Abstract:As a traditional medicinal herb and valuable natural spice in China, Aquilaria sinensis (Lour.) Gilg has many significant pharmacological effects. Agarwood is the resinous heartwood acquired from wounded A. sinensis trees, and is widely used in pharmaceuticals owing to its excellent medicinal value. In this study, the chemical composition of volatile components and alcohol extracts from different organs of A. sinensis and agarwoods grown in different regions were investigated using GC-MS. The results showed that Vietnam agarwood had the highest moisture content, which was attributed to the local climate, while the fruit and bark of A. sinensis had higher moisture contents than the other organs. The volatile components of A. sinensis organs included 3-ethyl-5-(2-ethylbutyl)-octadecane, oleic acid 3-(octadecyloxy) propyl ester, and docosanoic acid 1,2,3-propanetriyl ester, while the alcohol extracts of A. sinensis organs contained benzoic acid ethyl ester, hexadecanoic acid ethyl ester, oleic acid, and n-hexadecanoic acid. Furthermore, the main active ingredients in agarwood from different habitats were sesquiterpenoids, aromatic species, and chromone compounds. The role of chromone compound 2-phenylethyl-benzopyran as an elicitor and the mechanism of agarwood formation were also investigated. Antioxidant tests showed that essential oils from agarwood and A. sinensis had antioxidant capacities by comparison with butylated hydroxytoluene and vitamin E. An antibacterial activity test showed that the inhibition effect of the essential oil was better against Gram-positive bacteria than against Gram-negative bacteria.

The Characteristic Fragrant Sesquiterpenes and 2-(2-Phenylethyl)chromones in Wild and Cultivated “Qi-Nan” Agarwood

Abstract:Recently, cultivated “Qi-Nan” (CQN) agarwood has emerged as a new high-quality agarwood in the agarwood market owing to its similar characteristics, such as high content of resin and richness in two 2-(2-phenylethyl)chromone derivatives, 2-(2-phenylethyl)chromone (59) and 2-[2-(4-methoxyphenyl)ethyl]chromone (60), to the wild harvested “Qi-Nan” (WQN) agarwood. In this study, we compared the chemical constituents and fragrant components of two types of WQN agarwood from A. agallocha Roxb. and A. sinensis, respectively, with CQN agarwood and ordinary agarwood varieties. Additionally, we analyzed different samples of WQN agarwood and CQN agarwood by GC-MS, which revealed several noteworthy differences between WQN and CQN agarwood. The chemical diversity of WQN was greater than that of CQN agarwood. The content of (59) and (60) was higher in CQN agarwood than in WQN agarwood. For the sesquiterpenes, the richness and diversity of sesquiterpenes in WQN agarwood, particularly guaiane and agarofuran sesquiterpenes, were higher than those in CQN. Moreover, guaiane-furans sesquiterpenes were only detected by GC-MS in WQN agarwood of A. sinensis and could be a chemical marker for the WQN agarwood of A. sinensis. In addition, we summarized the odor descriptions of the constituents and established the correlation of scents and chemical constituents in the agarwood.

Chemical analysis of different parts from agarwood columns by artificially agarwood-inducing method based on GC-MS and UPLC-TOF-MS

Abstract:Agarwood is resin-containing wood produced by plants that have been injured. It is widely used in herbal medicine, incense, decorative items, and so on. In this study, we conducted resin area statistical analysis, determined starch particle and reducing sugar contents, and performed multivariate statistical analysis of chemical composition by GC-MS and UPLC-Q-TOF-MS to explore the different components in sections cut from an agarwood column, designated as A1-A4. The results showed that after stimulation by Agar-Bit inducer, the internal phloem parenchyma cells of the column started to form agarwood, and then starch granules were converted into soluble reducing sugars and agarwood resin. Section A1 showed rapid loss of starch granules, resulting in higher contents of reducing sugars and resin. The resin areas of agarwood in the respective sections were different, gradually decreasing on going from A1 to A4. Total numbers of metabolites of 87 and 63 were identified by GC-MS and UPLC-Q-TOF-MS, respectively. Of these, 10 and 16 metabolites with significant differences (variable importance projection >1) were selected through multivariate statistical analysis. These metabolites included chromones, sesquiterpenes, alkanes, and fatty acids. Among them, 6-methoxy-2-(2-phenylethyl)chromone and 6,7-dimethoxy-2-(2-phenylethyl)chromone were significant markers detected by both GC-MS and UPLC-Q-TOF-MS, which may be essential substances responsible for differences in the agarwood-forming capacities of the cut sections. In conclusion, there has been limited research on the different agarwood-forming capacities of agarwood columns. Here, we explored the differences in various sections of agarwood through chemical analysis to provide a more comprehensive and in-depth understanding of its constitution.

Abstract:Agarwood holds significant importance as a valuable resource for aromatic purposes, however, key components responsible for aroma and the differences between regions and grades remain to be deeply elucidated. Thus, the odors of agarwood sourced from typical zones, as well as the renowned Kynam agarwood, were analyzed by HS-SPME Arrow GC-MS in SCAN and MRM modes. The integrated strategy proposed herein exploits the respective advantages of non-targeted and targeted analysis. In addition to a total of 55 volatile components identified from the NIST database, 114 odor components were matched according to the Smart Aroma Database, and a series of differential compounds was also unearthed and quantified.

Abstract:Agarwood, derived from Aquilaria sinensis and Aquilaria malaccensis , is of medicinal and ecological value and religious importance as incense. The existing imbalance between short supply and increasing demand of this product remains to be solved. Thus, the biologically artificial agarwood-inducing methods commonly called whole-tree agarwood-inducing techniques (agar-wit) have been established to dramatically improve agarwood yield within a short period. However, several studies reported a lower content of ethanol-soluble extractive in the agar-wit agarwood than in the natural agarwood. To further understand the role of microorganisms in agarwood formation, we investigated and contrasted the endophytic bacteria and fungi between different types of agarwood from A. sinensis through high-throughput sequencing. Results showed that the same dominant phyla of bacteria consisting of Proteobacteria, Actinobacteria, and Acidobacteria were shared by the natural agarwood and agar-wit agarwood. Meanwhile, Ascomycota and Basidiomycota constituted the similar dominant fungal phyla of these two kinds of agarwood. However, the principal microbial communities at the genus or order level evidently varied from natural agarwood to agar-wit agarwood. Moreover, the bacterial communities are closely connected with terpenoid and carbohydrate metabolism, which indicated that the bacterial communities also play a vital role in agarwood formation. In conclusion, the higher concentrated abundance of the dominant microbial communities in agar-wit agarwood than in natural agarwood may promote agarwood formation, however, the low evenness of microbial communities also lowers the content of ethanol-soluble extractive.

Abstract:2-(2-Phenylethyl)chromones (PECs), the pivotal constituents responsible for the distinctive aroma and pharmacological potential of agarwood, are primarily obtained through extraction from natural materials. The restricted availability of agarwood has, however, hindered a comprehensive and systematic evaluation of their biological properties. In this study, we have chemically synthesized a total of 38 PEC derivatives, including 23 new compounds that had not been previously isolated from agarwood. These compounds were then evaluated for their inhibitory effects on nitric oxide release, neutrophil superoxide production, as well as their activities against tyrosinase, acetylcholinesterase, and α -glucosidase. The preliminary screening resulted in the identification of promising leads with potential therapeutic activities, and the structure-activity relationship of the synthesized PECs have also been briefly discussed. The results provide a foundation for the synthesis and pharmacological exploration of PECs, which sheds light on the pharmacological potential of agarwood's secondary metabolites.

Abstract:Agarwood is generally used to make incense sticks in China and Southeast Asia. It emits smoke with a pleasant odor when burned. There are few reports on the chemical components of smoke generated by burning or heating agarwood. The agarwoods were produced by the whole-tree agarwood-inducing technique (AWIT), agarwood induced by axe wounds (AAW), burning-chisel-drilling agarwood (BCDA), wood of Aquilaria sinensis trees (AS), respectively. Herein, we used GC-MS to analyze the chemical constituents of incense smoke generated from AWIT, AAW, BCDA, AS, and the extracts of sticks from agarwood produced by the whole-tree agarwood-inducing technique (EAWIT), and 484 compounds were identified. A total of 61 chemical constituents were shared among AWIT, AAW, and BCDA. The experimental data showed that aromatic compounds were the main chemical constituents in agarwood smoke and that some chromone derivatives could be cracked into low-molecular-weight aromatic compounds (LACs) at high temperature. Furthermore, agarwood incense smoke showed anti-inflammatory activities by inhibiting lipopolysaccharide- (LPS-) induced TNF-α and IL-1α release in RAW264.7 cells.
Investigation on Characteristic Constituents of Agarwood by SPME/GC-MS

Abstract:The volatile compositions of the Agarwoods from different production places were extracted by solid phase microextraction(SPME),and isolated and identified by GC-MS,then the quality of Agarwood was evaluated.The investigation showed that the Agarwoods from different places were all consisted of sesquiterpenoids and aromatic compounds,but their chemical compositions and contents were quite different.The six characteristic components,including 4-phenyl-2-butanone,p-methoxy-benzylacetone,agarospirol,agarofuran,hinesol and baimuxinal,and their contents were used as the evaluation indexes for the quality of the Agarwoods.The results indicated that the contents of characteristic constituents in Laos Agarwood were the highest(about 20％),the contents in the Cambodian Agarwood were the second(17％),while the contents in the Chinese Agarwoods(Hainan and Dongguan) were the lowest.Therefore,the Laos Agarwood had the best quality,the Cambodian Agarwood was slightly poorer,and they both were superior to the Chinese Agarwood.The method showed the advantages of low sample consumption and strong characteristic.

Abstract:Introduction: Agarwood is a traditional aromatic southern medicine. It has a long history of being used in traditional Chinese aromatherapy to treat insomnia, anxiety and depression. Due to the scarcity of wild resources, people have planted trees successfully and begun to explore various agarwood-inducing techniques. This study comparative analysis of volatile metabolites in agarwood produced by various inducing techniques and its potential sleep-promoting, anti-anxiety and anti-depressant network pharmacological activities. Methods: A total of 23 batches of two types of agarwood were collected, one of which was produced by artificial techniques, including 6 batches of TongTi (TT) agarwood produced by "Agar-Wit" and 6 batches of HuoLao (HL) agarwood produced by "burning, chisel and drilling", while the other was collected from the wild, including 6 batches of BanTou (BT) agarwood with trunks broken due to natural or man-made factors and 5 batches of ChongLou (CL) agarwood with trunks damaged by moth worms. The study employed metabolomics combined with network analysis to compare the differences in volatile metabolites of agarwood produced by four commonly used inducing techniques, and explored their potential roles and possible action targets in promoting sleep, reducing anxiety, and alleviating depression. Results: A total of 147 volatile metabolites were detected in agarwood samples, mainly including small aromatic hydrocarbons, sesquiterpenes and 2-(2-phenylethyl) chromone and their pyrolysis products. The results showed composition of metabolites was minimally influenced by the agarwood induction method. However, their concentrations exhibited significant variations, with 17 metabolites showing major differences. The two most distinct metabolites were 6-methoxy-2-(2-phenylethyl) chromone and 6,7-dimethoxy-2-(2-phenylethyl) chromone. Among the volatile metabolites, 142 showed promising potential in treating insomnia, anxiety, and depression, implicating various biological and signaling pathways, predominantly ALB and TNF targets. The top three active metabolites identified were 2-(2-phenylethyl) chromone, 1,5-diphenylpent-1-en-3-one, and 6-methoxy-2-[2-(4'-methoxyphenyl) ethyl] chromone, with their relative content in the four types of agarwood being TT>HL>CL>BT. Conclusion: The differences in the content of 2-(2-phenylethyl) chromones suggest that they may be responsible for the varying therapeutic activities observed in different types of agarwood aromatherapy. This study offers theoretical support for the selection of agarwood in aromatherapy practices.

Comprehensive Comparisons Between Grafted Kynam Agarwood and Normal Agarwood on Traits, Composition, and in Vitro Activation of AMPK

Abstract:Agarwood, a highly valuable resin/wood combination with diverse pharmacological activities but scarce supply, has a long history of being used as a medicine in several medical systems. Grafted Kynam agarwood (GKA) has been cultivated successfully recently and has the qualities meeting the definition of premium Kynam agarwood. However, there are few comprehensive comparisons between GKA and normal agarwood in terms of traits, global composition, and activity, and some key issues for GKA to be adopted into the traditional Chinese medical (TCM) system have not been elaborated. The two types of agarwood samples were evaluated in terms of trait characteristics, physicochemical indicators, key component groups, and global compositional profile. Furthermore, a molecular docking was performed to investigate the active ingredients. In vitro activity assays were performed to evaluate the activation of adenosine 5’-monophosphate (AMP)-activated protein kinase (AMPK) by GKA and normal agarwood. The results revealed that, overall, the traits, microscopic characteristics, chemical composition types, and bioactivity between GKA and normal agarwood were similar. The main differences were the content of resin (ethanolic extract content), the content of key component groups, and the composition of the different parent structural groups of 2-(2-phenethyl) chromones (PECs). The contents of total PEC and ethanol extract content of GKA were significantly higher than those of normal agarwood. The MS-based high-throughput analysis revealed that GKA has higher concentrations of sesquiterpenes and flindersia-type 2-(2-phenylethyl) chromones (FTPECs) (m/z 250-312) than normal agarwood. Molecular docking revealed that parent structural groups of FTPECs activated multiple signaling pathways, including the AMPK pathway, suggesting that FTPECs are major active components in GKA. The aim of this paper is to describe the intrinsic reasons for GKA as a high-quality agarwood and a potential source for novel drug development. We combined high-throughput mass spectrometry and multivariate statistical analysis to infer the different components of the two types of agarwood. Then we combined virtual screening and in vitro activity to construct a component/pharmacodynamic relationship to explore the causes of the activity differences between agarwood with different levels of quality and to identify potentially valuable lead compounds. This strategy can also be used for the comprehensive study of other TCMs with different qualities.
Remarkable Phytochemical Characteristics of Chi-Nan Agarwood Induced from New-Found Chi-Nan Germplasm of Aquilaria sinensis Compared with Ordinary Agarwood

Abstract:Wild Chi-Nan agarwood is regarded as the highest quality agarwood from Aquilaria spp. However, the comprehensive research on chemical composition of wild Chi-Nan agarwood is limited. An integrated strategy using SHS-GC-MS and UPLC-Q/Tof-MS was applied to explore the phytochemical characteristics of a kind of agarwood induced from a newly identified germplasm of Chi-Nan A. sinensis. Progenesis QI and MS-Dial were used to preprocess the UPLC-Q/Tof-MS and GC-MS raw data, respectively. Principle component analysis (PCA) and orthogonal partial least squares to latent structure-discriminant analysis (OPLS-DA) models were built to discriminate Chi-Nan agarwood from ordinary agarwood and to screen potential distinguishing components between them. In this study, we clarified the distinguishing differences between Chi-Nan agarwood and ordinary agarwood. The difference is mainly manifested in the average contents of 2-(2-phenylethyl)chromone and 2-[2-(4′-methoxybenzene)ethyl]chromone, which are 170 and 420 times higher in Chi-Nan agarwood than in ordinary agarwood, respectively, while the contents of 5,6,7,8-diepoxy-2-(2-phenylethyl)chromones(DEPECs), 5,6-epoxy-2-(2-phenylethyl)chromones(EPECs), and 5,6,7,8-tetrahydro-2-(2-phenylethyl)chromones(THPECs) such as agarotetrol are extremely low. The content of the main sesquiterpenes in Chi-Nan agarwood was higher than that in ordinary agarwood, especially in regard to guaiane and eudesmane derivatives. In addition, there were significant differences in the contents of low-molecular-weight aromatic compounds such as 2-methyl-4H-1-benzopyran-4-one, 4-methoxybenzaldehyde, and 2-hydroxybenzaldehyde between Chi-Nan agarwood and ordinary agarwood. All the mentioned main chemical characteristics of this new Chi-Nan agarwood were coincident with those of the rare wild Chi-Nan agarwood from A. malaccensis, A. sinensis, and A. crassna. We reported differences in 2-(2-phenylethyl)chromones, sesquiterpenes, and low-molecular-weight aromatic compounds between Chi-Nan agarwood and ordinary agarwood from A. sinensis for the first time; it is necessary to evaluate the agarwood from the new-found Chi-Nan germplasm.

Abstract:The present study is to investigate the chemical constituents and anti-inflammation of agarwood produced via whole-tree agarwood-inducing technique( Agar-Wit) from Aquilaria sinensis by column chromatographic technique and semi-preparation HPLC.Eleven sesquiterpenes were isolated from the agarwood produced by Agar-Wit,and their structures were identified on the basis of physiochemical characteristics and spectroscopic data analysis as baimuxinol( 1),5α,7α( H)-eudesm-11( 13)-en-4α-ol( 2),( 7 S,9 S,10 S)-( +)-9-hydroxy-selina-4,11-dien-14-al( 3),petafolia A( 4),7( 11)-eremophilen-8-one( 5),neopetasane( 6),petafolia B( 7),11-hydroxy-valenc-1( 10)-en-2-one( 8),( 4αβ,7β,8αβ)-3,4,4α,5,6,7,8,8α-octahydro-7-[1-( hydroxymethyl) ethenyl]-4α-methylnaphthalene-1-carboxaldehyde( 9),12-hydroxy-4( 5),11( 13)-eudesmadien-15-al( 10),and( 4 R,5 R,7 S,9 S,10 S)-(-)-eudesma-11( 13)-en-4,9-diol( 11). Among them,compound 1 was a new natural product,and this is the first time to report its13 CNMR spectroscopic data. Compounds 4,9 and 10 were reported from Aquilaria for the first time,and all the compounds are firstly isolated by Agar-Wit from A. sinensis. The anti-inflammatory activity of RAW264. 7 cells with lipopolysaccharide-induced was evaluated.As a result,1,4 and 9 showed potential anti-inflammatory activities with IC50 values( 2. 5±0. 35),( 3. 2±0. 2),( 4. 3±0. 56) μmol·L-1,respectively. This work provided scientific foundation for quality evaluation of the agarwood produced by Agar-Wit.
Chemical and Bioactivity Diversity of 2-(2-Phenylethyl)chromones in Agarwood: A Review

Abstract:2-(2-Phenylethyl)chromone derivatives are regarded as key components in agarwood. An oxygen-containing heterocycle with a benzoannelated gamma-pyrone moiety form the bioactive core of 2-(2-phenylethyl)chromones. With different substituents and positions, 2-(2-phenylethyl)chromone derivatives exhibit diverse biological properties, such as antioxidant, antimicrobial, neuroprotective, anti-inflammatory, and acetylcholinesterase inhibitory activities. In this review, we summarized the studies (from January 1976 to September 2021) on phytochemistry, bioactivity and quality control of 2-(2-phenylethyl)chromones. These studies aimed to clarify the chemical specificity, diversity and structure-activity relationship of 2-(2-phenylethyl)chromones. In addition, we assumed that diverse factors such as tree species, induction methods and formation time contribute to the chemical diversity of 2-(2-phenylethyl)chromones. Furthermore, this review contends that different types of 2-(2-phenylethyl)chromones should be utilized in the quality control methods of agarwood.
Salty treatment increased bioactive compounds accumulation during agarwood development in Aquilaria sinensis trees

Abstract:To compare the bioactive compounds in agarwood induced by different methods in Aquilaria sinensis(Lour.) Gilg trees, a two dimensional thin layer chromatograph(2D-TLC) combined with effect directive analysis(EDA) was developed. Three antioxidants were found by 2D-TLC-DPPH and further identified as 2-(2-phenylethyl) chromones(PECs) with LC-MS/MS. The 3 antioxidants decreased along agarwood formation and their compositions in drilling induced agarwood differed with those in microbe culture induced agarwood. Further study showed NaCl treatment promoted antioxidants accumulation in agarwood induced by drilling or hot drilling. Hot drilling combined with salty stimulation was most efficient in some chemicals accumulation, which were identified as PECs with antioxidant, tyrosinase or β-glucosidase inhibiting activities by 2D-TLC-EDA-LC-MS/MS. This study provided a 2D-TLC-EDA-LC-MS/MS method for bioactive compounds screen and qualification of agarwood. Based on this method, non-conventional methods were found to accelerate the accumulation of some bioactive PECs in A. sinensis trees.
Overview of Sesquiterpenes and Chromones of Agarwood Originating from Four Main Species of the Genus Aquilaria.

Abstract:The main chemical constituents of agarwood are sesquiterpenes and chromones, which can be divided into different categories depending on their molecular skeletons. Agarwoods are obtained from different plant species: Aquilaria sinensis, A. malaccensis, A. crassna, and A. subintegra. In this review, we systematically summarized the structures of 367 compounds isolated from agarwoods originating from four main species. We structurally classified all the components into 11 different types and summarized the number of compounds in each type. Different and identical components were obtained by enumerating the chemical compositions of the different species. Knowledge regarding the chemical constituents of agarwoods of different species will aid understanding of the chemical compositions of agarwoods and will subsequently identify similar compounds that can serve as standards for quality control to provide a reference for future studies on agarwoods from different species and to increase their usefulness.
iTRAQ-Based Quantitative Proteomic Analysis of Chemically Induced Aquilaria sinensis Provides Insights into Agarwood Formation Mechanism

Abstract:Agarwood is a precious traditional Chinese medicine with a variety of pharmacological effects. Although efforts have been made in elucidating the mechanism of agarwood formation, little progress is obtained till now. Therefore, the molecular mechanism of agarwood formation needs to be further explored using different biological approaches. In this study, the quantitative proteomic analysis using iTRAQ technology combined with transcriptomic and metabolomic analyses on chemically induced Aquilaria sinensis is performed to elucidate the agarwood formation mechanism by formic acid stimulus. Data are available via ProteomeXchange with identifier PXD007586; 1884 proteins are detected, 504 differential proteins that show at least twofold differences in their expression levels are selected based on GO annotations, KEGG, STRING analysis, and quantitative RT-PCR analysis. The results indicate that sesquiterpene synthase, germin-like protein, pathogenesis-related protein, 6-phosphogluconate dehydrogenase, lipoyl synthase, and superoxide dismutase play important roles in the agarwood formation, suggesting that the proteins related to the plant defensive response, the removal of peroxide, the disease-resistance, the biosythesis of glycan, fatty acids, and sesquiterpene are crucial for agarwood formation.
Remarkable Phytochemical Characteristics of Chi-Nan Agarwood Induced from New-Found Chi-Nan Germplasm of <i>Aquilaria sinensis</i> Compared with Ordinary Agarwood

Abstract:Wild Chi-Nan agarwood is regarded as the highest quality agarwood from Aquilaria spp. However, the comprehensive research on chemical composition of wild Chi-Nan agarwood is limited. An integrated strategy using SHS-GC-MS and UPLC-Q/Tof-MS was applied to explore the phytochemical characteristics of a kind of agarwood induced from a newly identified germplasm of Chi-Nan A. sinensis. Progenesis QI and MS-Dial were used to preprocess the UPLC-Q/Tof-MS and GC-MS raw data, respectively. Principle component analysis (PCA) and orthogonal partial least squares to latent structure-discriminant analysis (OPLS-DA) models were built to discriminate Chi-Nan agarwood from ordinary agarwood and to screen potential distinguishing components between them. In this study, we clarified the distinguishing differences between Chi-Nan agarwood and ordinary agarwood. The difference is mainly manifested in the average contents of 2-(2-phenylethyl)chromone and 2-[2-(4 '-methoxybenzene)ethyl]chromone, which are 170 and 420 times higher in Chi-Nan agarwood than in ordinary agarwood, respectively, while the contents of 5,6,7,8-diepoxy-2-(2-phenylethyl)chromones(DEPECs), 5,6-epoxy-2-(2-phenylethyl)chromones(EPECs), and 5,6,7,8-tetrahydro-2-(2-phenylethyl)chromones(THPECs) such as agarotetrol are extremely low. The content of the main sesquiterpenes in Chi-Nan agarwood was higher than that in ordinary agarwood, especially in regard to guaiane and eudesmane derivatives. In addition, there were significant differences in the contents of low-molecular-weight aromatic compounds such as 2-methyl-4H-1-benzopyran-4-one, 4-methoxybenzaldehyde, and 2-hydroxybenzaldehyde between Chi-Nan agarwood and ordinary agarwood. All the mentioned main chemical characteristics of this new Chi-Nan agarwood were coincident with those of the rare wild Chi-Nan agarwood from A. malaccensis, A. sinensis, and A. crassna. We reported differences in 2-(2-phenylethyl)chromones, sesquiterpenes, and low-molecular-weight aromatic compounds between Chi-Nan agarwood and ordinary agarwood from A. sinensis for the first time; it is necessary to evaluate the agarwood from the new-found Chi-Nan germplasm.
Effects of various artificial agarwood-induction techniques on the metabolome of Aquilaria sinensis

Abstract:Background: Agarwood is a highly sought-after resinous wood for uses in medicine, incense, and perfume production. To overcome challenges associated with agarwood production in Aquilaria sinensis, several artificial agarwood-induction treatments have been developed. However, the effects of these techniques on the metabolome of the treated wood samples are unknown. Therefore, the present study was conducted to evaluate the effects of four treatments: fire drill treatment (F), fire drill + brine treatment (FS), cold drill treatment (D) and cold drill + brine treatment (DS)) on ethanol-extracted oil content and metabolome profiles of treated wood samples from A. sinensis. Results: The ethanol-extracted oil content obtained from the four treatments differed significantly (F < D < DS < FS). A total of 712 metabolites composed mostly of alkaloids, amino acids and derivatives, flavonoids, lipids, phenolic acids, organic acids, nucleotides and derivatives, and terpenoids were detected. In pairwise comparisons, 302, 155, 271 and 363 differentially accumulated metabolites (DAM) were detected in F_vs_FS, D_vs_DS, F_vs_D and FS_vs_DS, respectively. The DAMs were enriched in flavonoid/flavone and flavonol biosynthesis, sesquiterpenoid and triterpenoid biosynthesis. Generally, addition of brine to either fire or cold drill treatments reduced the abundance of most of the metabolites. Conclusion: The results from this study offer valuable insights into synthetically-induced agarwood production in A. sinensis.

Abstract:This work is focused on the characterization of the composition of a CO2 supercritical fluid extract of Aquilaria sinensis (Chinese agarwood) collected in the Dongguan area (China) and infected by mechanical methods. The constituents of this extract were analyzed by gas chromatography-mass spectrometry (GC-MS) and quantified accurately by gas chromatography with a flame ionization detector (GC-FID), using an internal reference and predicted response factors. Since a significant number of components of this extract remained non-identified after the initial GC-MS analysis of the whole extract, its fractionation by chromatography on silica gel helped to characterize several additional constituents by isolation and structural analysis by NMR spectroscopy. The main components are the classical agarwood chromones (Flindersia chromone and its mono-, di-, and trimethoxylated analogues (respectively, 11.01% and 0.11-4.02%) along with sesquiterpenic constituents typically found in agarwood essential oils, like baimuxinal (1.90%) and kusunol (1.24%), as well as less common selinane dialdehydes (1.58-2.27%) recently described in the literature. Moreover, the structure and stereochemistry of a new sesquiterpenic alcohol, 14β,15β-dimethyl-7αH-eremophila-9,11-dien-8β-ol (0.67%), was determined unambiguously by the combination of structural analysis (NMR, MS), hemisynthesis, and total synthesis, leading to dihydrokaranone and a neopetasane epimer.
