Abstract:Fruit of jujube (Ziziphus jujube Mill. cv. Dongzao), known as a high-quality fresh edible variety, is popular among consumers on account of its bright color, crispy texture and abundant nutrients. However, the softening of jujube fruit after harvest seriously shortens its shelf life. To examine possible treatments to reduce softening rate, we evaluated the effect on fruit quality attributes of an elevated atmospheric O2 concentration during cold storage, ultimately unraveling its biological effect on jujube cell wall metabolism and antioxidant level. Our results demonstrated that an elevated O2 atmosphere during cold storage induced both the activity of antioxidant enzymes and relative expression levels of related genes, promoting the scavenging of hydrogen peroxide and superoxide anion. By the end of storage, the activities of superoxide dismutase, peroxidase, and catalase in elevated O2-treated jujubes were 1.9, 1.3, and 1.1 times, respectively, those in the control group. In addition, elevated O2 concentration inhibited the activities of polygalacturonase, pectin methyl esterase, cellulase and the relative abundance of related transcripts, thus delaying the degradation of cellulose and hemicelluloses and reducing the conversion of insoluble pectin to water-soluble pectin and ionic-soluble pectin. These changes presumably contribute to the maintenance of the cell wall components and cell microstructure. In conclusion, an elevated atmospheric O2 concentration of 60 % during cold storage promoted antioxidant capacity and inhibited cell wall metabolism in postharvest jujube fruit, reducing postharvest softening.

Abstract:This study investigated the effects of convective drying(CD) and freeze drying(FD) on bioactive and volatile components in jujube. No significant difference in total phenolic, total flavonoids and antioxidant capacity among CD60, CD70, CD80 and FD samples (<i>P</i> > 0.05). LC-MS/MS analysis showed that this trend mainly originated from the dynamic equilibrium relationships between caffeic acid, chlorogenic acid, <i>p</i>-hydroxybenzoic acid, rutin, epicatechin, and quercetin. HS-SPME-GC-MS identified 31 volatile organic compounds (VOCs) comprising more than 80% aldehydes and acids. Principal component analysis distinguished the VOC characteristics of samples subjected to different drying methods. Six VOCs had an odor activity value (OAV) >1, most of which were fatty acid oxidation or Maillard reaction products. Combined with the precursor components, these reactions were speculated to be the major VOC-producing pathways in dried jujube. Considering the bioactive components and flavor retention, CD at 60 °C was an effective drying method with potential to replace FD.

Abstract:Jujube has various functional properties and is a promising source of bioactive compounds and flavors. This study investigated the functional properties and flavor characteristics of milk from cows supplemented with jujube powder (JP). Here, milk volatile profiles and taste properties were analyzed by using an electronic nose, and headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS). Compared with the control group, the total antioxidant capacity, 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic free radical scavenging activity, lactoferrin, and IgG levels increased significantly in the JP group. Volatile flavor analysis indicated that ketone levels increased, while acid abundance decreased, and toluene and dimethyl sulfone significantly increased in the JP group. Taste profile analyses demonstrated that jujube supplementation altered the taste of the milk. In summary, dietary jujube powder supplementation affects the volatile flavor composition and aroma of milk, as well as the bioactive component and antioxidant properties. These findings enhance our understanding of milk production using direct dietary supplementation to produce sustainable dairy products.

Abstract:To investigate the effects of exogenous melatonin (MLT) treatment on the quality of postharvest jujubes, fresh 'Lingwuchangzao', and 'Dongzao' jujubes (Zizyphus jujuba Mill) were dipped in MLT solution at the dose of 0, 50,100 and 200 mu mol L-1 for 20 min. Results showed the exogenous MLT application significantly delayed the color change and firmness decline, and maintained the content of total soluble solids and titratable acidity of both jujube cultivars, (p < 0.05). It was demonstrated that the endogenous MLT content was increased by exogenous MLT treatment. Furthermore, phenolic compounds level was enhanced by MLT application, companied by the upregulated expression of main genes involved in phenolic biosynthesis, including phenylalanine ammonia-lyase, cinnamate 4-hydroxylase, chalcone synthase, flavonoid 3 beta-hydroxylase, leucoanthocyanidin reductase, and anthocyanidin synthase. Given the evidence from the present study, it's proposed that the exogenous MLT approach is a promising approach for maintaining quality attributes and delaying the senescence of postharvest jujubes.

Abstract:BACKGROUNDThe taste of dried jujube fruit when compared with fresh ones is less palatable, as it develops bitterness during drying and storage. Therefore, identifying the methods by which bitterness occurs is essential for developing strategies for processing and storage. RESULTSBitterness in fresh jujube fruit was negligible; however, it increased by 0.9-, 1.5- and 1.8-fold during drying and storage over 6 and 12months. The moisture significantly decreased during harvesting and drying. Free amino acids, except proline and tyrosine, significantly decreased during drying and storage. Fructose, glucose and sucrose hardly changed during harvest, drying and storage. Titratable acidity, total phenolic and total flavonoids contents were stable during harvest and drying, but increased upon storage. Additionally, protocatechuic and ellagic acids were not detected in fresh jujube fruit, however, were found to increase during drying and storage. CONCLUSIONBitterness in fresh jujube fruit tasted negligible because of meagre amount of phytochemicals, while the condensation effect of moisture reduction, the loss of free amino acids, and the formation of protocatechuic and ellagic acids could aggravate the bitterness of jujube fruit during drying and storage. Abstract:To understand the evolution of aroma in jujubes during dynamic freeze drying (FD), the relationship between aroma compounds, precursors, and related enzyme activities were analyzed. Fifty-three volatiles were identified during FD processing. After FD, the total aroma contents were increased from 11,004 to 14,603 μg/kg, ketones content was significantly decreased by 54.11 %, resulted in the loss of creamy note in freeze-dried jujube (FDJ). Through the network analysis, serine, glycine, proline, valine, cysteine, arginine, glutamic acid, lysine and leucine had the significant correlation with pyrazines, dominated the roasty note of FDJ. Linoleic acid, α-linolenic acid and oleic acid with lipoxygenase had important effects on the increase of esters (from 412 to 9,486 μg/kg), contributed fruity and sweet notes of FDJ. Besides, through the Mantel test, the influence degree of factors on the formation of FDJ aroma was ranked as temperature > enzyme activity > fatty acids > amino acids.

Abstract:Dried jujube fruit has been wildly used as flavoring in food preparation and food industry because of its unique and exquisite flavor. However, there is limited information available on the formation and alteration of jujube flavor during drying and storage. In this study, the effects of drying and storage on the volatiles in jujube fruit were investigated by electronic nose and GC-MS. Electronic nose and GC-MS could distinguish different jujube samples clearly; the ester and aldehydes showed significant decreases during drying, while furans and pyrazines were generated and increased with heat time; the esters, aldehydes, furans and pyrazines showed significant decreases, and some of them even disappeared during storage, while acids and terpenes increased with the extension of storage. Based on the present results, furans and pyrazines, generated during drying, are the predominant components of the unique and exquisite flavor of dried jujube, and these compounds could be negatively influenced because of evaporation, oxidation and degradation of jujube volatiles during storage.

Abstract:Previous studies demonstrated jujube blackening effectively increased cyclic adenosine phosphate and triterpene acid levels, improving its nutritional value. However, compositional changes during this process require further elucidation. The objective aimed to analyze compositional transformations during this process with SEM, TPA, UPLC-MS, E-nose. Results showed decreased hardness, springiness, and chewiness coupled with increased gumminess over blackening durations. Untargeted omics analysis revealed increases of 2-aminooctadec-8-ene-1,3,4-trioland carbendazim. Targeted organic acid analysis showed initial citric acid accumulation (1481.62 to 1645.78 mg/kg) in the first 24 h, then declines to 1072.96 mg/kg. Meanwhile, oxalic and lactic acids steadily rose, peaking at 96-120 h before slightly decreasing. E-nose analysis implied alterations in organic sulfide aromatics engendered the characteristic flavors. Organic acid fluctuations likely resulted from sugar biotransformation and thermal degradation. These comprehensive analyses demonstrate jujube blackening imparts a rich and unique flavor, providing theoretical support for investigating the mechanisms and products underlying this process.

Abstract:Color deterioration is one of the major symptoms of postharvest senescence in winter jujube fruit. Elevated O2 (60 %) was applied to winter jujube fruit during storage at 0 degrees C and 90 % relative humidity. The changes in three major skin pigments, chlorophyll, carotenoid, and anthocyanin, were quantified to explain changes in skin color. The results showed that elevated O2 maintained the contents of chlorophyll and carotenoid during the initial stages of storage, which were 25% higher chlorophyll a, 34% higher chlorophyll b, and 18 % higher carotenoid compared to the control on day 7. The maintenance of chlorophyll content under elevated O2 was due to the inhibition of chlorophyll catabolism. Additionally, elevated O2 treatment also maintained carotenoid content by activating its biosynthesis rather than inhibiting its degradation. During the later stages of storage, the accumulation of anthocyanin was enhanced by elevated O2, with a 1.3-fold increase in anthocyanin observed in elevated O2-treated fruit compared to the control fruit on day 21. Elevated O2 treatment effectively activated phenylpropanoid biosynthesis and flavonoid biosynthesis pathways, leading to the accumulation of anthocyanin. This study elucidated the mechanism by which elevated O2 regulated color deterioration in winter jujube fruit.

Abstract:Current research does not fully elucidate the key compounds and their mechanisms that define the aroma profile of fresh jujube fruits. Therefore, this study conducted a comprehensive analysis of both free and glycosidically bound aroma compounds in fresh jujube fruits of ten cultivars. Utilizing gas chromatography–mass spectrometry (GC-MS), we identified 76 volatile free aroma compounds and 19 glycosidically bound volatile compounds, with esters, aldehydes, and ketones emerging as the predominant volatile compounds in the jujube fruits. Odor activity value (OAV) analysis revealed that the primary aroma profile of the jujubes is characterized by fruity and fatty odors, with β-damascenone being a key contributor to the fruity aroma, and (E)-2-oct-en-1-al and nonanal significantly influencing the fatty aroma. Moreover, the integration of sensory evaluation and partial least squares regression (PLSR) analysis pinpointed octanal, (E)-2-oct-en-1-al, nonanal, β-damascenone, and pentanal as significant contributors to the jujube's characteristic aroma, while isoamyl acetate was identified as significantly influencing the fatty acid taste. This study not only underscores the complexity of the jujube aroma composition but also highlights the impact of environmental factors on aroma profiles, offering valuable insights into the sensory characteristics of jujube fruits.

Abstract:Jujube is a plant of the genus Ziziphus in the family Rhamnaceae; its fruit has high nutritional value, and it is rich in polyphenols, flavonoids, and other secondary metabolites. The color of its peel is an important indicator for evaluating the appearance of the fruit. However, the mechanism of the difference in color presentation between the seedling offspring of the 'Red Fruit' (TLHH) and the 'Green Fruit' (TLHL) of the fresh jujube cultivar 'Tailihong' is not clear. Therefore, this study used targeted metabolomics techniques to accurately and quantitatively analyze the metabolic pathways of carotenoid and anthocyanin metabolites during the ripening process of two color-presenting types of jujube fruits. Through the analysis of the dynamic changes in the pigment content of the jujube peel, it was found that 30 DAP (days after pollination), 80 DAP, and 110 DAP were the key periods for the development of the color of the peel of 'TLHL' and 'TLHH' jujube and that the substances responsible for the main differences were chlorophyll, carotenoids, and anthocyanins. Furthermore, we used an LC-MS/MS metabolic analysis to compare the differences in the carotenoids and anthocyanin metabolites between the two color-presenting types of jujube peels at the key periods of 30 DAP, 80 DAP, and 110 DAP. We detected 32 carotene metabolites and 75 anthocyanin metabolites, respectively, among which lutein had the highest content of carotenoids; it reached the maximum value (93.05 µg/g) and was higher than that of 'TLHH' (74.14 µg/g) at 30 DAP of 'TLHL'. Both showed a decreasing trend with fruit ripening. The anthocyanin with the highest content was cyanidin-3-O-(tartaryl)rhamnoside-5-O-glucoside, which reached the maximum value (258.32 µg/g) at 30 DAP of 'TLHH' and was 51.6 times that of 'TLHL'; similarly, both showed a decreasing trend with fruit ripening. These results elucidate the main metabolites of carotenoids and anthocyanins in the two types of jujube peel and their accumulation characteristics, suggesting that the key metabolites of the difference in color between 'TLHL' and 'TLHH' jujube fruits were lutein and cyanidin-3-O-(tartaryl)rhamnoside-5-O-glucoside, increasing the understanding of the color mechanism of jujube peel and providing a reference for targeted genetic breeding of jujube peel color.

Abstract:The purpose of this research is to investigate the kinetics of nutrient quality (Vitamin C (Vc), reducing sugar and total acidity) change of winter jujube slices that under different drying temperatures (55, 60, 65 and 70?) and different air velocities (3, 6 and 9m/s) during the air-impingement drying process. Results showed that the content of Vc, reducing sugar and total acidity decreased with increasing drying time. Furthermore, analysis of variances indicated that the drying temperature, air velocity and time had a significant effect on the loss of Vc, reducing sugar and total acidity (p<0.05). Zero order, first order and Weibull models were used to fit the experimental data, Weibull model was considered as the most suitable one to the degradation kinetics of Vc, reducing sugar and total acidity in dried samples at different temperatures and air velocities. According to the Arrhenius formula, the activation energy of Vc, reducing sugar and total acidity degradation kinetics were 63.78 kJ/mol, 36.48 kJ/mol and 153.51 kJ/mol, respectively. This research can provide some references for enhancing dried products quality in the jujube drying industry.

Abstract:The quality attributes of jujube fruit can be directly and indirectly affected by abiotic stresses associated with climate change. Increased temperature and drought are among the most important factors challenging sustainable jujube production in the temperate semi-arid region in northwest China. The main objective of the present study was to understand the effects of elevated air temperature and drought stress on sugar and acid accumulation and coloration of jujube fruits. The content of soluble sugar, organic acid and pigments of traditional jujube "Linwuchangzao" under different atmospheric temperatures and drought stresses were analyzed during three different fruit ripening stages. The elevated temperature (1.5-2.5° C than normal temperature) significantly increased the fruit sugar content, sugar-acid ratio, anthocyanins, flavonoids and carotenoids content. Under the drought stress where the soil moisture was 30% -50% of the field capacity, sugar content, anthocyanin, flavonoid and carotenoid content of the fruit were significantly reduced at the same temperature, but the chlorophyll and organic acid content increased. No significant interaction of Temperature x Drought was observed for all the analyzed quality parameters. The current results showed that the fruit quality of jujube variety "Lingwuchangzao" could be improved when the atmospheric temperature increases by 2° C in this region. However, drought stress had a negative impact on the fruit's sugar-acid ratio and pigment content. The present results also showed that the synthesis and accumulation of anthocyanins in jujube fruit were positively correlated with sugar content and related enzyme activities, especially Phenylalanine Ammonia-lyase (PAL) activity. This study, therefore, provides novel information for understanding the influence of growth environment on the quality properties of jujube fruits. This knowledge will help develop appropriate crop management practices for jujube production in arid and semi-arid areas in northwest China.

Abstract:Jujubes have been favored by consumers because of their rich nutrition and wide use. Hot air drying has been commercially and typically used to prolong shelf life and acquire the dried produce. Jujube slices were dried with hot air combined with radio frequency (RF) at different drying stages, namely, early (0–2 h, E-HA + RF), middle (2–4 h, M-HA + RF), later (4–6 h, L-HA + RF), and whole (0–6 h, W-HA + RF) stages. This study aimed to investigate the effects of different RF application stages on the microstructure, moisture absorption rate, color, and ascorbic acid of jujube slices. Compared with the hot air drying (HA) group, the E-HA + RF group obtained the best results among the experimental groups because it reduced the cells with a roundness of less than 0.4 by 5%. Moreover, the M-HA + RF group showed better results than those of other groups, with an 18.6% and 48.8% reduction in cells for a cross-sectional area less than 200 μm2 and a perimeter less than 25 μm, respectively. The minimum total color difference (ΔE = 9.21 ± 0.31) and maximum retention of ascorbic acid (285.06 mg/100 g) were also observed in this group. Therefore, the method of hot air drying assisted by phased RF is viable in the drying industry to improve the quality of dried agricultural products and reduce energy consumption.

Abstract:One of the most important qualities of jujube fruit is its color. Chlorophyll, carotenoid, and anthocyanin all play important roles in the coloring of jujube fruit. However, few studies have focused on the pigment molecular mechanism. In the present study, jujube peels of 'Sanbianhong' in three growth stages were evaluated for their gene expression characteristics and gene regulation related to pigment formation using the transcriptome sequencing analysis. A total of 84.86 Gb of clean data were obtained in the analysis. In the FS1 vs. FS3, FS1 vs. FS5, and FS3 vs. FS5, 4,530, 11,012, and 9,072 differentially expressed genes (DEGs) were identified, respectively. The inter-group screening among the three comparisons yielded 1430 common DEGs. Among these DEGs, 27, 16, and 28 genes were enriched in chlorophyll, carotenoid, and anthocyanin metabolic pathways, respectively. Twelve genes were chosen at random, and the accuracy of the transcriptome data were confirmed using qRT-PCR. The molecular mechanism underlying the pigmentation of jujube fruit was elucidated at the transcriptome level, which would provide a scientific basis for the subsequent functional studies on the color-regulating genes of jujube fruits.

Physicochemical Characteristics of 'Wushan Plum' Subjected to Three Drying Methods

Abstract:In this study, the effects of hot air drying (HAD), freeze‐drying (FD), and air‐impingement jet drying (JD) on the physicochemical characteristics of 'Wushan plum' were analyzed. The results indicated three drying methods affected the browning, ascorbic acids, phenolic compounds, and antioxidant activities. The ascorbic acid content in HAD, FD, and JD samples was1.75±0.38, 5.88±0.56, and 4.73±0.75 mg/100 g, respectively, and the content of total phenolic compounds was 2.76±0.10, 3.05±0.04, and 2.83±0.24 mg/g, respectively. HAD samples had the lowest antioxidant activities as determined by DPPH·, ABTS+·, and FRAP assays. In addition, 11 phenolic compounds in 'Wushan plum' were quantified by UPLC‐QqQ‐MS/MS. The results showed that quercetin glycoside was the main compound, and its content was significantly reduced by these three drying methods. In all, this result indicated that JD would be a good choice for dying 'Wushan plum' considering the drying time, drying efficiency, and the integrated quality of the dried sample.

Abstract:Aroma profile and key aroma-active of red jujube subjected to pilot scale freeze drying (FD) were investigated based on molecular sensory science. After FD, 41 aroma compounds were identified through gas chromatography-olfactometry-mass spectrometry (GC-O-MS) and quantified through calibration curves. The total aroma compounds content was decreased 26.71% compared with raw red jujube, of which ketones and acids contents were decreased 63.33% and 62.88%, while, the esters, lactones and alcohols contents were increased 34.10%, 8.52% and 480.17%, respectively. Through the GC-O-MS, odor active values combined with recombination and omission tests, 14 key aroma-active compounds were identification in freeze-dried red jujube. In which, 2-ethyl-3,5-dimethyl-pyrazine had the highest OAV (2687.00) and dominated the roast note of aroma profile. In addition, ethyl heptanoate, hexyl acetate, 2,3-butanediol, ethyl dodecanoate and 5-heptyloxolan-2-one were newly identified as the key aroma-active compounds in freeze-dried red jujube.

Abstract:The fruit flavor is a key economic value attribute of jujube. Here we compared metabolomes and transcriptomes of "Mazao" (ST) and "Ping'anhuluzao" (HK) with unique flavors during fruit development. We identified 437 differential metabolites, mainly sugars, acids, and lipids. Fructose, glucose, mannose and citric acid, and malic acid are the determinants of sugar and acid taste of jujube fruit. Based on the transcriptome, 16,245 differentially expressed genes (DEGs) were identified, which were involved in "glucosyltransferase activity," "lipid binding," and "anion transmembrane transporter activity" processes. Both transcriptome and metabolome showed that developmental stages 2 and 3 were important transition periods for jujube maturation. Based on WGCNA and gene-metabolite correlation analysis, modules, and transcription factors ( ZjHAP3 , ZjTCP14 , and ZjMYB78 ) highly related to sugar and acid were identified. Our results provide new insights into the mechanism of sugar and acid accumulation in jujube fruit and provide clues for the development of jujube with a unique flavor.

Abstract:Maillard reaction browning is one of the quality deterioration in dried fruit products, but how pectin affects Maillard reaction in the fruit drying and storage process is not clear. This study aimed at investigating the mechanism of pectin variation impact on the browning of Maillard reaction by using simulated system ( l -lysine, d -fructose and pectin) in thermal (60 °C and 90 °C for 8 h) and storage (37 °C for 14 days) process. Results showed that apple pectin (AP) and sugar beet pectin (SP) significantly enhanced the browning index (BI) of the Maillard reaction system by 0.01 to 134.51 in the thermal and storage processes, respectively, which were methylation degree of pectin-dependent. The pectin depolymerization product participated Maillard reaction by reacting with l -lysine, and increasing the 5-hydroxymethyl furfural (5-HMF) content (1.25–11.41-fold) and Abs 420nm (0.01–0.09). It also produced a new product ( m / z 225.1245), which finally increased browning level of the system.
