Abstract:The essential oil from the leaves of Blumea densiflora var. hookeri was analyzed using GC–MS, and bioassays were performed. GC‐MS identified thirty‐four constituents, representing 98.7% of the oil composition. The major compounds identified in the oil were (E)‐caryophyllene (15.3%), viridiflorol (5.5%), trans‐dauca‐4(11),7‐diene (4.6%), spathulenol (4.4%), and isolongifolan‐7‐α‐ol (4.3%). The antimicrobial activity of the oil was assessed to determine its efficacy against six bacterial strains and one fungal strain using minimum inhibitory concentration (MIC) methods. The essential oil demonstrated antimicrobial activity against all tested microorganisms. A cytotoxicity assay was performed on KB and A549 cancer cell lines using the MTT method. The essential oil displayed strong cytotoxic effects on both cell lines, with IC50 values of 1.40 ± 0.05 μg/mL for KB and 1.72 ± 0.04 μg/mL for A549.

Abstract:Plant based compounds are frequently modified, developed into drug components to treat different types of cancer. Celery is one of the most used ingredients in daily life, exhibits potential medicinal properties and myristic acid exhibits anti microbial and inflammatory properties making it ideal candidate for combinational therapy. This study demonstrates the formulation of celery oil-myristic acid based microemulsion system with sequence of weight/volume of components to optimize stable microemulsion. the sequence of formulations were performed, optimized through pseudo ternary phase diagram and stability analysis, of which CMA ME 4 formulation were observed to transparent and stable. This CMA ME 4 was analyzed using DLS for particle size which is found to be 14.0±27.1 and PDI 0.34±0.46 through 5th day to 30th day. In vitro drug release study has been performed with stimulated physiological condition (pH 5.6) and stimulated tumor tissue (pH 7.4), the microemulsion was withdrawn in time intervals and the kinetic model is constructed to detect the behaviour of the drug release profile. Further, anti inflammatory, anti diabetic and anti obesity assays were performed to analyse the potentiality of CMA ME 4 microemulsion formulation, where the results exhibited from 80 to 90% radical scavenging and inhibition compared with the synthetic drugs. A good cytotoxic activity of CMA ME 4 microemulsion was determined by positive results of the MTT assay performed with HeLa with 80 to 90% cell death, MCF7 with 80 to 90% cell death and HUVECs was used to determine the toxicity level when the microemulsion interacts with the normal cell which is found to be 80 to 90% cell viability over time. These findings more suitably suggest that combination of natural oil with synthetic components can be effective in treatment against diseases.

Abstract:Murraya paniculata (L.) Jack is commonly cultivated as ornamental plant in Assam and has been used as spice and phytomedicine traditionally for many healthcare purposes. The therapeutic potential and chemical constituents of the essential oil of M. paniculata leaf was investigated against several pathogenic microbial species and human cancer cell lines. 29 chemical compounds were identified by GC–MS analysis from the essential oil representing 97.62% of the oil. The major compound identified was caryophyllene (20.93%). Leaf essential oil exhibited promising antibacterial activity against Mycobacterium smegmatis (MIC = 4 µg/mL) and Pseudomonas aeruginosa (MIC = 4 µg/mL). Best anticancer activity of the oil was observed for HeLa cells (IC50 = 6.28 μg/mL). Further, scanning electron microscopic studies revealed that the oil kills micro-organisms with the deformation of cellular morphology on treatment of the oil. Thus, the essential oil of M. paniculata leaf can be an excellent alternative for development of new antimicrobials and anticancer chemotherapeutic agents for the pharmaceutical industries.

Abstract:Androgen levels in blood and the behaviour of male camels were determined during 1 year. In the months January to April large amounts of androgens were found in blood serum (approximately 30 ng/ml compared with approximately 2 ng/ml throughout the remaining months) as well as in a secretion from glands on the back of the neck (36 ng/ml). During this period when hormone levels were high the behaviour of the camels changed: they became unmanageable, blew-out a palatal flap from the mouth, vocalized, and spread urine over their backs with the aid of their tails. The changes in the sexual cycle of the male camels corresponded with the oestrous period in female camels.

Abstract:Aralia chinensis L. is a traditional Miao ethnic medicine known for its pain and inflammation relief and its ability to dispel wind and dampness. This study aimed to assess its antitumour activity and identify the chemical constituents of A. chinensis essential oils. Gas chromatography–mass spectrometry was used to analyse the volatile oil composition, which identified 35, 35, and 24 constituents in essential oils from roots, stems, and leaves, respectively. Network pharmacology predicted the possible key targets of common components in breast-cancer treatment, which revealed AKT1, SRC, EGFR, STAT3, and MAPK3 as high-priority targets with high active-constituent affinity. CCK-8 assay confirmed the inhibitory effect of the essential oils on MCF-7 breast-cancer cells, with oils from Aralia rhizomes, stems, and leaves inhibiting cell viability by 77%, 64%, and 62%, respectively. The active components of Aralia essential oils show promise for breast-cancer treatment by targeting AKT1, SRC, EGFR, and other key factors.

Abstract:Mikania micrantha Kunth (mile-a-minute) is a major invasive weed with medicinal properties. It causes destruction of forests and threat to crop plantations, thus affecting the agriculture-based economy of a country. As an attempt to mitigate this problem, we have studied the applicability of M. micrantha flower essential oil against disease causing microorganisms and inhibition of cancerous cells. Sixty-six compounds were characterized from the flower essential oil by gas chromatography–mass spectrometry. The major constituents were: Isoledene (16%), δ-Cadinene (11.2%), Debromofiliformin (9.4%), Trans-Caryophyllene (9.1%), β-Bisabolene (7.4%), Germacrene-D (5.8%), and Zingiberene (5.3%). The oil is a complex mixture containing mainly sesquiterpene hydrocarbons (62.9%) and oxygenated sesquiterpenes (23.9%) with the other terpene compounds. Well-diffusion and resazurin assay were conducted to determine antimicrobial activity and minimum inhibitory concentration (MIC) of the essential oil. Flower oil showed best activity against Mycobacterium smegmatis and Candida albicans with MIC 8 µg/mL. Moreover, it is the first report of antimicrobial activity of the oil towards M. smegmatis, C. albicans, Staphylococcus aureus, and Pseudomonas aeruginosa. Minimum bactericidal and fungicidal concentration was determined and mechanism of antimicrobial activity was investigated by Field Emission-Scanning Electron Microscopy and Transmission Electron Microscopy analysis. It is the first cytotoxicity report of M. micrantha flower essential oil for cervical, ovarian, and pancreatic cancer cell lines. Through MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, we have found that oil has excellent cytotoxicity against HeLa cervical cancer cell lines (IC50 5.44 ± 1.33 µg/mL) and PA1 ovarian cancer cell lines (IC5010.57 ± 1.44 µg/mL). The essential oil extracted from the invasive weed M. micrantha flower is now found to have antimicrobial and anticancer properties from our study. Thus, this harmful weed can be utilized as antimicrobial and anticancer agents for the healthcare of human being.
Effects of acute or 3-day treatments of Hypericum caprifoliatum Cham. & Schltdt. (Guttiferae) extract or of two established antidepressants on basal and stress-induced increase in serum and brain corticosterone levels

Abstract:Since depressive patients present alterations in the hypothalamo-pituitary-adrenal (HPA) axis that are normalised by antidepressants, this HPA axis has been considered as a target of their actions. We have investigated the mechanism of action of a cyclohexane extract of Hypericum caprifoliatum (HCP), which displays antidepressant like activity, by studying, in mice, the influence of HCP and of two established antidepressant drugs, imipramine and bupropion, administered either acutely or semi-chronically (once a day, three consecutive days), on serum and brain cortex corticosterone levels, either in basal conditions or shortly after a forced-swimming session (FSS). Administered acutely, imipramine (20 mg/kg, per os (p.o.)), bupropion (30 mg/kg, p.o.) and HCP (360 mg/kg, p.o.) significantly reduced the immobility time and had no effects on FSS-induced increase of serum and cortical corticosterone levels. Conversely, 3 days repeated treatment with imipramine or bupropion resulted in a significant reduction of immobility time and FSS-induced increase of serum and cortical corticosterone levels. In a different way, repeated treatment with HCP significantly reduced the immobility time and only cortical corticosterone levels in stressed mice. These results indicate that short-term treatments with antidepressants are sufficient to induce modifications in the HPA axis reactivity to stress; and that apparently HCP has an influence on corticosterone levels by a mechanism diverse from the other tested antidepressants.
Chemical Composition, In vitro and In silico Evaluation of Essential Oil from Ocimum tenuiflorum and Coriandrum sativum Linn for Lung Cancer

Abstract:Background: Medicinal plants play an essential role in everyday life; plants highly contain therapeutic phytoconstituents commonly used to treat various diseases. This paper discusses the Chemical composition, In vitro antiproliferative activity and In silico study of essential oil extracted from Ocimum tenuiflorum (family Lamiaceae) and Coriandrum sativum (family Apiaceae). Objective: In present study GC-MS was used to identify the chemical constituents from O. tenuiflorum and C. sativum. In vitro antiproliferative activity was performed on A549 cancer cell lines. In silico study was performed by Schrodinger's maestro software to identify chemical constituents in both plants as potential EGFR inhibitors for the treatment of lung cancer. Methods: The essential oil was extracted by hydro distillation from aerial parts of O. tenuiflorum and C. sativum. The volatile oil sample was analyzed by (GC-MS) Gas Chromatography- Mass Spectrometry. Different chemical constituents were identified based on the retention index and compared with the NIST library. The oil samples from O. tenuiflorum and C. sativum was also evaluated for antiproliferative activity against human lung cancer A549 cell lines. In silico study was performed by Schrodinger maestro software against EGFR (PDB ID 5HG8). Results: O. tenuiflorum essential oil contains Eugenol (42.90%), 2-β-Elemene (25.98%), β- Caryophyllene (19.12%) are the major constituents. On the other side, C. sativum contains nnonadecanol- 1 (16.37%), decanal (12.37%), dodecanal (12.27%), 2-Dodecanal (9.67%), Phytol (8.81%) as the major constituents. Both the oils have shown in vitro antiproliferative activity against human lung cancer cell lines A549 having IC50 values of 38.281 μg/ml (O. tenuiflorum) and 74.536 μg/ml (C. sativum). Molecular interactions of constituents hydro distilled from two oils was analysed by schrodinger maestro software against EGFR (PDB ID 5HG8). Conclusion: The oil sample extracted from O. tenuiflorum showed more antiproliferative activity than C. sativum. In silico study showed that two chemical constituents, namely di-isobutyl phthalate (-7.542 kcal/mol) and dibutyl phthalate (-7.181 kcal/mol) from O. tenuiflorum and one diethyl phthalate (-7.224 kcal/mol) from C. sativum having more docking score than standard Osimertinib which indicates the effectiveness of oils for lung cancer.

Abstract:The composition and the bioactivity of the essential oil of Artemisia anomala, a traditional medicinal herb in China, were investigated. The composition of the essential oil was analyzed by gas chromatography-mass spectometry (GC-MS), and a total of 39 components representing 98.1% in this plant were identified. The main constituents were camphor (18.3%), 1, 8-cineole (17.3%), -caryophyllene oxide (12.7%), and borneol (9.5%). The antimicrobial activities of the oil were separately evaluated against six micro-organisms, Bacillus cereus (CMCCB 63301), Micrococcus flavus (ATCC 14698), Escherichia coli (ATCC 11775), Pseudomonas aeruginosa (ATCC 10145), Staphylococcus epidermidis (ATCC 12228), and a clinical-isolated strain Staphylococcus aureus. The results showed that the oil possessed inhibitory activity against B. cereus, M. flavus, E. coli, S. epidermidis, and S. aureus, among which the best inhibition was observed against B. cereus with a MIC of 0.313L/mL. The antiproliferative activities of the oil were assessed against four human tumor cell lines, A459 (human lung adenocarcinoma), BRO (human melanoma), MCF7 (human breast cancer), and PC3 (human prostate). The oil showed the ability to inhibit proliferation of human cancer cells. And the MCF7 cell line was the most sensitive to the oil with an IC50 of 0.09.

Abstract:Bioactive components used to treat human ailments are abundant in medicinal plants. Mentha arvensis and Aegle marmelos have therapeutic properties due to the presence of many phytonutrients. Gas chromatography–mass spectrometry (GC-MS) was used to evaluate the properties of the antioxidant and anticancer bioactive components found in the ethanolic extracts of Mentha arvensis and Aegle marmelos leaves. The major constituents identified in the leaf extracts were octadecanoic acid, heptadecanoic acid, 15-methyl-9,12,15-octadecatrienoic acid, ethyl ester, hexadecanoic acid, neophytadiene, pentadecane, hexadecane, and 1-octadecanol. Some of these components have antibacterial, antioxidant, anticancer, and anti-inflammatory properties. Among the identified compounds, octadecanoic acid and n-hexadecanoic acid are pivotal for their anticancer and antioxidant effects. The GC–MS characterisation indicated the existence of abundant bioactive compounds, confirming their potential therapeutic activities. This study showed that the leaves of Mentha arvensis and Aegle marmelos could be used as novel sources of antioxidants and anticancer medicines.

Abstract:In this study, the volatile components of Erigeron sublyratus essential oils and their anti‐inflammatory and cytotoxic activities were investigated for the first time. Gas chromatography‐mass spectrometry (GC‐MS) analysis revealed that a total of 28 components were identified in the root and aerial part essential oils. Among them, cis‐lachnophyllum ester (53.4‐64.2%), followed by germacrene D (5.6‐8.6%), trans‐β‐ocimene (2.6‐7.5%), β‐caryophyllene (4.7‐6.8%), β‐myrcene (2.0‐6.3%), and (E)‐β‐famesene (4.8‐5.0%) were principal components. The root essential oil significantly inhibited nitric oxide (NO) production on LPS‐induced RAW264.7 cells (IC50 = 1.41 ± 0.10 μg/mL) as compared to standard, dexamethasone (IC50 = 5.43 ± 0.54 μg/mL). Besides, both root and aerial part essential oils exhibited cytotoxic activity against MCF‐7, SK‐LU‐1, and HepG2 (IC50 from 1.11 ± 0.04 to 1.70 ± 0.05 μg/mL). Molecular docking simulation results show that (E)‐β‐farnesene exhibits the strongest binding energy among the studied compounds with the VEGFR‐2 enzyme (ΔG = ‐7.295 kcal/mol), while δ‐cadinene demonstrates the strongest affinity (ΔG = ‐8.047 kcal/mol) towards the COX‐2 enzyme. Furthermore, hydrophobic interactions were indicated to be the main contributors to the binding ability in the studied protein‐ligand complex. These findings proposed that E. sublyratus can be exploited for its anti‐inflammatory and anti‐cytotoxicity potential.

Abstract:The ubiquitous occurrence of Lantana camara highlights its exceptional ecological versatility. Leaves of L. camara were obtained for this study from two different locations of NE, India. Hydro distillation was used to extract the essential oils from the leaves, and gas chromatography-mass spectrometry (GC-MS) and gas chromatography-flame ionization detection (GC-FID) were used to study the chemical profiles of the isolated oils. The essential oil analysis detected 35 components in total essential oil from Location 1 (BLCEO) while in Location 2 essential oil (JLCEO), 29 components were identified. Caryophyllene, humulene and sabinene were present in considerably major amounts in both BLCEO and JLCEO, though variation in area percentage was seen. The anti-oxidant properties of BLCEO were better than that of JLCEO. The BLCEO has also exhibited better results in inhibiting acetylcholinesterase. However, both the essential oils did not show any prominent results in α-amylase inhibitory assay and anti-tyrosinase activity. In this study, the phytochemical composition and their effect on the bioactivity of both samples were observed. The chemical constitution of L. camara essential oils (LCEO) and its possible ecological ramifications have also been well explained.

Abstract:This study was aimed to explore the chemical composition and biological activities of essential oil from aerial part of Mosla dianthera along with its major isolated compound, carvone. The hydro-distilled essential oil was analysed by GC-MS and biological activities were investigated in terms of antioxidant, anti-inflammatory, herbicidal, antibacterial, anti-fungal and anti-feedant properties. GC-MS analysis led to the identification of forty-nine components contributing 96.2% of essential oil with carvone (41.9%) as the most abundant constituent. The oil and carvone showed good to moderate antioxidant potentials determined by radical scavenging, reducing power and metal chelating activities. Carvone showed good anti-inflammatory activity (78.0%) compared to essential oil (74.2%). Both essential oil and carvone exhibited excellent herbicidal activity against Raphanus raphanistrum subsp. sativus seeds. The essential oil and carvone showed significant anti-bacterial efficacy against Bacillus cereus and Escherichia coli. It was observed that essential oil showed strong antifungal property than carvone against Alternaria alternata and Curvularia lunata. Both the samples exhibited anti-feedant activity in a dose dependent manner against third instar larvae of Spilosoma obliqua. Results obtained revealed the possible applications of essential oil and carvone as a bioactive source of natural antioxidants, excellent herbicide and an effective substance for antifungal and antifeedant activities.

Abstract:Artemisia vulgaris is an enormously useful aromatic plant known for its insecticidal, antifungal, parasiticidal, and medicinal values. The main aim of this study is to investigate phytochemical contents and the potential antimicrobial activities of Artemisia vulgaris essential oil (AVEO) from the fresh leaves of A. vulgaris grown in Manipur. The AVEO isolated by hydro-distillation from A. vulgaris were analyzed by gas chromatography/mass spectrometry and solid-phase microextraction-GC/MS to describe their volatile chemical profile. There were 47 components identified in the AVEO by GC/MS, amounting to 97.66% of the total composition, while 97.35% were identified by SPME-GC/MS. The prominent compounds present in AVEO analyzed by direct injection and SPME methods are found to be eucalyptol (29.91% and 43.70%), sabinene (8.44% and 8.86%), endo-Borneol (8.24% and 4.76%), 2,7-Dimethyl-2,6-octadien-4-ol (6.76% and 4.24%), and 10-epi-γ-Eudesmol (6.50% and 3.09%). The consolidated component in the leaf volatiles comes to the terms of monoterpenes. The AVEO exhibits antimicrobial activities against fungal pathogens such as Sclerotium oryzae (ITCC 4107) and Fusarium oxysporum (MTCC 9913) and bacterial cultures such as Bacillus cereus (ATCC 13061) and Staphylococcus aureus (ATCC 25923). The percent inhibition of AVEO against the S. oryzae and F. oxysporum was found up to 50.3% and 33.13%, respectively. The MIC and MBC of the essential oil tested for B. cereus and S. aureus were found to be (0.3%, 0.63%) and (0.63%, 2.5%), respectively. Finally, the results revealed that the AVEO characterized by the hydro-distillation and SPME extraction yielded the same chemical profile and showed potent antimicrobial activities. Further research into A. vulgaris's antibacterial properties can be performed in order to use it as a source for natural antimicrobial medications.

Abstract:Plasma air purifying systems present an interesting alternative to filters for purifying air. In this study, molecular processes in a commercially available ac driven plasma air purifier were studied in detail. This air purifier is supposed to reduce all air contaminants to small nontoxic molecules (e.g., H(2)O and CO(2)). However, degradation mechanisms are not yet fully understood. In this study, we investigated the exhaust of the plasma air purifier to determine which degradation products are formed. An interface was designed and constructed to allow the direct coupling of the plasma air purifier's exhaust to a mass spectrometer. The compounds studied, primary and secondary amines, were introduced at a concentration of 1 ppmV. Contrary to our expectations, polymerization instead of degradation was observed. The higher the ac voltage applied (max. 9.0 kV) to the plasma air purifier, the higher the mass of the oligomer distribution. Side chain oxidation products as well as oligomers could be observed for all compounds tested. Starting with amines of low mass (m/z < 200), compounds of molecular masses above 1000 Da were observed in the plasma air purifier. Detailed analysis of the observed mass spectra as well as experiments with deuterated dibutylamine helped to unravel the mechanism taking place in the plasma air purifier. Nitrate anions generated in the plasma air purifier (presumably from N(2)) are proposed to form ionic clusters with protonated amines.

Abstract:Cruciferous vegetables are rich source of glucosinolates (GSLs), which in presence of myrosinase enzyme cause hydrolytic cleavage and result in different hydrolytic products like isothiocyanates, thiocyanates, nitriles and epinitriles. The GSLs hydrolytic products are volatile compounds, which are known to exhibit bioactivities like antioxidant, fungicidal, bioherbicidal and anticancer. Among the Brassicaceae family, Brassica juncea is very well known for high content of GSLs. In the present study, the isolation of volatile oil of B. juncea var. raya was done by hydrodistillation method using clevenger apparatus and further there extraction was done by solvents ethyl acetate and dichloromethane. The volatile compounds present in the extract were analysed by gas chromatography/gas chromatography–mass spectrometry (GC/GC–MS). Fatty acid esters, sulphur and/or nitrogen compounds, carbonyl compounds and some other volatile compounds were also identified. Besides the analytical studies, the extracts were analysed for their bioactivities including radical scavenging activity by using DNA nicking assay and cytotoxic effect using different human cancer cell lines viz. breast (MCF-7 and MDA-MB-231), prostate (PC-3), lung (A-549), cervix (HeLa) and colon (HCT116) by MTT assay. The oil extracts were efficiently able to reduce the increase of cancer cells in a dose-dependent manner. Among all cell lines, the most effective anticancer activity was observed in case of breast (MCF-7) cancer cell line. So, MCF-7 cells were used for further mechanistic studies for analysing the mechanism of anticancer activity. Confocal microscopy was done for analysing morphological changes in the cells and the images confirmed the features typical of apoptosis. For evaluating the mode of cell death, spectrofluorometric determination of reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) was done. The volatile oil extract treated MCF-7 cells had a significant increase in number of ROS, also there was a rise in percentage of cells with increased disruption of MMP. So, the present study marks necessary indication that B. juncea (raya) oil extracts significantly induces apoptosis in all the above mentioned cancer cells lines through a ROS-mediated mitochondrial pathway and thus play a remarkable role in death of cancer cells.

Abstract:Cistus albidus : L., also known as Grey-leaved rockrose and locally addressed as šṭab or tûzzâla lbîḍa, is a plant species with a well-established reputation for its health-promoting properties and traditional use for the treatment of various diseases. This research delves into exploring the essential oil extracted from the aerial components of Cistus albidus (referred to as CAEO), aiming to comprehend its properties concerning antioxidation, anti-inflammation, antimicrobial efficacy, and cytotoxicity. Firstly, a comprehensive analysis of CAEO's chemical composition was performed through Gas Chromatography-Mass Spectrometry (GC-MS). Subsequently, four complementary assays were conducted to assess its antioxidant potential, including DPPH scavenging, β-carotene bleaching, ABTS scavenging, and total antioxidant capacity assays. The investigation delved into the anti-inflammatory properties via the 5-lipoxygenase assay and the antimicrobial effects of CAEO against various bacterial and fungal strains. Additionally, the research investigated the cytotoxic effects of CAEO on two human breast cancer subtypes, namely, MCF-7 and MDA-MB-231. Chemical analysis revealed camphene as the major compound, comprising 39.21% of the composition, followed by α-pinene (19.01%), bornyl acetate (18.32%), tricyclene (6.86%), and melonal (5.44%). Notably, CAEO exhibited robust antioxidant activity, as demonstrated by the low IC 50 values in DPPH (153.92 ± 4.30 μg/mL) and β-carotene (95.25 ± 3.75 μg/mL) assays, indicating its ability to counteract oxidative damage. The ABTS assay and the total antioxidant capacity assay also confirmed the potent antioxidant potential with IC 50 values of 120.51 ± 3.33 TE μmol/mL and 458.25 ± 3.67 μg AAE/mg, respectively. In terms of anti-inflammatory activity, CAEO displayed a substantial lipoxygenase inhibition at 0.5 mg/mL. Its antimicrobial properties were broad-spectrum, although some resistance was observed in the case of Escherichia coli and Staphylococcus aureus . CAEO exhibited significant dose-dependent inhibitory effects on tumor cell lines in vitro . Additionally, computational analyses were carried out to appraise the physicochemical characteristics, drug-likeness, and pharmacokinetic properties of CAEO's constituent molecules, while the toxicity was assessed using the Protox II web server.

Abstract:Ethnopharmacological relevance: The genus Hedychium of family Zingiberaceae comprises several perennial rhizomatous species widely used in perfumery and traditional folk medicine to treat diseases related to asthma, diarrhoea, nausea, stomach disorders, inflammation and tumours. Several species of Hedychium have remained under-explored with respect to their chemical composition and biological activities. Aim of the study: The current research aimed to explore the chemical composition and evaluate the antiproliferative and anti-inflammatory activities of rhizome essential oil from four Hedychium species (H. coccineum, H. gardnerianum, H. greenii and H. griffithianum). Materials and methods: Compound identification was accomplished using a Clarus 580 gas chromatography system in conjunction with mass spectrometry (GC-MS). The multivariate data statistics using chemometrics (PCA, PLS-DA, sPLS-DA) and cluster analysis (Dendrogram, Heat maps, K-means) were used to compare the similarity and relationship among Hedychium metabolomes. MTT assay was employed to visualize the antiproliferative property against MCF7, HepG2 and PC3 cancerous cell lines. The toxicity of essential oils was determined using 3T3-L1 non-tumorigenic/normal cells. Lipopolysaccharide (LPS)-induced RAW 264.7 cells were used to investigate the anti-inflammatory properties of Hedychium essential oils by measuring the production of nitric oxide (NO) using the Griess reagent method. Furthermore, the levels of prostaglandin (PGE2) and pro-inflammatory cytokines (TNF-α, IL-6, IL-1β) was assessed using the ELISA technique. Results: In total, 82 compounds were identified in four targeted species of Hedychium from which 1,8-cineole (52.71%), β-pinene (32.83%), α-pinene (19.62%), humulene epoxide II (19.86%) and humulene epoxide I (19.10%) were the major constituents. Monoterpenes (8.5-59.9%) and sesquiterpenes (2.87-54.11%) were the two class of compounds, found as the most prevalent in the extracted essential oils. The multivariate analysis classified the four Hedychium species into three different clusters. Hedychium essential oils exhibited potent antiproliferative activity against MCF7, HepG2 and PC3 cancer cell lines with IC50 values less than 150 μg/mL where H. gardnerianum exhibited the highest selective cytotoxicity against human breast and prostate adenocarcinoma cells with an IC50 value of 44.04 ± 1.07 μg/mL and 56.11 ± 1.44 μg/mL, respectively. The essential oils on normal (3T3-L1) cells displayed no toxicity with higher IC50 values thereby concluding as safe to use for normal human health without causing any side effects. Besides, the essential oils at 100 μg/mL concentration revealed remarkable anti-inflammatory activity in LPS-activated RAW 264.7 murine macrophages by inhibiting the production of inflammatory mediators, with H. greenii exhibiting the maximum anti-inflammation response by significantly suppressing the levels of NO (84%), PGE2 (87%), TNF-α (94.3%), IL-6 (95%) and IL-1β (85%) as compared to LPS treated group. Conclusion: The present findings revealed that the Hedychium species traditionally used in therapeutics could be a potential source of active compounds with antiproliferative and anti-inflammatory properties.

Abstract:Murraya koenigii (L.) Spreng., is a renowned medicinal and aromatic plant having antimicrobial and broad biological activities. In the present study, the essential oil chemical profile of the leaf and floral part were analysed using GC-FID and GC-MS, and the antimicrobial activities were determined by disk diffusion assay. The scientific analysis revealed α-pinene (40.41%) was the major compound of leaf essential oil (MKL) followed by sabinene (29.66%), β-pinene (7.92%), caryophyllene (4.24%), limonene (3.39%), terpinen-4-ol (3.25%), γ-terpinene (2.63%), terpinolene (1.58%), linalool (1.32%), along with other trace components. The floral part (MKF) contains the compounds α-pinene (74.02%) as the major compound followed by β-pinene (7.17%), sabinene (5.72%), limonene (4.19%), β-myrcene (1.98%), γ-terpinene (1.47%) and terpinen-4-ol (1.41%) along with other trace compounds. The MIC result revealed essential oil showed effectiveness against Escherichia coli (40 μg/mL), Micrococcus luteus (80 μg/mL), Aspergillus fumigatus (60 μg/mL) and Aspergillus brasiliensis (80 μg/mL) for MKL while MKF did not exhibit potent antimicrobial activities. The essential oils revealed potent antioxidant potential as determined by DPPH method with IC 50 values of 14.02 μL/mL (MKL) and 5.45 μL/mL (MKF) and the ABTS method with IC 50 values of 5.07 μL/mL and 8.38 μL/mL in MKL and MKF respectively as compared to that of the standards. The essential oils revealed potent antidiabetic activity with 6.02 μL/mL (MKL) and 8.18 μL/mL (MKF) as that of the standard. However, both MKL and MKF were not much effective in protease inhibitory activity and anti-tyrosinase activity. The study results exhibited a potent bioactivity profile and revealed many new utilization aspects of this essential oil.

Abstract:Spilanthes paniculata , a member of the Asteraceae family, is predominantly used as a traditional remedy in addressing oral ailments, particularly gum infections and sore throat. The flowers are chewed to alleviate toothache immediately. This study evaluated a comparison between the essential oils of leaf and flower derived from Spilanthes paniculata targeting the SCC9 oral cell lines using in vitro and in silico approaches. The anticancer activity was performed through an MTT assay, apoptosis assays using annexin V and orospheres formation assays. Molecular docking was performed between five selected phytocompounds against the p53 protein by using AutoDock 4.2.6 software. The results confirmed that the flower essential oil significantly reduced the cell viability in a dose-dependent manner, with an IC 50 value of 113.95 μg/mL. The apoptosis assays showed that the flower essential oil was approximately 2.5 times more effective in inducing early apoptosis at 50 μg/mL compared to the essential oil of the leaf. The orosphere formation assays further confirmed the anticancer potential of the flower essential oil. Spathulenol exhibited strong hydrogen bonding with the p53 protein. The ADMET prediction tools were used to predict the in silico pharmacokinetics and drug-like properties of the phytoconstituents. The results suggested that Spathulenol and Nerolidol have high gastrointestinal absorption (GIA), with estimated solubility (ESOL) values of − 3.17 and − 3.22, respectively, falling within the optimal range. These findings suggest that the flower's essential oil efficiently prevented the proliferation of oral cancer and observed a notable degree of cell death, inducing apoptosis and suggesting its significant antiproliferative activity against cancerous cell line which could be explored further for therapeutic applications.
