Abstract:Recently, the orientational immobilization of enzymes has attracted extensive attention. In this study, we report the development of a strategy combined with rational design to achieve precise site-specific covalent immobilization of β-agarase. We first rationally screened six surface sites that can be mutated to cysteine by combining molecular dynamics simulation and energy calculation. Site-specific immobilization was successfully achieved by Michael addition reaction of mutant enzymes and maleimide-modified magnetic nanoparticles (MAL-MNPs). The enzyme activity retention rate of R66C-MAL-MNPs and K588C-MAL-MNPs was greater than 96%. The thermal deactivation kinetics study revealed that the site-specific immobilization strategy significantly improved the thermal stability of Aga50D, resulting in a substantial increase in its antidenaturation activity at elevated temperatures, and the highest t1/2 of the immobilized mutant enzymes was increased by an impressive 21.25-fold at 40 °C. The immobilized mutant enzymes also showed significantly enhanced tolerance to metal ions and organic reagents. For instance, all of the immobilized enzymes maintained over 90% of their enzymatic activity in the 50% (v/v) acetone/water solution. The present work may pave the way for the design of precisely immobilized enzymes, which can help promote green manufacturing.

Abstract:In this work, a new way for enzyme immobilization was explored and properties of the enzyme immobilized on different polymer films were investigated. In the process, a polystyrene-based diazonium salt (PS-DAS) was synthesized and used as molecular adhesive to immobilize β-glucosidase on the polymeric supports (films of polyethylene, polypropylene and poly(ethylene terephthalate)). The immobilization of β-glucosidase on the polymer surfaces was achieved by sequential depositions of a piece of the polymer films in PS-DAS and the enzyme solutions. The surface modification was investigated by X-ray photoelectron spectroscopy (XPS), water contact angle measurement, and atomic force microscopy (AFM). The activity of the immobilized β-glucosidase was evaluated by measuring its enzymatic activity to the hydrolysis of p-nitrophenyl-β-d-glucopyranoside (pNPG). The optimized reaction conditions (such as pH and temperature), thermal stability, and reusability of the immobilized enzyme on PE films were assayed by using the enzyme-catalyzed reaction. Results show that the polymeric diazonium salt is firmly adhered on the polymer surfaces and the modified surfaces can react with the enzyme to form covalent bonds. The immobilized enzyme shows changes in the optimized pH and temperature for the hydrolysis reaction catalyzed by β-glucosidase. The kinetic parameter (Km) of the immobilized β-glucosidase is lower than that of its free counterpart. The immobilized enzyme shows significant enhancement in the thermal stability and reasonable reusability. This new approach can be used as a simple and versatile method for protein immobilization.

Abstract:Background Various cellulolytic enzymes have been studied to date for their application in biofuels, food, cosmetics, beverages and pharmaceutical industries. However, due to the difficulty in recovery, the direct use of free enzymes is not economically viable in large-scale industrial applications. To preserve enzyme stability, recovery and reusability, immobilization is an effective approach. Methods In this study, gene transformation was carried out using Escherichia coli DH5 host cells, and E. coli BL21(DE3), BW25113(DE3), ER2566(DE3), and JM109(DE3) were used to express recombinant β-glucosidase. The immobilized metal ion affinity membrane (IMAM) chromatography approach using modified regenerated cellulose membrane was applied to immobilize and purify recombinant β-glucosidase. Both free and immobilized β-glucosidases were analyzed for their pH/temperature stability, purification and kinetics. The reusability of immobilized β-glucosidase and regeneration of IMAM was also studied. Significant findings The results showed that the immobilized β-glucosidase had higher activity and more stability at a wide range of pH and temperature. Immobilized β-glucosidase retained more than 80 % of relative activity at 4 °C after storage for 20 days. Moreover, it maintained 60 % of relative activity after 5 cycles of repeated use. IMAM membrane was successfully regenerated 6 times with 100 % β-glucosidase adsorption and activity.

Immobilization of β-Galactosidase on Carboxymethyl Cellulose-chitosan Polyelectrolyte Complex

Abstract:β-Galactosidase was immobilized on carboxymethyl collulose-chitosan(CMC-CS) polyelectrolyte complex with glutaraldehyde by crosslinking reaction.The immobilization conditions and characterization of the immobilized enzyme were studied.The results indicated that the optimal activity was 0.023 5 U/g,when the concentrantion of glutaraldehyde was 0.5 %,dosage of β-galactosidase was 1 g/L per 0.5 g CMC-CS,immobilization time and crosslinking time were 9 and 3 h,respectively.The optimalreaction temperature and pH value of the immobilized enzyme were 50 ℃ and 7,respectively;and those of free enzyme were 30 ℃ and 9,respectively.The immobilized enzyme showed lower thermal stability than that of the free enzyme,because of irreversible reaction from carrier and enzyme,but pH value stability of the immobilized enzyme were better than that of the free enzyme.The immobilized β-galactosidase showed high stability when it was used repeatedly for three times.Michaelis constant(Km) of the immobilized enzyme was 0.705 1 mmol/L,which was higher than that of the free enzyme,which indicated the increased affinity of immobilized enzyme toward substrate which was beneficial to the enzymatic reaction.

Abstract:A simplified procedure for the preparation of immobilized beta-amylase using non-purified extract from fresh sweet potato tubers is established in this paper, using differently activated agarose supports. Beta-amylase glutaraldehyde derivative was the preparation with best features, presenting improved temperature and pH stability and activity. The possibility of reusing the amylase was also shown, when this immobilized enzyme was fully active for five cycles of use. However, immobilization decreased enzyme activity to around 15%. This seems to be mainly due to diffusion limitations of the starch inside the pores of the biocatalyst particles. A fifteen-fold increase in the Km was noticed, while the decrease of Vmax was only 30% (10.1Umg−1 protein and 7.03Umg−1 protein for free and immobilized preparations, respectively).

Abstract:Functionalized mesoporous materials have become a promising carrier for enzyme immobilization. In this study, Santa Barbara Amorphous 15 (SBA-15) was modified by N-aminoethyl-γ-aminopropyl trimethoxy (R). R-SBA-15 was employed to purify and immobilize recombinant β-glucosidase from Terrabacter ginsenosidimutans (BgpA) in one step for the first time. Optimum pH of the constructed R-SBA-15@BgpA were 7.0, and it has 20 ℃ higher optimal temperature than free enzyme. Relative activity of R-SBA-15@BgpA still retained > 70% at 42 ℃ after 8-h incubation. The investigation on organic reagent resistance revealed that the immobilized enzyme can maintain strong stability in 15% DMSO. In leaching test and evaluation of storage stability, only trace amount of protein was detected in buffer of the immobilized enzyme after storage at 4 ℃ for 33 days, and the immobilized BgpA still maintained > 50% relative activity. It also demonstrated good reusability, with 76.1% relative activity remaining after fourteen successive enzymatic hydrolyses of epimedin A to sagittatoside A. The newly proposed strategy is an effective approach for the purification and immobilization of BgpA concurrently. In addition, R-SBA-15@BgpA was demonstrated to have high efficiency and stability in this application, suggesting its great feasibility and potential to produce bioactive compounds such as secondary glycosides or aglycones from natural products.

Abstract:BackgroundGlucoamylase enzyme is one of the most important enzymes. It catalyzes the hydrolysis of starch into soluble sugars. It was covalently immobilized onto κ-carrageenan gel beads after activation by using polyethylenimine (PEI) followed by glutaraldehyde (GA). All parameters in activation process were studied.ResultsThe immobilized enzyme shows enhancement in temperature profile as the optimum temperature for the free enzyme was 60 °C, and it becomes 60–80 °C that means broader range and also shows stability in acidic conditions more than free enzyme. The apparent Km of immobilized enzyme, 147.46 mM, becomes higher than Km of the free one, 110 mM and the maximum reaction velocity (Vmax) values for the immobilized enzyme decreased from 2.28 to 1.11 μmol min−1. The immobilized enzyme can be reused and kept its activity (100%) till 11 successive cycles. The immobilization process steps were characterized by FTIR and SEM.ConclusionsThese results confirm the economic and biotechnical benefits of enzyme immobilization, particularly with regard to the number of enzyme reuses, which open the possibility of different industrial applications.

Abstract:Agarose from biomass can be used to synthesize the rare sugar 3,6-anhydro- L -galactose ( L -AHG), and the new synthesis route and functional properties of L -AHG have always been the focus of research. Here we developed a novel method to co-immobilize Aga50D and Bp GH117 onto streptavidin-coated magnetic nanoparticles and achieved the conversion of agarose to bioactive L -AHG in one pot. Results showed that enzymes were successfully immobilized on the carrier. The activity of co-immobilized enzymes was 2.5-fold higher than that of single immobilized enzymes. Compared with free enzymes, co-immobilized enzymes exhibited enhanced thermal stability. The co-immobilized enzymes retained 79.45 % relative activity at 40 °C for 3 h, while the free enzymes only possessed 21.40 % residual activity. After eight cycles, the co-immobilized enzymes still retained 73.47 % of the initial activity. After silica gel chromatography, the purity of L -AHG obtained by co-immobilized enzymes hydrolysis reached 83.02 %. Furthermore, bioactivity experiments demonstrated that L -AHG displayed better antioxidant and antibacterial effects than neoagarobiose. L -AHG had broad-spectrum antibacterial activity, while neoagarobiose and D -galactose did not show an obvious antibacterial effect. This study provides a feasible method for the production of L -AHG by a co-immobilized multi-enzyme system and confirms that L -AHG plays a key role in the bioactivity of neoagarobiose.

Abstract:Immobilized enzymes are better suited for industrial applications than free enzymes due to their favorable properties such as ease of separation and reuse, and enhanced stability and storage life. β-Galactosidases are an important class of glycosidases with hydrolysis and transglycosylation activities, which are applied in industries for lactose hydrolysis and prebiotics synthesis worldwide. The recent innovations in immobilized β-galactosidases have improved the performance of the immobilized enzymes and broadened their applications in the fields of food, energy, and medicine. Innovations in β-galactosidase immobilization methods include rational adsorption based on enzyme features, layer by layer adsorption for strengthened ionic bonding, 3-D printing for rapid and elaborate entrapment, modifications of either materials or enzymes for ingenious covalent binding, nontoxic crosslinking, carrier-free immobilization, and oriented immobilization either through protein engineering or enzyme display on cells, membranes, and phages, along with innovations in carrier materials involving the introduction of graphene derivatives, polyaniline nanomaterials, nanofibers, nucleotide molecules, Langmuir−Blodgett films, and so on. These innovations have partially solved the problems associated with traditional methods, resulting in enzymes with highly retained activity, excellent stability, reduced microbial contamination, enzyme leakage, and reagent toxicity. The immobilized β-galactosidases with potential economic and environmental benefits have been extendedly used for hydrolysis of prodrugs for disease treatment, assembly of biosensors for lactose detection, synthesis of bioactive carbohydrates, and even production of food additives and industrial products, such as tagatose and bioethanol. This review describes the innovations in β-galactosidases immobilization and the applications of these immobilized enzymes. It not only enables the fully understanding of β-galactosidases, but also provides a valuable reference for the immobilization of other industrially-important enzymes.

Abstract:The highly demanding conditions of industrial processes may lower the stability and affect the activity of enzymes used as biocatalysts. Enzyme immobilization emerged as an approach to promote stabilization and easy removal of enzymes for their reusability. The aim of this review is to go through the principal immobilization strategies addressed to achieve optimal industrial processes with special care on those reported for two types of enzymes: β-galactosidases and fructosyltransferases. The main methods used to immobilize these two enzymes are adsorption, entrapment, covalent coupling and cross-linking or aggregation (no support is used), all of them having pros and cons. Regarding the support, it should be cost-effective, assure the reusability and an easy recovery of the enzyme, increasing its stability and durability. The discussion provided showed that the type of enzyme, its origin, its purity, together with the type of immobilization method and the support will affect the performance during the enzymatic synthesis. Enzymes' immobilization involves interdisciplinary knowledge including enzymology, nanotechnology, molecular dynamics, cellular physiology and process design. The increasing availability of facilities has opened a variety of possibilities to define strategies to optimize the activity and re-usability of β-galactosidases and fructosyltransferases, but there is still great place for innovative developments.

Immobilization of Penicillin G Acylase on Mesostructured Cellular Foams through a Cross-Linking Network Method

Abstract:A novel immobilization method that combines the advantages of physical adsorption and covalent binding was developed for penicillin G acylase (PGA) immobilization. In this method, a polymer network was formed by linear polymer molecules and enzymes were connected to the network through multipoint immobilization. This method is different from the conventional covalent binding method that creates chemical bonds between PGA molecules and support. Initially, PGA was physically adsorbed into the support because of the interaction between Si–OH and enzyme molecules. Afterward, chitosan and glutaraldehyde were added to generate a network connected with PGA, thereby successfully preventing enzyme leaching. Using the novel method, the PGA loading amount reached 555.0 mg/g (dry support), the specific activity was up to 27.7 U/mg (enzyme), and 85% activity was retained after five reuses. In addition, the immobilized PGA showed relatively high operation stability in pH and thermal stability.

Abstract:The method of immobilized β-glucuronidase was studied in this paper,β-glucuronidase has been immobilized on chitin with glutaraldehyde as crosslinking agent.Some factors that had effect on activity of immobilized β-glucuronidase were studied.The optimal conditions of immobilization were achieved by using the method of orthogonal test,when glutaraldehyde concentration was 0.7%,cross-linking time was 4h,adsorption time was 9h and reaction temperature was 40℃,the recovery rate of immobilized β-glucuronidase activity was 67.32%.

Abstract:In this paper we successfully immobilized bilayer glucose isomerase in porous p-trimethylamine-polystyrene (TMPS) beads by use of a new method of immobilization of enzyme - molecular-deposition. The effects of immobilizing time, the concentration of nature enzyme, the concentration of hexamethylene bis (trimethylammonium bromide), the concentration BCD were investigated. The properties of this immobilized enzyme were also studied. The results obtained show that the activity and the protein loading of per weight bilayer immobilized enzyme are doubled as compared to monolayer immobilized enzyme. The activity of immobilized bilayer glucose isomerase is 1 200 IU/g wet gel.
Study on the Immobilization of Β-Glucuronidase by Sodium Alginate

Abstract:β-glucuronidase was immobilized by sodium alginate as the carrier.The method of cross-linking-embedding was adopted to immobilize β-glucuronidase.The effects of reaction conditions(the concentration of sodium alginate,glutaraldehyde concentration,CaCl2concentration,cross-linking time and immobilized time)on the relative activity of immobilized β-glucuronidase were tested.The orthogonal test was used to investigate the optimal condition of immobilization.The results showed that the best efficiency of immobilization was obtained when sodium alginate concentration was 35 g/L,β-glucuronidase dosage was 1600 U,glutaraldehyde concentration was 0.2%,CaCl2 concentration was 20g/L,and immobilized time was 2h.The recovery rate of immobilized enzyme activity was 71.66 %.The study showed that this immobilize method was simple and had a prospective industrial application.
A New Application of Aptamer: One-step Purification and Immobilization of Enzyme from Cell Lysates for Biocatalysis.

Abstract:Aptamers are nucleic acid-based high affinity ligands that are able to capture their corresponding target through molecular recognition. In this study, several DNA aptamers with high affinity and specificity for β-glucuronidases (PGUS-E) were obtained by our modified SELEX method. Among them, Apt5 and Apt9 were selected as representatives and covalently linked to magnetic beads, respectively. The aptamer-modified magnetic beads were characterized and successfully applied to one-step purification and immobilization of PGUS-E from the complex cell lysates. By conveniently adjusting the pH and ion strength, the PGUS-E purities reached 84% for Apt5-modified beads and 88% for Apt9-modified beads. Moreover, the maximum PGUS-E capturing capacity of the Apt5 and Apt9 modified magnetic beads were found to be 31.75μg/mg and 32.95μg/mg, respectively. The immobilized PGUS-E on aptamer-based magnetic beads showed good reusability, and the conversion of glycyrrhizin still remained more than 70% after 7 cycles. In addition, the aptamer-modified beads support can be easily regenerated, and the conversion rate of glycyrrhizin (GL) was still 62% after the 7th cycle of regeneration. This investigation can be easily extended to other enzyme systems and may help open a generic route to develop a novel enzyme immobilization technology for biocatalysis based on aptamer.

Abstract:Polyclonal antibody bound cellulose support has been exploited for the immobilization and stabilization of β galactosidase from Aspergillus oryzae. Immunoaffinity bound β galactosidase retained 96.5% of the initial activity on the support. Immobilized β galactosidase showed broad-spectrum pH optima, pH 4.6–5.5 and temperature at 50–60°C whereas the soluble enzyme exhibited activity peak at pH 4.6 and 50°C. Immunoaffinity bound enzyme preparation was quite stable to thermal denaturation and it retained 72% activity after 4h incubation at 60°C whereas under these conditions the soluble β galactosidase lost almost its full activity. IgG-cellulose immobilized β galactosidase was quite stable against the inactivation caused by proteolytic enzymes; trypsin and pepsin. Moreover, this immobilized enzyme preparation retained 64%, 71% and 65% activity in the presence of 4.0M urea, 5% CaCl2 and 5% galactose. IgG-cellulose bound β galactosidase exhibited 80% of its original activity after 2 months storage at 4°C while the soluble enzyme showed only 35% of the initial activity. After 10th repeated use immobilized β galactosidase retained 46% activity.

Abstract:With proper design, immobilization can be useful tool to improve the stability of enzymes, and in certain cases even their activity, selectivity, productivity and economic viability. An immobilized β-glucosidase (BGL, EC 3.2.1.21) through matrix adsorption and cross-linked enzyme aggregate (ad-CLEA) technology is presented in this work. After adsorption and precipitation, BGL was immobilized to poly(glycidyl methacrylate-co-ethylenedimethacrylate) (PGMA/EDMA) microparticles using glutaraldehyde as the cross-linker. Immobilized BGL exhibits lower apparent Km but much higher Vmax than that of the soluble enzyme, suggesting greater enzyme–substrate affinity and rapid velocity. Besides, ad-CLEA-BGL presents better thermostability retaining activity nearly 70% for 3 h and approximately 50% for 5 h at 70 °C, high operational reusability remaining more than 90% activity after nine uses and excellent storage stability holding about 95% activity after 45 days. Furthermore, the cellobiose is completely hydrolyzed within 1 h with ad-CLEA-BGL, which is significantly more efficient than soluble enzyme (about 4 h). Therefore, BGL was successfully immobilized on PGMA/EDMA microparticles with an ad-CLEA technology and the immobilization greatly enhances the biochemical characteristics. This work indicates promising application for ad-CLEA-BGL in utilizing agricultural remnants, bio-converting cellobiose to fermentable reducing sugar and ethanol production.

Abstract:In this study, a novel screening approach is presented, merging capillary electrophoresis analysis with an enzymatic assay employing immobilized α-glucosidase. A metal–organic framework composite membrane was devised for α-glucosidase immobilization. Employing polyvinylidene fluoride (PVDF) as the substrate material, it was functionalized with dopamine, creating a polydopamine composite layer. This layer acted as a covalent linker, anchoring Zn 2+ , which subsequently chelated with the organic ligand 2-ethylimidazole, leading to the self-assembly of a three-dimensional porous framework ZIF-8. This structure facilitated the in situ encapsulation of α-glucosidase. Characterization techniques including scanning electron microscopy, Fourier transform infrared-attenuated total reflectance spectroscopy, X-ray photoelectron spectroscopy, and X-ray diffraction validated the composite material's structure. A thorough investigation of various parameters affecting immobilization was conducted. The immobilized α-glucosidase exhibited enhanced pH tolerance, thermal stability, and remarkable reusability, retaining 79.3% of its original activity over 10 cycles, with a batch-to-batch reproducibility of 5.9% ( n = 5). Its Michaelis–Menten constant ( K m ) was determined to be 2.85 mM. Application of this immobilized enzyme in screening potential inhibitors from 13 traditional Chinese medicines revealed Sanguisorba radix as the most potent inhibitor. This study underscores the potential of this immobilized enzyme screening strategy for identifying enzyme inhibitors in traditional Chinese medicines.

Abstract:β-galactosidase from Aspergillus oryzae was immobilized in fibers composed of alginate and gelatin hardened with glutaraldehyde. Immobilization resulted in 56% relative activity retained for 35 days without decrease. The optimum conditions were not affected by immobilization, and the optimum pH and temperature for free and immobilized enzyme were 4.5 and 50°C, respectively. Immobilized β-galactosidase was more stable at high pH and temperatures. The kinetic parameters for soluble and immobilized β-galactosidase were also determined. The newly developed immobilization method is simple, yet effective and can be used for the immobilization of other enzymes.

Abstract:Industrial application of α-galactosidase requires efficient methods to immobilize the enzyme, yielding a biocatalyst with high activity and stability compared to free enzyme. An α-galactosidase from tomato fruit was immobilized on galactose-containing polymeric beads. The immobilized enzyme exhibited an activity of 0.62U/g of support and activity yield of 46%. The optimum pH and temperature for the activity of both free and immobilized enzymes were found as pH 4.0 and 37°C, respectively. Immobilized α-galactosidase was more stable than free enzyme in the range of pH 4.0–6.0 and more than 85% of the initial activity was recovered. The decrease in reaction rate of the immobilized enzyme at temperatures above 37°C was much slower than that of the free counterpart. The immobilized enzyme shows 53% activity at 60°C while free enzyme decreases 33% at the same temperature. The immobilized enzyme retained 50% of its initial activity after 17 cycles of reuse at 37°C. Under same storage conditions, the free enzyme lost about 71% of its initial activity over a period of 7 months, whereas the immobilized enzyme lost about only 47% of its initial activity over the same period. Operational stability of the immobilized enzyme was also studied and the operational half-life (t1/2 was determined as 6.72h for p-nitrophenyl α-d-galactopyranoside (PNPG) as substrate. The kinetic parameters were determined by using PNPG as substrate. The Km and Vmax values were measured as 1.07mM and 0.01U/mg for free enzyme and 0.89mM and 0.1U/mg for immobilized enzyme, respectively. The synthesis of the galactose-containing polymeric beads and the enzyme immobilization procedure are very simple and also easy to carry out.
