Abstract:The emergence of antimicrobial-resistant (AR) Salmonella has a major concern worldwide. This study was designed to determine the AR profiles and serovars distribution of Salmonella enterica isolated from different breeds of breeder chickens in the province of Henan, China. For this, 2,139 dead embryo samples were collected from 28 breeder chicken hatcheries, representing two domestic and four foreign breeds. The samples were subjected to the isolation and identification of Salmonella by PCR. The confirmed strains were serotyped according to the Kauffmann-White scheme and their AR profiles against 20 antimicrobial agents were determined by Kirby-Bauer (K-B) disc diffusion method. The results of this study showed the prevalence of Salmonella in 504 strains (23.56%) with a high abundance in southern regions of Yellow River (28.66%, n = 495, N = 1,727) compared to the northern regions (2.18%, n = 9, N = 412) (p < 0.0001). The domestic breeds were more contaminated than imported breeds (p < 0.0001). However, the contamination rate of samples recovered from M-hatcheries was the highest (p < 0.0001). Serotyping method identified 12 serovars, with the dominance of S. Pullorum (75.79%), followed by S. Enteritidis (7.14%). The AR assay showed high resistant to ciprofloxacin (77.00%), sulfisoxazole (73.00%), and ampicillin (55.60%), as well as 98.81% (n = 498) of the isolated strains, were resistant to at least one antimicrobial and 69.64% (n = 351) were resistant to three or more antimicrobials. Among them, one strain of S. Thompson was resistant to 15 antimicrobial agents belonging to eight different classes. In conclusion, Salmonella strains isolated in this study were multidrug-resistant (MDR), presenting a serious problem for human and animal health. Therefore, it is necessary to monitor, control, and rationalize the use of antimicrobials agents in chicken farms in order to limit the increasing resistance against the recent antimicrobial agents.
Antimicrobial Resistance Profiles and Genetic Typing of Salmonella Serovars from Chicken Embryos in China

Abstract:Salmonella continues to be a major food and public health burden worldwide that can threaten human health via eating contaminated meats, particularly those originating from chicken. In this study, the antimicrobial resistance profiles, epidemiological characteristics of resistance genes, and pulsed field gel electrophoresis (PFGE-XbaI) typing of 120 non-Pullorum/Gallinarum Salmonella isolates recovered from chicken embryos in Henan province were determined. The antimicrobial resistant phenotypes and evaluation of the extended-spectrum beta-lactamases (ESBLs) producing strains of Salmonella were investigated by the Kirby-Bauer test and the double-disk synergy test. Additionally, 37 antimicrobial resistance genes encoding resistance to five different categories, including aminoglycosides, cephalosporins, sulphonamides, tetracyclines, and beta-lactams, were examined by conventional PCR. However, genotyping analysis was conducted by macro-restriction using enzyme XbaI followed by the separation of the restricted DNA fragments by PFGE. The results of this study showed that the studied Salmonella strains were highly resistant to ampicillin (66.67%) and sulfisoxazole (66.67%), while they were all susceptible to meropenem, imipenem, colistin, and chloramphenicol. Additionally, 67.5% (81/120) of the studied strains were multidrug resistant, and 21.67% (26/120) were phenotypically confirmed as ESBLs positive. The statistical analysis showed that resistance depends on the serovars, and ESBLs positive strains showed more multi-resistance than ESBLs negative strains (p < 0.05). The genotypic antimicrobial resistance showed the detection of 14 among the 37 tested genes, and the concordance between genotypic and phenotypic antimicrobial resistance ranged from 0% to 100% depending on the serovars. However, the PFGE-XbaI typing results showed that the examined Salmonella strains were divided into 22 individual subtypes and were grouped in nine clusters, with similarity values ranging from 64.7% to 100%. From this study, we can conclude that the antimicrobial resistance of Salmonella serovars isolated from chicken embryos in Henan province was alarming, with rigorous multidrug resistance, which requires the urgent mitigation of the use of antimicrobial drugs in chicken hatcheries. Additionally, our results showed evidence of the presence of different PFGE patterns among the studied Salmonella serovars, suggesting the presence of different sources of contamination.</p>
Prevalence, virulence factor and antimicrobial resistance analysis of Salmonella Enteritidis from poultry and egg samples in Iran

Abstract:Abstract Background Salmonella enterica serovar Enteritidis ( S . Enteritidis) is one of the most common serovars, associated with human salmonellosis. The food-borne outbreak of this bacterium is mainly related to the consumption of contaminated poultry meat and poultry products, including eggs. Therefore, rapid and accurate detection, besides investigation of virulence characteristics and antimicrobial resistance profiles of S . Enteritidis in poultry and poultry egg samples is essential. A total of 3125 samples (2250 poultry and 875 poultry egg samples), sent to the administrative centers of veterinary microbiology laboratories in six provinces of Iran, were examined for Salmonella contamination, according to the ISO 6579 guideline. Next, duplex PCR was conducted on 250 presumptive Salmonella isolates to detect invA gene for identification of the genus Salmonella and sdf gene for identification of S . Enteritidis. Subsequently, the S. Enteritidis isolates were examined for detection of important virulence genes ( pagC , cdtB , msgA , spaN , tolC , lpfC , and spvC ) and determination of antibiotic resistance patterns against nalidixic acid, trimethoprim-sulfamethoxazole, cephalothin, ceftazidime, colistin sulfate, and kanamycin by the disk diffusion method. Results Overall, 8.7 and 2.3% of poultry samples and 6.3 and 1.3% of eggs were contaminated with Salmonella species and S. Enteritidis, respectively. The invA and msgA genes (100%) and cdtB gene (6.3%) had the highest and the lowest prevalence rates in S . Enteritidis isolates. The spvC gene, which is mainly located on the Salmonella virulence plasmid, was detected in 50.8% of S . Enteritidis isolates. The S . Enteritidis isolates showed the highest and the lowest resistance to nalidixic acid (87.3%) and ceftazidime (11.1%), respectively. Unfortunately, 27.0% of S . Enteritidis isolates were multidrug-resistant (MDR). Conclusion The rate of contamination with Salmonella in the poultry and egg samples, besides the presence of antimicrobial resistant and MDR Salmonella isolates harboring the virulence genes in these samples, could significantly affect food safety and subsequently, human health. Therefore, continuous monitoring of animal-source foods, enhancement of poultry farm control measures, and limiting the use of antibiotics for prophylactic purposes in food producing animals, are essential for reducing the zoonotic risk of this foodborne pathogen for consumers and also choosing effective antibiotics for the treatment of salmonellosis.

Prevalence of Salmonella contamination in consumed eggs in Iran: A systematic review and meta-analysis study on published studies from 1996 to 2018

Abstract:Background and Aim: Food poisoning caused by Salmonella is among the most common gastrointestinal discomfort resulted from egg consumption which can produce various syndromes. The present study is a systematic review and metaanalysis investigation on the published studies about the prevalence of Salmonella contamination in the consumed eggs in Iran. Materials and Methods: The data were collected and analyzed from four international search databases, including PubMed, Scopus, Science Direct, and Google Scholar and four Iranian databases comprising SID, MagIran, Civilica, and IranDoc. After searching all the databases, 303 articles were found, from which 31 articles were included in the final analysis. Results: According to the data analysis, the highest rate of contamination was belonged to the industrial eggs (7.49%), however, the prevalence rate was reported 13.61% in the eggshell part. The overall prevalence of Salmonella contamination in consumed eggs of Iran using culture of microbial, molecular, molecular-serological, culture-molecular, cultureserological, and culture -molecular-serological methods was obtained 11.33%, 5.52%, 0.37%, 1.91%, 5.52%, and 0.73%, respectively. Prevalence in the 21 geographical areas, where studies have been conducted, ranged from 0% (Zahedan) to 29.06% (Tabriz). The studies have also showed that eight different serotypes were among the major cause of Salmonella contamination in eggs. The most common Salmonella serotype was Salmonella Enteritidis and the highest diversity in Salmonella contaminant serotypes was recorded in Talesh (including S. Enteritidis, Salmonella Gallinarum, Salmonella Virchow, and Salmonella Newport). Conclusion: Results of this study revealed the high prevalence of Salmonella contamination in eggs, in Iran. Therefore, disinfection and cleaning bed, cleaning of equipment and supplies, and proper maintenance temperature and humidity of the eggs are recommended. In addition, proper personal hygiene and prohibition of consuming raw egg products are essential.
Prevalence of Salmonella Typhimurium and Salmonella Enteritidis isolated from poultry meat: virulence and antimicrobial-resistant genes

Abstract:Salmonellosis, a zoonotic disease, is one of the leading causes of foodborne illness worldwide. It is responsible for most infections caused by consumption of contaminated food. In recent years, a significant increase in the resistance of these bacteria to common antibiotics has been observed, posing a serious threat to global public health. The aim of this study was to investigate the prevalence of virulent antibiotic-resistant Salmonella spp. strains in Iranian poultry markets. A total of 440 chicken meat samples were randomly selected from meat supply and distribution facilities in Shahrekord and tested for bacteriological contamination. After culturing and isolating the strains, identification was performed using the classical bacteriological method and PCR. To determine antibiotic resistance, a disc diffusion test was performed according to the recommendations of the French Society of Microbiology. PCR was used to detect resistance and virulence genes. Only 9% of the samples were positive for Salmonella. These were Salmonella typhimurium isolates. All Salmonella typhimurium serotypes tested positive for the rfbJ, fljB, invA and fliC genes. Resistance to TET, cotrimoxazole, NA, NIT, piperacillin/tazobactam and other antibiotics was found in 26 (72.2%), 24 (66.7%), 22 (61.1%) and 21 (58.3%) isolates, respectively. The sul1, sul2 and sul3 genes were present in 20, 12 and 4 of 24 cotrimoxazole-resistant bacteria, respectively. Chloramphenicol resistance was found in six isolates, but more isolates tested positive for the floR and cat two genes. In contrast, 2 (33%) of the cat three genes, 3 (50%) of the cmlA genes and 2 (34%) of the cmlB genes were all positive. The results of this investigation showed that Salmonella typhimurium is the most common serotype of the bacterium. This means that most of the antibiotics commonly used in the livestock and poultry industries are ineffective against most Salmonella isolates, which is important for public health.

Plasmid-mediated quinolone resistance in Escherichia coli isolates from commercial broiler chickens in Semnan, Iran

Abstract:Background and objectives: Antibiotic resistance within the poultry sector presents a considerable health concern due to treatment inefficacy and resistance transmission to humans and the environment. The investigation of plasmid-mediated quinolone resistance (PMQR) in Escherichia coli, acknowledged for its role in advancing resistance, remains inadequately studied in Iranian poultry. This study aimed to evaluate PMQR gene prevalence as well as to determine correlation between resistance phenotype and genotype in E. coli obtained from poultry colibacillosis. Materials and methods: A collection of 100 E. coli isolates from the viscera of broilers suspected to colibacillosis was assessed. Using the Kirby-Bauer disk diffusion method, antimicrobial susceptibility tests were conducted for ofloxacin, nalidixic acid, levofloxacin, ciprofloxacin, and ampicillin. Additionally, PCR was employed to screen for qnrS, qnrB, and aac(6)Ib-cr genes. Results: Among the analyzed E. coli isolates, 51% demonstrated resistance to at least one of the tested antibiotics, with 17% exhibiting resistance to four different antibiotics. Nalidixic acid displayed the highest resistance rate at 48%, while ampicillin had the lowest at 16%. PMQR genes were detected in 28% of the E. coli isolates, with aac(6')-Ib-cr being the most prevalent at 14%, followed by qnrB in 13%, and qnrS in 7%. Conclusion: The study underscores the vital need for careful antibiotic usage in poultry to curb the emergence of antibiotic-resistant bacteria. The results illuminate the prevalence of PMQR genes and their association with resistance trends in Iranian poultry, forming a pivotal basis for forthcoming approaches to combat antibiotic resistance within the poultry sector.

Abstract:Homesickness is the distress or impairment caused by an actual or anticipated separation from home. It is characterized by preoccupying thoughts of home and attachment objects. In its severe form, homesickness is subjectively distressing and has clinically significant cognitive, emotional, and behavioral sequelae. This study provided 75 boys who ranged in age from 8 to 16 years and who were 1st-year campers at an overnight summer camp with an inexpensive, multimodal homesickness prevention package. Results suggest that combining environmental information, psychoeducation, social support, explicit coping instruction, caregiver education, practice time away from home, and surrogate caregiver training can reduce homesickness and associated behavior problems. These data support the theory that novelty reduction, attitudinal shifts, preseparation coping enhancement, and supportive social environments diminish the negative emotional intensity of homesickness.
The Antibiotic Resistance Pattern and Prevalence of blaTEM, blaSHV, blaCTX-M, blaPSE-1, sipB/C, and cmlA/tetR Genes in Salmonella typhimurium Isolated from Children with Diarrhea in Tabriz, Iran

Abstract:Background: Salmonella gastroenteritis is a global health concern. Recently, increased resistance to Salmonella typhimurium has been reported in several countries. Objectives: The present study aimed to evaluate the prevalence of blaTEM, blaSHV, blaCTX-M, blaPSE-1, sipB/C, and cmlA/tetR genes in S. typhimurium isolates and determine their antibiotic resistance. Methods: This cross-sectional descriptive study was conducted on 110 fecal samples, which were collected from the patients referred to the hospitals and medical centers in Tabriz, Iran during eight months. After phenotypic identification, the antibiogram test and double-disc synergy test were performed on the isolates. Following that, the prevalence of resistance genes was evaluated using multiplex PCR and specific primers. Results: Out of 110 fecal samples, 26 samples (23.63%) were positive for S. typhimurium. The highest resistance of the isolates was against ceftazidime, cefotaxime, amikacin, and tetracycline (100%), and the lowest resistance was against imipenem (3.85%) and nalidixic acid (7.69%). In total, 15 S. typhimurium isolates (57.69%) were positive for the extended-spectrum beta-lactamases. In addition, the most common resistance genes in the isolates were cmlA/tetR (38.46%), blaTEM (34.61%), and blaCTX-M (26.92%). Four isolates (15.38%) carried sipB, three isolates (11.53%) contained blaSHV, and two isolates (7.69%) carried blaPSE-1. Conclusions: The obtained results indicated the high prevalence of antibiotic-resistant S. typhimurium. Therefore, the identification of resistance genes is an important strategy for identifying and counteracting antibiotic resistance.
Occurrence, antimicrobial resistance, and molecular characterization of Salmonella enterica from chicken products and human in Wasit Governorate of Iraq

Abstract:Background: Salmonella infections are considered the most common foodborne pathogens responsible for zoonotic infections and food poisoning in humans and animal species such as birds. Antimicrobial resistance is considered a global anxiety because it causes human public health repercussions, as well as leads to an increase in animal morbidity and death. Aim: The aims of this study are the isolation and identification of Salmonella enterica, as well as to investigate the antimicrobial susceptibility test (AST) and the molecular characteristics using polymerase chain reaction (PCR) and sequences for isolates from chicken products (eggs, livers, and minced meat) and human in the Wasit Governorate of Iraq. Methods: A total of 300 samples (150 chicken product samples including eggs, livers, and minced meat, and 150 human fecal samples) were collected from the Wasit governorate of Iraq from January to December 2022. The bacterial isolation was done according to recommendations of ISO 6579 standard and the Global Foodborne Infections Network laboratory protocol. Serotyping test and AST were done by using 19 antibiotic agents according to the recommendations of the Clinical and Laboratory Standards Institute, 2022 by using disc diffusion susceptibility test and Vitik 2 test. Finally, the suspected isolates were confirmed using the conventional PCR method and sequencing for a unique rRNA gene. Results: The results showed that the isolation percentage of S. enterica in chicken products was 8.66% (12% eggs, 6% livers, and 8% minced meat), while in humans it was 4.6%. Also, showed 100% of Salmonella typhi in humans. While, in chicken eggs S. typhi, Salmonella typhimurium, and Salmonella enteritidis were 50%, 33.33%, and 16.66%, respectively. Also, showed 100% of S. typhimurium in both livers and minced meat. The AST in human isolates showed resistance to Ampicillin, Cefotaxime, Ceftazidime, Cefepime, Amikacin, Gentamicin, Ciprofloxacin, Norfloxacin, and Ceftriaxone, while no resistance to Amoxicillin, Pipracillin, Ertapenem, Imipenem, Meropenem, Fosfomycin, Nitrofurantoin, Trimethoprim, Azithromycin, and Tetracycline. In chicken products, isolates were resistant with different percentages to Amikacin, Gentamicin, Tetracycline, Ciprofloxacin, Norfloxacin, Nitrofurantoin, Ampicillin, Cefotaxime, Ceftazidime, Cefepime, and Trimethoprim; while no resistance to Amoxicillin, Pipracillin, Ertapenem, Imipenem, Meropenem, Fosfomycin, Azithromycin, and Ceftriaxone. Sequencing by using rRNA gene was done for four PCR products. Conclusion: This study showed the presence of genetic mutations for S. enterica which led to variations in the molecular characteristics, and antimicrobial drug resistance of S. enterica isolated from chicken products and humans.
Risk factors associated with Salmonella prevalence, its antibiotic resistance, and egg antibiotic residues in the layer farming environment

Abstract:Background and Aim: Human salmonellosis with non-typhoidal Salmonella remains a global public health concern related to the consumption of contaminated eggs and egg-based products. This study aimed to examine the prevalence of Salmonella, antimicrobial-resistant Salmonella, and egg antibiotic residues concerning risk factors associated with Salmonella contamination in eggs, the layer farming environment, and laying hens kept in battery-cage closed-housing systems. Materials and Methods: This study used a repeated cross-sectional design to collect 488 samples from eggs, laying hens, and the farm environment on one laying farm for Salmonella detection according to ISO 6579:2002/AMD 1:2007. Salmonella-positive samples were further tested for serotype and antimicrobial susceptibility using the disk diffusion test. The layer farm contact person was interviewed at the sampling time to evaluate the risk factors associated with Salmonella contamination using logistic regression analysis. For each month, 24 eggs (144 eggs in total) were also randomly sampled from the collection egg area at the farm for antibiotic residue detection using the European Four Plate Test. Results: The highest Salmonella prevalence rates were in the samples from the layer pen floors, followed by the egg sizing machine (ESM) and eggshells at 65.5%, 52.5%, and 15%, respectively. Salmonella enterica serovar Corvallis was the dominant serovar (48.38%), followed by Mbandaka (37.76%), Braenderup (14.29%), and Typhimurium (4.08%). Rodent presence at the farm and the frequency of changing the disinfectant foot dip were significant factors related to Salmonella contamination on the pen floors (odds ratio [OR]=22.5, 95% confidence interval [CI]=2.11-240.48, p=0.01; OR=24, 95% CI=2.78-206.96, p=0.004, respectively). Hand-washing before sorting and cleaning the ESM were the significant factors (OR=13, 95% CI=1.2-140.73, p=0.04). The most resistant Salmonella isolates were resistant to oxytetracycline. One isolate of S. enterica Typhimurium was resistant to cefotaxime, enrofloxacin, and oxytetracycline. The antibiotic residues in the egg yolks were streptomycin, enrofloxacin, and tetracycline at prevalence rates of 36.11%, 11.81%, and 7.64%, respectively. Streptomycin was the most abundant residue in the albumen and yolk, followed by tetracycline. Conclusion: Salmonella prevalence in layer farming with a closed-housing system is related to effective biosecurity and hygiene issues, such as rodent control, clean farm equipment, and good worker hygiene. In addition, eggs' antibiotic residues may be related to treating antimicrobial-resistant Salmonella isolates and medicated feed with inappropriate antibiotic withdrawal time.
High prevalence of antibiotic resistance and biofilm formation in Salmonella Gallinarum

Abstract:Background and Objectives: Antibiotic resistance is an indicator of the passively acquired and circulating resistance genes. Salmonella Gallinarum significantly affects the poultry food industry. The present study is the first study of the S. Gallinarum biofilm in Iran, which is focused on the characterization of the S. Gallinarum serovars and their acquired antibiotic resistance genes circulating in poultry fields in central and northwestern Iran. Materials and Methods: Sixty isolates of S. Gallinarum serovar were collected from feces of live poultry. The bacteria were isolated using biochemical tests and confirmed by Multiplex PCR. Biofilm formation ability and the antibacterial resistance were evaluated using both phenotypic and genotypic methods. The data were analyzed using SPSS software. Results: According to Multiplex PCR for ratA, SteB, and rhs genes, all 60 S. Gallinarum serovars were Gallinarum biovars. In our study, the antibiotic resistance rate among isolated strains was as follows: Penicillin (100%), nitrofurantoin (80%), nalidixic acid (45%), cefoxitin (35%), neomycin sulfate (30%), chloramphenicol (20%), and ciprofloxacin (5%). All isolates were susceptible to imipenem, ertapenem, ceftriaxone, ceftazidime, and ceftazidime+clavulanic acid. All sixty isolates did not express the resistance genes IMP, VIM, NDM, DHA, bla , and qnrA. On the other hand, they expressed GES (85%), OXA48 qnrB (75%), Fox M (70%), SHV (60%), CITM (20%), KPC (15%), FOX (10%), MOXM (5%), and qnrS (5%). All S. Galli- narum isolates formed biofilm and expressed sdiA gene. Conclusion: Considering that the presence of this bacteria is equal to the death penalty to the herd, the distribution of resis- tance genes could be a critical alarm for pathogen monitoring programs in the region. This study showed a positive correla- tion between biofilm formation and 50% of tested resistance genes. Also, it was found that the most common circulating S. gallinarum biovars are multidrug-resistant.
Resistance profiles, virulence and antimicrobial resistance genes of XDR S. Enteritidis and S. Typhimurium

Abstract:Avian salmonellosis is concomitant with high financial crises in the poultry industry as well as food-borne illness in man. The present study is designed to investigate the emergence of Salmonella Enteritidis and Salmonella Typhimurium in diseased broilers, resistance profiles, and monitoring virulence and antibiotic resistance genes. Consequently, 450 samples (cloacal swabs, liver, and spleen) were collected from 150 diseased birds from different farms in Giza Governorate, Egypt. Subsequently, the bacteriological examination was done. Afterward, the obtained Salmonella isolates were tested for serogrouping, antibiogram, PCR monitoring of virulence ( inv A, stn, hil A, and pef A), and antimicrobial resistance genes ( bla TEM , bla CTX-M , bla NDM , erm A, sul 1, tet A, and aad A1). The total prevalence of Salmonella in the examined diseased broilers was 9.3%, and the highest prevalence was noticed in cloacal swabs. Among the recovered Salmonella isolates ( n = 35), 20 serovars were recognized as S . Enteritidis and 15 serovars were identified as S . Typhimurium. Almost 60% of the retrieved S. Enteritidis serovars were extensively drug-resistant (XDR) to seven antimicrobial classes and inherited sul 1, bla TEM , tet A , bla CTX-M , ere A, and aad A1 genes. Likewise, 25% of the recovered S. Enteritidis serovars were multidrug-resistant (MDR) to six classes and have sul 1, bla TEM , tet A , bla CTX-M , and ere A resistance genes. Also, 66.7% of the retrieved S. Typhimurium serovars were XDR to seven classes and have sul 1, bla TEM , tet A , bla CTX-M , ere A, and aad A1 genes. Succinctly, this report underlined the reemergence of XDR S . Typhimurium and S. Enteritidis in broiler chickens. Meropenem and norfloxacin exposed a hopeful antimicrobial activity toward the re-emerging XDR S . Typhimurium and S. Enteritidis in broilers. Moreover, the recurrence of these XDR Salmonella strains poses a potential public health threat.

Abstract:Non-typhoid salmonella infections are one of the leading food borne infections worldwide. Similarly, ever increasing antimicrobial resistance has become a major problem to animal as well as human health worldwide. Poultry being the single largest animal protein source in Pakistan is one of the major suspects for both these public health concerns. This study was aimed to determine the prevalence of Salmonella Enteritidis and Salmonella Typhimurium in commercial poultry flocks of Punjab and to evaluate their antimicrobial susceptibility patterns. Specimens were collected from morbid or dead birds suspected for salmonella infection on the basis of clinical signs or post-mortem lesions brought to five poultry diagnostic laboratories in Punjab between 2014 and 2017. The samples were then processed for bacterial isolation and molecular confirmation through PCR. The isolates were then subjected to antibiotic sensitivity test using disc diffusion method. The susceptibility was determined using CLSI guidelines for antimicrobial susceptibility testing. The frequencies and percentages were calculated using Epi info. A total of 28150 samples were processed, among them 1.04% (294/28150) were positive. 89.5% (263/294) of the isolates were Salmonella Enteritidis while 10.5% (31/294) were Salmonella Typhimurium. The isolates were most sensitive to Ciprofloxacin and least to doxycycline with 96% (273/294) and 56% (166/294) response rates respectively. 7.8% (23/294) of the isolates were found to be resistant to three or more antibiotics. For other drugs the sensitivity percentages were Gentamicin 86% (254/294), Enrofloxacin 82% (241/294), Amoxicillin 77% (227/294), Norfloxacin 74% (219/294), Colistin 71% (208/294) and Neomycin 67% (197/294). Salmonella isolates were found more sensitive to Ciprofloxacin, followed by Gentamicin, Enrofloxacin and Amoxicillin while the isolates were least sensitive to Doxycycline.
Nrf 2 transcriptional derepression from Keap 1 by dietary polyphenols ( Running title : Polyphenols derepress Nrf 2 inhibition by Keap 1 )

Abstract:The liver expresses batteries of cytoprotective genes that confer cellular resistance to oxidative stress and xenobiotic toxins, and protection against cancer and other stress-related diseases. These genes are mainly regulated by Nrf2, making this transcription factor a target for small molecule discovery to treat such diseases. In this report, we identified dietary polyphenolic antioxidants that not only activated these genes but also relieved Nrf2 repression by Keap1, a Cul3dependent ubiquitin ligase adaptor protein that mediates its degradation. Analysis of postprandial liver RNA revealed a marked activation of both genes by all test polyphenols compared with controls. Nrf2 inhibition by RNA interference reduced polyphenol effects on its target gene expression. Our data suggest that polyphenols may induce cellular defense genes by derepressing Nrf2 inhibition by Keap1. We posit that this ability to derepress Nrf2 and reactivate its target genes may underlie the protection conferred by polyphenols against oxidative stress-related diseases.
Molecular characterization and antibiotic resistance of Salmonella serovars isolated from broiler chicken farms

Abstract:The current study aimed to survey the prevalence, antimicrobial resistance, and virulence-associated genes of Salmonella enterica recovered from broiler chickens farms located in North coast, Egypt. A total of 200 broiler chickens’ cloacal swabs and farm environmental samples were randomly collected from four small scale broiler chicken farms at 5 weeks of production cycle. It was recorded that out of 200 samples, Salmonella species were identified in 26 (13%) samples. Salmonella strains were observed in 16% (16/100) of cloacal swabs, 10% (10/100) of environmental samples. S. Enteritidis (6%) was the most common identified serotypes followed by S. Kentucky (4%), S. Shangani (2.5%) and S. Virchow (0.5%). The antimicrobial susceptibility testing revealed absolute resistance to SXT (88.5%) followed by AMC and AMP (65.4% and 61.5%, respectively). On the other hand, lower rates of resistance were observed for DO (50%), CTX (42.3%), CEC (30.8%) and CIP (19.2%). Interestingly, only one isolate was resistant to IMP (3.8%). PCR targeting 5 virulence genes (invA, ompA, ompF, stn and pefA) were performed to detect the virulence of identified Salmonella serovars. Generally, all Salmonella isolates showed at least two virulence-associated genes. Of note, invA gene (genus specific gene) was detected in 100% (26/26) of the isolates. On the contrary, pefA gene was completely absent in all isolates. Finally, it was concluded that S. Enteritidis was the most common identified serotypes in the examined sources. Circulation of such strains in broiler farms required introducing special biosecurity measures for control of Salmonella. Such measures might limit the adverse effects of antibiotics and ensure the safety of the environment and animal-derived food.

Abstract:Paratyphoid salmonellosis is a serious disease threatens the poultry industry worldwide, besides its public health hazard. The aims of this study were characterization of paratyphoid Salmonella spp. in chicken flocks of some Egyptian governorates, demonstration of the antimicrobial susceptibility of the isolated Salmonella spp., and detection of some virulence genes and antibiotic resistance genes using recent molecular techniques. A total of 238 organ samples were collected from 52 broiler, layer, and breeder chicken flocks, representing 9 Egyptian governorates. Conventional characterization of Salmonella isolates revealed a total isolation rate of 56.3% (134/238). Moreover, the isolation rates of Salmonella spp. were (49/79; 62%), (47/81; 58%), (10/18; 55.5%), (9/20; 45%), (2/6; 33.3%), (2/3; 66.7%), and (15/82; 53.6%) from liver, yolk sac, heart, spleen, caecum, ovary, and dead-in-shell embryos, respectively. A total of 32/238 (13.44%) isolates of Salmonella were found. Serological identification revealed presence of S. enteritidis (21.9%), S. kentucky (15.6%), S. typhimurium (12.5%), S. molade (12.5%), S. takoradi (9.4%), S. wingrove (6.3%), S. infantis (6.3%), S. tsevie (6.3), S. shangani (3.1%), S. bargny (3.1%), and S. papuana (3.1%). All Salmonella strains (32/32; 100%) were resistant to streptomycin, while almost all of them (31/32; 96.9%) were susceptible to meropenem. The amplification of 16S rRNA gene of Salmonella isolates using uniplex polymerase chain reaction (PCR) generated a specific Salmonella product of approximate 550 base pair. The multiplex PCR revealed presence of invA (100%), stn (65.6%), and sopB (40.6 %) virulence-associated genes as well as aadA1 (100%), blaTEM (59.4%), aadB (18.75%), and sul1 (28.1%) antibiotic resistance genes. In conclusion, virulent paratyphoid Salmonella spp. are circulating in the Egyptian flocks, causing economic loses. Additionally, they became resistant to the most commonly used field antibiotics. Therefore, regular molecular surveillance studies on the circulating Salmonella spp. and their resistance to the used antibiotics are of significant importance.
The Prevalence of Shiga Toxin-Producing Escherichia coli and Enteropathogenic Escherichia coli Isolated from Raw Chicken Meat Samples

Abstract:Background. Shiga toxin-producing Escherichia coli (STEC) is known as a crucial zoonotic food-borne pathogen. A total of 257 raw chicken meat samples were collected from different markets in Hamadan, west of Iran, from January 2016 to May 2017. Materials and Methods. The samples were cultured in selective and differential culture media, and the virulence genes of E. coli isolates were analyzed by PCR assay. The antibiotic resistance patterns of E. coli isolates were determined by the disk diffusion method. The genetic relatedness of the E. coli O157 isolates was analyzed by ERIC-PCR. Results. In total, 93 (36% ± 3.12) of the isolates were identified as E. coli in this study. Based on serological and microbiological tests, 36 (38.7% ± 9.9), 7 (7.5% ± 5.35), and 12 (12.9% ± 6.81) of the E. coli isolates were characterized as STEC, enteropathogenic E. coli (EPEC), and attaching and effacing E. coli (AEEC) strains, respectively. A high level of resistance to nalidixic acid (91.4% ± 5.7), tetracycline (89.2% ± 6.31), ampicillin (82.8% ± 7.67), and trimotoprime-sulfametoxazole (71% ± 9.22) was detected among the E. coli isolates. The analysis of the ERIC-PCR results showed five different ERIC types among the E. coli O157 isolates. Conclusions. Based on our findings, control and check-up of poultry meats should be considered as a crucial issue for public health.
Prevalence and genomic characterization of Salmonella isolates from commercial chicken eggs retailed in traditional markets in Ghana

Abstract:Salmonella enterica are important foodborne bacterial pathogens globally associated with poultry. Exposure to Salmonella-contaminated eggs and egg-related products is a major risk for human salmonellosis. Presently, there is a huge data gap regarding the prevalence and circulating serovars of Salmonella in chicken eggs sold in Ghana. In this study, 2,304 eggs (pools of six per sample unit) collected from informal markets in Accra, Kumasi and Tamale, representing the three ecological belts across Ghana, were tested for Salmonella. Antimicrobial susceptibility testing and Whole Genome Sequencing (WGS) of the isolates were performed using standard microdilution protocols and the Illumina NextSeq platform, respectively. The total prevalence of Salmonella was 5.5% with a higher rate of contamination in eggshell (4.9%) over egg content (1.8%). The serovars identified were S. Ajiobo (n = 1), S. Chester (n = 6), S. Hader (n = 7), S. enteritidis (n = 2); and S. I 4:b:- (n = 8). WGS analysis revealed varied sequence types (STs) that were serovar specific. The S. I 4:b:- isolates had a novel ST (ST8938), suggesting a local origin. The two S. enteritidis isolates belonged to ST11 and were identified with an invasive lineage of a global epidemic clade. All isolates were susceptible to ampicillin, azithromycin, cefotaxime, ceftazidime, gentamicin, meropenem, and tigecycline. The phenotypic resistance profiles to seven antimicrobials: chloramphenicol (13%), ciprofloxacin (94%), and nalidixic acid (94%), colistin (13%), trimethoprim (50%) sulfamethoxazole (50%) and tetracycline (50%) corresponded with the presence of antimicrobial resistance (AMR) determinants including quinolones (gyrA (D87N), qnrB81), aminoglycosides (aadA1), (aph(3")-Ib aph(6)-Id), tetracyclines (tet(A)), phenicols (catA1), trimethoprim (dfrA14 and dfrA1). The S. enteritidis and S. Chester isolates were multidrug resistant (MDR). Several virulence factors were identified, notably cytolethal distending toxin (cdtB gene), rck, pef and spv that may promote host invasion and disease progression in humans. The findings from this study indicate the presence of multidrug resistant and virulent strains of Salmonella serovars in Ghanaian chicken eggs, with the potential to cause human infections. This is a critical baseline information that could be used for Salmonella risk assessment in the egg food chain to mitigate potential future outbreaks.

Abstract:Background and Aim: Food poisoning caused by Salmonella enterica serovars is the most common type of foodborne illness. Tainted chicken meat is a major vector for spreading these serovars throughout the food supply chain. Salmonella isolates that developed resistance to commonly used antimicrobials pose a noteworthy risk to public health, yet there has been a lack of data on this issue in Iraq. Therefore, it is crucial to address these serious public health challenges with an adequate database on the occurrence and antibiotic resistance of these serovars. This study aimed to determine the frequency of occurrence of Salmonella enterica serovars Enteritidis and Typhimurium (S. Enteritidis and S. Typhimurium), antimicrobial resistance (AMR), and prevalence of multidrug resistance among S. Enteritidis and S. Typhimurium isolated from poultry meat collected in Wasit Province in Iraq. Materials and Methods: A total of 150 raw and frozen poultry meat samples were gathered from retail markets in various locales across the Wasit Governorate in Iraq. Salmonella spp. were successfully cultured and identified using the technique recommended by ISO 6579:2002, with minor modifications. The multiplex polymerase chain reaction approach was used to confirm Salmonella spp. (S. Enteritidis and S. Typhimurium). A disk diffusion test was performed to determine the susceptibility to particular antimicrobial agents, and 12 different antimicrobial agents were evaluated. Results: Only 19 of the 150 (12.7%) samples tested positive for Salmonella (16% and 11% were isolated from raw and frozen chicken meat, respectively). S. Enteritidis accounted for 63.2%, whereas S. Typhimurium accounted for 36.8%. Nalidixic acid resistance was the most common (73.7%), followed by sulfamethoxazole-trimethoprim (63.2%) and tetracycline (63.2%), but gentamicin and ciprofloxacin (up to 15.8%) only had modest resistance. Antibiogram of S. Enteritidis and S. Typhimurium yield 13 antibiotypes. Among the 19 Salmonella isolates, 12 of 19 (63.2%) established resistance to no less than three categories of antimicrobials. Conclusion: This study highlighted the necessity of limiting the utilization of antibiotics in animal production by providing vital information regarding the frequency and AMR of Salmonella at markets in Wasit Province. Therefore, risk assessment models could use these data to lessen the amount of Salmonella passed on to humans in Iraq from chicken meat. The Prevalence of Infection with Gastrointestinal Helminths in Free-Range Poultry of Zabol, Iran

Abstract:Background: As an ecological and ethical method employed for poultry farming, free-range chicken keeping is growing and is important in fostering rural farmers' livelihoods and community prosperity. This study aimed to determine the nature and prevalence of helminth infections in native poultry in Zabol, Iran. Methods: Between 2022 and 2023, 160 chickens were acquired and slaughtered ethically, and then their gastrointestinal tracts were taken and formalin preserved in Zabol, Iran. The parasites were isolated from gastrointestinal mucus in the laboratory and characterized using a microscope and specific methods such as Acetocarmine staining. Results: Among 160 dissected birds, 92 (57.5%) were presented with gastrointestinal parasites, with 64 (40%) containing cestodes, 16 (10%) containing nematodes, and 12 (7.5%) infected by both. Observations were made of seven different species. Ascaridia galli (A. galli) (10%), Subulura brumpti (7.5%), and Heterakis gallinarum (3.12%) represent the three main nematode species. While Raillietina tetragona (R. tetragona) (33.12%) was the most prevalent cestode, R. echinobothrida (6.25%), R. magninumida (5%), and Cotugnia digonopora (3.12%) were less common. Conclusion: The study revealed that free-range chickens in the arid Zabol region had a high prevalence of gastrointestinal parasites. There is a strong correlation between chicken foraging behavior and exposure to contaminated soil, with certain nematodes and cestodes constituting the most prevalent, negatively affecting bird growth, productivity, and health. Therefore, improving poultry welfare by applying parasite control measures and implementing appropriate management strategies is essential.
