Abstract:Sufu is a cheese-like product fermented with soybean curd in China, widely consumed for its unique nutritional and flavor properties. To investigate the associations among microbial communities, chemical properties, producing regions and dressing flavors of sufu, the high-throughput sequencing for 16 sufu products from different regions based on the bacterial 16S rRNA and fungal internal transcribed spacer 2 gene was performed. A sum of 368 bacterial genera and 299 fungal genera were identified. The fungal microflora in sufu samples contained many symbiont fungi of soybean. Aspergillus and Actinomucor were abundant in the sufu products as hydrolase-producing genera. The bacterial diversity result showed that lactic acid bacteria (LAB) were very abundant in all these sufu products and the Lactococcus was the most abundant genus. According to the principal coordinate analysis and hierarchical clustering analysis, the bacterial communities of sufu samples could be generally clustered into the LAB clade and Bacillus clade. Moreover, the fungal communities of sufu samples from eastern China could be generally clustered into one group, while the dressing flavor of sufu samples during post-fermentation was not a major factor for microbial communities shaping. This study could potentially serve as a reference for a rational design of microbial communities to improve the sufu quality.

Abstract:Multispecies microbial mixed fermentation is crucial for the formation of flavors in soy sauce . The present study applied metagenomic strategies to uncover the metabolic network for flavors formation in the non-Cantonese soy sauce in China. Twenty-nine major flavor compounds were identified by GC-MS and HPLC , representing the typical flavor compounds of the non-Cantonese soy sauce, including 3 organic acids, 8 amino acids, and 18 volatile compounds. Metagenomic sequencing and KEGG enrichment analysis revealed the specific functional microbiome, which constituted seven major flavor compounds associated genera, namely Leuconostoc , Staphylococcus , Tetragenococcus , Weissella , Aspergillus , Wickerhamycesa , and Zygosaccharomyces . Aspergillus was primarily responsible for the degradation of raw materials in the early fermentation stage, including the degradation of cellulose and lecithin. It also played an irreplaceable role in the formation of 1-octene-3-ol. Weissella , Staphylococcus, and Leuconostoc played essential metabolic roles in the whole fermentation process, particularly in the metabolism of amino acids and the formation of alcohols. Staphylococcus also participated in the degradation of nitrite/nitrate. Tetragenococcus became the dominant bacteria with the highest relative abundance in the late fermentation stage, which was mainly responsible for the synthesis of acetic acid, lactic acid, and other acids. Additionally, Wickerhamycesa and Zygosaccharomyces also contributed to the formation of alcohols, acids, and aldehydes during the late fermentation stage. This study provides a deeper understanding of the diverse roles in the flavor formation of microorganisms during the traditional condiment soy sauce fermentation process.

Abstract:Complex microbial metabolism is responsible for the unique flavor of soy sauce. In our earlier work, we unveiled the pivotal components that contribute to the unique flavor of soy sauce. However, it is challenging to comprehend the connection between microbes and the development of flavors during soy sauce fermentation. This work aims to characterize the taxonomic and functional attributes of soy sauce fermentation microbiota as well as to validate it. Samples from three representative fermentation stages were selected for metagenomic sequencing. Results demonstrated that in the metabolic network linked to the characteristic flavor formation, the functional genes are primarily connected to the metabolism of carbohydrates and amino acids. Analysis of microbial distribution in different flavor biosynthesis pathways revealed that 15 key genera were involved in the flavor generation. Finally, 6 screened potential flavor-producing microorganisms were validated by culturing them in a simulated high-salt fermentation environment of soy sauce, with several strains demonstrating the ability to produce the characteristic flavor of soy sauce, and Weissella paramesenteroides being the most remarkable. These findings are helpful for the selection of flavor-inducing starters for soy sauce industrial fermentation.

Abstract:Microorganisms play essential roles in flavor formation during soy sauce fermentation. Different soy sauce fermentation types significantly affect flavor formation. However, comparisons of microbial communities and metabolites between different fermentation types have been little studied. Here, we investigated variation in microbial communities, metabolite profiles, and metabolic pathways during Japanese-type (JP) and Cantonese-type (CP) fermentation. Free amino acids and volatile compound profiles varied significantly between fermentation types, with JP samples containing higher contents of esters (39.84%; p < 0.05), alcohols (44.70%; p < 0.05) in the 120 d fermentation samples. Volatile compound profiles varied significantly between fermentation types, with JP samples containing higher contents of esters, alcohols, and free amino acids ( p < 0.05). Metagenomic analysis indicated that both JP and CP communities were dominated by Tetragenococcus , Staphylococcus , Weissella (bacteria), and Aspergillus (fungi), but the two communities varied differently over time. Tetragenococcus drastically increased in abundance throughout the fermentation (from 0.02 to 59.2%) in JP fermentation, whereas Tetragenococcus (36.7%) and Staphylococcus (29.7%) dominated at 120 d of fermentation in CP fermentation. Metagenomic functional profiles revealed that the abundances of most genes involved with carbohydrate, amino acid, and lipid metabolism exhibited significant differences between fermentation types ( p < 0.05) during the middle to late fermentation stages. Furthermore, predicted metabolic pathways for volatile substance biosynthesis differed between JP and CP fermentation, likely explaining the differences in flavor metabolite profiles. In addition, most of the genes associated with flavor generation were affiliated with Tetragenococcus , Weissella , Staphylococcus , Bacillus , and Aspergillus , suggesting that these microbes play important roles in flavor production during soy sauce fermentation. This study significantly improves our understanding of microbial functions and their metabolic roles in flavor formation during different soy sauce fermentation processes.

Abstract:BS Sufu is a fermented food that is made by mixed black soybeans and soybeans. Microbial communities and metabolites play an important role for the final product. We characterized microbial diversity of BS Sufu during fermentation by high-throughput DNA sequencing. Meanwhile, volatile compounds were investigated by solid-phase microextraction (SPME) coupled with gas chromatography–mass spectrometry (GC-MS). The results showed that bacterial diversity was higher than that of fungi in BS Sufu. We found the existence of bacterial and fungal core communities, including Enterococcus , Enterobacter , Rhizopus , and Monascus . Network analysis indicated that bacterial and fungal communities maintain positive and negative interactions, which are important to shape the resident microbial communities in Sufu. In addition, 17 free amino acids (FAAs) were detected at the post-fermentation stage, and umami amino acid mainly contributed to taste of BS Sufu. Furtherly, a total of 79 volatile constituents in BS Sufu, including nine alcohols, 31 esters, and four aldehydes, form synergistically the unique odor of Sufu. Additionally, the correlations between microbiota and metabolites were analyzed. Our results suggested that these microbial taxa and metabolites contribute to the taste and flavor of BS Sufu. This study provided information for analysis of BS Sufu at different fermentation periods in terms of the microbial diversity and metabolites, and this information was important to understand the properties of mixed soybeans Sufu.

Abstract:Sufu, a Chinese traditional fermented soybean product, has a characteristic foul smell but a pleasant taste. We determined the core functional microbiota and their metabolic mechanisms during sufu fermentation by examining relationships among bacteria, characteristic flavor compounds, and physicochemical factors. Flavor compounds in sufu were detected through headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry, and the microbial community structure was determined through high-throughput 16S rRNA sequencing. The results showed that the fermentation process of sufu could be divided into early and late stages. The early stage was critical for flavor development. Seven microbiota were screened based on their abundance, microbial relevance, and flavor production capacity. Five microbes were screened in the early stage: Pseudomonas, Tetragenococcus, Lysinibacillus, Pantoea, and Burkholderia–Caballeronia–Paraburkholderia. Three microbes were screened in the late stage: Exiguobacterium, Bacillus, and Pseudomonas. Their metabolic profiles were predicted. The results provided a reference for the selection of enriched bacterial genera in the fermentation process and controlling applicable process conditions to improve the flavor of sufu.

Abstract:Ventriculoperitoneal shunt is one of the most frequently performed procedures by the neurosurgeons in clinical practice. This simple device results in great improvement in neurological functions and survival of patients with hydrocephalous. Infections associated with the shunt placement vary between 2 to 27%. Central nervous system infection is a cause of significant morbidity and mortality causing malfunction and chronic ill health and a few cases may even turn fatal. They are mainly caused by coagulase negative staphylococcus and rarely by other bacterias like streptococci, enterococci, Gram negative bacilli, fungus etc. We encountered a very rare case in which a 12 year-old female child developed an infection of subcutaneous tissue around the shunt tube that was consistent with M. tuberculosis.

Abstract:Summary Sufu, a traditional Chinese fermented soybean food, had its flavor and quality influenced by the specific strains and fermentation environment. To investigate how microbial community succession affects the volatile aroma substances of Mucor racemosus sufu during fermentation, the microbial community structure using high‐throughput sequencing (HTS) technology was analyzed. Additionally, volatile flavor compounds using solid‐phase microextraction gas chromatography mass spectrometry (SPME‐GC‐MS) were detected. Throughout the pre‐fermentation, salting, and post‐fermentation stages, the changes in the microbial communities over time and space due to environmental variations were observed. The colony compositions at the phylum and genus levels were also analyzed. The results revealed that Proteobacteria and Actinobacteria were the dominant bacterial phyla in the final fermented product. Furthermore, several microorganisms associated with flavor substances were identified, such as Lactobacillus, Pseudomonas, Wescheria, Hyphomyces, Candida, and Pichia. These microorganisms primarily influenced the synthesis of alcohols and acids, contributing to the overall flavor profile of the sufu. In the analysis of the final product, it identified 54 volatile flavor compounds, including alcohols, esters, phenolic acids, aldehydes, ketones, ether, alkenes, and other compounds. These findings have important implications for isolating specific strains, enhancing flavor, and providing a solid foundation for optimizing and ensuring the quality of sufu.

Abstract:Aspergillus sojae 3.495 and Aspergillus oryzae 3.042 are essential microbial starters in fermented foods due to their high hydrolytic enzyme activities. However, their metabolic characteristics and differences during soy sauce fermentation are still unclear. This study identified 42 non-volatile differential metabolites based on UHPLC-MS/MS, among which 10 amino acids and derivatives and 4 non-volatile acids were high in A. sojae 3.495 koji , while 3 sugar alcohols, 5 flavonoids, and 4 fatty acids were prominent in A. oryzae 3.042. Further analysis revealed that higher acidic protease (1887.43 U/g) in A. sojae 3.495 and higher glucoamylase (2413.36 U/g) and cellulase (890.78 U/g) in A. oryzae 3.042 dominated the formation of differential metabolites. Metabolic pathway analysis showed that shikimate and phenylalanine metabolic pathways in A. sojae 3.495 were more active and enhanced 3-methyl-butanal and 2-phenylethanal in moromi fermentation. The more prominent pathways of sugar alcohol metabolism and flavonoid biosynthesis in A. oryzae 3.042 koji enhanced 2-methyl-butanoic acid and ethyl 3-methylbutyrate in moromi and significantly elevated reducing sugars (1.56 g/100 mL). Overall, comparative metabolomics elucidated the metabolic framework and flavor formation mechanisms of different inocula in soy sauce koji , providing ideas for scientific control of soy sauce fermentation and multi-strain koji -making for flavor enhancement.

Abstract:Fermentation microbiota is specific microorganisms that generate different types of metabolites in many productions. In traditional solid-state fermentation, the structural composition and functional capacity of the core microbiota determine the quality and quantity of products. As a typical example of food fermentation, Chinese Maotai-flavor liquor production involves a complex of various microorganisms and a wide variety of metabolites. However, the microbial succession and functional shift of the core microbiota in this traditional food fermentation remain unclear. Here, high-throughput amplicons (16S rRNA gene amplicon sequencing and internal transcribed space amplicon sequencing) and metatranscriptomics sequencing technologies were combined to reveal the structure and function of the core microbiota in Chinese soy sauce aroma type liquor production. In addition, ultra-performance liquid chromatography and headspace-solid phase microextraction-gas chromatography-mass spectrometry were employed to provide qualitative and quantitative analysis of the major flavor metabolites. A total of 10 fungal and 11 bacterial genera were identified as the core microbiota. In addition, metatranscriptomic analysis revealed pyruvate metabolism in yeasts (genera Pichia, Schizosaccharomyces, Saccharomyces, and Zygosaccharomyces) and lactic acid bacteria (genus Lactobacillus) classified into two stages in the production of flavor components. Stage I involved high-level alcohol (ethanol) production, with the genus Schizosaccharomyces serving as the core functional microorganism. Stage II involved high-level acid (lactic acid and acetic acid) production, with the genus Lactobacillus serving as the core functional microorganism. The functional shift from the genus Schizosaccharomyces to the genus Lactobacillus drives flavor component conversion from alcohol (ethanol) to acid (lactic acid and acetic acid) in Chinese Maotai-flavor liquor production. Our findings provide insight into the effects of the core functional microbiota in soy sauce aroma type liquor production and the characteristics of the fermentation microbiota under different environmental conditions.
Microbiota dynamics and volatile metabolite generation during sausage fermentation

Abstract:Microorganism metabolic activity is critical for the formation of unique flavors in fermented meat products. To clarify the relationship between the formation of the special flavor of fermented meat and microorganisms, high-throughput sequencing and gas chromatography-ion mobility spectrometry were used to analyze microorganisms and volatile compounds in naturally fermented sausage. The findings revealed 91 volatile compounds and 4 key microorganisms, including Lactobacillus, Weissella, Leuconostoc, and Staphylococcus. The key microorganisms were positively correlated with the formation of 21 volatile compounds. The validation results showed that the contents of volatile compounds such as heptanal, octanal, 2-pentanone, and 1-octen-3-ol increased significantly after inoculation with Lb. sakei M2 and S. xylosus Y4. These two bacteria are the key microorganisms that produce the special flavor of fermented sausage. The present study can provide a theoretical basis for the directional development of fermented meat products, the preparation of special flavor enhancers, and expedited fermentation processes.

Abstract:Guizhou sour meat and sour fish, Chaoshan fish sauce, Sichuan sausage and bacon, Cantonese sausage, Jinhua ham, and Xinjiang air-dried beef are eight representatives of Chinese traditional fermented meat and fish products (FMFPs), which are favored by Chinese consumers due to their high nutritional value and quality. The quality of the spontaneously fermented Chinese traditional FMFP is closely correlated with microorganisms. Moreover, the dominant microorganisms are significantly different due to regional differences. The effects of microorganisms on the texture, color, flavor, nutrition, functional properties, and safety of Chinese traditional FMFPs have not been not fully described. Additionally, metabolic pathways for flavor formation of Chinese traditional FMFPs have not well been summarized. This article describes the seven characteristic Chinese traditional FMFPs and correlated dominant microorganisms in different regions of China. The effects of microorganisms on the texture, color, and flavor of Chinese traditional FMFPs are discussed. Furthermore, the metabolic pathways of microbial regulation of flavor formation in Chinese traditional FMFPs are proposed. This work provides a theoretical basis for improvement of Chinese traditional FMFPs by inoculating functional microorganisms isolated from Chinese traditional fermented foods.

Abstract:The effects of fermentation with lactic acid bacteria (LAB) and yeast on the aroma of samples were analyzed in this work. The volatile features of different soybean hydrolysates were investigated using both GC-MS and GC-IMS. Only 47 volatile flavor compounds (VFCs) were detected when using GC-IMS, while a combination of GC-MS and GC-IMS resulted in the identification of 150 compounds. LAB-yeast fermentation could significantly increase the diversity and concentrations of VFCs (p < 0.05), including alcohols, acids, esters, and sulfurs, while reduce the contents of aldehydes and ketones. Hierarchical clustering and orthogonal partial least squares analyses confirmed the impact of fermentation on the VFCs of the hydrolysates. Seven compounds were identified as significant compounds distinguishing the aromas of different groups. The partial least squares regression analysis of the 25 key VFCs (ROAV > 1) and sensory results revealed that the treatment groups positively correlated with aromatic, caramel, sour, overall aroma, and most of the key VFCs. In summary, fermentation effectively reduced the fatty and bean-like flavors of soybean hydrolysates, enhancing the overall flavor quality, with sequential inoculation proving to be more effective than simultaneous inoculation. These findings provided a theoretical basis for improving and assessing the flavor of soybean protein hydrolysates.

Abstract:Based on the widespread application and under-research of mechanized preparation Cantonese soy sauce koji (MP), absolute quantitative approaches were utilized to systematically analyze the flavor formation mechanism in MP. The results indicated that the enzyme activities increased greatly during MP fermentation, and 4 organic acids, 15 amino acids, and 2 volatiles were identified as significantly different flavor actives. The flavor parameters of MP4 were basically identical to those of MP5. Furthermore, microorganisms were dominated by Staphylococcus, Weissella, and Aspergillus in MP, and their biomass demonstrated an increasing trend. A precise enumeration of microorganisms exposed the inaccuracy of relative quantitative data. Concurrently, Staphylococcus and Aspergillus were positively correlated with numerous enzymes and flavor compounds, and targeted strains for enhancing MP quality. The flavor formation network comprises pathways including carbohydrate metabolism, lipid metabolism and oxidation, and protein degradation and amino acid metabolism. In summary, the fermentation period of MP can be substantially shortened without compromising the product quality. These findings lay the groundwork for refining parameters in modern production processes.

Abstract:Western Sichuan Yi suancai contains a group of highly functional microorganisms in an alpine and high-altitude ecological environment. Due to its unique salt-free fermentation process, it is nutritious and has a crispy texture. Six periods were established during fermentation: day 0, day 2, day 5, day 8, day 11, and day 14. The results showed that the antioxidant capacity and organic acid content increased during the fermentation process, while the pH gradually decreased, indicating that suancai from the later periods was better for supplementing the human body with acid substances and eliminating free radicals. Twenty-six flavour compounds were identified, including alcohols, esters, ketones, and acids. Ethyl alcohol, 3-methyl-1-butanol, ethyl methanoate, and acetic acid were the main contributors to the flavour, imparting floral and fruity notes to the suancai. Five dominant bacterial genera (Lactobacillus, Leuconostoc, Weissella, Klebsiella, and unclassified_o__Lactobacillales) were identified via high-throughput sequencing during the fermentation process, and there were nine dominant fungal genera (Dipodascaceae_gen_Incertae_sedis, Mucor, Pichia, unclassified__f__Dipodascaceaet, Cyberlindnera, Diutina, Trichosporon, Saccharomycopsis, and Wickerhamomyces). Correlation analysis showed that the antioxidant capacity was positively correlated with genera such as Lactobacillus, Mucor, and Alternaria, indicating that these microorganisms have important roles in enhancing the antioxidant properties of suancai. Meanwhile, some genera, such as Microbacterium, Herbaspirillum, Mortierella, and Sampaiozyma, promote the synthesis of alcohols, esters, acids, and ketones. This study revealed the interactions between microorganisms and metabolites during the fermentation of western Sichuan Yi suancai and provided a scientific basis for further understanding the fermentation mechanism of traditional suancai and improving the fermentation process.

Synthesis and Characterization of Molecular Hybrids of Insulin and Insulin-like Growth Factor I

Abstract:Insulin and insulin-like growth factor (IGF) I have 50% homology in their primary amino acid sequences. IGF-I differs structurally from insulin because it has a connecting peptide between the Aand B-chains and an octapeptide sequence at the COOH-terminal of the IGF-I molecule termed the "D" region. To determine the role of the "D" region of IGF-I in its biological activity, we have synthesized a hybrid molecule in which this octapeptide is linked to the COOH-terminal of the A-chain of pork insulin and tested its properties in radioimmunoassay, radioreceptor assays, and bioassays. A control insulin analogue, A21 asparaginimid, which is altered at A21 but without the addition of the octapeptide sequence of IGF-I "D" region, was similarly tested. When pork insulin, insulin-IGF-I hybrid, A21-asparaginimid insulin, IGF-I, and IGF-II were tested for the stimulation of glucose oxidation in rat adipocytes, their potencies were 100%, 20%, 25%, 0.2%, and 2%, respectively. Similarly, in radioimmunoassay for pork insulin, the insulin-IGF-I hybrid, A21-asparaginimid insulin, IGF-I, and IGF-II were 11%, 10%, 0.2%, and 0.2% as potent as pork insulin. This was similar to their relative potency in competing with '2 5I-pork insulin for binding to the insulin receptors on rat adipocytes; insulin-IGFI hybrid and A21-asparaginimid insulin were only 28% and 25% as potent. IGF-I did not compete with 25'Iinsulin for binding even at 200 ,pg/ml. By contrast, in a bioassay measuring stimulation of thymidine incorporation into the DNA of human fibroblasts, the relative order of potency for these peptides was reversed: IGFI was 10-fold more potent than pork insulin, followed by IGF-II and insulin-IGF-I hybrid which were 5and 2.8-fold more potent than insulin, respectively, whereas A21-asparaginimid insulin was only 10% as potent. These data suggest that the terminal octapeptide of IGF-I plays a role in the increased growth-promoting activity of the IGFs, while it decreased IGF-I's potency on metabolic effects.

Abstract:A multi-omics approach was applied to investigate the differences and correlations between characteristic volatile flavor substances and non-volatile metabolites in sausages fermented by Pediococcus pentosaceus (P. pentosaceus) and Staphylococcus carnosus (S. carnosus) alone and in a mixture. Twenty-seven volatile metabolites were identified by headspace solid-phase microextraction/gas chromatography-mass. According to orthogonal projections to latent structures-differential analysis, 17 characteristic volatile metabolites were detected in the sausages of different treatments. Utilizing ultra-high-performance liquid chromatography coupled with a mass spectrometer to analyze metabolite profiles, 42.03% of the non-volatile metabolites were classified as lipids and lipid-like molecules, 25.00% of organic acids and derivatives, and others. Seventeen characteristic flavor substances were significantly correlated with twenty differential non-volatile metabolites, and the non-volatile metabolites changed significantly. Differences in the characteristics and combinations of microorganisms themselves have a decisive role in the development of flavor substances and non-volatile metabolites in sausages.

Abstract:Compound fermented seasonings were increasingly favored by consumers. This study using Aspergillus oryzae and soybeans as raw materials to make koji . Lactobacillus plantarum and Pediococcus pentosaceus were used as starters for common fermentation with mature koji and beef to obtain a fermented beef-soybean paste. The physicochemical parameters, volatile flavor compounds, and microbial community of beef-soybean paste during fermentation were determined, and the relationship between flavor changes and microbial community changes was analyzed. Compared to the soybean paste, the addition of beef for fermentation can significantly enhanced the content of protein, amino acid nitrogen, and free amino acids in the compound fermented soybean paste, improved the nutritional and health value of the product. The addition of beef also caused a decrease in pH and total acid content in the compound soybean paste. The flavor changed during the fermentation process of beef-soybean paste were related to the changed in its microbial community.

Abstract:Microbial metabolism is considered to have an important influence on the formation of taste, aroma and flavour compounds during vegetable fermentation. However, the relationship between microbial gene expression and the biosynthetic sources of flavor compounds are still unclear. In this study, genetic and metabolite profiles were used to characterize flavour development during Sichuan paocai (SCP) fermentation. Metatranscriptomic results showed that gene expression and enzyme activity by Enterobacteriaceae and Lactobacillaceae were strongly involved in SCP fermentation. Differential expression of 666 genes involving 27 metabolic pathways - most associated with amino acids- accompanied the accumulation of 17 amino acids (from 31.5 mu g/mL to 738.4 mu g/ mL). In addition, 239 volatile organic compounds (VOCs) were identified and odour activity values showed 10 volatile organic compounds (include 5 sulphur compounds) exert an important influence on the aroma of SCP. Sulphur compounds, Glu, Asp, Lactic acid, acetic acid and maltic acid exert an important influence on the sour, umami and sulphur flavour of SCP. The cysteine and methionine metabolism asparate and glutamate biosynthesis and metabolism and pyruvate metabolism were responsible for the key flavours formation. This study provides significant guidance in the aroma, flavour and tastes regulation in SCP fermentation and new product development.
