Abstract:A central component of wayfinding is the ability to maintain a consistent representation of one's facing direction when moving about the world. In rodents, head direction cells are believed to support this "neural compass", but identifying a similar mechanism in humans during dynamic naturalistic navigation has been challenging. To address this issue, we acquired fMRI data while participants freely navigated through a virtual reality city. Encoding model analyses revealed voxel clusters in retrosplenial complex and superior parietal lobule that exhibited reliable tuning as a function of facing direction. Crucially, these directional tunings were consistent across perceptually different versions of the city, spatially separated locations within the city, and motivationally distinct phases of the behavioral task. Analysis of the model weights indicated that these regions may represent facing direction relative to the principal axis of the environment. These findings reveal specific mechanisms in the human brain that allow us to maintain a sense of direction during naturalistic, dynamic navigation.

Abstract:The complexity of the environment requires humans to solve problems collaboratively. The aim of this study was to investigate the neural mechanism of social navigation in group problem-solving situations. A novel cooperative task was designed in which dyadic participants assumed the role of an operator or a navigator with different skills and knowledge and worked together to complete the task. Using functional near-infrared spectroscopy-based hyperscanning, we found stronger interbrain neural synchronization of the right temporoparietal junction (rTPJ) between dyads when the operator received instructions from the navigator rather than from a computer. The functional connections between the rTPJ and the other brain areas indicated the involvement of the mirror neural system during the task. Further directional analysis using Granger causality analysis revealed a flow of information from the temporal to the parietal and then to the pre-motor cortex in the operator's brain. These findings provide empirical evidence for the neural mechanism of social navigation and highlight the importance of the rTPJ for communication and joint attention in uncertain group problem-solving situations.
Structural and Functional Neural Correlates of Spatial Navigation: a Combined Voxel-Based Morphometry and Functional Connectivity Study.

Abstract:Introduction: Navigation is a fundamental and multidimensional cognitive function that individuals rely on to move around the environment. In this study, we investigated the neural basis of human spatial navigation ability.Methods: A large cohort of participants (N > 200) was examined on their navigation ability behaviorally and structural and functional magnetic resonance imaging (MRI) were then used to explore the corresponding neural basis of spatial navigation.Results: The gray matter volume (GMV) of the bilateral parahippocampus (PHG), retrosplenial complex (RSC), entorhinal cortex (EC), hippocampus (HPC), and thalamus (THAL) was correlated with the participants' self-reported navigational ability in general, and their sense of direction in particular. Further fMRI studies showed that the PHG, RSC, and EC selectively responded to visually presented scenes, whereas the HPC and THAL showed no selectivity, suggesting a functional division of labor among these regions in spatial navigation. The resting-state functional connectivity analysis further revealed a hierarchical neural network for navigation constituted by these regions, which can be further categorized into three relatively independent components (i.e., scene recognition component, cognitive map component, and the component of heading direction for locomotion, respectively).Conclusions: Our study combined multi-modality imaging data to illustrate that multiple brain regions may work collaboratively to extract, integrate, store, and orientate spatial information to guide navigation behaviors.
Electrophysiological signatures of veridical head direction in humans

Abstract:Information about heading direction is critical for navigation as it provides the means to orient ourselves in space. However, given that veridical head-direction signals require physical rotation of the head and most human neuroimaging experiments depend upon fixing the head in position, little is known about how the human brain is tuned to such heading signals. Here we adress this by asking 52 healthy participants undergoing simultaneous electroencephalography and motion tracking recordings (split into two experiments) and 10 patients undergoing simultaneous intracranial electroencephalography and motion tracking recordings to complete a series of orientation tasks in which they made physical head rotations to target positions. We then used a series of forward encoding models and linear mixed-effects models to isolate electrophysiological activity that was specifically tuned to heading direction. We identified a robust posterior central signature that predicts changes in veridical head orientation after regressing out confounds including sensory input and muscular activity. Both source localization and intracranial analysis implicated the medial temporal lobe as the origin of this effect. Subsequent analyses disentangled head-direction signatures from signals relating to head rotation and those reflecting location-specific effects. Lastly, when directly comparing head direction and eye-gaze-related tuning, we found that the brain maintains both codes while actively navigating, with stronger tuning to head direction in the medial temporal lobe. Together, these results reveal a taxonomy of population-level head-direction signals within the human brain that is reminiscent of those reported in the single units of rodents.

Abstract:The efficient use of various spatial cues within a setting is crucial for successful navigation. Two fundamental forms of spatial navigation, landmark-based and self-motion-based, engage distinct cognitive mechanisms. The question of whether these modes invoke shared or separate spatial representations in the brain remains unresolved. While nonhuman animal studies have yielded inconsistent results, human investigation is limited. In our previous work (Chen et al., 2019), we introduced a novel spatial navigation paradigm utilizing ultra-high field fMRI to explore neural coding of positional information. We found that different entorhinal subregions in the right hemisphere encode positional information for landmarks and self-motion cues. The present study tested the generalizability of our previous finding with a modified navigation paradigm. Although we did not replicate our previous finding in the entorhinal cortex, we identified adaptation-based allocentric positional codes for both cue types in the retrosplenial cortex (RSC), which were not confounded by the path to the spatial location. Crucially, the multi-voxel patterns of these spatial codes differed between the cue types, suggesting cue-specific positional coding. The parahippocampal cortex exhibited positional coding for self-motion cues, which was not dissociable from path length. Finally, the brain regions involved in successful navigation differed from our previous study, indicating overall distinct neural mechanisms recruited in our two studies. Taken together, the current findings demonstrate cue-specific allocentric positional coding in the human RSC in the same navigation task for the first time and that spatial representations in the brain are contingent on specific experimental conditions.

Abstract:Objective:Our goal was to determine the incidence and outcomes of intramammary in-transit sentinel lymph nodes (IMSLN) from primary malignant melanoma (MM) of the trunk. We hypothesize that regional metastasis to the breast from anterior trunk MM also occurs via the lymphatic system to these intramammary in-transit sentinel lymph nodes. Background:MM is the most common solid tumor metastasis to the breast. The mechanism of intramammary (IM) metastasis is generally attributed to hematogenous rather than lymphatic spread. Methods:We retrospectively reviewed medical records from all patients who underwent selective sentinel lymph node dissection at the UCSF Melanoma Center from 1993 to 2008 after the approval of UCSF Committee on Human Research. Of the 1911 cases, we found 614 patients with primary MM located on the trunk, and queried their medical records for in-transit SLN and SLNs in the breast. Data from preoperative lymphoscintigraphy, intraoperative lymphatic mapping, operative notes, and pathology and clinic notes were gathered. Results:Of the 1911 patients with MM, 169 (8.9%) and 420 (22.0%) had anterior and posterior trunk lesions, respectively, and 25 patients (1.3%) with flank lesions (lateral abdominal wall below the rib cage, above the iliac crest). Of the anterior trunk population, 18 patients had in-transit SLNs. The vast majority of these patients (14 of 18, 77.8%) had in-transit IMSLN. Of patients with posterior trunk melanoma, 27 patients had in-transit nodes with 1 patient having IMSLNs. Of patients with flank melanomas, 3 patients had in-transit nodes with 1 patient having IMSLNs. Interestingly, all patients with IMSLNs had primary lesions located inferior to the breasts. Two of the 16 patients with IMSLNs had micrometastasis to IMSLN; 1 patient died and the other currently is disease free 4 years after initial SLND. Four of the 32 patients with non-IM in-transit nodes had micrometastases to these in-transit nodes. Of all patients with trunk melanomas, 4 patients had micrometastases to axillary SLNs (AxSLNs). Three of the 4 patients with positive AxSLNs also had positive in-transit nodes whereas only half of the patients with positive in-transit SLNs had positive AxSLNs. Conclusions:IMSLNs exist in the breast. Our results establish an anatomic basis for lymphatic metastasis to the breast from primary cutaneous melanoma mainly from the anterior trunk inferior to the breasts. For anterior trunk melanomas, IMSLNs should not be overlooked during SLND as they may harbor micrometastasis.
A neuro-cognitive robot for spatial navigation

Abstract:This paper presents a brain-inspired neural architecture with spatial cognition and navigation capability. It captures some navigation properties of rat brain in hidden goal hunting. The brain-inspired system consists of two main parts. One part is hippocampal circuitry and the other part is hierarchical vision architecture. The hippocampus is mainly responsible for the memory and spatial navigation in the brain. The vision system provides the key information about the environment. In the experiment, the cognitive model is implemented in a mobile robot which is placed in a spatial memory task. During the navigation, the neurons in CA1 area show a place dependent response. This place-dependent pattern of CA1 guides the motor neuronal area which then dictates the robot move to the goal location. The results of current study could contribute to the development of brain-inspired cognitive map which enables the mobile robot to perform a rodent-like behavior in the navigation task.

Abstract:Place cells of the rodent hippocampus constitute one of the most striking examples of a correlation between neuronal activity and complex behaviour in mammals1,2. These cells increase their firing rates when the animal traverses specific regions of its surroundings, providing a context-dependent map of the environment3,4,5. Neuroimaging studies implicate the hippocampus and the parahippocampal region in human navigation6,7,8. However, these regions also respond selectively to visual stimuli9,10,11,12,13. It thus remains unclear whether rodent place coding has a homologue in humans or whether human navigation is driven by a different, visually based neural mechanism. We directly recorded from 317 neurons in the human medial temporal and frontal lobes while subjects explored and navigated a virtual town. Here we present evidence for a neural code of human spatial navigation based on cells that respond at specific spatial locations and cells that respond to views of landmarks. The former are present primarily in the hippocampus, and the latter in the parahippocampal region. Cells throughout the frontal and temporal lobes responded to the subjects' navigational goals and to conjunctions of place, goal and view.

Abstract:Abstract It is currently unclear whether the human brain processes navigation tasks at different scales in the same way. According to the classical view, humans process navigation information using a hierarchical representation system in a unified way. Other theories, such as the dual and multiple systems theories, suggest that the processing of navigation tasks differs between spatial scales. In addition, previous human navigation studies have mainly focused on scales ranging from rooms to small neighborhoods. However, the neural mechanisms underlying the processing of large-scale spatial navigation tasks in the human brain, and the ways in which neural activation changes with scale, have not been examined in detail. In this study, we conducted a functional magnetic resonance imaging (fMRI) based multi-scale mental navigation experiment across six spatial scales. On the basis of cortical activity patterns, we discovered a scale-dependent neural system that included the parahippocampal gyrus, cingulate gyrus, parietal gyrus and temporal gyrus, revealing neural-based divisions based on spatial scale: small scale (room), medium scale (building), large scale (block), and huge scale (city, country and continent). In contrast, scale-free characteristics were observed in middle occipital gyrus. The current findings provide new insight into the neural mechanisms of scale-driven spatial navigation in humans.
Social navigation: distance and grid-like codes support navigation of abstract social space in human brain

Abstract:People form impressions about others during daily social encounters and infer personality traits from others’ behaviors. Such trait inference is thought to rely on two universal dimensions, i.e., competence and warmth. These two dimensions can be used to construct a ‘social cognitive map’ organizing massive information obtained from social encounters efficiently. Originated from spatial cognition, the neural codes supporting representation and navigation of spatial cognitive map has been widely studied. Recent studies suggest similar neural mechanism subserves the map-like architecture in social cognition as well. Here we investigated how spatial codes operate beyond physical environment and support the representation and navigation of social cognitive map. We designed a social value space defined by two dimensions of competence and warmth. Behaviorally, participants were able to navigate to a learned location from random starting locations in this abstract social space. At neural level, we identified representation of distance in precuneus, fusiform gyrus and middle occipital gyrus. We also found partial evidence of grid-like representation patterns in medial prefrontal cortex and entorhinal cortex. Moreover, the intensity of grid-like response scaled with performance of navigating in social space and social avoidance trait scores. Our findings suggest a neurocognitive mechanism by which social information can be organized into a structured representation namely cognitive map and its relevance to social well-being.

Human Navigation Network: the Intrinsic Functional Organization and Behavioral Relevance

Abstract:Spatial navigation is a crucial ability for living. Previous work has revealed multiple distributed brain regions associated with human navigation. However, little is known about how these regions work together as a network (referred to as navigation network) to support flexible navigation. In a novel protocol, we combined neuroimaging meta-analysis, and functional connectivity and behavioral data from the same subjects. Briefly, we first constructed the navigation network for each participant, by combining a large-scale neuroimaging meta-analysis (with the Neurosynth) and resting-state functional magnetic resonance imaging. Then, we investigated multiple topological properties of the navigation networks, including small-worldness, modularity, and highly connected hubs. Finally, we explored the behavioral relevance of these intrinsic properties in a large sample of healthy young adults (N = 190). We found that navigation networks showed small-world and modular organization at global level. More importantly, we found that increased small-worldness and modularity of the navigation network were associated with better navigation ability. Finally, we found that the right retrosplenial complex (RSC) acted as one of the hubs in the navigation network, and that higher betweenness of this region correlated with better navigation ability, suggesting a critical role of the RSC in modulating the navigation network in human brain. Our study takes one of the first steps toward understanding the underlying organization of the navigation network. Moreover, these findings suggest the potential applications of the novel approach to investigating functionally meaningful networks in human brain and their relations to the behavioral impairments in the aging and psychiatric patients.

Abstract:Abstract The retrosplenial complex (RSC) plays a crucial role in spatial orientation by computing heading direction and translating between distinct spatial reference frames based on multi-sensory information. While invasive studies allow investigating heading computation in moving animals, established non-invasive analyses of human brain dynamics are restricted to stationary setups. To investigate the role of the RSC in heading computation of actively moving humans, we used a Mobile Brain/Body Imaging approach synchronizing electroencephalography with motion capture and virtual reality. Data from physically rotating participants were contrasted with rotations based only on visual flow. During physical rotation, varying rotation velocities were accompanied by pronounced wide frequency band synchronization in RSC, the parietal and occipital cortices. In contrast, the visual flow rotation condition was associated with pronounced alpha band desynchronization, replicating previous findings in desktop navigation studies, and notably absent during physical rotation. These results suggest an involvement of the human RSC in heading computation based on visual, vestibular, and proprioceptive input and implicate revisiting traditional findings of alpha desynchronization in areas of the navigation network during spatial orientation in movement-restricted participants.

Leveraging Hyperscanning EEG and VR Omnidirectional Treadmill to Explore Inter-Brain Synchrony in Collaborative Spatial Navigation

Abstract:Navigating through a physical environment to reach a desired location involves a complex interplay of cognitive, sensory, and motor functions. When navigating with others, experiencing a degree of behavioral and cognitive synchronization is both natural and ubiquitous. This synchronization facilitates a harmonious effort toward achieving a common goal, reflecting how individuals instinctively align their actions and thoughts in collaborative settings. Collaborative spatial tasks, which are crucial in daily and professional settings, require coordinated navigation and problem-solving skills. This study explores the neural mechanisms underlying such tasks by using hyperscanning electroencephalography (EEG) technology to examine brain dynamics in dyadic route planning within a virtual reality setting. By analyzing intra- and inter-brain couplings across delta, theta, alpha, beta, and gamma EEG bands using both functional and effective connectivity measures, we identified significant neural synchronization patterns associated with collaborative task performance in both leaders and followers. Functional intra-brain connectivity analyses revealed distinct neural engagement across EEG frequency bands, with increased delta couplings observed in both leaders and followers. Theta connectivity was particularly enhanced in followers, whereas the alpha band exhibited divergent patterns that indicate role-specific neural strategies. Inter-brain analysis revealed increased delta causality between interacting members but decreased theta and gamma couplings from followers to leaders. Additionally, inter-brain analysis indicated decreased couplings in faster-performing dyads, especially in theta bands. These insights enhance our understanding of the neural mechanisms driving collaborative spatial navigation and demonstrate the effectiveness of hyperscanning in studying complex brain-to-brain interactions.

Abstract:Abstract Individuals use social information to guide social interactions and to update relationships along multiple social dimensions. However, it is unclear what neural basis underlies this process of abstract “social navigation”. In the current study, we recruited twenty-nine participants who performed a choose-your-own-adventure game in which they interacted with fictional characters during fMRI scanning. Using a whole-brain GLM approach, we found that vectors encoding two-dimensional information about the relationships predicted BOLD responses in the hippocampus and the precuneus, replicating previous work. We also explored whether these geometric representations were related to key brain regions previously identified in physical and abstract spatial navigation studies, but we did not find involvement of the entorhinal cortex, parahippocampal gyrus or the retrosplenial cortex. Finally, we used psychophysiological interaction analysis and identified a network of regions that correlated during participants’ decisions, including the left posterior hippocampus, precuneus, dorsolateral prefrontal cortex (dlPFC), and the insula. Our findings suggest a brain network for social navigation in multiple abstract, social dimensions that includes the hippocampus, precuneus, dlPFC, and insula.

Multivoxel Pattern Analysis Reveals 3D Place Information in the Human Hippocampus

Abstract:The spatial world is three dimensional (3D) and humans and other animals move both horizontally and vertically within it. Extant neuroscientific studies have typically investigated spatial navigation on a horizontal 2D plane, leaving much unknown about how 3D spatial information is represented in the brain. Specifically, horizontal and vertical information may be encoded in the same or different neural structures with equal or unequal sensitivity. Here, we investigated these possibilities using fMRI while participants were passively moved within a 3D lattice structure as if riding a rollercoaster. Multivoxel pattern analysis was used to test for the existence of information relating to where and in which direction participants were heading in this virtual environment. Behaviorally, participants had similarly accurate memory for vertical and horizontal locations and the right anterior hippocampus (HC) expressed place information that was sensitive to changes along both horizontal and vertical axes. This is suggestive of isotropic 3D place encoding. In contrast, participants indicated their heading direction faster and more accurately when they were heading in a tilted-up or tilted-down direction. This direction information was expressed in the right retrosplenial cortex and posterior HC and was only sensitive to vertical pitch, which could reflect the importance of the vertical (gravity) axis as a reference frame. Overall, our findings extend previous knowledge of how we represent the spatial world and navigate within it by taking into account the important third dimension.SIGNIFICANCE STATEMENT The spatial world is 3D. We can move horizontally across surfaces, but also vertically, going up slopes or stairs. Little is known about how the brain supports representations of 3D space. A key question is whether horizontal and vertical information is equally well represented. Here, we measured fMRI response patterns while participants moved within a virtual 3D environment and found that the anterior hippocampus (HC) expressed location information that was sensitive to the vertical and horizontal axes. In contrast, information about heading direction, found in retrosplenial cortex and posterior HC, favored the vertical axis, perhaps due to gravity effects. These findings provide new insights into how we represent our spatial 3D world and navigate within it.

Abstract:<p>Coupling behavioral measures and brain imaging in naturalistic, ecological conditions is key to comprehend the neural bases of spatial navigation. This highly‐integrative function encompasses sensorimotor, cognitive, and executive processes that jointly mediate active exploration and spatial learning. However, most neuroimaging approaches in humans are based on static, motion constrained paradigms and they do not account for all these processes, in particular multisensory integration. Following the Mobile Brain/Body Imaging approach, we aimed to explore the cortical correlates of landmark‐based navigation in actively behaving young adults, solving a Y‐maze task in immersive virtual reality. EEG analysis identified a set of brain areas matching state‐of‐the‐art brain imaging literature of landmark‐based navigation. Spatial behavior in mobile conditions additionally involved sensorimotor areas related to motor execution and proprioception usually overlooked in static fMRI paradigms. Expectedly, we located a cortical source in or near the posterior cingulate, in line with the engagement of the retrosplenial complex in spatial reorientation. Consistent with its role in visuo‐spatial processing and coding, we observed an alpha power desynchronization while participants gathered visual information. We also hypothesized behavior‐dependent modulations of the cortical signal during navigation. Despite finding few differences between the encoding and retrieval phases of the task, we identified transient time‐frequency patterns attributed, for instance, to attentional demand, as reflected in the alpha/gamma range, or memory workload in the delta/theta range. We confirmed that combining mobile high‐density EEG and biometric measures can help unravel the brain structures and the neural modulations subtending ecological landmark‐based navigation.</p>

Experience with the Cardinal Coordinate System Contributes to the Precision of Cognitive Maps

Abstract:The coordinate system has been proposed as a fundamental and cross-culturally used spatial representation, through which people code location and direction information in the environment. Here we provided direct evidence demonstrating that daily experience with the cardinal coordinate system (i.e., east, west, north, and south) contributed to the representation of cognitive maps. Behaviorally, we found that individuals who relied more on the cardinal coordinate system for daily navigation made smaller errors in an indoor pointing task, suggesting that the cardinal coordinate system is an important element of cognitive maps. Neurally, the extent to which individuals relied on the cardinal coordinate system was positively correlated with the gray matter volume of the entorhinal cortex, suggesting that the entorhinal cortex may serve as the neuroanatomical basis of coordinate-based navigation (the entorhinal coordinate area, ECA). Further analyses on the resting-state functional connectivity revealed that the intrinsic interaction between the ECA and two hippocampal sub-regions, the subiculum and cornu ammonis, might be linked with the representation precision of cognitive maps. In sum, our study reveals an association between daily experience with the cardinal coordinate system and cognitive maps, and suggests that the ECA works in collaboration with hippocampal sub-regions to represent cognitive maps.
A neural circuit architecture for rapid behavioral flexibility in goal-directed navigation

Abstract:Anchoring goals to spatial representations enables flexible navigation in both animals and artificial agents. However, using this strategy can be challenging in novel environments, when both spatial and goal representations must be acquired quickly and simultaneously. Here, we propose a framework for how use their internal representation of head direction to build a goal heading representation upon selective thermal reinforcement. We show that flies in a well-established operant visual learning paradigm use stochastically generated fixations and directed saccades to express heading preferences, and that compass neurons, which represent flies’ head direction, are required to modify these preferences based on reinforcement. We describe how flies’ ability to quickly map their surroundings and adapt their behavior to the rules of their environment may rest on a behavioral policy whose parameters are flexible but whose form and dependence on head direction and goal representations are genetically encoded in the modular structure of their circuits. Using a symmetric visual setting, which predictably alters the dynamics of the head direction system, enabled us to describe how interactions between the evolving representations of head direction and goal impact behavior. We show how a policy tethered to these two internal representations can facilitate rapid learning of new goal headings, drive more exploitative behavior about stronger goal headings, and ensure that separate learning processes involved in mapping the environment and forming goals within that environment remain consistent with one another. Many of the mechanisms we outline may be broadly relevant for rapidly adaptive behavior driven by internal representations.

Abstract:People form impressions about others during daily social encounters and infer personality traits from others' behaviors. Such trait inference is thought to rely on two universal dimensions: competence and warmth. These two dimensions can be used to construct a 'social cognitive map' organizing massive information obtained from social encounters efficiently. Originating from spatial cognition, the neural codes supporting the representation and navigation of spatial cognitive maps have been widely studied. Recent studies suggest similar neural mechanism subserves the map-like architecture in social cognition as well. Here we investigated how spatial codes operate beyond the physical environment and support the representation and navigation of social cognitive map. We designed a social value space defined by two dimensions of competence and warmth. Behaviorally, participants were able to navigate to a learned location from random starting locations in this abstract social space. At the neural level, we identified the representation of distance in the precuneus, fusiform gyrus, and middle occipital gyrus. We also found partial evidence of grid-like representation patterns in the medial prefrontal cortex and entorhinal cortex. Moreover, the intensity of grid-like response scaled with the performance of navigating in social space and social avoidance trait scores. Our findings suggest a neurocognitive mechanism by which social information can be organized into a structured representation, namely cognitive map and its relevance to social well-being.
