Abstract:Germination offers advantages to improve legume protein digestibility as it disintegrates seed structure and hydrolyzes proteins and anti-nutrients. Seed permeability (related to polyphenol content of seed coats) is an important factor affecting the duration of seed germination and its impact on protein digestibility and bioactivity. The objective was to compare the effect of seed germination on protease activity, structure, and proteolysis of four selected legumes with contrasting seed coat polyphenol profiles (gray zero-tannin lentil [GZL], beluga lentil [BL], and dehulled red lentil [DL]; and zero tannin/low vicine-convicine fava bean [ZF]). Protein hydrolysis was characterized during germination and digestion with respect to proteins, peptides, and free amino acids (FAAs). In vitro antihypertensive and antioxidant activities of digests were investigated, and the peptidomic characterization [high-performance liquid chromatography coupled to tandem mass spectrometry (HPLC-MS/MS)] and identification of bioactive fragments in intestinal digests were performed. Regardless of the seed type, germination increased protease activity and reduced the levels of phytic acid, trypsin inhibitors, and tannins (only in BL). A significant proteolysis of the 7S and 11S globulins and a concomitant increase of peptides and FAAs were observed in all sprouted legumes. Digestion kinetics in sprouts revealed a faster generation of FAAs and peptides than in dry seeds, with changes being more evident for DL, associated with a faster imbibition, germination, and sprout growth. In contrast, BL sprouts showed the lowest protein digestibility, likely due to a lower protease activity, seed structure disintegration, and higher anti-nutrient levels in comparison to GZL, DL, and ZF. Moreover, the digestion of sprouts resulted in a higher number of resistant peptides in DL and ZF that matched with previously reported bioactive sequences, suggesting a promising health potential of legume sprouts that was confirmed in vitro. The results suggested that the germination process improved protein digestibility and the health-promoting potential of lentil and fava bean proteins although these changes were more evident in DL due to its rapid imbibition, faster germination, and sprout development. This study will provide important information for either plant breeders to develop legume varieties with permeable seed coats or food producers that could use dehulled seeds for efficient production of sprouts as sustainable food sources of plant proteins with improved nutritional and healthy properties.
Impact of germination time on protein solubility and anti-inflammatory properties of Pisum sativum L grains

Abstract:During germination processes take place that modify the major components of the grain, such is the case of proteins that are hydrolyzed to generate peptides that can lead to the generation of bioactivity. The objective of the present work was to germinate grains of Pisum sativum to evaluate the effect on the soluble protein content and the anti-inflammatory activity. The grains were subjected to 10 days of germination at 24 °C and relative humidity of 75%. Sprouts were lyophilized, milled, and phenolic compounds were extracted to avoid interferences. Soluble protein content varied significantly during the 10 days of germination. In vitro assays indicate that sprouts protein inhibits thermal denaturation of proteins, protease activity, and stabilize cell membranes. The IC50 values indicate that after germination the bioactivity increased between 1.4 and 3.5 times, with respect to the ungerminated grains. Results indicated that Pisum sativum sprouts may constitute promising health-promoting foods.
Variation in Polyphenols, Tocols, Γ‐aminobutyric Acid, and Antioxidant Properties in Whole Grain Rice (oryza Sativa L.) As Affected by Different Germination Time

Abstract:Dynamic alterations in the polyphenols, tocols, antioxidant activities (AOA), and γ‐aminobutyric acid (GABA) contents of white, red, and black germinated rice upon different germination periods were studied. The results indicated that the contents of the free, bound, and total phenolics and AOA steadily increased during germination, even though the free part of colored germinated brown rice (GBR) showed a notable decline at the earlier stages, but still higher than the white rice. The compositions of the nine phenolic acids and six tocols generally accumulated in a time‐ and variety‐dependent manner during the germination process. However, the contents of p‐coumaric, ferulic, and sinapic acids displayed a pronounced increase with the duration of germination in all cultivars. The GABA in white, red, and black GBR accumulated to the highest content at the end of germination, which was 4.7, 14.2, and 6.7‐fold of their respective nongerminated counterparts. Interestingly, the total anthocyanin content in black GBR displayed a constant drop at earlier stages and then a rise at the later stages. These results suggest that prolonged germination may further increase the functional properties of GBR.

Abstract:In recent years, with the accelerated pace of life and the high incidence of chronic diseases, the demand for functional food has been increasingly urgent. Under such circumstances, germinated edible foods have attracted much attention because of their unique physiological effects. This review mainly summarizes recent studies about the physiological and biochemical changes during edible seeds germination. Further, the different germination treatments and the enrichment of γ-amino butyric acid are discussed. Finally, the applications of germinated seeds in the food sector are mentioned. Germination of edible seeds is a complex process of material transformation, which not only improves the nutritional value of raw seeds, but also accumulates a wide variety of bioactive compounds with health benefits. As a potential source of numerous functional ingredients, germinated edible foods have attracted widespread attention for its physiological benefits. Therefore, germinated grain and legume seeds are expected to be functional dietary supplements with a variety of health benefits and medicinal values. Appropriate germination treatments can enrich bioactive components, which is conducive to the efficient production of bioactive functional food. Research in this area is expected to serve the purpose of preventing and alleviating chronic diseases.

Abstract:Folate deficiency associated with low dietary intake is a well-documented public health problem, resulting in serious health and socioeconomic burdens. Therefore, optimization of the germination process of different cultivars of legume seeds in relation to the content and composition of folate, vitamin C, and total phenolics and total antioxidant capacity was carried out to maximize the health-promoting properties. The content and composition of folate, vitamin C, and total phenolic and total antioxidant capacities varied between species, among cultivars, and with germination time. During germination, total folate content was maximum at 815.2 μg/100 g fresh weight in soybean sprout and at 675.4 μg/100 g fresh weight in mungbean sprout on the fourth day, which were equivalent to, respectively, 3.5- and 3.9-fold increases in the seed's content, and total folate content strongly decreased thereafter. 5-CH(3)-H(4)folate was the most abundant folate species in legume sprouts and reached a maximum on the fourth day. Vitamin C was not detected in raw seeds, and its content increased sharply in soybean and mungbean sprouts and reached a maximum at the fourth day of germination (29 and 27.7 mg/100 g fresh weight, respectively). Germination of soybean and mungbean for 4 days provided the largest amount of total folate as well as the more stable species 5-CH(3)-H(4)folate and also brought about large amounts of vitamin C and total phenolics and substantial antioxidant capacities.

Abstract:The determination of the changes in the composition of bioactive phenolic compounds of germinating seeds which accumulate high levels of these compounds could contribute to the understanding of the germination mechanism and the development of markers for the selection of plant genotypes. In the current study, the changes in the phenolic composition and a-glucosidase inhibition activity, taking place during the germination of four wild forms and four varieties of common bean (Phaseolus vulgaris L.) from Durango Mexico, were determined. A total of 66 phenolic compounds (19 phenolic acids, 18 isoflavones, 18 flavonol glycosides, 3 flavonol aglycones, 3 flavones, 2 dihydroflavonoids, 2 chalcones and one non-identified type) were found by HPLC-DAD, which were differentially accumulated by the seeds, 24 h-soaked seeds, and 4 day-sprouts of each genotype. The accumulation of the flavonol aglycones, myricetin, quercetin and kaempferol was distinctive of the wild seeds. Soaking not only caused leaching and degradation but also triggered the synthesis of new phenolic compounds whereas germination diversified the composition of isoflavones and flavonol glycosides. The seeds of all genotypes analyzed were important inhibitors of a-glucosidase, improving their potential after soaking and germination. The results suggested that the structure rather than the concentration of the flavonoids and phenolic acids determined the inhibitory potential of a-glucosidase of samples. The principal component analysis and cluster analysis revealed HPLC-DAD phenolic profiles as genotype-specific chemomarkers at any of the states (seeds, soaked seeds, and sprouts). The results have wide implications on agronomy and food quality.

Abstract:There has been an increase in the use of adoptable bioprocessing methods for the development of high-quality leguminous ingredients. The potential use of germinated green Altamura lentils as a food ingredient is closely related to the resulting properties. The objective of this study was to evaluate the impact of two germination times -0 (C), 24 (G) and 48 (H) hours-on the physicochemical, microstructural, flavour, functional, and nutritional features of lentil flour samples (CF, GF and HF). Lentil flour samples were obtained by grinding both whole green seeds (C) and germinated seeds (G and H), and then sifting them to obtain a particle size < 300 μm. Germinated samples -GF (24 h) and HF (48 h)- exhibited differences (P < 0.05) in the physicochemical and bioactive properties of CF (control). Similarly, compared with those in the control sample, the total starch, amylose and total phenolic contents in the GF and HF samples decreased, while the protein content increased (p < 0.05). A decrease in the presence of intact starch granules was observed via SEM in the germinated samples. The germination time had a significant (P < 0.05) effect on the colour indices, L*, a*, b* of the samples. Flavour attributes were significantly influenced by the germination time. Overall, a total of 14 (CF) and 17 (GF and HF) aromatic compounds were identified. The technological characteristics of the CF, GF and HF doughs were studied by using a Brabender farinograph. Germination time affects flour properties, leading to a significant decrease in farinographic parameters such as water absorption (WA), dough development time (DT), and dough stability (DS) and an increase in the degree of dough weakening (DOS). A differential scanning calorimetry was employed to examine the gelatinization transition of the samples. Germination strongly influenced all the thermal properties of the samples. Germination also had a significant impact on the in vitro starch digestibility, starch fraction and glycaemic index (eGI) of the samples. In particular, the eGI of germinated lentils was lower than that of the CF. In conclusion, germination time could be a key factor for modulating some crucial lentil flour properties.

Abstract:MONARTICULAR bacterial arthritis of the sternoclavicular joint appears to be rare. Arlet and Ficat 1 reported one proved and one probable case of staphylococcal infection of this joint. Abstract:On October 29, 2009, researchers and physicians gathered at the Sheraton Four Points Hotel in Boston for 4 days to discuss a disease called multiple hereditary exostoses (MHE). MHE is an autosomal dominant disease that is associated with mutations in two enzymes that are required for heparan sulfate (HS) synthesis. Children with the disease form numerous benign bone tumors (osteochondromas) and have >2% chance of developing chondrosarcoma. The aim of the meeting was to generate new ideas for the diagnoses, treatment, and cure of this disease. Discussions ranged from orthopedic surgical treatment and patients' personal experiences to fundamental questions in skeletal biology and the precise molecular role that HS plays in developmental signaling pathways. Abstract:This study assesses the transformation and stability of polyphenols, sulforaphane, and indoles in a fermented beverage made from broccoli leaves during in vitro gastrointestinal digestion (GID). This process was simulated using a dialysis membrane to assess intestinal absorption. The total phenolic compounds (TPC) and antioxidant TEAC assays showed an increase in phytochemical content due to the GID process. The higher TPC and antioxidant activity observed after digestion was likely due to the enzymatic transformation of polyphenols in mildly alkaline conditions. Individual phytochemical analysis revealed that hydroxycinnamic acids, particularly 3CQa, remained stable initially but then decreased significantly during intestinal digestion. Acylated flavonoids exhibited a decrease during intestinal digestion, while deacylated flavonoids initially decreased before stabilising. This indicated the occurrence of enzymatic hydrolysis of more structurally complex flavonoids to glycosylated flavonoids such as kaempferol-3,7-diglucoside, and kaempferol-3-sophoroside-7-glucoside. Consequently, deacylated flavonoids were highlighted for their high bioaccessibility rate after in vitro GID. Glucosinolate-hydrolysis products, including sulforaphane and indoles, exhibited a general decrease during digestion, with sulforaphane showing 51% bioaccessibility. The study highlights the dialysed in vitro GID process, which affects the release and transformation of bioactive compounds, potentially increasing their bioaccessibility and the subsequent health benefits of the lactofermented beverage made from broccoli leaves.

Abstract:To the Editor: In a recent letter, Ferguson’ argued that the incremental potassium load produced by irradiation of blood would be clinically trivial. Rock and Shear2 replied that, in the situation of transfusing an infant, this would not be true. They based their argument on calculations in which the incremental potassium was confined to the infant’s plasma volume and resulted in an increase in serum potassium that might have clinical significance. There is a fundamental error in the latter calculations. Any potassium infused, whether in an intravenous solution or in association with a blood transfusion, would not be confined to the plasma space but would be distributed rapidly in the total potassium pool, which is approximately 98 percent intracellular. This error leads to a second error in which the total body potassium is underestimated by a factor of 50 times. A common method of calculating potassium distribution is to consider any increment as being distributed in total body water, which is also predominantly intracellular. Using the 50-mL plasma volume chosen by Rock and Shear,2 the effect of such an infusion can be reevaluated. Assuming a packed cell volume of 55 percent, the total blood volume would then be approximately 110 mL. Because the blood volume at birth is 75 mL per kg, this would indicate a birth weight of 1470 g. Total body water is 78 percent of the birth weight, or 1150 mL. If an incremental potassium load of 0.09 mM (as used by Ferguson) is distributed in this volume, it will result in an increase of 0.078 mM per L, which would raise the normal serum level from 3.5 to 3.578 mM per L. This is less than the minimum suggested increment for S y s t h e International units and, more important, has no clinical significance, as pointed out by Ferguson. No one can disagree with the need for careful transfusion practices, particularly in neonatal care. Abstract:Lima beans (Phaseolus lunatus) and adzuki beans (Vigna angularis) are some of the most nutritious underutilized pulses that are significant in being used as basic ingredients for the preparation of various food products. The present study aimed to determine the impact of soaking and germination on nutritional and bioactive components, in vitro protein digestibility, reducing power, metal chelating capacity, antioxidant activity, and anti-nutritional components of lima and adzuki beans. The findings showed that during the germination treatment, the in vitro protein digestibility of lima and adzuki beans increased by 14.75 and 10.98%, respectively. There was an increase in the antioxidant activity of lima beans by 33.48% and adzuki beans by 71.14% after 72 h of germination, respectively. The reducing power assay of lima and adzuki beans indicated an increase of 49.52 and 36.42%, respectively, during germination. Similarly, the flavonoid and metal chelating activity increased in lima and adzuki beans after 72 h of germination. In contrast, the anti-nutrients, such as phytic acid, tannin content, and trypsin inhibitor activity, decreased significantly p < 0.05 after 72 h of germination. These results are encouraging and allow for utilizing the flour obtained from the germinated beans in functional bakery products, which can contribute to eradicating protein deficiency among some population groups. At the same time, promoting soaking and germination of the beans as a way to enhance the nutritional quality and reduce anti-nutrients can contribute to the interest in these underutilized pulses. They could be seen as an additional tool to improve food security.
Influence of fermentation and germination on some bioactive components of selected lesser legumes indigenous to Nigeria

Abstract:This study assessed the effects of fermentation time (2, 3 and 4 days) and germination time (2, 3 and 4 days) on total phenolics, tannin, anthocyanin, carotenoids and flavonoids contents of lesser legumes: cowpea (Vigna unguiculata L.), bambaranut (Vigna subterranean L.), red bean (Phaseolus vulgaris), pigeonpea (Cajanus cajan), African breadfruit (Treculia africana) seeds, African yam bean (Sphenostylis stenocarpa) seed, African oil bean (Pentaclethra mycrophylla Benth.) seed and groundnut (Arachis hypogea L.). The antioxidant and reducing powers of a three day germinated samples were also evaluated. The result revealed that fermentation reduced the total phenolics, tannin, anthocyanin, carotenoid and flavonoid contents of the samples with increasing fermentation time; however, red bean showed minimal increase with increasing fermentation time. There was significant (p < 0.05) increase in the total phenolics of all the samples with increasing germination time but, tannin and flavonoid showed significant (p < 0.05) reduction with increasing germination time. Germination significantly (p < 0.05) reduced the carotenoid and anthocyanin levels in all the samples, however, red bean, pigeonpea and African oil bean showed increases with increasing germination time. Samples evaluated exhibited significantly (p < 0.05) different antioxidant capacities. African oil bean and groundnut had the highest antioxidant activities 52.18% and 52.16%, respectively while, African yam bean seed was the lowest (19.85%). Similar trend was observed in the reducing power of the raw samples where groundnut, bambaranut and African breadfruit showed significantly (p < 0.05) higher reducing power. Three (3) days germination significantly (p < 0.05) increased the antioxidant capacities by 14.65%, 18.42%, 53.58%, 52.84%, 17.24%, 14.56%, 53.18% and 43.03% in African oil bean, bambaranut, cowpea, red bean, African breadfruit, groundnut, African oil bean and pigeonpea, respectively. Therefore, for increase antioxidant activity in lesser known legumes, germination is more preferred over fermentation and for maximum yield of total phenolics, three and four days germination time are recommended.

Abstract:Phenolic compounds are secondary plant metabolites whose beneficial health effects make them of intense interest to researchers. The aim of the study presented here was to evaluate the change in the phenolic compound profile of lupin seed in in vitro digestion. The most abundant phenolic compounds in undigested lupin seeds were mostly apigenin derivatives. The in vitro digestion of lupin seeds resulted in qualitatively altered phenolic compound profiles. Approximately 80% of phenolic compounds were released from lupin seeds during the in vitro digestion, which simulated gastric processes. Continued digestion, imitating the intestinal phase, additionally increased the bioaccessibility of lupin seed polyphenols by about 10%. The in vitro gastrointestinal model was also used to elucidate how the content of native phenolic compounds affects the digestion susceptibility of lupin seed proteins. An inverse correlation between protein digestibility and phenolic compound content, was also demonstrated.

Impact of Germination on the Protein Solubility and Antinutritive Compounds of Lupinus angustifolius and Vicia faba in the Production of Protein-Rich Legume-Based Beverages

Abstract:Lupines and faba beans are protein-rich legumes, which can be utilized as a plant-based substitute for animal proteins in human nutrition in general and in the beverage industry in particular. However, their application is hampered by low protein solubility in the acidic pH range and by antinutrients such as flatulence-causing "raffinose family oligosaccharides" (RFOs). Germination is known in the brewing industry for increasing enzymatic activities and mobilizing storage compounds. Therefore, germinations of lupines and faba beans were performed at different temperatures, and their impact on protein solubility, the concentration of free amino acids, and degradation of RFOs, alkaloids, and phytic acid was evaluated. In general, changes were comparable for both legumes but less pronounced in faba beans. Germination depleted the RFOs entirely in both legumes. The protein size distribution was found to be shifted toward smaller fractions, the concentrations of free amino acids multiplied, and protein solubility increased. No substantial reductions in the binding capacity of phytic acid toward iron ions were observed, but a release of free phosphate in lupines was detected. The results prove that germination is an applicable process for refining lupines and faba beans for use not only in refreshing beverages or milk alternatives but also in other food applications.

Abstract:Edible sprouts with germinating seeds of a few days of age are naturally rich in nutrients and other bioactive compounds. Among them, the cruciferous (Brassicaceae) sprouts stand out due to their high contents of glucosinolates (GLSs) and phenolic compounds. In order to obtain sprouts enriched in these phytochemicals, elicitation is being increasing used as a sustainable practice. Besides, the evidence regarding the bioavailability and the biological activity of these compounds after their dietary intake has also attracted growing interest in recent years, supporting the intake of the natural food instead of enriched ingredients or extracts. Also, there is a growing interest regarding their uses, consumption, and applications for health and wellbeing, in different industrial sectors. In this context, the present review aims to compile and update the available knowledge on the fundamental aspects of production, enrichment in composition, and the benefits upon consumption of diverse edible cruciferous sprouts, which are sources of phenolic compounds and glucosinolates, as well as the evidence on their biological actions in diverse pathophysiological situations and the molecular pathways involved.

Evidence on the Bioaccessibility of Glucosinolates and Breakdown Products of Cruciferous Sprouts by Simulated In Vitro Gastrointestinal Digestion

Abstract:Cruciferous vegetables are gaining importance as nutritious and sustainable foods, rich in phytochemical compounds such as glucosinolates (GSLs). However, the breakdown products of these sulfur-based compounds, mainly represented by isothiocyanates (ITC) and indoles, can contribute to human health. In the human digestive system, the formation of these compounds continues to varying extents in the different stages of digestion, due to the contact of GSLs with different gastric fluids and enzymes under the physicochemical conditions of the gastrointestinal tract. Therefore, the aim of the present work was to uncover the effect of gastrointestinal digestion on the release of glucosinolates and their transformation into their bioactive counterparts by applying a simulated in vitro static model on a range of brassica (red radish, red cabbage, broccoli, and mustard) sprouts. In this sense, significantly higher bioaccessibility of ITC and indoles from GSLs of red cabbage sprouts was observed in comparison with broccoli, red radish, and mustard sprouts, due to the aliphatic GSLs proportion present in the different sprouts. This indicates that the bioaccessibility of GSLs from Brasicaceae sprouts is not exclusively associated with the initial content of these compounds in the plant material (almost negligible), but also with the release of GSLs and the ongoing breakdown reactions during the gastric and intestinal phases of digestion, respectively. Additionally, aliphatic GSLs provided higher bioaccessibility of their corresponding ITC in comparison to indolic and aromatic GSLs.

Effect of sprouting on the proteome of chickpea flour and on its digestibility by ex vivo gastro-duodenal digestion complemented with jejunal brush border membrane enzymes

Abstract:The demand for sustainably produced proteins is increasing with the world population and is prompting a dietary shift toward plant sourced proteins. Vegetable proteins have lower digestibility and biological value compared to animal derived counterparts. We explored sprouting of chickpea seeds as a strategy for improving digestibility. Protein evolution associated with by the sprouting process was assessed by proteomics. The sprouting induced breakdown of seed storage proteins and doubled the release of free alpha-amino nitrogen in sprouted chickpea flour. During sprouting, several enzymes involved in plant development were newly expressed. An ex vivo model of gastroduodenal and jejunal digestion was applied to assess the bioaccessibility of the protein digests. Proteins from chickpea sprouts showed a greater susceptibility to digestion with a 10% increase in alpha amino nitrogen. Peptides with potential immunoreactivity or bioactivity were catalogued in both digested chickpea sprouts and seeds using an in-silico approach. Peptides belonging to the non-specific transfer proteins, which are allergens in pulses, and peptides belonging to an IgE-binding hemagglutinin protein could only be identified in the digested chickpea sprouts. The observation collected paved the way to immune-based evaluations to assess the effect of germination on the allergenic potential.

Abstract:The influence of different germination conditions on isoflavone contents in chickpea sprouts was investigated in this study. Chickpea sprouts were germinated under different experimental conditions, including germination in the dark (GD), in the light (GL), under ethanol stress (GE), or under salt stress (GS) in the dark. The results demonstrated that the isoflavone contents in chickpea sprouts germinated with these various conditions significantly increased (p<0.05) compared to those in untreated chickpea seeds. The maximum amount of total isoflavones was obtained from chickpea sprouts in the GL group on day 8. The contents of formononetin and biochanin A in this group were 154 and 130 times higher, respectively, than in untreated seeds and 1.2 times higher than in sprouts in the GD group. Moreover, the isoflavone contents of chickpea sprouts in the GE and GS groups were also higher (p<0.05) than those in the GD group. A solution of 3% ethanol and 0.03 mol/L salt seemed to be the most optimal for isoflavone production among the solutions selected for this study. Most of the isoflavone contents significantly increased (p<0.05), especially formononetin and biochanin A, while the genistein content decreased with germination. Ononin, pseudobaptigenin, and glycitein glucoside acetylated were only detected in germinated chickpeas. This finding could expand the potential for the development of chickpea sprouts as a functional food.
