Abstract:Pyroptosis-based nanomedicine shows great potential to unchoke the bottlenecks of cancer immunotherapy, but hindered by precise and specific regulation of pyroptosis in tumor cells. Herein, we report an endogenous/exogenous stimulus-activatable unimolecular polyprodrug as nano-inducer (denoted as CCNP) to precisely regulate the process of gasdermin E (GSDME)-medicated tumor pyroptosis. This nano-inducer is specifically switched by noninvasive laser radiation and tumor microenvironment reactive oxygen species (ROS) and glutathione (GSH), and activates caspase-3, thus triggering GSDME-mediated pyroptosis. This three-pronged induced model of pyroptosis can fully play the therapeutic potency of combined chemotherapy and photodynamic therapy exported by nano-inducer, suggests a new perspective on how to multi-stimulus-responsive nanomedicine induces and precisely regulates the tumor cell pyroptosis for assisting immune checkpoint blockade therapy.

Abstract:Abstract Precise and efficient image-guided immunotherapy holds great promise for cancer treatment. Here, we report a self-accelerated nanoplatform combining an aggregation-induced emission luminogen (AIEgen) and a hypoxia-responsive prodrug for multifunctional image-guided combination immunotherapy. The near-infrared AIEgen with methoxy substitution simultaneously possesses boosted fluorescence and photoacoustic (PA) brightness for the strong light absorption ability, as well as amplified type I and type II photodynamic therapy (PDT) properties via enhanced intersystem crossing process. By formulating the high-performance AIEgen with a hypoxia-responsive paclitaxel (PTX) prodrug into nanoparticles, and further camouflaging with macrophage cell membrane, a tumor-targeting theranostic agent is built. The integration of fluorescence and PA imaging helps to delineate tumor site sensitively, providing accurate guidance for tumor treatment. The light-induced PDT effect could consume the local oxygen and lead to severer hypoxia, accelerating the release of PTX drug. As a result, the combination of PDT and PTX chemotherapy induces immunogenic cancer cell death, which could not only elicit strong antitumor immunity to suppress the primary tumor, but also inhibit the growth of distant tumor in 4T1 tumor-bearing female mice. Here, we report a strategy to develop theranostic agents via rational molecular design for boosting antitumor immunotherapy.

Abstract:Despite immunotherapy involving immune checkpoint inhibitors (ICIs) have revolutionized cancer therapy, the clinical efficacy is limited due to ICI resistance. Pyroptosis is a gasdermin-mediated programmed cell death that enhances responses to ICIs. However, nontargeted elicitation of pyroptosis may induce systemic side effects and toxicity. Therefore, we reasonably design and construct a tumor-specific prodrug that combines the heat shock protein 90 inhibitor tanespimycin (17-AAG) with the photosensitizer chlorin e6 (Ce6) to induce pyroptosis, by utilizing the high glutathione level in the tumor microenvironment. The released Ce6 and 17-AAG produce reactive oxygen species by laser triggering, which induces gasdermin E-mediated pyroptosis. Furthermore, 17-AAG reduces myeloid-derived suppressor cells and sensitizes tumors to anti-programmed death-1 (PD-1) therapy. Thus, our prodrug strategy achieves tumor-targeted pyroptosis to suppress tumor growth, thereby improving the response to anti-PD-1 therapy and extending the survival of 4T1 breast tumor-bearing mice. Consequently, this pyroptosis-based prodrug represents a novel strategy for enforcing immunogenic photodynamic therapy.

Abstract:The immune checkpoint blockade (ICB) antibody immunotherapy has demonstrated clinical benefits for multiple cancers. However, the efficacy of immunotherapy in tumors is suppressed by deficient tumor immunogenicity and immunosuppressive tumor microenvironments. Pyroptosis, a form of programmed cell death, can release tumor antigens, activate effective tumor immunogenicity, and improve the efficiency of ICB, but efficient pyroptosis for tumor treatment is currently limited. Herein, we show a mild hyperthermia-enhanced pyroptosis-mediated immunotherapy based on hollow carbon nanozyme, which can specifically amplify oxidative stress-triggered pyroptosis and synchronously magnify pyroptosis-mediated anticancer responses in the tumor microenvironment. The hollow carbon sphere modified with iron and copper atoms (HCS-FeCu) with multiple enzyme-mimicking activities has been engineered to induce cell pyroptosis via the radical oxygen species (ROS)-Tom20-Bax-Caspase 3-gasdermin E (GSDME) signaling pathway under light activation. Both in vitro and in vivo antineoplastic results confirm the superiority of HCS-FeCu nanozyme-induced pyroptosis. Moreover, the mild photothermal-activated pyroptosis combining anti-PD-1 can enhance antitumor immunotherapy. Theoretical calculations further indicate that the mild photothermal stimulation generates high-energy electrons and enhances the interaction between the HCS-FeCu surface and adsorbed oxygen, facilitating molecular oxygen activation, which improves the ROS production efficiency. This work presents an approach that effectively transforms immunologically "cold" tumors into "hot" ones, with significant implications for clinical immunotherapy.

Abstract:An ICD and pyroptosis‐induced nanoenzyme is first proposed for efficient tumor immunotherapy. The nanoenzyme IMZF demonstrates the SOD, CAT, POD, and GSHOx‐like catalytic ability; then thus, facilitating ROS generation, depleting GSH, and arousing ICD/pyroptosis pathways for efficient antigen presentation. This work provides a new design strategy for simultaneously enhancing antigen presentation and for reversal of immune suppression TME. Immunotherapy represents a widely employed modality in clinical oncology, leveraging the activation of the human immune system to target and eradicate cancer cells and tumor tissues via endogenous immune mechanisms. However, its efficacy remains constrained by inadequate immune responses within "cold" tumor microenvironment (TME). In this study, a multifunctional nanoscale pyroptosis inducer with cascade enzymatic activity (IMZF), comprising superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and glutathione oxidase (GSHOx), is dissociated within the acidic and glutathione‐rich TME. The vigorous enzymatic activity not only generates oxygen (O2) to alleviate hypoxia and promote M2 to M1 macrophage polarization but also yields reactive oxygen species (ROS) and depletes glutathione (GSH) within the TME. Functioning as an immunogenic cell death (ICD) activator and pyroptosis inducer, IMZF synergistically triggers dendritic cell maturation and inflammatory lymphocyte infiltration via ICD‐associated pyroptosis, thereby reversing immune suppression within the TMEs. Consequently, it exerts inhibitory effects on both primary and distal tumors. This cascade enzymatic platform‐based pyroptosis inducer offers an intelligent strategy for effectively overcoming immune suppression within "cold" tumors, thereby providing a promising avenue for advanced immunotherapeutic interventions.

Abstract:Clinical photodynamic therapy (PDT) only has a limited cancer therapeutic effect and typically leads to a more hypoxic milieu owing to the hypoxic conditions of the solid tumor microenvironment that limit the singlet oxygen (1O2), generation. To address this issue, the PDT, in combination with hypoxia-activated prodrugs, has recently been investigated as a possible clinical treatment modality for cancer therapy. By cross-linking the photosensitizer tetra(4-hydroxyphenyl)porphine (THPP) and a 1O2-cleavable thioketal (TK) linker, a multifunctional nanoscale covalent organic framework (COF) platform with a high porphyrin loading capacity was synthesized, which significantly improve the reactive oxygen species (ROS) generation efficiency and contributes to PDT. As-synthesized THPPTK-PEG nanoparticles (NPs) possess a high THPP photosensitizer content and mesoporous structure for further loading of the hypoxia-responsive prodrug banoxantrone (AQ4N) into the COF with a high-loading content. The nano-carriers surfaces are coated with a thick PEG coating to promote their dispersibility in physiological surroundings and therapeutic performance. When exposed to 660 nm radiation, such a nanoplatform can efficiently create cytotoxic 1O2 for PDT. Similarly, oxygen intake may exacerbate the hypoxic environment of the tumor, inducing the activation of AQ4N to achieve hypoxia-activated cascade chemotherapy and increased treatment efficacy. This study provides a new nanoplatform for photodynamic-chemical synergistic therapy and offers critical new insights for designing and developing a multifunctional supramolecular drug delivery system.Statement of significanceHere, we designed a laser-activated hypoxia-responsive nanoscale COF nanoplatform for hypoxia-activated cascade chemotherapy and PDT. When exposed to laser light, thus this nanoplatform can efficiently create cytotoxic 1O2 for PDT while consuming oxygen at the tumor location. However, increased oxygen consumption can exacerbate the tumor's hypoxic environment, causing AQ4N to become active, allowing for programmed hypoxia-triggered cascade chemotherapy and improved therapeutic efficacy. In addition, this innovative nanoscale COF nanoplatform allows for laser-controlled drug delivery in specific areas, which dramatically improves tumor inhibition. This research suggests a method for attaining ultrasensitive drug release and effective cascade therapy for cancer treatments.

Abstract:Pyroptosis is a gasdermin-mediated programmed necrosis that occurs via membrane perforation and that can be exploited for biomedical applications in cancer therapy. However, inducing specific pyroptotic cancer cell death while sparing normal cells is challenging. Here, we report an acid-activatable nanophotosensitizer library that can be used to spatiotemporally target distinct stages of endosomal maturation, enabling tunable cellular pyroptosis. Specific activation of phospholipase C signalling transduction in early endosomes triggers gasdermin-E-mediated pyroptosis, which is dramatically reduced when acid-activatable nanophotosensitizers are transported into late endosomes/lysosomes. This nanotuner platform induces pyroptotic cell death with up to 40-fold tunability in various gasdermin-E-positive human cancers, resulting in enhanced anti-tumour efficacy and minimized systemic side effects. This study offers new insights into how to engineer nanomedicines with tunable pyroptosis activity through specific targeting of distinct endocytic signalling for biomedical applications.

Abstract:A nano photoimmunotherapy system is fabricated that can promote rapid pyroptosis and modify the inflammatory microenvironment. The treatments between immunodeficient mice and normal mice prove that pyroptosis of cancer cells can better facilitate immune cell infiltration and then obtain tumor suppression, which provide the potential to address the problem of checkpoint‐inhibitor immunotherapy inefficacy in immune‐suppressed cold tumor care field. Immunotherapy, especially immune checkpoint blockade (ICB) antibody immunotherapy, has revolutionized the treatment ways of cancers and provided remarkable clinical benefits for multiple cancers. However, the efficacy of immunotherapy in tumors with an immune‐excluded or immune‐suppressed phenotype is dismal due to the lack or paucity of immune infiltration in the tumor microenvironment. Herein, an emerging photoimmunotherapy based on remodeling the inflammatory microenvironment is reported, ascribed to nanoagonist‐mediated gasdermin E (GSDME)‐dependent pyroptosis and providing positive feedback to activate anti‐PD‐1 immunotherapy. An iridium‐based photosensitizer (IrP) carrying methyltransferase inhibitor RG108 (R@IrP) lead to rapid cell pyroptosis via the caspase‐3/GSDME pathway under the light activation. Furthermore, light‐elicited pyroptosis synergized with anti‐PD‐1 to induce anti‐tumor photoimmunotherapy. The pro‐inflammatory factors released by pyroptotic cells remodel the inflammatory microenvironment and recruit immune cells to kill tumor cells, resulting in CD8+ cytotoxic T lymphocytes activation, PD‐1 expression enhancement, and dendritic cell slightly maturation. Collectively, these findings present a synergistic strategy of photoimmunotherapy, that is, turning immunological cold tumors into hot tumors that can respond to anti‐PD‐1‐based immunotherapy via precise pathway regulation.

Abstract:Immunotherapy shows remarkable efficacy in treating several types of cancer such as melanoma, leukemia, and lung carcinoma, but its therapeutic effect for most solid tumors is still limited. Various cancer therapies, such as chemotherapy, radiotherapy and phototherapy, kill solid tumors through non-inflammatory apoptosis or ablation, rather than making solid tumors immunogenic. As a highly-inflammatory programmed cell death (PCD), pyroptosis provides a great opportunity to alleviate immunosuppression and promote a systemic immune response in treating solid tumors. Herein, by fusing breast cancer membrane onto the poly(lactic-co-glycolic acid) polymeric core, we design a biomimetic nanoparticle (BNP) loaded with indocyanine green (ICG) and decitabine (DCT) for photo-activated cancer cell pyroptosis and solid tumor immunotherapy. The tumor-homing BNP effectively accumulate in tumor with low immunogenicity. ICG in BNP puncture cancer cell membranes induces a sharp cytoplasm Ca2+ concentration increase by low-dose NIR photo-activation, which promotes cytochrome c release followed by caspase-3 activation. DCT up-regulates GSDME expression synergistically via inhibiting DNA methylation, which enhances caspase-3 cleavage to GSDME and causes cancer cell pyroptosis. Finally, photo-activated pyroptosis mediated by BNP induces an impressive systemic antitumor immunity for inhibition of both primary tumor and distant tumors. Overall, pyroptosis-associated BNP shows a novel strategy for solid tumor immunotherapy with high compatibility and wide clinical applicability.

Abstract:The hydrogen peroxide responsive specific pyroptosis inducer triggers specific pyroptotic tumor cell death and maintains pyroptosis silence through differentially controlling reactive oxygen species levels in tumor and normal cells, precisely inhibiting the growth of primary and metastatic tumors with negligible side effect. Pyroptosis inducer, a powerful anti‐tumor agent that causes obvious programmed cell death and immune stimulation, has been challenged to trigger specific pyroptotic tumor cell death while keeping pyroptosis silence in normal cells. Here, an intelligent inducer is reported that acts as a reactive oxygen species (ROS) scavenger in normal cells to keep pyroptosis silence, while serving as ROS generator to induce obvious pyroptotic tumor cells death dependent on high hydrogen peroxide levels and near‐infrared laser irradiation. This switchable activity ensures this inducer to precisely kill the tumor cells with augmented immunogenicity while causing minimal damage to normal cells. Moreover, the catalase‐like activity endows this inducer to overcome limitation of tumor hypoxia on ROS generation and show significant pyroptosis activation, further initiating the immune response to inhibit the tumor metastases in vivo. This study provides valuable insights into design new pyroptosis inducer with controllable pyroptosis activity to specifically induce programmed tumor cell pyroptosis for precise and augmented tumor therapy with minimal side effects.

Abstract:Immunotherapy is considered to be an effective treatment for cancer and has drawn extensive interest. Nevertheless, the insufficient antigenicity and immunosuppressive tumor microenvironment often cause unsatisfactory therapeutic efficacy. Herein, we develop a photo‐activated reactive oxygen species (ROS) amplifying system (defined as "M‐Cu‐T") to induce anti‐tumor immune response by triggering a tumor‐specific immunogenic pyroptosis. In M‐Cu‐T, M1 macrophage membrane‐based vesicles are used for drug loading and tumor targeting, photosensitizers (meso‐tetra(4‐aminophenyl) porphyrin, TAPP) are used as a pyroptosis inducer, copper ions (Cu2+) can enhance ROS‐induced pyroptosis by consuming antioxidant systems in cells. As expected, the prepared M‐Cu‐T targets enrichment into tumor cells and cascades the generation of ROS, which further induces pyroptosis through caspase 3‐mediated gasdermin E (GSDME) cleavage under laser activation. The pyroptotic cancer cells accompanyingly secrete related pattern molecules, induce immunogenic cell death, and activate anti‐tumor immunity for immunotherapy. An effective tumor ablation is observed in LLC and CT26 cancer mouse model. Abstract:Cancer immunotherapy is a favorable strategy for facilitating anti‐tumor immunity, but it shows limited benefits in clinical practice owing to the immunosuppressive tumor microenvironment. Pyroptosis shows great immunostimulatory effect on tumor, whereas the lack of pyroptotic inducer with imaging property has restricted its progress in tumor theranostics. Herein, a mitochondria‐targeted AIEgen (TPA‐2TIN) with NIR‐II emission is designed for highly efficient induction of tumor cell pyroptosis. The fabricated TPA‐2TIN nanoparticles can be efficiently taken up by tumor cells and selectively accumulate in tumor for a long term observed by NIR‐II fluorescence imaging. More importantly, the TPA‐2TIN nanoparticles can effectively stimulate immune responses both in vitro and in vivo mediated by the mitochondrial dysfunctions and the subsequent activation of the pyroptotic pathway. Ultimately, the reversal of the immunosuppressive tumor microenvironment significantly enhances the immune checkpoint therapy. Our study paves a new avenue for adjuvant immunotherapy of cancer. Abstract:A pH‐responsive polydopamine nanocluster R‐P@PDA‐GC for enhanced phototherapy‐induced pyroptosis and anti‐tumor immunity via a two‐input programmed cascade therapy is rationally designed. This novel two‐input programmed cascade strategy of R‐P@PDA‐GC generated substantial ROS within tumor cells, initiating cell pyroptosis, remodeling the TME and facilitating potent anti‐tumor immunotherapy. Pyroptosis, an inflammatory cell death, plays a pivotal role in activating inflammatory response, reversing immunosuppression and enhancing anti‐tumor immunity. However, challenges remain regarding how to induce pyroptosis efficiently and precisely in tumor cells to amplify anti‐tumor immunotherapy. Herein, a pH‐responsive polydopamine (PDA) nanocluster, perfluorocarbon (PFC)@octo‐arginine (R8)‐1‐Hexadecylamine (He)‐porphyrin (Por)@PDA‐gambogic acid (GA)‐cRGD (R‐P@PDA‐GC), is rationally design to augment phototherapy‐induced pyroptosis and boost anti‐tumor immunity through a two‐input programmed cascade therapy. Briefly, oxygen doner PFC is encapsulated within R8 linked photosensitizer Por and He micelles as the core, followed by incorporation of GA and cRGD peptides modified PDA shell, yielding the ultimate R‐P@PDA‐GC nanoplatforms (NPs). The pH‐responsive NPs effectively alleviate hypoxia by delivering oxygen via PFC and mitigate heat resistance in tumor cells through GA. Upon two‐input programmed irradiation, R‐P@PDA‐GC NPs significantly enhance reactive oxygen species production within tumor cells, triggering pyroptosis via the Caspase‐1/GSDMD pathway and releasing numerous inflammatory factors into the TME. This leads to the maturation of dendritic cells, robust infiltration of cytotoxic CD8+ T and NK cells, and diminution of immune suppressor Treg cells, thereby amplifying anti‐tumor immunity.

A Formal High-level Modeling Approach to Develop Reliable Components in Vision-based Robotics

Abstract:This paper proposes the use of the control-state Abstract State Machines for a rigorous foundation in high- level modeling and validating component-based applications in Vision-Based Robotics. In particular, an extension of the classical flowchart notation for control-state ASMs is proposed to support modularization and reuse in a direct way. The resulting ASM models are to be intended as "ground models" that can be used as basis or patterns to practically model and formally validate the behavior of typical robotic control tasks, and to link (via successive refinements) these high-level models of components to their implementation code by making their functional correctness mathematically controllable. The proposed flowchart extension and the availability of reusable and validated ground models allow a better system design and speed up the development of the system. Keywords-components; abstract state machines; ground mod- eling; robotic control tasks. I. INTRODUCTION Vision-based robotics is a challenging research field (1). One of the open and commonly stated problems in the field is the need for exchange of experiences, best practices, and high-level models of robust, reliable and flexible robot control applications with visual servoing functions. Recently, we investigated (2) the use of the Abstract State Machine (ASM) formal method (3) for a systematic study and a rigorous foundation of modeling and validating Visual Servoing (VS) applications. The ASM method is a discipline for reliable system development, which allows to bridge the gap between informal requirements and executable code. The ASM formalism supports concurrency, heterogeneous state and modularity (compositional design and verification techniques). These features are essential to tailor ground models of control tasks definitions and associated syn- chronization/communication patterns of VS applications in rigorous, compositional and abstract terms. Ground models are blueprints of the to-be-implemented piece of "real world" that "ground the design in the reality" (4). In particular, we exploit the notion of control state ASMs (a class of ASMs (3)) as a natural extension of Finite State Machines. In this paper, we present an extension - called pattern- oriented control-state ASMs - of the classical flowchart notation for control-state ASMs to support modularization and reuse in a direct way. The proposed notation is useful to denote explicitly modeling elements to be further refined, to allow the definition/instantiation of recurring design so- lutions or patterns, to perform initial validation of separate high-level models and to improve model traceability between the flowchart diagrams and their concrete (textual) ASM specifications during the ground modeling and development process. In this context, the term "pattern" is to be intended to have its classical meaning, i.e., as a schema of a recurring solution, rather than the meaning of "design pattern" as in the book of the GoF (Gang of Four). We here repeat our previous experience in the embedded system-on-a-chip domain (5)(6) to shift the focus from implementation to design through high-level modeling. Our approach combines the expressive power and accuracy of control state ASMs with the intuition provided by visual flowchart descriptions to capture the behavioral view of task-level control of VS applications. As starting point, we manually extracted from the structure of existing (basically C/C++) code architectural descriptions (in terms of UML component diagrams) of high-level models of component control tasks and also recurring synchronization/communica- tion patterns between tasks that could be used for the ground modeling and analysis in ASM. We then defined these ASM abstract models using the pattern-oriented control-state ASM notation, transformed such models into executable ASM models using the notation ASMETA/AsmetaL (7)(8), and then validated them through basic formal analysis techniques (simulation and scenario-based simulation) (9). The resulting and validated ASM models are used as basis or patterns for high-level modeling and validating typical control tasks of VS applications in a formal way, thus leading from the abstract models to executable (C/C++) code by making their functional correctness mathematically controllable.
Pyroptosis Activation by Photodynamic-Boosted Nanocatalytic Medicine Favors Malignancy Recession

Abstract:Although pyroptosis as a newly-emerging but burgeoning programmed cell death fashion has been extensively accepted to outperform apoptosis in inducing rapid and complete cell deaths, potentiating immunity and mitigating resistance, its contribution is low. Therefore, the demand to elevate pyroptosis level is highly desirable and highlighted. Herein, a RGD-modified and pyroptosis-engineered theranostic agent (PETA) consisting of Fe3O4-embedded magnetic mesoporous silica nanoparticles (MMSN) vehicles and chlorin e6 (Ce6) photosensitizers (i.e., MMSN-cRGD@Ce6) is established to heighten ROS level for inducing pyroptosis by Ce6-mediated photodynamic process. Inspiringly, Fe3O4-mediated nanocatalytic medicine contributes to O-2 release and hypoxia mitigation, which liberates hypoxia-induced resistances to ROS production and further favors ROS production. Consequently, ROS-activated pyroptosis is accessible to circumvent the hypoxia-induced resistance to ROS-activated apoptosis especially when uniting with cRGD-enhanced active targeting, which significantly represses malignant breast cancer in vitro and in vivo. The design principles of targeted PETAs can serve as a general method to develop new pyroptosis-highlighted theranostic agents, and coincidently indicate that pyroptosis contribution should be surveyed to veritably and comprehensively reflect death pathway when investigating ROS-based anti-tumor pathways.

Abstract:Pyroptosis, as an inflammatory cell death, has been widely applied in tumor therapy, but its systemic adverse reactions caused by nonspecific activation still seriously hinder its application. Herein, a near‐infrared (NIR) light triggered thermoresponsive pyroptosis strategy is designed for on‐demand initiation of pyroptosis and synergistic tumor immunotherapy. Specifically, glucose oxidase (GOx) loaded and heat‐sensitive material p(OEOMA‐co‐MEMA) (PCM) modified mesoporous Pt nanoparticles (abbreviated as PCMPt/GOx) were prepared as the mild‐temperature triggered pyroptosis inducer. Pt nanoparticles can not only serve as nanozyme with catalase‐like activity to promote GOx catalytic reaction, but also act as photothermal agent to achieve mild‐temperature photothermal therapy (PTT) and thermoresponsive GOx release on‐demand under the irradiation of NIR light, thereby activating and promoting pyroptosis. In vitro and in vivo experiments prove that NIR light triggered thermoresponsive pyroptosis system exhibits excellent antitumor immunity activity as well as significantly inhibits tumor growth. The precise control of pyroptosis by NIR light as well as pyroptosis cooperated with mild‐temperature PTT for synergistically attenuated tumor immunotherapy are reported for the first time. This work provides a new method to initiate pyroptosis on demand, which is of great significance for spatiotemporally controllable pyroptosis and immunotherapy. Abstract:Promoting innate immunity through pyroptosis induction or cGAS‐STING pathway activation has emerged as a potent approach to counteract the immunosuppressive tumor microenvironment and elicit systemic antitumor immunity. However, current pyroptosis inducers and STING agonists often suffer from limitations including instability, unpredictable side effects, or inadequate intracellular expression of gasdermin and STING. Here, a tumor‐specific nanotheranostic platform that combines photodynamic therapy (PDT) with epigenetic therapy to simultaneously activate pyroptosis and the cGAS‐STING pathway in a light‐controlled manner is constructed. This approach involves the development of oxidation‐sensitive nanoparticles (NP1) loaded with the photosensitizer TBE, along with decitabine liposomes (NP2). NP2 enables the restoration of STING and GSDME expression, while NP1‐mediated PDT facilitates the release of DNA fragments from damaged mitochondria to potentiate the cGAS‐STING pathway, and promotes the activation of caspase‐3 to cleave the upregulated GSDME into pore‐forming GSDME‐N terminal. Subsequently, the released inflammatory cytokines facilitate the maturation of antigen‐presentation cells, triggering T cell‐mediated antitumor immunity. Overall, this study presents an elaborate strategy for simultaneous photoactivation of pyroptosis and the cGAS‐STING pathway, enabling targeted photoimmunotherapy in immunotolerant tumors. This innovative approach holds significant promise in overcoming the limitations associated with existing therapeutic modalities and represents a valuable avenue for future clinical applications. Abstract:The conventional molecular immunogenic cell death (ICD) inducers suffer from poor biocompatibility and unsatisfactory efficacy. Here, a biocompatible nanosized covalent organic framework (nCOF)–based pyroelectric catalyst (denoted as TPAD-COF NPs) is designed for pyroelectric catalysis-activated in situ immunotherapy. TPAD-COF NPs confine organic pyroelectric molecules to rigid TPAD-COF NPs to substantially reduce aggregation and enhance biocompatibility, thus improving pyroelectrocatalytic efficiency. After tumor internalization, TPAD-COF NPs facilitate photothermal tumor ablation under near-infrared (NIR) laser exposure, resulting in effective ICD induction. In addition, TPAD-COF NPs effectively catalyze the conversion of temperature changes to pyroelectric changes, which subsequently react with adjacent O 2 to generate reactive oxygen species, thus triggering robust ICD activation. In vivo evaluation using mouse models confirmed that TPAD-COF NPs evidently inhibited the proliferation of primary and distant tumors and prevented lung metastasis under NIR laser illumination. Therefore, this study opens an avenue for designing nCOF-based catalysts for pyroelectric catalysis-activated in situ immunotherapy.

An NIR-II Photothermally Triggered "Oxygen Bomb" for Hypoxic Tumor Programmed Cascade Therapy

Abstract:Hypoxia, as a characteristic feature of solid tumors, has a close relationship with tumor resistance to photodynamic therapy (PDT) and chemotherapy. Perfluorocarbon (PFC) is reported to relieve hypoxic in solid tumors by acting as an oxygen carrier via several nanostructures. However, the oxygen delivery process is mostly driven by a concentration gradient, which is uncontrollable. Herein, a photothermally controlled "oxygen bomb" PSPP-Au980 -D is designed by encapsulating a PFC core within a functionalized bilayer polymer shell. Near-infrared second window photothermal agent gold nanorods with excellent photo-to-heat energy-conversion ability are fabricated on the surface of the polymer shell via an innovative modified two-step seedless ex situ growth process to thermally trigger O2 release. Then, a programmed cascade therapy strategy is customized for hypoxic orthotopic pancreatic cancer. First, PSPP-Au980 -D is irradiated by a 980 nm laser to photothermally trigger O2 infusing into the hypoxic tumor microenvironment, which is accompanied by local hyperemia and doxorubicin release. Subsequently, a 680 nm laser is used to generate singlet oxygen in the oxygenated tumor microenvironment for PDT. This choreographed programmed cascade therapy strategy will provide a new route for suppressing hypoxic tumor growth under mild conditions based on controllable and effective oxygen release.

Abstract:Abstract Immunogenic programmed cell death, such as pyroptosis and ferroptosis, efficiently induces an acute inflammatory response and boosts antitumor immunity. However, the exploration of dual-inducers, particularly nonmetallic inducers, capable of triggering both pyroptosis and ferroptosis remains limited. Here we show the construction of a covalent organic framework (COF-919) from planar and twisted AIEgen-based motifs as a dual-inducer of pyroptosis and ferroptosis for efficient antitumor immunity. Mechanistic studies reveal that COF-919 displays stronger near-infrared light absorption, lower band energy, and longer lifetime to favor the generation of reactive oxygen species (ROS) and photothermal conversion, triggering pyroptosis. Because of its good ROS production capability, it upregulates intracellular lipid peroxidation, leading to glutathione depletion, low expression of glutathione peroxidase 4, and induction of ferroptosis. Additionally, the induction of pyroptosis and ferroptosis by COF-919 effectively inhibits tumor metastasis and recurrence, resulting in over 90% tumor growth inhibition and cure rates exceeding 80%.
