Abstract:Soil sterilization integrated with agronomic measures is an effective method to reduce soilborne replant diseases. However, the effect of vermicompost or biochar application after soil sterilization on soilborne diseases is poorly understood. A pot experiment was conducted in American ginseng to investigate the effects of vermicompost (VF), biochar (BF), and a combination of vermicompost and biochar (VBF) applied after soil sterilization on the incidence of Fusarium root rot using natural recovery (F) as control. After one growing season, the disease index of root rot, the phenolic acids, and the microbial communities of American ginseng rhizosphere soil were analyzed. The disease index of VF, BF, and VBF decreased by 33.32%, 19.03%, and 80.96%, respectively, compared with F. The highest bacterial richness and diversity were observed in the rhizosphere soil of VBF. Besides, VF and VBF significantly increased the relative abundance of beneficial bacteria (Pseudomonas, Lysobacter, and Chryseolinea) in the rhizosphere soil Higher concentrations of vanillin, one of the phenolic acids in the roots exudates, were recorded in the rhizosphere soils of BF and VBF. The vanillin concentration showed a significant negative correlation with the disease index. To conclude, vermicompost improved the beneficial bacteria of the rhizosphere soil, while biochar regulated the allelopathic effect of the phenolic acids. The study proposes a combined application of biochar and vermicompost to the rhizosphere soil to control Fusarium root rot of replanted American ginseng effectively.Key pointsVermicompost improves the relative abundance of rhizosphere beneficial bacteria.Biochar inhibits the degradation of phenolic acids by adsorption.The combination of vermicompost and biochar enhances the disease control effect.

Abstract:Root rot caused by Fusarium oxysporum severely reduces ginseng root yield and quality, leading to large economic losses. Biological control has attracted extensive attention because it is safe, environmentally friendly, and sustainable. Screening for biocontrol strains to suppress soil-borne diseases is important for improving ginseng quality. A Bacillus velezensis strain was isolated from the forest rhizosphere of Pinus sylvestris could inhibited F. oxysporum in ginseng. To explore the strain’s biocontrol effects and mechanism of action against F. oxysporum, four treatments were established in pot experiments: no inoculation (CK), F. oxysporum inoculation (F), B. velezensis YW 17 (B), and mixed inoculation of B. velezensis and F. oxysporum (BF). After 4 months of cultivation, the ginseng plants in the F treatment grew more slowly, and the withering degree was most serious among all the treatments. The results from high-throughput sequencing showed that the relative abundance of Fusarium in the roots of the BF treatment was significantly lower than that of the F treatment. B. velezensis inoculation improved the relative abundance of other beneficial microorganisms in the ginseng rhizosphere, such as Arthrobacter and Mortierella. The experimental results indicated that YW17 had strong abilities to produce biofilms and IAA, and could colonize roots. The whole genome sequencing results showed that some genes (trpA-C, gatA) were involved in the synthesis of the plant growth-promoting hormone IAA and other genes (efp, hfq, epsA-O, spo0A, sinI/sinR, lysC, yjbB, flgK, fliD, srfABC) were involved in biofilm formation and root colonization. The results of whole genome sequencing showed that there are 18 gene clusters related to the synthesis of secondary metabolite with antifungal activities. In addition, some genes involved in the biosynthetic pathway of acetoin, such as ilvB and alsD, were also found. Based on the above results, we infer that B. velezensis YW17 could inhibit pathogenic F. oxysporum by secreting antifungal lipopeptides, proteins and volatile substances, thereby indirectly protecting ginseng from pathogenic fungal infections. These results indicated that B. velezensis could control ginseng root rot indirectly by regulating the root and rhizosphere microbial community structures and directly by secreting antifungal substances.

Biochar increases Panax notoginseng's survival under continuous cropping by improving soil properties and microbial diversity

Abstract:Replant problem is widespread in agricultural production and causes serious economic losses, which has limited sustainable cultivation of Panax notoginseng (PN), a well-known medicinal plant in Asia. Here we conducted a field experiment to investigate the effectiveness and possible mechanisms of biochar to improve its survival under continuous cropping. Biochar from tobacco stems was applied at 4 rates of 9.0, 12, 15, and 18 t/ha to a soil where PN has been continuously cultivated for 10 years. After 18 months, soil properties, 5 allelochemicals, including phydroxybenzoic acid, vanillic acid, syringic acid, p-coumaric acid, and ferulic acid, key pathogen Fusarium oxysporum, microbial community, and PN survival rate were investigated. Our results show that 10 years' continuous PN cropping led to soil acidification, accumulation of NH4+-N and F. oxysporum, and low PN survival rate. However, biochar increased its survival rate from 6.0% in the control to 69.5% under 15 t/ha treatment. Moreover, soil pH, available P and K, organic matter content, and microbial diversity were increased while NH4+-N and allelochemicals vanillic acid and syringic acid contents were decreased under biochar treatment (P<0.05). Soil available K increased from 177 to 283 mg center dot kg(-1) while NH4+-N decreased from 6.73 to 4.79 mg center dot kg(-1) under 15 t/ha treatment. Further, soil pH, available P and K, and microbial diversity (bacteria and fungi) were positively correlated with PN survival rate, however, NH4+-N content was negatively correlated (P<0.05). Our study indicates that biochar effectively increased the survival rate of Panax notoginseng under continuous cropping by improving soil properties andmicrobial diversity.

Abstract:The role of biochar–microbe interaction in plant rhizosphere mediating soil-borne disease suppression has been poorly understood for plant health in field conditions. Chinese ginseng ( Panax ginseng C. A. Meyer) is widely cultivated in Alfisols across Northeast China, being often stressed severely by pathogenic diseases. In this study, the topsoil of a continuously cropped ginseng farm was amended at 20 t ha –1 , respectively, with manure biochar (PB), wood biochar (WB), and maize residue biochar (MB) in comparison to conventional manure compost (MC). Post-amendment changes in edaphic properties of bulk topsoil and the rhizosphere, in root growth and quality, and disease incidence were examined with field observations and physicochemical, molecular, and biochemical assays. In the 3 years following the amendment, the increases over MC in root biomass were parallel to the overall fertility improvement, being greater with MB and WB than with PB. Differently, the survival rate of ginseng plants increased insignificantly with PB but significantly with WB (14%) and MB (21%), while ginseng root quality was unchanged with WB but improved with PB (32%) and MB (56%). For the rhizosphere at harvest following 3 years of growing, the total content of phenolic acids from root exudate decreased by 56, 35, and 45% with PB, WB, and MB, respectively, over MC. For the rhizosphere microbiome, total fungal and bacterial abundance both was unchanged under WB but significantly increased under MB (by 200 and 38%), respectively, over MC. At the phyla level, abundances of arbuscular mycorrhizal and Bryobacter as potentially beneficial microbes were elevated while those of Fusarium and Ilyonectria as potentially pathogenic microbes were reduced, with WB and MB over MC. Moreover, rhizosphere fungal network complexity was enhanced insignificantly under PB but significantly under WB moderately and MB greatly, over MC. Overall, maize biochar exerted a great impact rather on rhizosphere microbial community composition and networking of functional groups, particularly fungi, and thus plant defense than on soil fertility and root growth.

Abstract:Medicine plants such as Panax notoginseng (Sanqi) frequently experience replant failure owing to the accumulation of soilborne pathogens and autotoxins in continuous cropping soil. Bacillus sp. is a commonly used biocontrol agent to combat soilborne pathogens, but further research is needed to understand its impact on controlling Sanqi root rot and the underlying mechanisms. Here, we isolated and screened a Bacillus strain, B19, from Sanqi rhizosphere soil that exhibited potential for inhibiting Sanqi root rot pathogens. We then tested the strain's effect on the incidence of Sanqi root rot in continuous cropping soil, along with its motility/chemotaxis and biofilm formation ability (i.e., potential for root colonization). We also analyzed the strain's genomic characteristics, including potential secondary metabolite synthesis gene clusters. Our results showed that the strain B19 exhibited high inhibitory activity against fungal pathogens (F. solani and F. oxysporum) and could metabolize saponins as a sole carbon source. The application of B19 to continuous cropping soil significantly reduced the incidence of root rot in Sanqi seedlings. B19 was identified as B. velezensis through morphological characteristics, 16S rRNA and gyrA gene sequence analysis, and genome-level annotation. Notably, B19′s genome encodes many genes associated with functions such as motility/chemotaxis and biofilm formation, as well as 13 biosynthetic gene clusters that could encode secondary metabolites, including surfactin, fengycin, and siderophore. Additionally, B19 demonstrated the capability to produce volatile organic compounds that inhibit pathogenic fungi. The motility/chemotaxis and biofilm-forming ability of B19 were enhanced by saponins. Collectively, these properties could enhance B19′s biocontrol ability, thereby reducing the incidence of root rot.

Abstract:Bacillus subtilis as microbial fertilizers contribute to avoiding the harmful effects of traditional agricultural fertilizers and pesticides. However, there are many restrictions on the practical application of fertilizers. In this study, microbial biochar formulations (BCMs) were prepared by loading biochar with B. subtilis SL-44. Pot experiments were conducted to evaluate the effects of the BCMs on soil fertility, Fusarium wilt control, and radish plant growth. The application of BCMs dramatically improved soil properties and favored plant growth. Compared with SL-44 and biochar treatments, the BCMs treatments increased radish plant physical-chemical properties and activities of several enzymes in the soil. What’s more, Fusarium wilt incidence had decreased by 59.88%. In addition, the BCMs treatments exhibited a significant increase in the abundance of bacterial genera in the rhizosphere soil of radish. Therefore, this study demonstrated that BCMs may be an eco-friendly strategy for improving soil fertility, reducing Fusarium wilt, and promoting radish plant growth.
Effects of Biochar on the Growth, Ginsenoside Content, and Soil Microbial Community Composition of Panax Quinquefolium L

Abstract:Biochar has shown great potential for use in agricultural systems. However, few studies have focused on its effects on the growth, active ingredients, and soil microbial community composition of Chinese medicinal herbs. The growth, photosynthetic capacity, content of active ingredients of Panax quinquefolium L, soil enzyme activity, and soil microbial community composition treated with different concentrations of biochar (0, 0.6%, 1.2%, 1.8%, and 2.4%) were investigated. Compared with the control, the plant growth, net photosynthetic rate, stomatal conductance, transpiration rate, photosynthetic carbon assimilation, chlorophyll b content, and ginsenoside (active ingredients) content of P. quinquefolium were promoted by biochar amendment. The activities of soil sucrase, urease, and acid phosphatase of P. quinquefolium were promoted by biochar application, whereas soil catalase activity and the richness and diversity of soil microbial communities in biochar treatments were reduced relative to the control. However, the dominant soil bacteria at the phylum level (such as Proteobacteria) and fungi at the genus level (such as Chalara and Sistotrema) differed significantly between the control and 1.8% biochar treatment. The linear discriminant analysis effect size (LEfSe) analysis showed that the relative abundance of some microbial taxa in the rhizosphere soil was significantly different among the control, 1.8% and 2.4% biochar additions (p < 0.05). Furthermore, the correlation analysis showed that the ginsenoside content of P. quinquefolium roots was correlated with Acidobacteria, Actinobacteria, and Verrucomicrobia, etc. (p < 0.05). The growth and ginsenoside accumulation of P. quinquefolium could be promoted, and the rhizosphere soil microbial community composition could be improved by an appropriate rate (1.8%) of biochar application.
Inoculation with Penicillium citrinum aids ginseng in resisting Fusarium oxysporum by regulating the root and rhizosphere microbial communities

Abstract:Ginseng root rot disease caused by Fusarium oxysporum severely decreases root yield and quality, causing huge economic losses. Biological control is safe, environmentally friendly and sustainable. The results from bananas, sunflowers and chickpeas showed that Penicillium citrinum could control root diseases. However, its potential to control root rot disease of the medicinal plant is unclear. Here, the effects of P. citrinum YW322 isolated from ginseng rhizosphere soil on controlling root rot disease of ginseng were evaluated using the pot experiment. Four treatments were set up: no inoculation (CK), P. citrinum inoculation (PC), F. oxysporum inoculation (FO) and mixed inoculation of P. citrinum and F. oxysporum (PCFO). The results showed that the PC treatment did not affect ginseng growth when compared with the CK treatment. The plants in FO grew more slowly and rotted more severely. Distinct from FO, the ginseng plants in the PCFO treatment all grew similarly to CK. Plate confrontation experiments showed that P. citrinum YW322 inhibited the mycelium growth of F. oxysporum YFW32 by 95.75%. After 7 days of cultivation, the filtrate secreted nonvolatile inhibitory substances with 58.82% inhibition. The results from high-throughput sequencing showed that the structure of the bacterial community in ginseng roots and the fungal community in rhizosphere soil in the PCFO treatment were significantly different compared to the FO treatment (p < 0.05). Compared with FO, the abundance of Clostridium was significantly suppressed in the ginseng roots of PCFO treatment (93.5% reduction) (p < 0.05); significant enrichment of fungi such as Mortierella, Trichoderma and Penicillium was observed in ginseng rhizosphere soil in the PCFO treatment (8.26%, 17.44% and 2.80% increase in relative abundance, respectively). The relative abundance of the pathogenic fungus Fusarium decreased 31.85% compared with FO (p < 0.05). Soil physicochemical analysis showed that compared to the FO treatment, the soil pH and S-CAT activity in the PCFO treatment significantly increased (p < 0.05). Therefore, P. citrinum has potential to control ginseng root rot disease by directly inhibiting the growth of F. oxysporum and regulating the community structure of root bacteria and rhizosphere fungi to help ginseng resist F. oxysporum.

Abstract:Root rot is a main disease in Panax notoginseng , which is severe and difficult to control. Arbuscular mycorrhizal fungi (AMF) are known to effectively prevent plant diseases caused by soil-borne pathogens. However, the potential role of AMF as a biocontrol agent against root rot in P. notoginseng is still unclear. This study aims to investigate the efficacy of AMF, including Funneliformis mosseae, Rhizophagus intraradices and Claroideoglomus etunicatum , individually and in combination, for controlling root rot in P. notoginseng seedlings. The effect of different AMF on the indicators of P. notoginseng growth, superoxide dismutase, peroxidase activity, proline, malondialdehyde, soluble sugars and proteins were measured. The effects of different AMF on root rot resistance were comprehensively evaluated using the subordinate function method. The results showed that the incidence of root rot decreased from 75% to 12.5%–50.0% after inoculation with different AMF agents, and the disease index decreased from 22.92% to 6.25%–16.67%. Moreover, AMF significantly increased the growth parameters of P. notoginseng , such as plant height (6.35%–41.73%), stem diameter (12.98%–45.96%), fresh weight (6.28%–95.35%), and leaf area (8.89%–97.71%). Moreover, AMF also improved the content of chlorophyll (2.40%–50.67%) and rate of net photosynthetic (52.08%– 279.17%), enhancing superoxide dismutase (4.02%–114.89%) and peroxidase activity (85.60%–139.98%), as well as promoting the accumulation of soluble sugars (30.21%–90.00%), proteins (3.31%–65.81%), and proline (4.02%–114.89%) in the root and leaf, while reducing the malondialdehyde content (19.99%–139.98%). Particularly, C. etunicatum was found to be the most effective in promoting growth and disease resistance in P. notoginseng . These findings provide insights into the potential use of AMF as an effective biocontrol agent to manage root rot.
Effects of a co-bacterial agent on the growth, disease control, and quality of ginseng based on rhizosphere microbial diversity

Abstract:Background: The ginseng endophyte Paenibacillus polymyxa Pp-7250 (Pp-7250) has multifaceted roles such as preventing ginseng diseases, promoting growth, increasing ginsenoside accumulation, and degrading pesticide residues, however, these effects still have room for improvements. Composite fungicides are an effective means to improve the biocontrol effect of fungicides, but the effect of Pp-7250 in combination with its symbiotic bacteria on ginseng needs to be further investigated, and its mechanism of action has not been elucidated. In this study, a series of experiments was conducted to elucidate the effect of Paenibacillus polymyxa and Bacillus cereus co-bacterial agent on the yield and quality of understory ginseng, and to investigate their mechanism of action. Results: The results indicated that P. polymyxa and B. cereus co-bacterial agent (PB) treatment improved ginseng yield, ginsenoside accumulation, disease prevention, and pesticide degradation. The mechanism is that PB treatment increased the abundance of beneficial microorganisms, including Rhodanobacter, Pseudolabrys, Gemmatimonas, Bacillus, Paenibacillus, Cortinarius, Russula, Paecilomyces, and Trechispora, and decreased the abundance of pathogenic microorganisms, including Ellin6067, Acidibacter, Fusarium, Tetracladium, Alternaria, and Ilyonectria in ginseng rhizosphere soil. PB co-bacterial agents enhanced the function of microbial metabolic pathways, biosynthesis of secondary metabolites, biosynthesis of antibiotics, biosynthesis of amino acids, carbon fixation pathways in prokaryotes, DNA replication, and terpenoid backbone biosynthesis, and decreased the function of microbial plant pathogens and animal pathogens. Conclusion: The combination of P. polymyxa and B. cereus may be a potential biocontrol agent to promote the resistance of ginseng to disease and improve the yield, quality, and pesticide degradation.
Impact of rhizosphere microorganisms on arsenic (As) transformation and accumulation in a traditional Chinese medical plant

Abstract:Panax notoginseng is an important traditional medicinal plant, but the commercial value is threatened by root-rot disease caused by rhizosphere microbes and a potential health risk caused by plant arsenic (As) accumulation. Whether rhizospheric microbes isolated from P. notoginseng rhizosphere soil could impact As uptake and transport into P. notoginseng is not yet known. Among the three root-rot disease-causing pathogens Fusarium flocciferum (PG 1), Fusarium oxysporum (PG 2), and Fusarium solani (PG 3) and one root-rot disease biocontrol fungus Trichoderma koningiopsis (FC 1) and five biocontrol-exerting bacterial species Bacillus siamensis (BC 1), Delftia acidovorans (BC 2), Brevibacillus formosus (BC 3), Mortierella alpine (BC 4), and Bacillus subtilis (BC 5), one As-resistant pathogen and four biocontrol microorganisms with As-resistant ability were identified. The As-transforming ability of the identified fungi and bacteria was ranked in the order of FC 1 > PG 1 and BC 2 > BC 3 > BC 1, respectively. Then, the As-resistant biocontrol and pathogenic microbes were initiated to colonize the rhizosphere of 1-year-old P. notoginseng seedlings growing in artificially As(V)-contaminated soil to evaluate the impact of microbe inoculation on P. notoginseng As uptake and transport capacity. Concentration of As in P. notoginseng tissues decreased in the order of the sequence stem > root > leaf. Compared to treatment without colonization by microorganism, inoculation with microorganisms increased As root uptake efficiency and root As concentration, especially under treatment of inoculation by BC 2 and PG 1 + BC 2. As transport efficiency from root to stem decreased by inoculation with microorganism, especially under treatment with inoculation of BC 2 and PG 1 + BC 2. However, the impact of microorganism colonization on As stem to leaf transport efficiency was not obvious. In summary, inoculation with rhizosphere microbes may increase As accumulation in P. notoginseng root, especially when using bacteria with high As transformation ability. Therefore, it is necessary to evaluate the As transformation capacity before applying biological control microorganism to the rhizosphere of P. notoginseng.

Abstract:Bioorganic fertilizers can alleviate (a) biotic stresses and sustainably increase crop yields. The effect of bioorganic fertilizers on the rhizosphere bacterial community of Panax notoginseng and soil metabolism remains unknown. Here, we tracked the changes in the soil physicochemical properties, bacterial microbiota responses, and soil metabolic functions after the addition of a bioorganic fertilizer in a P. notoginseng field. The application of a bioorganic fertilizer reduced the soil acidification, improved the organic matter, and increased the contents of the total/available soil nutrients. Soil amendment with a bioorganic fertilizer significantly affected the structure of the rhizosphere bacterial community, leading to the enrichment of specific bacterial consortia such as Rhodanobacter, Arthrobacter, Sphingomonas, Devosia, Pseudolabrys, Luteimonas, Lysobacter, Nitrosospira, and Nakamurella. Previously, many of these genera have been associated with nutrient cycling, plant productivity, and disease suppression. Metabolome analysis further highlighted that the bioorganic fertilizer treatment significantly reduced phenolic acids and flavonoids and enhanced organic acids, saccharides and alcohols, and amino acids. This result indicates a high survival of bacterial microbiota in the rhizosphere and an availability of nutrients for P. notoginseng growth. This work showed that the application of bioorganic fertilizers significantly improves soil health status, alters soil metabolic functions, and stimulates a specific subset of rhizosphere microbiota for nutrient cycling and disease protection in P. notoginseng.

Abstract:There is currently increasing pressure on agriculture to simultaneously remediate soil and ensure safe agricultural production. In this study, we investigate the potential of a novel combination of biochar and plant growth-promoting bacteria (PGPB) as a promising approach. Two types of biochar, corn stover and rice husk-derived, were used in combination with a PGPB strain, Bacillus sp. PGP5, to remediate Cd and Pb co-contaminated soil and enhance lettuce performance. The contaminated soil was pre-incubated with biochar prior to PGP5 inoculation. The combined application of biochar and PGPB reduced the diethylenetriaminepentaacetic acid (DTPA) -extractable Cd and Pb concentrations in the soil by 46.45%-55.96% and 42.08%-44.83%, respectively. Additionally, this combined application increased lettuce yield by 23.37%-65.39% and decreased Cd and Pb concentrations in the edible parts of the lettuce by 57.39%-68.04% and 13.57%-32.50%. The combined application showed a better promotion on lettuce growth by facilitating chlorophyll synthesis and reducing oxidative stress. These demonstrated a synergistic effect between biochar and PGPB. Furthermore, our study elucidated the specific role of the biochar-PGPB combination in soil microbial communities. Biochar application promoted the survival of PGP5 in the soil. The impact of biochar or PGPB on microbial communities was found to be most significant in the early stage, while the development of plants had a greater influence on rhizosphere microbial communities in later stage. Plants showed a tendency to recruit plant-associated microbes, such as Cyanobacteria, to facilitate growth processes. Notably, the combined application of biochar and PGPB expedited the assembly of microbial communities, enabling them more closely with the rhizosphere microbial communities in late stage of plant development and thus enhancing their effects on promoting plant growth. This study highlights the "accelerating" advantage of the biochar-PGPB combination in the assembly of rhizosphere microbiomes and offers a new strategy for simultaneous soil remediation and safe agricultural production.

Abstract:ABSTRACT Black shank disease in tobacco, caused by Phytophthora nicotianae , can lead to yield losses of 30%–50% upon outbreak. Recently, biochar derived from agricultural waste has shown significant potential in controlling soil-borne diseases, though its mechanisms remain unclear. Over a 3-year observation period, we found that the incidence of black shank was significantly lower in plots amended with biochar compared with normal cultivation plots. To investigate the underlying mechanisms, we studied both the direct and indirect effects of biochar on black shank. Direct antifungal assays indicated that biochar reduced the total number of sporangia by 53.91%. Further pot experiments revealed a 62.34% reduction in the P. nicotianae population in the soil following biochar application. Additionally, biochar application led to notable changes in soil physicochemical properties and microbial community composition. Microbial species analysis showed that biochar promoted the aggregation of beneficial microbes such as Sphingomonas , Flavisolibacter , and Mucoromycota . Functional predictions using the PICRUSt 2 software revealed that biochar enhances bacterial functions related to antimicrobial substance synthesis (Tetracycline biosynthesis), detoxification metabolism (D-arginine and D-ornithine metabolism, arginine and proline metabolism), and lipid and fatty acid metabolism (Lipopolysaccharide biosynthesis, fatty acid biosynthesis), while fungal functions showed no significant changes. This suggests that rhizosphere bacteria play a more prominent role in the suppression of black shank by biochar, a finding supported by partial least squares path modeling analysis. Therefore, we hypothesize that biochar not only directly inhibits P. nicotianae growth but also regulates the composition of the rhizosphere microbial community, inducing the production of antimicrobial substances by rhizosphere bacteria, effectively preventing P. nicotianae invasion. IMPORTANCE Black shank, a global soil-borne fungal disease in tobacco, currently lacks effective control methods. Notably, biochar derived from agricultural waste has shown significant potential in controlling soil-borne diseases. Over a 3-year observation period, we found that plots amended with biochar had a significantly lower incidence of black shank compared with normal cultivation plots. However, the mechanisms of disease suppression remained unclear. Through in vitro antifungal assays and pot experiments, we discovered that tobacco-derived biochar can directly inhibit the growth of the pathogen. Additionally, biochar regulates the composition of the rhizosphere microbial community, inducing rhizosphere bacteria to produce antimicrobial substances, effectively preventing pathogen invasion. This discovery reveals both the direct and indirect mechanisms by which biochar suppresses black shank in tobacco. It provides a scientific basis for developing green control technologies for black shank and offers theoretical support for the application of biochar in managing soil-borne diseases in tobacco cultivation areas.

Abstract:Rusty root rot is the most destructive disease in ginseng production. The bacterial strain FG14 isolated from the rhizosphere of Panax ginseng was characterised as Bacillus amyloliquefaciens. The inhibition rates of the FG14 bacterial suspension against the mycelial growth, spore production and spore germination of Ilyonectria robusta which causes ginseng rusty root rot were all more than 90% at a concentration of 108 cfu/mL. The sterile fermentation broth of strain FG14 also had significant inhibitory ability. The results of plantgrowth promotion showed that the fresh weight and root length of ginseng seedlings treated with 3 × 108 cfu/mL FG14 bacterial suspension were 4.49 times and 2.12 times greater than the seedlings treated with I. robusta. The soil enzyme activities of urease, phosphatase, invertase, and catalase of strain FG14 and the expression of seven lipopeptide biosynthesis genes were up-regulated, and antifungal metabolite production was stable. The diversity and abundance of bacterial and fungal communities in rhizosphere soil changed during different growth stages of ginseng after inoculation with strain FG14. The relative abundance of the Ilyonectria genus was significantly lower than the water control at the leaf spreading stage of P. ginseng. These results indicate the great potential of B. amyloliquefaciens FG14 for use as a biological agent for the green control of ginseng rusty root rot.

Impact of Biochar on Rhizosphere Bacterial Diversity Restoration Following Chloropicrin Fumigation of Planted Soil

Abstract:Chloropicrin (CP) can effectively combat soil-borne diseases but has significant side effects on nontarget microorganisms. The rhizosphere microflora play a crucial role in promoting plant growth and protecting plants from infection by soil-borne pathogens. We conducted a laboratory pot experiment to evaluate the effect of CP on the rhizosphere soil bacterial flora and the effect of biochar amendments on the reconstruction of microbial communities. Our results show that CP fumigation and biochar additions promoted the growth of cucumber plants in the later stage of the pot experiment. CP significantly inhibited the rhizobacterial diversity and changed the community composition. Biochar amendments after CP fumigation shortened the time for the rhizobacterial diversity to recover to unfumigated levels. Biochar amendments promoted the transplantation of new populations to empty microbiome niches that were caused by CP and, in particular, stimulated many beneficial microorganisms to become the predominant flora. The relative abundances of many functional taxa related to plant-disease suppressiveness and pollutant bioremediation increased, including Pseudomonas, Stenotrophomonas, Bacillus, Massilia, Acinetobacter, Delftia, Micromonospora, Cytophagaceae, and Flavisolibacter. These changes stimulated by biochar amendments would promote multifunctionality in the soil rhizosphere and benefit plant growth and disease resistance.
Improved ginseng production under continuous cropping through soil health reinforcement and rhizosphere microbial manipulation with biochar: a field study of Panax ginseng from Northeast China

Abstract:ABSTRACT The production of ginseng, an important Chinese medicine crop, has been increasingly challenged by soil degradation and pathogenic disease under continuous cropping in Northeast China. In a field experiment, an Alfisol garden continuously cropped with Chinese ginseng (Panax ginseng C. A. Meyer) was treated with soil amendment at 20 t ha−1 with maize (MB) and wood (WB) biochar, respectively, compared to conventional manure compost (MC). Two years after the amendment, the rooted topsoil and ginseng plants were sampled. The changes in soil fertility and health, particularly in the soil microbial community and root disease incidence, and in ginseng growth and quality were portrayed using soil physico-chemical assays, biochemical assays of extracellular enzyme activities and gene sequencing assays as well as ginsenoside assays. Topsoil fertility was improved by 23% and 39%, ginseng root biomass increased by 25% and 27%, and root quality improved by 6% and 18% with WB and MB, respectively, compared to MC. In the ginseng rhizosphere, fungal abundance increased by 96% and 384%, with a significant and insignificant increase in bacterial abundance, respectively, under WB and MB. Specifically, the abundance of Fusarium spp. was significantly reduced by 19–35%, while that of Burkholderia spp. increased by folds under biochar amendments over MC. Relevantly, there was a significant decrease in the abundance proportion of pathotrophic fungi but a great increase in that of arbuscular mycorrhizal fungi, along with an enhanced microbial community network complexity, especially fungal community complexity, under biochar amendments. Thus, biochar, particularly from maize residue, could promote ginseng quality production while enhancing soil health and ecological services, including carbon sequestration, in continuously cropped fields.

Abstract:Excessive N critically depresses soil properties. Biochar has the potential to enhance soil health. A two-year field experiment was conducted to analyze the effects of biochar (0, 10, 20 and 40 t ha −1 , namely, B0, B10, B20 and B40) combined with N (180, 144 and 108 kg ha −1 , namely, N100, N80 and N60) on soil microorganisms. Results showed that B10 significantly promoted soil microbial biomass and had a strong correlation with most microbial groups, mainly reflected in a15:00, 10Me17:0, i18:0, i19:0, i16:0, 16:1w5c, 10Me18:0, 18:0 2OH and 18:00, while N60 decreased fungi and actinomycete content. Soil microbial functional activity was also significantly increased under B10 and B20 treatments and decreased under N80 and N60 treatments, compared to B0 treatment and N100 treatment, respectively. Microbial functional composition was also changed greatly by biochar, and B10 and B20 were closely related to most carbon sources. The combination of B10 and N80 significantly increased soil microbial biomass (total PLFA by 1.4 nM kg −1 ) and functional activity (Shannon index and Evenness index by 0.8%) compared with conventional N management (N100B0). Our findings reveal that biochar can achieve N reduction and enhance soil microbial diversity and community stability in purple soil regions of southwest China.

Abstract:Applying plant growth-promoting rhizobacteria (PGPR) improves the efficiency of soil-borne disease control and is considered a sustainable practice. However, the effect of PGPR on the fungal community, especially pathogenic fungi and arbuscular mycorrhizal fungi (AMF), remains unclear. In this study, we examined the effects of a compound microbial agent (consisting of Bacillus subtilis HG-15 and Bacillus velezensis JC-K3) on the incidence and yield of wheat under low salt stress, as well as compared the diversity and community composition of the rhizosphere fungal and AMF communities of wheat in the CK (not inoculated bacterial agent) and BIO (inoculated with a bacterial agent) groups. Chlorophyll relative content (SPAD), net photosynthesis rate ( Pn ), transpiration rate ( Tr ), leaf water use efficiency ( WUE L ), grains per spike and wheat yield in the BIO group increased more than in the CK group. The number of diseased plants and disease incidence was observed to be reduced. The relative efficacy reached 79.80%. We classified 1007 fungal operational taxonomic units (OTU) based on Miseq sequencing data: 11 phyla, 173 families, 319 genera, and 521 species. Fifty-four OTUs were classified from the AMF effective sequences, including 1 phylum, 3 families, 3 genera, and 17 species. The inoculation of bacterial agents reduced the relative abundance of pathogen genera such as Gibberella , Fusarium , Cladosporium , and Alternaria in wheat rhizosphere. It increased the relative abundance of AMF species such as Glomus-group-B-Glomus-lamellosu-VTX00193, Glomus-viscosum-VTX00063, and Glomus-Glo2-VTX00280. In addition, pH, EC, exchangeable K, available N, total N, organic matter, and olsen P were the main driving forces for shaping wheat rhizosphere fungi. The pH value was positively correlated with the relative abundance of fungal communities in soil, especially Gibberella , Cladosporium , Fusarium , and Alternaria . In summary, inoculation with Bacillus subtilis HG-15 and Bacillus velezensis JC-K3 affected wheat yield, incidence, rhizosphere soil chemical properties, rhizosphere fungi, and AMF fungal diversity and community. The findings may provide a theoretical foundation and strain support for constructing efficient PGPR-community and clarifying its mechanism of pathogenic bacteria inhibition.

Abstract:Usher's syndrome (US) is an autosomal recessive disorder characterized by congenital sensorineural hearing loss and retinitis pigmentosa. The majority of affected individuals have severe to profound sensorineural hearing loss and are therefore potential cochlear implant candidates. A goal of our otology program has been early identification and implantation of children with US. At our center, early diagnosis of US has been achieved by comprehensive ophthalmologic evaluation including electroretinography. Four of our 19 pediatric cochlear implant recipients with congenital sensorineural hearing loss were identified with US and implanted prior to the onset of signs or symptoms of visual loss. All children have received measurable benefit from cochlear implantation. In light of the dual sensory deficits present in US, the need for early diagnosis and cochlear implantation is of special importance in this population.
