Abstract:In recent years, the novel minimally invasive combination therapy for tumors has garnered widespread attention due to its controllable therapeutic target, effective combination therapy outcome, and minimal toxic side effects. This study integrated inorganic nanospheres of ferric oxide (Fe 3 O 4 ) with mesoporous silica nanoparticles (MSN), along with polymeric organic polymers—polyaniline (PANI) and hyaluronic acid (HA)—to create a multifunctional tumor treatment platform (Fe 3 O 4 @MSN@PANI@HA), aiming to enhance the efficacy of therapies that have monotherapy limitations. The Fe 2+ within the Fe 3 O 4 @MSN@PANI@HA nanoparticles can react with the unique hydrogen peroxide present in the tumor microenvironment to generate hydroxyl radicals (·OH), which possess cytotoxic properties that can selectively eradicate tumor cells, thereby facilitating chemodynamic therapy (CDT). Moreover, Fe 3 O 4 @MSN@PANI@HA nanoparticles, when loaded with the chemotherapy drug doxorubicin hydrochloride (DOX), yield Fe 3 O 4 @MSN@PANI@HA-DOX nanoparticles. It has been demonstrated that these nanoparticles exhibit commendable drug-loading efficiency and controlled drug-release capabilities, contributing to the chemotherapy of tumors. The inclusion of PANI in the Fe 3 O 4 @MSN@PANI@HA nanoparticles endows them with an exceptional photothermal conversion capability. Upon exposure to an 808-nm near-infrared laser (NIR), the photothermal conversion efficiency of the composite nanomaterials reaches 42.76%, thereby enabling photothermal therapy (PTT) through the thermal ablation of tumor cells. The HA component of the composite nanoparticles provides excellent biocompatibility and a sustained drug-release effect. Simultaneously, it could target tumor cells by recognizing specific receptors that are overexpressed on the cell surfaces, directing the transport of the composite nanoparticles to the tumor site, and selectively destroying tumor cells. This paper confirms the potent therapeutic impact of composite nanoparticles on tumors and offers a novel perspective for designing multifunctional composite nanomaterials.

Abstract:Breast cancer is one of the most prevalent malignancies, necessitating the exploration of more effective synergistic treatment strategies to overcome the limitations of conventional therapies. Chemodynamic therapy (CDT) is an innovative antitumor approach that can be combined with chemotherapy to achieve potent synergistic effects. Mesoporous silica nanomaterials (MSNs) are ideal drug delivery vehicles in cancer therapy due to their unique advantages. This study presents an effective and straightforward strategy to design an intelligent drug delivery system (DDS) activated by the tumor-specific weakly acidic microenvironment to achieve efficient cancer treatment. By incorporating the chemotherapeutic drug doxorubicin (DOX) and divalent iron (Fe 2+ ) into the unique mesoporous channels of MSNs, we fabricated MSNs@Fe 2+ @DOX. Under weakly acidic pH conditions, the functional components Fe 2+ and DOX of MSNs@Fe 2+ @DOX are gradually released at the tumor site. The released Fe 2+ can then consume hydrogen peroxide (H 2 O 2 ) in tumor cells, increasing the levels of reactive oxygen species (ROS) and lipid peroxides through the Fenton reaction, thereby inducing ferroptosis. The combination of DOX-induced apoptosis and ferroptosis results in further enhanced cancer treatment. In vitro and in vivo experiments demonstrated that MSNs@Fe 2+ @DOX had an excellent therapeutic effect on breast cancer cells and tumor-bearing nude mice. We anticipate that this study will provide a promising biotechnological platform for combined breast cancer treatment by inducing CDT and chemotherapy.

Abstract:Breast cancer, a major health concern globally, often faces challenges in the non-invasive elimination of cancer cells. This has prompted researchers to explore alternative strategies, such as photothermal therapy (PTT), which takes advantage of the location of breast cancer lesions. Additionally, the tumor microenvironment with high hydrogen peroxide (H 2 O 2 ) provides a key prerequisite for treating tumors with nanozymes, especially those with peroxidase (POD)-like activity. However, the overexpression of TrxR1, which maintains redox homeostasis in tumor cells, limits reactive oxygen species (ROS)-based cytotoxic therapy. This study aims to present molybdenum selenide nano urchins (MoSe 2 NUs) with excellent photothermal conversion efficiency, POD-like activity and selective inhibition of TrxR1 to achieve photothermal enhanced nanocatalytic therapy for combating breast cancer.

Construction of Methotrexate-Loaded Bi2S3 Coated with Fe/Mn-Bimetallic Doped ZIF-8 Nanocomposites for Cancer Treatment Through the Synergistic Effects of Photothermal/Chemodynamic/Chemotherapy

Abstract:A combination of therapeutic modalities in a single nanostructure is crucial for a successful cancer treatment. Synergistic photothermal therapy (PTT) can enhance the effects of chemodynamic therapy (CDT) and chemotherapy, which could intensify the therapeutic efficacy to induce cancer cell apoptosis. In this study, Fe and Mn on a zeolitic imidazolate framework (ZIF-8) (Fe/Mn-ZIF-8; FMZ) were synthesized through ion deposition. Furthermore, bismuth sulfide nanorods (Bi2S3 NRs; BS NRs) were synthesized via a hydrothermal process and coated onto FMZ to generate the core-shell structure of the Bi2S3@FMZ nanoparticles (B@FMZ). Next, methotrexate (MTX) was loaded effectively onto the porous surface of ZIF-8 to form the B@FMZ/MTX nanoparticles. The Fenton-like reaction catalyzes Fe2+/Mn2+ ions by decomposing H2O2 in the tumor microenvironment, resulting in the formation of toxic hydroxyl radicals (·OH), which promotes the CDT effect of killing cancer cells. Furthermore, under 808 nm laser irradiation, these B@FMZ nanoparticles showed a strong PTT effect, owing to the presence of intense BS NRs as a photothermal agent. The B@FMZ nanoparticles exhibited a prominent drug release efficiency of 87.25% at pH 5.5 under near-infrared laser irradiation due to the PTT effect can promote the drug delivery performance. The B@FMZ nanoparticles were subjected to dual-modal imaging, guided magnetic resonance imaging, and X-ray computed tomography imaging. Both in vitro and in vivo results suggested that the B@FMZ/MTX nanoparticles exhibited enhanced antitumor effects through the combined therapeutic effects of PTT, CDT, and chemotherapy. Therefore, these nanoparticles exhibit good biocompatibility and are promising candidates for cancer treatment.

Abstract:Multimodal therapeutic agents based on novel nanomaterials for multidrug resistance have attracted increasing attention in cancer therapy. In this study, we describe the construction of a programmed mesoporous silica-capped gold nanorod covered with nano-selenium overcoat (Se@Au@mSiO2) nanoparticles as a multifunctional nanoplatform to incorporate materials with specific chemotherapeutic, chemoprevention, and photoablation/hyperthermia functions that collectively contribute to enhance anticancer efficacy in multidrug-resistant breast cancer. The triple-combination-based nanosized Se@Au@mSiO2/DOX effectively accumulates in the tumor and the release of the therapeutic cargo could be remotely manipulated by mild near-infrared (NIR) irradiation. Se@Au@mSiO2/DOX notably enhances the cell killing effect through induction of cell apoptosis. In addition, Se@Au@mSiO2/DOX inhibits tumor cell growth through cell cycle arrest and induction of apoptosis via suppression of the Src/FAK/AKT signaling pathways. Synergistic Se-photothermal-chemotherapy combination exhibits significant tumor growth suppression and delayed tumor progression in vivo. Immunohistochemistry analysis shows elevated numbers of caspase-3 and PARP-immunolabeled cells and decreased Ki-67 + and CD31 + cancer cells in the tumor mass. No noticeable signs of organ damage or toxicity are observed after treatment with Se@Au@mSiO2/DOX (NIR+), which is further supported by hematology and biochemical analyses. Thus, Se@Au@mSiO2/DOX has potential for the clinical treatment of metastatic breast cancers with little or no adverse effects.

Abstract:IntroductionTumor cells can effectively be killed by heat, e.g. by using magnetic hyperthermia. The main challenge in the field, however, is the generation of therapeutic temperatures selectively in the whole tumor region. We aimed to improve magnetic hyperthermia of breast cancer by using innovative nanoparticles which display a high heating potential and are functionalized with a cell internalization and a chemotherapeutic agent to increase cell death.MethodsThe superparamagnetic iron oxide nanoparticles (MF66) were electrostatically functionalized with either Nucant multivalent pseudopeptide (N6L; MF66-N6L), doxorubicin (DOX; MF66-DOX) or both (MF66-N6LDOX). Their cytotoxic potential was assessed in a breast adenocarcinoma cell line MDA-MB-231. Therapeutic efficacy was analyzed on subcutaneous MDA-MB-231 tumor bearing female athymic nude mice.ResultsAll nanoparticle variants showed an excellent heating potential around 500 W/g Fe in the alternating magnetic field (AMF, conditions: H = 15.4 kA/m, f = 435 kHz). We could show a gradual inter- and intracellular release of the ligands, and nanoparticle uptake in cells was increased by the N6L functionalization. MF66-DOX and MF66-N6LDOX in combination with hyperthermia were more cytotoxic to breast cancer cells than the respective free ligands. We observed a substantial tumor growth inhibition (to 40% of the initial tumor volume, complete tumor regression in many cases) after intratumoral injection of the nanoparticles in vivo. The proliferative activity of the remaining tumor tissue was distinctly reduced.ConclusionThe therapeutic effects of breast cancer magnetic hyperthermia could be strongly enhanced by the combination of MF66 functionalized with N6L and DOX and magnetic hyperthermia. Our approach combines two ways of tumor cell killing (magnetic hyperthermia and chemotherapy) and represents a straightforward strategy for translation into the clinical practice when injecting nanoparticles intratumorally.

Abstract:The pursuit of mesoporous Fe-based nanoagents addresses the field of developing alternative Fe-bearing nanoagents for synergistic cancer therapy with the expectation that the use of an essential element may avoid the issues raised by the exogenous administration of other metal element-based nanoagents. Herein, we highlight the interface-engineered mesoporous FeB (mFeB) where the core mFeB is interfacially oxidized into an FeOOH nanosheet loaded with the chemotherapeutic drug doxorubicin (DOX) and further encapsuled within the double-sulfide-bonded SiO2 outer layer, denoted as mFeB@DOX-ss-SiO2, which can realize programmed drug release for synergistic cancer theranostics. When only in a tumor microenvironment, the nanoagent can be activated to release DOX from the mFeB and FeOOH nanosheets as well as expose the easily oxidized mFeB to spontaneously transform to FeOOH nanosheets with Fenton activity to facilitate chemodynamic therapy (CDT). In addition, the high photothermal conversion efficiency of mFeB@DOX-ss-SiO2 would promote CDT. Also, owing to the inherent nature of ferromagnetism and red fluorescence of DOX, mFeB@DOX-ss-SiO2 can realize T2-weighted magnetic resonance imaging and fluorescence imaging. In vivo mouse model experiments demonstrate that mFeB@DOX-ss-SiO2 with good biocompatibility realizing CDT/photothermal therapy/chemotherapy achieved complete tumor suppression. This study opens up a new way to explore theranostic nanoagents.
A theranostic nanocomposite system based on radial mesoporous silica hybridized with Fe3O4 nanoparticles for targeted magnetic field responsive chemotherapy of breast cancer

Abstract:Achieving accumulation of a target drug in tumor tissue, optimal release behavior and concurrent diagnostic imaging with therapy (theranostic) requires an ideal chemotherapy protocol that involves the responsive control of drug delivery. In the current study, we report the construction of a novel theranostic drug delivery system based on radial mesoporous silica, which is hybridized with multiscale magnetic nanoparticles for MRI-guided and alternative magnetic field (AMF) responsive chemotherapy for breast cancer. Superparamagnetic iron oxide nanoparticles (IONPs) with multiscale sizes were prepared via a hydrothermal method (for larger IONPs, lIO NPs) and a thermal decomposition method (for ultra-small IONPs, uIO NPs). lIO NPs act as the heat mediator that is responsive to the AMF, while uIO NPs act as the T-2 contrast agent for MRI. Mesoporous silica nanoparticles with radially oriented mesochannels were further in situ grown onto the surfaces of the lIO NPs, and both the uIO NPs and doxorubicin, an anti-cancer drug, can be readily incorporated within the mesochannels. To endow the as-prepared doxorubicin-loaded nano-hybrids with a good targeting ability, the surfaces of the particles were modified with folic acid (FA). This prepared drug delivery system based on magnetic/silica nanocomposites showed high drug loading efficiency and AMF stimuli responsive release properties. Meanwhile, in vitro MRI measurements revealed an excellent MRI contrast effect with a high relaxation value of 308 mM(-1) s(-1). Furthermore, both in vitro cytotoxicity analysis and in vivo anti-tumor effect evaluation demonstrated that the drug delivery system possesses great potential as an MRI-guided stimuli-responsive theranostic platform for effective active targeting chemotherapy of cancer.

Abstract:Selecting subcellular organelles as therapeutic targets have drawn great interest for cancer treatment. Herein, novel nanotheranostic agents FeF2/Fe1–xS nanorods modified by polyethylene glycol (FFSNRs‐PEG) are first synthesized for tumor microenvironments (TME) responsive magnetic resonance imaging (MRI) guided synergetic cancer therapy focused on the mitochondria. In TME, the Fe2+ and F− ions released from FFSNRs‐PEG can make mitochondrial damage via the production of toxic ·OH through Fenton reaction and activation of caspase‐3, resulting in apoptosis and cell death. Taking advantage of the strong absorption of FFSNRs‐PEG in the second biowindow (1000–1700 nm), the photothermal effect boosts the efficiency of Fenton reaction to achieve synergetic chemodynamic therapy and photothermal therapy under near‐infrared light (1064 nm) irradiation. In addition, benefiting from the self‐oxidation effect, the FFSNRs‐PEG can serve as a novel TME‐responsive contrast agent for T2‐weighted MRI. Overall, this work highlights an innovative “weapon” to realize TME‐responsive tumor diagnosis and synergetic therapies focused on the mitochondria. This work reports novel nanotheranostic agents FeF2/Fe1–xS nanorods modified by polyethylene glycol for tumor microenvironment responsive magnetic resonance imaging‐guided antitumor therapy focused on the mitochondria that is based on the synergetic effect of chemodynamic therapy, photothermal therapy, and fluoride‐induced mitochondrial dysfunction, which presents the new insights for exploring the nanotheranostic agents focused on mitochondria.

Abstract:2D MXene, Ti3C2 (TC), has displayed enormous potential in applications in photothermal therapy (PTT), attributing to its biocompatibility and outstanding photothermal conversion capability. However, some tumor ablations are difficult to be realized completely by monotherapy due to the essential defects of monotherapy and intricate tumor microenvironment (TME). In this work, the appropriate doped Fe2+ ions are anchored into the layers of 2D ultrathin TC nanosheets (TC NSs) to synthesize a novel multifunctional nanoshell of Fe(II)-Ti3C2 (FTC) through interlayer electrostatic adsorption. FTC possesses superior photothermal conversion efficiency (PTCE) than TC NSs, attributing to the enhanced conductivity promoted by interlaminar ferrous ion-channels. Moreover, Fenton reaction based on ferrous ions endows FTC the abilities of reactive oxide species (ROS) releasing and glutathione (GSH) suppression triggered by near-infrared (NIR) laser, featuring splendid biocompatibility and curative effect in hypoxic TME. Meanwhile, magnetic resonance imaging (MRI) responding in FTC reveals the potential as an integrated diagnosis and treatment nanoplatform. FTC could provide new insights into the development of multimoded synergistic nanoplatform for biological applications, especially breaking the shackles of MXenes merely used as a photo-thermal agent (PTA), adopting it to bioimaging sensor and drug loading.
Magnetic and Ph Dual-Responsive Nanoparticles for Synergistic Drug-Resistant Breast Cancer Chemo/Photodynamic Therapy

Abstract:Background Drug resistance is one of the prime reasons of chemotherapy failure in breast cancer and is also an important factor affecting prognosis. Purpose In this study, we constructed a functional magnetic mesoporous silica-based nanocomposite (MMSN) for breast cancer chemotherapy/photodynamic therapy. Methods MMSN was characterized by scanning electron microscopy and transmission electron microscopy to observe the morphology. The size distribution and zeta potential of the MSNs were determined using Malvern Particle Size Analyzer. Anti-tumor activity in vitro was investigated by CCK-8 assay, flow cytometry and transwell experiment, and the anti-tumor activity in vivo was probed into by magnetic targeting, toxicity, and antitumor effects in breast cancer-bearing BABL/c nude mice. Results The results showed that the release of doxorubicin in the nanocomposites was pH sensitive, and the cumulative release rate reached 80.53% at 60 h under acidic conditions. The nanocomposites had a high cellular uptake ability in MCF-7/ADR cells, and the IC50 value of the nanocomposites on MCF-7/ADR cells was 4.23 μg/mL, much smaller than that of free DOX (363.2 μg/mL). The nanocomposites could effectively reverse resistance and induce apoptosis of MCF-7/ADR cells. The blood biochemistry parameters and H&E staining results showed no serious adverse effects after treatment with the nanocomposites. Prussian blue staining showed that the nanocomposites were able to target tumor tissues in tumor-bearing mice under a magnetic field. The combined chemical/photodynamic therapy significantly inhibited tumor growth in vivo. Conclusion Nanocomposites with magnetic and pH dual-responsive performance has shown a promising platform for enhanced drug-resistant breast cancer treatment.

Abstract:Recently, magnetic photothermal nanomaterials have emerged as a new class of bio-nanomaterials for application in cancer diagnosis and therapy. Hence, we developed a new kind of magnetic nanomaterials, iron diselenide (FeSe2) nanoparticles, for multimodal imaging-guided photothermal therapy (PTT) to improve the efficacy of cancer treatment. By controlling the reaction time and temperature, FeSe2 nanoparticles were synthesized by a simple solution-phase method. After modification with polyethylene glycol (PEG), the obtained FeSe2-PEG nanoparticles showed high stability under various physiological conditions. FeSe2-PEG could serve as a T2-weighted magnetic resonance (MR) imaging contrast agent because of its strong superparamagnetic properties, with its r(2) relaxivity determined to be 133.38 mM(-1) S-1, a value higher than that of the clinically used Feridex. On the other hand, with high absorbance in the near-infrared (NIR) region, FeSe2-PEG also appeared to be a useful contrast agent for photoacoustic imaging (PA) as well as an effective photothermal agent for PTT cancer treatment, as demonstrated in our animal tumor model experiments. Moreover, long-term toxicity tests have proven that FeSe2-PEG nanoparticles after systematic administration rendered no appreciable toxicity to the treated animals, and could be gradually excreted from the major organs of mice. Our work indicates that FeSe2-PEG nanoparticles would be a new class of theranostic agents promising for application in bioimaging and cancer therapy.

Abstract:Nanotechnology possesses the potential to revolutionize the diagnosis and treatment of tumors. The ideal nanoparticles used for in vivo cancer therapy should have long blood circulation times and active cancer targeting. Additionally, they should be harmless and invisible to the immune system. Here, we developed a biomimetic nanoplatform with the above properties for cancer therapy. Macrophage membranes were reconstructed into vesicles and then coated onto magnetic iron oxide nanoparticles (Fe3O4 NPs). Inherited from the Fe3O4 core and the macrophage membrane shell, the resulting Fe3O4@MM NPs exhibited good biocompatibility, immune evasion, cancer targeting and light-to-heat conversion capabilities. Due to the favorable in vitro and in vivo properties, biomimetic Fe3O4@MM NPs were further used for highly effective photothermal therapy of breast cancer in nude mice. Surface modification of synthetic nanomaterials with biomimetic cell membranes exemplifies a novel strategy for designing an ideal nanoplatform for translational medicine.

Abstract:Cancer comprehensive treatment has been fully recognized as it can provide an effective multimodality approach for fighting cancers. In therapeutic oncology, hyperthermic adjuvant chemotherapy termed as thermochemotherapy plays an increasing role in multimodality cancer treatment. Currently, targeted nanothermotherapy is one of the effective hyperthermia approach based on magnetic nanoparticles (MNPs), which can be achieved by applying biocompatible nanoscaled metallic particles that convert electromagnetic energy into heat, for instance, magnetic fluid hyperthermia (MFH) mediated by superparamagnetic iron oxide nanoparticles (SPIONs). Upon exposure under alternative magnetic field (AMF), SPIONs can generate heat through oscillation of their magnetic moment. Nowadays, clinical trials at phase II are now under investigations for MFH on patients in Germany and Japan and demonstrate very inspiring for cancer therapy. In this work we explore the feasibility and effectiveness of magnetic thermochemotherapy mediated by magnetic nanoparticles combined with methotrexate, an anti-cancer drug, for breast cancer comprehensive treatment. Amino silane coated MNPs as agent of MFH were prepared by the chemical precipitation method. Physiochemical characterizations on MNPs have been systematically carried out by various instrumental analyses. Inductive heating property of the MNPs was evaluated by monitoring the temperature increase of the MNPs suspension under AMF. The in-vitro cytotoxicity results on human breast cancer cell MCF-7 by CCK-8 assay indicated the bi-modal cancer treatment approach for combined MFH and chemotherapy is more effective than mono-modal treatment, indicating a thermal enhancement effect of hyperthermia on drug cytocoxicity. The magnetic thermochemotherapy mediated by MNPs combined with methotrexate can realize cancer comprehensive treatment thus has great potential in clinical application.
Biomimic FeS2 Nanodrug with Hypothermal Photothermal Effect by Clinical Approved NIR-Ⅱ Light for Augmented Chemodynamic Therapy

Abstract:Hypothermal photothermal therapy (HPTT) employs hyperthermia (< 45 degrees C) to destroy tumor cells with negligible side effects to the surrounding normal tissues. Despite extensive studies, the clinical translation of HPTT is severely hindered, owing to the discounted therapeutic effect and maximum permissible power of near-infrared (NIR) laser by food and drug administration (FDA). Herein, we report a rational design of red blood cell membranes (RBCs) coated FeS2 (FeS2@RBCs) with strong absorption at NIR-II window for effective HPTT augmented chemodynamic therapy (CDT). FeS2(@)RBCs exhibits prolonged blood circulation and negligible immune response, leading to improved tumor accumulation for enhanced HPTT. Furthermore, the CDT effect of FeS2@RBCs is significantly augmented by the temperature elevation in the tumor region, which leads to the synergetic HPTT and CDT. Lipidomics analysis reveals that the damage of tumor cells by CDT is via the lipid peroxidation. In addition, FeS2@RBCs exhibits self-enhanced magnetic resonance imaging after reacting with H2O2 in tumor region for imaging-guided laser irradiation. Thus, FeS2@RBCs achieves remarkable inhibition of subcutaneous 4T1 breast tumor growth without obvious side effects by a 1064 nm laser irradiation of 1.0 W/cm(2) (FDA approved power density). Overall, this work provides a HPTT augmented CDT strategy for effective cancer therapy with a clinical approved laser power, which may pave the way for the clinical application of HPTT augmented CDT in the future.
Injectable Liquid Metal- and Methotrexate-Loaded Microsphere for Cancer Chemophotothermal Synergistic Therapy.

Abstract:With inherent fluidity and prominent photothermal conversion efficiency, multifunctional liquid metals (LMs) have been increasingly explored for various biomedical applications, especially as cancer therapeutic materials. Here, a series of LM- and/or methotrexate (MTX)-loaded microspheres, which are prepared by chitosan (CS) and formaldehyde, are employed to realize cancer chemophotothermal synergistic therapy under near-infrared (NIR) laser irradiation. The size of these microspheres is controllable and adjustable, and the preparation process is rather convenient. The LM- and MTX-loaded microspheres (CS/LM/MTX microspheres) are in the size range of 6-80 μm and exhibit an excellent photothermal effect of LM droplets, and MTX release, and thereby efficient in the inhibition of 4T1 cancer cell growth in vitro and in vivo. Further, the CS/LM/MTX microspheres were applied to tumor-bearing mice with intratumoral injection. The CS/LM/MTX microspheres can raise the local temperature of tumors to 60 °C in 1 min under NIR laser irradiation. Combined with the drug release of MTX, these microspheres showed better antitumor efficiency in vivo. Moreover, these microspheres display low toxicity and good biocompatibility, which are significant for practices in vivo. Hence, the demonstrated LM- and/or MTX-loaded microspheres reveal their superiority for cancer chemophotothermal synergistic therapy with NIR laser irradiation.
Temperature and Tumor Microenvironment Dual Responsive Mesoporous Magnetic Nanospheres for Magnetothermal Effect Induced Cancer Theranostics

Abstract:The development of smart drug delivery systems (SDDSs) based on engineered nanomaterials is important for clinical applications. Nevertheless, controllable administration of chemotherapeutic drugs for deep tumors and the avoidance of side effects caused by off-targeting during delivery remain a great challenge. Herein, a stimulus-responsive system of mesoporous nanospheres (composed of Cu@Fe2C@mSiO(2)) with good magnetothermal effect is introduced into the tumor microenvironment. This system plays an important role in image-guided controllable targeted drug delivery that is independent of tumor depth. Aggregation-induced emission luminogen-based fluorescence imaging and magnetic resonance imaging were utilized since these techniques visualize the delivery process in real time. In addition, the degraded nanocarriers showed high catalytic activity for Fenton and Fenton-like reactions, upregulating the level of hydroxyl radicals (center dot OH) in cancer cells to realize chemodynamic therapy. The induced center dot OH led to the overexpression of pho-STAT3, activating the STAT3 signaling pathway, eventually inducing cancer cell apoptosis. Through metabolic monitoring, this SDDS is removed from the body after its degradation in vivo. The synergistically enhanced therapeutic effect was obtained in the chemo-chemodynamic therapy of 4T1 tumor-bearing mice, offering a platform for efficient cancer therapy with a personalized theranostic strategy. Abstract:Magnetite‐based nanozymes have attracted great interest for catalytic cancer therapy enabled by catalyzing hydrogen peroxide (H2O2) to produce highly toxic hydroxyl radicals (•OH) to kill tumor cells. However, their therapeutic efficacies remain low due to insufficient •OH. Here, we report a light‐responsive carbon‐encapsulated magnetite nanodoughnuts (CEMNDs) with dual‐catalytic activities for photothermal‐enhanced chemodynamic therapy (CDT). The CEMNDs can accumulate in tumor and get into tumor cells and effectively act as peroxidase to convert H2O2 to hydroxyl radicals (•OH) that cause tumor cell death. The CEMNDs also possess intrinsic glutathione oxidase‐like activity that which catalyzes the oxidation of reduced glutathione and produce lipid peroxidase for enhanced catalytic therapy. Furthermore, the CEMNDs can absorb 1064 nm light to elevate local temperature and increase release of Fe ions for photothermal therapy (PTT) and enhanced CDT respectively. Our in vivo experiments in an aggressive and drug‐resistant metastatic mouse model of triple negative breast cancer (TNBC) model demonstrate excellent synergistic anti‐tumor function and no measurable systemic toxicity of CEMNDs. Synergistic Theranostics of Magnetic Resonance Imaging and Photothermal Therapy of Breast Cancer Based on the Janus Nanostructures Fe3O4-Aushell-PEG.

Abstract:Background: Satisfactory prognosis of breast cancer (BC) is limited by difficulty in early diagnosis and insufficient treatment. The combination of molecular imaging and photothermal therapy (PTT) may provide a solution. Methods: Fe3O4-Aushell nanoparticles (NPs) were prepared by thermal decomposition, seeded growth and galvanic replacement and were comprehensively characterized. After conjugated to PEG, NPs were used as MRI and PTT agents in vitro and in vivo. Results: Fe3O4-Aushell NPs which had uniform Janus-like morphology were successfully synthesized. The Fe3O4 had a size of 18 +/- 2.2 nm, and the Aushell had an outer diameter of 25 +/- 3.3 nm and an inner diameter of 20 +/- 2.9 nm. The NPs showed superparamagnetism, a saturation magnetization of 36 emu/g, and an optical absorption plateau from 700 to 808 nm. The Fe3O4-Aushell NPs were determined to possess good biocompatibility. After PEG coating, the zeta potential of NPs was changed from -23.75 +/- 1.37 mV to -13.93 +/- 0.55 mV, and the FTIR showed the characteristic C-O stretching vibration at 1113 cm(-1). The NPs' MR imaging implied that the T-2 can be shortened by Fe3O4-Aushell NPs in a concentration dependent manner, and the Fe3O4-Aushell NPs coated with PEG at the molar ratio of 160 (PEG: NPs) showed the highest transverse relaxivity (r2) of 216 mM-1s-1. After irradiation at 0.65 W/cm(2) for 5 min, all cells incubated with the Fe3O4-Aushell-PEG160 NPs (Fe: 30 ppm, Au: 70 ppm) died. After administrated intratumorally, Fe3O4-Aushell-PEG160 notably decreased the signal intensity of tumor in T2WI images. Under the same irradiation, the temperature of tumors injected with Fe3O4-Aushell-PEG160 quickly rose to 54.6 degrees C, and the tumors shrank rapidly and were ablated in 6 days. Conclusion: Fe3O4-Aushell-PEG NPs show good r2 and PTT performance and are promising synergistic theranostic agents of MRI and PTT for BC.
