Abstract:Micro/nanorobots hold exciting prospects for biomedical and even clinical applications due to their small size and high controllability. However, it is still a big challenge to maneuver micro/nanorobots into narrow spaces with high deformability and adaptability to perform complicated biomedical tasks. Here, we report a light-controlled soft bio-microrobots (called "Ebot") based on Euglena gracilis that are capable of performing multiple tasks in narrow microenvironments including intestinal mucosa with high controllability, deformability and adaptability. The motion of the Ebot can be precisely navigated via light-controlled polygonal flagellum beating. Moreover, the Ebot shows highly controlled deformability with different light illumination duration, which allows it to pass through narrow and curved microchannels with high adaptability. With these features, Ebots are able to execute multiple tasks, such as targeted drug delivery, selective removal of diseased cells in intestinal mucosa, as well as photodynamic therapy. This light-controlled Ebot provides a new bio-microrobotic tool, with many new possibilities for biomedical task execution in narrow and complicated spaces where conventional tools are difficult to access due to the lack of deformability and bio-adaptability.

Abstract:The effect of dynamic shape switching of hydrogel bilayers on the performance of self-folding microrobots is investigated for navigation in body orifices and drug release on demand. Tubular microrobots are fabricated by coupling a thermoresponsive hydrogel nanocomposite with a poly(ethylene glycol)diacrylate (PEGDA) layer, to achieve spontaneous and reversible folding from a planar rectangular structure. Graphene oxide (GO) or silica-coated superparamagnetic iron oxide nanoparticles are dispersed in the thermoresponsive hydrogel matrix to provide near-infrared (NIR) light sensitivity or magnetic actuation, respectively. The NIR light-responsive microstructures are fabricated for triggered drug delivery while magnetic nanocomposite-based microrobots are used to analyze the role of shape in locomotion. Experimental analysis and computational simulations of tubular structures show that drug release and motility can be optimized through controlled shape change. These concepts are finally applied to helical microrobots to show a possible way to achieve autonomous behavior.

Shape-programmable Adaptive Multi-material Microrobots for Biomedical Applications

Abstract:Flagellated microorganisms can swim at low Reynolds numbers and adapt to changes in their environment. Specifically, the flagella can switch their shapes or modes through gene expression. In the past decade, efforts have been made to fabricate and investigate rigid types of microrobots without any adaptation to the environments. More recently, obtaining adaptive microrobots mimicking real microorganisms is getting more attention. However, even though some adaptive microrobots achieved by hydrogels have emerged, the swimming behaviors of the microrobots before and after the environment-induced deformations are not predicted in a systematic standardized way. In this work, experiments, finite element analysis, and dynamic modeling are presented together to realize a complete understanding of these adaptive microrobots. The above three parts are cross-verified proving the success of using such methods, facilitating the bio-applications with shape-programmable and even swimming performance-programmable microrobots. Moreover, an application of targeted object delivery using the proposed microrobot has been successfully demonstrated. Finally, cytotoxicity tests are performed to prove the potential for using the proposed microrobot for biomedical applications.

Abstract:Microrobots have attracted considerable attention due to their extensive applications in microobject manipulation and targeted drug delivery. To realize more complex micro-/nanocargo manipulation (e.g., encapsulation and release) in biological applications, it is highly desirable to endow microrobots with a shape-morphing adaptation to dynamic environments. Here, environmentally adaptive shape-morphing microrobots (SMMRs) have been developed by programmatically encoding different expansion rates in a pH-responsive hydrogel. Due to a combination with magnetic propulsion, a shape-morphing microcrab (SMMC) is able to perform targeted microparticle delivery, including gripping, transporting, and releasing by "opening-closing" of a claw. As a proof-of-concept demonstration, a shape-morphing microfish (SMMF) is designed to encapsulate a drug (doxorubicin (DOX)) by closing its mouth in phosphate-buffered saline (PBS, pH ∼ 7.4) and release the drug by opening its mouth in a slightly acidic solution (pH < 7). Furthermore, localized HeLa cell treatment in an artificial vascular network is realized by "opening-closing" of the SMMF mouth. With the continuous optimization of size, motion control, and imaging technology, these magnetic SMMRs will provide ideal platforms for complex microcargo operations and on-demand drug release.

Abstract:In the daily life of mankind, microrobots can respond to stimulations received and perform different functions, which can be used to complete repetitive or dangerous tasks. Magnetic driving works well in robots that are tens or hundreds of microns in size, but there are big challenges in driving microrobots that are just a few microns in size. Therefore, it is impossible to guarantee the precise drive of microrobots to perform tasks. Acoustic driven micro-nano robot can achieve non-invasive and on-demand movement, and the drive has good biological compatibility, but the drive mode has low resolution and requires expensive experimental equipment. Light-driven robots move by converting light energy into other forms of energy. Light is a renewable, powerful energy source that can be used to transmit energy. Due to the gradual maturity of beam modulation and optical microscope technology, the application of light-driven microrobots has gradually become widespread. Light as a kind of electromagnetic wave, we can change the energy of light by controlling the wavelength and intensity of light. Therefore, the light-driven robot has the advantages of programmable, wireless, high resolution and accurate spatio-temporal control. According to the types of robots, light-driven robots are subdivided into three categories, namely light-driven soft microrobots, photochemical microrobots and 3D printed hard polymer microrobots. In this paper, the driving materials, driving mechanisms and application scenarios of light-driven soft microrobots are reviewed, and their advantages and limitations are discussed. Finally, we prospected the field, pointed out the challenges faced by light-driven soft micro robots and proposed corresponding solutions.

Abstract:The field of microrobotics has emerged as a promising area of research with significant applications in biomedicine, both in vitro and in vivo, such as targeted cargo delivery, microsurgery, and cellular manipulation. Microrobots actuated with multiple modalities have the potential for greater adaptability, robustness, and capability to perform various tasks. Modular units that can reconfigure into various shapes, create structures that may be difficult to fabricate as one whole unit, and be assembled on-site, could provide more versatility by assembly and disassembly of units on demand. Such multi-modal modular microrobots have the potential to address challenging applications. Here, we present a biocompatible cylindrical microrobot with a dome-shaped cavity. The microrobot is actuated by both magnetic and acoustic fields and forms modular microstructures of various shapes. We demonstrate the use of these microrobots for cellular manipulation by creating patterns on a surface. Abstract:Medical microrobots are distinguished from other robotic systems in that they must function in the human body. As such, they exhibit special characteristics of size, function, and material choice. Recent advances have focused on fabrication techniques, locomotion at microscale environment, and targeted drug delivery. In this article, we will review these advances and examine related developments in material science and strategies for wireless control systems for communication, as well as data acquisition and processing. We focus on medical microrobots made of soft materials (with an emphasis on polymers and hydrogels), which are mechanically robust and deformable, offer tunable biophysical properties, and are highly biocompatible. Integration of the four design elements of locomotion, feedback and control, functionality, and biocompatibility will be needed to develop a fully functioning medical microrobot with clinical utility in the human body, such as diagnostics and treatment of disease, maintenance of wellness, and improvement of overall performance.

Abstract:Microrobots with simultaneously improved degradability and mechanical strength are highly demanded in performing in vivo delivery tasks in clinical applications. The properties of degradability and mechanical strength are contradictory for many materials used to make microrobots. This article proposes a new design that can result in 3D cell culture microrobots with improved degradability and mechanical strength from the following perspectives. First, the mechanical strength of a microrobot is improved using triangle patterns to replace hexagon pattern in the microrobot structure, which can provide more supporting grids to obtain increased mechanical strength. Second, the relationship between structural design and material composition in relation to the mechanical strength of microrobot is investigated. The study reveals that triangle‐patterned microrobots have increased mechanical strength compared with hexagon‐patterned microrobots, thereby allowing high composition of degradable material that leads to the fast degradation of the microrobot. It is also shown that the triangle‐patterned microrobots can maintain the same structural integrity and cell capacity as hexagon‐patterned microrobots. Finally, the demonstration shows that the triangle‐patterned microrobot can be precisely navigated in microfluidic channels. This article successfully demonstrates that the degradability and mechanical strength can be improved simultaneously through the microrobot structural design.

Abstract:Systems with programmable and complex shape morphing are highly desired in many fields wherein sensing, actuation, and manipulation must be performed. Living organisms use nonuniform distributions of their body structural composition to achieve diverse shape morphing, motion, and functionality. However, for the microrobot fabrication, these designs often involve complicated robotic architectures requiring time-consuming and arduous fabrication processes. This paper proposes a single-step aniso-electrodeposition method for fabricating modular microrobots (MMRs) with distinct functions in each modular segment. By programming the electric field, the microscale stripe-shaped structure can be endowed with diverse shape-morphing capabilities, such as spiraling, twisting, bending, and coiling. The proposed fabrication method can develop MMRs with multiple independent modules onto which cells, drugs, and magnetic nanoparticles can be loaded to achieve multifunctionality. Thus, MMRs can perform multiple tasks, such as propulsion, grasping, and object delivery, simultaneously under magnetic control and ionic and pH stimuli.
Biohybrid Microrobots Based on Jellyfish Stinging Capsules and Janus Particles for In Vitro Deep-Tissue Drug Penetration

Abstract:Microrobots engineered from self-propelling active particles, extend the reach of robotic operations to submillimeter dimensions and are becoming increasingly relevant for various tasks, such as manipulation of micro/nanoscale cargo, particularly targeted drug delivery. However, achieving deep-tissue penetration and drug delivery remain a challenge. This work developed a novel biohybrid microrobot consisting of jellyfish stinging capsules, which act as natural nanoinjectors for efficient penetration and delivery, assembled onto an active Janus particle (JP). While microrobot transport and navigation was externally controlled by magnetic field-induced rolling, capsule loading onto the JP surface was controlled by electric field. Following precise navigation of the biohybrid microrobots to the vicinity of target tissues, the capsules were activated by a specific enzyme introduced to the solution, which then triggered tubule ejection and release of the preloaded molecules. Use of such microrobots for penetration of and delivery of the preloaded drug/toxin to targeted cancer spheroids and live Caenorhabditis elegans was demonstrated in-vitro. The findings offer insights for future development of bio-inspired microrobots capable of deep penetration and drug delivery. Future directions may involve encapsulation of various drugs within different capsule types for enhanced versatility. This study may also inspire in-vivo applications involving deep tissue drug delivery.

Abstract:As a new micromanipulation tool with the advantages of small size, flexible movement and easy manipulation, light-driven microrobots have a wide range of prospects in biomedical fields such as drug targeting and cell manipulation. Recently, microrobots have been controlled in various ways, and light field has become a research hotspot by its advantages of noncontact manipulation, precise localization, fast response, and biocompatibility. It utilizes the force or deformation generated by the light field to precisely control the microrobot, and combines with the drug release technology to realize the targeted drug application. Therefore, this paper provides an overview of light-driven microrobots with drug targeting to provide new ideas for the manipulation of microrobots. Here, this paper briefly categorizes the driving mechanisms and materials of light-driven microrobots, which mainly include photothermal, photochemical, and biological. Then, typical designs of light-driven microrobots with different driving mechanisms and control strategies for multiple physical fields are summarized. Finally, the applications of microrobots in the fields of drug targeting and bioimaging are presented as well as the future prospects of light-driven microrobots in the biomedical field are demonstrated.

Abstract:Sub-millimeter untethered locomoting robots hold promise to radically change multiple areas of human activity such as microfabrication/assembly or health care. To overcome the associated hurdles of such a degree of robot miniaturization, radically new approaches are being adopted, often relying on soft actuating polymeric materials. Here, we present light-driven, crawling microrobots that locomote by a single degree of freedom actuation of their light-responsive tail section. The direction of locomotion is dictated by the robot body design and independent of the spatial modulation of the light stimuli, allowing simultaneous multidirectional motion of multiple robots. Moreover, we present a method for steering such robots by reversibly deforming their front section, using ultraviolet (UV) light as a trigger. The deformation dictates the robot locomotion, performing right- or left-hand turning when the UV is turned on or off respectively. The robots' motion and navigation are not coupled to the position of the light sources, which enables simultaneous locomotion of multiple robots, steering of robots and brings about flexibility with the methods to deliver the light to the place of robot operation.

Abstract:Abstract Using materials with properties similar to those of cells and microorganisms together with innovative fabrication methods, soft and smart microrobots can be developed, with increased adaptability and flexibility toward in vivo applications. These tiny robots are designed to carry out difficult tasks such as noninvasive microsurgery, diagnosis and therapy in complex environments, including viscous media and intricate channels. Moreover, the novel property of the soft materials to respond to stimuli has paved the way for the creation of reconfigurable and smart microrobots with both actuation and function (e.g., sensing, drug delivery) capabilities. This feature article aims to give an overview of the different soft and smart swimming microrobots (less than 1 mm in all dimensions), highlighting some aspects of new materials, their development and the challenges in their processing to obtain highly functional microrobots.

Stratified Disk Microrobots with Dynamic Maneuverability and Proton-Activatable Luminescence for in Vivo Imaging

Abstract:Microrobots can expand our abilities to access remote, confined, and enclosed spaces. Their potential applications inside our body are obvious, e.g., to diagnose diseases, deliver medicine, and monitor treatment efficacy. However, critical requirements exist in relation to their operations in gastrointestinal environments, including resistance to strong gastric acid, responsivity to a narrow proton variation window, and locomotion in confined cavities with hierarchical terrains. Here, we report a proton-activatable microrobot to enable real-time, repeated, and site-selective pH sensing and monitoring in physiological relevant environments. This is achieved by stratifying a hydrogel disk to combine a range of functional nanomaterials, including proton-responsive molecular switches, upconversion nanoparticles, and near-infrared (NIR) emitters. By leveraging the 3D magnetic gradient fields and the anisotropic composition, the microrobot can be steered to locomote as a gyrating "Euler's disk", i.e., aslant relative to the surface and along its low-friction outer circumference, exhibiting a high motility of up to 60 body lengths/s. The enhanced magnetomotility can boost the pH-sensing kinetics by 2-fold. The fluorescence of the molecular switch can respond to pH variations with over 600-fold enhancement when the pH decreases from 8 to 1, and the integration of upconversion nanoparticles further allows both the efficient sensitization of NIR light through deep tissue and energy transfer to activate the pH probes. Moreover, the embedded down-shifting NIR emitters provide sufficient contrast for imaging of a single microrobot inside a live mouse. This work suggests great potential in developing multifunctional microrobots to perform generic site-selective tasks in vivo.

Abstract:Microrobots have shown great potential in many applications, such as non-invasive surgery, tissue engineering, precision medicine, and environmental remediation. Within the past decade, soft microrobot has become one of the important branches. It is aimed to create soft and deformable microrobots with high bioaffinity, which can perform complex tasks noninvasively in inaccessible small spaces in the body. Herein, the latest research progress of soft microrobots regarding the three cornerstones of this field is reviewed: material, fabrication, and actuation. First, various materials that are used for the fabrication of soft microrobots are summarized, and their characteristics and functions are discussed. Second, various fabrication methods of soft microrobots are introduced, and their applicability to different materials is discussed. Third, the actuation methods of soft microrobots are discussed, as well as their pros, cons, and adaptability. Moreover, the outstanding behaviors of soft microrobots in biomedical and environmental applications are introduced with some typical examples published recently. Finally, current clinical use challenges of soft microrobots are pointed out, and their intelligentization is proposed and discussed for further innovative development. Soft microrobots have emerged as a significant branch of microrobots due to their low invasiveness and safety, making them highly beneficial for biomedicine applications. The development of soft microrobots relies on the innovations of their materials, fabrication, and actuation.
