degradation of naphthenic acids using constructed wetland treatment systems: metabolic and genomic insights for improved bioremediation of process-affected water

Abstract:Naphthenic acids (NAs) are a complex mixture of organic compounds released during bitumen extraction from mined oil sands that are important contaminants of oil sands process-affected water (OSPW). NAs can be toxic to aquatic organisms and, therefore, are a main target compound for OSPW. The ability of microorganisms to degrade NAs can be exploited for bioremediation of OSPW using constructed wetland treatment systems (CWTS), which represent a possible low energy and low-cost option for scalable in situ NA removal. Recent advances in genomics and analytical chemistry have provided insights into a better understanding of the metabolic pathways and genes involved in NA degradation. Here, we discuss the ecology of microbial NA degradation with a focus on CWTS and summarize the current knowledge related to the metabolic pathways and genes used by microorganisms to degrade NAs. Evidence to date suggests that NAs are mostly degraded aerobically through ring cleavage via the beta-oxidation pathway, which can be combined with other steps such as aromatization, alpha-oxidation, omega-oxidation, or activation as coenzyme A (CoA) thioesters. Anaerobic NA degradation has also been reported via the production of benzoyl-CoA as an intermediate and/or through the involvement of methanogens or nitrate, sulfate, and iron reducers. Furthermore, we discuss how genomic, statistical, and modeling tools can assist in the development of improved bioremediation practices.
Optimal floc structure for effective dewatering of polymer treated oil sands tailings

Abstract:Synthetic polymeric flocculants such as anionic polyacrylamides are widely used for dewatering mature fine tailings (MFT) generated from oil sands mining operations. Many studies have investigated the effects of polymer properties on dewatering efficiency. However, only a few studies have investigated the reasons for effective dewatering at optimum dosages from the perspective of understanding changes in the internal and external microstructure of the floc. In MFT, clay solids are dispersed in brine (along with trace amounts of bitumen) and are known to have a house-of-card like microstructural arrangement due to electrostatic stabilization. In this study, we investigated why there is maximum dewatering in the specific polyacrylamide (PAM) polymer-MFT system at optimum dosage. To understand this, we investigated the morphological changes in the house-of-cards microstructure in terms of porosity and floc density when flocculated at different polymer dosages as well as the corresponding floc strength, size, and re-flocculation tendencies. SEM imaging results showed that at the optimum dosage of 1000 ppm the internal floc microstructure had two different characteristic zones of reduced porosity and open channels, respectively, indicating that this arrangement could have led to an effective squeeze-out of water from the pores. Focused Beam Reflectance Microscopy results indicated that optimum dosage leads to effective dewatering due to flocs formed in the size range of 10–50 μm and 50–150 μm having reduced re-flocculation tendencies. Optimum dosage resulted in the least number of flocs and the reduction of flocs in the size range of 10–50 μm and < 10 μm was maximum while the corresponding increase of flocs in the size range of 50–150 μm was maximum as compared to other dosages. Additionally, the flocs at optimum dosage did not change in number for a given size range of flocs even after 1-hour stirring indicating strong floc formation. These observations are specific for the particular pairing and could form a basis for investigating other polymer-tailing systems.

Abstract:Anaerobic digestion provides a solution for the treatment of vegetable waste water (VWW), but there are currently limited targeted treatment methods available. Building upon previous studies, this research investigated the effects of polyacrylamide-modified magnetic micro-particles (MMP) on anaerobic digestion (AD) of VWW. Three variations of these particles were created by grafting anionic, cationic, and non-ionic polyacrylamide (PAM) onto the MMPs' surfaces, resulting in aPAM-MMP, cPAM-MMP, and nPAM-MMP, respectively. In AD experiments, the addition of aPAM-MMP notably enhanced the degradation of chemical oxygen demand (COD) in VWW. COD decreased to 1290 mg/L in the reactor with aPAM-MMP by day 12 and remained low, while the other reactors had COD concentrations of 4137.5, 5510, and 3010 mg/L on the same day, decreasing thereafter. This modification also improved the production and utilization of hydrogen gas and volatile fatty acids (VFAs), along with the conversion of methane. When tested for bioaffinity using fluorescent GFP-E.coli bacteria, the aPAM-MMP, cPAM-MMP, and nPAM-MMP demonstrated increases in fluorescence intensity by 51.66%, 36.13%, and 37.02%, respectively, compared to unmodified MMP when attached with GFP-E.coli. Further analyses of microbial community revealed that the reactor with aPAM-MMP had the highest microbial richness and enriched bacteria capable of organic matter degradation, such as Bacteroidota, Synergistota, Chloroflexi, Halobacterota phyla, and Parabacteroides, Muribaculaceae, and Azotobacter genera. In conclusion, our experiment verifies that APAM-MMP promotes anaerobic treatment of VWW and provides a novel reference point for enhancing VWW degradation.

Abstract:The wide utilization of poly dimethyl diallyl ammonium chloride (polyDADMAC) in industrial conditions leads to its accumulation in waste activated sludge (WAS), thereby affecting subsequent WAS treatment processes. This work investigated the interaction between polyDADMAC and WAS components from the perspective of anaerobic digestion (AD) performance and anaerobes adaptability variation. The results showed that polyDADMAC decreased the content of biodegradable organic substrates (i.e., soluble protein and carbohydrate) by binding with the functional groups and then settling to the solid phase, thus impeding the subsequent utilization. Higher concentrations of polyDADMAC prompted an initial protective response of excreting organic substrates into extracellular environment, but its toxicity to archaea was irreversible. Consequently, polyDADMAC inhibited the processes of AD and induced a 30 % reduction in methane production with 0.05 g polyDADMAC/g total suspended solid (TSS) addition. Changes in microbial community structure indicated that archaea involved in methane production (e.g., Anaerolineaceae sp. and Methanosaeta sp.) were inhibited when exposed to polyDADMAC. However, several adaptive bacteria with the ability of utilizing complex organics and participating in nitrogen cycle (e.g., Aminicenantales sp. and Ellin6067 sp.) were enriched with the above dosage. Specifically, the decreased abundance of genes relevant to methane metabolism pathway (i.e., mer and cdh) and increased abundance of genes involved in metabolism of cofactors and vitamins (e.g., nad and thi) indicated the toxicity of polyDADMAC and the irritant response of microflora. Moreover, polyDADMAC underwent degradation in AD system, resulting in a 12 % reduction in 15 days, accompanied by an increase in the -NO2 functional group. In general, this study provided a thorough understanding of the interaction between polyDADMAC and WAS components, raising concerns regarding the elimination of endogenous pollutants during AD.
