Abstract:The formation of acrylamide under three asparagine–sugar low-moisture systems were analyzed by the response surface methodology (RSM) and orthogonal array methodology (OAM) in the present work. The acrylamide content was quantified by a novel validated ultra-performance liquid chromatography tandem mass spectrometry (UPLC–MS/MS). Results of RSM study showed that in the asparagine–glucose system, the acrylamide content achieved a maximum level (442.7μmol/mol Asn) when the system was heated 18min at about 199°C; in the asparagine–fructose system, the acrylamide content increases in the combined condition of higher temperature and shorter heating time (>200°C, <15min) or lower temperature and longer heating time (<150°C, >35min); in the asparagine–sucrose system, the amount of acrylamide enhanced with the increase of temperature and the decrease of time, but declined with the decrease of temperature and the increase of heating time. The coefficients of determination (R2) of the model indicated that all the three models adequately represented the real relationship among the parameters chosen. OAM study indicated that the molar quantity of asparagine monohydrate became the most important parameter contributing to the formation of acrylamide. Meanwhile, acrylamide was readily formed via heating binary precursors 15min at 180°C in the glucose and fructose system while acrylamide was readily generated when the binary precursors were heated 15min at 210°C in the sucrose system.

Abstract:Acrylamide is mainly generated from asparagine and reducing sugars. The comprehensive kinetic profile of acrylamide and Maillard reaction products (MRP) is important for understanding the control of acrylamide. We unraveled the role of flavonoids in the kinetic process of acrylamide via a potato-based asparagine-glucose microwave heating system. Ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS) was employed for the simultaneous determination of asparagine, glucose, fructose and acrylamide. The multi-response non-linear regression models were used for kinetic parameter estimation. The results indicated a positive role of flavonoids in the fructose-participating (k(3): 0.461-0.674; P < 0.05, P < 0.01, compared to the control) instead of the glucose-participating Maillard reaction (k(1): 0.128-0.148; P > 0.05). The addition of flavonoids significantly reduces the formation of acrylamide during the advanced stage (k(4): 2.311-4.327; P < 0.05, P < 0.01) but did not affect its elimination process (k(6): 0.076-0.113; P > 0.05). Treatment of flavonoids has no significant impact on the formation of melanoidins (k(5): 3.645-4.368; P > 0.05) and retains the original sensory attributes of MRP. These findings favor thinking mitigation strategies via the use of additives during food processing.
Attitude tracking control of a GroundGen airborne wind energy system

Abstract:An existing attitude tracking control method developed for a FlyGen airborne wind energy (AWE) system is extended to control a GroundGen AWE system. The GroundGen system produces energy through kite-generated aerodynamic forces that tension a flexible tether attached between the kite center of gravity and a ground generator system. The kite is modelled using a six degree of freedom aircraft dynamics model. We first provide an extension of a previously developed attitude tracking theorem which shows the possibility of simultaneously tracking both the effective angle of attack and sideslip angle on the kite. A detailed aerodynamic model with empirical airfoil data and three dimensional wing tip effects is included. A feedback linearization controller is designed, and a baseline simulation is studied for confirming the tracking behavior.
Relationship between Antioxidants and Acrylamide Formation

Abstract:The relationship between antioxidants and acrylamide formation deserves investigation. Both positive and negative results were obtained. Antioxidants with different structures or functional groups can react with acrylamide precursors, reaction intermediates, or acrylamide itself, leading to either reducing or promoting effects. Acrylamide content, as the final result of both positive and negative reactions, depends on the predominant reaction that antioxidants and substrates undergo, which may change when the antioxidant concentration varies. The fact that the same antioxidant or that extract and its representative components behave discordantly might be due to the different reaction conditions among studies, the antioxidant concentrations, and the preparation methods. The relationship between antioxidant capacity and acrylamide inhibition remains controversial. A close correlation between ΔTEAC and acrylamide inhibition was later found. However, more in-depth studies are still needed.

Abstract:Microwave heating can be regarded as a possible way to produce a considerable amount of acrylamide. The present study investigated the formation of acrylamide in asparagine–glucose, asparagine–fructose and asparagine–sucrose microwave heating systems by the response surface methodology (RSM) and the orthogonal array methodology (OAM). The acrylamide content was rapidly quantified by a validated ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) method. Results of RSM study indicated that in the asparagine–glucose system, the acrylamide content increased in the combined condition of high temperature accompanying with short heating time (>190°C, <20min) or low temperature accompanying with long heating time (<180°C, >30min). In the asparagine–fructose system, the similar conclusion was made in the combined condition of high temperature accompanying with short heating time (>175°C, <20min) or low temperature accompanying with long heating time (<170°C, >25min). In the asparagine–sucrose system, the amount of acrylamide enhanced with the increase of both heating temperature and heating time. The fitted mathematic models were successfully applied to the quantification of acrylamide formation when the heating temperature and heating time fell into the ranges of 120–240°C and 5–35min simultaneously. OAM study showed that acrylamide is readily formed via heating binary precursors 5min at 180°C in the asparagine–glucose and asparagine–fructose systems. However, acrylamide is readily generated when the binary precursors are heated 15min at 180°C in the asparagine–sucrose system.

Abstract:Objective:To examine the effect of processing conditions on the formation of acrylamide in green tea.Methods:The quantitative analysis of acrylamide was conducted by liquid chromatography-triple quadrupole mass spectrometry (LC-TQ-MS).The optimization of the processing conditions with respect to fixation methods,rolling time and drying methods was investigated for reduced formation of acrylamide using an L9 (34) orthogonal array design.Results:Hot air drying temperature was found to have the greatest influence on acrylamide formation,followed sequentially by rolling time and fixing temperature,and the optimal values of these variables were 150 ℃,30 min and 150 ℃,respectively.Conclusions:Microwave treatment was more conducive to the formation of acrylamide in green tea than conventional heat treatment.More acrylamide was formed with increasing fixing temperature,and less acrylamide was formed with increasing drying temperature and rolling time.

Abstract:The recent report of elevated acrylamide levels in heat processing foods evoked an international health alarm. Acrylamide, an acknowledged potential genetic and reproductive toxin with mutagenic and carcinogenic properties in experimental mammalians, has been found in various heat processing foods. Many original contributions reported their findings on the formation mechanism and possible reduction methods of acrylamide. The aim of this review article is to summarize the state-of-the-art about the formation and reduction of acrylamide in the Maillard reaction. This research progress includes mechanistic studies on the correlation between the Maillard reaction and acrylamide, the formation mechanism of acrylamide, the main pathways of formation and impact factors on formation including cultivars, storage temperature, storage time, heat temperature, heat time, environmental pH, concentration of precursors, effects of food matrixes, type of oil, etc. Meanwhile, primary mechanisms on the reduction of acrylamide as well as reduction pathways including material and processing related ways and use of exogenous chemical additives are systematically reviewed. The mitigation studies on acrylamide are also summarized by the Confederation of the Food and Drink Industries of the EU (CIAA) "Toolbox" approach.

Abstract:The effect of dietary flavone C- and O-glycosides on the formation of acrylamide contaminants has been investigated in the present work. Flavone glycosides were added in different concentrations, and the acrylamide levels were quantified in a potato-based equimolar asparagine-reducing sugar model system via microwave heating. Results indicated a non-linear relationship between the addition levels of flavone glycosides and inhibitory rates of acrylamide formation in the Maillard reaction. The maximum inhibitory rate (25.3-63.5%) was observed when the addition levels of all flavone glycosides was 10(-9) mol L-1. A structure-activity analysis of the ability of flavonoids to reduce the formation of acrylamide revealed that both the number and position of phenol hydroxyl functional groups play an important role in the inhibition ability of flavonoids. Furthermore, flavone C-glycosides are more effective at inhibiting the formation of acrylamide than flavone O-glycosides, although they share the same aglycone structure. The rate of inhibition of acrylamide formation correlated well with changes in trolox equivalent antioxidant capacity (Delta TEAC) measured by DPPH (R-2 = 0.934), ABTS (R-2 = 0.897) or FRAP (R-2 = 0.912) assay. Using Delta TEAC as variables, a multiple linear regression (MLR) model could effectively serve as a predictive tool for estimating the reduction of acrylamide by flavone glycosides during microwave heat processing. These observations are important for our understanding and future development of agents that might decrease the formation of this hazardous toxin.

Abstract:The detection of high level of the neurotoxic and carcinogenic acrylamide in heat-treated foodstuffs in 2002 led to an intensive research effort. Among the parameters affecting the level of acrylamide in foods, the effect of antioxidants had not yet been elucidated satisfactorily. Several researchers have investigated the effect of various antioxidants and antioxidative extracts on acrylamide formation, but the data were discordant. Some studies claimed mitigation while others no effect or even an increase. It can be attributed to the ability of antioxidants with different structures or functional groups to react with acrylamide precursors, with intermediates of the reaction or with acrylamide itself, leading to either reducing or promoting effects. The fact that the same kind of antioxidant, or extract and its representative components behaves differently in different studies might be due to the different reaction conditions among the studies, concentrations of the antioxidant, as well as preparative methods of the extract. Effect of natural antioxidants on kinetic behavior of acrylamide formation and elimination in low-moisture asparagine–glucose model system

Abstract:The present study investigated the effect of two natural antioxidants, i.e. antioxidant of bamboo leaves (AOB) and extract of green tea (EGT), on the mitigation and the kinetic behavior of acrylamide in an equimolar asparagine–glucose model system (pH 6.80). The substrates with addition of AOB or EGT were oven-heated at 180°C under low-moisture conditions and the acrylamide content in final reaction products was quantified by ultra-performance liquid chromatography tandem mass spectrometry (UPLC–MS/MS). Results of mitigation effect on the generation of acrylamide showed that both AOB and EGT could effectively reduce the formation of acrylamide and achieve a maximum reduction rate (74.4% for the use of AOB and 74.3% for the use of EGT) when the addition levels of AOB and EGT were both 10−4mg. Such mitigation effect in the model system was further confirmed by addition of homoorientin and epigallocatechin gallate (EGCG), the representative components in AOB and EGT. To describe the kinetic behavior of acrylamide, a logistic-exponential model was selected among three kinetic model candidates while relative kinetic rate constants were calculated by the Marquardt non-linear regression method. Results indicated that there were significant differences on kinetic parameters (kg and tcg) between AOB/EGT-spiked and non-spiked model system studies, which suggested that addition of AOB or EGT could reduce the formation of acrylamide during the formation-predominant kinetic stage and prolong the formation progress of acrylamide in the low-moisture reaction system. However, such addition method was not able to reduce the acrylamide content during the elimination-predominant kinetic stage of acrylamide.
Coaggregation between Porphyromonas gingivalis and Treponema denticola.

Abstract:To elucidate an ecological profile of several periodontopathogens, the authors examined the coaggregation between cells of Porphyromonas gingivalis and oral bacterial strains including Treponema denticola in vitro. Coaggregation between cells of plaque bacteria was examined by visual assay and phase-contrast microscope. P. gingivalis cells coaggregated with strains of T. denticola and Treponema socranskii subspecies socranskii, but did not coaggregate with T. socranskii subspecies buccale, T. socranskii subspecies paredis, Treponema vincentii, or Treponema pectinovorum. The extracted hemagglutinin from P. gingivalis was active agglutinating T. denticola cells. Addition of serum and saliva somewhat affected the coaggregation, but no effects of tested sugars or amino acids were found. Heat treatment of T. denticola cells did not reduce the coagregation: heat treatment of P. gingivalis cells eliminated it. Growth inhibitory activity among these bacterial species was examined by the stab culture method. Strains of T. denticola ATCC 35404 and 35405 and T. vincentii inhibited the growth of some P. gingivalis strains, but not others. No strain of Treponema was inhibited by black-pigmented anaerobic rods. The coaggregation observed between P. gingivalis and T. denticola indicates the potential importance of their simultaneous existence in human periodontal pockets and development of the disease.
Towards P2P-based Computer Network Management *

Abstract:Nowadays, the rapid development of computer networks calls for highly distributed management technologies that aim at integration and automation. This paper then gives an overview of current management architectures for computer networks, and tries to apply P2P technologies to make up existing limitations in term of performance, scalability, flexibility, maintainability and reliability. Due to their characteristics, P2P technologies provide a prospective approach to implement inter-domain network management, moving towards cooperative management. Performance issues show that, appropriate manipulations especially by means of compressing the P2P traffic and P2P messages, are of great significance in utilizing P2P technologies for computer network management.

Abstract:We reported a structure–activity relationship study on unravelling phenolic hydroxyls instead of alcoholic hydroxyls contribute to the reduction of acrylamide formation by flavonoids. The dose-dependent study shows a close correlation between the number of phenolic hydroxyls of flavonoids and their reduction effects. In view of positions of hydroxyls, the 3′,4′(ortho)-dihydroxyls in B cycle, 3-hydroxyl or hydroxyls of 3-gallate in C cycle, and 5,7(meta)-dihydroxyls in A cycle of flavonoid structures play an important role in the reduction of acrylamide. Flavone C-glycosides are more effective at reducing the formation of acrylamide than flavone O-glycosides when sharing the same aglycone. The current multiplex quantitative structure–activity relationship (QSAR) equations effectively predict the inhibitory rates of acrylamide using selected chemometric parameters (R2: 0.835–0.938). This pioneer study opens a broad understanding on the chemoprevention of acrylamide contaminants on a structural basis.
Antioxidant-capacity-based Models for the Prediction of Acrylamide Reduction by Flavonoids.

Abstract:The aim of this study was to investigate the applicability of artificial neural network (ANN) and multiple linear regression (MLR) models for the estimation of acrylamide reduction by flavonoids, using multiple antioxidant capacities of Maillard reaction products as variables via a microwave food processing workstation. The addition of selected flavonoids could effectively reduce acrylamide formation, which may be closely related to the number of phenolic hydroxyl groups of flavonoids (R: 0.735-0.951, P < 0.001). The rate of inhibition of acrylamide formation correlated well with the change of trolox equivalent antioxi'dant capacity (Delta TEAC) measured by DPPH (R-2 = 0.833), ABTS (R-2 = 0.860) or FRAP (R-2 = 0.824) assay. Both ANN and MLR models could effectively serve as predictive tools for estimating the reduction of acrylamide affected by flavonoids. The current predictive model study provides a low-cost and easy-to-use approach to the estimation of rates at which acrylamide is degraded, while avoiding tedious sample pretreatment procedures and advanced instrumental analysis. Abstract:Study Design. Multicenter, prospective analysis of consecutive patients with adult spinal deformity (ASD). Objective. Identify age-related radiographical parameters associated with poor health-related quality of life (HRQOL) and treatment preferences for ASD. Summary of Background Data. Patients with ASD report discrepant severities of disability. Understanding age-associated differences for reported disability and treatment preferences may improve ASD evaluation and treatment. Methods. Baseline demographic, radiographical, and HRQOL values were evaluated in a multicenter, prospective cohort of consecutive patients with ASD. Inclusion criteria: ASD, age more than 18 years, and no prior spine surgery. Patients were grouped into those treated operatively (OP) or nonoperatively (NON) and stratified into 3 age groups: G1, 50 years or less; G2, 50 to 65 years; G3, 65 years or more. HRQOL measures included Scoliosis Research Society-22r questionnaire, Oswestry Disability Index, and Short Form-36 Health Survey. Results. Four hundred ninety-seven patients (OP = 156, NON = 341) with a mean age of 50.4 years met inclusion criteria. The OP group was older (53.3 vs. 49.0 yr), had larger scoliosis (49.3° vs. 43.3°), larger sagittal vertical axis (SVA, 33.2 vs. 13.7 mm), greater pelvic incidence-lumbar lordosis mismatch (6.6°vs. 3.1°), and worse HRQOL scores than the NON group, respectively (P < 0.05). Age stratification demonstrated worsening of SVA, spinopelvic alignment (SPA), and HRQOL scores with increasing age (P < 0.05). Age/treatment stratification demonstrated that younger OP had greater scoliosis than NON (G1OP = 49.9°vs. G1NON = 42.2°; G2OP = 56°vs. G2NON = 47.2°; P < 0.05) but similar SPA as NON. Older OP had similar scoliosis, but larger SVA than NON (G3OP = 100.6 vs. G3NON = 66.4 mm; P < 0.05). OP in all age groups reported worse HRQOL than NON (P < 0.05). Conclusion. Poor HRQOL uniformly determined operative treatment for ASD. Spinal deformities differed between age groups. Younger OP had larger scoliosis but similar SPA and SVA than NON. Older OP had similar scoliosis but worse SVA than NON. Age-associated differences for poor HRQOL must be considered when evaluating patients with ASD. Level of Evidence: 2

Abstract:The objectives of this study were to evaluate the effect of 0.5% chitosan incorporation on acrylamide development in a food model solution containing 0.5% fructose and asparagine after heating for 30 min at 180 °C. All the solutions were investigated for the following characteristics: acrylamide, asparagine, reducing sugar content, color, kinematic viscosity, Maillard reaction products (MRPs), and pH every 10 min. After heating for 10 min, the viscosity of chitosan-containing solutions reduced significantly. The investigational data confirmed that chitosan may have decomposed into lower molecular structures, as demonstrated by the reduced viscosity of the solution at pH < 6 and a decrease in the acrylamide content during 30 min of heating in a fructose–asparagine system. This study also confirms that the formation of ultraviolet-absorbing intermediates and browning intensity of MRPs containing acrylamide prepared by fructose–asparagine was more than those of MRPs prepared by glucose–asparagine solution system. MRPs containing acrylamide resulted from the reaction of asparagine with fructose (ketose) rather than glucose (aldose). Acrylamide formation could be significantly mitigated in the fructose–asparagine–chitosan model system as compared to the fructose–asparagine model system for possible beverage and food application.

Abstract:Acrylamide (AA) is known as a neurotoxin in humans and it is classified as a probable human carcinogen by the International Agency of Research on Cancer. AA is produced as by-product of the Maillard reaction in starchy foods processed at high temperatures (>120 °C). This review includes the investigation of AA precursors, mechanisms of AA formation and AA mitigation technologies in potato, cereal and coffee products. Additionally, most relevant issues of AA risk assessment are discussed. New technologies tested from laboratory to industrial scale face, as a major challenge, the reduction of AA content of browned food, while still maintaining its attractive organoleptic properties. Reducing sugars such as glucose and fructose are the major contributors to AA in potato-based products. On the other hand, the limiting substrate of AA formation in cereals and coffee is the free amino acid asparagine. For some products the addition of glycine or asparaginase reduces AA formation during baking. Since, for potatoes, the limiting substrate is reducing sugars, increases in sugar content in potatoes during storage then introduce some difficulties and potentially quite large variations in the AA content of the final product. Sugars in potatoes may be reduced by blanching. Levels of AA in different foods show large variations and no general upper limit is easily applicable, since some formation will always occur. Current policy is that practical measures should be taken voluntarily to reduce AA formation in vulnerable foods since AA is considered a health risk at the concentrations found in foods.

Pyrolytic acrylamide formation from purified wheat gluten and gluten-supplemented wheat bread rolls

Abstract:Recent studies have revealed different acrylamide formation mechanisms, e. g. from carnosine (N-beta-alanyl-L-histidine) and aminopropionamide as additional precursors. The occurrence of acrylamide in food matrices devoid of common precursors such as meat supports an additional formation pathway. Gluten was recovered from wheat flour by water extraction. Starch, reducing sugars and amino acids were removed using alpha-amylase and NaCl solution and were completely absent in the purified gluten fraction. The gluten was dry heated at temperatures ranging from 160 to 240 degrees C for 8 to 12 min and analyzed for acrylamide and cinnamic amide using liquid chromatography-tandem mass spectrometry. Acrylamide could be detected up to 3997 microg/kg gluten dry weight. Cinnamic amide was detected and unambiguously identified in the gluten samples, thus confirming the proposed formation of acrylamide from proteins. After gluten addition to bread roll dough, protein pyrolysis to form acrylamide in the complex food matrix was assessed. Contents of asparagine and reducing sugars were diminished due to the addition of the gluten. In contrast to the expectation with respect to the well-established common formation mechanism of acrylamide, it increased from 53.4 to 63.9 microg/kg (+20%), which was in good correlation with the higher proportion of gluten. As demonstrated by the t-test, the increase in acrylamide was significant when comparing 0 and 15% gluten addition. Additionally, cinnamic amide could be found in crusts of bread rolls. Thus, evidence for pyrolytic formation of acrylamide from wheat gluten was provided.

Abstract:Acrylamide is a monomer which is associated with asparagine especially when proteins react to carbohydrate during Maillard reaction. The issue of this carcinogen was highlighted by Swedish Nation Food Administration SNFA and the cause progressed by European food industries, especially spiked the high treatment to high saccharon foods. Experiments replicated to find end products of trio effects of high saccharides, free protein subunits the aminoacids and high temperature, revealing formation of acrylamide in research institutes and snack food industries. Mini review highlights formation, consumption, and health impacts of acrylamide.
