Abstract:Triclosan (TCS), a kind of pharmaceuticals and personal care products (PPCPs), is widely used and has had a large production over years. It is an emerging pollutant in the water environment that has attracted global attention due to its toxic effects on organisms and aquatic ecosystems, and its concentrations in the water environment are expected to increase since the COVID-19 pandemic outbreak. Some researchers found that microbial degradation of TCS is an environmentally sustainable technique that results in the mineralization of large amounts of organic pollutants without toxic by-products. In this review, we focus on the fate of TCS in the water environment, the diversity of TCS-degrading microorganisms, biodegradation pathways and molecular mechanisms, in order to provide a reference for the efficient degradation of TCS and other PPCPs by microorganisms.

Abstract:Triclosan (TCS), a halogenated aromatic hydrocarbon, is a highly persistent personal care product (PCP) with a varying concentration range, i.e. ng*L −1 − μg*L −1 in environmental matrices. TCS exhibits lethal effects on the environmental ecosystem and human health due to its antimicrobial resistance, carcinogenic effects, bioaccumulation, biomagnification, and endocrine disruptive activities. Since the sudden occurrence of the novel coronavirus disease 2019 (COVID-19), the worldwide production rate of TCS is very high because of the large requirement for disinfection, which may have led to its elevated concentration. Thus, the monitoring and regulation of TCS are of utmost importance from the perspective of its potential damage. The current review article deliberates on its persistence in aqueous and solid systems as well as its toxicological impact on algal, vertebrate, and invertebrate systems. The advanced monitoring techniques for detection up to nanogram levels, such as chromatographic techniques, electrochemical, optical, and electrophoresis methods, etc., have been discussed. The negative impacts of TCS on the environment raised a great concern about developing waste water treatment methods because of the inefficacy of conventional methods. The review also focuses on the removal and remediation techniques so as to develop better research directions for phasing out TCS.

Abstract:Triclosan (TCS), a lipophilic broad-spectrum biocide is widely used in personal care, acrylic, veterinary, medical and household products. It has been observed to be present in aquatic environments, animal and plant tissues around the world, and even in human blood, urine and breast milk. Under natural conditions, TCS degrades photolytically as well as through microbial action into more persistent and toxic byproducts like dioxins. Moreover, accumulation in deep water bodies or soil strata where light is not adequately available makes its degradation even more prolonged. Present review has been undertaken with an objective to highlight the concerns surrounding TCS exposure to aquatic organisms, the infiltration routes into the food chain, its persistence and accumulation, teratogenic, biochemical and cytogenic effects on a wide range of aquatic species. The widespread use of products containing TCS and potential toxicity at lethal concentrations makes it a compound of utmost concern worldwide and hence its use under permissible levels, proper disposal needs to be regulated.

Abstract:Triclosan (TCS) is an antibacterial agent commonly used in personal care products. Due to its widespread use and improper disposal, it is also a pervasive contaminant, particularly in aquatic environments. When released into water bodies, TCS can induce deleterious effects on developmental and physiological aspects of aquatic organisms and also interact with environmental stressors such as weather, metals, pharmaceuticals, and microplastics. Multiple studies have described the adverse effects of TCS on aquatic organisms, but few have reported on the interactions between TCS and other environmental conditions and pollutants. Because aquatic environments include a mix of contaminants and natural factors can correlate with contaminants, it is important to understand the toxicological outcomes of combinations of substances. Due to its lipophilic characteristics, TCS can interact with a wide range of substances and environmental stressors in aquatic environments. Here, we identify a need for caution when using TCS by describing not only the effects of exposure to TCS alone on aquatic organisms but also how toxicity changes when it acts in combination with multiple environmental stressors.

Abstract:The increasing discharge of pharmaceuticals and personal care products (PPCPs) into the environment has generated serious public concern. The recent awareness of the environmental impact of this emerging class of pollutants and their potential adverse effects on human health have been documented in many reports. However, information regarding uptake and intracellular distribution of PPCPs in hydrophytes under hydroponic conditions, and potential human exposure is very limited. A laboratory experiment was conducted using C-14-labeled triclosan (TCS) to investigate uptake and distribution of TCS in six aquatic plants (water spinach, purple perilla, cress, penny grass, cane shoot, and rice), and the subcellular distribution of C-14-TCS was determined in these plants. The results showed that the uptake and removal rate of TCS from nutrient solution by hydrophytes followed the order of cress (96%) > water spinach (94%) > penny grass (87%) > cane shoot (84%) > purple perilla (78%) > rice (63%) at the end of incubation period (192 h). The range of C-14-TCS content in the roots was 94.3%-99.0% of the added C-14-TCS, and the concentrations in roots were 2-3 orders of magnitude greater than those in shoots. Furthermore, the subcellular fraction-concentration factor (3.6 x 10(2)-2.6 x 10(3) mL g(-1)), concentration (0.58-4.47 mu g g(-1)), and percentage (30%-61%) of C-14-TCS in organelles were found predominantly greater than those in cell walls and/or cytoplasm. These results indicate that for these plants, the roots are the primary storage for TCS, and within plant cells organelles are the major domains for TCS accumulation. These findings provide a better understanding of translocation and accumulation of TCS in aquatic plants at the cellular level, which is valuable for environmental and human health assessments of TCS. Abstract:The halogenated antimicrobial triclocarban (TCC) has large production and consumption over last decades. Its extensive utilization in personal care products and insufficient treatment in conventional wastewater treatment plants (WWTPs) has led to its listing as one of emerging organic contaminants (EOCs). Due to the hydrophobicity and chemical stability of TCC, it has been omnipresent detected in terrestrial and aquatic environments, and its prolonged exposure has thrown potential pernicious threat to ecosystem and human health. Considering its recalcitrance, especially under anoxic conditions, both biological and non-biological methods have been exploited for its removal. The efficiency of advanced oxidation processes was optimistic, but complete removal can rarely be realized through a single method. The biodegradation of TCC either with microbial community or pure culture is feasible but efficient bacterial degraders and the molecular mechanism of degradation need to be further explored. This review provides comprehensive information of the occurrence, potential ecological and health effects, and biological and non-biological removal of TCC, and outlines future prospects for the risk evaluation and enhanced bioremediation of TCC in various environments.

Abstract:Triclosan (TCS) is an anti-microbial widely used in personal care and medical antibacterial products. Despite the widespread occurrence of TCS in municipal sewage sludge, understanding toward the fate of TCS within sewage treatment and environmental risks in the eventual land application is still limited. This review summarizes the TCS loads and transfer mechanisms in the sewage treatment process, sludge management process, land application, and its potential environmental impacts. TCS transfer from sewage to sludge mainly occurs in the primary sedimentation process, representing 2.50 to 4.58 times more compared to the secondary sedimentation process. This transfer is facilitated through adsorption because of the presence of humic acid-like and protein-like substances in sludge. Both anaerobic digestion and aerobic composting contribute to the degradation of TCS with aerobic composting being more effective, exhibiting TCS degradation rates 1.04–2.87 times higher than those observed in anaerobic digestion. After sludge land application, TCS majorly dissipates in the soil through biodegradation by fungi and bacteria, potentially posing environmental risks, such as inhibiting the seedling growth of plant species. Additionally, the degradation of TCS, coupled with the formation and subsequent degradation of MeTCS, is observed, with MeTCS exhibiting a higher half-life and greater toxicity than its parent compound (TCS). Overall, this research offers vital insights to enhance understanding of TCS's migration and degradation processes in sewage treatment and soil. It also provides guidance in environmental protection and sustainable resource management.

Abstract:Triclosan [5-chloro-2-(2,4-dichlorophenoxy)phenol; TCS] is a broad spectrum antibacterial agent used in personal care, veterinary, industrial and household products. TCS is commonly detected in aquatic ecosystems, as it is only partially removed during the wastewater treatment process. Sorption, biodegradation and photolytic degradation mitigate the availability of TCS to aquatic biota; however the by-products such as methyltriclosan and other chlorinated phenols may be more resistant to degradation and have higher toxicity than the parent compound. The continuous exposure of aquatic organisms to TCS, coupled with its bioaccumulation potential, have led to detectable levels of the antimicrobial in a number of aquatic species. TCS has been also detected in breast milk, urine and plasma, with levels of TCS in the blood correlating with consumer use patterns of the antimicrobial. Mammalian systemic toxicity studies indicate that TCS is neither acutely toxic, mutagenic, carcinogenic, nor a developmental toxicant. Recently, however, concern has been raised over TCS's potential for endocrine disruption, as the antimicrobial has been shown to disrupt thyroid hormone homeostasis and possibly the reproductive axis. Moreover, there is strong evidence that aquatic species such as algae, invertebrates and certain types of fish are much more sensitive to TCS than mammals. TCS is highly toxic to algae and exerts reproductive and developmental effects in some fish. The potential for endocrine disruption and antibiotic cross-resistance highlights the importance of the judicious use of TCS, whereby the use of TCS should be limited to applications where it has been shown to be effective.

Abstract:Triclosan (TCS) is a multi-purpose antimicrobial agent used as a common ingredient in everyday household personal care and consumer products. The expanded use of TCS provides a number of pathways for the compound to enter the environment and it has been detected in sewage treatment plant effluents; surface; ground and drinking water. The physico-chemical properties indicate the bioaccumulation and persistence potential of TCS in the environment. Hence, there is an increasing concern about the presence of TCS in the environment and its potential negative effects on human and animal health. Nevertheless, scarce monitoring data could be one reason for not prioritizing TCS as emerging contaminant. Conventional water and wastewater treatment processes are unable to completely remove the TCS and even form toxic intermediates. Considering the worldwide application of personal care products containing TCS and inefficient removal and its toxic effects on aquatic organisms, the compound should be considered on the priority list of emerging contaminants and its utilization in all products should be regulated.

Abstract:Contaminants of emerging concern (CECs) including endocrine disrupting compounds (EDCs) and pharmaceuticals and personal care product chemicals (PPCPs) have recently received more attention because of their occurrence in water bodies and harmful impacts on human health and aquatic organisms. Triclosan is widely used as a synthetic broad-spectrum antimicrobial agent due to its antimicrobial efficacy. However, triclosan detected in aquatic environment has been recently considered as one of CECs, because of the potential for endocrine disruption, the formation of toxic by-products and the development of cross-resistance to antibiotics in aquatic environment. This comprehensive review focuses on the regulations, toxicology, fate and transport, occurrence and removal efficiency of triclosan. Overall, this review aims to provide better understanding of triclosan and insight into application of biological treatment process as an efficient method for triclosan removal.

Abstract:The angular distribution of myosin cross-bridges in muscle fibers was investigated in four physiological states using a multiple probe analysis of varied extrinsic probes of the cross-bridge [Burghardt & Ajtai (1994) Biochemistry (preceding paper in this issue)]. The analysis combines data of complementary techniques from different probes giving the highest possible angular resolution. Four extrinsic probes of the fast reactive sulfhydryl (SH1) on myosin subfragment 1 (S1) were employed. Electron paramagnetic resonance (EPR) spectra from paramagnetic probes, deuterium- and 15N-substituted for greater sensitivity to orientation, on S1 were measured when the protein was freely tumbling in solution and when it was decorating muscle fibers. The EPR spectra from labeled S1 tumbling in solution were measured at X- and Q-band microwave frequencies to uniquely specify the orientation of the probe relative to the S1 principal hydrodynamic frame. The EPR spectra from labeled S1 decorating muscle fibers in rigor and in the presence of MgADP were measured at X-band and used in the multiple probe analysis of cross-bridge orientation. The time-resolved fluorescence anisotropy decay (TRFAD) of fluorescent probes on S1 was measured when the protein was freely tumbling in solution, and fluorescence polarization (FP) intensities from fluorescent probes modifying SH1 in intact muscle fibers were measured for fibers in rigor, in the presence of MgADP, in isometric contraction, and in relaxation at low ionic strength. The TRFAD measurements limit the range of possible orientations of the probe relative to the S1 principal hydrodynamic frame. The FP intensity measurements were used in the multiple probe analysis of cross-bridge orientation. The combination of the EPR and FP data determined a highly resolved cross-bridge angular distribution in rigor, in the presence of MgADP, in isometric contraction, and in relaxation at low ionic strength. These findings confirm earlier observations of a rigid body rotation of the SH1 region in the myosin head group upon physiological state changes and indicate the path and extent of cross-bridge rotation during contraction. The rotation of the cross-bridge is visualized with computer-generated space-filling models of actomysin in six states of the contraction cycle.

Abstract:Due to the widespread use of tetracyclines antibiotics (TCs) in livestock breeding, TCs has always caused soil pollution. TCs cannot only enter soil and water bodies through multiple channels but also accumulate in them over a long period. It is more abundant in soils, agricultural land. Primarily, should explore efficient methods for removing or degrading TCs. This review provides an overview of the birth, development and problem of soil contamination of TCs. Then focuses on the toxicity and effects of TCs on microorganisms, animals, and plants. Furthermore, in the following chapters, microbial -, plant -, animal – biodegradation methods are introduced to summarize the species and plants applied in soil TCs removal at this stage. Meanwhile, the principle mechanism of advanced oxidation processes is analyzed deeply to fill the gap. In addition, a case of microbial fuel cells based on sustainable bioenergetics with promising prospects for soil TCs removal is presented. Due to the interaction between carbonaceous materials and soil, the characteristics of adsorption method under soil environment are deeply analyzed.

Abstract:Triclosan (TCS), an antimicrobial agent extensively incorporated into pharmaceuticals and personal care products, poses significant environmental risks because of its persistence and ecotoxicity. So far, a few microorganisms were suggested to degrade TCS, but the microbial degradation mechanism remains elusive. Here, a two-component angular dioxygenase (TcsAaAb) responsible for the initial TCS degradation was characterized in Sphingomonas sp. strain YL-JM2C. Whole-cell biotransformation and crude enzyme assays demonstrated that TcsAaAb catalyzed the conversion of TCS to 4-chlorocatechol and 3,5-dichlorocatechol rather than the commonly suggested product 2,4-dichlorophenol. Then two intermediates were catabolized by tcsCDEF cluster via an ortho -cleavage pathway. Critical residues (N262, F279, and F391) for substrate binding were identified via molecular docking and mutagenesis. Further, TcsAaAb showed activity toward methyl triclosan and nitrofen, suggesting its versatile potential for bioremediation. In addition, TCS-degrading genes were also present in diverse bacterial genomes in wastewater, ocean and soil, and a relatively high gene abundance was observed in marine metagenomes, revealing the transformation fate of TCS in environments and the microbial potential in pollutant removal. These findings extend the understanding of the microbe-mediated TCS degradation and contribute to the mining of TCS-degrading strains and enzymes, as well as their application in the bioremediation of contaminated environments.

Abstract:Tetracycline antibiotics (TCs) are widely used all over the world in recent decades. TCs are a family used as broadspectrum antibiotics and animal veterinary drugs. Among the TCs, tetracycline (TC) is the most use. Due to the rapid development of antibiotics industry, the dosage standards of TC are not yet clearly defined in most countries and regions. TC is hard to degrade in living organisms and can even be converted to more toxic substances. The overuse and wanton discharge of TC, also caused serious pollution of groundwater. This article attempts to summarize the latest knowledge on the nature, sources, pollution status, the impact on water environment toxicity of TC respectively. Meanwhile, there are many technologies to remove TC. This paper mainly included 12 kinds of degradation methods, including photodegradation, microbial removal, adsorption, electrochemical and sludge digestion. This review will provide a reference for the study of the basic properties and removal methods of TC.

Abstract:Triclosan (TCS) is an antibacterial and antifungal agent widely used in personal care products, and it has been frequently detected in the aquatic environment. In the present study, the acute toxicity of TCS to Daphnia magna, Photobacterium phosphoreum, Danio rerio, and Limnodrilus hoffmeisteri was assessed under different pH conditions. Generally, TCS was more toxic to the four aquatic organisms in acidic medium. The LC50 values for D. magna and D. rerio were smaller among the selected species, suggesting that D. magna and D. rerio were more sensitive to TCS. In addition, the oxidative stress-inducing potential of TCS was evaluated in Carassius auratus at three pH values. Changes of superoxide dismutase (SOD) and catalase (CAT) activity, glutathione (GSH) level, and malondialdehyde (MDA) content were commonly observed in all TCS exposure groups, indicating the occurrence of oxidative stress in the liver of C. auratus. The integrated biomarker response (IBR) index revealed that a high concentration of TCS induced great oxidative stress in goldfish under acidic condition. This work supplements the presently available data on the toxicity data of TCS, which would provide some useful information for the environmental risk assessment of this compound.

Abstract:Triclosan (TCS), a broad-spectrum antimicrobial agent, has been widely used in personal care products, medical products, plastic cutting boards, and food storage containers. Colgate Total® toothpaste, containing 10 mM TCS, is effective in controlling biofilm formation and maintaining gingival health. Given its broad usage, TCS is present ubiquitously in the environment. Given its strong lipophilicity and accumulation ability in organisms, it is potentially harmful to biohealth. Several reports suggest the toxicity of this compound, which is inserted in the class of endocrine disrupting chemicals (EDCs). In September 2016, TCS was banned by the U.S. Food and Drug Administration (FDA) and the European Union in soap products. Despite these problems, its application in personal care products within certain limits is still allowed. Today, it is still unclear whether TCS is truly toxic to mammals and the adverse effects of continuous, long-term, and low concentration exposure remain unknown. Indeed, some recent reports suggest the use of TCS as a repositioned drug for cancer treatment and cutaneous leishmaniasis. In this scenario it is necessary to investigate the advantages and disadvantages of TCS, to understand whether its use is advisable or not. This review intends to highlight the pros and cons that are associated with the use of TCS in humans.

Abstract:<p>Triclocarban (TCC), a typical emerging contaminant, was abundantly released into environment and frequently detected in practical wastewater treatment plants. However it is also an important material when being added to personal skin care products as a antibacterial agent. In this work, the behavior of TCC in wastewater treatment process was investigated. Experiments showed that ~82% of influent TCC was removed by activated sludge adsorption and its adsorption isotherm was well fitted with Linear model and Freundich model. High levels of TCC had seriously impact on the settleability, dewaterability and extracellular polymetric substance (EPS) of activated sludge, even on effluent turbidity after a long-term exposure. Furthermore, the performance of biological wastewater treatment was damaged by TCC long-term exposure as well. The removal rates of total nitrogen and phosphorus decreased from 91.2 ± 2.1% to 72.6 ± 2.2% and from 94.7 ± 3.1% to 78.4 ± 2.3%, respectively, with TCC level increasing from 0 to 100 μg/L. Mechanism analysis showed that TCC exposure significantly inhibited the relevant biological processes, such as ammonia oxidation, denitrification, phosphorus release and uptake, which were closely relevant to nitrogen and phosphorus removal.</p>

Abstract:The release of tetracyclines (TCs) in the environment is of significant concern because the residual antibiotics may promote resistance in pathogenic microorganisms, and the transfer of antibiotic resistance genes poses a potential threat to ecosystems. Microbial biodegradation plays an important role in removing TCs in both natural and artificial systems. After long-term acclimation, microorganisms that can tolerate and degrade TCs are retained to achieve efficient removal of TCs under the optimum conditions (e.g. optimal operational parameters and moderate concentrations of TCs). To date, cultivation-based techniques have been used to isolate bacteria or fungi with potential degradation ability. Moreover, the biodegradation mechanism of TCs can be unveiled with the development of chemical analysis (e.g. UPLC-Q-TOF mass spectrometer) and molecular biology techniques (e.g. 16S rRNA gene sequencing, multi-omics sequencing, and whole genome sequencing). In this review, we made an overview of the biodegradation of TCs in different systems, refined functional microbial communities and pure isolates relevant to TCs biodegradation, and summarized the biodegradation products, pathways, and degradation genes of TCs. In addition, ecological risks of TCs biodegradation were considered from the perspectives of metabolic products toxicity and resistance genes. Overall, this article aimed to outline the research progress of TCs biodegradation and propose future research prospects.
