Abstract:Biodegradable polymer nanoparticle drug delivery systems provide targeted drug delivery, improved pharmacokinetic and biodistribution, enhanced drug stability and fewer side effects. These drug delivery systems are widely used for delivering cytotoxic agents. In the present study, we synthesized GC/5-FU nanoparticles by combining galactosylated chitosan (GC) material with 5-FU, and tested its effect on liver cancer in vitro and in vivo. The in vitro anti-cancer effects of this sustained release system were both dose- and time-dependent, and demonstrated higher cytotoxicity against hepatic cancer cells than against other cell types. The distribution of GC/5-FU in vivo revealed the greatest accumulation in hepatic cancer tissues. GC/5-FU significantly inhibited tumor growth in an orthotropic liver cancer mouse model, resulting in a significant reduction in tumor weight and increased survival time in comparison to 5-FU alone. Flow cytometry and TUNEL assays in hepatic cancer cells showed that GC/5-FU was associated with higher rates of G0-G1 arrest and apoptosis than 5-FU. Analysis of apoptosis pathways indicated that GC/5-FU upregulates p53 expression at both protein and mRNA levels. This in turn lowers Bcl-2/Bax expression resulting in mitochondrial release of cytochrome C into the cytosol with subsequent caspase-3 activation. Upregulation of caspase-3 expression decreased poly ADP-ribose polymerase 1 (PARP-1) at mRNA and protein levels, further promoting apoptosis. These findings indicate that sustained release of GC/5-FU nanoparticles are more effective at targeting hepatic cancer cells than 5-FU monotherapy in the mouse orthotropic liver cancer mouse model.

Abstract:The primary objectives of the present work are to synthesize chitosan nanoparticles (CS-NPs) loaded with 5-fluorouracil, characterize the nanoparticles using various techniques, assess the impact of 5-FU concentration on encapsulation efficiency, study the sustained drug release from the nanoparticles, and evaluate their biocompatibility and cytotoxicity against specific cancer cell lines (MCF-7, U266, HEP G2). 5-FU are formulated with CS-NPs using sodium tripolyphosphate (STTP) and sodium hexametaphosphate (SHMP) through ionic crosslinking and are characterized via FTIR, XRD, SEM, TEM, and DLS measurements. From the SEM and TEM results, it was determined that the particle size of 5-FU-CS-NPs-SHMP air-dried and freeze-dried samples ranged between 90–120 nm and 60–80 nm, respectively. For the 5-FU-CS-NPs-TPP air-dried and freeze-dried formulation, the particle size ranged between 30–50 nm and 10–20 nm, respectively. Loading measurements confirmed a slight reduction in encapsulation efficiency from 99.8 to 98.6% when increasing the concentration of 5-fluorouracil from a ratio of 0.1 to 0.3 relative to the concentration of CS and TPP/SHMP. The in vitro drug release studies demonstrated sustained drug release, and the resulting data was fitted with kinetic models. The cytotoxicity assay indicated that the prepared drug-loaded formulation exhibited greater toxicity towards MCF-7 cancer cells compared to U266 and HEP G2 cancer cells.

Abstract:Our current study reports the successful synthesis of thiolated chitosan-based nanoparticles for targeted drug delivery of 5-Fluorouracil. This process was achieved through the ionic gelation technique, aiming to improve the efficacy of the chemotherapeutic moiety by modifying the surface of the nanoparticles (NPs) with a ligand. We coated these NPs with hyaluronic acid (HA) to actively target the CD44 receptor, which is frequently overexpressed in various solid malignancies, including breast cancer. XRD, FTIR, SEM, and TEM were used for the physicochemical analysis of the NPs. These 5-Fluorouracil (5-FU) loaded NPs were evaluated on MDA-MB-231 (a triple-negative breast cell line) and MCF-10A (normal epithelial breast cells) to determine their in vitro efficacy. The developed 5-FU-loaded NPs exhibited a particle size within a favorable range (< 300 nm). The positive zeta potential of these nanoparticles facilitated their uptake by negatively charged cancer cells. Moreover, they demonstrated robust stability and achieved high encapsulation efficiency. These nanoparticles exhibited significant cytotoxicity compared to the crude drug (p < 0.05) and displayed a promising release pattern consistent with the basic diffusion model. These traits improve the pharmacokinetic profile, efficacy, and ability to precisely target these nanoparticles, offering a potentially successful anticancer treatment for breast cancer. However, additional in vivo assessments of these formulations are obligatory to confirm these findings.

Abstract:Conventional chemotherapy is ineffective for colorectal cancer as the drug moiety does not reach its intended site. 5-Fluorouracil (5-FU) microspheres aimed at colon-specific drug delivery have been developed by using the solvent evaporation technique in this study. The influence of utilizing different percentages of resistant starch-pectin polymer on the drug loading, encapsulation efficiency, and release behavior of the drug was investigated. The characterization of prepared microspheres was done by shape, surface morphology, drug loading, encapsulation efficiency and in vitro drug release studies. Drug-polymer interactions and drug stability were investigated by Differential Scanning Calorimetry (DSC) and FTIR spectroscopy. The 5-Fluorouracil polymeric microspheres that were formulated utilizing a resistant starch (RS) and pectin polymer displayed ideal physicochemical properties and spherical particles. The targeted microspheres demonstrated a high drug loading, encapsulation efficiency, and a satisfactory drug release pattern over the time period of 8 hours. Studies on drug release conducted under conditions that simulate stomach to colon transit have demonstrated that the drug was prevented from being released in the physiological conditions of the stomach and small intestine. Creating a new multiparticulate system using RS-pectin microspheres in order to forecast the protection that the combination might provide to the dosage form that will ensure localized drug release in the colon. Finally, it was found that the 5-Fluorouracil polymeric microsphere is a suitable microcarrier for effective colon-specific drug delivery tools with increased chemotherapeutic efficacy. Abstract:Colorectal cancer (CRC) is the third most common cancer worldwide, acting as a significant public health problem. 5-Fluorouracil (5-FU) is a key chemotherapy for various types of cancer, due to its broad anticancer activity. However, the emergence of drug resistance is a considerable limitation in the clinical application of 5-FU. Quercetin (QC) is proposed as an adjuvant therapy to minimize drug resistance to chemotherapeutics and enhance their pharmacological efficacy. The oral delivery of 5-FU and QC is challenged by poor aqueous solubility of QC and poor cellular permeability of 5-FU. To solve this issue, novel polylactide-co-glycolide (PLGA) combinatorial nanoparticles loading 5-FU and QC were prepared to deliver them directly to the colon. These sustained-release combinatorial nanoparticles recorded a significant decrease in cancer cell proliferation, C-reactive protein (CRP) level, and Interleukin-8 (IL-8) expression by 30.08%, 40.7%, and 46.6%, respectively. The results revealed that this combination therapy may offer a new strategy for the targeted delivery of chemotherapeutics to the colon.

Abstract:Chitosan succinate is distinguished by its ability to shield the loaded drug from the acidic environment, localize and keep the drug at the colon site, and release the drug over an extended time at basic pH. The current study attempts to develop polyelectrolyte liposomes (PEL), using chitosan and chitosan succinate (CSSC), as a carrier for liposomal-assisted colon target delivery of 5 fluorouracil (5FU). The central composite design was used to obtain an optimized formulation of 5FU-chitosomes. The chitosan-coated liposomes (chitosomes) were prepared by thin lipid film hydration technique. After that, the optimized formulation was coated with CSSC, which has several carboxylic (COOH) groups that produce an anionic charge that interacts with the cation NH2 in chitosan. The prepared 5FU-chitosomes formulations were evaluated for entrapment efficiency % (EE%), particle size, and in vitro drug release. The optimized 5FU-chitosomes formulation was examined for particle size, zeta potential, in vitro release, and mucoadhesive properties in comparison with the equivalent 5FU-liposomes and 5FU-PEL. The prepared 5FU-chitosomes exhibited high EE%, small particle size, low polydispersity index, and prolonged drug release. PEL significantly limited the drug release at acidic pH due to the deprotonation of carboxylate ions in CSSC, which resulted in strong repulsive forces, significant swelling, and prolonged drug release. According to a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay, PEL treatment significantly decreased the viability of HT-29 cells. When compared to 5FU-liposome and 5FU-chitosome, the in vivo pharmacokinetics characteristics of 5FU-PEL significantly (p < 0.05) improved. The findings show that PEL enhances 5FU permeability, which permits high drug concentrations to enter cells and inhibits the growth of colon cancer cells. Based on the current research, PEL may be used as a liposomal-assisted colon-specific delivery.

Abstract:Targeting critical pathways of cancer cells using combination drugs is gaining significant importance as drug resistance is lowered and drug efficacy is improved even at low concentrations. Docetaxel and 5-fluorouracil are chemotherapeutic drugs prescribed for patients with solid tumours. While coated polymeric nanoparticles can combat drug resistance by sequential release of drugs, this paper proposes to encapsulate docetaxel and 5-fluorouracil in chitosan nanoparticles. To further improve the solubility of chitosan nanoparticles, they are crosslinked with polyethylene glycol using formaldehyde as crosslinker. Various techniques were used to characterize the nanoparticles. HR-SEM images indicated the formation of nanoparticles, drug loading into the chitosan nanoparticles and crosslinking of polyethylene glycol to chitosan nanoparticles. The experimental strategies were designed to evaluate the synergistic combination of the drugs in nanoparticles against AGS cell lines. It was found that the drugs loaded in chitosan nanoparticles synergistically inhibited the cell viability of AGS cells with a combination effect at 0.414. The combined effect of the drugs was even more effective, with value at 0.23 in the case of polyethylene glycol crosslinked-drug-loaded chitosan nanoparticles. The DPPH scavenging activity of drugs encapsulated in polyethylene glycol crosslinked chitosan nanoparticles was more effective at 57.39% compared to drug-alone counter parts. To further confirm the synergistic efficacy of the drugs loaded in nanoparticles in inducing apoptosis, changes were observed using acridine orange /ethidium bromide staining. Results indicated that combination effect of drugs in nanoparticles was much higher in AGS cells when compared to drug-alone incubated groups.

Abstract:The goal of the current study was to target 7-ethyl-10-hydroxycamptothecin (SN38) orally to colon tumours by synthesizing a targeting polymer. To achieve the optimum delivery for SN38, initially methoxy-polyethylene glycol (mPEG)-chitosan was synthesized and then nanoparticles were developed through ionic gelation between mPEG-chitosan and hyaluronic acid as a ligand for cell-surface glycoprotein CD44 receptor. The SN38 was loaded in nanoparticles (SN38-NPs) using the non-covalent physical adsorption method. The size of the optimized SN38-NPs was 226.7 nm, encapsulation efficiency was 89.23% and drug content was 7.98 ± 0.54% in the optimum formulation. The attachment of mPEG to chitosan was confirmed by proton nuclear magnetic resonance. The results of differential scanning calorimetry and Fourier transforms infra-red analysis indicated that SN38 existed in amorphous form and functional groups of SN38 protected in the formulations which could be a sign of suitable encapsulation of SN38 in SN38-NPs. In vitro study indicated that SN38-NPs were more potent against the cancer cells than free SN38. The cellular uptake of SN38-NPs improved up to 1.6-fold against human colorectal adenocarcinoma (Caco-2) cells. Moreover, SN38-NPs remarkably demonstrated superior anti-tumor efficacy in contrary to pure SN38. This suggests the advantage of SN38-NPs as a potent oral drug carrier which could be further explored for clinical investigations.
Targeted anticancer drug delivery using chitosan, carbon quantum dots, and aptamers to deliver Ganoderic acid and 5‐fluorouracil

Abstract:Breast cancer is a deadly disease that usually affects women. The nano‐drug delivery system's ligand‐functionalized nanoparticles have great cancer‐treatment potential. This study developed a potential method for drug loading and sustained release of breast cancer model medications 5‐fluorouracil (5‐FU) and Ganoderic acid (GA). CS/Apt/COQ NPs were produced. FT‐IR, XRD, DLS, zeta potentiometer, and SEM analyzed NP's chemical structure, particle size, and shape. In vitro MTT experiments examined MCF‐7 cell viability. Real‐time PCR quantified cell apoptosis. XRD and FTIR showed nanocarrier generation, crystalline structure, and molecular interactions. DLS indicated that nanocarriers contain 250.6 nm NPs with PDI 0.057. Zeta potential and SEM indicated their spherical shape and +37.8 mV surface charge. pH 5.4 released most 5‐FU and GA in 48 h. 5‐FU entrapment efficiency was 84.7 ± 5.2 and GA 80.2% ± 2.3. The 5‐FU‐GA‐CS‐CQD‐Apt group killed the most MCF‐7 cells, 57.9%. 5‐FU and GA in CS‐CQD‐Apt increased flow cytometry‐induced apoptosis. 5‐FU‐GA‐CS‐CQD‐Apt increased Caspase 9 and decreased Bcl2. Thus, pH‐sensitive NPs may release 5‐FU and GA to treat breast cancer.

A novel and simple oral colon-specific drug delivery system based on the pectin/modified nano-carbon sphere nanocomposite gel films

Abstract:The 3-aminopropyltriethoxysilane modified nano-carbon sphere (MNCS) was added into pectin-Ca2+ film to improve the controlled release properties of the pectin-based oral colon-specific drug delivery system (OCDDS). The FT-IR measurements indicated the successful modification of nano-carbon sphere via silylation reaction and the electrostatic interaction between the pectin molecules and MNCS in the composite film. The FE-SEM showed the pore structure when the MNCS was mingled with the pectin. The 5-fluorouracil (5-FU) was employed as the drug model and the controlled release properties of the corresponding OCDDSs were determined. The values of the encapsulation efficiency ranged from 30.1% to 52.6%. All composite film based OCDDSs presented higher encapsulation efficiency than single pectin-Ca2+ based OCDDS. The drug release studies emerged that almost all the OCDDSs from composite films presented better release properties than single pectin-Ca2+ based OCDDS. The sample C revealed best release performance with the cumulative release rate of 32.17%, 22.77% and 63.89% in the simulated gastric fluid, small intestinal fluid and colon fluid, respectively. In addition, the kinetics studies were performed to analyze the release data. The cytotoxicity assay indicated good biocompatibility of the composite carriers.

Abstract:Targeted drug delivery to tumor cells may be possible using nanoparticles containing human therapeutic drugs. The present study was carried out to develop cisplatin (CP) and 5-fluorouracil (FA) encapsulated chitosan nanoparticles (CSNPs), crosslinked with sodium tripolyphosphate (TPP) by an ionic gelation method and in vitro release, promoting antibacterial and anticancer activities. The prepared CSNPs, before and after CP and FA encapsulation, have been studied using various characterization techniques such as FTIR, XRD, SEM, and TEM-SAED patterning. The composites were well-dispersed, with an average particle size diameter of about 395.3 ± 14.3 nm, 126.7 ± 2.6 nm, and 82.5 ± 2.3 nm, respectively. In vitro release studies indicated a controlled and sustained release of CP and FA from the CSNPs, with the release amounts of 72.9 ± 3.6% and 94.8 ± 2.9%. The antimicrobial activity of the CSNPs-FA (91.37 ± 4.37% and 89.28 ± 3.19%) showed a significantly better effect against E. coli and S. aureus than that shown by the CSNPs-CP (63.41 ± 3.84% and 57.62 ± 4.28%). The HCT-116 cell lines were selected for in vitro cell cytotoxicity and live/dead assay to evaluate the preliminary anticancer efficacy of the CSNPs-CP and CSNPs-FA towards successfully inhibiting the growth of cancer cells.

Abstract:Ursolic acid is widely used as an effective anticancer drug for the treatment of various cancers. However, its poor water solubility, short circulation time in vivo, and lack of targeting have made it a burden for clinical applications. We report a self-assembled folate-modified pectin nanoparticle for loading ursolic acid ( NPs) and embed the anticancer drug hydroxycamptothecin to achieve synergistic treatment with ursolic acid. In addition, the galactose residue of the pectin molecule can be recognized by the asialoglycoprotein receptor on the surface of the liver cancer cell, promoting the rapid penetration and release of NPs intracellularly. In particular, the introduction of multiarm polyethylene glycol can improve the uniformity (106 nm) and concealment of the nanoparticles and avoid the early release of the drug or the toxicity to normal cells. NPs have a high drug loading (7.27 wt %) and embedding efficiency (19.84 wt %) and continuous circulation up to 80 h, leading to more apoptosis (91.61%). NP intracellular drug delivery will be a promising strategy.This article has not yet been cited by other publications.

Abstract:Green nanotechnology is considered a promising method to construct functional materials with significant anticancer activity, while overcoming the shortcomings of traditional synthesis process complexity and high organic solvents consumption. Thus, in this study, we report for the first time the rational design and green synthesis of functionalized 5-fluorouracil and curcumin co-loaded lysozyme-hyaluronan composite colloidal nanoparticles (5-Fu/Cur@LHNPs) for better targeted colorectal cancer therapy with minimized side effects. The functionalized 5-Fu/Cur@LHNPs exhibit stabilized particle size (126.1 nm) with excellent homogeneity (PDI = 0.1), favorable colloidal stabilities, and excellent re-dispersibility. In vitro cell experiments illustrate that the cellular uptake of 5-Fu/Cur@LHNPs was significantly improved and further promoted a higher apoptosis ratio of HCT-116 cells. Compared with the control group, the 5-Fu/Cur@LHNPs formulation group achieved effective inhibition (60.1 %) of colorectal tumor growth. The alcohol-free self-assembly method to construct 5-Fu/Cur@LHNPs is simple and safe for a translational chemotherapy drug, also to promote more robust delivery systems for treating colorectal cancer.

The resulting 5FU‐loaded magnetic nanocomposites (Cs‐Alg/IO 5FU NCs) show higher selectivity against colon cancer cells (HCT116, HT‐29) in 2D and 3D models, partially attributed to the synergistic effect of peel extract in Fe3O4 NPs, lower drug loading capacity, and encapsulation efficiency. This study aimed to synthesize dual‐crosslinked chitosan‐alginate nanocomposites (Cs‐Alg/IO NCs) using green‐synthesized iron oxide nanoparticles (IO, Fe3O4 NPs) for drug delivery application. The NCs, formed via ionotropic gelation with sodium tripolyphosphate (TPP) and calcium chloride (CaCl2) as crosslinking agents for Cs and Alg, respectively, and further characterized for physiochemical properties. The presence of Fe3O4 NPs and 5‐Fluorouracil (5FU) increased hydrodynamic size (153.12 ± 0.57 to 207.64 ± 3.59 nm). Morphological studies revealed NC agglomeration within nanoscales. 5FU‐loaded Cs‐Alg/IO NCs (Cs‐Alg/IO 5FU NCs) exhibited superparamagnetic behavior and sustained drug release under various pH conditions. Cytotoxicity assays on colon cancer cells (HCT116 and HT‐29) and normal colon cells (CCD‐112) indicated higher cancer cell selectivity (selectivity index, SI = 1.91) in 2D models. Elevated IC50 values in 3D models were linked to lower drug loading capacity (LC) and encapsulation efficiency (EE). Overall, the study highlights the potential of dual‐crosslinked biopolymer NCs with green‐synthesized Fe3O4 NPs for developing an anticancer drug delivery system in colorectal cancer treatment. Abstract:The present study aimed to formulate 5-fluorouracil loaded cross linked chitosan nanoparticles based on chemical cross-linking of low molecular weight chitosan with glutaraldehyde by reverse micelles technique as 5-FU is less hydrophobic, relatively potent, has a shorter half-life, is rapidly metabolized, less tolerated, and has low oral bioavailability; therefore, we aimed to formulate potential nanocarriers of 5-FU for efficient drug delivery to specific targeted areas of action, reduce oral toxicity, improve tolerability and therapeutic outcomes of 5-FU, in a restricted fashion to enhance the bioavailability of 5-FU. Nanoparticles were formulated by the reverse micelle method based on the chemical cross-linking of glutaraldehyde (25% aqueous solution) into a w/o emulsion in different ratios. LMWCH-NPs were characterized for post-formulation parameters by mean particle size, zeta potential, %age yield, loading/entrapment efficiency, Fourier transform infrared spectroscopy (FTIR), DSC/TGA, TEM, PXRD, drug release at pH 1.2, and pH 7.4. 5-FU loaded NPs showed a size range (198 nm-200 nm) and zeta potential (−39mV to −41mV), which ensured mechanical stability and increased retention time in blood vessels by the sustained release properties of biodegradable nanocarrier drug delivery systems. % age yield showed the range 92% to 96% while % LC ranged 2.0% to 3.4% and %EE ranged 40% to 43%. The TEM images showed spherical nanoparticles. FTIR revealed the compatibility between the drug and the cross-linked polymer. DSC/TGA ensured the thermal stability of the drug, while the solid-state stability of the drug-loaded cross-linked chitosan nanoparticles was evaluated by powder X-ray diffraction (PXRD) analysis. Drug release studies were performed using the dialysis bag technique at both pH (1.2 and 7.4) to mimic the gastrointestinal tract. Highly stable NPs displayed targeted release in phosphate buffer pH 7.4 at 37°C. Fickian diffusion was the predominant release with an R 2 value of 0.9975-0.9973—and an N value 0.45-0.53. Prepared nanoparticles are inert, biodegradable, and biocompatible drug delivery systems for sustained release of 5-FU with maximum therapeutic efficacy and bioavailability.

Abstract:The nanoparticle drug delivery system, which uses natural or synthetic polymeric material as a carrier to deliver drugs to targeted tissues, has a broad prospect for clinical application for its targeting, slow-release, and biodegradable properties. Here, we used chitosan (CTS) and hepatoma cell-specific binding molecule glycyrrhetinic acid to synthesize glycyrrhetinic acid-modified chitosan (GA-CTS). The synthetic product was confirmed by infrared (IR) spectra and hydrogen-1 nuclear magnetic resonance. The GA-CTS/5-fluorouracil (5-FU) nanoparticles were synthesized by combining GA-CTS and 5-FU and conjugating 5-FU onto the GA-CTS nanomaterial. The central composite design was performed to optimize the preparation process as CTS:tripolyphosphate sodium (TPP) weight ratio =5:1, 5-FU:CTS weight ratio =1:1, TPP concentration =0.05% (w/v), and cross-link time =50 minutes. GA-CTS/5-FU nanoparticles had a mean particle size of 193.7 nm, a polydispersity index of 0.003, a zeta potential of +27.4 mV, and a drug loading of 1.56%. The GA-CTS/5-FU nanoparticle had a protective effect on the drug against plasma degrading enzyme, and provided a sustained release system comprising three distinct phases of quick, steady, and slow release. Our study showed that the peak time, half-life time, mean residence time and area under the curve of GA-CTS/5-FU were longer or more than those of the 5-FU group, but the maximum concentration (C(max)) was lower. We demonstrated that the nanoparticles accumulated in the liver and have significantly inhibited tumor growth in an orthotropic liver cancer mouse model.

Abstract:The study aimed to develop and evaluate chitosan-based nanoparticles coated with TPGS for the targeted delivery of imatinib mesylate to colon cancer cells. Particle size and zeta potential analysis were within the acceptable range for targeting colon cancer. CS-IMT-TPGS-NPs had a significant positive zeta potential of 30.4 mV, suggesting improved cellular intake. FE-SEM and TEM demonstrated that the nanoparticles appeared spherical, smooth, and did not aggregate, with a visible TPGS coating. XRD confirmed that crystalline imatinib transitioned to an amorphous state during nano formulation. In-vitro tests on HCT-116 cells demonstrated that CS-IMT-TPGS-NPs outperformed free IMT regarding cytotoxicity, apoptosis induction, cellular uptake, and cell migration inhibition. Additionally, the nanoparticles were examined in vitro using mitochondrial membrane potential, DNA fragmentation, GAPDH relative gene expression, ROS estimation, and cell cycle analysis. The effect of therapy on expected colon-associated bacterial strains was also investigated. The biocompatibility of nanoparticles was assessed by hemolysis and platelet aggregation experiments. The anti-inflammatory impact was determined using the HET-CAM test. Non-Fickian diffusion at pH 5.5 resulted in sustained in-vitro drug release, with no initial burst. In-vivo investigations using albino Wistar rats suggest pharmacokinetic properties for produced nanoparticles, whereas histopathological examinations assess acute toxicity.

Abstract:Zinc-phosphate/hydroxyapatite hybrid form (ZP/HP) in core-shell nanostructure was developed and functionalized with both chitosan (CS@ZP/HP) and β-cyclodextrin (CD@ZP/HP) as bio-composite of enhanced physicochemical and biological properties. These structures were assessed as potential deliveries of 5-fluorouracil, exhibiting enhanced loading, release, and anti-cancer behaviors. The functionalization strongly prompted the loading effectiveness to be 301.3 mg/g (CS@ZP/HP) and 342.8 mg/g (CD@ZP/HP) instead of 238.9 mg/g for ZP/HP. The loading activities were assessed based on the hypotheses of traditional kinetic and isotherm models, alongside the computational variables of the monolayer model with a single energetic site as an advanced isotherm model. The functionalized versions exhibit much greater loading efficacy compared to ZP/HP as a result of the increment in the density of the existing loading sites [Nm(5-Fu) = 78.85 mg/g (ZP/HP), 93.87 mg/g (CS@ZP/HP), and 117.8 mg/g (CD@ZP/HP)]. Furthermore, the loading energies of approximately 40 kJ/mol, together with the loading potential of each receptor (n > 1) and Gaussian energies of approximately 8 kJ/mol, indicate the physical entrapment of 5-Fu molecules according to a vertical orientation. The materials mentioned verify long-term and continuous release characteristics. Following the modification processes, this behavior became faster as both CS@ZP/HP and CD@ZP/HP displayed complete release within 120 h at pH 1.2. The kinetic studies and diffusing exponent (>0.45) indicate that release characteristics are controlled by both diffusion and erosion processes. These carriers also markedly increase the cytotoxicity of 5-Fu against HCT-116 colorectal cancer cell lines: 5-Fu-ZP/HP (3.2% cell viability), 5-Fu-CS@ZP/HP (1.12% cell viability), and 5-Fu-CD@ZP/HP (0.63% cell viability).

Abstract:In recent years nanotechnologies have been applied to human health with promising results, especially in the field of drug delivery. Polymeric nanoparticles (NPs) have garnered much importance in controlled drug delivery owing to their size. Chitosan (Cs) is a well-recognized biopolymer and Cs NPs have been widely explored in drug delivery. Nonetheless, reports pertaining to green synthesis of Cs NPs are scarce. Thus, in this study, green synthesis of Cs NPs was accomplished from raw mango peel extract. Spherical Cs NPs with positively charged surface of 33.4 mV was accomplished by this process. Cs NPs, in varied content, were integrated in a guar gum network matrix resulting in a nanocomposite hydrogel. The mechanical and thermal stability of the hydrogel improved upon addition of Cs NPs. The hydrogel exhibited smart swelling, good antioxidant and anti-inflammatory propensities. Cs NPs encapsulating 5-Fluorouracil demonstrated a controlled release drug profile in the colorectum and the kinetics implied the anomalous nature of drug release mechanism. The exposure of the drug-loaded nanocomposite hydrogel displayed improved anticancer effects in HT-29 colon cancer cells. Taken altogether, this study puts forth the greater efficacy of Cs NPs in controlled drug delivery for anticancer therapy.
