Abstract:Aims: The effects of exogenously applied salicylic acid (SA) on gas exchange characteristics, photosynthetic pigments and chloroplast ultrastructure were investigated in gerbera at their reproductive stage under salt-stressed conditions. Methodology: A pot experiment was conducted under glasshouse conditions at the Zhejiang University, Hangzhou, China, (30° N/120° E) between February 2008 and March 2009.Plants, pretreated with foliar applications of 0, 0.5, and 1.0 mmoldm-3 SA at the onset of flower initiation were irrigated with 100 mmoldm-3NaCl(aq) for two weeks, starting after three days from the SA pretreatment. Control did not receive either NaCl or SA.Photosynthetic rate, gas exchange, photosynthetic pigments content and chloroplast ultrastructure were investigated against treatments. All data were subjected to analysis of variance (ANOVA) and Generalized Linear Model (GLM) using SAS statistical software. Pearson’s correlation test was carried out to study the relationships among the parameters. The means were compared using Duncan’s multiple range test (DMRT). For all the tests, P< .05 was considered statistically significant. Results: Salt stress adversely affected the gas exchange characteristics, photosynthetic pigment contents and chloroplast ultrastructure. SA application significantly increased the net photosynthesis, stomatal conductivity, intra-cellular CO2 content and transpiration rate but decreased the stomatal limitation, compared to those of untreated salt-stressed plants. Further, the enhanced photosynthetic pigment contents and notably undamaged chloroplast ultrastructure were evident of the ameliorative effects of SA on photosynthetic system under salt stress. Of the two concentrations tested, 0.5 mmoldm-3 SA concentration seemed to have greater effect throughout the experiment showing no significant variation from control in some attributes (chlorophyll contents and chloroplast ultrastructure). Conclusion: Responses of plants pretreated with SA spraying and significant correlation among them plausibly suggest SA-induced enhancement of photosynthetic system as another target for conferring salt tolerance in crop plants.
Exogenous Salicylic Acid Alleviates Salt Stress by Improving Leaf Photosynthesis and Root System Architecture in Cucumber Seedlings

Abstract:Exogenous salicylic acid (SA) can alleviate salt stress in plants. Previous researches have been focused on SA's role in leaf morphological and physiological characteristics, however, little information is available on its role in both leaf photosynthesis and root system architecture (RSA). A hydroponic study was conducted to analyze biomass, leaf photosynthesis, RSA parameters and expression of RSA genes in cucumber seedlings grown under four treatments, including control (CK), 50 mM NaCl (NaCl), 0.3 mM SA (SA) and 50 mM NaCl plus 0.3 mM SA (NaCl + SA). The results showed that compared with CK, both NaCl and SA treatments severely reduced relative growth rate (RGR) and leaf photosynthetic parameters (F-v/F-m, Phi(II) and Pn). However, compared with NaCl treatment, NaCl + SA treatment significantly increased RGR, leaf photosynthetic (F-v/F-m, Phi(II) and Pn) and RSA (total root length, root surface area, number and total length of primary and secondary LR) parameters. In addition, compared with CK, although both NaCl and SA treatments down-regulated expression of GL2 gene and up-regulated expression of RHD2 gene, NaCl + SA treatment notably up-regulated expression of NAC1, NAC2, GL2, EXP and down-regulated expression of RHD2. It is concluded that exogenous SA alleviated salt stress via effectively promoting leaf photosynthesis and RSA in cucumber seedlings. SA-mediated increment in expression of RSA genes, total length and number of primary and secondary LR could contribute to the improved RSA.

Sc., 16: 49-58 Responses of photosynthesis rate, stomatal conductance, chlorophyll fluorescence characteristics and chlorophyll content of maize (Zea mays L.) seedlings under combined salinity-alkalinity stress were examined. Investigation revealed that the net photosynthesis rate (P-n) and stomatal conductance (g(s)) decreased rapidly with increasing of salinity-alkalinity stress. The intercellular CO2 concentration (C-i) decreased at low salinity-alkalinity and increased at high salinity-alkalinity. The maximal efficiency of photosystem II (PSII) photochemistry (F-V/F-M) decreased only at high salinity-alkalinity. The comprehensive photosynthesis performance index (PIABS) gradually decreased with increasing of salinity-alkalinity concentration. The JIP test showed that the electric transfer of donor side and acceptor side of PSII in maize seedlings were inhibited by high salinity-alkalinity stress. Stomatal limitation is the main reason of decreased photosynthesis rate at low salinity-alkalinity, and non-stomatal limitation, i.e. decreased photosynthesis activity in PSII plays an important role in decreased photosynthesis rate at high salinity-alkalinity.

Abstract:Soil salinization has been considered as a major environmental threat to plant growth. Different types of salt in saline soil have different effects on germination and seedling growth. Effect of NaCl on germination and seedling establishment in Suaeda liaotungensis have been reported, but its response to alkali stress remains unclear. Our results showed that brown seeds had higher germination rate, however, black seeds had higher germination recovery percentage under alkali stress. Na 2 CO 3 had stronger inhibitory effect on germination and seedling growth than NaHCO 3 . As the concentration of alkali stress increased, the ROS level of brown seeds gradually ascended, while that of black seeds decreased first and then ascended. MDA content of dimorphic seeds significantly increased under alkali stress. The trend of SOD, POD and CAT activity between dimorphic seeds was similar under the same type of alkali stress. Alkali stress enhanced proline content of dimorphic seeds, and dimorphic seeds in NaHCO 3 solution had higher proline content than Na 2 CO 3 solution. Moreover, radicle and shoot tolerance indexes of seedlings in NaHCO 3 solution were significantly higher than that of Na 2 CO 3 solution. Under strong alkali stress, seedlings in NaHCO 3 solution had significantly lower ROS level and MDA content as well as higher antioxidant enzyme activity than Na 2 CO 3 solution. This study comprehensively compared the morphological and physiological characteristics in germination and seedlings to better reveal the saline-alkali tolerance mechanisms in S. liaotungensis .

Abstract:Sunflower seedlings were treated under 30 different conditions of alkalinity and salinity, which were established by mixing NaCl, NaHCO3, Na2SO4, and Na2CO3, at various proportions. The treatments included a salt concentration range of 50–250mmol and pH values from 7.12 to 10.72. Several physiological indices of seedlings stressed—including relative growth rate (RGR), leaf area, electrolyte leakage rate, proline content, citric acid content, and contents of Na+ and K+—were determined to analyze the characteristics of the stresses due to the salt–alkali mixes and their main stress factors.The results showed that the physiological responses of sunflower closely correlated not only with salinity (the total concentration of stress salt) but also with the pH (or alkalinity) of the treatment solution. RGR, leaf area, and of K+ content decreased with increasing salinity and pH. Electrolyte leakage rate, proline content, citric acid content, and Na+ content increased with increasing salinity and pH. The deleterious effects of a high pH value or salinity alone were significantly less than those of high pH in combination with salinity. This result suggested that for a salt–alkali mix stress, a reciprocal enhancement between salt stress and alkali stress was a characteristic feature.The buffer capacity of the treatment solution was taken as a stress factor in order to simplify the stress factor analysis. The results of the statistical analysis showed that for the stress factors of the salt–alkali mix stress, [CO32−] and [HCO3−] could be fully represented by the buffer capacity; [Na+] could be fully represented by salinity; whereas [SO42−] was negligible. Therefore, four factors, salinity, buffer capacity, pH and [Cl−], could reflect all of stress factors. Perfect linear correlations were observed between all strain indices and the four stress factors. However, the effects of the four stress factors on the strain indices were significantly different in magnitude. Buffer capacity and salinity were dominant factors for all strain indices. Thus, it is reasonable to consider the sum of salinity plus buffer capacity as the strength value of salt–alkali mix stress. Furthermore, the relationships between different strain indices and various stress factors were shown to be different.

Effects of Salt Stress on Photosynthetic Characters and Chloroplast Ultra-structure of Almond

Abstract:150,300 and 350 mmol/L NaCl and Na_2SO_4 were used to treat the seed-originated seedlings of Amygdalus communis L.and A.communis var.persiooides in greenhouse.Then the chlorophyll contents,net photosynthetic rates and stomatal conductance and intercellular CO_2 concentrations in their leaves were determined and the ultra-structures of the leaves were observed.It was shown that(1) both Amygdalus communis L.and A.communis var.persiooides presented the highest chl a and chl b contents at low concentrations of the salts and the lowest chl a and chl b contents at higher concentrations of the salts,and the chlorophyll contents with Na_2SO_4 were lower than those with NaCl;(2) The net photosynthetic rates of Amygdalus communis L.and A.communis var.persiooides declined with increased salt concentration and the decrement in net photosynthetic rate was larger in Amygdalus communis L.;(3) The intercellular CO_2 concentration of A.communis var.persiooides increased with increased salt concentrations and the variation did not appeared regular;(4) The leaf stomatal conductance of the two declined with increased salt concentrations;(5) The salt stresses resulted in the chloroplast grana and stroma distorted and the choloroplast thylakoids swelling and the effects become intensified with increased salt concentrations,the chloroplasts swelling from elliptical shape to round shape and the chloroplast membrane disintegrating,and the chloroplasts of Amygdalus communis L.were more sensitive to the salt stresses.These results indicated that two salt stresses had different action mechanisms to damage the plants.A.communis var.persiooides was more tolerant to the salt stresses than Amygdalus communis L.

Abstract:High salinity is one of the major abiotic stresses that limit crop production. Salicylic acid (SA) has been shown to ameliorate the adverse effects of environmental stress on plants. However, the molecular basis of salicylic acid-mediated salinity tolerance in kenaf is still unclear. To uncover the toxic alleviation effects of SA on kenaf salt stress, the morphological, physiological indexes, and transcriptomic regulating were assayed under NaCl stress with (or without) SA pretreated. The result showed that application of exogenous SA significantly alleviated the repressive effects of kenaf under salt stress, including agronomic traits, antioxidant enzyme systems, and several important pathways and involved differentially expressed genes (DEGs). In addition, some transcription factors (TFs) such as NAC , MYB , bHLH , ERF and enzyme activity-related genes such POD were also significantly changed with exogenous SA application. Virus‐Induced Gene Silencing (VIGS) of a differentially expressed TFs , HcNAC29, reduced salt tolerance in kenaf. Integrating physiological and transcriptomic analyses, a number of important pathways were found to be important for SA-mediated salinity tolerance in kenaf, including phenylpropanoid biosynthesis, flavonoid biosynthesis, plant hormone signaling pathway, and some transcription factors such as NAC . The data presented here may be useful in further elucidating the multiple regulatory roles of SA in plant responses to abiotic stresses.

Abstract:This work mainly investigated the interferences of NaCl treatment to major metabolite accumulation, individual flavonoid biosynthesis and photosynthetic activity in Tetrastigma hemsleyanum. The results showed that the accumulation of phenolic acids, steroids and polysaccharides in T. hemsleyanum was significantly enhanced after the treatment of 50 or 100 mM NaCl for 12 days, and after 150 mM NaCl treatment for 3 days, the total content of flavonoids reached the highest value of 219.5 μg·g‐1−1 and was higher by 3.8 times in comparison with the control (CK). In addition, the salt stress would generally enhance the reaction selectivity of Path II (RPSII) to produce flavonol derivatives (Rut, IsoQ, Km3rut, Ast, Qu, Km) and reduce the reaction selectivity of Path I (RPSI) to produce flavone derivatives (IsoO, Or, Ap), and after the treatment of 150 mM NaCl for 3 days, the RPSII and the RPSI reached the highest level of 0.785 and the lowest level of 0.215, respectively. Moreover, salt stress demonstrated significant and diverse effects on photosynthetic characteristics of T. hemsleyanum. Compared with CK, the values of Pn, Cond, Tr, Fv/Fm, Y(Ⅱ) and qP increased first and then declined with increasing NaCl concentration and stress time, and the highest level of Pn, Fv/Fm and Y(II)occurred after 50 mM NaCl treatment. These findings clearly showed that 50 mM NaCl treatment results in favorable changes in photosynthesis and secondary metabolites accumulation in T. hemsleyanum.

Abstract:Abstract Background Application of exogenous glycine betaine (GB) and exogenous salicylic acid (SA) mitigates the adverse effects of salinity. Foliar spraying with exogenous GB or SA alleviates salt stress in plants by increasing leaf gas exchange and stimulating antioxidant enzyme activity. The effects of foliar application of exogenous GB and SA on the physiology and biochemistry of cotton seedlings subjected to salt stress remain unclear. Results Results showed that salt stress of 150 mM NaCl significantly reduced leaf gas exchange and chlorophyll fluorescence and decreased photosynthetic pigment quantities and leaf relative water content. Foliar spray concentrations of 5.0 mM exogenous GB and 1.0 mM exogenous SA promoted gas exchange and fluorescence in cotton seedlings, increased quantities of chlorophyll pigments, and stimulated the antioxidant enzyme activity. The foliar spray also increased leaf relative water content and endogenous GB and SA content in comparison with the salt-stressed only control. Despite the salt-induced increase in antioxidant enzyme content, exogenous GB and SA in experimental concentrations significantly increased the activity of glutathione reductase, ascorbate peroxidase, superoxide dismutase, catalase and peroxidase, and decreased malondialdehyde content under salt stress. Across all experimental foliar spray GB and SA concentrations, the photochemical efficiency of photosystem II ( F V / F M ) reached a peak at a concentration of 5.0 mM GB. The net photosynthetic rate ( P n ) and F V / F M were positively correlated with chlorophyll a and chlorophyll b content in response to foliar spraying of exogenous GB and SA under salt stress. Conclusions We concluded, from our results, that concentrations of 5.0 mM GB or 1.0 mM SA are optimal choices for mitigating NaCl-induced damage in cotton seedlings because they promote leaf photosynthesis, increase quantities of photosynthetic pigments, and stimulate antioxidant enzyme activity. Among, 5.0 mM GB and 1.0 mM SA, the best performance in enhancing endogenous GB and SA concentrations was obtained with the foliar application of 1.0 mM SA under salt stress.

Plant growth regulators mitigate oxidative damage to rice seedling roots by NaCl stress

Abstract:The aim of this experiment was to investigate the effects of exogenous sprays of 5-aminolevulinic acid (5-ALA) and 2-Diethylaminoethyl hexanoate (DTA-6) on the growth and salt tolerance of rice ( Oryza sativa L.) seedlings. This study was conducted in a solar greenhouse at Guangdong Ocean University, where ‘Huanghuazhan’ was selected as the test material, and 40 mg/L 5-ALA and 30 mg/L DTA-6 were applied as foliar sprays at the three-leaf-one-heart stage of rice, followed by treatment with 0.3% NaCl (W/W) 24 h later. A total of six treatments were set up as follows: (1) CK: control, (2) A: 40 mg⋅ L −1 5-ALA, (3) D: 30 mg⋅ L −1 DTA-6, (4) S: 0.3% NaCl, (5) AS: 40 mg⋅ L −1 5-ALA + 0.3% NaCl, and (6) DS: 30 mg⋅ L −1 DTA-6+0.3% NaCl. Samples were taken at 1, 4, 7, 10, and 13 d after NaCl treatment to determine the morphology and physiological and biochemical indices of rice roots. The results showed that NaCl stress significantly inhibited rice growth; disrupted the antioxidant system; increased the rates of malondialdehyde, hydrogen peroxide, and superoxide anion production; and affected the content of related hormones. Malondialdehyde content, hydrogen peroxide content, and superoxide anion production rate significantly increased from 12.57% to 21.82%, 18.12% to 63.10%, and 7.17% to 56.20%, respectively, in the S treatment group compared to the CK group. Under salt stress, foliar sprays of both 5-ALA and DTA-6 increased antioxidant enzyme activities and osmoregulatory substance content; expanded non-enzymatic antioxidant AsA and GSH content; reduced reactive oxygen species (ROS) accumulation; lowered malondialdehyde content; increased endogenous hormones GA3, JA, IAA, SA, and ZR content; and lowered ABA content in the rice root system. The MDA, H 2 O 2 , and O 2− contents were reduced from 35.64% to 56.92%, 22.30% to 53.47%, and 7.06% to 20.01%, respectively, in the AS treatment group compared with the S treatment group. In the DS treatment group, the MDA, H 2 O 2 , and O 2− contents were reduced from 24.60% to 51.09%, 12.14% to 59.05%, and 12.70% to 45.20%. In summary, NaCl stress exerted an inhibitory effect on the rice root system, both foliar sprays of 5-ALA and DTA-6 alleviated damage from NaCl stress on the rice root system, and the effect of 5-ALA was better than that of DTA-6.

Abstract:Salt and alkali stress are considered major abiotic stresses in agriculture. Tea plant ( Camellia sinensis ), an acidophilic economic crop, is seriously affected by salt and alkali stress, which severely restricts its widespread planting. However, the mechanisms underlying the response of tea plants to salt and alkali stress remain unclear. To understand the physiological and molecular responses of tea plants to salt and alkali stress, tea plants were treated with NaCl and NaHCO 3 to study the effects of salt and alkali stresses, respectively. Tea plants exhibited different symptoms under the NaCl and NaHCO 3 treatments. The leaves of tea plants suffered varying degrees of damage, and concentrations of the quality components epigallocatechin gallate, caffeine, and theanine in the leaves decreased significantly under the NaCl treatment. However, after NaHCO 3 treatment, epigallocatechin and theanine levels were significantly reduced. Photosynthesis and antioxidant enzymes changed significantly to varying degrees under both the NaCl and NaHCO 3 treatments. The stomata closed under both NaCl and NaHCO 3 treatments. After the transcriptome analysis of tea samples treated for one, three, and 7 days with NaCl or NaHCO 3 , a large number of differentially expressed genes related to photosynthesis and the antioxidant system were identified. Analysis of the promoters of key differentially expressed genes revealed many light-responsive, hormone-responsive, and stress-responsive elements, and many corresponding upstream transcription factors were also differentially expressed. These results provide a basis for understanding the physiological and molecular responses of tea plants to salt and alkali stress.

Abstract:Xanthoceras sorbifolium Bunge is priced for its medical and energetic values. The species also plays a key role in stabilizing ecologically fragile areas exposed to excess soil salinity. In this study, the effects of salinity on the growth, physiological, and photosynthetic parameters of X. sorbifolium Bunge were investigated. The X. sorbifolium seedlings were subjected to five salt treatments: 0 (control, CK), 70, 140, 210, and 280 mM of sodium chloride (NaCl) solutions. NaCl caused a decrease in plant height, specific leaf area, biomass, and root parameters. Leaf wilting and shedding and changes in root morphology, such as root length, root surface area, and root tips were observed. This study found that X. sorbifolium is tolerant to high salinity. Compared with the CK group, even if the concentration of NaCl was higher than 210 mM, the increase of the relative conductivity was also slow, while intercellular CO 2 concentration had a similar trend. Moreover, NaCl stress caused an increase in the malondialdehyde (MDA), soluble proteins, and proline. Among the enzymes in the plant, the catalase (CAT) activity increases first and decreased with the increase in the intensity of NaCl stress, but the salt treatment had no significant effect on superoxide dismutase (SOD) activity. The peroxidase (POD) showed an increasing trend under salt stress. It was found that the photosynthesis of X. sorbifolium was notably impacted by saline stress. NaCl toxicity induced a noticeable influence on leaf net photosynthetic rate (Pn), stomatal conductance (Gs), intercellular CO 2 concentration (Ci), transpiration rate (E), and water use efficiency (Wue). As salt concentration increased, the content of chlorophyll decreased. It can be found that a low concentration of NaCl induced the increase of photosynthetic capacity but a high-intensity exposure to stress resulted in the reduction of photosynthetic efficiency and SOD activity, which had a positive correlation. In summary, salt-induced ionic stress primarily controlled root morphology, osmotic adjustment, and enzyme activities of salt-treated X. sorbifolium leaves, whereas the low salt load could, in fact, promote the growth of roots.

Abstract:Salt stress is one of the major adverse factors affecting plant growth and crop production. Rapeseed is an important oil crop, providing high-quality edible oil for human consumption. This experiment was conducted to investigate the effects of salt stress on the phenotypic traits and physiological processes of rapeseed. The soil salinity was manipulated by setting three different levels: 0 g NaCl kg−1 soil (referred to as S0), 1.5 g NaCl kg−1 soil (referred to as S1), and 3.0 g NaCl kg−1 soil (referred to as S2). In general, the results indicated that the plant height, leaf area, and root neck diameter decreased with an increase in soil salinity. In addition, the biomass of various organs at all growth stages decreased as soil salinity increased from S0 to S2. The increasing soil salinity improved the distribution of biomass in the root and leaf at the seedling and flowering stages, indicating that rapeseed plants subjected to salt stress during the vegetative stage are capable of adapting their growth pattern to sustain their capacity for nutrient and water uptake, as well as leaf photosynthesis. However, as the soil salinity increased, there was a decrease in the distribution of biomass in the pod and seed at the maturity stage, while an increase was observed in the root and stem, suggesting that salt stress inhibited carbohydrate transport into reproductive organs. Moreover, the C and N accumulation at the flowering and maturity stages exhibited a reduction in direct correlation with the increase in soil salinity. High soil salinity resulted in a reduction in the C/N, indicating that salt stress exerted a greater adverse effect on C assimilation compared to N assimilation, leading to an increase in seed protein content and a decrease in oil content. Furthermore, as soil salinity increased from S0 to S2, the activity of superoxide dismutase (SOD) and catalase (CAT) and the content of soluble protein and sugar increased by 58.39%, 33.38%, 15.57%, and 13.88% at the seedling stage, and 38.69%, 22.85%, 12.04%, and 8.26% at the flowering stage, respectively. In summary, this study revealed that salt stress inhibited C and N assimilation, leading to a suppressed phenotype and biomass accumulation. The imbalanced C and N assimilation under salt stress contributed to the alterations in the seed oil and protein content. Rapeseed had a certain degree of salt tolerance by improving antioxidants and osmolytes.

Abstract:The influence of salt (0,200 and 400 mmol/L NaCl;0,200 and 400 mmol/L KCl)on growth,ion accumulation,chlorophyll fluorescence and gas exchange in leaves and antioxidant levels was assessed in Suae-da salsa plants from different habitats;intertidal and inland saline.200 mmol/L NaCl significantly increased shoot fresh weight and dry weight and succulence of S .salsa plants,while 400 mmol/L NaCl and all KCl treat-ments resulted in reduced growth.The concentrations of Na+ and Cl- were increased by NaCl treatments while the concentration of K+ significantly decreased.Similarly KCl treatments resulted in increasing K+ and Cl-concentrations accompanied by a marked reduction in Na+ concentration.The 200 mmol/L NaCl treatment had no effect on net photosynthesis,photosynthetic oxygen evolution rate,F v/F m and φPSⅡin S .salsa,but KCl treatments resulted in a significant decrease in these traits.In addition,200 mmol/L NaCl resulted in a signifi-cant increase in the SOD activity whereas 400 mmol/L NaCl and all KCl treatments markedly decreased SOD activity.Interestingly,the POD activity in S .salsa was significantly increased by all NaCl and KCl treat-ments.Shoot fresh weight and dry weight,succulence,net photosynthesis,photosynthetic oxygen evolution rate and ion concentration (Na+ and Cl- )of S .salsa plants from the intertidal zone were lower than that those from an inland saline areas.The salt stress effect of KCl was greater than that of NaCl.Saline stress in S .sal-sa plants from saline inland habitats was significantly greater than plants from an intertidal zone.
Canola (Brassica napus) enhances sodium chloride and sodium ion tolerance by maintaining ion homeostasis, higher antioxidant enzyme activity and photosynthetic capacity fluorescence parameters

Abstract:Under salt stress, plants are forced to take up and accumulate large amounts of sodium (Na+ ) and chloride (Cl- ). Although most studies have focused on the toxic effects of Na+ on plants, Cl- stress is also very important. This study aimed to clarify physiological mechanisms underpinning growth contrasts in canola varieties with different salt tolerance. In hydroponic experiments, 150mM Na+ , Cl- and NaCl were applied to salt-tolerant and sensitive canola varieties. Both NaCl and Na+ treatments inhibited seedling growth. NaCl caused the strongest damage to both canola varieties, and stress damage was more severe at high concentrations of Na+ than Cl- . High Cl- promoted the uptake of ions (potassium K+ , calcium Ca2+ ) and induced antioxidant defence. Salt-tolerant varieties were able to mitigate ion toxicity by maintaining lower Na+ content in the root system for a short period of time, and elevating magnesium Mg2+ content, Mg2+ /Na+ ratio, and antioxidant enzyme activity to improve photosynthetic capacity. They subsequently re-established new K+ /Na+ and Ca2+ /Na+ balances to improve their salt tolerance. High concentrations of Cl salts caused less damage to seedlings than NaCl and Na salts, and Cl- also had a positive role in inducing oxidative stress and responsive antioxidant defence in the short term.

Abstract:BACKGROUND: Salt stress is a major factor limiting plant growth and productivity. Salicylic acid (SA) has been shown to ameliorate the adverse effects of environmental stress on plants. To investigate the protective role of SA in ameliorating salt stress on Torreya grandis (T. grandis) trees, a pot experiment was conducted to analyze the biomass, relative water content (RWC), chlorophyll content, net photosynthesis (Pn), gas exchange parameters, relative leakage conductivity (REC), malondialdehyde (MDA) content, and activities of superoxide dismutase (SOD) and peroxidase (POD) of T. grandis under 0.2% and 0.4% NaCl conditions with and without SA.METHODOLOGY/PRINCIPAL FINDINGS: The exposure of T. grandis seedlings to salt conditions resulted in reduced growth rates, which were associated with decreases in RWC and Pn and increases in REC and MDA content. The foliar application of SA effectively increased the chlorophyll (chl (a+b)) content, RWC, net CO2 assimilation rates (Pn), and proline content, enhanced the activities of SOD, CAT and POD, and minimized the increases in the REC and MDA content. These changes increased the capacity of T. grandis in acclimating to salt stress and thus increased the shoot and root dry matter. However, when the plants were under 0% and 0.2% NaCl stress, the dry mass of the shoots and roots did not differ significantly between SA-treated plants and control plants.CONCLUSIONS: SA induced the salt tolerance and increased the biomass of T. grandis cv. by enhancing the chlorophyll content and activity of antioxidative enzymes, activating the photosynthetic process, and alleviating membrane injury. A better understanding about the effect of salt stress in T. grandis is vital, in order gain knowledge over expanding the plantations to various regions and also for the recovery of T. grandis species in the future.

Abstract:Abstract This pot experiment was to evaluate how salts (NaCl, Na 2 SO 4 ) and alkali (Na 2 CO 3 +NaHCO 3 ) affect the physiological and biochemical characteristics during the seedling stage of two cotton cultivars (salt-tolerant, L24; salt-sensitive, X45). Salt and alkali stress reduced seedling emergence rate, relative biomass, and chlorophyll content, however, the REC and MDA content increased. Salt and alkali stress increased markedly superoxide dismutase (SOD) activity. Peroxidase (POD) activity increased first and then decreased as the increase of salt and alkali stress. Catalase (CAT) activity initially increased and then decreased as NaCl stress increased. In addition, the SOD activity, REC, and MDA content was markedly higher in salt stress than that in alkali stress. The proline content of L24 was higher than that of X45 under salt and alkali stress. However, glycine betaine and soluble sugar content of L24 was lower than that of X45 under alkali stress. The REC and MDA content of L24 were lower than those of X45, however, the relative biomass, chlorophyll content, SOD, POD, CAT, and Pro were higher than those of X45. In conclusion, salt tolerant cotton cultivars may possess a superior protection effect by increasing antioxidant enzymes activity under salt and alkali stress.

Abstract:In order to compare the stress effect of NaCl, Na+-salt and Cl--salt on the physiological responses in Chrysanthemum, the seedlings of Chrysanthemum okiense and C. ornatum were cultured in 0.75 fold diluted Hogland solution supplemented with NaCl, Na+(without Cl-) and Cl-(without Na+) with the osmotic potential of -0.64 Mp. Plants cultured in 0.75 fold diluted Hogland solution were used as control. The plant biomass, chlorophyll content, malondialdehyde (MDA) content, proline content and electrolyte leakage were investigated in the control plant and salt stressed plants. The plant biomass and chlorophyll content were decreased, while MDA content, proline content and electrolyte leakage were increased under either NaCl, Na+-salt or Cl--salt stress. The changes in five parameters were obviously affected by Cl--salt, moderately by NaCl, and lesser by Na+-salt in turn. C. ornatum was more affected by the three kinds of salt stress than C. okiense in terms of the five parameters. In sum, Cl- is the main cause for salt stress compared to Na+, and C. okiense is more salt tolerant than C. ornatum.
The Enhancement of Salt Stress Tolerance by Salicylic Acid Pretreatment in Arabidopsis Thaliana

Abstract:Salicylic acid (SA) is an important plant hormone involved in the activation of defense responses against environmental stresses. However, there are still large of unsolved mysteries about how SA pretreatment affects the establishment of plant stress tolerance. In this study, application of SA at different concentrations and different times were conducted to investigate their effects on the response of Arabidopsis seedlings to salt stress. The pretreatment with 10 or 20 μM SA for more than 6 h promoted Arabidopsis seedlings resistance to salt stress. On the other hand, pretreatment with 200 μM SA reduced Arabidopsis resistance to salt stress and aggravated oxidative damage to the seedlings. At all concentrations used, SA pretreatment inhibited the total respiration and promoted reactive oxygen species (ROS) generation. However, the ROS content in 10 or 20 μM SA pretreated seedlings decreased to the basal level within 6 h and high activities of antioxidant enzymes and alternative oxidase were maintained. Notably, the SA-enhanced salt stress resistance was significantly impaired by blocking alternative oxidase (AOX) pathway. Our findings indicate that SA-mediated salt stress response is in a dose- and time-dependent manner and that the effects were related to the induction of AOX capacity and antioxidant system.
