Abstract:In acidic mine wastewater, Mn and Fe are among the prime heavy metals and pose a great threat to the ecological environment and human health. Combining biological and physical methods to treat heavy metal contaminated water bodies is a promising remediation technique. In this study, grapefruit peel biochar immobilized Acinetobacter sp. AL-6 (co-system) was applied to treat the contaminated water with co-existing Mn2+, Fe2+ and NH4 +-N. The results showed that almost 100% of Mn2+, Fe2+, Zn2+ and Cu2+ were removed in bioreactor, and was transferred to Mn2+, Fe3+, Zn2+ and Cu2+ precipitates on the surface of co-system without N2O accumulation. Especially, Mn2+ and Fe2+ were adsorbed in extracellular polymers (EPS) of strain AL-6. The present study mainly confirmed that the combined system of biochar and bacteria had great potential in the complex water removal of multiple heavy metals and NH4 +-N, and also revealed the regulation of heavy metals by the EPS of strain.

Abstract:Denitrification is an important biological process in wastewater treatment plants (WWTPs). However, a low carbon-to-nitrogen (C/N) ratio limits the availability of organic carbon, potentially reducing denitrification efficiency. This study investigates the impact of sludge-derived biochar on the nitrogen removal of activated sludge for low C/N ratio municipal wastewater. Sludge-based biochar was characterized by its physicochemical properties, revealing that biochar prepared at 400 °C exhibited the highest specific surface area and the most favorable surface functional groups for electron transfer. The results from batch tests showed that adding 4 g/L of biochar dosage enhanced denitrification rates and total nitrogen (TN) removal efficiency the most. Sequencing batch reactors (SBRs) experiments further confirmed that biochar dosgae improved the removal efficiencies of COD, NH 4 + -N, and TN, achieving stable values of 97.2 ± 1.2 %, 99.2 ± 0.6 %, and 83.8 ± 2.4 %, respectively. Metabolic and electrochemical analyses revealed that biochar addition enhanced the activity of denitrification enzymes, increasing the ammonia oxidation rate by 12.9 ± 0.7 %, nitrite oxidation rate by 14.7 ± 1.2 %, nitrate reduction rate by 36.9 ± 1.5 %, and nitrite reduction rate by 16.4 ± 0.8 %. The relative abundance of denitrification functional genes ( amoA, nirS, nirK, narG, nosZ ) increased, and the activities of the corresponding enzymes (AMO, NXR, NAP, NIR) rose by 23±6 %, 53±5 %, 260±15 %, and 55±7 %, respectively. This increase in enzyme activity suggested enhanced denitrification processes, which was further supported by the 60.1 ± 3.7 % increase in electron transfer system activity (ETSA), indicating that biochar acted as an electron shuttle. This study proposes a potential sustainable approach for sludge recycling and enhanced wastewater nitrogen removal under low C/N conditions.

Abstract:Microbial remediation is an eco-friendly and promising approach for the decontamination of wastewater containing sulfamethoxazole (SMX) and Zn2+. However, free bacteria are sensitive to environmental variation, limiting the application. In this study, biochar-immobilized Bacillus paramycoides SDB4 (ISDB4) was used to overcome the disadvantages. The results showed that the removal by ISDB4 was significantly increased under different pH conditions and initial combined contaminant concentrations. Notably, at pH 7, SMX (20 mg L-1) and Zn2+ (100 mg L-1) were completely removed by ISDB4, and their levels were 29.28% and 92.34% higher than that of SDB4, respectively. Besides, ISDB4 showed excellent reusability with 43.24% of SMX and 50.34% of Zn2+ removal efficiencies after five continuous cycles. Such improvements might be related to enhanced biotransformation of SMX from 45.81% with SDB4 to 89.98% with ISDB4. Compared with SDB4, the contributions of ion exchange and complexation of ISDB4 with Zn2+ increased by 9.53% and 6.72%, respectively. In case of ISDB4, SEM-EDS and FTIR showed that biochar provided sufficient space for the attachment and proliferation of SDB4, and the removal of combined contaminants was enhanced by SDB4 due to the presence of functional groups and light metals on biochar. This study provides a green and effective strategy for the bioremediation of antibiotic and heavy metal co-contaminated aquaculture wastewater.
Metopic suture and RUNX2, a key transcription factor in osseous morphogenesis with possible important implications for human brain evolution.

Abstract:BACKGROUND Overall, the comparative data available on the timing of metopic suture closure in present-day and fossil members of human lineage, as well as great apes, seem to indicate that human brain evolution occurred within a complex network of fetopelvic constraints, which required modification of frontal neurocranial ossification patterns, involving delayed fusion of the metopic suture. It is very interesting that the recent sequencing of the Neanderthal genome has revealed signs of positive selection in the modern human variant of the RUNX2 gene, which is known to affect metopic suture fusion in addition to being essential for osteoblast development and proper bone formation. It is possible that an evolutionary change in RUNX2, affecting aspects of the morphology of the upper body and cranium, was of importance in the origin of modern humans. Thus, to contribute to a better understanding of the molecular evolution of this gene probably implicated in human evolution, we performed a comparative bioinformatic analysis of the coding sequences of RUNX2 in Homo sapiens and other non-human Primates. RESULTS We found amino-acid sequence differences between RUNX2 protein isoforms of Homo sapiens and the other Primates examined, that might have important implications for the timing of metopic suture closure. CONCLUSIONS Further studies are needed to clear the potential distinct developmental roles of different species-specific RUNX2 N-terminal isoforms. Meantime, our bioinformatic analysis, regarding expression of the RUNX2 gene in Homo sapiens and other non-human Primates, has provided a contribution to this important issue of human evolution.
Synthesis of acidified magnetic sludge-biochar and its role in ammonium nitrogen removal: Perception on effect and mechanism

Abstract:An acidified magnetic sludge-biochar (MSB) was prepared to enhance ammonium nitrogen (AN) removal efficiency in eutrophic water, and MSB was obtained by secondary pyrolysis of sludge biochar powder. A series of MSB were prepared under 300, 400, 500, 600 °C and different valence states of iron ions by impregnation pyrolysis, which is based on the deposition of unstable iron minerals on biochar matrix. Physicochemical properties of pristine biochar and MSB were revealed through characterization analysis, suggesting that MSB prepared by ferric chloride at 400 °C presented the largest adsorption capacity, and the acid-modification enhanced the ammonium adsorption capacity by 10.7%. Electrostatic attraction and ion-exchange processes were identified as the main adsorption mechanisms of MSB on AN. As the most dominant mechanism, ion exchange of AN with functional groups containing -OH and CO on the surface of MSB resulted in the relative content of -OH (61.3%) and CO (11.5%) bonds reduced to 34.2% and 7.0% respectively. The novel magnetic sludge-biochar with acid-modification possessed enhanced electron transfer capacity, revealing a removal pathway of ammonium by nitrification. The findings above demonstrated that MSB is a promising adsorbent for ammonium removal and can be applied to the natural nitrogen-rich water regulation.

Biochar derived from alkali-treated sludge residue regulates anaerobic digestion: Enhancement performance and potential mechanisms

Abstract:Biochar produced from bio-wastes has been widely used to promote the performance of anaerobic digestion. Waste activated sludge (WAS) is considered as a kind of popular precursor for biochar preparation, but the abundant resources in WAS were neglected previously. In this study, the roles of biochar prepared from raw, pretreated, and fermented sludge on anaerobic digestion were investigated. That is, parts of carbon sources and nutrients like polysaccharides, proteins, and phosphorus were firstly recovered after sludge pretreatment or fermentation, and then the sludge residuals were used as raw material to prepare biochar. The methane yield improved by 22.1% with adding the biochar (AK-BC) prepared by sludge residual obtained from alkaline pretreatment. Mechanism study suggested that the characteristics of AK-BC like specific surface area and defect levels were updated. Then, the conversion performance of intermediate metabolites and electro-activities of extracellular polymeric substances were up-regulated. As a result, the activity of electron transfer was increased with the presence of AK-BC, with increase ratio of 21.4%. In addition, the electroactive microorganisms like Anaerolineaceae and Methanosaeta were enriched with the presence of AK-BC, and the potential direct interspecies electron transfer was possibly established. Moreover, both aceticlastic and CO2-reducing methanogenesis pathways were improved by up-regulating related enzymes. Therefore, the proposed strategy can not only obtain preferred biochar but also recover abundant resources like carbon source, nutrients, and bioenergy.

Abstract:Conventional coagulation or enhanced coagulation water treatment produced mass floc sludge, which contained plenty of metals and organic matter. Floc sludge disposal and resource recycling was vital relevance and urgently needed. Here, we reported on the waste flocs recycling through in-situ manganese-aluminum-iron biochar (MAFBC) synthesized via one-step pyrolysis using potassium permanganate (KMnO 4 ) oxidation- aluminum-iron composite flocculant coagulation for humic acid removal floc sludge as raw material. Compared with ex-situ loaded or similar types of biochar, MAFBC showed faster and efficient activation effect of peroxymonosulfate (PMS) to degrade tetracycline (TC). TC was rapidly removed within 1 min in MAFBC/PMS system with degradation rate constant K obs was 1.825 min −1 , which mainly attributed to the multiple promoting effects of manganese-aluminum-iron. Aluminum oxides facilitated TC adsorption and synergistic Fe/Mn further significantly promoted electron transfer on the surface of the MAFBC. Additionally, bimetallic structure of Fe/Mn and the CO functional group on MAFBC accelerated the formation of reactive species. The redox cycles of Fe 2+ /Fe 3+ and Mn 2+ /Mn 3+ /Mn 4+ promoted the formation of free radicals OH and SO 4 − . Moreover, O 2 – and 1 O 2 also assumed an important role for TC removal during active species evolution drived by Fe/Mn. This research provides a new perspective for in-situ design of economical and stable metal carbon-based water treatment activator, and make contributions to the utilization of waste flocs in water treatment.

Synergistic enhancement of microbes-to-pollutants and inter-microbes electron transfer by Fe, N modified ordered mesoporous biochar in anaerobic digestion

Abstract:Extracellular electron transfer was essential for degrading recalcitrant pollutants by anaerobic digestion (AD). Therefore, existing studies improved AD efficiency by enhancing the electron transfer from microbes-to-pollutants or inter-microbes. This study synthesized a novel Fe, N co-doped biochar (Fe, N-BC), which could enhance both the microbes-to-pollutants and inter-microbes electron transfer in AD. Detailed characterization data indicated that Fe, N-BC has an ordered mesoporous structure, high specific surface area (463.46 m2/g), and abundant redox functional groups (Fe2+/Fe3+, pyrrolic-N), which translate into excellent biocompatibility and electrochemical properties of Fe, N-BC. By adding Fe, N-BC, the stability and efficiency of the medium-temperature AD system in the treatment of methyl orange (MO) wastewater were improved: obtained a high degradation efficiency of MO (96.8 %) and enhanced the methane (CH4) production by 65 % compared to the control group. Meanwhile, Fe, N-BC reduced the accumulation of volatile fatty acids in the AD system, and the activity of anaerobic granular sludge electron transport system and coenzyme F420 was enhanced. In addition, Fe, N-BC showed positive enrichment of azo dyes decolorization bacteria (Georgenia) and direct interspecies electron transfer (DIET) synergistic partners (Syntrophobacter, Methanosarcina). Overall, the rapid degradation of MO and enhanced CH4 production in AD systems by Fe, N-BC is associated with enhancing two electronic pathways, i.e., microbes to MO and DIET between syntrophic bacteria and methanogenic archaea. This study introduced an enhanced "two-pathways of electron transfer" theory, realized by Fe, N-BC. These findings provided new insights into the interactions within AD systems and offer strategies for enhancing their performance with recalcitrant pollutants.
Excess sludge-based biochar loaded with manganese enhances catalytic ozonation efficiency for landfill leachate treatment

Abstract:This study developed an efficient and stable landfill leachate treatment process, which was based on the combination of biochar catalytic ozonation and activated sludge technology for intensive treatment of landfill leachate, aiming to achieve the standard discharge of leachate. The focus is to investigate the effect of manganese loading on the physicochemical properties of biochar and the mechanism of its catalytic ozonation. It was found that more surface functional groups (CO, Mn–O, etc.) and defects (I D /I G = 1.27) were exposed via the change of original carbon structure by loading Mn, which is conducive to the generation of lattice oxygen. Meanwhile, generating different valence states of Mn metal can improve the redox properties and electron migration rate, and encourage the production of reactive oxygen species (ROS) during the reaction process and enhance the catalytic efficiency. The synergistic action of microorganisms, especially denitrifying bacteria, was found to play a key role in the degradation of nitrogenous pollutants during the activated sludge process. The concentration of NH + 4 -N was reduced from the initial 1087.03 ± 9.56 mg/L to 9.05 ± 1.91 mg/L, while COD was reduced from 2290 ± 14.14 mg/L to 86.5 ± 2.12 mg/L, with corresponding removal rates of 99.17% and 99.20%, respectively. This method offers high efficiency and stability, achieving discharge standards for leachate (GB16889-2008). The synergy between Mn-loaded biochar and microorganisms in the activated sludge is key to effective treatment. This study offers a new approach to solving the challenge of waste leachate treatment.

Abstract:Membrane bioreactors (MBRs) are frequently used to treat municipal wastewater, but membrane fouling is still the main weakness of this technology. Additionally, the low carbon-nitrogen (C/N) ratio influent has been shown to not only increase the membrane fouling, but also introduce challenges to meet the effluent discharge standard for nitrogen removal. Herein, the authors addressed the challenges by adding cost-effective biochar. The results suggested that the biochar addition can enable membrane fouling alleviation and nitrogen removal improvement. The reduced membrane fouling can be ascribed to the biochar adsorption capacity, which facilitates to form bigger flocs with carbon skeleton in biochar as a core. As a result, the biochar addition significantly altered the mixed liquor suspension with soluble microbial product (SMP) concentration reduction of approximately 14%, lower SMP protein/polysaccharide ratio from 0.28 ± 0.02 to 0.22 ± 0.03, smaller SMP molecular weight and bigger sludge particle size from 67.68 ± 6.9 μm to 113.47 ± 4.8 μm. The nitrogen removal is also dramatically improved after biochar addition, which can be due to the initial carbon source release from biochar, and formation of aerobic–anaerobic microstructures. Microbial diversity analysis results suggested more accumulation of denitrification microbes including norank_f__JG30-KF-CM45 and Plasticicumulans. Less relative abundance of Aeromonas after biochar addition suggested less extracellular polymer substance (EPS) secretion and lower membrane fouling rate.

Abstract:For antibiotic-enriched waste activated sludge, classical iron-based chemical conditioning significantly enhanced sludge dewaterability. Nevertheless, the intricate constituents within sludge rapidly depleted reactive oxygen species (ROS), leading to challenges such as excessive production of iron sludge and inadequate elimination of antibiotics from sludge. Herein, we proposed an innovative strategy integrating biochar with Fe(II) for peroxymonosulfate (PMS) activation, aiming to enhance both sludge dewaterability and antibiotics elimination simultaneously. Compared to classical chemical conditioning of Fe(II)/PMS, the presence of biochar not only reduced bound water content of sludge from 1.36 g/g DS to 0.97 g/g DS, but also enhanced sulfamethoxazole (SMX) degradation rate constant from 0.015 min -1 to 0.042 min -1 . Mechanism studies disclosed the essential roles of biochar in modulating Fe oxidative states distribution and reaction sites in multiphase. Initially, biochar elevated Fe(II)/Fe(III) ratio from 0.38 to 0.78 by abundant carbon defects, which significantly promoted the cumulative concentration of predominant ROS, hydroxyl radicals (•OH), from 4.6 mM to 8.1 mM. Subsequently, EPS underwent destruction by •OH, leading to the liberation of antibiotics and negatively charged polysaccharides (PS), proteins (PN). Secondly, biochar enriched hydrophobic PN with an elevated ratio of PN/PS from 0.92 to 1.50, while the charge neutralization occurred between Fe(II)/Fe(III) and PN, PS, leading to sludge particles granulation. Finally, the mesoporous structure of biochar not only achieved SMX enrichment, but also enhanced the mass transfer of Fe(II)/Fe(III) from sludge aqueous phase to its surface, ensuring that the in-situ generated •OH efficiently targets the locally concentrated SMX. Overall, this work provides a new guidance for developing biochar-mediated chemical conditioning, aiming to enhance the generation and utilization of •OH for antibiotics elimination from sludge.

Abstract:Biochar has been proved to improve methane production in high solids anaerobic co-digestion (HS-AcoD) of dewatered sludge (DS) and food waste (FW), but its potential mechanism for simultaneous methane production and phosphorus (P) transformation has not been sufficiently revealed. Results showed that the optimal preparation temperature and dosage of sludge-based biochar were selected as 300 °C and 0.075 g·g −1 , respectively. Under this optimized condition, the methane production of the semi-continuous reactor increased by 54 %, and the active phosphorus increased by 18 %. The functional microorganisms, such as Methanosarcina , hydrogen-producing, sulfate-reducing, and iron-reducing bacteria, were increased. Metabolic pathways associated with sulfate reduction and methanogenesis, especially hydrogenotrophic methanogenesis, were enhanced, which in turn promoted methanogenesis and phosphorus transformation and release. This study provides theoretical support for simultaneously recovery of carbon and phosphorus resources from DS and FW using biochar.

Unacclimated Activated Sludge Improved Nitrate Reduction and N2 Selectivity in Iron Filling/biochar Systems

This study aimed to improve nitrate reduction and N 2 selectivity in iron filling/biochar (Fe/BC) systems with a simplified approach by coupling unacclimated microbes (M) in the system. Factors such as initial pH, Fe/BC ratio, and Fe/BC dosage on nitrate removal efficiency and N 2 selectivity were evaluated. Results show that the introduction of microbes significantly enhanced nitrate removal and N 2 selectivity, achieving 100 % nitrate removal and 79 % N 2 selectivity. The Fe/BC/M system exhibited efficient nitrate reduction at pH of 2 - 10. Moreover, the Fe/BC/M system demonstrated an improved electrochemical active surface area (ECSA), lower electron transfer resistance and lower corrosion potential, leading to enhanced nitrate reduction. The high i 0 value in Fe/BC/M system means more H ads could be generated, thus improving the N 2 selectivity. This study provides valuable insights into a novel approach for effective nitrate removal, offering a potential solution to the environmental challenges posed by excessive nitrate in wastewater, surface water and ground water.

Results show that the introduction of microbes significantly enhanced nitrate removal and N 2 selectivity, achieving 100 % nitrate removal and 79 % N 2 selectivity. The Fe/BC/M system exhibited efficient nitrate reduction at pH of 2–10. Moreover, the Fe/BC/M system demonstrated an improved electrochemical active surface area (ECSA), lower electron transfer resistance and lower corrosion potential, leading to enhanced nitrate reduction. The high i 0 value in Fe/BC/M system means more H ads could be generated, thus improving the N 2 selectivity. This study provides valuable insights into a novel approach for effective nitrate removal, offering a potential solution to the environmental challenges posed by excessive nitrate in wastewater, surface water and ground water.

Effects of methanol, sodium citrate, and chlorella powder on enhanced anaerobic treatment of coal pyrolysis wastewater

Abstract:To better promote environment friendly development of the coal chemical industry, this study investigated effects of methanol, sodium citrate, and chlorella powder (a type of microalgae) as co-metabolic substances on enhanced anaerobic treatment of coal pyrolysis wastewater with anaerobic sludge. The anaerobic sludge was loaded into four 2 L anaerobic reactors for co-metabolism enhanced anaerobic experiments. Anaerobic reactor 1 (R1) as control group did not add a co-metabolic substance; anaerobic reactor 2 (R2) added methanol; anaerobic reactor 3 (R3) added sodium citrate; and anaerobic reactor 4 (R4) added chlorella powder. In the blank control group, the removal ratios of total phenol (TPh), quinoline, and indole were only 12.07%, 42.15%, and 50.47%, respectively, indicating that 50 mg/L quinoline, 50 mg/L indole, and 600 mg/L TPh produced strong toxicity inhibition function on the anaerobic microorganism in reactor. When the concentration of methanol, sodium citrate, and chlorella was 400 μg/L, the reactors with co-metabolic substances had better treatment effect on TPh. Among them, the strengthening effects of sodium citrate (TPh removal ratio: 44.87%) and chlorella (47.85%) were better than that of methanol (38.72%) and the control group (10.62%). Additionally, the reactors with co-metabolic substances had higher degradation ratios on quinoline, indole, and chemical oxygen demand (COD). The data of extracellular polymeric substances showed that with the co-metabolic substances, anaerobic microorganisms produced more humic acids by degrading phenols and nitrogen-containing heterocyclic compounds (NHCs). Compared with the control group, the reactors added with sodium citrate and chlorella had larger average particle size of sludge. Thus, sodium citrate and chlorella could improve sludge sedimentation performance by increasing the sludge particle size. The bacterial community structures of reactors were explored and the results showed that Aminicenantes genera incertae sedis, Levinea, Geobacter, Smithella, Brachymonas, and Longilinea were the main functional bacteria in reactor added with chlorella.

Abstract:Although anaerobic digestion is a promising way for valorization of waste activated sludge , the process severely deteriorates the sludge dewaterability. Herein, a novel strategy of iron and nitrogen co-doped biochar (Fe-N-BC) activated peroxydisulfate (PDS) was developed to improve anaerobically digested sludge (ADS) dewaterability. Results showed that Capillary suction time (CST) and moisture content (MC) of ADS after conditioned with Fe-N 2 -BC-800/PDS system dropped to 43.4 s and 66.8% when the initial pH was 6.0, dose of Fe-N 2 -BC-800 was 0.6 g/g VS, dose of PDS was 0.9 mmol/g VS, and reaction time was 30 min, respectively. Quantitative analysis indicated that the heterogeneous oxidation initiated by Fe-N 2 -BC-800 primarily contributed to the enhancement of ADS dewaterability. Mechanistic results revealed that ( S•O4− , O•H , and O21 were generated as reactive oxygen species (ROS) through multiple active sites of Fe-N 2 -BC-800 during catalytic reaction. These highly ROS efficiently decomposed extracellular polymeric substances (EPS) and disintegrated ADS cells, thereby releasing bound and intracellular water into free water. Meanwhile, Fe-N 2 -BC-800/PDS significantly reduced hydrophilic biopolymers , improved hydrophobicity of ADS through adjusting protein secondary structure and enriching hydrophobic groups in sludge pellets. Furthermore, Fe-N 2 -BC-800 produced rigid and porous drainage routes in ADS as skeleton builder, inducing enhancement of ADS dewaterability combing with the strong oxidation. The proposed Fe-N 2 -BC-800/PDS treatment offers a novel and promising technology for high-efficiency enhancement of ADS dewaterability, providing a valuable reference for ADS deep dehydration.

Abstract:The Multilingual Aphasia Examination, a short battery of language measures, was administered to 61 healthy, community-dwelling adults (42 female, 19 male) 70 to 90 years old, and their performances were compared with the norms for younger participants. The intent was to investigate the decline of verbal abilities with advancing age and to consider the implications of such a decline for the interpretation of the performances of other elderly individuals. A significant age-related decline in performance was found on only one (Sentence Repetition) of the nine tests. Of note, Sentence Repetition is also the only test that makes specific demands on both short-term memory and serial auditory information processing. The relative stability of verbal performances from the early adult years to the ninth decade suggests that the occurrence of defective performances in older individuals is more likely to reflect the presence of "age-associated" disease rather than "normal" aging. Further, the findings suggest that normative standards derived from the performances of unselected, representative samples of elderly adults may not be adequate bases for clinical interpretation.

Abstract:Two integrins, α3β1 and α6β4, are high‐affinity receptors for laminin 332, the major laminin isoform of the dermal–epidermal junction, although they are thought to have different functions. Biological and genetic studies have firmly established that the α6β4 integrin is indispensable for the stable anchorage of the epidermis to the underlying dermis. In contrast, the α3β1 integrin is thought to be important for cell migration, although the issue is controversial, and both positive and negative effects have been reported. To address the function of α3β1 integrin, we used small interfering RNA to down‐regulate the α3 subunit in human keratinocytes. The resulting phenotype indicates that lack of α3β1 integrin compromises intercellular adhesion and collective migration, while it enhances single cell migration with a concomitant increase of both focal adhesion kinase and extracellular signal‐regulated kinase. In addition, down‐regulation of integrin α3 subunit results in an increased expression of fibronectin and precursor laminin 332, two extracellular matrix proteins known to be up‐regulated during wound healing. Thus, down‐regulation of α3β1 integrin recapitulates crucial events governing keratinocyte migration associated with wound healing and tissue repair.
COD inhibition alleviation and anammox granular sludge stability improvement by biochar addition

Abstract:High chemical oxygen demand (COD) in anaerobic ammonium oxidation (anammox) reactors may destroy the structure or inhibite the activity of anammox granular sludge (AnGS). Herein, the effects of adding different biochars on the alleviation of COD inhibition, performance of anammox reactors in phases I (fed with an initial COD = 100 mg L−1) and II (= 150 mg L−1), as well as their effect on the properties of AnGS granules were investigated. Results showed that nitrogen removal in biochar-added reactors was superior to that in reactors without biochar (the control). AnGS with biochar addition outperformed the control in terms of stability (EPS and particle size distribution) as the initial COD concentration increased. In the reactors amended with litchi biochar and chinar biochar, the respective average TN removal efficiencies were 86.49% and 85.70% in phase Ⅰ as well as 86.54% and 85.51% in phase Ⅱ; the proportion of AnGS particles with a diameter of 1–3 mm reached a respective maximum of 80.97% and 73.74% (much higher than 67.69% in the control); and the AnGS granules had better stability with a little change in the protein (PN) to polysaccharide (PS) (PN/PS) values, the PN/PS values were 4.18 and 4.77 in phase Ⅰ as well as 4.21 and 4.70 in phase Ⅱ. Even at increasing COD concentrations, no significant change in the relative abundance of Planctomycetes was observed in all the biochar-containing reactors (P value over 0.9), and the relative abundance of anammox-related functional genes (hzs, hdh) was higher than that in the control.

Abstract:This study aimed to investigate the effectiveness and mechanism of biochar and modified magnetic biochar addition in improving the anaerobic digestion (AD) efficiency of swine wastewater under ammonia stress. Two batches of serial experiments were carried out with ammonia levels of 3000, 4500, and 6000 mg/L. The results indicated that a one-time addition of 15 g/L of both types of carbon materials could sustainably optimize the AD performance of swine wastewater at ammonia nitrogen levels of 3000 and 4500 mg/L. In the second batch of experiments, at 4500 mg/L ammonia, groups with C and FeC additions showed 6.5%, 16.2% higher cumulative methane yield and 45.5%, 60.0% greater maximum gas production rates than controls. However, under the ammonia stress condition of 6000 mg/L, the reactor exhibited a mismatch between the carbon material addition and the degree of stress. The maximum methane production rate was intensified only in the first batch, which was 71.8% and 105.1% higher in the C and FeC groups, respectively, than in the control group. The microbiological analysis revealed that biochar optimized the AD performance by enriching the abundance of fermentative bacteria and enhancing the activity of Methanosaeta . In contrast, modified magnetic biochar promoted the enrichment of acetogens and shifted the dominant methanogens from sensitive Methanosaeta to the highly ammonia-tolerant Methanosarcina , thus causing the optimization of AD performance. The findings of this study provided new insights into the mitigation of inhibition and enhancement of efficacy in the anaerobic digestion of swine wastewater.
