Abstract:This study investigated the occurrence of hexabromocyclododecanes (HBCDs) in soil-maize system around Baiyangdian Lake. The total concentration of ΣHBCDs was in the order of industrial area > residential area > marginal area > Fuhe River estuary in soil. γ-HBCD was predominated in soils, roots and stems, while α-HBCD was the main diastereoisomer in leaves and kernels. Concentration of ΣHBCDs and three diastereoisomer concentrations in soils were significantly reduced and remained low level from 2018 to 2019. Selectivity enrichment of (+)α- and (-)γ-HBCD was found in soils, roots, stems and leaves, whereas only (+)β-HBCD dominated in stems. Most of the total root bioaccumulation factors (ΣRCFs) were less than 1.0, but no significant correlation was showed between translocation factors (TFs) and log K ow . RCFs and TFs of enantiomers suggested (-)β- and (-)γ-HBCD were easily translocated from soil to roots, while (+)α-, (-)β- and (-)γ-HBCD tended to translocate from stems to leaves. Estimated daily intake (EDI) and of ΣHBCDs, diastereoisomers and enantiomers were all lower than the threshold value, while the Calculated margins of exposure (MOE) were well above the threshold value, which demenstrited the safe consumption of Maize around Baiyangdian Lake.

Abstract:The phytotoxicities of TBECH diastereomers to plants at the biochemical and molecular levels were investigated in a hydroponic study by using maize as a model plant. The results showed that TBECH could induce the production of two species of reactive oxygen species (ROS), O2•− and H2O2, in maize tissues. The accumulation of ROS was the highest when maize was exposed to β-TBECH. TBECH enhanced the phosphorylation of plant histone, and the contents of γ-H2AX in maize followed the order β-TBECH > αβ-TBECH > γδ-TBECH > γ-TBECH. Transcriptome profiling revealed that antioxidant enzyme genes (AEGs) were over-expressed in maize when stressed by technical grade TBECH. The RT-PCR detection further validated that three typical AEGs, including CAT, SOD, and POD genes, were time-dependent and selectively expressed under the influence of TBECH diastereomers. Molecular compositions of maize root exudates characterized by FT-ICR-MS were significantly different among the four groups of TBECH diastereomer treatments. TBECH diastereomers specifically affected the chemical diversity and abundance of root exudates. New insights into the biochemical effects of TBECH on plants are provided in this work, which is helpful to deepening the understanding of their stereo-selectivity.

Abstract:The bioaccumulation and biotransformation of tetrabromoethylcyclohexane (TBECH) in maize were investigated. Furthermore, the roles of plant biomacromolecules such as lipid transfer proteins (LTPs), CYP and GST enzymes in driving the biological processes of TBECH stereoisomers were explored. The uptake and translocation of TBECH in maize were diastereo- and enantio-selective. Isomerization from α- to δ-TBECH and β- to γ-TBECH, and metabolites of debromination, hydroxylation and TBECH-GSH adducts were identified in maize roots. The gene expressions of LTPs, CYPs and GSTs were extensively changed in maize after exposure to technical TBECH. CYP and GST enzyme activities as well as GST31 and CYP71C3v2 gene expressions were selectively induced or inhibited by TBECH diastereomers over time. TBECH was able to dock into the active sites and bind with specific residues of the typical biomacromolecules ZmLTP1.6, GST31 and CYP71C3v2, indicating their roles in the bioaccumulation and metabolization of TBECH. Binding modes and affinities to biomacromolecules were significantly different between α- and β-TBECH, which contributed to their stereo-selectivity. This study provided a deep understanding of the biological fate of TBECH, and revealed the driving molecular mechanisms of the selectivity of TBECH stereoisomers in plants.

Abstract:Hexabromocyclododecanes (HBCDs) are additive brominated flame retardants mainly used in plastics and textiles. At the present time, these compounds are found in almost all environmental and human samples. In order to evaluate the environmental safety and health risk of HBCDs, the enantiomerically pure alpha-, beta-, and gamma-HBCD were prepared using high performance liquid chromatography (HPLC) on a PM-beta-CD column and the cytotoxicities of their enantiomers were evaluated in Hep G2 cells. Results from the 3-(4,5-dimethylthioazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), resazurin reduction and lactate dehydrogenase (LDH) release assays showed a good agreement that the order of cytotoxicity was gamma-HBCDbeta-HBCD>alpha-HBCD, and that significantly lower cell viability and higher LDH release were observed in all (+)-enantiomers ((+) alpha-, (+) beta- and (+) gamma-HBCD) than the corresponding (-)-forms ((-) alpha-, (-) beta- and (-) gamma-HBCD). Additionally, the formation of reactive oxygen species (ROS) induced by these HBCD enantiomers were detected. The positive correlation between the LDH release and ROS formation demonstrated that the toxic mechanism might be mediated by oxidative damage. These results suggest that environmental and human health risks of HBCDs must be evaluated at the level of individual enantiomers.
Antioxidant gene expression and metabolic responses of earthworms (Eisenia fetida) after exposure to various concentrations of hexabromocyclododecane

Abstract:Hexabromocyclododecane (HBCD), a ubiquitous suspected contaminant, is one of the world's most prominent brominated flame retardants (BFRs). In the present study, earthworms (Eisenia fetida) were exposed to HBCD. The expression of selected antioxidant enzyme genes was measured, and the metabolic responses were assessed using nuclear magnetic resonance (NMR) to identify the molecular mechanism of the antioxidant stress reaction and the metabolic reactions of earthworms to HBCD. A significant up-regulation (p < 0.05) of superoxide dismutase (SOD) gene expression was detected, with the highest gene expression level of SOD appearing at a dose of 400 mg kg-1 dw (2.06-fold, p < 0.01). However, the glutathione transferase (GST) gene expression levels did not differ significantly (p > 0.05). Principal component analysis (PCA) of the metabolic responses showed that all groups could be clearly differentiated, and the highest concentration dose group was the most distant from the control group. Except for fumarate, the measured metabolites, which included adenosine triphosphate (ATP), valine, lysine, glycine, betaine and lactate, revealed significant (p < 0.05) increases after 14 days of exposure to HBCD. HBCD likely induces high levels of anaerobic respiration, which would result in high levels of ATP and lead to the disintegration of proteins into amino acids, including valine and lysine, to produce energy. The observed changes in osmotic pressure were indicative of damage to the membrane structure. Furthermore, this study showed that NMR-based metabolomics was a more sensitive tool than measuring the gene expression levels for elucidating the mode of toxicity of HBCD in earthworm exposure studies.
Enantioselective Toxicity of Metolachlor to Scenedesmus Obliquus in the Presence of Cyclodextrins

Abstract:Cyclodextrins (CDs) possess a variety of chiral centers and are capable of recognizing enantiomeric molecules through the formation of inclusion complexes. Two types of CDs, α-cyclodextrin (α-CD) and β-cyclodextrin (β-CD), were selected to evaluate the effects of the enantioselective ecotoxicity of racemic metolachlor (Rac-metolachlor) and its S-enantiomer (S-metolachlor) on the freshwater algae Scenedesmus obliquus (S. obliquus) by acute toxicity test. The results showed that the aquatic toxicity of S-metolachlor was higher than Rac-metolachlor and that CDs enhanced the toxicity of metolachlor enantioselectively by increasing the aquatic toxicity of Rac-metolachlor rather than that of S-metolachlor to S. obliquus. The equilibrium constant for Rac-metolachlor-CD complexes was higher than that of S-metolachlor-CDs, which was responsible for the greater aquatic toxicity shift effect of Rac-metolachlor. Thermodynamic studies of CD complexes showed that inclusion for all of the complexes was primarily a spontaneous, enthalpy-driven process. These results will help to understand the preliminary mechanism of shifting aquatic toxicity of metolachlor by CDs and the CDs mediated environmental processes of metolachlor, to correctly apply CDs to chiral pesticides formulation and environmental remediation of chiral contaminants.
Different Cytotoxicity Induced by Hexabromocyclododecanes on Mouse Neuroblastoma N2a Cells via Oxidative Stress and Mitochondrial Apoptotic Pathway

Abstract:Hexabromocyclododecane (HBCD) is widely used in polystyrene foams, building materials, and electrical equipment as a brominated flame retardant (BFR) and persists in the environment and human body matrix. It has attracted increased attention since its neuroendocrine disorder effects have been observed in humans and animals. However, studies evaluating the neurotoxicity of HBCD diastereoisomers and the potential mechanisms involved are still limited. In this study, we compared the cytotoxicity induced by the three HBCD diastereoisomers (i.e., α-, β-, and γ-HBCD) in N2a cells and further investigated the underlying molecular mechanism. Our results showed that HBCD diastereoisomers decreased cell viability in the order of β-HBCD > α-HBCD > γ-HBCD. Moreover, α-HBCD and β-HBCD exposure led to different degrees of cell cycle disruption and oxidative stress of N2a cells, implying that oxidative stress-mediated differential cytotoxicity of HBCD diastereoisomers. The expressions of caspases and Bcl-2 were differentially regulated by α-HBCD and β-HBCD, suggesting that the mitochondrial apoptosis pathway may be critical in HBCDs-mediated N2a cell toxicity. Therefore, our studies provided novel evidence for the underlying mechanisms of the distinct cytotoxicity of HBCD diastereoisomers.

Abstract:ZDAX5 is a variety of herbicide-tolerant maize that contains the modified P450-N-Z1 gene isolated from Cynodon dactylon (L.) Pers. and the cp4 epsps gene isolated from the Agrobacterium tumefaciens strain CP4 and exhibits high tolerances to flazasulfuron and glyphosate under field conditions. Once ZDAX5 corn is available on the market, the evolution of herbicide-resistant weeds will be delayed by applying glyphosate and flazasulfuron to corn fields. Prior to commercialization, it is critical to assess the safety of ZDAX5 maize. Compositional analysis and feed consumption studies in rodents are an important consideration in the safety assessment of genetically modified crops. The nutritional components of ZDAX5 were analyzed and compared with those of its non-transgenic counterpart. The data showed that all the analyzed components in the herbicide-tolerant maize plants were substantially equivalent to those of its non-transgenic counterpart. Furthermore, most of the measured values from ZDAX5 were within the range of values reported for other commercial maize varieties. The sub-chronic feeding trial was carried out with grains from GM, and non-GM maize were independently added into rodent diets at concentrations of 12.5%, 25% and 50%. As a control, another set of rats was fed with a marketed diet. At the end of the 90-day feeding study, no negative effects associated with the consumption of GM maize were found. These results indicate that the herbicide-tolerant maize ZDAX5 is as nutritious and safe as non-transgenic maize.
Phenotypic and Proteomic Insights into Differential Cadmium Accumulation in Maize Kernels

Abstract:The contamination of agricultural soil with cadmium (Cd), a heavy metal, poses a significant environmental challenge, affecting crop growth, development, and human health. Previous studies have established the pivotal role of the ZmHMA3 gene, a P-type ATPase heavy metal transporter, in determining variable Cd accumulation in maize grains among 513 inbred lines. To decipher the molecular mechanism underlying mutation-induced phenotypic differences mediated by ZmHMA3, we conducted a quantitative tandem mass tag (TMT)-based proteomic analysis of immature maize kernels. This analysis aimed to identify differentially expressed proteins (DEPs) in wild-type B73 and ZmHMA3 null mutant under Cd stress. The findings demonstrated that ZmHMA3 accumulated higher levels of Cd compared to B73 when exposed to varying Cd concentrations in the soil. In comparison to soil with a low Cd concentration, B73 and ZmHMA3 exhibited 75 and 142 DEPs, respectively, with 24 common DEPs shared between them. ZmHMA3 showed a higher induction of upregulated genes related to Cd stress than B73. Amino sugar and nucleotide sugar metabolism was specifically enriched in B73, while phenylpropanoid biosynthesis, nitrogen metabolism, and glyoxylate and dicarboxylate metabolism appeared to play a more significant role in ZmHMA3. This study provides proteomics insights into unraveling the molecular mechanism underlying the differences in Cd accumulation in maize kernels.
Cadmium toxicity in Maize (Zea mays L.): consequences on antioxidative systems, reactive oxygen species and cadmium accumulation

Abstract:Increased cadmium (Cd) accumulation in soils has led to tremendous environmental problems, with pronounced effects on agricultural productivity. Present study investigated the effects of Cd stress imposed at various concentrations (0, 75, 150, 225, 300, 375 μM) on antioxidant activities, reactive oxygen species (ROS), Cd accumulation, and productivity of two maize (Zea mays L.) cultivars viz., Run Nong 35 and Wan Dan 13. Considerable variations in Cd accumulation and in behavior of antioxidants and ROS were observed under Cd stress in both maize cultivars, and such variations governed by Cd were concentration dependent. Exposure of plant to Cd stress considerably increased Cd concentration in all plant parts particularly in roots. Wan Dan 13 accumulated relatively higher Cd in root, stem, and leaves than Run Nong 35; however, in seeds, Run Nong 35 recorded higher Cd accumulation. All the Cd toxicity levels starting from 75 μM enhanced H2O2 and MDA concentrations and triggered electrolyte leakage in leaves of both cultivars, and such an increment was more in Run Nong 35. The ROS were scavenged by the enhanced activities of superoxide dismutase, peroxidase, catalase, ascorbate peroxidase, and glutathione peroxidase in response to Cd stress, and these antioxidant activities were higher in Wan Dan 13 compared with Run Nong 35 at all Cd toxicity levels. The grain yield of maize was considerably reduced particularly for Run Nong 35 under different Cd toxicity levels as compared with control. The Wan Dan 13 was better able to alleviate Cd-induced oxidative damage which was attributed to more Cd accumulation in roots and higher antioxidant activities in this cultivar, suggesting that manipulation of these antioxidants and enhancing Cd accumulation in roots may lead to improvement in Cd stress tolerance.

Abstract:To assess the toxicity of Hexabromocyclododecane (HBCD), the population, individual, and cellular biochemical parameters of the rotifer Brachionus plicatilis exposed to different concentrations of HBCD were investigated. The results showed that the population growth rate, reproductive period, and offspring number in B. plicatilis significantly decreased under 324 μg/L and 648 μg/L HBCD. Antioxidant enzyme activity and mRNA expression of CAT and Mn-SOD were promoted at low concentrations (32 μg/L and 64 μg/L) and inhibited at high concentrations (324 μg/L and 648 μg/L), while MDA content accumulated continuously with increasing HBCD concentrations, indicating that HBCD induced oxidation imbalance in rotifers. Further evidence was provided by the correlation between DNA fragmentation and physiological changes. The increased intercellular concentration of Ca 2+ and the expression of CaM mRNA suggested that HBCD activated pathways related to calcium signaling. In summary, the excessive production of ROS induced by HBCD was considered to be the main cause of reproductive toxicity.

Abstract:Microplastics (MPs) and cadmium (Cd) has attracted increasing attention due to their combined toxicity to terrestrial vegetation. Photosynthesis which utilizes light energy to synthesize organic substances is crucial for crop production. However, the plant photosynthetic response to the joint toxicity of MPs and Cd is still unknown. Here, we studied the effects of polyethylene (PE) MPs on the photosynthetic performance of two maize cultivars Xianyu 335 (XY) and Zhengdan 958 (ZD) grown in a Cd contaminated soil. Results showed that the leaf Cd concentration in XY and ZD reached 26.1 and 31.9 μg g −1 , respectively. PE-MPs did not influence the leaf Cd content, but posed direct and negative effects on photosynthesis by increasing the malondialdehyde content, reducing the chlorophyll content, inhibiting photosynthetic capacity, disrupting the PSII donor side, blocking electron transfer in different photosystems, and suppressing the oxidation and reduction states of PSI. Transcriptomic analysis revealed that the inhibitory effect of combined PE-MPs and Cd on maize photosynthesis was attributed to suppressed expression of the genes encoding PSII, PSI, F-type ATPase, cytochrome b 6/f complex, and electron transport between PSII and PSI. Using WGCNA, we identified a MEturquoise module highly correlated with photosynthetic traits. Hub genes bridging carbohydrate metabolism, amino acid metabolism, lipid metabolism, and translation provided the molecular mechanisms of PE-MPs and Cd tolerance in maize plants. The comprehensive information on the phytotoxicity mechanisms of Cd stress in the presence or absence of PE-MPs on the photosynthesis of maize is helpful for cloning Cd and PE-MP resistance genes in the future.

Variability in cadmium stress tolerance among four maize genotypes: Impacts on plant physiology, root morphology, and chloroplast microstructure

Abstract:Cadmium (Cd) is detrimental to both plants and humans. Maize (Zea mays L.) genotypes exhibit variations in Cd accumulations. This study examined variations in Cd accumulation and tolerance among four maize genotypes with contrasting root morphology. The four maize genotypes were cultivated in a semi-hydroponic system with three Cd concentrations (0, 10, 20 μmol L-1). The effects of Cd on plant growth and physiology were assessed 39 days after transplanting. Results showed that root characteristics were positively correlated with root Cd accumulation and the bioconcentration factor under Cd20 treatment. Genotypes Shengrui999 and Zhengdan958 exhibited higher total Cd content than Xundan29 and Zhongke11 under Cd20 conditions. Cd toxicity led to membrane degradation of chloroplast mesophyll cells, loosening and swelling of grana lamella, and reduced starch reserves. The greater tolerance of Shengrui999 and Zhengdan958 was contributed to factors such as root biomass, shallower root depth, higher Cd content, accumulation of osmolyte such as soluble protein, antioxidant activities such as catalase (CAT), and the presence of phytohormone gibberellic acid. The study establishes a link between root morphology, Cd accumulation, and tolerance in maize plants, as demonstrated by the higher Cd accumulation and shallower root system in Cd-tolerant genotypes. This research provides a foundation for breeding maize cultivars better suited for adaptation to moderate Cd-contaminated environments.
Developmental Toxicity Evaluation of Three Hexabromocyclododecane

Abstract:Structural dissimilarities of hexabromocyclododecane diastereoisomers could raise substantial differences in physicochemical, biological and toxicological properties. In order to fully assess the environmental safety and health risk of hexabromocyclododecanes (HBCDs), zebrafish embryos were used to evaluate the developmental toxicity of individual HBCD diastereoisomers (α-HBCD, β-HBCD and γ-HBCD). Four-hour post-fertilization (hpf) zebrafish embryos were exposed to different concentrations of HBCD diastereoisomers (0, 0.01, 0.1 and 1.0 mg/l) until 120 hpf. The results showed that exposure to HBCDs can affect the development of zebrafish embryos/larvae in a dose-dependent and diastereoselective manner. The diastereoisomers α-, β- and γ-HBCD at 0.01 mg/l had little effect on the development of zebrafish embryos except that exposure to 0.01 mg/l γ-HBCD significantly delayed hatching ( P P P P β-HBCD > α-HBCD.
Stereoselective Phytotoxicity of HCH Mediated by Photosynthetic and Antioxidant Defense Systems in Arabidopsis Thaliana

Abstract:BACKGROUND:Hexachlorocyclohexane (HCH) has been used for plant protection and sanitation world-widely, and its isomers have been detected in water, soil, and air as well as in vegetation. As a sink for lipophilic pollutants, vegetation is very important for the degradation and fate of organic contamination; however, little was known about their phytotoxicity and mechanisms of toxic effect. In this study, the stereoselective phototoxicity of four isomers (α, β, γ, and δ) of HCHs mediated by independent as well as interconnecting systems of photosynthesis and enzymatic antioxidant defense system in Arabidopsis thaliana were assessed.PRINCIPAL FINDINGS:Our results revealed that all the HCHs not only stimulated the activities of catalase (CAT) and peroxidase (POD), but also inhibited the activity of superoxide dismutase (SOD). In photosynthesis system, the photosynthetic efficiency of PSI and PSII were all down regulated. Meanwhile, results from both systems showed that δ-HCH was the most toxic one, while α-HCH the least in Arabidopsis thaliana.CONCLUSIONS:For the first time, stereoselective effects of different isomers of HCH in plant were demonstrated. And the results suggest that it requires further research to fully elucidate the environmental toxicity and their mechanisms.
Systemic evaluation of novel acaricide hexythiazox for bioactivity improvement and risk reduction at the enantiomer level

Abstract:Traditional risk assessments of chiral pesticides mainly depend on racemic form, which is often incomprehensive. This study conducted systemic investigations on the bioactivity, toxicity, and ecotoxicological effects of hexythiazox (HTZ) at the enantiomer level. The elution order and absolute configuration of HTZ enantiomers were determined. ( 4R, 5R )-(+)-HTZ exhibited 708 and 1719 times higher bioactivity against Tetranychus cinnabarinus and Tetranychus urticae eggs than ( 4S, 5S )-(−)-HTZ, respectively. Molecular docking indicated greater interactions between ( 4R, 5R )-(+)-HTZ and chitin synthase leading to higher bioactivity of ( 4R, 5R )-(+)-HTZ. However, ( 4S, 5S )-(−)-HTZ induced greater changes in protein and malondialdehyde content, and antioxidant and detoxification enzyme activities than ( 4R, 5R )-(+)-HTZ in earthworms. Furthermore, integrated biomarker response results indicated ( 4S, 5S )-(−)-HTZ exhibited higher toxic effects on earthworms than ( 4R, 5R )-(+)-HTZ. Finally, significant differentially expressed genes (DEGs) were observed in earthworms after exposure to ( 4R, 5R )-(+)-HTZ and ( 4S, 5S )-(−)-HTZ, respectively. These DEGs were mainly enriched in glycolysis/gluconeogenesis and purine metabolism pathways in earthworms. Additionally, six metabolism pathways were also enriched, including pyruvate metabolism, fatty acid biosynthesis, oxidative phosphorylation, citric acid cycle, fatty acid degradation, and ATP-binding cassette transporters. These findings suggest that earthworms exhibited enantiomer-specific responses to ( 4R, 5R )-(+)-HTZ and ( 4S, 5S )-(−)-HTZ. This study provides systemic insight into the toxicity mechanism of HTZ at the enantiomer level and the potential to develop ( 4R, 5R )-(+)-HTZ as a high-efficiency and low-risk pesticide.

Abstract:Different maize varieties respond differentially to cadmium (Cd) stress. As the first organ in contact with the soil, the response of the root is particularly important. However, the physiological mechanisms that determine the response are not well defined. Here, we compared the differences in Cd-induced related gene expression, ionic homeostasis, and ultrastructural changes in roots of Cd-tolerant maize variety (XR57) and Cd-sensitive maize variety (LY296), and assessed their effects on Cd uptake and accumulation. Our findings indicate that XR57 absorbed a significantly lower Cd than LY296 did, and that the expression levels of genes related to Cd uptake (ZmNRAMP5 and ZmZIP4) and efflux (ZmABCG4) in the root were consistent with the Cd absorption at the physiological levels. Compared with LY296, the lower Cd concentration in the roots of XR57 caused less interference with the ion balance. Transmission electron microscope images revealed that the roots from XR57 exposed to Cd had developed thicker cell walls than LY296. In addition, the large increase ZmABCC1 and ZmABCC2 expression levels in XR57 mediated the appearance of numerous electron-dense granules in the vacuoles present in the roots. As a result, the high Cd tolerance of XR57 is the result of a multi-level response that involves increased resistance to Cd uptake, a stronger capacity for vacuolar regionalization, and the formation of thicker cell walls. These findings may provide a theoretical basis for maize cultivation in Cd-contaminated areas.

Abstract:Background: The continued uses of dichlordiphenyltrichloroethane (DDT) for indoor vector control in some developing countries have recently fueled intensive debates toward the global ban of this persistent legacy contaminant. Current approaches for ecological and health risk assessment has ignored the chiral nature of DDT. In this study by employing an array of cytotoxicity related endpoints, we investigated the enantioselective cytotoxicity of o,p'-DDT.Principal Findings: we demonstrated for the first time that R-(2)-o,p'-DDT caused more neuron cell death by inducing more severe oxidative stress, which selectively imbalanced the transcription of stress-related genes (SOD1, SOD2, HSP70) and enzyme (superoxide dismutase and lactate dehydrogenase) activities, and greater cellular apoptosis compared to its enantiomer S-(+)-o,p'-DDT at the level comparable to malaria area exposure (parts per million). We further elucidated enantioselective modes of action using microarray combined with enzyme-linked immunosorbent assay. The enantioselective apoptosis might involve three signaling pathways via caspase 3, tumor protein 53 (p53) and NFkB.Conclusions: Based on DDT stereochemistry and results reported for other chiral pesticides, our results pointed to the same directional enantioselectivity of chiral DDT toward mammalian cells. We proposed that risk assessment on DDT should consider the enantiomer ratio and enantioselectivities.
Enantioselective residues and toxicity effects of the chiral triazole fungicide hexaconazole in earthworms (Eisenia fetida)

Abstract:The enantioselective toxic effect and environmental behavior of chiral pesticides have attracted increasing research attention. In this study, the enantioselective toxicity and residues of hexaconazole (HEX) in earthworms (Eisenia fetida) were investigated. In the present study, significant enantioselective degradation characteristics were observed in artificial soil with the R-enantiomer preferentially degrading (p < 0.05); however, no significant enantioselective bioaccumulation was observed in the earthworms (p > 0.05). The acute toxicity of S-(+)-HEX was higher than that of R-(-)-HEX in earthworms, with 48-h LC50 values of 8.62 and 22.35 μg/cm2, respectively. At 25 mg/kg, enantiospecific induction of oxidative stress was observed in earthworms; moreover, S-(+)-HEX had a greater influence on the contents of malonaldehyde, cytochrome P450, and 8-hydroxy-2-deoxyguanosine than R-(-)-HEX. These results were consistent with those of the enrichment analysis of differentially expressed genes. The transcriptome sequencing results showed that S-(+)-HEX had a more significant influence on steroid biosynthesis, arachidonic acid metabolism, and cell cycle processes than R-(-)-HEX, leading to abnormal biological function activities. These results indicate that S-(+)-HEX may pose a higher risk to soil organisms than R-(-)-HEX. This study suggests that the environmental risk of chiral pesticides to nontarget organisms should be assessed at the enantiomeric level.
Stereoisomer-Specific Trophodynamics of the Chiral Brominated Flame Retardants HBCD and TBECH in a Marine Food Web, with Implications for Human Exposure

Abstract:Stereoisomers of 1,2,5,6,9,10-hexabromocyclododecane (HBCD) and 1,2-dibromo-4-(1,2-dibromoethyl)-cyclohexane (TBECH) were determined in sediments and 30 marine species in a marine food web to investigate their trophic transfer. Lipid content was found to affect the bioaccumulation of ΣHBCD and ΣTBECH in these species. Elevated biomagnification of each diastereomer from prey species to marine mammals was observed. For HBCD, biota samples showed a shift from γ- to α-HBCD when compared with sediments and technical mixtures; trophic magnification potential of (-)-α- and (+)-α-HBCD were observed in the food web, with trophic magnification factors (TMFs) of 11.8 and 8.7, respectively. For TBECH, the relative abundance of γ- and δ-TBECH exhibited an increasing trend from abiotic matrices to biota samples; trophic magnification was observed for each diastereomer, with TMFs ranging from 1.9 to 3.5. The enantioselective bioaccumulation of the first eluting enantiomer of δ-TBECH in organisms at higher TLs was consistently observed across samples. This is the first report on the trophic transfer of TBECH in the food web. The estimated daily intake of HBCD for Hong Kong residents was approximately 16-times higher than that for the general population in China, and the health risk to local children was high, based on the relevant available reference dose.
