Abstract:In this work, we report the fabrication and functional demonstration of a kind of dually responsive nanoparticles (NPs) as a potential drug delivery vector. The pH value, corresponding to the acidic microenvironment at the tumor site, and mannitol, to the extracellular trigger agent, were employed as the dually responsive factors. The function of dual responses was achieved by breaking the dynamic covalent bonds between phenylboronic acid (PBA) groups and diols at low pH value (pH 5.0) and/or under the administration of mannitol, which triggered the decomposition of the complex NPs and the concomitant release of anticancer drug of doxorubicin (DOX) loaded inside the NPs. The NPs were composed of modified chitosan (PQCS) with quaternary ammonium and PBA groups on the side chains, heparin (Hep), and poly(vinyl alcohol) (PVA), in which quaternary ammonium groups offer the positive charge for the cell-internalization of NPs, PBA groups serve for the formation of dynamic bonds in responding to pH change and mannitol addition, PVA furnishes the NPs with diol groups for the interaction with PBA groups and the formation of dynamic NPS, and Hep plays the roles of reducing the cytotoxicity of highly positively-charged chitosan and forming of complex NPs for DOX up-loading. A three-step fabrication process of drug-loaded NPs was described, and the characterization results were comprehensively demonstrated. The sustained drug release from the drug-loaded NPs displayed obvious pH and mannitol dependence. More specifically, the cumulative DOX release was increased more than 1.5-fold at pH 5.0 with 20 mg mL−1 mannitol. Furthermore, the nanoparticles were manifested with effective antitumor efficient and apparently enhanced cytotoxicity in response to the acidic pH value and/or mannitol.
β-Cyclodextrin-based supramolecular nanoparticles: a pH-sensitive nanocarrier for the sustained release of the anti-tumor drug

Abstract:Chemotherapy is one of the effective ways to treat cancer because of its ability to control lesions quickly. However, chemotherapy can have many side effects due to chemotherapy indiscriminately killing cancer cells as well as normal cells. Targeted chemotherapy strategies may be able to improve this problem effectively. In this work, a pH-responsive nanoparticle based on sulfobutyl ether-β-CD (SBE7-β-CD) and cetylbenzyldimethylammonium chloride (HDBAC) were designed and prepared. The obtained nanoparticles showed assemble and disassemble behaviors with the pH alternating between 10.0 and 2.0, which were adopted to encapsulate and release the anti-tumor drug celastrol (CSL). Moreover, CSL-loaded nanoparticles exhibited sustained release action throughout 82 h, which suggests it may possess the potential to improve therapeutic effects. In cell experiments, CSL-loaded nanoparticles displayed fewer cytotoxic effects on healthy cells, while significant cytotoxicity on five human cancer cells. Notably, CSL-loaded nanoparticles showed a higher apoptosis rate on liver cancer SMMC-7721cells than free CSL. We hope that the nanoparticles can be clinically used as a sustained-release carrier for anticancer drugs after further research.

Abstract:Controlled drug release systems are pivotal in optimizing therapeutic outcomes and mitigating side effects in treatment protocols. While traditional delivery vectors such as liposomes, micro/nanoparticles, and microspheres are effective, they often struggle with consistency in drug release rates. This study addresses these issues by integrating stimuli-responsive elements specifically magnetic, thermal, and pH-responsive components into drug delivery systems for precise control. Central to our approach is the use of silk fibroin (SF), chosen for its superior biocompatibility and tunable degradation kinetics. We developed uniform carrier microspheres (CMs) by embedding polydopamine nanoparticles (PDA NPs) into SF microspheres using a custom-designed microfluidic platform. The development process and the application of this platform are detailed, highlighting the precision in control achievable. These CMs showcased enhanced photothermal effects, with the thermal response finely adjustable by altering the PDA NPs concentration, achieving a notable temperature increase of 24.5°C at 7.4 wt% concentration. High drug loading capacity (7.5%) and encapsulation efficiency (91.6%) were achieved, along with a pH-responsive release profile under near-infrared irradiation, paving the way for targeted anticancer drug delivery systems using the model drug doxorubicin hydrochloride. These findings underscore the potential of the developed CMs for external topical application, offering promising prospects for targeted cancer therapy utilizing drug-loaded microspheres.

Abstract:To efficiently deliver anti-cancer drug to tumor site and reduce its toxic side effects on normal tissues, a polyethylene glycol (PEG) shielding and tumor microenvironment triggering cascade pH-responsive hollow mesoporous silica nanoparticles (HMSNs) drug delivery system was fabricated. 3-(3, 4-dihydroxyphenyl) propionic acid (DHPA) functionalized beta-cyclodextrin (β-CD) was grafted onto the surfaces of HMSNs via boronic acid-catechol ester bonds. Then, PEG conjugated adamantane (Ada) was anchored on HMSNs-β-CD nanocarrier via host-gust interaction. Various techniques proved the successful fabrication of the system. The in vitro tests confirmed that the system was biocompatible. After the system permeating into tumor via enhanced permeability and retention (EPR) effect, the benzoic-imine bonds between the PEG and Ada were cleaved under weak acid condition in tumor microenvironment (pH 6.8), while the dissociated PEG protective layer facilitating cellular uptake of HMSNs system. Subsequently, the boronic acid-catechol ester bonds linkers further hydrolyzed under even low endosomal pH (4.5-6.5) condition for intracellular drug delivery, leading to efficient cell apoptosis. The in vivo results demonstrated that drug loaded HMSNs significantly inhibited tumor growth while only with minimal toxic side effects. The strategy provides new insight into the development of new generation of drug delivery carriers triggering by tumor microenvironment.
Cytochrome C Capped Mesoporous Silica Nanocarriers for Ph-Sensitive and Sustained Drug Release

Abstract:In this paper, pH-responsive drug nanocarriers based on mesoporous silica nanoparticles (MSNs) capped with a natural, nontoxic protein cytochrome C (CytC) are designed and demonstrated for cancer therapy. At neutral pH the positively charged CytC can prevent the premature release of a preloaded anti-cancer drug. The results show that the CytC capped nanocarriers have excellent doxorubicin (DOX) loading efficiency (414 μg mg-1 MSN) and the leakage of the drug is only 16% at pH 7.4 phosphate-buffered saline for 72 h. Simultaneously, the DOX release percentage can reach 54% by decreasing the pH to 5.5. In contrast, unsealed MSNs show a fast DOX release rate at pH 7.4 and a slight pH-response. Confocal laser scanning microscopy demonstrates that the nanocarriers can enter human breast cancer MCF-7 cells and the DOX is sustained released from the drug carriers. Cytotoxicity tests and histological assays confirm that the constructed CytC capped nanocarriers possess lower toxicity than free DOX and unsealed drug carriers. Furthermore, intratumoral administration of the nanocarriers is significantly more efficacious in tumor reduction than free DOX and unsealed drug carriers in the xenograft models of MCF-7 cancer. Overall, this study demonstrates new drug nanocarriers with pH-sensitive and sustained drug release properties by using natural and nontoxic proteins as pore blockers to achieve highly efficient cancer treatment.

Abstract:We prepared novel DOX-loaded HMSNs coated with PDA and PEGMA.High loading capacity and entrapment efficiency were due to incorporation of HMSNs.The material exhibited pH and GSH mediated DOX controlled release profiles.HMSNs function as drug storage platforms and PDA and PEGMA act as gatekeepers.The drug carriers have great potentials to mediate drug release for cancer therapy.The development of multifunctional nano-drug carriers has become one of the most effective and prevailing approaches to overcome the non-selectivity and low cell uptake efficiency of drugs, as well as various side effects of traditional chemotherapy drugs. Herein, we reported novel hollow mesoporous silica nanoparticles (HMSNs) coated with polydopamine (PDA) and poly(poly(ethylene glycol) methacrylate) (PPEGMA) (PPEGMA) for the treatment of cancer. The HMSNs mainly served as a warehouse loading anticancer drug doxorubicin (DOX); the covering of PDA acted as pH-sensitive gatekeepers to control the diffusion and/or release of DOX from HMSNs in different pH stimuli environments, whilst the grafting of PPEGMA on the surface of by disulfide bonds and atom transfer radical polymerization (ATRP) increased the stability under physiological conditions and showed reductant sensitive responsiveness. The PPEGMA had a high loading capacity and entrapment efficiency of drugs through loading twice but low cytotoxicity. The in vitro release results indicated that the PPEGMA exhibited a high sensitivity to pH and glutathione (GSH) due to the PDA coating and disulfide bonds, which could control DOX release as intelligent "switches". The drug-loaded formulations could better be taken up by the cells, formed a higher concentration of DOX in the cells, and produced more remarkable apoptosis, thus achieving better treatment of tumors with high therapy efficacy. Therefore, the developed multifunctional materials could be used as a promising nano-drug carrier for cancer treatment.

Abstract:BACKGROUND: Although pH and redox sensitiveness have been extensively investigated to improve therapeutic efficiency, the effect of disulfide bonds location and pH-triggered charge-reversal on cascade-targeting still need to be further evaluated in cancer treatment with multi-responsive nanoparticles.PURPOSE: The aim of this study was to design multi-responsive DOX@MSNs-COS-NN-CMC, DOX@MSNs-COS-SS-CMC and DOX@MSNs-COS-CMC-SS and systematically investigate the effects of disulfide bonds location and charge-reversal on the cancer cell specificity, endocytosis mechanisms and antitumor efficiency.RESULTS: In vitro drug release rate of DOX@MSNs-COS-SS-CMC in tumor environments was 7-fold higher than that under normal physiological conditions after 200 h. Furthermore, the fluorescence intensity of DOX@MSNs-COS-SS-CMC and DOX@MSNs-COS-CMC-SS was 1.9-fold and 1.3-fold higher than free DOX at pH 6.5 and 10 mM GSH. In addition, vesicular transport might be a factor that affects the uptake efficiency of DOX@MSNs-COS-SS-CMC and DOX@MSNs-COS-CMC-SS. The clathrin-mediated endocytosis and endosomal escape of DOX@MSNs-COS-SS-CMC enhanced cellular internalization and preserved highly controllable drug release into the perinuclear of HeLa cells. DOX@MSNs-COS-SS-CMC exhibited a synergistic chemotherapy in preeminent tumor inhibition and less side effects of cardiotoxicity.CONCLUSION: The cascade-targeting of charge-reversal and disulfide bonds shielding would be a highly personalized strategy for cervical cancer treatment.

A pH-Responsive Nanoplatform Based on Fluorescent Conjugated Polymer Dots for Imaging-Guided Multitherapeutics Delivery and Combination Cancer Therapy

Abstract:For cancer treatment, nanocarriers were designed with cationic lipids and polymers to improve the cytosolic delivery efficiency of siRNA. Though the positively charged nanocarriers showed great potential for RNA therapy, it was inevitable to generate the potential cytotoxicity. We constructed a pH-responsive nanoplatform, which co-carried siRNA and anticancer drug (hydroxycamptothecine, HCPT), to integrate gene therapy and chemotherapy for combination cancer therapy. The fluorescent conjugated polymer nanoparticles (CPNPs) modified with cell-penetrating peptides were employed as cores to carry siRNA molecules (siRNA-CPNPs) and track the biodistribution of nanotherapeutics by virtue of fluorescence. Calcium phosphate (CaP) nanocoatings were deposited on the surface of siRNA-CPNPs, followed by loading with HCPT and aptamers targeting cancer cells to obtain a targeted and tumor acid-responsive biocompatible nanoplatform. After the uptake of cancer cells, the CaP nanocoatings were decomposed in the acidic endo/lysosomes to release HCPT, and the siRNA-CPNPs were exposed to facilitate the siRNA endo/lysosome escape and cytoplasm delivery. Results obtained from both in vitro and in vivo studies in tumor inhibition expressed that the combined therapy exhibited a better therapeutic efficacy than any monotherapy.

Abstract:Nanodrug delivery systems (NDSs), such as mesoporous silica, have been widely studied because of their high specific surface area, high loading rate, and easy modification; however, they are not easily metabolized and excreted by the human body and may be potentially harmful. Hence, we aimed to examine the synergistic anti-tumor effects of ex vivo chemo-photothermal therapy to develop a rational and highly biocompatible treatment protocol for tumors. We constructed a biodegradable NDS using organic mesoporous silica with a tetrasulfide bond structure, copper sulfide core, and folic acid-modified surface (CuS@DMONs-FA-DOX-PEG) to target a tumor site, dissociate, and release the drug. The degradation ability, photothermal conversion ability, hemocompatibility, and in vitro and in vivo anti-tumor effects of the CuS@DMONs-FA-DOX-PEG nanoparticles were evaluated. Our findings revealed that the nanoparticles encapsulated in copper sulfide exhibited significant photothermal activity and optimal photothermal conversion rate. Further, the drug was accurately delivered and released into the target tumor cells, annihilating them. This study demonstrated the successful preparation, safety, and synergistic anti-tumor effects of chemo-photothermal therapeutic nanomaterials.

Abstract:The majority of current nanocarriers in cancer treatment fail to deliver encapsulated cargos to their final targets at therapeutic levels, which decreases the ultimate efficacy. In this work, a novel core–shell nanocarrier with a biodegradable property was synthesized for efficient drug release and subcellular organelle delivery. Initially, silver nanoparticles (AgNPs) were grafted with terminal double bonds originating from N, N′-bisacrylamide cystamine (BAC). Then, the outer coatings consisting of chitosan (CTS) and polyvinyl alcohol (PVA) were deposited on the surface of modified AgNPs using an emulsion method. To improve the stability, disulfide-containing BAC was simultaneously reintroduced to cross-link CTS. The as-prepared nanoparticles (CAB) possessed the desired colloidal stability and exhibited a high drug loading efficiency of cationic anticancer agent doxorubicin (DOX). Furthermore, CAB was tailored to transform their size into ultrasmall nanovehicles responding to weak acidity, high glutathione (GSH) levels, and overexpressed enzymes. The process of transformation was accompanied by sufficient DOX release from CAB. Due to the triple sensitivity, CAB enabled DOX to accumulate in the nucleus, leading to a great effect against malignant cells. In vivo assays demonstrated CAB loading DOX held excellent biosafety and superior antitumor capacity. Incorporating all the benefits, this proposed nanoplatform may provide valuable strategies for efficient drug delivery.

Abstract:The safe and efficient delivery of drugs is regarded as a fundamental strategy for enhancing the low effectiveness of cancer treatments. Herein, a simple preparation approach for a multifunctional drug delivery system was reported. The novel structure of spiropyran (SP), 1-(6 ́-bromo-hexyl)−3,3-dimethyl indoline-6 ́ -methoxy benzopyran, was used to modify sodium carboxymethyl cellulose (NaCMC) for controlling drug release. SP has a heightened sensitivity to variations in pH, with a linear response range of .2–5.5. The elevated drug loading (31.68 %) can be attributed to the hydrophobic interaction of SP and the adsorption of NaCMC. Porous materials composed of zinc ions and dimethylimidazole (ZIF-8) as an embedded shell, which serves as a barrier that effectively reduces drug leakage from 20 % to 12 %, thus mitigating the side effects of camptothecin (CPT) in alkaline conditions. In acidic conditions, the collapse of ZIF-8 provides zinc ions that act as crosslinking agents for NaCMC, thereby improving the channel of drug release and enhancing the release amount from 51.9 % to 66.7 % compared with SP-grafted NaCMC micelles. The inherent biological adhesion of NaCMC ensures sustained drug release for a period of up to 40 h. These pH-responsive carriers hold great promise in the safe and effective treatment of cancer in the gastrointestinal tract, with CPT emerging as a highly promising candidate.

Supramolecular Modular Approach toward Conveniently Constructing and Multifunctioning a pH/Redox Dual-Responsive Drug Delivery Nanoplatform for Improved Cancer Chemotherapy

Abstract:Because heterogeneity affects many functional aspects of a tumor, a way to overcome it is to arm nanosized drug delivery systems (nanoDDS) with diverse functions required to shatter heterogeneity. However, it remains technically challenging to fabricate a nanocarrier possessing all required functions. Here, we propose a modular strategy for generating a supramolecular, multifunctional, and stimuli-responsive nanoDDS through docking a parental core nanoDDS with various daughter function-prebuilt modules. Doxorubicin (DOX)-loaded mesoporous silica nanoparticles (MSNs) as the parental nanocore are wrapped by poly(β-cyclodextrin) (PCD) as a gatekeeper through host-guest interactions between cyclodextrin units and pyridine groups of pyridine-disulfide bonds that confers pH/redox dual responsiveness, thus constructing stimuli-responsive nanoDDS (DOX@PRMSNs). Meanwhile, PCD's free cyclodextrin is tightly caged by adamantyl (Ad)-terminated daughter modules via host-guest interactions, achieving convenient multifunctionalization of this nanoDDS. DOX@PRMSNs rapidly released DOX in lysosomal pH/redox microenvironment, potently killing drug-resistant cancer cells. Further, three different types of Ad-terminated daughter modules, including two targeting ligands (Ad-PEG-FA and Ad-PEG-LA), a cationic polymer (Ad-PEI), and a fluorescence agent (Ad-FITC), are utilized to functionalize PRMSNs via cyclodextrin-Ad self-assembly, endowing the nanoDDS with cell-targeting capability, gene codelivery property, and imaging function. Thus, this work develops a supramolecular modular self-assembly approach for constructing and multifunctionalizing stimuli-responsive "smart" nanoDDSs.

Abstract:It is urgent to develop functional nanocarriers for targeted drug therapy of cancer. This work developed a smart drug controlled release system based on the dendritic mesoporous silica nanoparticle (DMSN) for dual gatekeepers drug delivery. The DMSN-Cys grafted with disulfide bonds appeared as spheres of uniform size with a porous structure, which enabled a drug loading efficiency of 68.32%. Since the protonated amino groups of chitosan were responsive to pH and the disulfide bonds were sensitive to glutathione, the modified 5-Fu@DMSN-Cys/CS can be used for pH/GSH responsive drug delivery. Release behaviors showed excellent drug controlled release effects. Good biocompatibility and pronounced cytotoxicity of 5-Fu@DMSN-Cys/CS were confirmed by cytotoxicity test. Noted specially, the microcosmic adsorption interaction between drug molecules and DMSN-Cys has been evaluated through molecular dynamic simulation. Electrostatic potential and adsorption simulation confirmed that the driving force of 5-Fu adsorption was originated from electrostatic force and hydrogen bonding interaction between the nanocarriers and 5-Fu molecules. These results indicated that 5-Fu@DMSN-Cys/CS could be potential carriers for anticancer drug delivery.

Multiple-Responsive Mesoporous Silica Nanoparticles for Highly Accurate Drugs Delivery to Tumor Cells

Abstract:A core-shell nanocarrier with triple layers, where each layer is sensitive to one specific physiological stimulus, has been fabricated for highly accurate cancer therapy. The nanocarrier consists of mesoporous silica nanoparticles (core structure for drug loading), fluorescein isothiocyanate-labeled hyaluronan (FITC-HA, first shell for imaging with enzymatic response), disulfide bond-embedded silica (SiO2, second layer with glutathione response), and switchable zwitterionic surface (third layer with pH response). The nanocarrier decorated with zwitterionic surface is able to offer long blood circulation time due to the weak nonspecific protein absorption. After these nanocarriers were gradually gathered around tumor cells through enhanced permeability and retention effect, the zwitterionic surface could switch to positive charge in low-pH environment, which was in favor of cellular uptake due to the strengthened positive nanocarrier-negative cellular membrane interaction. Once internalized into tumor cells, the high concentration of glutathione in cytoplasm could cleave disulfide bonds to remove the SiO2 shell and the HA layer would be exposed, which would be further degraded by hyaluronidase to trigger payload release. The fluorescent spectrum and images reveal that both glutathione and hyaluronidase are required for the release of preloaded drugs from these nanocarriers. By employing the multiple response, our nanocarriers could achieve effective antibiofouling ability while maintaining enhanced cellular internalization and targeted drug delivery, resulting in preferred cancer cell cytotoxicity, which is much higher than that of free doxorubicin. The in vitro data exhibited that our nanocarriers may provide an effective strategy for accurate cancer treatment.
Acid Susceptible Ultrathin Mesoporous Silica Coated On Layered Double Hydroxide Nanoplates For Ph Responsive Cancer Therapy

Abstract:Bioresponsive drug delivery systems that can modulate drug release profiles according to different tumor microenvironment are highly desired for improving cancer therapy. In this work, a pH-responsive nanocarrier, layered double hydroxide (LDH) nanoplates coated with ultrathin mesoporous silica layer (LDH@MS), was fabricated with total thickness of around 9 nm. The coating of ultrathin porous silica significantly improved the stability of nanoplates. Moreover, the LDH@MS exhibited pH responsive functionality due to the degradation of silica shell and LDH under moderately acidic pH condition. Notably, the curcumin loaded LDH@MS displayed nearly five-fold greater antitumor efficacy against human breast cancer cells in vitro and marked tumor inhibition in vivo compared to free curcumin under the same drug dosage, most likely due to high dispersibility of the nanocarrier, as well as responsive and steady release of drug molecules. This study opens new avenues to design safer and more effective drug delivery systems with improved therapeutic outcomes.

Abstract:Mesoporous silica nanoparticles (MSNs), an excellent carrier material, have been widely used in tumor therapy as a vector for numerous therapeutic substances to boost therapeutical efficiency and specificity, such as loading them with chemotherapy drugs to improve the efficacy of chemotherapy. Nevertheless, they still face hurdles, such as lack of specificity and poor efficacy of monotherapy. The construction of multifunctional MSNs with excellent therapeutic effects by introducing metal ions has attracted the attention of many researchers. Herein, we demonstrated a calcium doped, chemotherapy drug doxorubicin (Dox) loaded, specific degradation nanoplatform, prepared using the sol–gel method by introducing calcium ions into an MSN framework, which enabled the doped nanoplatform to enhance chemotherapy and activate anti-tumor immune response. As a proof of concept, the doping of Ca2+ endowed MSNs with excellent specific degradation and pH responsive drug release, and enabled the synergy of chemotherapy and calcicoptosis. Furthermore, this nanoplatform also effectively elicited immunogenic cell death (ICD) and promoted the maturation of dendritic cells (DCs), realizing the activation of the anti-tumor immune system. The Ca2+ doped MSNs (CMSNs), that can activate immune response with specific degradation capability, demonstrate a practical strategy for the effective synergy between chemotherapy and calcicoptosis, providing a new paradigm for promoting chemotherapy-related treatment.
Cu2+-Chelating Mesoporous Silica Nanoparticles for Synergistic Chemotherapy/Chemodynamic Therapy

Abstract:In this study, a pH-responsive controlled-release mesoporous silica nanoparticle (MSN) formulation was developed. The MSNs were functionalized with a histidine (His)-tagged targeting peptide (B3int) through an amide bond, and loaded with an anticancer drug (cisplatin (CP)) and a lysosomal destabilization mediator (chloroquine (CQ)). Cu2+ was then used to seal the pores of the MSNs via chelation with the His-tag. The resultant nanoparticles showed pH-responsive drug release, and could effectively target tumor cells via the targeting effect of B3int. The presence of CP and Cu2+ permits reactive oxygen species to be generated inside cells; thus, the chemotherapeutic effect of CP is augmented by chemodynamic therapy. In vitro and in vivo experiments showed that the nanoparticles are able to effectively kill tumor cells. An in vivo cancer model revealed that the nanoparticles increase apoptosis in tumor cells, and thereby diminish the tumor volume. No off-target toxicity was noted. It thus appears that the functionalized MSNs developed in this work have great potential for targeted, synergistic anticancer therapies.

Abstract:Mesoporous silica nanoparticle (MSN), sodium hyaluronate (SH), silk fibroin (SS), and oxidized sodium carboxymethyl cellulose (O-CMC) hybrids were used to develop an intelligent drug delivery platform that may be employed for pH and redox-responsive bi-drug administration. The first drug, cytarabine (Cyt), was loaded with amino-functionalized mesoporous silica (MSN-NH 2 ) encased by the hydrogel of cystamine (Cys) and SH cross-linked by amide bonds. Hydrophobic doxorubicin (DOX) was co-loaded with Cyt/MSN-NH 2 /SA in the hydrogel of SS and O-CMC in the Cyt- loaded hydrogel. Dual-responsive drug delivery may be achieved by encapsulating SS and O-CMC in a hydrogel, including Cyt/MSN-NH 2 /SA/DOX/SS/O-CMC, which has acyl hydrazone bonds (-HC = N) and disulfide bond (-S-S-) exchange reaction with glutathione (GSH). Compared to hydrogels encapsulating only one drug (Cyt or DOX), cell survival analysis revealed that the newly fabricated hydrogels have significantly greater chemotherapeutic efficacy. The cell proliferation of the fabricated nanoparticles was examined in MCF-7 and MDA-MB-231 cells, which indicates that the nanoparticles effectively kill the cancer cells without affecting non-cancerous cells. Further, we effectively investigated the morphological changes, and various biochemical staining methods examined nuclear fragmentation/condensation. Furthermore, the biosafety of the nanoparticles was investigated by the in vivo animal model, which reveals that they remarkably enhanced the safety profile in various organs. These outcomes demonstrated that this nanoparticle platform was a promising beneficial agent for improving breast cancer treatment.

Abstract:Promising drug nanocarriers consisting of mono-dispersed and pH sensitive chitosan–silica hollow nanospheres (CS–SiO2 HNPs) suitable for breast cancer therapy are produced and investigated. The SiO2 HNPs are fabricated using a one-step, one-medium process which obviates the need for post-treatment to remove the templates, additional dissolution, or calcination. Taking advantage of the cross-linking reaction with (3-Glycidyloxypropyl) trimethoxysilane (GTPMS), cationic polysaccharide-chitosan decorates the surface and produces pH sensitive CS–SiO2 HNPs. The materials enable controlled release of loaded drugs in pericellular and interstitial environments. In particular, the antibody molecule (to ErbB 2) can be conjugated onto the surface of the CS–SiO2 HNPs thereby allowing the hollow nanospheres to serve as a targeted delivery agent to breast cancer cells. TNF-α are delivered to MCF-7 breast cancer cells under both in vitro and in vivo conditions to suppress the growth of cancerous cells and even kill them with high therapeutic efficacy. Owing to their hollow inner cavity and porous structures, the CS–SiO2 HNPs are excellent pH-responsive targeted nanocarriers.
