Abstract:Current serology tests for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibodies mainly take the form of enzyme-linked immunosorbent assays, chemiluminescent microparticle immunoassays or lateral flow assays, which are either laborious, expensive or lacking sufficient sensitivity and scalability. Here we present the development and validation of a rapid, low-cost, solution-based assay to detect antibodies in serum, plasma, whole blood and to a lesser extent saliva, using rationally designed split luciferase antibody biosensors. This new assay, which generates quantitative results in 30 min, substantially reduces the complexity and improves the scalability of coronavirus disease 2019 (COVID-19) antibody tests. This assay is well-suited for point-of-care, broad population testing, and applications in low-resource settings, for monitoring host humoral responses to vaccination or viral infection.

Abstract:Despite the promising performance of nanomaterial-based electrochemical detection techniques, product-level point-of-care sensor systems are still evolving. Unique approaches toward nanoscale tunability, enriched measurements, and data analysis are therefore imperative. The recent outbreak of coronavirus disease provides a suitable test bench toward this pursuit, attracting several innovative efforts from the sensor research community. Accordingly, this work reports an ultrasensitive and selective molecular biosensor for RT-PCR-confirmed hospital samples of SARS-CoV-2 built on ZnO nanoflowers. The unique experimental and analytical methodologies drastically improve the device efficiency, relying on three conserved genomic sequences (E, N, and ORF1ab genes). It involves multiple techniques like cyclic voltammetry, electrochemical impedance spectroscopy, and differential pulse voltammetry where normalized responses of these techniques are used to create 1D, 2D, and 3D data points representing each sample for clustering both by heuristic (2D separation plane) and ML approach (logistic regression) for accurate diagnosis. Enhancing data features also enlarges the Euclidean distances (ED) of data points, which raises the separation efficiency of the device to 93.75%. As the 3D space also allows the data (with nearly identical EDs) to have different spatial orientations for efficient clustering, samples were dichotomized into positive and negative with 100% efficiency, sensitivity, and specificity. Furthermore, owing to the ultrasensitive nanostructured transducer, the viral load was successfully quantified in the spiked control samples with a commendable detection sensitivity and limits of detection as low as 14.137 fM (E-gene). The device’s fast response time (<45 min) justifies its potential on-field applicability.

Abstract:The emergence of COVID-19 caused by the coronavirus SARS-CoV-2 has prompted a global pandemic that requires continuous research and monitoring. This study presents a design of an electrochemical biosensing platform suitable for the evaluation of monoclonal antibodies targeting the SARS-CoV-2 nucleocapsid (N) protein. Screen-printed carbon electrodes (SPCE) modified with gold nanostructures (AuNS) were applied to design a versatile and sensitive sensing platform. Electrochemical techniques, including electrochemical impedance spectroscopy (EIS) and square wave voltammetry (SWV), were used to investigate the interactions between immobilised recombinant N (rN) protein and several monoclonal antibodies (mAbs). The electrochemical characterisation of SPCE/AuNS/rN demonstrated a successful immobilisation of rN, enhancing the electron transfer kinetics. Affinity interactions between immobilised rN and four mAbs (mAb-4B3, mAb-4G6, mAb-12B2, and mAb-1G5) were explored. Although mAb-4B3 showed some non-linearity, the other monoclonal antibodies exhibited specific and well-defined interactions followed by the formation of an immune complex. The biosensing platform demonstrated high sensitivity in the linear range (LR) from 0.2 nM to 1 nM with limits of detection (LOD) ranging from 0.012 nM to 0.016 nM for mAb-4G6, mAb-12B2, and mAb-1G5 and limits of quantification (LOQ) values ranging from 0.035 nM to 0.139 nM, as determined by both EIS and SWV methods. These results highlight the system's potential for precise and selective detection of monoclonal antibodies specific to the rN. This electrochemical biosensing platform provides a promising route for the sensitive and accurate detection of monoclonal antibodies specific to the rN protein.

Abstract:The rampant spread and death rate of the recent coronavirus pandemic related to the SARS-CoV-2 respiratory virus have underscored the critical need for affordable, portable virus diagnostics, particularly in resource-limited settings. Moreover, efficient and timely monitoring of vaccine efficacy is needed to prevent future widespread infections. Electrochemical immunosensing poses an effective alternative to conventional molecular spectroscopic approaches, offering rapid, cost-effective, sensitive, and portable electroanalysis of disease biomarkers and antibodies; however, efforts to improve binding efficiency and sensitivity are still being investigated. Graphene quantum dots (GQDs) in particular have shown promise in improving device sensitivity. This study reports the development of a GQD-functionalized point-of-contamination device leveraging the selective interactions between SARS-CoV-2-specific Spike (S) Protein receptor binding domain (RBD) antigens and IgG anti-SARS-CoV-2-specific S-protein antibodies at screen-printed carbon electrode (SPCE) surfaces. The immunocomplexes formed at the GQD surfaces result in the interruption of the redox reactions that take place in the presence of a redox probe, decreasing the current response. Increased active surface area, conductivity, and binding via EDC/NHS chemistry were achieved due to the nanomaterial inclusion, with 5 nm, blue luminescent GQDs offering the best results. GQD concentration, EDC/NHS ratio, and RBD S-protein incubation time and concentration were optimized for the biosensor, and inter- and intra-screen-printed carbon electrode detection was investigated by calibration studies on multiple and single electrodes. The single electrode used for the entire calibration provided the best results. The label-free immunosensor was able to selectively detect anti-SARS-CoV-2 IgG antibodies between 0.5 and 100 ng/mL in the presence of IgM and other coronavirus antibodies with an excellent regression of 0.9599. A LOD of 2.028 ng/mL was found, offering comparable findings to the literature-reported values. The detection sensitivity of the sensor is further compared to non-specific IgM antibodies. The developed GQD immunosensor was compared to other low-oxygen content carbon nanomaterials, namely (i) carbon quantum dot (CQD), (ii) electrochemically reduced graphene oxide, and (iii) carbon black-functionalized devices. The findings suggest that improved electron transfer kinetics and increased active surface area of the CNs, along with surface oxygen content, aid in the detection of anti-SARS-CoV-2 IgG antibodies. The novel immunosensor suggests a possible application toward monitoring of IgG antibody production in SARS-CoV-2-vaccinated patients to study immune responses, vaccine efficacy, and lifetime to meet the demands for POC analysis in resource-limited settings.

Abstract:Efficient monitoring and assessing the efficacy of vaccines against SARS‐CoV‐2 hinges on swiftly detecting its antibodies. Current methods, often intricate and reliant on nanomaterials, struggle with sensitivity and complexity, limiting their broad use. The immunosensing platform utilized in this work is tailored for quick and precise identification of SARS‐CoV‐2‐specific IgG and Nucleocapsid antibodies. Noteworthy, this platform marks only the second attempt at detecting N‐protein antibodies. Unlike its counterparts, it employs traditional self‐assembled monolayers to foster a selective bond between SARS‐CoV‐2‐specific Nucleocapsid antibodies and a gold electrode coated with the N‐protein antigen. Remarkably, despite eschewing nanomaterials, it matches existing sensors′ sensitivity over a broad detection range (0.025 to 1 ng/mL) with an outstanding limit of detection (0.019 ng/mL). The method's simplicity, reliant solely on immunocomplex reactions, underscores the possibility of achieving effective binding without complex functionalization, rendering it affordable, specific, and highly sensitive. By streamlining the process and eliminating additional steps, this platform presents a practical solution for SARS‐CoV‐2 antibody detection. Its success in N‐protein antibody detection suggests promise for widespread application, especially in resource‐limited settings, for outbreak surveillance and vaccine efficacy evaluation. Highly sensitive, scalable, and rapid SARS-CoV-2 biosensor based on In2O3 nanoribbon transistors and phosphatase

Abstract:Developing convenient and accurate SARS-CoV-2 antigen test and serology test is crucial in curbing the global COVID-19 pandemic. In this work, we report an improved indium oxide (In2O3) nanoribbon field-effect transistor (FET) biosensor platform detecting both SARS-CoV-2 antigen and antibody. Our FET biosensors, which were fabricated using a scalable and cost-efficient lithography-free process utilizing shadow masks, consist of an In2O3 channel and a newly developed stable enzyme reporter. During the biosensing process, the phosphatase enzymatic reaction generated pH change of the solution, which was then detected and converted to electrical signal by our In2O3 FETs. The biosensors applied phosphatase as enzyme reporter, which has a much better stability than the widely used urease in FET based biosensors. As proof-of-principle studies, we demonstrate the detection of SARS-CoV-2 spike protein in both phosphate-buffered saline (PBS) buffer and universal transport medium (UTM) (limit of detection [LoD]: 100 fg/mL). Following the SARS-CoV-2 antigen tests, we developed and characterized additional sensors aimed at SARS-CoV-2 IgG antibodies, which is important to trace past infection and vaccination. Our spike protein IgG antibody tests exhibit excellent detection limits in both PBS and human whole blood ((LoD): 1 pg/mL). Our biosensors display similar detection performance in different mediums, demonstrating that our biosensor approach is not limited by Debye screening from salts and can selectively detect biomarkers in physiological fluids. The newly selected enzyme for our platform performs much better performance and longer shelf life which will lead our biosensor platform to be capable for real clinical diagnosis usage.

Abstract:Development of specific signal reporters with signal amplification effect are highly needed for sensitive and accurate detection of pathogen. Herein, we design a colorimetric immunosensing nanosystem based on liposome encapsulated quantum dots-sized MnO2 nanozyme (MnO2QDs@Lip) as a signal reporter for ultrasensitive and fast detection of SARS-CoV-2 antigen. The pathogenic antigens captured and separated by antibody-conjugated magnetic beads (MBs) are further connected with antibody-modified MnO2QDs@Lip to form a sandwich-like immunocomplex structure. After triggered release, MnO2 QDs efficiently catalyze colorless 3,3',5,5'-tetramethylbenzidine (TMB) to blue oxidized TMB, which can be qualitatively observed by naked eyes and quantitatively analyzed by UV-Vis spectra or smartphone platforms. By taking advantages of immuno-magnetic separation, excellent peroxidase-like catalytic activity of MnO2 QDs, and high encapsulation efficiency of MnO2QDs@Lip, ultrasensitive detection of SARS-CoV-2 antigen ranging from 0.1 pg/mL to 100 ng/mL is achieved within 20 min. The limit of detection (LOD) is calculated to be 65 fg/mL in PBS buffer. Furthermore, real clinical samples of SARS-CoV-2 antigens can be effectively identified by this immunosensing nanosystem with excellent accuracy. This proposed detection nanosystem provides a strategy for simple, rapid and ultrasensitive detection of pathogens and may shed light on the development of new POCT detection platforms for early diagnosis of pathogens and surveillance in public health.

Abstract:Following the emergence of the worldwide severe respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic, the need for innovative strategies and methodologies to facilitate cost-effective and early stage diagnosis has become evident. To prevent the outbreak of such contagious diseases, an efficient approach is systematic testing of the population. Here, we introduce a planar organic electrochemical transistor (OECT)-based immunosensor for the detection of SARS-CoV-2. The gold gate electrode of the poly(3,4-ethylenedioxy-thiophene):polystyrene sulfonate (PEDOT:PSS)-based OECTs was functionalized with SARS-CoV-2 antibodies. The detection mechanism is based on the specific interaction of the antibodies with the spike protein of the virus, allowing its direct detection and not requiring the prior formation of antibodies in the patient’s body. As a proof of concept, the ability of the immunosensor to detect the SARS-CoV-2 spike protein is assessed. The sensor exhibits a remarkably low limit of detection (LOD) of 10–17 M, with an incubation time of 30 min. Furthermore, the sensors demonstrate selectivity when exposed to similar proteins and stability, retaining their LOD after 20 days of storage. Lastly, the functionalization protocol may easily be adapted for other pathogens/biomarkers, enabling not only a point-of-care device for SARS-CoV-2 detection but also a versatile platform for biosensing applications.

Abstract:Rapid and reliable detection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antibody can provide immunological evidence in addition to nucleic acid test for the early diagnosis and on-site screening of coronavirus disease 2019 (COVID-19). All-solid-state biosensor capable of rapid, quantitative SARS-CoV-2 antibody testing is still lacking. Herein, we propose an electronic labelling strategy of protein molecules and demonstrate SARS-CoV-2 protein biosensor employing colloidal quantum dots (CQDs)-modified electrode. The feature current peak corresponding to the specific binding reaction of SARS-CoV-2 antigen and antibody proteins was observed for the first time. The unique charging and discharging effect depending on the alternating voltage applied was ascribed to the quantum confinement, Coulomb blockade and quantum tunneling effects of quantum dots. CQDs-modified electrode could recognize the specific binding reaction between antigen and antibody and then transduce it into significant electrical current. In the case of serum specimens from COVID-19 patient samples, the all-solid-state protein biosensor provides quantitative analysis of SARS-CoV-2 antibody with correlation coefficient of 93.8% compared to enzyme-linked immunosorbent assay (ELISA) results. It discriminates patient and normal samples with accuracy of about 90%. The results could be read within 1 min by handheld testing system prototype. The sensitive and specific protein biosensor combines the advantages of rapidity, accuracy, and convenience, facilitating the implement of low-cost, high-throughput immunological diagnostic technique for clinical lab, point-of-care testing (POCT) as well as home-use test.

Abstract:An electrochemical biosensor was constructed by convectively assembling polystyrene (PS) beads on a gold electrode to detect SAR‐Cov‐2 virus via partial nanochannel blockage by virus particles. Nanochannel blockage hinders the diffusion of a redox probe and causes attenuation of oxidation current, allowing quantification of the viral load by measuring the changes in oxidation currents before and after virus incubation. The development of simple, cost‐effective, rapid, and quantitative diagnostic tools remains critical to monitor infectious COVID‐19 disease. Although numerous diagnostic platforms, including rapid antigen tests, are developed and used, they suffer from limited accuracy, especially when tested with asymptomatic patients. Here, a unique approach to fabricate a nanochannel‐based electrochemical biosensor that can detect the entire virion instead of virus fragments, is demonstrated. The sensing platform has uniform nanoscale channels created by the convective assembly of polystyrene (PS) beads on gold electrodes. The PS beads are then functionalized with bioreceptors while the gold surface is endowed with anti‐fouling properties. When added to the biosensor, SARS‐CoV‐2 virus particles block the nanochannels by specific binding to the bioreceptors. The nanochannel blockage hinders the diffusion of a redox probe; and thus, allows quantification of the viral load by measuring the changes in the oxidation current before and after virus incubation. The biosensor shows a low limit of detection of ≈1.0 viral particle mL−1 with a wide detection range up to 108 particles mL−1 in cell culture media. Moreover, the biosensor is able to differentiate saliva samples with SARS‐CoV‐2 from those without, demonstrating the potential of this technology for translation into a point‐of‐care biosensor product.

Abstract:In this study, a metasurface plasmon resonance (MetaSPR) competitive or surrogate virus neutralization test (MSPR VNT) is established. Novel nanomaterial is combined with the optimized MetaSPR chip for dual signal amplification. This process has a low non‐specific reaction and can accurately calculate the IC50 value. In addition, the MSPR VNT miniaturized optical biosensor instrument has a high throughput, rapid, and accurate quantification of SARS‐CoV‐2 variants or other infectious diseases. A robust serological test is required to detect neutralizing antibodies (NAbs) against severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2). A nanomaterial‐coupled metasurface plasmon resonance (MetaSPR) biosensor is developed for rapid multiplexing quantitative tests of the half maximal inhibitory concentration (IC50) value of SARS‐CoV‐2 NAbs in human sera. This biosensor is applied to a MetaSPR competitive or surrogate virus neutralization test (MSPR VNT) used for quantifying NAb‐mediated blockage of the interaction between the human angiotensin‐converting enzyme 2 (hACE2) receptor and the receptor‐binding domain (RBD) of the SARS‐CoV‐2 spike protein. Novel nanomaterial, gold platinum nanoflowers (Au@Pt NFs), is combined with the optimized MetaSPR chip for dual signal amplification. The clinical utility of this biosensor is validated in 40 participants, and the results are confirmed with the IC50 values calculated using the pseudovirus neutralization test. Similarly, the neutralization titer of Omicron and Delta variants and the ability of NAbs to resist these variants are monitored. The MSPR VNT miniaturized optical biosensor instrument has a high throughput and enables one‐step, rapid, and accurate quantification of SARS‐CoV‐2 variants or viruses causing other infectious diseases, which is applicable in vaccine trials, immune response studies, and epidemiological studies.

Abstract:The current global outbreak of coronavirus (COVID-19) continues to be a severe threat to human health. Rapid, low-cost, and accurate antigen detection methods are very important for disease diagnosis. The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) nucleocapsid protein (N-Protein) is often used as the diagnostic and screening for coronavirus detection. To this end, we propose and experimentally validate a highly sensitive whispering gallery mode (WGM) optical cylindrical micro-resonator (CMR) for bioimmunoassay detection. To study the biokinetic process of immunoassay, the surface of the WGM micro-resonator is functionalized with N-Protein monoclonal antibody (N-Protein-mAb), which led to the specific detection of N-Proteins. The spectral characteristics of the WGM resonance dip were investigated, and it is found that the transmission spectrum of WGM shows a monotonically increasing red-shift as a function of recording time. The WGM red-shift is due to the antibody-antigen reaction and can be used for the analysis of the immunoassay process. The wavelength shift is shown to be proportional to the concentration of N-Protein, which ranges between 0.1 and 100 $mu ext{g}$ /mL. Finally, different types of samples (concentration of $10~mu ext{g}$ /mL of N-Protein) were prepared and tested to simulate the specificity of the sensor in the practical application environment. This method has the merits of a rapid assay, lower expense, easy preparation, and miniaturization, which makes the sensor have the potential for broad applications in the field of biochemistry and biomedical detection.

Abstract:Background: The pandemic of the new coronavirus infection, COVID-19, is currently ongoing in the world. Over the years, the pathogen, SARS-CoV-2, has undergone a series of mutational genome changes, which has led to the spread of various genetic variants of the virus. Meanwhile, the methods used to diagnose SARS-CoV-2, to establish the disease stage and to assess the immunity, are nonspecific to SARS-CoV-2 variants and time-consumable. Thus, the development of new methods for diagnosing COVID-19, as well as their implementation in practice, is currently an important direction. In particular, application of systems based on chemically modified fluorescent microspheres (with a multiplex assay for target protein molecules) opens great opportunities. Aim: development of a microfluidic diagnostic test system based on fluorescent microspheres for the specific detection of immunoglobulins G (IgG) to SARS-CoV-2. Methods: A collection of human serum samples was characterized using enzyme-linked immunosorbent assay (ELISA) and commercially available reagent kits. IgG to SARS-CoV-2 in the human serum were detected by the developed immunofluorescent method using microspheres containing the chemically immobilized RBD fragment of the SARS-CoV-2 (Kappa variant) viral S-protein. Results: The level of IgG in the blood serum of recovered volunteers was 9-300 times higher than that in apparently healthy volunteers, according to ELISA (p0.001). Conjugates of fluorescent microspheres with the RBD-fragment of the S-protein, capable of specifically binding IgG from the blood serum, have been obtained. The immune complexes formation was confirmed by the fluorescence microscopy data; the fluorescence intensity of secondary antibodies in the immune complexes formed on the surface of microspheres was proportional to the content of IgG (r 0.963). The test system had a good predictive value (AUC 70.3%). Conclusion: A test system has been developed, based on fluorescent microspheres containing the immobilized RBD fragment of the SARS-CoV-2 S-protein, for the immunofluorescent detection of IgG in the human blood serum. When testing the system on samples with different levels of IgG to SARS-CoV-2, its prognostic value was shown. The obtained results allow us to present the test system as a method to assess the level of immunoglobulins to SARS-CoV-2 in the human blood serum for the implementation in clinical practice. The test system can also be integrated into various microfluidic systems to create chips and devices for the point-of-care diagnostics.

Abstract:Persistent luminescence nanomaterials can remain luminescence when the light source is turned off, which exhibits promise in biosensor and bioimaging fields since they have the ability to completely eradicate tissue autofluorescence. Although significant progress has been made in the persistent luminescence biosensing, there is still a dearth of long-afterglow detection platform with low limit of detection (LOD) and high sensitivity. Herein, Zn 2 GeO 4 :Mn, Cr persistently luminescent nanorods (PLNRs) with superior persistent luminescence and long afterglow time were developed. The addition of Cr 3+ manifestly improves persistent luminescence intensity and afterglow duration through creating a deep defect trap. Then the biosensors were constructed by combining the Zn 2 GeO 4 :Mn,Cr PLNRs-antibody and Fe 3 O 4 magnetic nanoparticles (MNPs)-antibody for nucleocapsid protein detection based on electrostatic attraction. The LOD value for nucleocapsid protein realizes as low as 39.82 ag/mL, which is much lower than the previously reported persistent luminescent-based biosensors. Accordingly, the low detection sensitivity is attributed to fluorescence resonance energy transfer. In addition, high specificity is also achieved. Therefore, the as-prepared Zn 2 GeO 4 :Mn,Cr persistently luminescent materials can act as the promising candidate in biosensors applications. This strategy provides effective guidance for the development of biosensing platforms with high sensitivity and specificity.

Abstract:This article aims to present a comparative study of three polypyrrole-based molecularly imprinted (MIP) systems for the detection of the recombinant severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) nucleocapsid protein (rN). The rN is known for its relatively low propensity to mutate compared to other SARS-CoV-2 antigens. The aforementioned systems include screen-printed carbon electrodes (SPCE) modified with gold nanostructures (MIP1), platinum nanostructures (MIP2), and the unmodified SPCE (MIP3) for control. Pulsed amperometric detection (PAD) was employed as the detection technique, offering the advantage of label-free detection without the need for an additional redox probe. Calibration curves were constructed using the obtained data to evaluate the response of each system. Non-imprinted systems were also tested in parallel to evaluate the contribution of non-specific binding and assess the affinity sensor's efficiency. The analysis of calibration curves revealed that the AuNS-based MIP1 system exhibited the lowest contribution of non-specific binding and displayed a better fit with the chosen fitting model compared to the other systems. Further analysis of this system included determining the limits of detection (LOD) (51.2 ± 2.8 pg/mL), the limit of quantification (LOQ) (153.9 ± 8.3 pg/mL), and a specificity test using a recombinant receptor-binding domain of SARS-CoV-2 spike protein as a control. Based on the results, the AuNS-based MIP1 system demonstrated high specificity and sensitivity for the label-free detection of SARS-CoV-2rN. The utilization of PAD without the need for additional redox probes makes this sensing system convenient and valuable for rapid and accurate virus detection.

Abstract:The rapid spread of the Coronavirus Disease 2019 (COVID-19) pandemic, caused by the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) pathogen has generated a huge international public health emergency. Currently the reference diagnostic technique for virus determination is Reverse Transcription Polymerase Chain Reaction (RT-PCR) real time analysis that requires specialized equipment, reagents and facilities and typically 3–4 h to perform. Thus, the realization of simple, low-cost, small-size, rapid and point-of-care diagnostics tests has become a global priority. In response to the current need for quick, highly sensitive and on-site detection of the SARS-CoV-2 virus in several aqueous solutions, a specific molecularly imprinted polymer (MIP) receptor has been designed, realized, and combined with an optical sensor. More specifically, the proof of concept of a SARS-CoV-2 sensor has been demonstrated by exploiting a plasmonic plastic optical fiber sensor coupled with a novel kind of synthetic MIP nano-layer, especially designed for the specific recognition of Subunit 1 of the SARS-CoV-2 Spike protein. First, we have tested the effectiveness of the developed MIP receptor to bind the Subunit 1 of the SARS-CoV-2 spike protein, then the results of preliminary tests on SARS-CoV-2 virions, performed on samples of nasopharyngeal (NP) swabs in universal transport medium (UTM) and physiological solution (0.9% NaCl), were compared with those obtained with RT-PCR. According to these preliminary results, the sensitivity of the proposed optical-chemical sensor proved to be higher than the RT-PCR one. Furthermore, a relatively fast response time (about 10 min) to the virus was obtained without the use of additional reagents.

Abstract:ABSTRACT Spreading manure containing antibiotics in agriculture is assumed to stimulate the dissemination of antibiotic resistance in soil bacterial populations. Plant roots influencing the soil environment and its microflora by exudation of growth substrates might considerably increase this effect. In this study, the effects of manure from pigs treated with sulfadiazine (SDZ), here called SDZ manure, on the abundance and transferability of sulfonamide resistance genes sul1 and sul2 in the rhizosphere of maize and grass were compared to the effects in bulk soil in a field experiment. In plots that repeatedly received SDZ manure, a significantly higher abundance of both sul genes was detected compared to that in plots where manure from untreated pigs was applied. Significantly lower abundances of sul genes relative to bacterial ribosomal genes were encountered in the rhizosphere than in bulk soil. However, in contrast to results for bulk soil, the sul gene abundance in the SDZ manure-treated rhizosphere constantly deviated from control treatments over a period of 6 weeks after manuring, suggesting ongoing antibiotic selection over this period. Transferability of sulfonamide resistance was analyzed by capturing resistance plasmids from soil communities into Escherichia coli. Increased rates of plasmid capture were observed in samples from SDZ manure-treated bulk soil and the rhizosphere of maize and grass. More than 97% of the captured plasmids belonged to the LowGC type (having low G+C content), giving further evidence for their important contribution to the environmental spread of antibiotic resistance. In conclusion, differences between bulk soil and rhizosphere need to be considered when assessing the risks associated with the spreading of antibiotic resistance.

Abstract:The prompt and accurate point-of-care test (POCT) for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in infected persons or virus-containing environmental samples is of great importance. The present work reports a highly integrated electrochemiluminescence/electrochemical (ECL/EC) sensor for determination of SARS-CoV-2 pseudoviruses, in which bio-recognition element (SARS-CoV-2 IgG antibody), bifunctional probe (tris (2,2'-bipyridyl) ruthenium (Ru(bpy)32+)), and amplification material (gold nanoparticles (Au NPs)) are designed into bipolar silica nanochannel array (bp-SNA). bp-SNA consisting of homogeneous two-layer mesoporous silica films bears inner silanol groups and outer amino groups, generating a solid "electrostatic nanocage" for stable confinement of Ru(bpy)32+ and Au NPs inside the nanochannels and further providing functional sites for covalent modification of SARS-CoV-2 IgG antibody. Owing to the preconcentration capacity of bp-SNA and amplified effect of Au NPs, ECL or EC signals of Ru(bpy)32+ can be remarkably promoted and thereby increase the analytical performance, which can be diminished by immunorecognization of target SARS-CoV-2 pseudoviruses on the sensing interface. The developed integrated ECL/EC sensor based on Ru@AuNPs/bp-SNA modified solid indium tin oxide electrode enables the sensitive analysis of SARS-CoV-2 pseudoviruses by ECL mode with a linear range of 50 TU mL-1-5000 TU mL-1, as well as the EC mode with a linear range of 100 TU mL-1-5000 TU mL-1. Moreover, the designed sensor showed satisfactory results in the analyses of saliva and pond water samples. When flexible electrode substate (polyethylene terephthalate) is employed, Ru@AuNPs/bp-SNA has great potential to integrate with KN95 face masks for direct detection of SARS-CoV-2 pseudoviruses produced from breathing, talking and coughing processes, which could provide an efficient platform for POCT diagnosis.
