Abstract:In this work, novel cuprous oxide-demethyleneberberine (Cu2O-DMB) nanomaterials are successfully synthesized for photoresponsive-enhanced enzymatic synergistic antibacterial therapy under near-infrared (NIR) irradiation (808 nm). Cu2O-DMB has a spherical morphology with a smaller nanosize and positive ζ potential, can trap bacteria through electrostatic interactions resulting in a targeting function. Cu2O-DMB nanospheres show both oxidase-like and peroxidase-like activities, and serve as a self-cascade platform, which can deplete high concentrations of GSH to produce O2˙- and H2O2, then H2O2 is transformed into ˙OH, without introducing exogenous H2O2. At the same time, Cu2O-DMB nanospheres become photoresponsive, producing 1O2 and having an efficient photothermal conversion effect upon NIR irradiation. The proposed mechanism is that the generated ROS (O2˙-, ˙OH and 1O2) and hyperthermia can have synergetic effects for killing bacteria. Moreover, hyperthermia is not only beneficial for destroying bacteria, but also effectively enhances the efficiency of ˙OH production and accelerates GSH oxidation. Upon NIR irradiation, Cu2O-DMB nanospheres exhibit excellent antibacterial ability against methicillin-resistant Staphylococcus aureus (MRSA) and ampicillin-resistant Escherichia coli (AREC) with low cytotoxicity and bare bacterial resistance, destroy the bacterial membrane causing an efflux of proteins and disrupt the bacterial biofilm formation. Animal experiments show that the Cu2O-DMB + NIR group can efficiently treat MRSA infection and promote wound healing. These results suggest that Cu2O-DMB nanospheres are effective materials for combating bacterial infections highly efficiently and to aid the development of photoresponsive enzymatic synergistic antibacterial therapy.
Nanoarchitectonics of nitric oxide releasing supramolecular structures for enhanced antibacterial efficacy under visible light irradiation

Abstract:Light-controlled therapies offer a promising strategy to prevent and suppress infections caused by numerous bacterial pathogens. Excitation of exogenously supplied photosensitizers (PS) at specific wavelengths elicits levels of reactive oxygen intermediates toxic to bacteria. Porphyrin-based supramolecular nanostructure frameworks (SNF) are effective PS with unique physicochemical properties that have led to their widespread use in photomedicine. Herein, we developed a nitric oxide (NO) releasing, biocompatible, and stable porphyrin-based SNF (SNF-NO), which was achieved through a confined noncovalent self-assembly process based on π-π stacking. Characterization of the SNFs via scanning electron microscopy (SEM) and transmission electron microscopy (TEM) analysis showed the formation of three-dimensional, well-defined octahedral structures. These SNF-NO were shown to exhibit a red shift due to the noncovalent self-assembly of porphyrins, which also show extended light absorption to broadly cover the entire visible light spectrum to enhance photodynamic therapy (PDT). Under visible light irradiation (46 J cm-2), the SNF generates high yields of singlet oxygen (1O2) radicals, hydroxyl radicals (HO), superoxide radicals (O2), and peroxynitrite (ONOO-) radicals that have shown potential to enhance antimicrobial photodynamic therapy (APDT) against Gram-positive methicillin-resistant Staphylococcus aureus (MRSA) and Gram-negative Escherichia coli (E. coli). The resulting SNFs also exhibit significant biofilm dispersion and a decrease in biomass production. The combination of robust photosensitizer SNFs with nitric oxide-releasing capabilities is dynamic in its ability to target pathogenic infections while remaining nontoxic to mammalian cells. The engineered SNFs have enormous potential for treating and managing microbial infections.
Oxygen‐Tolerant Photoredox Catalysis Triggers Nitric Oxide Release for Antibacterial Applications

Abstract:Photoredox catalysis has emerged as a robust tool for chemical synthesis. However, it remains challenging to implement photoredox catalysis under physiological conditions due to the complex microenvironment and the quenching of photocatalyst by biologically relevant molecules such as oxygen. Here, we report that UV-absorbing N,N '-dinitroso-1,4-phenylenediamine derivatives can be selectively activated by fac-Ir(ppy)(3) photocatalyst within micellar nanoparticles under visible light irradiation (e.g., 500 nm) through photoredox catalysis in aerated aqueous solutions to form quinonediimine (QDI) residues with concomitant release of NO. Notably, the formation of QDI derivatives can actively scavenge the reactive oxygen species generated by fac-Ir(ppy)(3), thus avoiding oxygen quenching of the photocatalyst. Further, we exemplify that the oxygen-tolerant photoredox catalysis-mediated NO release can not only kill planktonic bacteria in vitro but also efficiently treat MRSA infections in vivo.

Abstract:After numerous efforts to elucidate the biological role of nitric oxide (NO), NO treatments have become a hotspot at the forefront of medicine. NO-releasing substances are constantly needed, while the direct use of NO gas is unattainable in bio-systems. An ideal NO donor should possess controllable and visible NO-release capability. The reported NO donating nanoparticles, prepared via encapsulating a hydrophobic NO-releasing compound into DSPE-PEG 2000 , meet the criteria mentioned previously. The localization and flux of NO released from these nanoparticles could be manipulated by UV or blue light. Meanwhile, NOD-NPs emit a dose-dependent fluorescence intensity to calibrate the generation of NO. While the good biocompatibility of NOD-NPs has been validated, the NO from our nanoparticles demonstrates efficient anti-bacterial and anti-biofilm effects toward Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Therefore, the NOD-NPs developed in this work have potential application in evaluating the regulation of microbes by NO.

Abstract:This study investigates the incorporation of active secondary amine moieties into the polymer backbone by co‐polymerizing 2,4,6‐tris(chloromethyl)‐mesitylene (TCM) with three diamines, namely 1,4‐diaminobutane (DAB), m‐phenylenediamine (MPD), and p‐phenylenediamine (PPD). This process results in the stabilisation of the amine moieties and the subsequently introduced nitroso groups. Charging bioactive nitric oxide (NO) into the polymers is accomplished by converting the amine moieties into N‐nitroso groups. The ability of the polymers to store and release NO depends on their structures, particularly the amounts of incorporated active secondary amines. With grafting photosensitive N‐nitroso groups into the polymers, the derived NO@polymers exhibit photoresponsivity. NO release is completely regulated by adjusting UV light irradiation. These resulting polymeric NO donors demonstrate remarkable bactericidal and bacteriostatic activity, effectively eradicating E. coli bacteria and inhibiting their growth. Abstract:The strict dependence of the biological effects of nitric oxide (NO) on its concentration and generation site requires this inorganic free radical to be delivered with precise spatiotemporal control. Light-activation by suitable NO photoprecursors represents an ideal approach. Developing strategies to activate NO release using long-wavelength excitation light in the therapeutic window (650–1300 nm) is challenging. In this contribution, we demonstrate that NO release by a blue-light activatable NO photodonor (NOPD) with self-fluorescence reporting can be triggered catalytically by the much more biocompatible red light exploiting a supramolecular photosensitization process. Different red-light absorbing photosensitizers (PS) are co-entrapped with the NOPD within different biocompatible nanocarriers such as Pluronic micelles, microemulsions and branched cyclodextrin polymers. The intra-carrier photosensitized NO release, involving the lowest, long-lived triplet state of the PS as the key intermediate and its quenching by the NOPD, is competitive with that by molecular oxygen. This allows NO to be released with good efficacy, even in aerobic conditions. Therefore, the adopted general strategy provides a valuable tool for generating NO from an already available NOPD, otherwise activatable with the poorly biocompatible blue light, without requiring any chemical modification and using sophisticated and expensive irradiation sources.

Abstract:Novel materials for the phototherapeutic release of the bioregulator nitric oxide (nitrogen monoxide) are described. Also reported is a method for scanning these materials with a focused NIR beam to induce photouncaging while minimizing damage from local heating. The new materials consist of poly(dimethylsiloxane) composites with near-infrared-to-visible upconverting nanoparticles (UCNPs) that are cast into a biocompatible polymer disk (PD). These PDs are then impregnated with the photochemical nitric oxide precursor Roussin's black salt (RBS) to give UCNP_RBS_PD devices that generate NO when irradiated with 980 nm light. When the UCNP_RBS_PD composites were irradiated with NIR light through filters composed of porcine tissue, physiologically relevant NO concentrations were released, thus demonstrating the potential of such devices for minimally invasive phototherapeutic applications.

Abstract:The peroxynitrite anion (ONOO- ) is closely associated with many diseases and the creation of ONOO- donors is an essential means of understanding its pathophysiological functions. However, it is challenging to develop ONOO- donors due to the difficulties in simultaneously producing highly reactive and short-lived nitric oxide (NO) and superoxide anion (O2 ⋅- ). Here, we report a novel strategy for constructing ONOO- donors by combining near-infrared (NIR)-mediated type I photosensitization and photoredox catalysis. The key design using a Nile blue analogue that can serve as both a type I photosensitizer and a metal-free photocatalyst. Intriguingly, the formation of O2 ⋅- via type I photosensitization avoids oxygen interference and instead activates nitrobenzofurazan-based NO donors via oxygen-tolerant NIR photoredox catalysis. The simultaneous release of O2 ⋅- and NO leads to ONOO- release, showing both antibacterial and antibiofilm activities.

Abstract:As an emerging bactericidal strategy, photodynamic therapy (PDT) exhibits great potential to combat various pathogenic infections, but its antibacterial efficacy may be compromised, generally owing to its shallow penetrability of short-wavelength (UV/Vis) light as well as short diffusion length and lifespan of the generated reactive oxygen species (ROS). Herein, we present a method for developing a nitric oxide (NO)-assisted PDT nanocomposite membrane with significantly enhanced bactericidal performances, upon a single near-infrared (NIR) light irradiation. Hierarchically structured nanoparticles (UCNP@PCN), composed of upconversion nanoparticles (UCNPs) and porphyrinic MOFs (PCN-224) are initially prepared. After being doped with L-arginine (LA), the nanoparticles are incorporated into polyvinylidene fluoride (PVDF) matrix to get electrospun nanocomposite membrane (UCNP@PCN@LA-PVDF). Upon NIR light irradiation (980 nm), this UCNP@PCN@LA-PVDF can generate sufficient ROS, which not only acts as main bactericidal agent in PDT but also induces the loaded LA to produce NO, eventually realizing the NO-assisted PDT antibacterial behavior. The generated NO exhibits a larger sterilization area as compared to that of the ROS, demonstrating a vital complementary advantage in facilitating PDT antibacterial efficacy as proven in vitro and in vivo tests. This type of nanocomposite membrane may find unique biomedical applications where both highly enhanced PDT antibacterial effects and deep penetration of NIR light are desired.
Lignin-Based Visible Light-Triggered Nitric Oxide Nanogenerator for Antibacterial Applications

Abstract:Nitric oxide (NO) has received growing attention as an effective antibacterial agent with broad-spectrum activity and a low risk of resistance. However, it remains challenging to develop effective, controllable, and biocompatible NO-releasing materials. Here, we report a novel NO nanogenerator (AL-BNN6-PEG) self-assembled by lignin, a UV-absorbing and hydrophobic NO donor (N,N'-disec-butyl-N,N'-dinitroso-1,4-phenylenediamine, BNN6), and PEG-DSPE2000. It was discovered that upon visible light irradiation (450-460 nm), BNN6 can be decomposed by lignin within micellar nanoparticles via a photoinduced electron transfer mechanism in the aqueous medium. Lignin not only served as a sustainable carrier, enhancing the water dispersity of BNN6, but also acted as a biocompatible photosensitizer, triggering BNN6 decomposition with the concomitant release of NO. As a result, the micellar nanoparticles displayed superior antibacterial effects against Gram-negative and Gram-positive bacteria upon visible light illumination. Moreover, MTT assay revealed the negligible cytotoxic effect of the micellar nanoparticles to the mouse fibroblast cells (L929). This research provides more insight into the BNN6 decomposition mechanism and demonstrates a straightforward, effective, and biocompatible strategy for controlled NO-mediated antibacterial applications.
Photo-Controllable Burst Generation of Peroxynitrite Based on Synergistic Interactions of Polymeric Nitric Oxide Donors and IR780 for Enhancing Broad-Spectrum Antibacterial Therapy

Abstract:The newly attractive peroxynitrite (ONOO − ) therapy can prominently enhance antibacterial therapeutic efficacy. However, it is a great challenge but urgently needed to generate ONOO − with adjustable release rate and dosage in order to satisfy personalized treatments for different disease types and severities. Herein, nanoparticles are fabricated via co-assembly of an amphiphilic PEG- b -PAASNO block copolymer grafted with abundant nitric oxide (NO) donor units and IR780 as a photothermal and photodynamic agent. Photo-controllable burst generation of ONOO − from nanoparticles could be realized based on synergistic reactions of rapid NO release induced by increased local temperature and efficiently produced superoxide anion radical (O 2 •− ) from IR780. The maximum ONOO − release dosage is up to 6.73 ± 0.07 μM and release rate is up to 98.1 ± 1.38 nM/s. Furthermore, the ONOO − release behavior can be precisely manipulated by varying sample concentrations, irradiated durations, output power densities, and laser switchs, respectively. Ultra-efficiently generated ONOO − from biocompatible nanoparticles significantly elevated broad spectrum antibacterial efficiency through damaging bacterial membranes. Thus, nanoparticles may present a new insight into preparation of burst and controllable generating ONOO − materials, and provide new opportunities for antibacterial therapy. Statement of significance : 1. Polymeric NO donor (PEG- b -PAASNO) grafted with abundant NO donor units was synthesized. 2. nanoparticles were prepared by co-assembly of IR780 and amphiphilic PEG- b -PAASNO polymer. 3. Abstract:Bacterial infection poses a massive threat to our society, and bacterial biofilm is a major cause of chronic and recurrent infections. The treatment of bacterial biofilms represents a challenging task, and the development of antibacterial materials that can not only disperse bacterial biofilms but also kill bacteria is of increasing interest. Herein, we report the fabrication of well-defined nitric oxide (NO)-releasing amphiphiles, poly(ethylene oxide)-b-polyCouNO (PEO-b-PCouNO), where CouNO is an N-nitrosoamine-based NO donor containing a coumarin chromophore, exhibiting visible-light-mediated and self-reporting NO-release behavior. Unlike conventional polymeric NO donors derived from N-diazeniumdiolate (NONOates) or N-nitrosothiol (SNOs) that could be only synthesized via the postmodification procedure due to poor stability, the newly developed Nnitrosoamine-based NO donors can be directly polymerized into amphiphiles using reversible addition-fragmentation chain transfer (RAFT) polymerization. The NO-releasing amphiphiles self-assembled into micelles and selective NO release in aqueous medium was achieved by irradiating the micelle solution with visible light, which was characterized by a remarkable fluorescence turn-on (>185-fold), thereby enabling in situ self-reporting NO release. The photoinduced NO release can efficiently disperse bacterial biofilm of Pseudomonas aeruginosa. Moreover, antibiotics (e.g., Ciprofloxacin, Cip) could be loaded into the NO-releasing micelles, and co-delivery of NO and Cip was achieved, allowing for simultaneous biofilm dispersal and bacterial killing. This work provides a new strategy to fabricate macromolecular NO donors, which can efficiently avoid uncontrolled NO leakage and display promising antibacterial applications.

Abstract:The pressing need for highly efficient antibacterial strategies arises from the prevalence of microbial biofilm infections and the emergence of rapidly evolving antibiotic-resistant strains of pathogenic bacteria. Photodynamic therapy represents a highly efficient and compelling antibacterial approach, offering promising prospects for effective control of the development of bacterial resistance. However, the effectiveness of many photosensitizers is limited due to the reduced generation of reactive oxygen species (ROS) in hypoxic microenvironment, which commonly occur in pathological conditions such as inflammatory and bacteria-infected wounds. Herein, we designed and prepared two phenothiazine-derived photosensitizers (NB-1 and NB-2), which can effectively generate superoxide anion radicals (O 2 ●– ) through the type I process. Both photosensitizers demonstrate significant efficacy in vitro for the eradication of broad-spectrum bacteria. Moreover, NB-2 possesses distinct advantages including strong membrane binding and strong generation of O 2 ●– , rendering it an exceptionally efficient antibacterial agent against mature biofilms. In addition, laser activated NB-2 could be applied to treat MRSA -infected wound in vivo , which offers new opportunities for potential practical applications.

Abstract:Background: Periodontal disease, an oral disease that initiates with plaque biofilm infection, affects 10% of the global population. Due to the complexity of tooth root anatomy, biofilm resistance and antibiotic resistance, traditional mechanical debridement and antibiotic removal of biofilms are not ideal. Nitric oxide (NO) gas therapy and its multifunctional therapy are effective methods to clear biofilms. However, large and controlled delivery of NO gas molecules is currently a great challenge. Methods: The core-shell structure of Ag2S@ZIF-90/Arg/ICG was developed and characterized in detail. The ability of Ag2S@ZIF-90/Arg/ICG to produce heat, ROS and NO under 808 nm NIR excitation was detected by an infrared thermal camera, probes and Griess assay. In vitro anti-biofilm effects were evaluated by CFU, Dead/Live staining and MTT assays. Hematoxylin-eosin staining, Masson staining and immunofluorescence staining were used to analyze the therapeutic effects in vivo. Results: Antibacterial photothermal therapy (aPTT) and antibacterial photodynamic therapy (aPDT) could be excited by 808 nm NIR light, and the produced heat and ROS further triggered the release of NO gas molecules simultaneously. The antibiofilm effect had a 4-log reduction in vitro. The produced NO caused biofilm dispersion through the degradation of the c-di-AMP pathway and improved biofilm eradication performance. In addition, Ag2S@ZIF-90/Arg/ICG had the best therapeutic effect on periodontitis and NIR II imaging ability in vivo. Conclusions: We successfully prepared a novel nanocomposite with NO synergistic aPTT and aPDT. It had an outstanding therapeutic effect in treating deep tissue biofilm infection. This study not only enriches the research on compound therapy with NO gas therapy but also provides a new solution for other biofilm infection diseases.

Abstract:The achievement of smart pharmaceuticals whose bioactivity can be spatiotemporally controlled by light stimuli is known as photopharmacology, an emerging area aimed at improving the therapeutic outcome and minimizing side effects. This is especially attractive for antibiotics, for which the inevitable development of multidrug resistance and the dwindling of new clinically approved drugs represent the main drawbacks. Here, we show that nitrosation of the fluoroquinolone norfloxacin ( NF ), a broad-spectrum antibiotic, leads to the nitrosated bioconjugate NF–NO , which is inactive at the typical minimum inhibitory concentration of NF . Irradiation of NF–NO with visible blue light triggers the simultaneous release of NF and nitric oxide (NO). The photouncaging process is accompanied by the revival of the typical fluorescence emission of NF , quenched in NF–NO , which acts as an optical reporter. This permits the real-time monitoring of the photouncaging process, even within bacteria cells where antibacterial activity is switched on exclusively upon light irradiation. The mechanism of photorelease seems to occur through a two-step hopping electron transfer mediated by the lowest triplet state of NF–NO and the phosphate buffer ions or aminoacids such as tyrosine. Considering the well-known role of NO as an "unconventional" antibacterial, the NF–NO conjugate may represent a potential bimodal antibacterial weapon activatable on demand with high spatio-temporal control.

However, finding effective strategies to mitigate the detrimental effects of excessive temperature and elevated concentrations of reactive oxygen species (ROS) remains a pressing issue that requires immediate attention. Methods: In this study, we designed a pH-responsive cationic polymer sodium nitroside dihydrate/branched polyethylenimine–indocyanine green@polyethylene glycol (SNP/PEI-ICG@PEG) nanoplatform using the electrostatic adsorption method and Schiff's base reaction. Relevant testing techniques were applied to characterize and analyze SNP/PEI-ICG@PEG, proving the successful synthesis of the nanomaterials. In vivo and in vitro experiments were performed to evaluate the antimicrobial properties of SNP/PEI-ICG@PEG. Results: The morphology and particle size of SNP/PEI-ICG@PEG were observed via TEM. The zeta potential and UV-visible (UV‐vis) results indicated the synthesis of the nanomaterials. The negligible cytotoxicity of up to 1 mg/mL of SNP/PEI-ICG@PEG in the presence or absence of light demonstrated its biosafety. Systematic in vivo and in vitro antimicrobial assays confirmed that SNP/PEI-ICG@PEG had good water solubility and biosafety and could be activated by near-infrared (NIR) light and synergistically treated using four therapeutic modes, photodynamic therapy (PDT), gaseous therapy (GT), mild photothermal therapy (PTT, 46 °C), and cation. Ultimately, the development of Gram-positive (G + ) Staphylococcus aureus ( S. aureus ) and Gram-negative (G − ) Escherichia coli ( E. coli ) were both completely killed in the free state, and the biofilm that had formed was eliminated. Conclusion: SNP/PEI-ICG@PEG demonstrated remarkable efficacy in achieving controlled multimodal synergistic antibacterial activity and biofilm infection treatment. The nanoplatform thus holds promise for future clinical applications. The severity of infection primarily relies on the virulence of bacteria and may involve biofilm formation. Biofilms, as umbrellas for free-living bacteria, are microcommunities of large number of free-living bacteria encapsulated in the extracellular polymeric substance (EPS) they produce. Biofilms tend to resist the host's immune system, reduce antibiotic susceptibility, and impede antibiotic leakage, making them difficult to treat. Abstract:It is of particular interest to develop new antibacterial agents with low risk of drug resistance development and low toxicity toward mammalian cells to combat pathogen infections. Although gaseous signaling molecules (GSMs) such as nitric oxide (NO) and formaldehyde (FA) have broad-spectrum antibacterial performance and the low propensity of drug resistance development, many previous studies heavily focused on nanocarriers capable of delivering only one GSM. Herein, we developed a micellar nanoparticle platform that can simultaneously deliver NO and FA under visible light irradiation. An amphiphilic diblock copolymer of poly(ethylene oxide)-b-poly(4-((2-nitro-5-(((2-nitrobenzyl)oxy)methoxy)benzyl)(nitroso)amino)benzyl methacrylate) (PEO-b-PNNBM) was successfully synthesized through atom transfer radical polymerization (ATRP). The resulting diblock copolymer self-assembled into micellar nanoparticles without premature NO and FA leakage, whereas they underwent phototriggered disassembly with the corelease of NO and FA. We showed that the NO- and FA-releasing micellar nanoparticles exhibited a combinatorial antibacterial performance, efficiently killing both Gram-negative (e.g., Escherichia coli) and Gram-positive (e.g., Staphylococcus aureus) bacteria with low toxicity to mammalian cells and low hemolytic property. This work provides new insights into the development of GSM-based antibacterial agents.

Abstract:Biofilm has resulted in numerous obstinate clinical infections, posing severe threats to public health. It is urgent to develop original antibacterial strategies for eradicating biofilms. Herein, we develop a surface charge switchable supramolecular nanocarrier exhibiting pH-responsive penetration into an acidic biofilm for nitric oxide (NO) synergistic photodynamic eradication of the methicillin-resistant Staphylococcus aureus (MRSA) biofilm with negligible damage to healthy tissues under laser irradiation. Originally, by integrating the glutathione (GSH)-sensitive α-cyclodextrin (α-CD) conjugated nitric oxide (NO) prodrug (α-CD-NO) and chlorin e6 (Ce6) prodrug (α-CD-Ce6) into the pH-sensitive poly(ethylene glycol) (PEG) block polypeptide copolymer (PEG-(KLAKLAK)2-DA) via host-guest interaction, the supramolecular nanocarrier α-CD-Ce6-NO-DA was finely prepared. The supramolecular nanocarrier shows complete surface charge reversal from negative charge at physiological pH (7.4) to positive charge at acidic biofilm pH (5.5), promoting efficient penetration into the biofilm. Once infiltrated into the biofilm, the nanocarrier exhibits rapid NO release triggered by the overexpressed GSH in the biofilm, which not only produces abundant NO for killing bacteria but also reduces the biofilm GSH level to improve photodynamic therapy (PDT) efficiency. On the other hand, NO can react with reactive oxygen species (ROS) to produce reactive nitrogen species (RNS), further improving the PDT efficiency. Due to the effective penetration into the biofilm and depletion of biofilm GSH, the surface charge switchable GSH-sensitive NO nanocarrier can greatly improve the PDT efficiency at a low photosensitizer dose and laser intensity and cause negligible side effect to healthy tissues. Considering the above advantages, the strategy developed in this work may offer great possibilities to fight against biofilm infections.
