Abstract:Lotus rhizome soup, renowned as a unique delicacy made from lotus rhizome, is cherished by many consumers. However, the varying colors of soup are sure to influence individuals' appetites. This research focused on the Miancheng lotus Rhizome (MLR) and Elian No. 6 lotus rhizome (ELR) to investigate the relationship between the variations in polyphenols, organic acids and iron levels in MLR and ELR soups during the cooking process. The findings indicated that cooking MLR for 12 min resulted in a red soup with a* value of 8.65 ± 0.12, whereas the soup made from ELR remained white with a* value of 3.43 ± 0.08. The correlation analysis results indicated that polyphenols, specifically epigallocatechin (0.0169 ± 0.00029 μg/100 mL FW after cooking for 12 min) and epicatechin (0.0211 ± 0.00047 μg/100 mL FW after cooking for 12 min), exhibited a significant positive relationship with a* (p < 0.05). Moreover, lowering the pH, removing polyphenols and incorporating metal-chelating agents can also prevent the development of red pigment. The analysis from HPLC-MS, UV-Vis, FT-IR spectra and ESI-Q-TOF-MS indicated that the development of the red soup color involved i) the generation of epicatechin gallate through the enantiomeric reaction of epigallocatechin, ii) the co-chromic red interaction between anthocyanin cations and catechol structures, and iii) the formation of polyphenol oligomers (i.e., procyanidin A2, (+)-procyanidin B2, procyanidin C1 and prodelphinidin B4) due to heating, while the creation of phenol-iron chelates could inhibit the development of red coloration. In sum, this research introduces a new idea for managing the color of lotus rhizome soup and similar soup products.
Formation mechanism of blue pigment in boiled lotus rhizome discs: Insight into the chelation of polyphenols and iron

Abstract:Boiled lotus rhizome discs (BLRDs), as common processed products of lotus rhizome, have gained increasing attention from consumers and food manufacturers. However, the blue pigment formed during boiling affects its appearance and reduces the appetite of BLRDs. In this study, the effects of polyphenols and iron contents on blue pigment formation in BLRDs in different regions and months were investigated. Results revealed that blue variation was more serious in March and April of the second year in Wuhan, and polyphenols and iron contents in these two months were significantly higher than those in other months. Then, UPLC and UV-Vis analysis showed that polyphenols causing the formation of blue pigment in BLRDs were L-dopa, gallocatechin, catechin, epigallocatechin, chlorogenic acid and epicatechin, among which L-dopa (52.450 mg/100 g in fresh lotus rhizome (FLR)) and gallocatechin (36.210 mg/100 g in FLR) possessed the greatest effect. Moreover, the ESI-Q-TOF-MS analysis of L-dopa-iron chelate and gallocatechin-iron chelate suggested that the blue pigment of BLRDs was mainly in the form of bis-complexes under boiling conditions. The study on formation mechanism of blue pigment in BLRDs can provide a reference for lotus rhizome processing.
Green Extraction of Phenolic Compounds from Lotus Seedpod (receptaculum Nelumbinis) Assisted by Ultrasound Coupled with Glycerol

Abstract:Lotus Receptaculum Nelumbinis has been sparking wide research interests due to its rich phenolic compounds. In the present work, ultrasonic-assisted extraction coupled with glycerol was employed to extract phenolic compounds from Receptaculum Nelumbinis and the process was optimized using a response surface methodology with Box-Behnken design (BBD). The optimal conditions for the total phenolic content (TPC) extract were obtained: glycerol concentration of 40%, an extraction temperature of 66 °C, ultrasonic time of 44 min, and the solvent-to-solid ratio of 55 mL/g. Under these optimum extraction conditions, the extraction yield of TPC was 92.84 ± 2.13 mg gallic acid equivalents (GAE) /g. Besides, the antioxidant activities demonstrated the ability of free radical scavenging by four different methods that included 2,2-Diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and reducing activity (RA) were 459.73 ± 7.07, 529.97 ± 7.30, 907.61 ± 20.28, and 983.66 ± 11.80 μmol TE/g, respectively. Six phenolic compounds were identified by ultra-high pressure liquid chromatography combined with triple-time-of-flight mass spectrophotometry (UPLC-Triple-TOF/MS) from the extracts. Meanwhile, Fourier transform infrared (FTIR) was conducted to identify the characteristic functional groups of the extracts and thus reflected the presence of polyphenols and flavonoids. Scanning electron microscopy (SEM) illustrated the microstructure difference of four treatments, which might explain the relationships between antioxidant activities and the structures of phenolic compounds.
Evaluation of the Antioxidant Properties of Litchi Fruit Phenolics in Relation to Pericarp Browning Prevention.

Abstract:Phenolics were extracted from litchi fruit pericarp (LFP) tissues, purified and their antioxidant properties analyzed. LFP phenolics strongly inhibited linoleic acid oxidation and exhibited a dose-dependent free-radical scavenging activity against α,α-diphenyl-β-picrylhydrazyl (DPPH·) and hydroxyl radicals and superoxide anions. The degradation of deoxyribose by hydroxyl radicals was inhibited by phenolics acting mainly as iron ion chelators, rather than by directly scavenging the radicals. Phenolics from litchi fruit pericarp were found to display similar reducing power activity as ascorbic acid. The effect of phenolic compound treatment on pericarp browning and electrolyte leakage of litchi fruit was also evaluated and it was observed that application of exogenous litchi phenolics to harvested litchi fruit significantly prevented pericarp browning and delayed increases in electrolyte leakage. These results suggest that litchi pericarp tissue phenolics could be beneficial in scavenging free radicals, maintaining membrane integrity and, thereby inhibiting pericarp browning of litchi fruit.

Comprehensive Reviews in Food Science and Food Safety

Abstract:Nelumbo nucifera Gaertn., also known as the sacred lotus, is extensively cultivated in Southeast Asia, primarily for food and as an herbal medicine. This article reviews studies published between 1995 and 2017, on flavonoid and phenolic acid profiles and contents of 154 different cultivars of lotus. So far, some 12 phenolic acids and 89 to 90 flavonoids (47 flavonols, 25 to 26 flavons, 8 flavan-3-ols, 4 flavanons, and 5 anthocyanins) have been isolated from different parts of the lotus plant, including its leaves (whole leaf, leaf pulp, leaf vein, and leaf stalk), seeds (seedpod, epicarp, coat, kernel, and embryo), and flowers (stamen, petal, pistil, and stalk), although not all of them have been quantified. Factors affecting flavonoids and phenolic acid profiles, including types of tissues and extracting factors, are discussed in this review, in order to maximize the application of the lotus and its polyphenols in the food industry. Health promotion activities, attributed to the presence of flavonoids and phenolic acids, are described along with toxicology studies, illustrating appropriate usage and safe consumption dosages of lotus extracts. This review also presents the controversies and discusses the research gaps that limit our ability to obtain a thorough understanding of the bioactivities of lotus extracts.
Improvement of Phenolic Compounds Extraction from High-Starch Lotus (nelumbo Nucifera G.) Seed Kernels Using Glycerol: New Insights to Amylose/amylopectin - Phenolic Relationships

Abstract:Lotus seed kernel (LSK) has high starch content and possesses many bioactivities, which are mainly attributed to its phenolics. In the present study, LSK phenolics were extracted using aqueous glycerol, and the obtained phenolics and its starch properties were observed for their correlations. Response surface methodology indicated significant quadratic effect of temperature and significant linear effect of glycerol on LSK phenolics (p < 0.05). Temperature also had a significant effect on LSK starch solubility and swelling power and % leaching amylose (p < 0.05). Leaching amylose in the presence of glycerol and swelling power showed positive correlations with LSK phenolics in a linear relationship (r = 0.825, p < 0.01) below 50 degrees C and a monotonic relationship (r(s) = 0.933, p < 0.01) above 60 degrees C, respectively. A building block backbone model was adopted to illustrate the interaction between amylose/amylopectin-glycerol, and most importantly, their relationship to LSK phenolics.
Recovery of Lotus (nelumbo Nucifera Gaertn.) Seedpod Flavonoids Using Polar Macroporous Resins: the Updated Understanding on Adsorption/desorption Mechanisms and the Involved Intermolecular Attractions and Bonding

Abstract:Macroporous resins have been employed in the effective recovery of flavonoids from plants. In this study, S8 polar resins were used to recover flavonoids and procyanidins from lotus seedpods. Adsorption kinetics, isotherms, and thermodynamics studies revealed that the adsorption process involved physico-chemical interactions, including flavonoid-resin and flavonoid-flavonoid electrostatic interactions, pi - pi aromatic stacking, moderate and strong hydrogen bonding, and repulsive forces. These forces worked complementarily in adsorption, except for the repulsive force, which opposed the adsorption. Further, adsorption temperature determined the adsorption behavior, with multilayer adsorption enhancing adsorption capacity. In dynamic desorption tests, an acetone/water/acetic acid mixture (58.77: 39.34: 1.89) designed by the D-optimal design method was able to desorb 95.57% and 89.85% of total flavonoids and procyanidins, respectively, using less than two bed volumes of solvent. Ultra-performance liquid chromatography triple-time of flight/mass spectrometry (UPLC-TOF/MS) analysis showed that 26 flavonoids, including 5 procyanidins, were detected after the recovery.

Abstract:In this paper, the gas-assisted combined with glycerol extraction (GAGE) for polyphenol recovery from lotus seedpods (LSPs) was modeled and optimized. Box–Behnken design was applied to optimize the total polyphenol content (TPC) of LSP along with enhancing antioxidant activities using response surface methodology based on the TPC extraction yield (%), which was affected by glycerol concentration, time, temperature, and glycerol-to-solid ratio. The optimal conditions for the LSP extract were glycerol-to-solid ratio, 42 mL/g; time, 50 min; concentration of glycerol, 45%; and temperature, 70 °C. Ultra-high-pressure liquid chromatography integrated with triple-time-of-flight mass spectrophotometry (UPLC-Triple-TOF/MS) analysis revealed nine biologically active polyphenols. Furthermore, Fourier-transform infrared spectroscopy and scanning electron microscopy results demonstrated the effect and influence during extraction. The findings suggested that GAGE is a potential, green, and high-efficiency alternative that could be used to recover polyphenols from plant source byproducts.

Abstract:In this study, the synthesis parameters of the lotus root polysaccharide iron complex (LRPF) were determined and optimized by response surface methodology. Under the optimum preparation conditions, the pH of the solution was 9, the ratio of M (trisodium citrate): m (lotus root polysaccharide) was 0.45, the reaction time was 3 h. UV spectroscopy, thermogravimetry, FT-IR spectroscopy, X-ray diffraction, CD, and NMR were used for the characterization of the LRPF. LRPF has good stability and easily releases iron ions under artificial gastrointestinal conditions. LRPF exhibited antioxidant activity in vitro and can significantly improve the antioxidant activity in vivo. In addition, LRPF has a good effect in the treatment of iron deficiency anemia in model mice, impacts the gut microbiome, and reduces the iron deficiency-induced perniciousness by regulating steroid hormone biosynthesis. Therefore, LRPF can be used as a nutritional supplement to treat and prevent iron-deficiency anemia and improve human immunity.

Ultrasound-assisted extraction of polyphenols from lotus rhizome epidermis by alcohol/salt-based aqueous two-phase system: Optimization, extraction mechanism and antioxidant activities

Abstract:In this study, ultrasonic-assisted (UA) alcohol/salt-based aqueous two-phase system (ATPS) method was constructed to extract lotus rhizome epidermis (LRE) polyphenols. The extraction conditions were optimized as salt concentration 26.75 %, ethanol concentration 25.45 %, ultrasonic power 487 W and liquid-solid ratio 35.33 mL/g by comparing response surface methodology (RSM) and artificial neural network (ANN) models. Then, l-dopa (2.35 ± 0.036 mg/g dw), gallocatechin (1.66 ± 0.0035 mg/g dw) and epigallocatechin (1.37 ± 0.0035 mg/g dw) were determined as major polyphenols in LRE by using UA-ATPS method. Moreover, study showed that ultrasound, van der Waals force, hydrogen bond and salting out could accelerate the mass transfer and extraction of polyphenols in LRE cells. The high-pressure cavity and collapse effect of ultrasound could also accelerate the extraction of polyphenols. In vitro antioxidant experiments showed that LRE polyphenols have good antioxidant ability. In sum, this study developed a green and efficient extraction method to enhance the profitability of LRE in food and medicine industries.
Antioxidant, Bacteriostatic and Preservative Effects of Extractable Condensed Tannins Isolated from Longan Pericarps and Seeds

Abstract:In the process of longan production and processing, a large amount of remnants is produced, such as dried longan pericarps and seeds, which have been reported to be rich in polyphenols but not effectively utilized. In this paper, the total phenolic contents in the remnants of longan pericarps and seeds were found to be 39.58 ± 3.54 and 69.53 ± 1.99 mg/g (DW), respectively, accounting for 60–80% of those in fresh samples. The contents of extractable condensed tannins (ECTs) in the remnants of longan pericarps and seeds were 19.25 ± 6.71 mg/g (DW) and 44.59 ± 2.05 mg/g (DW), respectively, accounting for 60–70% of the fresh samples. These data indicate that the polyphenols in the remnants of the sampled longan pericarps and seeds were effectively retained. The antioxidant capacity of ECTs from the longan pericarps and seeds was more than 60% of the fresh samples measured with the 1, 1-diphenyl-2-trinitrophenylhydrazine and ferric reducing ability of plasma methods. Further exploration showed that ECTs from the longan pericarps and seeds had significant inhibitory effects on Pseudomonas aeruginosa, Escherichia coli, Salmonella and Staphylococcus aureus. The minimum inhibitory concentration (MIC) of the longan pericarp ECTs on all four studied bacteria was 3 mg/mL. The MIC of longan seed ECTs on Salmonella was 3 mg/mL, and that of the other three bacteria was 1.5 mg/mL. In view of the good antioxidant and antibacterial activities of longan pericarps and seeds, we applied them to the preservation of fresh-cut lotus roots. When the concentration of ECTs in the longan pericarps and seeds was 2 mg/mL and 1 mg/mL, respectively, the two kinds of ECTs showed an obvious preservative effect. After the ECT treatment of the lotus roots, their browning degree was reduced, their color was better maintained, their respiration was inhibited and their nutrient loss was reduced. Bacterial reproduction was inhibited, and cell senescence was slowed. Accordingly, the shelf life of ECT-treated fruits and vegetables can be effectively extended. Overall, we can suggest that ECTs from the remnants of dried longan pericarps and seeds could be used as natural preservatives for fresh-cut fruits and vegetables.

Abstract:Objective: To optimize the extracting process of procyanidins and nuciferine from the lotus leaf simultaneously.Method: Orthogonal design was used to optimize the extracting process of procyanidins and nuciferine with the transfer rate as the evaluation index, The effect of several important factors such as the pH of extracting solvent,the volume fraction of acid ,the extraction time,the material-liquid ratio and the extract times of procyanidins and nuciferine were studied. Result.. The optimal extraction process was as follows: 0.3% (v/v)of phosphate water solution ; the material- liquid ratio was 1 : 25 ; two times with 90min per time. The transfer rate of procyanidins was 89.2%, while the transfer rate of nuciferine was 69.9%. Conclusion: The optimum extraction processes were stable, economical ,feasible and adapts for industrialized production.
Regulations and mechanisms of 1-methylcyclopropene treatment on browning and quality of fresh-cut lotus (Nelumbo nucifera Gaertn.) root slices

Abstract:The quality of fresh-cut lotus root slices is greatly affected by enzymatic browning. In this study, we investigated the effects of 1-methylcyclopropene treatment on browning and quality deterioration of lotus root slices. When fresh-cut lotus root slices were immersed in 0.1 mg L−1 1-MCP for 1 h and stored at 10 ℃ for twelve days, microbial growth, respiration, total phenolic content, phenylalanine ammonia lyase activity, polyphenol oxidase activity, and soluble quinone levels in fresh-cut lotus root slices were suppressed. Furthermore, 1-MCP treatment improved the antioxidant capacities, peroxidase activities, superoxide dismutase activities, catalase activities, and DPPH free radical scavenging rates in fresh-cut lotus root slices. In addition, 1-MCP treatment inhibited the production of reactive oxide species, which delayed quality deterioration of fresh-cut lotus root slices. Based on transcriptomics and real-time quantitative PCR results, gene expression levels of NnmetK2, NnACO, NnETR2, NnEIN3, and NnERF1 were up-regulated while expression levels of NnEBF1 were down-regulated. These findings indicate that 1-MCP inhibits ethylene biosynthesis and signal transduction, which suppresses a series of ethylene-induced physiological and biochemical reactions. Based on our findings, we propose a new strategy for inhibiting enzymatic browning in fresh-cut lotus root slices.

Abstract:This study presents operating factors affecting yield of total phenol and ferric reducing antioxidant power (FRAP) value of extracts from lotus rhizome using orthogonal design experiment (L-9(3(4))) based on single factor experiments. Optimum extraction conditions on yield of total phenol and FRAP value, respectively, were found as follows: ratio of material to liquid, 1 g: 6 ml, 1 g: 8 ml; extraction temperature, 70 degrees C, 50 degrees C; extraction time, 2 h, 0.5 h; and ethanol concentration, 40%, 40%.

Abstract:Lotus (Nelumbo) leaves are rich in polyphenols and dietary fiber, which have the potential as a high-quality fiber material in functional food. However, lotus leaves exhibit dense structure and poor taste, it is vital to develop appropriate modification methods to improve the properties of lotus leaves dietary fiber. In this study, the effects of three modification methods with shear emulsifying(SE), ball milling (BM), and autoclave treatment (AT) on structure, physicochemical properties, phenolic compounds, and antioxidant capacity of lotus leave dietary fiber (LDF) were evaluated. SEM indicated that there were significant differences in the microstructure of modified LDFs. FT-IR spectra and X-ray diffraction pattern of modified LDFs revealed similar shapes, while the peak intensity and crystalline region changed by modification. SE showed the greatest effect on crystallization index. SE-LDF had the highest water holding capacity, water swelling capacity, and bound phenolic content in LDFs, which increased by 15.69, 12.02, and 31.81%, respectively, compared with the unmodified LDF. BM exhibited the most dramatic effect on particle size. BM-LDF had the highest free phenolic and total phenolic contents in LDFs, which increased by 32.20 and 29.05% respectively, compared with the unmodified LDF. Phenolic compounds in LDFs were mainly free phenolic, and modifications altered the concents of flavonoids. The BM-LDF and SE-LDF exhibited higher antioxidant capacity than that of AT-LDF. Overall, SE-LDF showed better physical properties, and BM-LDF showed better bioactive components. SE and BM were considered to be appropriate modification methods to enhance the properties of LDF with their own advantages

Abstract:Lotus rhizome powder was extracted with solvents of different polarities. Antioxidant activities of the extracts were evaluated by a 2, 2'-diphenylpicylhydrazyl (DPPH) assay and a beta-carotene bleaching assay, and compared with that of butylated hydroxyanisole (BHA) and ascorbic acid. Methanol showed the highest extract yield among, all of solvents. Although acetone extract had the highest total phenolics content, methanol extract had the highest total phenolics recovery from lotus powder (20.1 mg catechin equivalents/100g lotus powder). Extract of either methanol or acetone demonstrated the highest DPPH scavenging activity at both 66,7 mg/L and 133.3 mg/L. All extracts exhibited higher antioxidant activity coefficient (AAC) than that of ascorbic acid, furthermore, dichloromethane and petroleum extracts had comparable AAC with BHA by the P-carotene bleaching assay. The properties of the extracting solvents significantly affected the yield, total phenolics content and antioxidant activity of lotus rhizome extracts.

Abstract:Abstract In this study, the antioxidant activities of different parts of lotus (Nelumbo nuficera Gaertn) rhizome were compared. The total phenolic content of lotus rhizome was determined, and Ferric reducing antioxidant power (FRAP) assay, 1,1-diphenyl-2-picrylhydrazyl hydrate (DPPH) radical-scavenging assay and β-carotene-linoleic acid assay were performed to assess the antioxidant activity of lotus rhizome. Results showed that there was a significant difference in total phenolic content and antioxidant activity between any two of four parts of lotus rhizome. The order of total phenolic content and antioxidant activity in different parts of lotus rhizome was as follows: peel of old lotus rhizome > peel of young lotus rhizome > flesh of old lotus rhizome > flesh of young lotus rhizome. The total phenol content is significantly positive correlated with the antioxidant activity in different parts of lotus rhizome. This study has provided a basis for further exploring the antioxidant components in lotus rhizome.

Abstract:Callus browning is a major drawback to lotus callus proliferation and regeneration. However, the underlying mechanism of its formation remains largely unknown. Herein, we aimed to explore the metabolic and molecular basis of lotus callus browning by combining histological staining, high‐throughput metabolomics, and transcriptomic assays for lotus callus at three browning stages. Histological stained brown callus cross sections displayed severe cell death symptoms, accompanied by an obvious accumulation of polyphenols and lignified materials. Widely targeted metabolomics revealed extensively decreased accumulation of most detected flavonoids and benzylisoquinoline alkaloids (BIAs), as well as a few phenolic acids, amino acids and their derivatives in callus with browning symptoms. Conversely, the contents of most detected tannins were significantly increased. Subsequent comparative transcriptomics identified a set of differentially expressed genes (DEGs) associated with the biosynthesis and regulation of flavonoids and BIAs in lotus. Notably, callus browning was coupled with significantly up‐regulated expression of two polyphenol oxidase (PPO) and 17 peroxidase (POD) encoding genes, while the expression of ethylene associated genes remained at marginal levels. These results suggest that lotus callus browning is primarily controlled at the level of metabolism, wherein the oxidation of flavonoids and BIAs is crucially decisive.

Abstract:In this study, we extracted lotus root polysaccharide (LRP) and synthesized carboxymethylated lotus root polysaccharide (CM-LRP) using response surface methodology (RSM). The monosaccharide component of LRP was analyzed by pre-column derivatization high performance liquid chromatography (PCD-HPLC). The polysaccharide was characterized by ultraviolet (UV) spectroscopy scan, Fourier transform infrared spectroscopy (FTIR), circular dichroism spectrometer (CD), and scanning electron microscopy (SEM). It was found that the main monosaccharide component of LRP was glucose, accounting for more than 98% of the total polysaccharide component. RSM analyses revealed that the optimal conditions for CM-LRP synthesis were a reaction duration of 2.22 h, the chloroacetic acid dosage is 2.02 g, and a temperature of 46.87 °C. Under these conditions, the predicted degree of substitution (DS) was determined to be 0.5101. The superoxide anion, ferrous ion and hydroxyl radical antioxidant system were constructed to study the antioxidant activity of LRP and CMLRP. The activity of CMLRP to remove ferrous ions and hydroxyl radicals were significantly stronger than that of LRP, and it also showed a certain concentration-dependent. LRP has a better scavenging ability than CMLRP in scavenging superoxide anion free radicals. Our data revealed that CM-LRP is a promising natural antioxidant with potential value as a food supplement.

Comparison of fresh and browning lotus roots (Nelumbo nucifera Gaertn.) on modulating cholesterol metabolism via decreasing hepatic cholesterol deposition and increasing fecal bile acid excretion

Abstract:Lotus root (LR) is prone to browning after harvest due to the oxidation of phenolic compounds by polyphenol oxidase (PPO). This study compared the effects of LR extract and BLR extract on cholesterol metabolism in high-fat diet (HFD) mice. Our findings highlighted the innovative potentiality of BLR extract in effectively regulating cholesterol metabolism via inhibiting the intestinal FXR-FGF15 signaling pathway and boosting probiotics in gut microbiota, offering valuable insights for hypercholesterolemia and metabolic disorders. In detail, catechin was the main phenolic compound in LR, while after browning, theaflavin was the main oxidation product of phenolic compounds in BLR. Both the intake of LR extract and BLR extract regulated the disorder of cholesterol metabolism induced by HFD. In particular, BLR extract intake exhibited more robust effects on increasing the BAs contents synthesized in the liver and excreted in feces compared with LR extract intake. Furthermore, the consumption of BLR extract was more effective than that of LR extract in reducing the ileal protein expressions of FXR and FGF15 and shifting BAs biosynthesis from the classical pathway to the alternative pathway. Moreover, LR extract and BLR extract had distinct effects on the gut microbiota in HFD-fed mice: BLR extract significantly elevated probiotics Akkermansia abundance, while LR extract increased Lactobacillus abundance. Therefore, both LR extract and BLR extract improved the cholesterol deposition effectively and BLR extract even showed a stronger effect on regulating key gene and protein expressions of cholesterol metabolism.
