Abstract:The toxic chemical element cadmium (Cd) in paddy fields triggered increasing problems of growth inhibition and food security in rice consistently. In this study, we found Metarhizium robertsii, which is widely used as a bioinsecticide and biofertilizer in agriculture and recently found to be resistant to Cd, developed intraradical and extraradical symbiotic hyphae in rice seedlings, and successfully colonized in the rice rhizosphere soil to more than 103 CFUs g(-1) soil at harvesting. M. robertsii colonization significantly reduced Cd accumulations in both hydroponically cultured seedlings and the matured rice cultured in Cd contaminated potting soil (2 ppm). Notably, Cd accumulation reduction of the roots, stems, leaves, husks and grains of the matured rice induced by the fungus were 44.3%, 32.1%, 35.3%, 31.9% and 24.7%, respectively. It was caused by the M. robertsii-induced suppression of Cd intake transporter gene osNramp5 in the rice roots, and the chemical stabilizing of Cd to the residual fraction in the rhizosphere soil. In addition, the colonization of M. robertsii significantly promoted the growth characters and the photosynthesis of the rice plants. This is achieved by the increase of endogenous hormone levels of indole-3-acetic, gibberellin A3 and brassinolide induced by M. robertsii. Furthermore, the fungus enhanced the antioxidative capacities via increasing enzyme activities of catalase, peroxidase and the production of glutathione, ascorbic acid, proline in the rice plants. Our work provides theoretical basis for expanding the use of M. robertsii as in situ Cd accumulation reduction and detoxification agents for rice in contaminated paddy fields.
Biochar Significantly Alters Rhizobacterial Communities and Reduces Cd Concentration in Rice Grains Grown on Cd-contaminated Soils.

Abstract:Cadmium (Cd) contamination poses a serious problem in paddy soils. Biochar is frequently reported to deactivate Cd in soils and reduce Cd accumulation in rice plants, but few studies have addressed whether and how biochar affected the microbial communities in rice rhizosphere, which was an important factor determining the metal bioavailability and plant growth. In this study, biochar was pyrolyzed from bamboo (Phyllostachys heterocycla) chips at 350 °C. By using ICP-MS analysis and 16S rRNA gene sequencing, the impact of the biochar on Cd uptake by rice and on rhizospheric bacterial communities was investigated in both high-accumulating (HA) and low-accumulating (LA) rice cultivars grown in soils artificially contaminated with different Cd levels. Applied biochar significantly reduced Cd contents in rice plants of both cultivars, with substantially lower grain Cd contents for LA grown in highly contaminated soil. Soil pH was slightly increased by the applied biochar. Cd bioavailability was somehow reduced in soils, but not as significant as the reduction of Cd contents in rice plants. More interestingly, biochar application significantly altered the rhizobacterial community: it stimulated growth-promoting bacteria, such as Kaistobacter, Sphingobium (order Sphingomonadales), and Rhizobiaceae (order Rhizobiales); improved natural barrier formation and the transformation of metal mobilization around the rhizosphere mediated by, e.g., Rhodocyclaceae (class Betaproteobacteria) and Geobacter (class Deltaproteobacteria); and enhanced colonization of the LA rhizosphere possibly by taxa involved in Cd immobilization (Desulfovibrionales and Desulfobacterales). These results indicate that biochar application significantly reduces Cd uptake and accumulation by altering the rhizosphere bacterial community in rice grown on Cd-contaminated soils. The baseline data generated in this study provide insights that pave the way toward safer rice production.
Community-based mechanisms underlying the root cadmium uptake regulated by Cd-tolerant strains in rice (Oryza sativa. L)

Abstract:In recent years, the problem of Cd pollution in paddy fields has become more and more serious, which seriously threatens the safe production of food crops and human health. Using microorganisms to reduce cadmium pollution in rice fields is a green, safe and efficient method, the complicated interactions between the microbes in rice roots throughout the process of cadmium absorption by rice roots are poorly understood. In this investigation, a hydroponic pot experiment was used to examine the effects of bacteria R3 ( Herbaspirillum sp) and T4 ( Bacillus cereus ) on cadmium uptake and the endophytic bacterial community in rice roots. The results showed that compared with CK (Uninoculated bacterial liquid), the two strains had significant inhibitory or promotive effects on cadmium uptake in rice plant, respectively. Among them, the decrease of cadmium content in rice plants by R3 strain reached 78.57-79.39%, and the increase of cadmium content in rice plants by T4 strain reached 140.49-158.19%. Further investigation showed that the cadmium content and root cadmium enrichment coefficient of rice plants were significantly negatively correlated with the relative abundances of Burkholderia and Acidovorax , and significantly positively correlated with the relative abundances of Achromobacter , Agromyces and Acidocella . Moreover, a more complex network of microbes in rice roots inhibited rice plants from absorbing cadmium. These results suggest that cadmium uptake by rice plants is closely related to the endophytic bacterial community of roots. This study provides a reference scheme for the safe production of crops in cadmium contaminated paddies and lays a solid theoretical foundation for subsequent field applications.

Abstract:It is unknown how the Cd-mediated changes in the rhizospheric nitrifier community affect plant growth under different water management practices. This study examined the effect of Cd on microbial N transformation and its association with rice growth in two water regimes, continuous flooding (CF) and alternate wetting and drying (AWD), in a paddy soil. The 16S rRNA, amoA genes in the rhizosphere and endosphere, and OsNTRs were determined using qPCR. Nitrifier community analysis was performed by sequencing the amoA genes. Concentrations of N and Cd in shoots and roots, and IAA in the roots were determined by flow injection analyzer, ICP-MS and enzyme-linked immunosorbent (ELISA), respectively. The direct and indirect negative effects of Cd bioavailability in the rhizosphere on plant growth and ultra-structure of roots and leaves were greater under AWD than CF after 56-day growth. Nitrifier community was changed by water and Cd treatments. Crenarchaeota, Nitrosopumilus, Nitrosopira, and Nitrosovibrio tenuis related species were the dominant nitrifier players. Increasing Cd level decreased plant N uptake, root-IAA concentration, and the abundance of 16S rRNA gene in the rhizosphere and endosphere to a greater extent under AWD than under CF. This in turn decreased the colonization of endophytes and the surface area of the roots, leading to decreased N uptake from the soil. The results imply that appropriate water management and rhizospheric engineering in Cd-contaminated paddy fields may be important approaches to decreasing plant Cd uptake and rhizospheric nitrification.
Special Microbial Communities Enhanced the Role of Aged Biochar in Reducing Cd Accumulation in Rice

Abstract:Biochar exhibits a good adsorption ability for heavy metals in soil and has been widely used as a remediation material in Cd-contaminated soil. However, the status of Cd uptake by rice driven by soil physicochemical properties and rhizosphere microbial communities after years of biochar application is not well understood. In this study, the relationship between the rhizosphere microbial community and soil physicochemical properties and rice Cd accumulation were investigated during the main rice growth stages. The results showed that in comparison to the non-biochar treatment (control), a noticeable reduction in Cd content in rice stem sheaths, leaves, rice husks and milled rice with different growth stages were observed in the biochar treatment after four years, which decreased by 38.76–66.18%, 40.93–70.27%, 43.64–47.92% and 31.91–34.38%, respectively. Compared to non-biochar treatment (control), the properties of the soil in different growth stages by biochar treatment of the soil pH, soil organic matter (SOM), total nitrogen (TN) and available phosphorus (AP) were significantly increased, which increased by 10.5–16.13%, 8–25%, 75–130.13% and 132.95–191.43%, respectively. The content of available Cd (ACd) concentration in different stages by biochar treatment was significantly decreased, which decreased by 26.57–44.24%. Biochar application after four years changed the rhizosphere bacterial community structure composition (phyla level) in all stages. The relative abundance of Proteobacteria, Bacteroidetes and Nitrospirae was increased, while the relative abundance of Chloroflexi, Acidobacteria and Actinobacteria was decreased. Meanwhile, the biochar application enriched Rhodocyclaceae, Burkholderiaceae, Nitrosomonadaceae, Anaerolineaceae, Ignavibacteriales and Bacteroidales, which may contribute to the reduction of Cd uptake and accumulation in rice. These results suggest that biochar treatment after four years changed the rhizosphere microbial community structure and soil physicochemical properties and promoted the colonization of specific microbial populations in the rice rhizosphere to form a special protective system in the rice rhizosphere, which reduced Cd uptake by rice.

Abstract:Soil cadmium (Cd) contamination is becoming a widespread concern because of its threat to global ecosystem health and food security. Co-incorporation of Chinese milk vetch (MV) and rice straw (RS) is a common agricultural practice in Southern China; however, the effects of combining these two materials with biochar on Cd bioavailability remain unclear. This study investigated the effects of MV, RS, rape straw biochar (RB), iron-modified biochar (FB), and their combinations on Cd uptake by rice through incubation and field experiments. The results showed that compared with the control without material input (CK), MV + RS (MR), MV + RS + RB (MRRB), and MV + RS + FB (MRFB) considerably reduced the Cd concentration in brown rice by 61.20 %, 65.38 %, and 62.65 %, respectively. Furthermore, the treatments increased the formation of iron‐manganese plaque (IMP) at different growth stages; MRRB and MRFB exhibited the highest increase rates among the treatments. Quantitatively, the Fe plaque and Mn plaque were increased by 20.61 %–47.23 % and 80.18 %–172.74 %, respectively. Compared with CK, the MRRB and MRFB treatments reduced the soil available Cd by 35.09 %–54.45 % and 38.20 %–50.20 %, respectively, at all stages. This decrease was substantially lower than that observed in the MV, RS, and MR treatments. Similar trends were observed in the incubation experiment. Additionally, the Community Bureau of Reference Sequential Extraction Analysis indicated that the MRRB and MRFB treatments converted the bioavailable Cd fractions into a stable form. Partial least squares path model and redundancy analysis revealed that pH was the major factor influencing Cd bioavailability. This study emphasized that the dual impact factors from the enhancement of Cd passivation capability and IMP formation jointly result in the reduction of Cd uptake by rice. Consequently, the co-incorporation of MV, RS, and biochar is promising for remediating Cd-contaminated paddy soils in Southern China.

Abstract:Excessive heavy metals in soils can threaten food security and soil health. New practical technology is urgently needed to remediate cadmium (Cd) contaminated paddies in many parts of the world. Chinese milk vetch (M), rice straw (R), and soil amendments can reduce Cd activity in soil; however, the mechanism underlying this reduction is not well understood. This study explored the impact of co-incorporation of milk vetch, rice straw, and either lime (L), sesbania biochar (B), or sepiolite on soil Cd bioavailability through field experiments. The results indicated that the rice grain Cd concentrations in soil treated with milk vetch + rice straw + fertilizer (MRF, 16.6 %), milk vetch + rice straw + fertilizer + sesbania biochar (MRFB, 50.1 %), and milk vetch + rice straw + fertilizer + lime (MRFL, 48.3 %) were significantly lower than those in soil treated with fertilizer (F). The acid-soluble Cd concentrations influenced rice grain Cd uptake and were 33.9 % and 47.5 % lower for the MRFB and MRFL treatments, respectively, than for F alone. A decrease in acid-soluble Cd (AciCd) was accompanied by a decrease in Eh and increases in pH, Fe 2+ , cation exchange capacity, and dissolved organic carbon. The MRFB treatment promoted iron plaque (IP) formation on the rice root surface. The relative abundances of Desulfobacterota and Verrucomicrobiota were higher for the MRFB treatment than for the other treatments. A partial least squares path model confirmed that Aci-Cd and low-crystalline IP (IP-Feh) influenced the rice grain Cd concentration.

Abstract:Biochar, as a sustainable amendment, effectively remediates soils contaminated with potentially toxic metals. However, the immobilization efficiencies of biochar can vary according to the soil properties. To investigate the critical impact factor of soil properties on the immobilization of cadmium (Cd) and lead (Pb) in soil by biochar, this study was conducted with an incubation batch experiment and a pot experiment by using an acid soil sample (soil A), a weakly acidic soil sample (soil B), and an alkaline soil sample (soil C). The results showed that the CaCl 2 -extracted Cd in the three soil samples was reduced by 15.2% (soil A), 44.3% (soil B), and 22.0% (soil C) with biochar application, and the decrease rate of the available Cd concentration is significantly negatively ( P < 0.05) related with soil pH, cation exchange capacity, and the proportion of illite and illite–smectite mixed layers of clay minerals. Biochar significantly reduced the soil available Pb concentration of soil A, soil B, and soil C by 57.2%, 52.3%, and 40.3%, respectively. And the decreased rate of available Pb concentration is significantly negatively related to soil pH and soil organic materials ( P < 0.05). The Cd concentration of rice shoots is positively related to the amount of iron plaque. Biochar application decreased the formation of iron plaque on rice roots grown in soils A and B due to the biochar slowing down the decreasing trend of the redox potential during flooding. However, biochar increased the amount of iron plaque on rice roots grown in soil C, which had a higher pH. As a result, biochar reduced the accumulation of Cd and Pb in rice shoots and promoted the biomass of rice grown in soils A and B (acid soils) but had the opposite effect on soil C (an alkaline soil).

Synergistic effects of combined application of biochar and arbuscular mycorrhizal fungi on the safe production of rice in cadmium contaminated soil

Abstract:Arbuscular mycorrhizal fungi (AMF) have been shown to effectively mitigate the detrimental effects of heavy metal stress on their plant hosts. Nevertheless, the biological activities of AMF were concurrently compromised. Biochar (BC), as an abiotic factor, had the potential compensate for this limitation. To elucidate the synergistic effects of biotic and abiotic factors, a pot experiment was conducted to assess the impact of biochar and AMF on the growth, physiological traits, and genetic expression in rice plants subjected to Cd stress. The results demonstrated that biochar significantly increased the mycorrhizal colonization rate by 22.19 %, while the combined application of biochar and AMF led to a remarkable enhancement of rice root biomass by 42.2 %. This resulted in a shift in spatial growth patterns that preferentially promoted enhanced underground development. Biochar effectively mitigated the stomatal limitations imposed by Cd on photosynthetic processes. The decrease in IBRv2 (Integrated Biomarker Response version 2) values suggested that the antioxidant system was experiencing a state of remission. An increase of Cd content within the rice root systems was observed, ranging from 33.71 % to 48.71 %, accompanied by a reduction in Cd bioavailability and mobility curtailed its translocation to the aboveground tissues. Under conditions of low soil Cd concentration (Cd ≤ 1 mg·kg-1), the Cd content in rice seeds from the group subjected to the combined treatment remained below the national standard (Cd ≤ 0.2 mg·kg-1). Furthermore, the combined treatment modulated the uptake of Fe and Zn by rice, while simultaneously suppressing the expression of genes associated with Cd transport. Collectively, the integration of biological and abiotic factors provided a novel perspective and methodological framework for safe in-situ utilization of soils with low Cd contamination.

Abstract:Biochar is an eco-friendly amendment for the remediation of soils contaminated with cadmium (Cd). However, little attention has been paid to the influence and underlying mechanisms of the co-pyrolyzed biochar on the bioavailability and uptake of Cd in paddy soils. The current study explored the effects of biochar co-pyrolyzed from peanut shells (P) and maize straw (M) at different mixing ratios (1:0, 1:1, 1:2, 1:3, 0:1, 2:1 and 3:1, w/w), on the bacterial community and Cd fractionation in paddy soil, and its uptake by rice plant. Biochar addition, particularly P1M3 (P/M 1:3), significantly elevated soil pH and cation exchange capacity, transferred the mobile Cd to the residual fraction, and reduced Cd availability in the rhizosphere soil. P1M3 application decreased the concentration of Cd in different rice tissues (root, stem, leaf, and grain) by 30.0%− 49.4%, compared to the control. Also, P1M3 enhanced the microbial diversity indices and relative abundance of iron-oxidizing bacteria in the rhizosphere soil. Moreover, P1M3 was more effective in promoting the formation of iron plaque, increasing the Cd sequestration by iron plaque than other treatments. Consequently, the highest yield and lowest Cd accumulation in rice were observed following P1M3 application. This study revealed the feasibility of applying P1M3 for facilitating paddy soils contaminated with Cd.

Biochar decreases cadmium uptake in indica and japonica rice (Oryza sativa L.): Roles of soil properties, iron plaque, cadmium transporter genes and rhizobacteria

Abstract:Biochar is an effective and economical strategy for in situ soil cadmium (Cd) remediation. It is essential to comprehensively investigate how biochar mitigates Cd uptake of the main rice subspecies. A pot experiment was established via adding corn stalk biochar into Cd-contaminated soil growing indica Yangdao 6 (YD) and japonica Nangeng 9108 (9108). 9108 had lower shoot biomass (-17.9%) but higher root biomass (+14.4%) and shoot Cd concentration (+29.4%) than YD. Biochar decreased soil available Cd by 25.2% and shoot Cd concentration by 13.6% through the liming and passivation effects. Biochar also favored Cd mitigation by recruiting Fe reducer, Cd remover and plant growth-promoting rhizobacteria (e.g. Bacteroides, Deferrisomatota, Bacillus and Allorhizobium). Besides, biochar reduced Cd uptake by stimulating iron plaques formation for 9108. Moreover, biochar did not reduce Cd uptake by inhibiting Cd transporter genes' expressions and it increased OsHMA2 expression in YD. In conclusion, biochar had great capacity in mitigating Cd pollution and rice subspecies responded differently to biochar in iron plaque formation and Cd transporter genes. The research established a comprehensive understanding of the mechanisms underlying Cd mitigation by biochar and helped to breed low Cd-accumulated rice cultivars to safeguard rice production.
Bioremediation of paddy soil with amphitropic mixture markedly attenuates rice cadmium: Effect of soil cadmium removal and Fe/S-cycling bacteria in rhizosphere

Abstract:Cadmium (Cd) pollution in paddy soil can easily lead to excessive Cd in rice, thereby considerably threatening human health. Microbial leaching is an effective pathway for the mobilization and removal of Cd from soil. In this study, an amphitropic mixture (AM) composed of autotrophic and heterotrophic microbial strains was used to leach Cd-contaminated paddy fields. Chemical analysis showed that the AM effectively removed 52 % of the total Cd, 39 % of the available Cd, and 60 % of the exchangeable and carbonate-bound Cd from the paddy soil. After bioleaching, the Cd in the discarded AM solution was adsorbed using a metal adsorbent. Effects of remediation on the soil nutrients or secondary pollution were not significant. Microbial analysis showed that >96 % and 67 % of the indigenous bacteria and fungi, respectively, remained in the AM-remediated soil. Double-cropped rice was cultivated to evaluate the Cd removal efficiency of grains using AM remediation. The Cd in early and late brown rice decreased by 86 % and 56 %, respectively, which was higher than that found for a series of biochemical remediation materials reported in other studies. Furthermore, the AM remediation promoted the growth of iron (Fe)- and sulfur ( S )-cycling bacteria in the rice rhizosphere, such as Sulfuricurvum , Desulfurivibrio and Geobacter etc., which reduced the Cd availability in the soil and rice uptake. This study shows that AM has potential applications in the remediation of Cd-contaminated paddy fields and provides a new pathway for safe rice production.

Abstract:The combination of biochar and specific bacteria has been widely applied to remediate Cadmium-contaminated soil. But little is known about how such composites affect the dynamic distribution of metal fractions. This process is accompanied by the alternations of soil properties and microbial community structures. Composite of rice straw biochar and Bacillus cereus RC-1 were applied to investigate its impacts on Cd alleviation and soil microbial diversity and structure. The bacterial/biochar composite treatment decreased the fraction of HOAc-extractable Cd by 38.82%, and increased residual Cd by 23.95% compared to the untreated control. Moreover, compared with the untreated control, the composite treatment significantly increased the soil pH by about 1.5 units, and the activities of catalase, urease and invertase enzymes were increased by 42.39%, 30.50% and 31.20%, respectively. Composite treatment increased soil bacterial and fungal alpha diversity, the relative abundance of Bacillus, Streptomyces, Arthrobacter, and Aspergillus species were also increased. Mantel test and correlation analysis indicated that the effects associated with fungal communities in influencing soil properties were lower than that those of bacterial communities by different treatment. Aggregated boosted tree (ABT) models analysis showed that soil chemical proprieties (as determined by SOM, CEC, AN, etc.,) contributed over 50% of the changes in bacterial and fungal communities by the composite treatment. The co-occurrence network results showed that all treatments enhanced the correlation between OUT groups and improved the possible relationships in the bacterial and fungal communities, especially the interrelationships between bacteria and fungi after the Cd fractions stabilized. These findings provide a new insight to optimal strategies for the remediation of Cd-contaminated soil.

Abstract:The application of Fe–Mn-modified biochar for the remediation of Cd-contaminated soil over long time periods has been little studied. In this paper, we describe the performance of coconut-shell-derived biochar modified with ferromanganese in relation to soil Cd stabilization and rice Cd bioaccumulation during a 3-year laboratory study. Different application dosages (0.05–0.5 wt%) and different rice varieties (the early and late rice) are also considered. The results show that ferromanganese is mainly loaded in biochar pores as MnFe2O4, and that it decreases the specific surface area (SSA) and total pore volume of biochar. Ferromanganese biochar (0.5 wt%) applied to paddy soil is more effective than the same dose of pristine biochar in decreasing the soil-exchangeable Cd fraction (27.42–41.92% vs 22.56–33.85%), predominantly by decreasing soil Eh and increasing root Fe plaques. Ferromanganese biochar application helps to reduce Cd bioaccumulation in rice, especially in the grain (up to 48.6%–61.0%), and grain Cd levels (0.2 mg/kg) are all within the acceptable limit for food security in China. It is shown that ferromanganese modification and application can maintain soil at low redox status, keep root Fe plaques at a high level, and may also increase the stability of pristine biochar. All of these effects contribute to maintaining its high remediation efficiency over a 3-year inoculation period. The results presented in this paper demonstrate the potential applications of ferromanganese biochar in soil remediation and the improvement of food safety.

Abstract:Iron plaque is believed to be effective in reducing the accumulation of heavy metals in rice. In this work, a known soil-derived Mn(II)-oxidizing bacterium, LLDRA6, which represents the type strain of Providencia manganoxydans , was employed to investigate the feasibility of decreasing cadmium (Cd) accumulation in rice by promoting the formation of iron plaque on the root surface. Firstly, the Fe(II) oxidation ability of LLDRA6 was evaluated using various techniques including Fourier Transform infrared spectroscopy, X-ray diffraction, X-ray photoelectron spectroscopy, phenanthroline photometry, and FeS gel-stabilized gradient assays. Subsequently, the formation of iron plaque on the root surface by LLDRA6 was investigated under hydroponic and pot conditions. Finally, Cd concentrations were examined in rice with and without iron plaque through pot and paddy-field tests. The results showed that LLDRA6 played an efficient role in the formation of iron plaque on seedling roots under hydroponic conditions, generating 44.87 and 36.72 g kg − 1 of iron plaque on the roots of Huazhan and TP309, respectively. In pot experiments, LLDRA6 produced iron plaque exclusively in the presence of Fe(II). Otherwise, it solely generated biofilm on the root surface. Together with Fe(II), LLDRA6 effectively reduced the concentrations of Cd in Huazhan roots, straws and grains by 25%, 46% and 44%, respectively. This combination also demonstrated a significant decrease in the Cd concentrations of TP309 roots, straws and grains by 20%, 52% and 44%, respectively. The data from the Cd translocation factor indicate that obstruction of Cd translocation by iron plaque predominantly occurred during the root-to-straw stage. In paddy-field tests, the Cd concentrations of grains harvested from the combination treatment of LLDRA6 and Fe(II) exhibited a decline ranging from 40 to 53%, which fell below the maximum acceptable value for Cd in rice grains (0.2 mg kg − 1 ) as per the China national standard for food security (GB2762-2017). Meanwhile, the relevant phenotypic traits regarding the yield were not adversely affected. These findings have demonstrated that LLDRA6 can impede the uptake of Cd by rice in Cd-contaminated soils through the formation of iron plaque on roots, thus providing a promising safe Cd-barrier for rice production.

Abstract:This work has developed a new strategy of biogeochemical Fe(II) generators for activating microbial Fe(II) generation to immobilize Cd in soils through protons scavenging and coprecipitation. A new biochar modified magnetite (FeBC15) has been fabricated through a top-down method, with which microbial respiration can be stimulated in paddy soil. The FeBC15 exhibits a higher adsorption capacity for Cd than pristine magnetite (1.7 times). The results show that the available Cd can be reduced by 14.4% after adding FeBC15 compared to the control. More importantly, FeBC15 particles promote the conversion of MgCl2 - Cd to stable crystalline Fe/Al bound Cd under the incubation period. The enhanced pH and Fe(II) leads to a comparably lower Cd availability in soils than in pristine soils, which are supported by the enhanced relative abundance of Geobacter and Clostridium with the FeBC15 treatment (i.e. up to 7.44-7.68 × 109 copies/g soil). The Diffusive Gradients in Thin-films (DGT) study indicates that FeBC15 can lower the replenish capacity of soils (i.e. KdL values of 0.2-3.6 mL/g) to soil pore waters and limit root absorption. Pot experiments demonstrate that this strategy can alleviate the rice Cd content by 38.4% (< 0.2 mg/kg). This work paves a new pathway for reducing Cd uptake in rice, enabling sustainable remediation of paddy soil.

Abstract:Soil amendments play a pivotal role in ensuring the safety of food production by inhibiting the transfer of heavy metal ions from soils to crops. Nevertheless, their impact on soil characteristics and the microbial community and their role in reducing cadmium (Cd) accumulation in rice remain unclear. In this study, pot experiments were conducted to investigate the effects of three soil amendments (mineral, organic, and microbial) on the distribution of Cd speciation, organic components, iron oxides, and microbial community structure. The application of soil amendments resulted in significant reductions in the soil available Cd content (16 %-51 %) and brown rice Cd content (16 %-78 %), facilitating the transformation of Cd from unstable forms (decreasing 10 %-20 %) to stable forms (increasing 77 %-150 %) in the soil. The mineral and organic amendments increased the soil cation exchange capacity (CEC) and plant-derived organic carbon (OC), respectively, leading to reduced Cd accumulation in brown rice, while the microbial amendment enhanced OC complexity and the abundances of Firmicutes and Bacteroidota, contributing to the decreased rice Cd uptake. The synchrotron radiation-based Fourier transform infrared (SR-FTIR) spectroscopy indicated that soil amendments regulated soil Cd species by promoting iron oxides and OC coupling. Moreover, both organic and microbial amendments significantly reduced the diversity and richness of the bacterial communities and altered their compositions and structures, by increasing the relative abundances of Bacteroidota and Firmicutes and decreasing those of Acidobacteria, Actinobacteria, and Myxococcota. Soil microbiome analysis revealed that the increase of Firmicutes and Bacteroidota associated with Cd adsorption and sequestration contributed to the suppression of soil Cd reactivity. These findings offer valuable insights into the potential mechanisms by which soil amendments regulate the speciation and bioavailability of Cd, and improve the bacterial communities, thereby providing guidance for agricultural management practices.

Abstract:Toxic metal-contaminated farmland from Cadmium (Cd) can enhance the accumulation of Cd and impair the absorption of mineral elements in brown rice. Although several studies have been conducted on Cd exposure on rice, little has been reported on the relationship between Cd and mineral elements in brown rice and the regulatory mechanism of rhizosphere microorganisms during element uptake. Thus, a field study was undertaken to screen japonica rice cultivars with low Cd and high mineral elements levels, analyze the quantitative relationship between Cd and seven mineral elements, and investigate the cultivar-specific response of rice rhizosphere bacterial communities to differences in Cd and mineral uptake in japonica rice. Results showed that Huaidao-9 and Xudao-7 had low Cd absorption and high amounts of mineral nutrient elements (Fe, Zn, Mg, and Ca, LCHM group), whereas Zhongdao-1 and Xinkedao-31 showed opposite accumulation characteristics (HCLM group). Stepwise regression analysis showed that zinc, iron, and potassium are the key minerals that affect Cd accumulation in japonica rice and zinc was the most important factor, accounting for 68.99 %. The accumulation of Cd and mineral elements is potentially associated with rhizosphere soil bacteria. Taxa enriched in the LCHM rhizosphere (phyla Acidobacteriota and MBNT15) indicated the high nutrient characteristics of the soil and reduced activity of Cd in soil. The HCLM rhizosphere was highly colonized by metal-activating bacteria (Actinobacteria), lignin-degrading bacteria (Actinobacteria and Chlorofexi), and bacteria scavenging nutrients and trace elements (Anaerolinea and Ketobacter). Moreover, the differences in the uptake of Cd and mineral elements affected predicted functions of microbial communities, including sulfur oxidation and sulfur derivative formation, human or plant pathogen, and functions related to the iron oxidation and nitrate reduction. The results indicate a potential association of Cd and mineral elements uptake and accumulation with rhizosphere bacteria in rice, thus providing theoretical basis and a new perspective on the maintenance of rice security and high quality simultaneously.

Abstract:Cadmium (Cd) contamination poses a significant threat to plants and human, as it can easily accumulate in plant tissues, leading to biochemical and physiological disorders. There is a growing interest in using biochar to mitigate the absorption of heavy metals by rice plants. This study tested peach biochar (PB) and its various levels of applications to evaluate the promising level for Cd remediation in contaminated soil. The application of PB3 had a significant impact on Cd mitigation, with extractable Cd (AB-DTPA) in soil decreasing from 66 mg kg-1 to 18 mg kg-1. Cd content in shoots decreased from 2.5 mg kg-1 to 0.9 mg kg-1, and in grains decreased from 1.1 mg kg-1 to 0.5 mg kg-1. Moreover, the PB treatment led to increased rice yield, from 4.9 to 10 g pot-1, and biological yield, from 4 to 20 g pot-1. The soil also showed improved organic matter content, increasing from 0.4% to 0.7%, and enhanced levels of nitrogen (N), phosphorus (P), and potassium (K), by increases from 2.1 g pot-1 to 5 g pot-1, 58 mg kg-1 to 83 mg kg-1, and 40 mg kg-1 to 63 mg kg-1, respectively. These findings demonstrate the potential of PB in mitigating Cd contamination in soil and reducing its uptake by rice plants.
Effect of sulfur and sulfur-iron modified biochar on cadmium availability and transfer in the soil–rice system

Abstract:Cadmium (Cd) contamination in paddy soils has aroused global concern. Sulfur modified biochar (BC) could combine the benefits of BC and S for Cd remediation. However, no information is available on the impact of sulfur modified biochar on Cd phytoavailability in paddy soils. In this study, a pot experiment was conducted to investigate the effect of sulfur modified biochar (S-BC) and sulfur and iron (Fe) modified biochar (S-Fe BC) on Cd mobility and Cd transfer in the soil-rice system. The application of S-BC and S-Fe BC effectively reduced pore water Cd in the rhizosphere and non-rhizosphere pore water throughout the rice growth stages. S-BC and S-Fe BC addition increased the total chlorophyll content, as well as the root, shoot and grain biomasses of rice. Furthermore, S-BC and S-Fe BC amendments greatly increase the formation of Fe plaque on rice root surface, thus decreasing Cd accumulation in different rice tissues. In particular, S-Fe BC supplementation significantly reduced the Cd concentration in rice grains to 0.018 mg kg-1 in Cd-contaminated soil, which was lower than the China National standard for food contamination limit (0.2 mg kg-1 Cd). Sequential extraction results showed that S-BC and S-Fe BC can promote the transfer of exchangeable Cd to Fe-Mn oxide, organic and residual bound forms which reduce Cd in paddy soils. Thus, the amendment of S-Fe BC to Cd-contaminated paddy soil is an effective strategy to decrease Cd accumulation in rice grains and thereby protect public health.
